Information on the sex, age, and ethnic compositions; reproductive parameters; intensity of natural selection (Crow's indices); and surname diversity of three rural populations (the Byadi, Dyupsya, and Cheriktey villages) of the Ust-Aldan ulus (district) of Sakha Republic (Yakutia) has been analyzed. The rural Yakut population of the Ust-Aldan ulus is demographically young (the mean age 25-31 years) and characterized by low outbreeding, unfavorable sex ratio in both prereproductive and reproductive ages, and high fertility (3.58-5.45 children surviving until the reproductive age per woman that has completed the reproductive period), although the actual reproductively active period is shorter than half its physiological duration. In the structure of total selection, the differential-fertility component is considerably greater than the differential-mortality component (Itot = 0.625, Im = 0.093, and If = 0.487). In the villages studied, some surnames are accumulated (45-65% of the population have five most frequent surnames), which determines the low surname diversity (alpha = 11.62-25.19) and high random isonymy (Ir = 0.0391-0.0823).  相似文献

The genetic demographic parameters of rural populations of Kyrgyzstan have been studied. The rural population of Kyrgyzstan has the following demographic characteristics: (1) it is demographically young (the population mean age is 25–28 years); (2) the sex ratio in reproductive age groups is unfavorable; (3) the fertility is high (5.78–7.68 pregnancies and 4.27–5.20 surviving children per woman of postreproductive age; (4) interethnic marriages are rare (1.0–5.9%), whereas consanguineous marriages are frequent (1.4–12%); (5) Crow's indices are relatively low (I tot = 0.24–0.29, I m = 0.05–0.13, I f = 0.14–0.22); and (6) both male and female contributions of different regions of the republic make unequal contributions to the gametic structure of the population, with the gametic structure varying from generation to generation. Migrations, dynamics of the gametic structure of rural populations, and marriage structure with respect to birthplaces and inbreeding estimated from isonymy have been studied in the Ust-Aldan ulus (administrative district) of Sakha Republic (Yakutia). The villages studied (Byadi, Dyupsya, and Cheriktey) are characterized by intense migration; however, the migration radius is small (most migrations occur within the district). The rural populations studied differ in the intensities and directions of gamete flows and their dynamics. There is no substantial gamete flow into the Ust-Aldan population from outside Sakha Republic. About 50% of marriages contracted in this population are homolocal (between residents of the same district); the endogamy is low (15%). In most cases of heterolocal marriages (contracted between residents of different districts), one of the spouses is a local resident. The inbreeding estimated from isonymy is F IT = 0.002930 in Yakuts; it is mainly accounted for by the nonrandom component (F IS = 0.002232 and F ST = 0.000700).  相似文献

The sex, age, tribal, and surname compositions of the populations of three villages of Altai Republic, Kulada (Ongudaisk raion), Beshpeltir (Chemal raion), and Kurmach-Baigol (Turochak raion) have been studied. Altaian populations are characterized by a high proportion of persons under 20 years of age (35.3–46.1%); however, there is a tendency towards a narrow base of the sex-age pyramid. The sex ratios in the total populations and in individual age groups are unfavorable. The rural populations studied differ in the spectrum and pattern of surname accumulation. The Kurmach-Baigol population (which consists of Northern Altaians) considerably differs from the Beshpeltir and Kulada populations (which are mostly Southern Altaian) with respect to the calculated parameters characterizing the population structure (random isonymy, migration index, the parameter of tribe diversity, entropy, and the redundancy of surname distribution). Isonymy coefficients of relationship between individual populations have been calculated from the data on tribes (surnames). Summary We have studied current (I Str) through the Na, K pump in amphibian oocytes under conditions designed to minimize parallel undesired currents. Specifically,I Str was measured as the strophanthidin-sensitive current in the presence of Ba2–, Cd2+ and gluconate (in place of external Cl). In addition,I Str was studied only after the difference currents from successive applications and washouts of strophanthidin (Str) were reproducible. The dose-response relationship to Str in four oocytes displayed a meanK 0.5 of 0.4 m, with 2–5 m producing 84–93% pump' block. From baseline data with 12 Na+-preloaded oocytes, voltage clamped in the range [–170, +50 mV] with and without 2–5 m Str, the averageI Str depended directly onV m up to a plateau at 0 mV with interpolated zero current at –165 mV. In three oocytes, lowering the external [Na+] markedly decreased the voltage sensitivity ofI p , while producing only a small change in the maximal outwardI Str. In contrast, decreasing the external [K+] from 25 to 2.5mm reducedI Str at 0 mV without substantially affecting its voltage dependence. At K+ concentrations of 1mm, both the absolute value ofI Str at 0 mV and the slope conductance were reduced. In eight oocytes, the activation of the averagedI Str by [K+] o over the voltage interval [–30, +30 mV] was well fit by the Hill equation, with K=1.7±0.4mm andnH (the minimum number of K+ binding sites) =1.7±0.4. The results unequivocally establish that the cardiotonic-sensitive current ofRana oocytes displays only a positive slope conductance for [K+] o >1mm. There is therefore no need to postulate more than one voltage-sensitive step in the cycling of the Na, K pump under physiologic conditions. The effects of varying external Na+ and K+ are consistent with results obtained in other tissues and may reflect an ion-well effect.  相似文献

Genetic and demographic characteristics for urban and rural population of the Chuvash Republic (Chuvashes and Russians) were calculated based on 1122 questionnaires. The sibship sizes for Chuvashes were 2.05 (urban) and 2.78 (rural). For Russians these indices were 1.75 (urban) and 2.00 (rural), respectively. The effect of nitroglycerin (NG) on inward voltage-activated calcium current (I Ca) was studied in isolated smooth muscle cells (SMC) of the guinea pigtaenia coli with the voltage clamp technique in an intracellular dialysis mode. Addition of NG (10–7 to 10–4 M) to the extracellular solution reduced theI Ca amplitude and increased theI Ca inactivation rate. The maximum inhibition ofI Ca (on the average, by 41.7 ± 4.8%,n=13) was produced by 10–4 M NG; the inhibition was dose-dependent. No shift of theI Ca current-voltage curve under the NG influence was observed. Application of dibutyril-cGMP (2·10–4 M), a membrane-penetrating analog of cyclic 3,5-guanosine monophosphate (cGMP), likewise decreased theI Ca amplitude and increased its inactivation rate. Summary The effects of epinephrine, glucagon and vasoactive intestinal polypeptide on chloride secretion by chloride cell-containing isolated opercular membranes from the seawater-adapted euryhaline teleost, the tilapiaSarotherodon mossambicus, have been examined. Epinephrine inhibits chloride secretion, measured as the short-circuit current (I sc), via -receptors, in a dose-dependent fashion. The minimum effective dose is 10–9 M, ED50 equals 2×10–7 M and maximal inhibition at 10–5 M is nearly 80%. Inhibition of phosphodiesterase by isobutylmethylxanthine (IBMX; 10–4 M), does not alterI sc in untreated tissues, but it completely reverses the epinephrine inhibition ofI sc, suggesting that hormones which modulate cAMP in chloride cells may alter chloride secretion. Glucagon and vasoactive intestinal polypeptide also stimulateI sc in epinephrine-inhibited tissues, an effect potentiated by IBMX. The effect of glucagon is dose-dependent with a minimum effective dose of 10–9 M, ED50 equal to 8×10–8 M and a maximum stimulation of 72% at 10–5 M.Analysis of the effects of epinephrine and IBMX onI sc and tissue conductance suggests that these agents act antagonistically on a nonconductive transport mechanism. Summary The electrically excitable salivary cells of the giant Amazon leech, Haementeria, display a time-dependent inward rectification. Under voltage clamp, hyperpolarizing steps to membrane potentials negative to about –70 mV were associated with the activation of a slow inward current (I h) which showed no inactivation with time. The time course of activation of I hwas described by a single-exponential function and was strongly voltage dependent. The activation curve ofhranged from –72 to –118 mV, with half-activation occurring at –100 mV. Ion-substitution experiments indicated that I his carried by both Na+ and K+ ions. 5-Hydroxytryptamine (5-HT) increased the amplitude of I hand its rale of activation. It also produced a positive shift of the activation curve of the conductance underlying I h Ghwithout altering the slope factor, thus indicating that the voltage dependence of I hwas modulated by 5-HT. Cs+ blocked both I hand the 5-HT-polentiated current in a voltage-independent manner, whereas Ba2+ had little effect. It is concluded that 5-HT increases I hby modulating the inwardly rectifying Na+-K+ channels in the salivary cells. The effect of 5-HT may be mediated by an increase in adenylate cyclase activity since I hwas increased by 8-bromocyclic AMP and by the phosphodiesterase inhibitor, 3-isobutyl-l-methylxanthine. In contrast, I hwas reduced by 8-bromo-cyclic GMPand by zaprinast (an inhibitor of cyclic GMP-scnsitive phosphodieslerase). Cyclic GMP itself also reduced I h, and the effect was specific to the 3,5 form; 2,3-cyclic GMP was inactive. Summary The influence of nerve growth factor (NGF) on Na currents of rat dorsal root ganglia (DRG) was studied in neurons obtained from newborns and cultured for 2–30 hr inserum-free defined medium (SFM). Cell survival for the period studied was 78–87% both with and without NGF. Na currents were detected in all cells cultured for 6–9 hr. They were also detected after 2 hr in culture in 21.5% of the cells cultured without NGF (–NGF cells), and in 91.5% of the cells cultured with NGF (+NGF cells). Current density of the -NGF cells was 2.3 and 2 pA/m2 after growth for 2 and 6–9 hr, respectively, compared to 3.0 and 3.9 pA/m2 for the +NGF cells. The +NGF cells were separated into fast (F), Intermediate (I) and slow (S) cells, based on the Na current they expressed, while -NGF cells were all of theI type.F, I andS currents differed in their voltage-dependent inactivation (Vh 50=–79, –28 and –20 mV), kinetics of inactivation (tau h =0.55, 1.3 and 7.75 msec), and TTX sensitivity (K i=60, 550 and 1100nm). All currents were depressed by [Ca] o with aKd Ca of 22, 17 and 8mm forF, I andS currents, respectively. Current density ofF andS currents was 5.5 and 5 pA/m2 for theI current. The concentration-dependent curve ofI currentvs. TTX indicated thatI current has two sites: one withF-like and another withS-likeK i for TTX. Hybridization ofF andS currents yieldI-like currents. Thus, the major effect of NGF on Na currents in SFM is the accleration of Na current acquisition and diversity, reflected in an increase of either theS orF type in a cell.  相似文献

Summary Using the patch-clamp technique, we recorded whole-cell calcium current from isolated cardiac myocytes dissociated from the apical ventricles of 7-day and 14-day chick embryos. In 70% of 14-day cells after 24 hr in culture, two component currents could be separated from totalI Ca activated from a holding potential (V h) of –80 mV. L-type current (I L) was activated by depolarizing steps fromV h –30 or –40 mV. The difference current (I T) was obtained by subtractingI L, fromI Ca.I T could also be distinguished pharmacologically fromI L in these cells.I T was selectively blocked by 40–160 m Ni2+, whereasI L was suppressed by 1 m D600 or 2 m nifedipine. The Ni2+-resistant and D600-resistant currents had activation thresholds and peak voltages that were near those ofI T andI L defined by voltage threshold, and resembled those in adult mammalian heart. In 7-day cells,I T andI L could be distinguished by voltage threshold in 45% (S cells), while an additional 45% of 7-day cells were nonseparable (NS) by activation voltage threshold. Nonetheless, in mostNS cells,I Ca was partly blocked by Ni2+ and by D600 given separately, and the effects were additive when these agents were given together. Differences among the cells in the ability to separateI T andI L by voltage threshold resulted largely from differences in the position of the steady-state inactivation and activation curves along the voltage axis. In all cells at both ages in which the steady-state inactivation relation was determined with a double-pulse protocol, the half-inactivation potential (V 1/2) of the Ni2+-resistant currentI L averaged –18 mV. In contrast,V 1/2 of the Ni2+-sensitiveI T was –60 mV in 14-day cells, –52 mV in 7-dayS cells, and –43 mV in 7-day NS cells. The half-activation potential was near –2 mV forI L at both ages, but that ofI T was –38 mV in 14-day and –29 mV in 7-day cells. Maximal current density was highly variable from cell to cell, but showed no systematic differences between 7-day and 14-day cells. These results indicate that the main developmental change that occurs in the components ofI Ca is a negative shift with, embryonic age in the activation and inactivation relationships ofI T along the voltage axis.  相似文献

The chick blastoderm at the stage of late gastrula is a flat disc formed by three cell layers and exhibiting epithelial properties. Blastoderms were cultured in miniature chambers and their electrophysiological characteristics were determined under Ussing conditions.Under open-circuit condition and identical physiological solutions on both sides, spontaneous transblastodermal potential difference (V oc) of –7.5±3.3 mV (ventral side positive) was measured. Under short-circuit condition (transblastodermal V = 0 mV), the blastoderm generated short-circuit current (I sc) of 21±8 A/cm2, which was entirely dependent on extracellular sodium, sensitive to ouabain applied ventrally and independent of extracellular chloride. The net transblastodermal Na+ flux fully accounted for the measured I sc, both under control conditions and with ouabain. The total transblastodermal resistance (R tot) was 390±125 cm2.Frequently, the V oc, I sc and R tot showed spontaneous oscillations with a period of 4–5 min. Removal of endoderm and mesoderm did not significantly affect the electrical properties, indicating that the electrogenic sodium transport is generated by the ectoderm.The V oc and I sc measured in the area pellucida (–1.3±0.8 mV, 9.3±4.4 A/cm2) and extraembryonic area opaca (–7.8±1.1 mV, 31.2±12.7 A/cm2) were significantly different. Summary We have investigated the pacemaker properties of aggregates of cells dissociated from the atria and ventricles of 10 to 14-day-old chick embryonic hearts using a two-microelectrode current and voltage-clamp technique. These preparations usually beat spontaneously and rhythmically in tissue culture medium containing 1.3mm potassium with a beat rate typically in the range of 15–60 beats per minute. The beat rate results show considerable variability, which precludes any statistically significant comparison between the spontaneous activity of atrial and ventricular cell preparations at 10–14 days of development. However, the shapes of pacemaker voltage changes do exhibit differences characteristic of cell type. Spontaneous atrial preparations rapidly depolarize from maximum diastolic potential (–90 mV) to a plateau range of pacemaker potentials (–80 to –75 mV). The membrane subsequently depolarizes more gradually until threshold (–65 mV) is reached. In contrast, spontaneously beating ventricular cell preparations slowly hyperpolarize after maximum diastolic potential to the –100 to –95 mV range before gradually depolarizing toward threshold. Voltage-clamp analysis reveals a virtual lack of any time-dependent pacemaker current in atrial preparations. These preparations are characterized by an approximately linear background current (I bg) having a slope resistance of 100 K cm2. Ventricular preparations have a potassium ion pacemaker current with slow kinetics (I K 2), and a second time-dependent component (I x) which is activated at potentials positive to –65 mV. The background current of these preparations displays inward rectification. Computer simulations of pacemaking reveal that the initial rapid phase of pacemaker depolarization in atrial cells is determined by the membrane time constant, which is the product of membrane capacitance and the slope resistance ofI bg. The hyperpolarization after maximum diastolic potential of ventricular cells is caused byI K 2. The final slow phase of depolarization in both cell types is caused in part by the steady-state amplitude of the fast inward sodium current (I Na). This component has negative slope conductance which effectively increases the slope resistance in the vicinity of threshold compared to TTX-treated preparations. This mechanism is sufficient to produce interbeat intervals several seconds in duration, even in the absence of time-dependent pacemaker current, provided that the background current is at the appropriate level.  相似文献

Synopsis Length at age and growth rates for 59 bull sharks, Carcharhinus leucas, collected from the northern Gulf of Mexico were estimated from the band patterns formed seasonally in the vertebral centra. The combined age at length data for both sexes were applied to a von Bertalanffy growth model producing parameter estimates of L = 285 cm TL, K = .076, t0 = –3.0 yr. Lengths at age for males and females were similar except that males did not attain as great a length as females. Growth was apparently slow and varied among individuals, but in general, was estimated to be 15–20 cm yr–1 for the first five years, 10 cm yr–1 for years 6–10, 5–7 cm yr–1 for years 11–16, and less than 4–5 cm yr–1 thereafter. Males mature at 210–220 cm TL or 14–15 yr of age; females mature at>225 cm TL or 18+ yr of age. The largest male (245 cm TL) was 21.3 yr old; the largest female (268 cm TL) was 24.2 yr old.  相似文献

We determined the effect of environmental stressors on the physiological condition of Brachionus rotundiformis. For two morphologically distinct B. rotundiformis strains: Hawaii (average lorica length = 222 m) and Langkawi strains (average lorica length 180 m), neonates hatched from resting eggs were exposed to different levels of unionized ammonia (0.7–9.8 mg l–1), viscosity (relative viscosity against natural seawater = 1–1.17) and Euplotes sp. (protozoan) contamination (1–40 cells ml–1). Increasing stress decreased fecundity and lifespan of both rotifer strains. Glucosidase and phospholipase activities were correlated with reproductive responses of both the strains exposed to unionized ammonia. When culture water viscosity was changed, the activity of esterase and phospholipase was correlated with reproductive responses of the Hawaiian strain, and glucosidase activity was correlated with those of Langkawi strain.With the protozoan contamination, esterase and glucosidase activities were correlated only with reproductive responses of the Hawaiian strain, while activity of all three enzymes was correlated to those of the Langkawi strain. Glucosidase activity proved to be a reliable indicator of stress for cultured B. rotundiformis.  相似文献

Atherina presbyter is a common fish off the Canary Islands. Age, growth, reproduction, and mortality of the species are studied based on sampling carried out from July 1995 to June 1996. The parameters of the total length–total weight relationship are: a=0.004521, and b=3.0771. Otoliths age readings indicate that the sampled population consists of four age groups (0–III years). The von Bertalanffy growth parameters for all individuals are: L=122mm total length, k=0.79year–1, and t0=–0.21 years. Individuals grow quickly in their immature first year, attaining approximately 60% of their maximum length. After the first year, the annual growth rate drops rapidly, because the energy is probably diverted to reproduction. It is a gonochoristic species with no evidence of sexual dimorphism. The gonad is present as a single diffuse testis in males and as a single discrete ovary in females. The overall ratio of males to females is not significantly different from 1:1. The reproductive period of the species is protacted (February to June). The peak of the reproductive effort occurs in April–May. The size at first maturity is 68mm. The population is being heavily exploited.  相似文献

Summary A stopped-flow rapid reaction apparatus was used for measuring changes in extracellular pH (pH o ) of red cell suspensions under conditions wheredpH o /dt was determined by the rate of HCO 3 /X exchange across the membrane (X =Cl, Br, F, I, NO 3 or SCN). The rate of the exchange at 37°C decreased forX in the order: Cl>Br>F>I>NO 3 >SCN, with rate constants in the ratios 10.860.770.550.520.31. When HCO 3 is exchanged for Cl, Br, F, NO 3 or SCN, a change in the rate-limiting step of the process takes place at a transition temperature (T T ) between 16 and 26°C. In I medium, however, no transition temperature is detected between 3 and 42°C. AlthoughT T varies withX , the activation energies both above and belowT T are similar for Cl, Br, NO 3 and F. The values of activation energy are considerably higher whenX =I or SCN. The apparent turnover numbers calculated for HCO 3 /X exchange (except forX =I) at the correspondingT T ranged from 140 to 460 ions/site ·sec for our experimental conditions. These findings suggest that: (i) HCO 3 /X exchange for allX studied takes place via the rapid anion exchange pathway; (ii) the rate of HCO 3 /X exchange is influenced by the specific anions involved in the 11 obligatory exchange; and (iii) the different transition temperatures in the Arrhenius diagrams of the HCO 3 /X exchange do not seem to be directly related to a critical turnover number, but may be dependent upon the influence ofX on protein-lipid interactions in the red blood cell membrane.  相似文献

Summary Two new forms of the plasma membrane ATP-ase ofMicrococcus lysodeikticus NCTC 2665 were isolated from a sub-strain of the microorganism by polyacrylamide gel electrophoresis. One of them had a mol.wt of 368,000 and a very low specific activity (0.80 µ mol.min–1.mg protein–1) that could not be stimulated by trypsin. This form has been called BI (strain B, inactive). If the electrophoresis was carried out in the presence of reducing agents (i.e., dithiothreitol) and the pH of the effluent maintained at a value of 8.5 another form of the enzyme was obtained. This had a mol.wt of 385,000 and a specific activity of 2.5–5.0 µ mol.min–1.mg protein–1 that could be stimulated by trypsin to 5–10 µ mol.min–1.mg protein–1. This preparation of the ATPase has been called form BA (strain B, enzyme active). The subunit composition of both forms has been studied by sodium dodecyl sulphate and urea gel electrophoresis and compared to that of the enzyme previously purified from the original strain (form A). The three forms of the enzyme had similar and subunits, with mol.wt of about 50,000 and 30,000 dalton, respectively. They also had in common the component(s) of relative mobility 1.0, whose status as true subunit(s) of the enzyme remains yet to be established. However, subunit, that had a mol.wt of about a 52,500 in form A (Andreu et al. Eur. J. Biochem. (1973) 37, 505–515), had a mol.wt similar to in form BI and about 60,000 in form BA. Furthermore BA usually showed two types of this subunit ( and) and an additional peptide chain () with a mol.wt of about 25,000 dalton. This latter subunit seemed to account for the stimulation by trypsin of form BA.Forms BA could be converted to BI by storage and freezing and thawing. Conventional protease activity could not be detected in any of the purified ATPase forms and addition of protease inhibitors to form BA failed to prevent its conversion to form BI. The low activity form (BI) was more stable than the active forms of the enzyme and also differed in its circular dichroism. These results show thatM. lysodeikticus ATPase can be isolated in several forms. Although these variations may be artifacts caused by the purification procedures, they provide model systems for understanding the structural and functional relationships of the enzyme and for drawing some speculations about its functionin vivo.  相似文献

We have applied enzyme kinetic analysis to electrophysiological steady-state data of Zhou et al. (Zhou, J.J., Trueman, L.J., Boorer, K.J., Theodoulou, F.L., Forde, B.G., Miller, A.J. 2000. A high-affinity fungal nitrate carrier with two transport mechanisms. J. Biol. Chem. 275:39894–9) and to new current-voltage-time records from Xenopus oocytes with functionally expressed NrtA (crnA) 2H+-NO 3 symporter from Emericella (Aspergillus) nidulans. Zhou et al. stressed two Michaelis-Menten (MM) mechanisms to mediate the observed nitrate-induced currents, I NO 3 . We show that a single straightforward reaction cycle describes the data well, pointing out that during exposure to external substrate, S = (2H++NO 3 )o, the product concentration inside, [P] = [H+] i 2 · [NO 3 i, may rise substantially near the plasma membrane, violating the condition [P] [S] for MM kinetics. Here, [P] and its changes during experimentation are treated explicitly. K 1/2 20 µM for I NO 3 at pHo from Zhou et al. is confirmed. According to our analysis, NrtA operates between about 0.2 and 0.6 of the electrical distance in the membrane (outside 0, inside 1). In absence of thermodynamic gradients, the predominant orientation of the binding site(s) is probably inwards. The activity of the enzyme is sensitive to the transmembrane voltage, V, with an apparent gating charge of +1.0 ± 0.5 for inactivation, and transition probabilities of 0.3–1.3 s–1 at V = 0. This gating mode impedes loss of cellular NO 3 during depolarization.  相似文献

The pH-sensitivity of transepithelial K+ transport was studied in vitro in isolated vestibular dark cell epithelium from the gerbil ampulla. The cytosolic pH (pH iwas measured microfluorometrically with the pH-sensitive dye 2,7-bicarboxyethyl-5(6)-carboxyfluorescein (BCECF) and the equivalent short-circuit current (I sc), which is a measure for transepithelial K+ secretion, was calculated from measurements of the transepithelial voltage (V t)and the transepithelial resistance (R t) in a micro-Ussing chamber. All experiments were conducted in virtually HCO 3 -free solutions. Under control conditions, pH iwas 7.01±0.04 (n=18), V twas 9.1±0.5 mV, R t16.7±0.09 cm2, and I sc was 587±30 A/cm2 (n=49). Addition of 20 mm propionate caused a biphasic effect involving an initial acidification of pH i, increase in V tand I sc and decrease in R tand a subsequent alkalinization of pH i, decrease of V tand increase of R t. Removal of propionate caused a transient effect involving an alkalinization of pH i, a decrease of V tand I sc and an increase in R t. pH iin the presence of propionate exceeded pH iunder control conditions. Effects of propionate on V t, R tand I sc were significantly larger when propionate was applied to the basolateral side rather than to the apical side of the epithelium. The pH i-sensitivityof I sc between pH 6.8 and 7.5 was –1089 A/(cm2 · pH-unit) suggesting that K+ secretion ceases at about pH i7.6. Acidification of the extracellular pH (pH o)caused an increase of V tand I sc and a decrease of R tmost likely due to acidification of pH i. Effects were significantly larger when the extracellular acidification was applied to the basolateral side rather than to the apical side of the epithelium. The pH osensitivity of I sc between pH 7.4 and 6.4 was –155 A/(cm2 · pH unit). These results demonstrate that transepithelial K+ transport is sensitive to pH iand pH oand that vestibular dark cells contain propionate uptake mechanism. Further, the data suggest that cytosolic acidification activates and that cytosolic alkalinization inactivates the slowly activating K+ channel (I sK)in the apical membrane. Whether the effect of pH ion the I sK channel is a direct or indirect effect remains to be determined.The authors wish to thank Drs. Daniel C.
