We investigated the usefulness of mitochondrial cytochrome c oxidase (COI) DNA barcoding of the genus Bradysia for the detection of immature stages and cryptic species complex. Although the larvae of some species in this genus are agricultural pests, immature stages are rarely identified due to the lack of key morphological characteristics. We constructed partial sequences of the COI gene for 25 species of Bradysia as a first step towards a DNA barcode. Using these data, Bradysia impatiens, B. procera and Bperaffinis were identified from larval specimens collected, respectively, from paprika, ginseng and oak sawdust beds used for cultivating shiitake. Our findings reveal a complex of three species within the Btilicola group. These species were all identified as important pest Bocellaris based on the morphology of male genital structures; however, the interspecific genetic divergence of the COI region was significantly greater (16.1–19.4%) than the intraspecific variation in each species. Therefore, Bocellaris may consist of at least three species. The results demonstrate that COI DNA barcodes are useful for Bradysia species identification.  相似文献

Flea beetles (Chrysomelidae: Galerucinae: Alticini), with ~8,000 species worldwide, include pest species causing substantial economic damage to crops. The genera Phyllotreta and Chaetocnema include both pest and non‐pest species. An accurate and fast taxonomic identification approach is required for discriminating among taxa for non‐expert taxonomists; moreover, the utility of this approach spans from biodiversity conservation to the monitoring of pest species. DNA barcoding represents a reliable and easy identification tool based on the use of short DNA sequences. In this study, 45 new COI sequences of 13 Phyllotreta and five Chaetocnema species, representing ~30% and ~20% of the Turkish species belonging to these genera, were provided. These sequences increased by ~18% and ~25% the number of species of these genera whose sequences are available in BOLD. In order to test DNA barcoding efficiency in Phyllotreta and Chaetocnema species identification, we created a data set consisting of sequences belonging to species present in the Middle East and available in BOLD plus the sequences developed in this study (36 species). The efficiency of species identification, estimated using best close match analysis (with the ad hoc calculated optimal distance threshold of 1.5%), was 99%. The overall intraspecific and interspecific mean nucleotide divergences were 1.4% and 20%, respectively. Interestingly, COI sequences of Phyllotreta nigripes clustered into two well‐separated groups with a high value of the between‐group nucleotide distance (11.4%), which suggests the presence of cryptic species. In addition, information was provided on the crops exploited by the collected organisms and the observed damage.  相似文献

The interaction between agricultural production and wildlife can shape, and even condition, the functioning of both systems. In this study, we i) explored the degree to which a widespread European bat, namely the common bent‐wing bat Miniopterus schreibersii, consumes crop‐damaging insects at a continental scale, and ii) tested whether its dietary niche is shaped by the extension and type of agricultural fields. We employed a dual‐primer DNA metabarcoding approach to characterize arthropod 16S and COI DNA sequences within bat faecal pellets collected across 16 Southern European localities, to first characterize the bat species’ dietary niche, second measure the incidence of agricultural pests across their ranges and third assess whether geographical dietary variation responds to climatic, landscape diversity, agriculture type and vegetation productivity factors. We detected 12 arthropod orders, among which lepidopterans were predominant. We identified >200 species, 44 of which are known to cause agricultural damage. Pest species were detected at all but one sampling site and in 94% of the analysed samples. Furthermore, the dietary diversity of M. schreibersii exhibited a negative linear relation with the area of intensive agricultural fields, thus suggesting crops restrict the dietary niche of bats to prey taxa associated with agricultural production within their foraging range. Overall, our results imply that M. schreibersii might be a valuable asset for biological pest suppression in a variety of agricultural productions and highlight the dynamic interplay between wildlife and agricultural systems.  相似文献

Genomic DNA samples from larvae of the beet webworm Loxostege sticticalis collected in the south‐western Russia were used to amplify mitochondrial cytochrome oxidase unit I (COI) gene. In a small proportion of samples, the sequenced product showed considerable heterogeneity due to admixture of a minor sequence. A preliminary BLAST analysis of a 100‐bp‐long fragment of this minor sequence showed its maximal similarity to the COI gene region of Cotesia, a genus of braconid larval endoparasitoids of Lepidoptera. An additional primer was designed to specifically amplify ca 300 bp of the COI gene region from Braconidae. As many as seven of 25 samples were positive by PCR. Sequencing of the amplified products in all these samples showed nucleotide sequence identity to the COI region of Cotesia vestalis (Cotesia plutellae) and the presence of two molecular haplotypes among individual parasitoid samples.  相似文献

The prey range of the invasive Asian paper wasp, Polistes chinensis antennalis, was studied using molecular diagnostics. Nests of paper wasps were collected from urban residential and salt marsh habitats, larvae were removed and dissected, and DNA in the gut of the paper wasp larvae was amplified and sequenced with cytochrome c oxidase subunit I (COI). Seventy percent of samples (211/299) yielded medium‐to high‐quality sequences, and prey identification was achieved using BLAST searches in BOLD. A total of 42 taxa were identified from 211 samples. Lepidoptera were the majority of prey, with 39 taxa from 91% of samples. Diptera was a relatively small component of prey (three taxa, 19 samples). Conclusive species‐level identification of prey was possible for 67% of samples, and genus‐level identification, for another 12% of samples. The composition of prey taken was different between the two habitats, with 2.5× more native prey species being taken in salt marsh compared with urban habitats. The results greatly extend the prey range of this invasive species. The technique is a more effective and efficient approach than relying on the collection of “prey balls”, or morphological identification of prey, for the study of paper wasps.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is oligophagous on plants in the Brassicaceae, and is considered one of the most significant insect pests of canola (Brassica napus L.), a major oilseed crop grown in the Prairie Provinces of Canada. The bertha armyworm (BAW), Mamestra configurata Walker (Lepidoptera: Noctuidae), is a generalist herbivore that preferentially feeds on canola plants. In the canola growing season in the Prairie Provinces of Canada, DBM feeding occurs prior to BAW herbivory. In this study, we test the effect of DBM herbivory on subsequent host use by BAW. Oviposition by female BAW was not influenced by previous DBM‐herbivory or mechanical damage of canola plants. Bertha armyworm larvae were deterred from feeding on canola previously damaged by DBM and larvae developed into lighter pupae when reared on DBM‐damaged plants. Bertha armyworm pupae that developed from larvae fed on mechanically damaged plants had similar pupal weights to those fed undamaged plants. Adult BAW reared on canola with previous DBM feeding damage had marginally smaller wings than moths reared on canola treated differently. The combination of these results suggests that herbivory by the brassicaceous specialist, DBM, negatively influences host use and larval performance by the generalist, BAW, on canola.  相似文献

Rice planthoppers are notorious plant sap‐feeding pests which cause serious damage. While several microbes in rice planthoppers have been broadly characterized, the abundance and diversity of bacteria and fungi in field planthoppers are largely unknown. This study investigated the bacterial and fungal community compositions of Chinese wild rice planthoppers Laodelphax striatellus and Sogatella furcifera using parallel 16S rRNA gene amplicon and internal transcribed space region sequencing. The bacteria varied significantly between the species and were partitioned significantly by sex, tissues and host environments in each species. The majority of bacteria were affiliated with the genera Wolbachia, Cardinium, Rickettsia and Pantoea. The abundance of Wolbachia was negatively correlated with that of Cardinium in both planthopper species. Compared with bacteria, the abundance and diversity of fungi did not differ between sexes but both were enriched in the gut. The bacterial community as a whole showed no significant correlation with the fungal community. The majority of fungi were related to Sarocladium, Alternaria, Malassezia, Aspergillus and Curvularia. A phylogenetic analysis revealed that these fungi were closely related to botanic symbionts or pathogens. Our results provide novel insights into the bacteria and fungi of rice planthoppers.  相似文献

Calanoid copepods play an important role in the pelagic ecosystem making them subject to various taxonomic and ecological studies, as well as indicators for detecting changes in the marine habitat. For all these investigations, valid identification, mainly of sibling and cryptic species as well as early life history stages, represents a central issue. In this study, we compare species identification methods for pelagic calanoid copepod species from the North Sea and adjacent regions in a total of 333 specimens. Morphologically identified specimens were analysed on the basis of nucleotide sequences (i.e. partial mitochondrial cytochrome c oxidase subunit I (COI) and complete 18S rDNA) and on proteome fingerprints using the technology of matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). On all three molecular approaches, all specimens were classified to species level indicated by low intraspecific and high interspecific variability. Sequence divergences in both markers revealed a second Pseudocalanus species for the southern North Sea identified as Pseudocalanus moultoni by COI sequence comparisons to GenBank. Proteome fingerprints were valid for species clusters irrespective of high intraspecific variability, including significant differences between early developmental stages and adults. There was no effect of sampling region or time; thus, trophic effect, when analysing the whole organisms, was observed in species‐specific protein mass spectra, underlining the power of this tool in the application on metazoan species identification. Because of less sample preparation steps, we recommend proteomic fingerprinting using the MALDI‐TOF MS as an alternative or supplementary approach for rapid, cost‐effective species identification.  相似文献

The morphologically convergent larvae of the echinoderm classes Ophiuroidea and Echinoidea have been suggested to be functionally dissimilar when it comes to their capacities to feed, but little is known about whether these larvae are similar in terms of energetics. Here, we compare the energetics of early development of a tropical ophiuroid, Ophiocoma alexandri, and a temperate to tropical echinoid, Arbacia punctulata, two species with similarly sized eggs. Measurements of respiration and constituent analyses were performed on eggs and unfed larvae of both species. Members of both species showed an increase in oxygen consumption during morphogenesis followed by a lower, static rate once morphogenesis was complete (3 d for O. alexandri and 1.3 d for A. punctulata). Compared to the echinoid larvae, the ophiuroid larvae developed more slowly and had peak respiration rates that were 3.1× lower. Eggs of O. alexandri contained significantly more protein and significantly less triacylglycerol than eggs of A. punctulata. Energy utilization, as calculated via respiration measurements, closely matched decreases in energy content from the eggs to larvae as measured with biochemical constituent assays. Larvae of A. punctulata used 1.4× more energy to reach the pluteus stage than larvae of O. alexandri, and used 4× more energy during the first 9 d of larval life. These data suggest that echinoid larvae require more energy to develop to the feeding stage than ophiuroid larvae, and likewise have higher requirements for maintenance metabolism. Ophiuroid larvae may be more tolerant of low food levels due to their very low metabolic rates, but this advantage may be offset by their slower rate of development.  相似文献

The Neotropical‐native figitid Aganaspis pelleranoi (Brèthes) and the Asian braconid Diachasmimorpha longicaudata (Ashmead) are two parasitoids of Tephritidae fruit flies with long and recent, respectively, evolutionary histories in the Neotropics. Both species experienced a recent range of overlap. In Argentina, A. pelleranoi is a potential species in biological control programs against the pestiferous tephritid species, Anastrepha fraterculus (Wiedemann) and Ceratitis capitata (Wiedemann), whereas D. longicaudata is already used in open‐field releases against Medfly in central‐western Argentina. To characterize the host‐foraging strategies of A. pelleranoi and D. longicaudata, olfactometer experiments were conducted comparing responses to C. capitata and A. fraterculus larvae, in two kinds of food substrate: fruit and artificial larval medium. To control the possible influence of host larvae used for parasitoid rearing on olfactory response, two strains of both parasitoid species, reared on both tephrtid species, were studied. Volatiles directly emanating either from A. fraterculus or C. capitata larvae may be detected by both A. pelleranoi and D. longicaudata, although chemical stimuli originating from the combination of host larvae and the habitat of the host were preferred. However, olfactory cues associated with host larvae probably play a relevant role in host searching behaviour of A. pelleranoi, whereas for D. longicaudata, the host‐habitat olfactory stimuli would be highly essential in short‐range host location. The strain of the parasitoids did not affect host search ability on the two tephritid species evaluated. These evidences are relevant for mass production of both parasitoids and their impact following open‐field augmentative releases.  相似文献

Cerithium koperbergi is a rare gastropod of the family Cerithiidae from the tropical Indo‐West Pacific. The species has a small, unusual shell and often inhabits deeper water, fore‐reef habitats that are atypical for the genus. Anatomical investigations reveal that it possesses a combination of features heretofore considered diagnostic of two main cerithiid subfamilies: Cerithiinae and Bittiinae. While the shell is bittiine, the animal lacks mesopodial pedal glands and possesses a seminal receptacle (vs. a spermatophore bursa) in the lateral lamina of the oviduct, which are considered to be cerithiine features. Re‐evaluation of the anatomy of Bittium reticulatum, the type species of Bittium, indicates the defining anatomical difference in oviduct anatomy between the two subfamilies does not stand up to closer scrutiny. Partial mitochondrial cytochrome c oxidase I (COI) sequences support the interpretation that C. koperbergi is a species complex around the western Pacific rim comprising three divergent mitochondrial lineages. Bayesian analysis of partial mitochondrial COI and 16S rRNA sequences confirm the placement of the C. koperbergi complex within a monophyletic Bittiinae, despite the apparent absence of a unifying anatomical feature. Species in the C. koperbergi complex are here united in Pictorium nov. gen. and two species are described as new. It is hypothesized that features of the midgut may be diagnostic of the Bittiinae, but more comparative data are needed.  相似文献

Lepidopteran defoliators are the most important pests of cultivated amaranths causing severe losses in cultivated fields worldwide. Leaf‐webbers, whose larvae fold, web or glue amaranth leaves using their silken webs as they feed and leaf‐worms which cause windowing but do not glue or fold leaves are mainly reported. Sustainable management strategies for these pests are still lacking given the adverse effects of synthetic pesticides. Field experiments were conducted during two seasons at two different sites in Central Kenya, to assess amaranth lepidopteran pests and their natural enemies’ population dynamics, evaluate the efficacy of phenylacetaldehyde (PAA) floral lure as attractant and the effects of three amaranth lines (Abuk1, Abuk2 and Abuk8) on the pests’ abundance and damage. Abundance of leaf‐webbers (p = .537), leaf‐worms (p = 1) and their associated parasitoids (p = .083) did not differ between the dry and wet seasons. The parasitoids Atropha tricolor and Apanteles sp. caused parasitism of 6.2% and 33.3% on Spoladea recurvalis and Choristoneura sp., respectively. PAA incorporated traps attracted moths that were largely unrelated to the damaging larvae observed on the crops with only 0.5% of total trap catches being S. recurvalis. Sub‐sites in which PAA were incorporated had significantly higher number of leaf‐webber larvae on the crops compared to control sub‐sites (p = .014). Amaranth lines studied had significant (p = .007) effect on lepidopteran defoliators’ abundance and damage, with fewer leaf‐webbers and lower severity of damage recorded on Abuk2 compared to Abuk8. The implication of these findings for the control of lepidopteran defoliators in East Africa is discussed.  相似文献

Pigeon pea is an important legume. Yield losses due to insect pests are enormous in the cultivation of this crop. Expression of cry proteins has led to increased resistance to pests in several crops. We report in this paper, expression of a chimeric cry1AcF (encoding cry1Ac and cry1F domains) gene in transgenic pigeon pea and its resistance towards Helicoverpa armigera. PCR, Southern hybridization, RT‐PCR and Western analysis confirmed stable integration and expression of the cry1AcF gene in pigeon pea transgenics. When screened for efficacy of the transformants for resistance against H. armigera, the transgenics showed not only high mortality of the larva but could also resist the damage caused by the larvae. Analysis for the stable integration, expression and efficacy of the transgenics resulted in the identification of four T3 plants arising from two T1 backgrounds as highly promising. The results demonstrate potentiality of the chimeric cry1AcF gene in developing H. armigera‐resistant pigeon pea.  相似文献

The tomato borer Tuta absoluta, native to western South America, is an extremely devastating pest in tomato crops in most of South America, Europe and Africa North of the Sahel, causes yield losses up to 100% and decreases fruit quality in open field and greenhouse crops if control methods are not applied. In Brazil two other important lepidopteran pests – Neoleucinodes elegantalis and Helicoverpa zea – occur in tomato, as well as thrips, whiteflies and aphids. For control of these pests, frequent applications of pesticides of up to 5 times per week are needed, and these resulted in the appearance of resistant populations to a number of active ingredients and decimation of natural enemies. Biological control may offer a better, safer and more sustainable opportunity for pest management. Mirid predatory bugs are currently used with success in southern Europe to control T. absoluta and other pests. In Brazil, four Hemipteran predatory bugs, not yet known to attack T. absoluta, were found to successfully prey on eggs and larvae of this pest. The first results on their predation capacity, development, survival and reproduction on T. absoluta on tomato plants are presented.  相似文献

Nuclear copies of mitochondrial DNA (NUMTs or mitochondrial pseudogenes) are known to impede the detection of interspecific genetic diversity. But the effect of these artifacts on phylogeographic reconstruction remains under evaluated. In this study, we analysed a set of 115 sequences of a fragment of the cytochrome c oxidase subunit I gene (COI) of Monochamus galloprovincialis (Coleoptera, Cerambycidae) for which overlapping signals in sequencing electropherograms were observed. Comparison of full and corrected ‘ambiguities‐free’ data sets reveals the prevalence of numerous supernumerary haplotypes that deeply affect genetic diversity indices and phylogeographic patterns of this species. Slightly divergent pseudogenes were recovered in 49 of the 115 sequences. These results highlight the potential misdetection of NUMTs using current control methods and the consequences on phylogeographic structure. To test the frequency of unintended amplification of NUMTs, a cloning was performed on 15 individuals. An average of 3.72 and a maximum of six paralogous sequences with different levels of divergence were identified among individual cloned. Within individual pairwise distance between paralogs raised 1.4%. This work calls for awareness to the presence of undetected NUMTs within mitochondrial data sets, especially at infra‐specific level.  相似文献

Eucryptorrhynchus scrobiculatus (Olivier) and E. brandti (Harold) are two wood boring pests of Ailanthus altissima (Mill.) Swingle (tree of heaven) and the variety Ailanthus altissima var. Qiantouchun. These beetles attack healthy trees and bore into roots and trunks during the larval stage. We studied the typology, distribution and morphostructure of the sensilla on the antennae, maxillary palps and labial palps of E. scrobiculatus and E. brandti larvae using scanning and transmission electron microscopy. The results showed the following: (i) the antennae of the two weevil larvae had two types of sensilla, sensilla basiconica (S.b.1 and S.b.2) and sensilla twig basiconica (S.tb.1‐S.tb.3), with S.tb.4 observed only on the antennae of E. brandti larvae; (ii) the maxillary palps had three types of sensilla, S.b.2, S.tb. (S.tb.2, S.tb.3 and S.tb.5) and digitiform sensilla; (iii) the labial palps had two types of sensilla, S.b.2 and S.tb. (S.tb.2, S.tb.3 and S.tb.5); (iv) the quantity and distribution of sensilla on the antennae, maxillary palps and labial palps remained constant between E. scrobiculatus and E. brandti larvae; and (v) sensilla basiconica had distinct sidewall pores, an apical pore was observed on sensilla twig basiconica, and digitiform sensilla were oval in shape, with a distinct apical pore. Based on the microstructure of the cuticle wall and dendrite, we hypothesized that these sensilla functioned as olfactory, gustatory and hygro‐/thermo‐receptors, respectively. We discuss the relationships among types of sensilla and the types of damage caused by the larvae inside the host tree to understand olfactory and gustatory receptor mechanisms. The results of this study will provide a firm basis for future electrophysiological studies.  相似文献

The leaf miner Coptodisca lucifluella and the carpophagous Rhagoletis completa, of American origin, are two non‐native walnut (Juglans spp.) pests in Italy. For the Friuli Venezia Giulia region (FVG) (N‐E Italy), C. lucifluella is not yet listed, while R. completa has been present for over 25 years. During 2015, samples of leaves and fruits were collected from 219 single old common walnut trees (Juglans regia) placed at different altitudes (0–1,073 m a.s.l.) in FVG to detect the distribution and abundance of both pests. Samples of leaf miner larvae and their parasitoids were subjected to mitochondrial DNA analysis for identification. C. lucifluella has been found in 55 out of 219 sites. The species has been identified by typical leaf symptoms and by its DNA barcode. This is the first report of the species for FVG. The leaf miner distribution was negatively correlated with altitude. The species has not been observed at sites over 600 m a.s.l. Larvae were parasitized by native parasitoids identified as belonging to the genus Chrysocharis by morphological features and by DNA barcode. R. completa has been found in 89 out of 165 sites on walnut trees with fruits. The infestation level was very high in lowland localities. At sites over 700 m a.s.l., no infestation was found. Both species are widespread in the region, and their occurrence is significantly affected by altitude. Data suggest that walnut trees could be cultivated in some mountain areas without the need to control R. completa with insecticides.  相似文献

China is the largest walnut producer in the world, and walnut trees, especially English walnut, are widely distributed in the country. Species of Botryosphaeriaceae include important plant pathogens that can cause diseases on many tree crops including English walnut. Recently, disease symptoms caused by Botryosphaeriaceae were observed on English walnut branches or kernels from Beijing, Henan and Sichuan provinces in China. Based on morphological characteristics and phylogenetic analyses of the ITS rDNA sequences and translation elongation factor 1‐alpha (TEF‐1α) gene regions, Botryosphaeria dothidea and Lasiodiplodia pseudotheobromae were identified. Pathogenicity tests showed that both species are virulent to English walnut. To our knowledge, this is the first report of L. pseudotheobromae infecting English walnut in the world.  相似文献

Species richness and similarity in metazoan parasite communities of fishes can be influenced by several biotic (age, body size, vagility, feeding and social behavior, among others), and local abiotic (temperature, salinity, etc.) factors. The parasite communities of three species of Oligoplites, marine fishes from the Pacific coast of Mexico, were quantified and analyzed. Four hundred sixty‐eight leatherjackets (O. altus, n=94; O. saurus, n=260; and Orefulgens, n=114) were collected from February 2016 to June 2017 from five locations. Twenty‐eight species of metazoan parasites were recovered and identified: four species of Monogenea (adults), nine of Digenea (seven adults and two metacercariae); two of Cestoda (larvae); four of Nematoda (two adults and two larvae); four of Acanthocephala (two adults, one juvenile, and one cystacanth); four of Copepoda; and one Pentastomida (larvae). At the component community level, species richness ranged from 9 in O. saurus to 19 in O. altus. Different species of helminth dominated the component communities of each species of host. Community composition and species richness of parasites differed among the three species of host, locations, and sampling years. Host feeding behavior, body size, and vagility had the most influence on these differences.  相似文献
