目的:探讨高危型人乳头瘤病毒(high-risk human papillomavirus,HR-HPV)检测在宫颈病变筛查中的应用价值.方法:采用液基细胞学(TCT)对1175例妇女宫颈癌及其癌前病变进行初筛,细胞学异常者或TCT正常、HR-HPV DNA阳性且高度怀疑宫颈病变患者进行阴道镜和宫颈多点活检组织病理学检查,结合病理结果分析宫颈病变.用二代杂交捕获法(hybird captureⅡ,HC-Ⅱ)对所有标本进行高危型HPV DNA的检测,对结果进行回顾性分析.结果:1175例样本中TCT检测结果正常或炎症968例,ASC-US(未明确诊断意义的不典型鳞状上皮细胞)62例,ASC-H(不典型鳞状上皮细胞,不能除外高度鳞状上皮内病变)39例,LSIL(低度鳞状上皮内病变)87例,HSIL(高度鳞状上皮内病变)19例.207例细胞学异常者经阴道镜下多点组织活检证实炎症116例,宫颈上皮内瘤变(CIN)Ⅰ级27例,Ⅱ级34例,Ⅲ级19例,浸润癌5例,湿疣6例.HC-Ⅱ法检测HR-HPV DNA发现207例细胞学异常者HPV感染率分别为:正常或炎症17.24%,CIN I 22.22%,CIN Ⅱ32.35%,CIN Ⅲ61.39%,浸润癌100.10%,湿疣30.23%.82例TCT正常、HR-HPV DNA阳性患者病理结果显示炎症67例,CIN Ⅰ 11例,CIN Ⅱ 6例.结论:随着病变的加重,HR-HPV感染率逐渐增高,其感染与宫颈病变级别相关,HR-HPV检测可辅助筛查宫颈病变,与细胞学联合检测为较好的宫颈癌筛查方案.  相似文献

探讨ASCUS细胞中p16的表达与子宫颈CIN患者的相关性。选取2014年2月至2015年10月本院经宫颈液基细胞学检查诊断为非典型鳞状细胞患者137例,所有患者均经阴道镜取活组织标本行组织病理学确诊,分析不同病检结果的非典型鳞状细胞患者的p16 mRNA和p16蛋白表达水平以及HR-HPV感染率的差异,并分析非典型鳞状细胞患者的p16 mRNA及其蛋白表达与HR-HPV感染率的相关性。研究显示,137例非典型鳞状细胞患者病理结果为阴性38例(27.74%),CINⅠ35例(25.55%),CINⅡ28例(20.44%),CINⅢ22例(16.06%),宫颈鳞癌14例(10.22%)。病检阴性、CINⅠ、CINⅡ、CINⅢ和宫颈鳞癌患者的p16 mRNA阳性表达率(10.53%,45.71%,57.14%,63.64%,71.43%)、p16蛋白阳性表达率(0%,40.0%,78.57%,90.91%,100%)、HR-HPV阳性率(0%,8.57%,35.71%,54.55%,57.14%)均随着宫颈病变程度的加深逐渐上升,且差异具有统计学意义(p0.05);HR-HPV阳性的非典型鳞状细胞患者p16 mRNA表达阳性率为84.85%,p16蛋白表达阳性率为75.76%,非典型鳞状细胞患者的p16 mRNA、p16蛋白表达阳性率与HR-HPV阳性率呈正相关(rs=0.325,p0.001和rs=0.303,p0.001)。p16 mRNA及蛋白表达阳性率随着宫颈病变程度的加深而逐渐上升,可能是由于HR-HPV感染引起p16表达增加,p16 mRNA和蛋白表达阳性率与HR-HPV阳性率呈正相关性,进一步提示了HPV16感染以及其他类型的高危HPV分型对于宫颈病变发生发展的促进作用。  相似文献

目的:评价高危型人类乳头瘤病毒(high-risk human papilloma virus,HR-HPV)DNA检测对宫颈病变的诊断价值.方法:对门诊450例细胞学异常的患者,采用杂交捕获试验法(Hybrid Capture Ⅱ,HCⅡ)检测HR-HPV-DNA,并行阴道镜检查.结果:细胞学诊断中非典型鳞状细胞332例,低度鳞状上皮内病变87例,高度鳞状上皮内病变31例,HPV-DNA阳性率分别为30.7%(102/332),44.8%(39/87),90.3%(28/31);病理学诊断中无上皮内病变或恶性病变、子宫颈上皮瘤(cervical intra-qaithelial neoplasia,CIN)Ⅰ/Ⅱ、CIN Ⅲ和原位癌HPV-DNA阳性率分别为31.2%(73/219)、47.3%(96/203)、100%(28/28),随细胞学检查的级别增高,HPV-DNA高危型阳性率增加(P<0.05).结论:高危型HPV-DNA检测是宫颈癌筛查的有效方法,建议临床推广应用.  相似文献

目的:研究不同程度子宫颈病变中高危型人乳头瘤病毒HR-HPV感染和端粒酶活性的表达,以探讨两者在宫颈癌及宫颈上皮内瘤变中的作用及相关性。方法:采用第二代杂交捕获技术检测宫颈脱落细胞HPV-DNA含量,并用免疫组织化学EnVision二步法检测宫颈组织标本中端粒酶的表达。结果:(1)端粒酶阳性表达率在对照组、CINⅠ、CINⅡ、CINⅢ和宫颈癌组分别为10.00%、16.67%、40.00%、70.00%、95.00%,宫颈癌组高于CINⅢ,CINⅢ高于CINⅡ,CINⅡ高于CINⅠ,差异均有统计学意义(x2=4.329,P=0.037;x2=4.327,P=0.038;x2=4.022,P=0.045)。(2)随着宫颈病变级别的增加,高危型HPV的阳性率和病毒负荷量均增高。高危型HPV的阳性率在宫颈癌和CINⅢ组明显高于对照组、CINⅠ及CINⅡ(x2=29.501~7.414,P<0.01)。高危型HPV的病毒负荷量在对照组与其他4组比较,差异均有统计学意义(P<0.05);CINⅠ组分别与CINⅡ、CINⅢ及宫颈癌组比较差异均有统计学意义(P<0.05)。(3)随着宫颈病变级别的增加,高危型HPV的阳性率和端粒酶阳性表达率依次递增,两者有明显的相关性(r=0.943,P<0.01)。结论:高危型HPV感染和端粒酶活性均与宫颈癌前病变及宫颈癌的发生发展密切相关,有望作为子宫颈癌前病变和宫颈癌筛查的监测指标。  相似文献

目的:比较酶切信号放大法(Cervista)与导流杂交基因芯片技术(Hybri Ma)检测高危型人乳头状瘤病毒(HR-HPV)诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的临床价值。方法:随机选择288例2012年3月至2013年1月在哈尔滨医科大学附属第一医院妇科门诊进行新柏氏液基细胞学检查的年龄在20~65岁的宫颈细胞学检测未明确意义的不典型鳞状细胞(ASCUS)的患者,采用Cervista技术与Hybri Ma技术进行高危型HPV检测,并对入组的患者行阴道镜下宫颈活组织检查。以病理学诊断结果为金标准,比较Cervista技术与Hybri Ma技术诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的敏感度、特异度及ROC曲线。结果:在入组的288例患者中,Cervista技术和Hybri Ma技术检出高危型HPV的阳性率分别为49.31%和51.39%(P0.05),其诊断CIN2+的敏感度分别为95.65%和91.30%(P0.05),特异度分别为59.50%和56.20%(P0.05),阳性预计值分别为30.99%和28.38%(P0.05),阴性预计值分别为98.63%和97.14%(P0.05)。两组ROC曲线下面积分别为0.776和0.738(P0.05)。结论:Cervista技术与Hybri Ma技术诊断CIN2+的临床价值相当。  相似文献

目的研究人乳头瘤病毒(HPV)在宫颈炎症、宫颈癌前病变及宫颈癌中的水平及临床意义。方法分别选取宫颈炎症278例、宫颈癌前病变174例、宫颈腺癌82例、宫颈鳞癌183例患者,并以218例宫颈无病变的病例作为对照,所有患者均行薄层液基细胞学(TCT)检查及阴道镜下病理活检,检测各组患者间HPV阳性水平并对统计结果加以分析。结果 218例宫颈无病变病例中高危性HPV阳性率为9.6%(21/218),其余717例宫颈出现病变的患者中高危型HPV筛查结果阳性例数为405例,阳性检出率56.5%,且随着宫颈病变严重程度的加重,高危型HPV检出率逐渐升高(P0.01),但是宫颈腺癌和宫颈鳞癌组的高危型HPV阳性率比较差异无统计学意义(P0.05)。结论 HPV是宫颈癌前病变及宫颈癌的主要致病因素,对宫颈炎症、宫颈癌前病变及宫颈癌患者行HPV检测对于患者早日确诊及早日治疗具有重要意义。  相似文献

目的探讨人乳头状瘤病毒(human papillomavirus,HPV)感染与宫颈高级别病变(highgrade squamous intraepithelial lesion,HSIL)的关系。方法选取2003年1月到2016年8月在首都医科大学附属北京天坛医院行宫腔镜辅助宫颈冷刀锥切术的患者181例,患者行宫腔镜辅助宫颈冷刀锥切术前均有HPV检测、薄层液基细胞学(TCT)以及阴道镜活检及免疫组化的结果。依据病检结果分析HPV感染与宫颈高级别病变的关系。结果在181例宫颈高级别病变中,HPV感染165例,HPV检测阳性率为91.16%。其中高危型HPV感染160例,阳性率为88.40%,占阳性标本的96.97%。HPV16阳性率57.46%,占高危型HPV感染的63.03%。40~49岁高危型HPV检测阳性率最高,但各年龄组比较差异均无统计学意义(各年龄组P值均0.05)。HPV感染在宫颈高级别病变和子宫颈癌中差异均有统计学意义(P0.05)。结论 HPV感染在宫颈高级别病变的发生和发展中起关键作用。高危型HPV基因分型诊断在宫颈癌筛查及防治过程中具有重要意义。  相似文献

目的:探讨乳头瘤病毒定性检测( HPV- DNA)与液基薄层细胞学检查技术(TCT)在宫颈癌前病变中的诊断价值.方法:分别采用HPV- DNA及TCT法检测1536例患者,对TCT阳性、HPV- DNA阳性病例者进行阴道镜下活检.结果:随着病理诊断级别的升高,高危型HPV感染率上升,而低危型HPV感染多见于轻度不典型增生(CINⅠ)及其以下病变,HPV混合感染在各组间未见明显趋势.HPV-DNA检测与宫颈活检的符合率为66.78%.随着病理级别的升高,不典型鳞状细胞(ASCUS)组的感染率呈降低趋势,鳞状上皮内低度病变(LSIL)组感染率亦呈下降趋势,而鳞状上皮内高度病变(HSIL)组和鳞癌(SCC)组感染率均呈上升趋势,TCT检测与组织学诊断的符合率为66.67%,与HPV-DNA检查比较,差异无统计学意义(x=0.001,P>0.05).结论:HPV-DNA、TCT及宫颈活检三者检测相结合,能明显提高诊断的正确性.  相似文献

目的:探讨高危型人乳头瘤病毒(HPV)感染对宫颈病变组织干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)的影响。方法:收集我院2013年1月到2015年1月妇科门诊行宫颈活检患者150例,根据病理检查结果,将患者分为观察组(宫颈癌33例、CINⅠ期26例、CINⅡ期28例、CINⅢ期23例)110例,对照组(慢性宫颈炎患者)40例。提取两组患者宫颈组织标本,检测观察组HPV-DNA、HPV分型,及两组IFN-γ、IL-10蛋白及m RNA表达,分析高危型HPV感染与宫颈病变组织IFN-γ、IL-10表达的关系。结果:观察组HPV-DNA阳性87例(79.1%),其中HPV16型47例(42.7%)、HPV18型20例(18.2%)感染率最高;且HPV16、18感染率在CINⅠ期、CINⅡ期、CINⅢ期以及宫颈癌组中的感染率差异均有统计学意义(P0.05),Spearman相关性分析显示,HPV16、18与宫颈病变程度均呈现正相关关系(r=0.896,0.786;均P0.05)。IFN-γ、IL-10表达水平在CINⅠ期、CINⅡ期、CINⅢ期、宫颈癌、对照组表达水平差异均具有统计学意义(P0.05),Spearman相关性分析显示,IFN-γ与宫颈病变程度呈现负相关关系(r=-0.567,P0.05),IL-10呈现正相关关系(r=0.678,P0.05)。且HPV16、HPV18感染率与IFN-γ表达呈现负相关关系(r HPV16/18=-0.678,-0.675,均P0.05),与IL-10呈现正相关关系(r HPV16/18=0.582,0.778,均P0.05)。结论:IFN-γ表达随着宫颈病变加重或者HPV16、HPV18感染率升高逐渐降低,IL-10则逐渐升高。  相似文献

目的:调查安康地区女性人乳头瘤病毒(HPV)感染的基因型别及年龄分布特征,分析其与宫颈癌的关系,为宫颈癌防治及HPV疫苗研发提供可靠的依据。方法:收集2010年6月-2012年8月间在本院及安康市部分县级医院妇产科就诊的2736名女性的液基细胞学和组织学标本,分为8个年龄组:16-24岁119例、25-29岁230例、30-34岁343例、35-39岁472例、40-44岁574例、45-49岁512例、50-54岁206例、55-86岁280例,进行病理学分类及HPV分型检测,分析不同年龄组及不同类型宫颈组织中的HPV感染率。结果:2736例女性中发生HPV感染720例(26.32%),共检出21种型别,感染率最高的基因型别是HPV16(25.05%),其他常见型别依次为HPV58、HPV52、HPV6、HPV11。单一感染占76.25%,多重感染占23.75%。HPV感染率在16-24岁、35-39岁和55-86岁三个年龄段出现高峰;而高危型HPV的感染率在35-39岁和55-86岁两个年龄段分别出现高峰。HPV的检出率随着宫颈病变的严重程度而增加,其中正常或炎症人群的HPV感染率显著低于宫颈病变及宫颈鳞状细胞癌患者(均P0.05),且意义未明的不典型鳞状细胞(ASCUS)、CIN1-3及宫颈鳞状细胞癌患者的HPV感染率对比结果存在显著差异(P0.05)。CIN1组、CIN2-CIN3组及宫颈鳞状细胞癌组单一感染率逐渐增加(P0.05),且其二重、三重感染率比较差异均有统计学意义(P0.05)。结论:安康地区HPV16型别感染较广,临床需加强对HPV16型单一感染宫颈病变患者的癌症预防工作。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献
