The molecular basis of BLAD is the D128G mutation of the gene coding for the CD18 subunit of beta-2 integrin. This mutation is lethal, since homozygous (BL/BL) animals die before they reach sexual maturity. In the 1990s, BLAD was the most widespread genetic disease in HF cattle worldwide. The aim of the present study was to determine the frequency of BLAD carriers among 4645 young breeding bulls in Poland in 1995-2006. The frequency of carriers of the mutated allele showed a clear decreasing trend. The highest frequency (7.9%) was recorded while implementing the BLAD control program (1995-1997). Regular monitoring has enabled a great reduction of this threat to the tested population. Today only sporadic cases of BL/TL heterozygotes are reported (ca. 0.8% in 2004-2006).  相似文献

牛白细胞黏附缺陷症(BLAD)是由常染色体上CD18基因单碱基突变(A→G)引起的隐性遗传疾病,隐性基因纯合时导致白细胞表面的β2整合素表达明显减少或缺乏而引起临床发病,患病牛的主要特征是机体免疫力降低、易患病从而影响其生产性能的表现,严重影响了奶牛场的经济效益。该工作利用自行设计的特异性PCR引物,通过对PCR、PCR-SSCP条件的筛选和优化,并结合DNA序列测定,建立了PCR-SSCP检测BLAD 有害基因的新方法,该方法简便快捷、准确率高,使用样品宽泛,适合大样本筛选,为奶牛BLAD有害基因的分型和筛选提供了新的方法和思路,为我国奶牛的分子选育提供了可靠的理论依据。  相似文献

Cutaneous evaporation is the main avenue by which cattle dissipate heat via the involvement of sweat glands and other skin components. The difference in skin morphology between B. indicus and B. taurus has been recognized, as well as differences in their ability to tolerate heat. The objective of this study was to compare skin morphology between B. indicus, B. taurus, and their crossbreds. Skin samples of Sahiwal (B. indicus) (n?=?10, reddish brown skin) and Holstein Friesian (HF) (B. taurus) (n?=?10, black and white skin) and crossbred of HF75% (n?=?10, black and white skin) and HF87.5 % (n?=?10, black and white skin) were biopsied for histological study, followed by measurement of skin components. The results indicated that breed significantly affected sweat gland morphology. The shape of the sweat gland, as indicated by the ratio of length/diameter, in Sahiwal was baggier in shape compared to HF (5.99 and 9.52) while values for crossbreds were intermediate (7.82, 8.45). The density and volume of sweat glands in Sahiwal (1,058 glands/cm2; 1.60 μ3?×?10?6) were higher than in HF (920 glands/cm2; 0.51 μ3x10?6) and crossbreds, both HF 75 % (709 glands/cm2; 0.68 μ3?×?10?6) and HF 87.5 % (691 glands/cm2; 0.61 μ3?×?10?6) respectively. However, capillary surface area was greater for HF (2.07 cm2) compared to Sahiwal (1.79 cm2); accordingly, the lower genetic fraction of HF in crossbred cattle showed less capillary surface area (1.83 and 1.9 cm2 for HF75% and HF87.5 %) (P?<?0.01). Nerve density was not significantly different between Sahiwal and HF but was higher in the crossbred (P?<?0.01) cattle. Moreover, the effect of skin color (black and white) was evaluated and it was found that there was an interaction (P?<?0.01) between breed and skin color on the skin components. This study reveals that there are differences in skin morphology among B. indicus, B. taurus and their crossbreds, with these differences being more or less related to the genetic fraction of HF. This may imply that capability for cutaneous evaporative heat loss and tolerance to heat in crossbred cattle could be related to skin morphology.  相似文献

A study on butyrophilin (BTN) gene was conducted to detect variability at nucleotide level between cattle and buffalo. Hae III PCR-RFLP was carried out in crossbred cattle and it revealed polymorphism at this locus. Three genotypes namely, AA, BB and AB and two alleles were observed with frequencies 0.78, 0.17, 0.04 and 0.87, 0.13, respectively. The sequences of different cattle, buffalo and sheep breeds have been reported in the EMBL gene bank with accession numbers: AY491468 to AY491475. The nucleotides, which have been substituted from allele A to B, were found to be C to G (71st nucleotide), C to T (86th nucleotide), A to T (217th nucleotide), G to A (258th nucleotide), A to C (371st nucleotide) and C to T (377th nucleotide). The nucleotide substitution at 71st, 86th and 377th position of the fragment were expected to be a silent mutation where as nucleotide changes at 217th, 258th and 371st positions were expected to be substituted by lysine with arginine, valine with isoleucine and leucine with proline in allele B. The differences of nucleotides and amino acids between cattle, buffalo and sheep breeds have been revealed and on the basis of nucleotide as well as protein variability the phylogenetic diagram have been developed indicating closeness between cattle and buffalo.  相似文献

Background

The accuracy of genomic prediction depends largely on the number of animals with phenotypes and genotypes. In some industries, such as sheep and beef cattle, data are often available from a mixture of breeds, multiple strains within a breed or from crossbred animals. The objective of this study was to compare the accuracy of genomic prediction for several economically important traits in sheep when using data from purebreds, crossbreds or a combination of those in a reference population.

Methods

The reference populations were purebred Merinos, crossbreds of Border Leicester (BL), Poll Dorset (PD) or White Suffolk (WS) with Merinos and combinations of purebred and crossbred animals. Genomic breeding values (GBV) were calculated based on genomic best linear unbiased prediction (GBLUP), using a genomic relationship matrix calculated based on 48 599 Ovine SNP (single nucleotide polymorphisms) genotypes. The accuracy of GBV was assessed in a group of purebred industry sires based on the correlation coefficient between GBV and accurate estimated breeding values based on progeny records.

Results

The accuracy of GBV for Merino sires increased with a larger purebred Merino reference population, but decreased when a large purebred Merino reference population was augmented with records from crossbred animals. The GBV accuracy for BL, PD and WS breeds based on crossbred data was the same or tended to decrease when more purebred Merinos were added to the crossbred reference population. The prediction accuracy for a particular breed was close to zero when the reference population did not contain any haplotypes of the target breed, except for some low accuracies that were obtained when predicting PD from WS and vice versa.

Conclusions

This study demonstrates that crossbred animals can be used for genomic prediction of purebred animals using 50 k SNP marker density and GBLUP, but crossbred data provided lower accuracy than purebred data. Including data from distant breeds in a reference population had a neutral to slightly negative effect on the accuracy of genomic prediction. Accounting for differences in marker allele frequencies between breeds had only a small effect on the accuracy of genomic prediction from crossbred or combined crossbred and purebred reference populations.  相似文献

In vivo responsiveness to epinephrine, expression of L-selectin on neutrophils, changes in intracellular calcium ([Ca2+]i), sulfatide-induced superoxide production and tyrosine phosphorylation in neutrophils were evaluated to elucidate the role of L-selectin-associated functions of normal and CD18-deficient bovine neutrophils. The number of neutrophils in peripheral blood was significantly increased (P < 0.05) in four normal calves at 5-20 min after in vivo administration of epinephrine; however, no significant increase of neutrophils was found in three calves with bovine leucocyte adhesion deficiency (BLAD). Expression of L-selectin on neutrophils from three calves with BLAD was 61-77% of that of normal calves. Pretreatment of neutrophils with phorbol myristate acetate caused a marked decrease in the expression of L-selectin on neutrophils from both normal and BLAD calves. The sulfatide-induced sustained phase of [Ca2+]i concentration in neutrophils from calves with BLAD was significantly (P < 0.05) decreased. Following stimulation with aggregated IgG, the transient phase of [Ca2+]i in neutrophils from normal and BLAD calves was increased; however, the sustained phase of [Ca2+]i in BLAD neutrophils was significantly lower (P < 0.05) than that of controls. Sulfatide-induced O2- production and chemiluminescent response in neutrophils from calves with BLAD were 48-51% of those of normal calves and were inhibited by genistein and wortmannin, respectively, in a dose-dependent manner. The amount of tyrosine phosphorylated 100 kDa protein in neutrophils from BLAD calves stimulated with sulfatides was 57% of that of controls. The degree of L-selectin expression on neutrophils was correlated with the intracellular signalling events and the related superoxide production.  相似文献

An experiment was carried out to study the glucose kinetics of crossbred Holstein cattle feeding on either hay or 5% urea treated rice straw during late pregnancy (21 days prepartum) and early lactation (30 days postpartum). In all 16 pregnant heifers (23–25 months of age) were selected for the experiments, including eight animals of two breed types, Holstein Friesian×Red Sindhi (50:50=50% HF) and Holstein Friesian×Red Sindhi (87.5:12.5=87.5% HF). They were divided into four groups of four animals each. Animals from the same breed type in each group were fed with either rice straw treated with 5% urea or pangola hay (Digitaria decumbens) as the source of roughage throughout the experiments. The glucose turnover rate in both types of crossbred Holstein cattle was determined using a continuous infusion of [U-14C] and 3-[3H]glucose during late pregnancy and early lactation. Total glucose entry and utilization rates increased significantly during lactation for all groups. Recycling of [C]glucose was, approximately, 20% in both crossbred cattle fed either hay or urea treated rice straw and was unaffected by the stage of late pregnancy or early lactation. Comparing 50 and 87.5% HF animals, arterial plasma glucose concentrations were slightly higher during pregnant periods but significantly higher in lactating periods in 50% HF animals. The ratio of specific radioactivity of arterial blood bicarbonate relative to that of arterial blood [14C]glucose in the lactating period, significantly decreased in 50% HF animals fed either urea treated rice straw or hay. An increase in udder blood flow during early lactation was significantly higher in 87.5% HF animals than in 50% HF animals. The uptake, arteriovenous differences and extraction ratio for glucose across the udder, significantly increased in the lactating period for all crossbred animals. Glucose uptake by the udder of 87.5% HF animals accounted for 65% of the total glucose turnover rate compared to a value of 46% in the lactating 50% HF animals. It can be concluded that both crossbred cattle fed either urea treated rice straw or hay exhibit the same body glucose turnover rate. The 87.5% HF animal has the genetic potential for a high milk yield and has high body and udder glucose metabolisms compared with 50% HF animals.  相似文献

Many local livestock breeds in developing countries are being replaced by exotic breeds, leading to a loss of genetic resources. In southern Mali, for the past two decades, a trend towards increasing crossbreeding between the trypanotolerant N’Dama cattle and the trypano-susceptible Fulani Zebu cattle has been taking place. A survey with 160 farmers owning a cattle herd was carried out in southern Mali to investigate their production objectives, as well as trait and breed preferences and correlated socio-economic determinants in order to understand farmers’ breeding decisions and to identify comparative advantages of three breed groups (N’Dama, Fulani Zebu and crossbreds) raised in the study area. Data were analyzed using an exploded logit model. The reasons for raising cattle, as well as trait and breed preferences reflected the multiple objectives of the farmers. Draught power and savings were the most important production objectives. Productive traits were ranked highest; farmers reported large body size as the most preferred trait, followed by fertility, draught ability and milk yield. Crossbreds were the favored breed group. Breed preferences were mainly explained by ‘resistance to disease’ for N’Dama cattle and ‘high market price’ for Fulani Zebu and crossbred cattle. Production objectives, trait and breed preferences were mainly influenced by farmer group (local farmers and settled transhumants). Local farmers put comparatively more emphasis on livestock functions linked to crop production such as draught power. They had a higher preference for traction ability as a selection trait and preferred N’Dama over Fulani Zebu cattle. Settled transhumants emphasized milk yield as a selection trait and preferred Fulani Zebu over N’Dama. The results indicate that the trend towards more crossbreeding will continue putting the N’Dama breed under high risk of genetic dilution in southern Mali. The N’Dama cattle remain a valuable breed due to their adaptive traits such as disease and drought tolerance and their good traction ability, fulfilling the diverse objectives of local farmers. Crossbreeding was found to be a promising breeding strategy, which might contribute to the maintenance of the local breed, provided that breeding schemes are thoroughly planned and organized.  相似文献

BLAD is a hereditary disease in Holstein dairy cattle. The defective allele of CD18 gene which is responsible for this disease has a recessive inheritance. The recessive homozygous form (BL/BL) is lethal and since carrier animals have viability, BLAD frequency increases by use of carrier bulls in Artificial Insemination (AI). BLAD carriers can be detected easily by means of polymerase chain reaction followed by restriction analysis of the amplicons. In this study DNA samples from Holstein (n = 30) and Brown Swiss (n = 10) bulls from Abbas Abad AI center (Khorasan state of Iran) were analysed. A 101 bp fragment from the polymorphic region of CD18 gene located on chromosome 1 was amplified by PCR. Restriction enzymes TaqI and HaeIII were used to identify genotypes. Digestion products were screened by electrophoresis on 8% non-denaturing polyacrylamide gel and visualized by ethidium bromide staining. Frequencies of BL/TL (carrier) genotypes in Holstein and Brown Swiss bulls were 3.33% and 0%, respectively. Our pedigree studies of the carrier bull in this experiment revealed that the mutation inherited to him from Hawkeye bull (CANM 369995, BL). Although the elimination of BLAD-carrier bulls from the Holstein world would be the most efficient method to control this genetic disorder, many BLAD-carrier bulls are still listed commercially for AI and BLAD is still occurring in Iran. Monitoring the prevalence of BLAD-carriers in random selected herds may be helpful in judging the effectiveness of the BLAD-control program.  相似文献

The expression of Fc receptors for immunoglobulin G(IgG) and concanavalin A (con A)-binding receptors, luminol-dependent chemiluminescent (LDCL) responses, and the effect of anti-bovine IgG on LDCL responses were evaluated in neutrophils from Holstein calves with leukocyte adhesion deficiency (BLAD). Neutrophils from affected calves showed a 2.1- to 2.5-fold increase in Fc receptor expression compared with those of control calves by flow cytometric analysis. Con A-binding activities of neutrophils from affected calves were similar to those of control calves. Neutrophils from a calf with BLAD, when stimulated with zymosan opsonized with bovine serum (OPZ), heat-aggregated bovine IgG (Agg-bovine IgG), sheep red blood cells (SRBC) sensitized with anti-SRBC antibody (SRBC-anti-SRBC Ab), or con A had LDCL responses of 36 (P<0.05), 77, 126 and 119% of peak LDCL values of controls, respectively. The NBT-reducing value of neutrophils from a calf with BLAD when stimulated with Agg-bovine IgG after pretreatment with anti-bovine IgG was 116.5% of the values of neutrophils from control calves, but the difference was not significant. The LDCL responses of neutrophils from a control calf and a calf with BLAD stimulated with OPZ were inhibited markedly by pre-incubation with anti-bovine IgG antiserum at concentrations ranging from 1.25 to 20 or 40 μg/ml. Although an increase in Fc receptor expression on neutrophils from calves with BLAD was observed, the LDCL responses stimulated with SRBC-anti-SRBC Ab and NBT-reducing activity stimulated with Agg-bovine IgG after pretreatment with anti-bovine IgG did not correlate significantly with the increased Fc receptor expression. These results support that neutrophil functions mediated by the Fc receptors are associated synergistically with the presence of the complement receptor type 3 (CR3)(CD11b/CD18).  相似文献

The accurate supply of energy is essential to optimize livestock productivity and profitability. Furthermore, replacing empty BW gain (EBG) with carcass gain (CG) might be a suitable alternative to estimate the retained energy (RE) of beef cattle. Thus, this multi-analysis study was conducted aiming to estimate and validate new equations to predict carcass weight (CW), EBG, and RE of Zebu, beef crossbred, and dairy crossbred. A database composed by 1 112 animals encompassing bulls, steers, heifers of different genetic groups (Zebu, beef crossbred, and dairy crossbred), and two types of slaughter plants (commercial and experimental) was used for generating the new CW equation. For the development of the EBG and RE equations, a database of 636 observations composed of bulls, steers, and heifers of different genetic groups (Zebu, beef crossbred, and dairy crossbred) was assembled. The validation of new equations was performed using independent databases composed by 137 observations (80 for CW and 57 for EBG and RE). The new approaches for EBG and RE validation also included data from our research group studies (Inside) and independent data from literature publications (Outside). Furthermore, the new RE equation was compared to the current model devised by the nutritional requirements, diet formulation, and performance prediction of Zebu and crossbred cattle (BR-CORTE, 2016). Validation analyses were performed by using the Model Evaluation System (MES; 3.1.13, College Station, US). The CW was accurately estimated by the new equation when using both commercial and experimental data. Also, the equations developed in this study accurately estimated EBG and RE using both inside and outside data. In conclusion, equations proposed in this study accurately and precisely estimated CW, EBG, and RE of Zebu beef cattle that composed validation data set. Therefore, we suggest the following equations to estimate CW, EBG, and RE of Zebu cattle: CW, kg = − 11.0±1.56 + P + ((0.609±0.005 + G + B) × SBW); EBG (kg) = 0.044±0.017 + 1.47±0.026 × CG; RE (MJ/d) = 4.184 × (0.082±0.002 × EQEBW0.75 × CG0.777±0.039), where P = slaughter plant effect, if commercial = − 10.98, if experimental = 0; G = gender effect, if steer = 0, if bull = 0.008169 and if heifer = − 0.00612; B = genotype effect, if Zebu = 0, if dairy crossbreds = − 0.03301 and if beef crossbreds = − 0.01595; SBW = shrunk BW; CG = carcass gain; EQEBW = equivalent empty BW.  相似文献

Scrotal circumference (SC) and plasma concentrations of luteinizing hormone (LH) and follicle stimulating hormone (FSH) were measured in 19 Finnish Landrace (Finn) and 29 Rambouillet crossbred ram lambs raised in confinement and in 21 Finn and 19 Dorset crossbred ram lambs raised on pasture. SC was larger (P<0.05) for Finn crossbreds than for Rambouillet crossbreds only at 70 and 100 days of age. Finn crossbreds had more (P<0.05) plasma LH and FSH than did Rambouillet crossbreds at 20 and 44 days but not at 70 days. SC was larger for Dorset crossbreds than for Finn crossbreds at the same age, but SC was larger (P<0.05) for Finn crossbreds than for Dorset crossbreds at the same weight. Finn and Dorset crossbreds differed (P<0.10) in plasma LH and FSH concentration at 77 days only (Finn>Dorset).  相似文献

牛白细胞粘附缺陷病(BLAD)是一种常染色体单基因隐性遗传疾病, 病因为白细胞表面整合素CD18亚单位基因突变所致。为了解我国奶牛群中BLAD的携带和发生情况, 本研究采用RT-PCR扩增CD18基因片段后, 用TaqⅠ内切酶对扩增产物进行酶切的方法检测了1 000头1~6岁的奶牛, 结果有19头奶牛为BLAD携带牛, 即BLAD携带率为1.9%; 1头为BLAD病牛。  相似文献

The legendary Vechur cattle of Kerala, described as a very short breed, and the crossbred (CB) Sunandini cattle population exhibited great phenotypic variation; hence, the present study attempted to analyze the genetic diversity existing between them. A set of 14 polymorphic microsatellites were chosen from FAO-ISAG panel and amplified from genomic DNA isolated from blood samples of 30 Vechur and 64 unrelated crossbred cattle, using fluorescent labeled primers. Both populations revealed high genetic diversity as evidenced from high observed number of alleles, Polymorphic Information Content and expected heterozygosity. Observed heterozygosity was lesser (0.699) than expected (0.752) in Vechur population which was further supported by positive FIS value of 0.1149, indicating slight level of inbreeding in Vechur population. Overall, FST value was 0.065, which means genetic differentiation between crossbred and Vechur population was 6.5%, indicating that the crossbred cattle must have differentiated into a definite population that is different from the indigenous Vechur cows. Structure analysis indicated that the two populations showed distinct differences, with two underlying clusters. The present study supports the separation between Taurine and Zebu cattle and throws light onto the genetic diversity and relationship between native Vechur and crossbred cattle populations in Kerala state.  相似文献

FABP4 is a protein primarily expressed in adipocytes and macrophages that plays a key role in fatty acid trafficking and lipid hydrolysis. FABP4 gene polymorphisms have been associated with meat quality traits in cattle, mostly in Asian breeds under feedlot conditions. The objectives of this work were to characterize FABP4 genetic variation in several worldwide cattle breeds and evaluate possible genotype effects on fat content in a pasture-fed crossbred (Angus-Hereford-Limousin) population. We re-sequenced 43 unrelated animals from nine cattle breeds (Angus, Brahman, Creole, Hereford, Holstein, Limousin, Nelore, Shorthorn, and Wagyu) and obtained 22 single nucleotide polymorphisms (SNPs) over 3,164?bp, including four novel polymorphisms. Haplotypes and linkage disequilibrium analyses showed a high variability. Five SNPs were selected to perform validation and association studies in our crossbred population. Four SNPs showed well-balanced allele frequencies (minor frequency?>?0.159), and three showed no significant deviations from Hardy-Weinberg proportions. An experiment was performed to study the glucose kinetics of crossbred Holstein cattle feeding on either hay or 5% urea-treated rice straw during early lactation (30 days post partum), mid-lactation (120 days post partum) and late lactation (210 days post partum). Two breeds: Holstein FriesianxRed Sindhi (50:50 = 50% HF) and Holstein FriesianxRed Sindhi (87.5:12.5 = 87.5% HF) were used. In early lactating 87.5% HF animals feeding on either hay or urea-treated rice straw, the high milk yields and lactose secretion were related to glucose uptake by the udder and udder blood flow as compared with those of 50% HF animals. Marked decreases in udder blood flow, glucose uptake, lactose secretion and milk yield were apparent in mid- and late lactation of both groups of 87.5% HF animals. In contrast, both groups of 50% HF animals showed no significant changes in udder blood flow, udder glucose uptake, lactose secretion and milk yields throughout the course of lactation. Total glucose entry rate using 3-[3H] glucose infusion, recycling of glucose carbon and plasma glucose clearance significantly increased during late lactation for 50 and 87.5% HF animals feeding on urea-treated rice straw. The utilization rates of glucose using [U-(14)C] glucose infusion were not significantly different among groups of animals and periods of lactation. It can be concluded that 87.5% HF animals have the genetic potential for a higher milk yield, but a shorter peak yield and poorer persistence in comparison with 50% HF animals. Changes in the utilization of glucose by the mammary gland for milk production in both groups of crossbred animals during feeding on either hay or urea-treated rice straw would be dependent on intramammary changes.  相似文献

The social behaviour of Polish Black-and-White dairy cattle (BW) was compared with that of the F1-crossbreds of BW with Jersey (BW × J), Ayrshire (BW × Ay), Holstein-Friesian (BW × HF) and Swedish Black-and-White (BW × SLB). The ethological observations were made under conditions of loose housing.The crossbreds BW × J proved to be most aggressive, excitable and susceptible, as far as milk yield was concerned, to the social tensions connected with moving cows between groups. The BW × Ay were most dominant, least attacked by their group-mates and least susceptible to a group change. The pure-bred BW were calm and subordinate as compared with the crossbreds.The social tensions after a group change caused a mean decrease in milk yield of about 4%. There was a low correlation between the parameters of social behaviour and physical or production traits. The dominance relationships based on spontaneous aggressive interactions differed slightly from those based on interactions motivated by feeding.  相似文献

A screening program for bovine leukocyte adhesion deficiency (BLAD) in Danish Holstein-Friesian cattle has been initiated. During the first months 1611 animals were tested by a PCR based assay. Of these animals 1256, 346, and 8 were assigned normal, BLAD carriers, and BLAD affected animals, respectively One bull, born as a co-twin, showed weak reaction for the BLAD allele on DNA isolated from leukocytes, but a normal genotype on DNA isolated from semen. Chromosome analysis showed that this bull was a blood chimaera. Estimation of the BLAD allele frequency upon the PCR test results showed that around 450 Danish calves born in 1991 might have been affected with the recessive disorder.  相似文献
