一株高含玫红品的红树林海洋紫色硫细菌分离鉴定及特性   总被引:3,自引:0,他引:3  
摘要:【目的】挖掘我国海洋紫色硫细菌物种资源、深入理解紫色硫细菌在红树林生态系统中的作用。【方法】采用琼脂振荡稀释法、显微技术、紫外可见吸收光谱法、TLC、HPLC 和MS 法。【结果】从福建泉州洛阳桥红树林潮间带泥水样分离获得一株细胞内含多个硫粒的细菌菌株,光合内膜呈囊状、含细菌叶绿素a 和螺菌黄质系类胡萝卜素,结合16S rRNA 基因序列分析和系统发育分析,表明该菌株属于海洋着色菌属(Marichromatium)。该菌株细胞球杆状;最适pH 范围5.7-6.7;最适盐度范围2%-3.5%;温度范围20℃ -35℃;能耐受3.6 mmol/L 硫化物;主要积累玫红品类胡萝卜素,而不是螺菌黄质;3 种细菌叶绿素组分中,一种为BChl aP,另2 种未见报道;不需要生长因子;可光同化固定CO2、能很好利用多种有机酸盐、多价态氮化物和硫化物,尤其能利用柠檬酸、葡萄糖、蔗糖、果糖和丙醇;对氯霉素、头孢唑林、苯、对羟基联苯、恩诺沙星、啶虫脒、HgCl2 和CdCl2的IC50 值分别为70、100、20、20、3、170、5 mg /L和25 mg/L。【结论】该菌株是一株轻度耐酸、高含玫红品类胡萝卜素的紫色硫细菌,被鉴定为Marichromatium gracile 新菌株,编号YL28。该菌株具有广泛利用多种碳源、氮源和硫源物质的能力,对抗生素氯霉素和头孢唑啉、农药啶虫脒、重金属汞和镉具有较强耐受性,对抗生素恩诺沙星较敏感。  相似文献

冲绳海槽热液区可培养硫氧化细菌多样性及其硫氧化特性   总被引:2,自引:0,他引:2  
冲绳海槽热液区独特的地质环境孕育了特殊的生物群落,硫氧化细菌作为生物地球化学循环的重要参与者在热液生态系统中发挥着至关重要的作用。【目的】通过硫氧化菌株的分离培养揭示冲绳海槽热液区可培养硫氧化细菌的多样性和硫氧化活性。【方法】采用多种培养基对冲绳海槽热液区不同沉积物样品中的硫氧化细菌进行富集培养和分离纯化;利用16S rRNA基因序列确定硫氧化细菌的分类地位并进行系统发育分析;采用碘量法对典型硫氧化菌株硫氧化活性进行检测。【结果】本研究从冲绳海槽热液区样品中共分离鉴定85株硫氧化细菌,分属于α-变形菌纲、γ-变形菌纲、放线菌门和厚壁菌门,优势属为氢弧菌属(Hydrogenovibrio)、拉布伦氏菌属(Labrenzia)、深海海旋菌属(Thalassospira)和海杆状菌属(Marinobacter)。硫氧化活性检测结果表明,7株典型硫氧化菌株对硫代硫酸钠的降解活性介于31%–100%之间,其中泰坦尼克号盐单胞菌SOB56 (Halomonas titanicae SOB56)、南极海杆状菌SOB93(Marinobacter antarcticus SOB93)、印度硫氧化粗杆菌SOB107 (Thioclava indica SOB107)和嗜温氢弧菌CJG136 (Hydrogenovibrio thermophiles CJG136)可以完全降解硫代硫酸钠。【结论】冲绳海槽热液区可培养硫氧化细菌的多样性丰富,为研究该热液区的硫循环过程提供了实验材料和理论基础,多种硫氧化活性菌株的获得极大地丰富了菌种资源,为探究深海热液区硫循环的能量代谢途径和分子机制奠定基础。  相似文献

【目的】利用培养法从日本三宅岛火山土壤(堆积年限131年)中分离到一株能氧化分解硫代硫酸盐的细菌MU2A-22T。【方法】用培养法对该菌株MU2A-22T进行了生理生化性质以及分类学位置上的确定。【结果】菌株MU2A-22T为革兰氏阴性,短杆状或球状。理化性质表明该菌株能利用葡萄糖、L-阿拉伯糖、葡萄糖酸盐、己二酸酯、dL-苹果酸钠、硫代硫酸钠(最适浓度为2.5 mmol/L)为唯一碳源进行自养生长。最适生长温度为25°C 30°C,最适pH为6.0 8.0。菌株MU2A-22T的16S rRNA序列与菌株Paracoccus solventivorans 6637T亲缘关系最近,序列相似性为97%,编码核酮糖-1,5-二磷酸羧化酶/加氧酶(Rubisco)的基因也被确定。对Paracoccus属内几种近缘菌的脂肪酸分析,证明菌株MU2A-22T中含有Paracoccus属的特征氨基酸,其中含量大于10%的分别为C18:1(74.7%)和C18:0(12.1%)。DNA-DNA杂交实验表明,菌株MU2A-22T与Paracoccus solventivorans 6637TDNA的相似度为49.3%。MU2A-22T菌株G+C含量为66.5%66.7%。【结论】菌株MU2A-22T为Paracoccus属内的一新种菌(登录号GQ452286),命名为Paracoccus scorialis sp.nov.。  相似文献

【目的】开展具有硫氧化能力的嗜酸硫杆菌属(Acidithiobacillus)的分离及其比较基因组学分析,不仅可以丰富硫氧化细菌菌种资源,而且有助于加深理解嗜酸硫杆菌的分子进化与生态适应机制。【方法】利用以硫代硫酸钠为唯一能源的培养基分离具有硫氧化能力的细菌;利用Illumina HiSeq X和Oxford Nanopore测序平台对一株嗜酸硫杆菌M4-422-6进行全基因组测序;利用相关生物信息学分析软件对原始数据进行组装和基因组注释,并与一株亲缘关系最近的菌株Igneacidithiobacillus copahuensis VAN18-1进行比较基因组学分析。【结果】分离获得一株具有硫氧化能力的嗜酸硫杆菌M4-422-6。基因组注释结果显示,菌株M4-422-6基因组由1个染色体和2个质粒组成,基因组大小为2 917 823 bp,G+C含量为58.54%,共编码2 925个蛋白。16S rRNA基因和基因组系统发育树显示,菌株M4-422-6代表嗜酸硫杆菌属的一个潜在新种。功能基因注释结果显示,菌株Acidithiobacillus sp. M4-422-6拥有与菌株特性相关的众多基因,包括硫氧化相关基因、CO2固定相关基因和耐酸相关基因。比较基因组学分析发现,虽然菌株M4-422-6与VAN18-1的亲缘关系最近,但两者仍拥有众多的差异基因,主要包括噬菌体抗性相关基因和移动元件编码基因。【结论】菌株M4-422-6代表嗜酸硫杆菌属的一个潜在新种,该菌株具有同种内菌株所不具有的特有基因,并据此推测嗜酸硫杆菌种内分化可归因于对特定生态位的适应。  相似文献

从内蒙古碱湖水样中分离得到一株紫色非硫光合细菌,命名为JH1-6.对该菌株进行了形态学观察、生理生化鉴定、活细胞吸收光谱以及16S rDNA序列分析.16S rDNA序列分析结果表明该菌株与沼泽红假单胞菌的16S rDNA序列同源性高达99%,结合形态特征和生理生化特性以及活细胞吸收光谱特征等,确定菌株JH1- 6在分类地位上属于沼泽红假单胞菌(Rhodopseudomonas palustris).  相似文献

新疆泥火山产酶嗜盐放线菌的筛选及多样性   总被引:2,自引:0,他引:2  
[目的]了解新疆乌苏泥火山嗜盐放线菌及其产酶功能多样性.[方法]分别采用含有5%与10%NaCl的5种分离培养基,稀释平板涂布法对泥火山土壤样品进行分离;利用五种筛选培养基定性检测酶活性;在形态特征、耐盐性实验及16S rDNA基因测序的基础上进行系统发育学分析.[结果]获得嗜盐放线菌43株,极端嗜盐放线菌3株.4株嗜盐放线菌产脂肪酶,30株产半乳糖苷酶,27株产淀粉酶,6株产酯酶,4株产纤维素酶,1株同时产4种酶.系统发育学分析结果表明其中24株为拟诺卡氏菌属(Nocardiopsis),1株为链霉菌属(Streptomyces).产两种酶的菌株10006与Nocardiopsis exhalans(AY03600)相似性为96.64%(小于97%),可能是潜在的新种.[结论]本研究表明新疆乌苏泥火山中存在大量的产半乳糖苷酶及淀粉酶的嗜盐放线菌,所分离到的拟诺卡氏菌属产酶多样性比较高,并且潜藏着新的微生物资源.  相似文献

[目的]对从健康桑树叶片中分离到的一株内生拮抗细菌Lu10-1进行鉴定,并探讨该菌株在桑树体内的定殖.[方法]通过形态观察、生理生化指标测定及16S rRNA基因序列同源性分析,结合recA基因特异引物PCR检测法对菌株Lu10-1进行分类学鉴定;以抗利福平(Rif)和氨苄青霉素(Amp)双抗药性为标记,采用浸种、浸根、涂叶和针刺等方法接种,测定Lu10-1菌株在桑树体内的定殖.[结果]结果表明,菌株Lu10-1属于伯克霍尔德氏菌属(Burkholderia),与亲缘关系较近菌株B.cepacia(X80284)的同源性达98%,该菌株的16S rDNA序列已在GenBank中注册,登录号为EF546394;Lu10-1菌株浸种接种后,菌株在桑苗组织中的数量总体上呈现下降趋势,到第20天后菌量趋于稳定;细菌浸根接种后,菌株在茎叶部定殖的菌量均呈现出"先增后降"的趋势.[结论]内生拮抗细菌Lu10-1归属于洋葱伯克霍尔德氏菌基因型Ⅰ(Burkholderia cepacia genomovar Ⅰ);该菌株可在桑树体内长期定殖并传导,且在定殖过程中菌株的拮抗性能未改变;为将该菌株导入桑树体内进行病害的生物防治提供了理论依据.  相似文献

从山西运城盐厂解化池分离获得一株嗜盐嗜碱细菌,编号为Y。其纯培养物经形态学、生理生化特性和DNA G+C含量等特征分析,结果表明,该菌株可在盐度ρ(NaCl)/gL-1为160和pH9.0碱性条件下生长。单细胞为杆状,大小为0.4-0.8×0.9-1.5μm。二分裂繁殖。革兰氏阴性。光合内膜为片层堆积并与细胞质膜相连,但并不与细胞质膜平行。细胞含有细菌叶绿素a,液体培养物呈玫瑰红色。Y菌株不仅可将硫化物氧化为元素硫沉积于细胞外,也可光同化多种有机物,并具有固氮和产H2特性。DNA中 G+C含量为61.6%。根据以上鉴定特征及相关资料,Y菌株应归于嗜盐红螺菌属(Halorhodospira)。但其生长所依赖的盐度、pH、细胞内色素成分及鞭毛着生方式与该属正式承认的3个种有明显的不同。该菌株独特的生理生化特性,对于极端环境微生物的资源开发和利用有重要的理论意义和应用价值。  相似文献

[目的]以结瘤豆科植物紫花苜蓿根际土壤为研究材料,筛选具有ACC脱氨酶活力的氢氧化细菌,探索氢氧化细菌植物促生作用机制.[方法]利用持续通H2 的气体循环培养体系、矿质盐固体培养基,分离、培养氢氧化细菌,观察菌株形态并测定生理生化特征;16S rDNA序列分析法构建系统发育树;采用薄层层析法筛选ACC脱氨酶阳性菌株,茚三酮显色法测定ACC脱氨酶活力.[结果]分离的37株细菌中有8株菌氧化氢和自养生长能力较强,初步确定为氢氧化细菌,从中筛选出1株ACC脱氨酶阳性菌株WMQ-7.菌株WMQ-7的形态特征、生理生化特征与恶臭假单胞菌(Pseudomonas putida)的特征基本一致;16s rDNA序列(GenBank登录号为EU807744)在系统发育树中与恶臭假单胞菌同属一个类群,序列同源性99%.鉴定菌株WMQ-7为恶臭假单胞菌,其ACE脱氨酶活力为0.671 U/μg[结论]采用气体循环培养体系分离氢氧化细菌,克服了传统配气法的局限.ACC脱氨酶阳性菌株的筛选,为深入研究氢氧化细菌作为植物根际促生菌的菌株特性和促生机制提供理论依据.  相似文献

硇洲岛潮汐带牡蛎相关可培养细菌多样性   总被引:1,自引:0,他引:1  
[目的]了解南海硇洲岛潮汐带香港巨牡蛎(Crassostrea hongkongensis)相关可培养细菌的多样性.[方法]应用纯培养法分离样品中的细菌(含放线菌),采用基于16S rRNA基因序列的系统发育分析方法研究这些分离菌株的生物多样性.[结果]用补充0-25%(W/V) NaC1的MA、MH和NA培养基从样品中分离到102株细菌,在形态观察和部分生理生化实验结果的基础上去冗余,选取74个代表性菌株进行基于16S rRNA基因序列的系统发育分析.结果表明,这些菌株归为38个物种,属于4个大的系统发育类群(Gamma-Proteobacteria,Alpha-Proteobacteria,Bacteroidetes,Firmicutes)的16个科、18个属.大多数菌株属于Gamma-Proteobacteria亚门(45株,60.8%),其余依次是Firmicutes门(12株,16.2%)、Bacteroidetes门(11株,14.9%)和Alpha-Proteobacteria亚门(6株,8.1%).大多数菌株与其系统发育关系最密切的有效发表种典型菌株之间(16S rRNA基因序列相似性为94.3%-99.8%)存在一定的遗传差异,其中JSM 111069可能代表Carnobacteriaceae科的潜在新属;菌株JSM 111039和JSM 111085可能分别代表Bacillus属的两个新物种,菌株JSM 111020、JSM 111072和JSM 111090可能分别代表Pseudoalteromonas、Proteus和Idiomarina属的新物种.[结论]南海硇洲岛潮汐带牡蛎中存在较为丰富的原核生物多样性,并潜藏着一定数量的微生物新类群(物种).  相似文献

A new strain of purple sulfur bacterium was isolated from a marine microbial mat sampled in Great Sippewissett Salt Marsh at the Atlantic coast (Woods Hole, Mass., USA). Single cells of strain AZ1 were coccus-shaped, highly motile by means of a single flagellum, and did not contain gas vesicles. Intracellular membranes were of the vesicular type. However, additional concentric membrane structures were present. The photosynthetic pigments were bacteriochlorophyll a and carotenoids of the normal spirilloxanthin series, with rhodopin as the dominant carotenoid. Hydrogen sulfide (up to 11 mM), sulfur, thiosulfate, and molecular hydrogen were used as electron donors during anaerobic phototrophic growth. During growth on sulfide, elemental sulfur globules were transiently stored inside the cells. Strain AZ1 is much more versatile than most other Chromatiaceae with respect to electron donor and organic substrates. In the presence of CO(2), it is capable of assimilating C(1)-C(5) fatty acids, alcohols, and intermediates of the tricarboxylic acid cycle. Strain AZ1 could also grow photoorganotrophically with acetate as the sole photosynthetic electron donor. Chemotrophic growth in the dark under microoxic conditions was not detected. Optimum growth occurred at pH 6.5-6.7, 30-35 degrees C, > or =50 micro mol quanta m(-2) s(-1), and 2.4-2.6% NaCl. The DNA base composition was 64.5 mol% G+C. Comparative sequence analysis of the 16S rRNA gene confirmed that the isolate is a member of the family Chromatiaceae. Sequence similarity to the most closely related species, Thiorhodococcus minor DSMZ 11518(T), was 97.8%; however, the value for DNA-DNA hybridization between both strains was only 20%. Because of the low genetic similarity and since strain AZ1 physiologically differs considerably from all other members of the Chromatiaceae, including Trc. minor, the new isolate is described as a new species of the genus Thiorhodococcus, Thiorhodococcus drewsii sp. nov.  相似文献

A new marine phototrophic purple sulfur bacterium (strain CE2203) was isolated in pure culture from a man-made coastal lagoon located on the Atlantic coast (Arcachon Bay, France). Single cells were coccus-shaped, did not contain gas vesicles, and were highly motile. Intracellular photosynthetic membranes were of the vesicular type. Bacteriochlorophyll a and carotenoids of the normal spirilloxanthin series were present as photosynthetic pigments. Hydrogen sulfide, thiosulfate, elemental sulfur, and molecular hydrogen were used as electron donors during photolithotrophic growth under anoxic conditions, while carbon dioxide was utilized as carbon source. Acetate, propionate, lactate, glycolate, pyruvate, fumarate, succinate, fructose, sucrose, ethanol, and propanol were photoassimilated in the presence of hydrogen sulfide. During growth on sulfide, elemental sulfur globules were stored inside the cells. Chemotrophic growth under microoxic conditions in the dark was possible. The DNA base composition was 66.9 mol% G+C. Comparative sequence analysis of the 16S rRNA gene confirmed the membership of strain CE2203 in the family Chromatiaceae. Morphological characteristics of strain CE2203 indicated a close affiliation to the genera Thiocystis and Thiocapsa. However, the phylogenetic treeing revealed no closer relationship to Thiocystis spp. than to Thiocapsa roseopersicina or other known members of the Chromatiaceae. Strain JA430(T) is a Gram-negative, vibrioid to spiral shaped phototrophic purple sulfur bacterium isolated from anoxic sediment of a saltern at Kanyakumari in a mineral salts medium that contained 2% NaCl (w/v). Strain JA430(T) grows optimally at 5-6% NaCl and tolerates up to 12% NaCl. Intracellular photosynthetic membranes were of the lamellar type. Bacteriochlorophyll a and carotenoids of the spirilloxanthin series are present as photosynthetic pigments. Major cellular fatty acids are C(18:1)ω7c, C(16:0), C(19:0)cycloω8c and C(16:1)ω7c/C(16:1)ω6c. Strain JA430(T) exhibits photoorganoheterotrophy and chemoorganoheterotrophy and requires para-aminobenzoic acid, pantothenate and pyridoxal phosphate for growth. Phylogenetic analysis on the basis of 16S rRNA gene sequence analysis showed that strain JA430(T) forms monophyletic group in the genus Ectothiorhodospira. The highest sequence similarity for strain JA430(T) was found with the type strains of Ectothiorhodospira variabilis DSM 21381(T) (96.1%) and Ectothiorhodospira haloalkaliphila ATCC 51935(T) (96.2%). Morphological and physiological characteristics discriminate strain JA430(T) from other species of the genus Ectothiorhodospira, for which we describe this as a novel species, Ectothiorhodospira salini sp. nov. A new phototrophic purple bacterium was isolated from a flat, laminated microbial mat in a salt marsh near Woods Hole, Mass., USA. The spiral-shaped bacterium was highly motile and had bipolar tufts of flagella and intracytoplasmic membranes of the vesicular type. The major photosynthetic pigments were identified as the carotenoid tetrahydrospirilloxanthin and bacteriochlorophyll b. The long wavelength in vivo absorption maximum of the bacteriochlorophyll was at 986 nm. The marine bacterium showed optimal growth in the presence of 2% NaCl. It utilized a number of organic substrates as carbon and energy sources and required vitamins and sulfide as a reduced sulfur source for growth. In the presence of sulfide, elemental sulfur globules were formed outside the cells. Elemental sulfur was not further oxidized to sulfate. The new isolate had a unique lipid and fatty acid composition, and according to the 16S rRNA gene sequence, it is most similar to Rhodospirillum rubrum. A purple sulfur bacterium, strain SW26, was isolated in pure culture from intertidal sediments from the Sylt-Rømø Basin, German Wadden Sea, sharing many properties with validated Marichromatium species, but differing significantly by possessing a plasmid, by tolerating up to 16 mM sulfide, and up to 44 °C for growth. Strain SW26 has a DNA base composition of 68.3 mol% G+C, a 16S rRNA gene sequence similarity of >99% to those of Marichromatium species, and shows the highest level of genomic relationship with Marichromatium gracile, despite its remarkably different phenotypic characters. Based upon high genomic similarity but different physiological properties of strain SW26 with respect to the type strain of M. gracile, a novel biotype, designated as M. gracile biotype thermosulfidiphilum is described.  相似文献

During the oxidation of sulfide and thiosulfate purple and green sulfur bacteria accumulate globules of 'elemental' sulfur. Although essential for a thorough understanding of sulfur metabolism in these organisms, the exact chemical nature of the stored sulfur is still unclear. We applied sulfur K-edge X-ray absorption near edge spectroscopy (XANES) to probe the forms of sulfur in intact cells. Comparing XANES spectra of Allochromatium vinosum, Thiocapsa roseopersicina, Marichromatium purpuratum, Halorhodospira halophila and Chlorobium vibrioforme grown photolithoautotrophically on sulfide with reference probes (fingerprint method), we found sulfur chains with the structure R-S(n)-R. Evidence for the presence of sulfur rings, polythionates and anionic polysulfides in the sulfur globules of these bacteria was not obtained.  相似文献

A dense 1 m thick layer of phototrophic purple sulfur bacteria is present at the pycnocline (17.8 m depth) in the meromictic South Andros Black Hole cave system (Bahamas). Two purple sulfur bacteria present in samples collected from this layer have been identified as belonging to the family Chromatiaceae. One isolate (BH-1), pink coloured, is non-motile, non-gas vacuolated, 2-3 microm in diameter and surrounded by a capsule. The other isolates (BH-2 and BH-2.4), reddish-brown coloured, are small celled (4 microm x 2 microm), motile by means of a single polar flagellum. In both isolates (BH-1 and BH-2), the intracellular photosynthetic membranes are of the vesicular type and bacteriochlorophyll a and carotenoids of the normal spirilloxanthin series are present. Both isolates grow well in the presence of sulfide and carbon dioxide in the light. During photoautotrophic growth sulfur globules are stored intracellularly as intermediate oxidation products. According to the 16S rRNA gene sequence data the isolates belong to the genera Thiocapsa and Allochromatium. However, at the species level a number of inconsistencies exist between the phenotypic and phylogenetic data, highlighting taxonomic problems within these genera. These inconsistencies may have implications for microbiologists studying the ecology of anoxygenic phototrophs. For ecologists studying the functioning of an ecosystem it may not be particularly important to know whether a specific isolate belongs to one species or another. However, if one wants to study the role of different populations within a particular functional group then the species concept is important. This study demonstrates that further work is still required on the taxonomy of purple sulfur bacteria in order that microbial ecologists are able to accurately identify a population/species isolated from hitherto undescribed aquatic ecosystems.  相似文献

A new type of phototrophic purple bacterium, strain 930I, was isolated from a microbial mat covering intertidal sandy sediments of Great Sippewissett Salt Marsh (Woods Hole, Mass., USA). The bacterium could only be enriched at a wavelength of 932 (± 10) nm. Cells were vibrioid- to spirilloid-shaped and motile by means of bipolar monotrichous flagellation. The intracytoplasmic membranes were of the lamellar type. Photosynthetic pigments comprised bacteriochlorophyll a and the carotenoids spirilloxanthin and lycopenal. The isolated strain exhibited an unusual, long-wavelength absorption maximum at 911 nm. Sulfide or thiosulfate served as electron donor for anoxygenic phototrophic growth. During growth on sulfide, elemental sulfur globules formed outside the cells. Elemental sulfur could not be further oxidized to sulfate. In the presence of sulfide plus bicarbonate, fructose, acetate, propionate, butyrate, valerate, 2-oxoglutarate, pyruvate, lactate, malate, succinate, fumarate, malonate, casamino acids, yeast extract, L(+)-alanine, and L(+)-glutamate were assimilated. Sulfide, thiosulfate, or elemental sulfur served as a reduced sulfur source for photosynthetic growth. Maximum growth rates were obtained at pH 7.9, 30 °C, 50 μmol quanta m–2 s–1 of daylight fluorescent tubes, and a salinity of 1–2% NaCl. The strain grew microaerophilically in the dark at a partial pressure of 1 kPa O2. The DNA base composition was 71.2 mol% G + C. Sequence comparison of 16S rRNA genes indicated that the isolate is a member of the α-Proteobacteria and is most closely related to Rhodobium orientis at a similarity level of 93.5%. Because of the large phylogenetic distance to known phototrophic species of the α-Proteobacteria and of its unique absorption spectrum, strain 930I is described as a new genus and species, Roseospirillum parvum gen. nov. and sp. nov. 从云南腾冲热海酸性温泉中分离纯化出一株极端嗜酸热菌株K4-1,并对其进行形态观察、生长特征、碳源和能源利用及16S rRNA基因分析.该菌株细胞形态为不规则球形,有单生鞭毛,严格好氧,兼性自养,能利用元素硫作为能源,也能利用酵母膏、精氨酸或核糖作为碳源和能源.其最适生长温度为75℃,最适pH为3.5.通过16S rRNA基因序列相似性对比对该菌株进行鉴定,结果表明该菌株与硫化叶茵属标准菌株的相似性介于86.6%~94.3%之间,与分离自腾冲热海的腾冲硫化叶菌Sulfolobus tengchongensis RT8-4菌株序列相似性最高,达到98.9%,可将菌株K4-1鉴定为硫化叶菌属菌株.菌株K4-1的16S rRNA基因序列号为EU729124.  相似文献

A new purple sulfur bacterium (strain M9) was isolated from the steppe soda Lake Dzun Uldziin Nur (pH 9.4; mineralization, 3.3%) situated in southeastern Mongolia. Individual cells appear as vibrios 0.3-0.5 x 0.7-1 micron in size. The dividing cells often do not separate from each other, forming an almost closed ring. The internal photosynthetic membranes are represented by concentric lamellae lining the cell wall. Photosynthetic pigments are bacteriochlorophyll a and carotenoids of the spirilloxanthin series. The main carotenoid (> 96%) is spirilloxanthin. Two typical light-harvesting complexes (LH1 and LH2) are present in the membranes in a 1:1 ratio. The bacterium is an anaerobe and facultative photoorganoheterotroph. Photolithoautotrophic growth on sulfide is scarce. Thiosulfate is utilized as an electron donor only in the presence of organic matter. Globules of elemental sulfur are formed as an intermediary product of sulfide and thiosulfate oxidation and are deposited outside the cells. The end product of oxidation is sulfate. In the presence of sulfide and carbonates, acetate, lactate, malate, pyruvate, propionate, succinate, and fumarate are used as the additional sources of carbon in anoxygenic photosynthesis. Vitamin are not required. The bacterium is an alkaliphile the pH optimum is at 8.3-9.1, the pH range is 7.6-10.1. The optimum NaCl concentration in the medium is 1 to 7%; the range is 0.5 to 0.9%. The optimum carbonate content in the medium is 2%; the range is 1 to 10%. The best growth occurs at 30-35 degrees C. The DNA G + C content is 57.5 mol%. According to the results of analysis of the 16S rRNA gene sequences, the new isolate M9 belongs to the phylogenetic cluster containing representatives of the family Ectothiorhodospiraceae within the class "Gammaproteobacteria." In this class, the new isolate forms a new branch, which occupies an intermediate position between the representatives of the genera Ectothiorhodospira and Thiorhodospira. Based on the phenotypic and genetic characteristics, the new purple sulfurbacterium was assigned to a new species of a new genus of the family Ectothiorhodospiraceae, Ectothiorhodosinus mongolicum gen. nov., sp. nov.  相似文献
