Background and Aims

Several strains of rhizobacteria may be found in the rhizospheric soil, on the root surface or in association with rice plants. These bacteria are able to colonize plant root systems and promote plant growth and crop yield through a variety of mechanisms. The objectives of this study were to isolate, identify, and characterize putative plant growth-promoting rhizobacteria (PGPR) associated with rice cropped in different areas of southern Brazil.

Methods

Bacterial strains were selectively isolated based on their growth on three selective semi-solid nitrogen-free media. Bacteria were identified at the genus level by PCR-RFLP 16S rRNA gene analysis and partial sequencing methodologies. Bacterial isolates were evaluated for their ability to produce indolic compounds and siderophores and to solubilize phosphate. In vitro biological nitrogen fixation and the ability to produce 1-aminocyclopropane-1-carboxylate deaminase were evaluated for each bacterial isolate used in the inoculation experiments.

Results

In total, 336 bacterial strains were isolated representing 31 different bacterial genera. Strains belonging to the genera Agrobacterium, Burkholderia, Enterobacter, and Pseudomonas were the most prominent isolates. Siderophore and indolic compounds producers were widely found among isolates, but 101 isolates were able to solubilize phosphate. Under gnotobiotic conditions, eight isolates were able to stimulate the growth of rice plants. Five of these eight isolates were also field tested in rice plants subjected to different nitrogen fertilization rates.

Conclusions

The results showed that the condition of half-fertilization plus separate inoculation with the isolates AC32 (Herbaspirillum sp.), AG15 (Burkholderia sp.), CA21 (Pseudacidovorax sp.), and UR51 (Azospirillum sp.) achieved rice growth similar to those achieved by full-fertilization without inoculation, thus highlighting the potential of these strains for formulating new bioinoculants for rice crops.  相似文献

Background and aims

Burkholderia tuberum STM678T was isolated from a South African legume, but did not renodulate this plant. Until a reliable host is found, studies on this and other interesting beta-rhizobia cannot advance. We investigated B. tuberum STM678T’s ability to induce Fix+ nodules on a small-seeded, easy-to-propagate legume (Macroptilium atropurpureum). Previous studies demonstrated that B. tuberum elicited either Fix- or Fix+ nodules on siratro, but the reasons for this difference were unexplored.

Methods

Experiments to promote effective siratro nodule formation under different environmental conditions were performed. B. tuberum STM678T’s ability to withstand high temperatures and desiccation was checked as well as its potential for promoting plant growth via mechanisms in addition to nitrogen fixation, e.g., phosphate solubilization and siderophore production. Potential genes for these activities were found in the sequenced genomes.

Results

Higher temperatures and reduced watering resulted in reliable, effective nodulation on siratro. Burkholderia spp. solubilize phosphate and produce siderophores. Genes encoding proteins potentially involved in these growth-promoting activities were detected and are described.

Conclusions

Siratro is an excellent model plant for B. tuberum STM678T. We identified genes that might be involved in the ability of diazotrophic Burkholderia species to survive harsh conditions, solubilize phosphate, and produce siderophores.  相似文献

Purpose

This study investigated the residual contribution of legume and fertilizer nitrogen (N) to a subsequent crop under the effect of elevated carbon dioxide concentration ([CO2]).

Methods

Field pea (Pisum sativum L.) was labeled in situ with 15N (by absorption of a 15N-labeled urea solution through cut tendrils) under ambient and elevated (700 μmol mol–1) [CO2] in controlled environment glasshouse chambers. Barley (Hordeum vulgare L.) and its soil were also labeled under the same conditions by addition of 15N-enriched urea to the soil. Wheat (Triticum aestivum L.) was subsequently grown to physiological maturity on the soil containing either 15N-labeled field pea residues (including 15N-labeled rhizodeposits) or 15N-labeled barley plus fertilizer 15N residues.

Results

Elevated [CO2] increased the total biomass of field pea (21 %) and N-fertilized barley (23 %), but did not significantly affect the biomass of unfertilized barley. Elevated [CO2] increased the C:N ratio of residues of field pea (18 %) and N-fertilized barley (19 %), but had no significant effect on that of unfertilized barley. Elevated [CO2] increased total biomass (11 %) and grain yield (40 %) of subsequent wheat crop regardless of rotation type in the first phase. Irrespective of [CO2], the grain yield and total N uptake by wheat following field pea were 24 % and 11 %, respectively, higher than those of the wheat following N-fertilized barley. The residual N contribution from field pea to wheat was 20 % under ambient [CO2], but dropped to 11 % under elevated [CO2], while that from fertilizer did not differ significantly between ambient [CO2] (4 %) and elevated [CO2] (5 %).

Conclusions

The relative value of legume derived N to subsequent cereals may be reduced under elevated [CO2]. However, compared to N fertilizer application, legume incorporation will be more beneficial to grain yield and N supply to subsequent cereals under future (elevated [CO2]) climates.  相似文献

Background

Endophytic diazotrophic bacteria colonize several non-leguminous plants and promote plant growth. Different mechanisms are involved in bacteria-induced plant growth promotion, including biological nitrogen fixation (BNF), mineral solubilization, production of phytohormones, and pathogen biocontrol. Herbaspirillum seropedicae is a broad-host-range endophyte that colonizes sugarcane, rice, wheat, sorghum, and maize, and has been used as a biofertilizer. Contrasting results between greenhouse and field experiments have prompted efforts to improve the consistency of the plant response to microbial stimulation.

Aims

The aim of this study was to evaluate the effect of the presence of humic substances on inoculation of maize (Zea mays L.) with H. seropedicae.

Methods

Two experiments were conducted: one in the greenhouse using sand and nutrient solution and the other a field trial in soil with low natural fertility and to which was applied N in the form of urea (50 kg ha?1). In the greenhouse, pre-emerging seeds were inoculated with a solution of H. seropedicae (109 cells mL?1) in the presence of humic substances isolated from vermicompost (10, 20, or 30 mg C?L?1); in the field trial, bacteria combined with humate were added as a foliar spray (450 L?ha?1).

Results

At early stages (7 and 45 days old) in the greenhouse, the treatment activated plant metabolism including enhancement of plasma membrane H+-ATPase activity, alteration of sugar and N metabolism, and greater net photosynthesis. The number of viable bacterial cells was higher in root tissues when inoculation was in the presence of soluble humic substances. Foliar application of endophytic diazotrophic bacteria and humic substances increased maize grain production 65 % under field conditions. These results show a promising use of humic substances to improve the benefit of endophytic diazotrophic inoculation.  相似文献

Aims

A pot study spanning four consecutive crop seasons was conducted to compare the effects of successive rice straw biochar/rice straw amendments on C sequestration and soil fertility in rice/wheat rotated paddy soil.

Methods

We adopted 4.5 t ha?1, 9.0 t ha?1 biochar and 3.75 t ha?1 straw for each crop season with an identical dose of NPK fertilizers.

Results

We found no major losses of biochar-C over the 2-year experimental period. Obvious reductions in CH4 emission were observed from rice seasons under the biochar application, despite the fact that the biochar brought more C into the soil than the straw. N2O emissions with biochar were similar to the controls without additives over the 2-year experimental period. Biochar application had positive effects on crop growth, along with positive effects on nutrient (N, P, K, Ca and Mg) uptake by crop plants and the availability of soil P, K, Ca and Mg. High levels of biochar application over the course of the crop rotation suppressed NH3 volatilization in the rice season, but stimulated it in the wheat season.

Conclusions

Converting straw to biochar followed by successive application to soil is viable for soil C sequestration, CH4 mitigation, improvements of soil and crop productivity. Biochar soil amendment influences NH3 volatilization differently in the flooded rice and upland wheat seasons, respectively.  相似文献

Background and purpose

Rapid increases in atmospheric carbon dioxide concentration ([CO2]) may increase crop residue production and carbon: nitrogen (C:N) ratio. Whether the incorporation of residues produced under elevated [CO2] will limit soil N availability and fertilizer N recovery in the plant is unknown. This study investigated the interaction between crop residue incorporation and elevated [CO2] on the growth, grain yield and the recovery of 15N-labeled fertilizer by wheat (Triticum aestivum L. cv. Yitpi) under controlled environmental conditions.

Methods

Residue for ambient and elevated [CO2] treatments, obtained from wheat grown previously under ambient and elevated [CO2], respectively, was incorporated into two soils (from a cereal-legume rotation and a cereal-fallow rotation) 1 month before the sowing of wheat. At the early vegetative stage 15N-labeled granular urea (10.22 atom%) was applied at 50 kg?N ha?1 and the wheat grown to maturity.

Results

When residue was not incorporated into the soil, elevated [CO2] increased wheat shoot (16 %) and root biomass (41 %), grain yield (19 %), total N uptake (4 %) and grain N removal (8 %). However, the positive [CO2] fertilization effect on these parameters was absent in the soil amended with residue. In the absence of residue, elevated [CO2] increased fertilizer N recovery in the plant (7 %), but when residue was incorporated elevated [CO2] decreased fertilizer N recovery.

Conclusions

A higher fertilizer application rate will be required under future elevated [CO2] atmospheres to replenish the extra N removed in grains from cropping systems if no residue is incorporated, or to facilitate the [CO2] fertilization effect on grain yield by overcoming N immobilization resulting from residue amendment.  相似文献

The objective of this work was to evaluate the effect of endophytic diazotrophic bacteria Herbaspirillum and Burkholderia on the Fusarium oxysporum f. sp. cubense (Foc) establishment and on the plantlets growth of the ‘Maçã’ banana (Musa spp., group ABB). Two assays were carried out in a greenhouse at the National Center of Tropical Agroindustry in Fortaleza city, Ceará State (Brazil), using randomized block designs in the factorial arrangements 4?×?2 and 8?×?2. On the first trial plantlets were inoculated with the Burkholderia sp. AB202; Herbaspirillum sp., BA227, both of strains and controls bacteria, with and without Foc and cultivated in pots filled with an autoclaved mixture of washed sand and vermiculite (ratio 3:2 v v?1), during 4 months. On a second assay, plants were subjected to following conditions: absence and presence of strains AB202, AB213 (Burkholderia spp.), BA227, BA234 (Herbaspirillum-like), AB202 plus BA227, AB213 plus BA234, the mixture of the four bacterial strain, absence and presence of the Foc; and cultivated in pots filled with autoclaved Haplic Arenosol, during 2 months. The plant association with diazotrophs and the Foc was confirmed, and factors interacted significantly on the most probable number of bacteria and the colony forming units of the pathogen on roots and plant rhizomes. The potential of the endophytes on the inhibition of Foc propagate units and on the plant growth promotion was demonstrated. The higher biomass was observed four and 2 months after plant inoculation with AB202 and BA234. Results showed that these endophytes may be used as potential biofertilizer and biocontrol agents.  相似文献

Background and aims

Whereas the expression patterns and kinetic properties of the rice (Oryza sativa) phosphate transporter gene OsPht1; 6 (OsPT6) are well documented, little is known about the biological functions of this gene. The aim of this study was to investigate the roles of OsPT6 in inorganic phosphate (Pi) acquisition and mobilization, and examine its potential to enhance agricultural production.

Methods

Here, we generated OsPT6 overexpression transgenic plants using Wuyujing 7, a widely cultivated variety of japonica rice, and then treated transgenic lines and wild type with different Pi supply in hydroponic and soil experiments to explore the functions of OsPT6 in rice.

Results

The OsPT6-overexpressing rice lines grew better and accumulated more biomass than wild-type plants, and exhibited significant increases in P accumulation in various tissues, including reproductive tissues under both hydroponic and soil culture conditions. Phosphate-uptake experiment using radiolabeled Pi (33P) showed that the rate of Pi uptake was 75 % and 73 % greater in transgenic plants grown under Pi-sufficient and -deficient conditions, respectively, than the wild-type controls, and that the shoot/root ratio of 33P was 104 % and 42 % greater, respectively. In addition, the grain yield per transgenic plant was much higher than that of the wild-type plants under field conditions.

Conclusions

Taken together, our results demonstrate that OsPT6 plays a vital role in Pi acquisition and mobilization in rice and suggest that this gene may be used for genetic engineering rice plants that require less Pi fertilizer.  相似文献

Aims

Carbon (C) bio-sequestration within the phytoliths of plants, a mechanism of long-term biogeochemical C sequestration, may play a major role in the global C cycle and climate change. In this study, we explored the potential of C bio-sequestration within phytoliths produced in cultivated rice (Oryza sativa), a well known silicon accumulator.

Methods

The rice phytolith extraction was undertaken with microwave digestion procedures and the determination of occluded C in phytoliths was based on dissolution methods of phytolith-Si.

Results

Chemical analysis indicates that the phytolith-occluded C (PhytOC) contents of the different organs (leaf, stem, sheath and grains) on a dry weight basis in 5 rice cultivars range from 0.4 mg?g?1 to 2.8 mg?g?1, and the C content of phytoliths from grains is much lower than that of leaf, stem and sheath. The data also show that the PhytOC content of rice depends on both the content of phytoliths and the efficiency of C occlusion within phytoliths during rice growth. The biogeochemical C sequestration flux of phytoliths in 5 rice cultivars is approximately 0.03–0.13 Mg of carbon dioxide (CO2) equivalents (Mg-e-CO2) ha?1?year?1. From 1950 to 2010, about 2.37?×?108?Mg of CO2 equivalents might have been sequestrated within the rice phytoliths in China. Assuming a maximum phytoliths C bio-sequestration flux of 0.13 Mg-e-CO2 ha?1?year?1, the global annual potential rate of CO2 sequestrated in rice phytoliths would approximately be 1.94?×?107?Mg.

Conclusions

Therefore rice crops may play a significant role in long-term C sequestration through the formation of PhytOC.  相似文献

Aim

In rice, the top two leaves are the major carbohydrate source during grain filling. Physiological performance of these leaves under salinity may allow estimate stress-induced yield loss.

Methods

Greenhouse grown rice plants (cv. Taipei 309) were subjected to 10 and 20 mM NaCl stress levels from germination till maturity. Plant development was measured at the flowering stage and yield parameters were quantified after complete ripening of panicles.

Results

Gas exchange in the main source leaves were not significantly affected by any of the stress levels. However, growth parameters as well as total metabolizable carbohydrates content, chlorophyll content (CCI), maximal efficiency of PSII photochemistry in dark-adapted state (F v/F m) and lipid peroxidation were significantly affected. Rice yield, measured as total panicle production, declined to 78 and 21 % of controls in 10 and 20 mM NaCl stress, respectively. Stress-induced yield loss was positively related with the decline in CCI, F v/F m and K+/Na+ ratio as well as with the increase in lipid peroxidation and total soluble carbohydrate contents.

Conclusions

Though the stress levels used in this work are below what is considered the minimal critical threshold of toxicity for rice, they induce significant negative effects on plant development and yield, when present along the whole plant life cycle.  相似文献

Aims

Previous studies have shown that elephant grass is colonized by nitrogen-fixing bacterial species; however, these results were based on culture-dependent methods, an approach that introduces bias due to an incomplete assessment of the microbial community. In this study, we used culture-independent methods to survey the diversity of endophytes and plant-associated bacterial communities in five elephant grass genotypes used in bioenergy production.

Methods

The plants of five genotypes of elephant grass were harvested from the experimental area of Embrapa Agrobiologia and divided into stem and root tissues. Total DNA and RNA were extracted from plant tissues and the bacterial communities were analyzed by DGGE and clone library of the 16S rRNA and nifH genes at both the cDNA and DNA levels.

Results

Overall, the patterns based on DNA- and RNA-derived DGGE-profiles differed, especially within tissue samples. DNA-based DGGE indicated that both total bacterial and diazotrophic communities associated with roots (rhizoplane?+?endophytes) differed clearly from those obtained from stems (endophytes). These results were confirmed by the phylogenetic analyses of RNA-derived sequences of 16S rRNA (total bacteria; 586 sequences), but not for nifH (186). In fact, rarefaction analyses showed a higher diversity of diazotrophic organisms associated with stems than roots. Based on 16S rRNA sequences, the clone libraries were dominated by sequences affiliated to members of Leptotrix (12.8 %) followed by Burkholderia (9 %) and Bradyrhizobium (6.5 %), while most of the nifH clones were closely related to the genus Bradyrhizobium (26 %).

Conclusions

Our results revealed an unexpectedly large diversity of metabolically active bacteria, providing new insights into the bacterial species predominantly found in association with elephant grass. Furthermore, these results can be very useful for the development of new strategies for selection of potential bacteria that effectively contribute to biological nitrogen fixation and enhance the sustainable production of elephant grass as bioenergy crop.  相似文献

Background and aims

Some elephant grass (Pennisetum purpureum) genotypes are able to produce large amounts of biomass and accumulate N derived from BNF when growing in soil with low N levels. However, information about the diazotrophic bacteria colonizing this C4 plant is still very scarce. This study aimed to characterize the plant growth promoting traits of a fraction of culturable diazotrophs colonizing the genotypes CNPGL F06-3 and Cameroon.

Methods

A total of 204 isolates were obtained from surface sterilized leaves, stems and roots after culturing on five different N-free semisolid media. These were then analyzed by BOX-PCR, and the 16S rRNA and nifH sequences of representative isolates were obtained. The functional ability of the isolates to reduce acetylene, produce indole and to solubilize phosphate was also determined.

Results

The diazotrophic bacterial population varied from 102 up to 106 bacteria g?1 fresh tissues of both genotypes. The BOX-PCR analysis suggested a trend in the genetic diversity among the 204 diazotrophic strains colonizing the different genotypes and plant tissues. Sequencing of 16S rRNA fragments confirmed the presence of Azospirillum brasilense and Gluconacetobacter diazotrophicus and revealed for the first time the occurrence of G. liquefaciens, G. sacchari, Burkholderia silvatlantica, Klebsiella sp., Enterobacter cloacae and E. oryzae in elephant grass. Interestingly, several nifH sequences from isolates identified as G. liquefaciens and G. sacchari showed homologies with nifH sequences of Enterobacter species. The majority of the isolates (97%) produced indole compounds, 22% solubilized phosphate and 6.4% possessed both characteristics.

Conclusions

The results showed the occurrence of novel diazotrophic bacterial species colonizing different tissues of both genotypes of elephant grass. In addition, the study revealed the presence of several bacteria with growth promoting traits, and highlighted their potential to be exploited as biofertilizers.  相似文献

Background and Aims

The accumulation of cadmium and lead in rice (Oryza sativa L.) grains is a potential threat to human health. In this study, the effect of selenium fertilization on the uptake and translocation of cadmium and lead in rice plants was investigated.

Methods

Rice plants were cultivated using cadmium and lead contaminated soils with selenium addition at three concentrations (0, 0.5 and 1 mg kg?1). At maturity, plants were harvested, and element concentrations in rice tissues were analyzed by using ICP-MS.

Results

Selenium application significantly increased selenium accumulation in rice grain, and markedly decreased cadmium and lead concentrations in rice tissues. In brown rice grains, selenium application reduced cadmium concentrations by 44.4 %, but had no significant effect on lead accumulation. Selenium application significantly decreased metal mobility in soils, at 0.5 mg kg?1 treatment, the translocation factor of cadmium and lead from soil to iron plaque decreased by 71 and 33 % respectively.

Conclusions

The mechanism of selenium mitigating of heavy metal accumulation in rice could be decreasing metal bioavailability in soil. Selenium fertilization could be an effective and feasible method to enrich selenium and reduce cadmium levels in brown rice.  相似文献

Background and aims

The rice production is experiencing a shift from conventionally seedling-transplanted (TPR) to direct-seeded (DSR) cropping systems in Southeast Asia. Besides the difference in rice crop establishment, water regime is typically characterized as water-saving moist irrigation for DSR and flooding-midseason drainage-reflooding and moist irrigation for TPR fields, respectively. A field experiment was conducted to quantify methane (CH4) and nitrous oxide (N2O) emissions from the DSR and TPR rice paddies in southeast China.

Methods

Seasonal measurements of CH4 and N2O fluxes from the DSR and TPR plots were simultaneously taken by static chamber-GC technique.

Results

Seasonal fluxes of CH4 averaged 1.58 mg m?2 h?1 and 1.02 mg m?2 h?1 across treatments in TPR and DSR rice paddies, respectively. Compared with TPR cropping systems, seasonal N2O emissions from DSR cropping systems were increased by 49 % and 46 % for the plots with or without N application, respectively. The emission factors of N2O were estimated to be 0.45 % and 0.69 % of N application, with a background emission of 0.65 and 0.95 kg N2O-N ha?1 under the TPR and DSR cropping regimes, respectively. Rice biomass and grain yield were significantly greater in the DSR than in the TPR cropping systems. The net global warming potential (GWP) of CH4 and N2O emissions were comparable between the two cropping systems, while the greenhouse gas intensity (GHGI) was significantly lower in the DSR than in the TPR cropping systems.

Conclusions

Higher grain yield, comparable GWP, and lower GHGI suggest that the DSR instead of conventional TPR rice cropping regime would weaken the radiative forcing of rice production in terms of per unit of rice grain yield in China, and DSR rice cropping regime could be a promising rice development alternative in mainland China.  相似文献

Background and aims

Herbaspirillum seropedicae (Hs) Z67 a diazotrophic endophyte was genetically engineered for secretion of 2-keto-D-gluconic acid by heterologous expression of genes for pqq synthesis and gluconate dehydrogenase to study its beneficial effect on plants.

Methods

Two plasmids, pJNK5, containing a 5.1 Kb pqq gene cluster of Acinetobacter calcoaceticus and pJNK6, carrying in addition the Pseudomonas putida KT2440 gluconate dehydrogenase (gad) operon were constructed in pUCPM18Gmr under Plac promoter. H. seropedicae Z67 transformants were monitored for P and K solubilization, cadmium (Cd) tolerance and rice growth promotion.

Results

Hs (pJNK5) secreted 23.5 mM gluconic acid and Hs (pJNK6) secreted 3.79 mM gluconic acid and 15.8 mM 2-ketogluconic acid respectively. Under aerobic conditions, Hs (pJNK5) and Hs (pJNK6) solubilized 239.7 μM and 457.7 μM P on HEPES rock phosphate and, 76.7 μM and 222.7 μM K on HRPF (feldspar), respectively, in minimal medium containing 50 mM glucose. Under N free minimal medium, similar effects of P and K solubilization were obtained. Hs (pJNK5) and Hs (pJNK6) inoculation increased the biomass, N, P, K content of rice plants (Gujarat – 17). These plants also accumulated 0.73 ng/g PQQ, and had improved growth and tolerance to CdCl2.

Conclusions

Incorporation of pqq and gad gene clusters in H. seropedicae Z67 imparted additional plant growth promoting traits of P and K solubilization and ability to alleviate Cd toxicity to the host plant.

Aims

Effects of different soil amendments were investigated on methane (CH4) emission, soil quality parameters and rice productivity in irrigated paddy field of Bangladesh.

Methods

The experiment was laid out in a randomized complete block design with five treatments and three replications. The experimental treatments were urea (220 kg ha?1) + rice straw compost (2 t ha?1) as a control, urea (170 kg ha?1) + rice straw compost (2 t ha?1) + silicate fertilizer, urea (170 kg ha?1) + sesbania biomass (2 t ha?1 ) + silicate fertilizer, urea (170 kg ha?1) + azolla biomass (2 t ha?1) + cyanobacterial mixture 15 kg ha?1 silicate fertilizer, urea (170 kg ha?1) + cattle manure compost (2 t ha?1) + silicate fertilizer.

Results

The average of two growing seasons CH4 flux 132 kg ha?1 was recorded from the conventional urea (220 kg ha?1) with rice straw compost incorporated field plot followed by 126.7 (4 % reduction), 130.7 (1.5 % reduction), 116 (12 % reduction) and 126 (5 % reduction) kg CH4 flux ha?1 respectively, with rice straw compost, sesbania biomass, azolla anabaena and cattle manure compost in combination urea and silicate fertilizer applied plots. Rice grain yield was increased by 15 % and 10 % over the control (4.95 Mg ha?1) with silicate plus composted cattle manure and silicate plus azolla anabaena, respectively. Soil quality parameters such as soil organic carbon, total nitrogen, microbial biomass carbon, soil redox status and cations exchange capacity were improved with the added organic materials and azolla biofertilizer amendments with silicate slag and optimum urea application (170 kg ha?1) in paddy field.

Conclusion

Integrated application of silicate fertilizer, well composted organic manures and azolla biofertilizer could be an effective strategy to minimize the use of conventional urea fertilizer, reducing CH4 emissions, improving soil quality parameters and increasing rice productivity in subtropical countries like Bangladesh.  相似文献

This study was aimed at assessing the diversity of putatively diazotrophic rhizobacteria associated with sunflower (Helianthus annuus L.) cropped in the south of Brazil, and to examine key plant growth promotion (PGP) characteristics of the isolates for the purposes of increasing plant productivity.

Methods

299 strains were isolated from the roots and rhizosphere of sunflower cultivated in five different areas using N-free media. 16S rDNA PCR-RFLP and 16S rRNA partial sequencing were used for identification and the Shannon index was used to evaluate bacterial diversity. Production of siderophores and indolic compounds (ICs), as well phosphate solubilization activities of each isolate were also evaluated in vitro. On the basis of multiple PGP activities, eight isolates were selected and tested for their N-fixation ability, and their capacity as potential PGPR on sunflower plants was also assessed.

Results

All except three Gram-positive strains (phylum Actinobacteria) belonged to the Gram-negative Proteobacteria subgroups [Gamma (167), Beta (78), and Alpha (50)] and the family Flavobacteriaceae (1)]. Shannon indexes ranged from 0.96 to 2.13 between the five sampling sites. Enterobacter and Burkholderia were the predominant genera isolated from roots and rhizosphere, respectively. Producers of siderophores and ICs were widely found amongst the isolates, but only 19.8% of them solubilized phosphate. About 8% of the isolates exhibited all three PGP traits, and these mostly belonged to the genus Burkholderia. Four isolates were able to stimulate the growth of sunflower plants under gnotobiotic conditions.

Conclusions

Enterobacter and Burkholderia were the dominant rhizospheric bacterial genera associated with sunflower plants. Inoculation with isolates belonging to the genera Achromobacter, Chryseobacterium, Azospirillum, and Burkholderia had a stimulatory effect on plant growth.  相似文献

Directed evolution of β-xylosidase XylBH43 using a single round of gene shuffling identified three mutations, R45K, M69P, and L186Y, that affect thermal stability parameter K t 0.5 by ?1.8 ± 0.1, 1.7 ± 0.3, and 3.2 ± 0.4 °C, respectively. In addition, a cluster of four mutations near hairpin loop-D83 improved K t 0.5 by ~3 °C; none of the individual amino acid changes measurably affect K t 0.5 . Saturation mutagenesis of L186 identified the variant L186K as having the most improved K t 0.5 value, by 8.1 ± 0.3 °C. The L186Y mutation was found to be additive, resulting in K t 0.5 increasing by up to 8.8 ± 0.3 °C when several beneficial mutations were combined. While k cat of xylobiose and 4-nitrophenyl-β-d-xylopyranoside were found to be depressed from 8 to 83 % in the thermally improved mutants, K m, K ss (substrate inhibition), and K i (product inhibition) values generally increased, resulting in lessened substrate and xylose inhibition.  相似文献
