Pseudomonas sp. B21C9, a new nitrile-degrading microorganism, was isolated from a soil sample. By the hydrolysis of (RS)-iso-propyl- 4´-chlorophenylacetonitrile using cells of B21C9, (S)-2- isopropyl-4´ -chlorophenylacetic acid having excellent optically purity could be obtained. It appeared that the microbial hydrolysis proceeded via stepwise reactions by a nitrile hydratase having poor (S)-selectivity and an amidase having strict (S)-selectivity  相似文献

The hydrolysis of PEtOx is studied to evaluate the potential toxicity of partially hydrolyzed polymers that might interfere with its increasing popularity for biomedical applications. The hydrolysis of PEtOx is studied in the presence of digestive enzymes (gastric and intestinal) and at 5.8?M hydrochloric acid as a function of temperature (57, 73, 90, and 100?°C). It is found that PEtOx undergoes negligible hydrolysis at 37?°C and that thermal and solution properties are not altered when up to 10% of the polymer backbone is hydrolyzed. Mucosal irritation and cytotoxicity is also absent up to 10% hydrolysis levels. In conclusion, PEtOx will not decompose at physiological conditions, and partial hydrolysis will not limit its biomedical applications.  相似文献

 The analogy between H-bonded nucleobase pairs and their metalated analogues is extended to the hemiprotonated pair of 7,9-dimethylguanine (7,9-DimeG) and the Watson-Crick and reversed Watson-Crick pair between 7,9-dimethylguaninium (7,9-DimeGH+) and 1-methylcytosine (1-MeC). The crystal structure analyses of two model compounds, trans–[Pt(CH3NH2)2(7,9-DimeG-N1)2](NO3)2 (1) and trans–[Pt(NH3)2(1-MeC-N3)(7, 9-DimeG-N1)](PF6)2· 2.5 H2O (3a) are reported. Pt binding is through N1 of 7,9-DimeG and N3 of 1-MeC. In solution, 3a exists in a mixture with Watson-Crick and reversed Watson-Crick arrangements of the two bases, depending on solvent, concentration and anions. Abstract

Liquid chromatography was used to follow the degradation of monoamino analogues of 2′- or 3′-deoxyadenosine and of 9-(2-deoxy-β-D-threo-pentofuranosyl) adenine or 9-(3-deoxy-β-D-threo-pentofuranosyl) adenine in buffers of different pH and constant ionic strength (μ). Comparison of stabilities of some of the compounds under study with those of corresponding hydroxyl analogues showed that at acid pH the aminated compounds are more stable than the corresponding hydroxyl compounds. The higher stability associated with the presence of an amino group in the sugar is explained in function of pKa values, which were determined by 13C NMR.  相似文献

A novel series of cis-3,4-diphenylpyrrolidines were designed as RORγt inverse agonists based on the binding conformation of previously reported bicyclic sulfonamide 1. Preliminary synthesis and structure–activity relationship (SAR) study established (3S,4S)-3-methyl-3-(4-fluorophenyl)-4-(4-(1,1,1,3,3,3-hexafluoro-2-hydroxyprop-2-yl)phenyl)pyrrolidine as the most effective scaffold. Subsequent SAR optimization led to identification of a piperidinyl carboxamide 31, which was potent against RORγt (EC50 of 61 nM in an inverse agonist assay), selective relative to RORα, RORβ, LXRα and LXRβ, and stable in human and mouse liver microsomes. Furthermore, compound 31 exhibited considerably lower PXR Ymax (46%) and emerged as a promising lead. The binding mode of the diphenylpyrrolidine series was established with an X-ray co-crystal structure of 10A/RORγt.  相似文献

An unprecedented octanuclear aggregate, [{Co(phen)2}6{W(CN)8}2Cl2] · 2Cl, 2, resulted from the assembling of {Co(phen)2Cl2}, 1, and {W(CN)8}4?. Surprisingly, the reaction with the paramagnetic {Nb(CN)8}4? unit did not afford the homologous {Co–Nb} cluster. Instead the latter building unit undergoes dissociation which led to the formation of a mixed-valence [{CoII(phen)2}{CoIII(phen)(CN)4}2], 3. This observation is in contrast to the usual trend that {NbIV(CN)8}4? forms compounds isostructural to that observed for {MoIV(CN)8}4? and {WIV(CN)8}4?. The structures of the compounds 2 and 3 have been established by single crystal X-ray diffraction. Magnetic behaviors for compounds 13 are reported.  相似文献

Enzymes and microorganisms were screened for the asymmetric hydrolysis of (R, S)-5-acetoxymethyl-3-tert-butyl-oxazolidin-2-one 1. Lipases from Pseudomonas aeruginosa and Alcaligenes species, and microorganisms which belong to Enterobacter species or Klebsiella species were found to hydrolyze 1 enantioselectively to give (R)-5-hydroxymethyl-3-tert-butyl-oxazolidin-2-one (R)-2 and (S)-l. (S)-2, one of the typical intermediates for preparing optically active β-blocking agents (β-blockers), was obtained with high enantiomeric excess (91~98% e.e.) from (S)-1.  相似文献

Ce(NH4)2(NO3)6对蔓茎堇菜悬浮细胞凋亡和总黄酮含量的影响   总被引:1,自引:0,他引:1  
用DNA Laddering和DAPI荧光检测法对Ce(NH4)2(NO3)6诱导的蔓茎堇菜(Viola diffusa Ging.)悬浮细胞的凋亡情况进行研究,同时还测定了诱导过程中悬浮细胞的生长量和总黄酮含量。结果表明,Ce(NH4)2(NO3)6可诱导蔓茎堇菜悬浮细胞发生凋亡,1.0 mmol.L-1Ce(NH4)2(NO3)6胁迫诱导培养8 d时,细胞凋亡率最高(55.7%);总黄酮含量显著增加,达2.820%。表明Ce(NH4)2(NO3)6在诱导蔓茎堇菜悬浮细胞凋亡并抑制其生长的同时,还能促进黄酮类化合物的合成。  相似文献

 The synthesis of cis-Pt(NH3)2(dCMP) is reported and by various physico-chemical methods it is demonstrated that it is a macrochelate in which Pt(II) is bound simultaneously to the N3 site of cytosine in dCMP2– and to a phosphate-oxygen atom. According to the NOESY spectra (cross-peaks between cytosine H6 and H2′ and H3′) the cytosine ring adopts an anti orientation. Highly unusual is the significant (1 ppm) downfield shift of the sugar proton H5″ in the 1H-NMR spectrum and the sensitivity of the cytosine H6 resonance on the protonation state of the phosphate group. Based on these three features a geometry for the macrochelate is proposed. The compound is a major product of the reaction of cis-[Pt(NH3)2(H2O)2]2+ with dCMP2– at neutral pH, but it even forms at pH 5. By applying pD-dependent NMR spectroscopy (1H, 31P) and potentiometric pH titration, it is demonstrated that the Pt-coordinated phosphate group can be protonated (pK a/1=3.21±0.10 and 3.31±0.05, respectively), and 1H- and 31P-NMR spectra also indicate deprotonation (pK a/2=13.35±0.25) of the exocyclic amino group of the cytosine moiety. The metal ion binding affinity of cis-Pt(NH3)2(dCMP) is very small, as shown for Cu2+ (log K<0.6). The cis-Pt(NH3)2(dCMP) complex reacts with nucleosides and nucleotides (L′) by losing its chelate structure and forming mixed ligand complexes, cis-Pt(NH3)2(dCMP)(L′); this means that the phosphate group is released from the coordination sphere of Pt(II), indicating that the Pt(II)-O(phosphate) bond is not very strong. The synthetic catechol, U-0521, (3′,4′-dihydroxy-2-methylpropiophenone) is a competitive inhibitor of both tyrosine hydroxylase and catechol-O-methyltransferase. Continuous subcutaneous administration of 10 μmoles per day of U-0521 via Alzet osmotic minipumps to adult male Spontaneously Hypertensive Rats (SHR) reduced blood pressure from 160 mmHg to 125 mmHg. This effect occurred within two days, persisted for the two weeks that the pumps were in place, and reversed gradually upon cessation of U-0521 administration. Similar treatment of U-0521 to normotensive Wistar Kyoto rats (WKY) did not result in a similar hypotentensive effect. Subcutaneous administration of the same dose to juvenile SHRs led to a blockade in the expression of hypertension. After the five week treatment period, the blood pressure of the U-0521 treated animals escalated rapidly to match the saline treated controls. The antihypertensive effect of U-0521 on SHRs also occured when the compound was delivered by the oral route at the rate of 50 mg/kg/day.  相似文献

Abstract

The degradation of diamino analogues of 2′- or 3′-deoxyadenosine and of 9-(2-deoxy-β-D-threo?pentofuranosyl)adenine or 9-(3-deoxy-β-D-threo-pentofuranosyl)adenine in buffers of acid, neutral and alkaline pH and constant ionic strength was followed by liquid chromatography. The rate of hydrolysis at acid pH was found to be related to the position and configuration of the amino group on the sugar moiety. The compounds under study were found to be more stable than corresponding monoaminated nucleosides, which have been reported to be more stable than the hydroxyl nucleosides. Liquid chromatographic analyses indicate that acid hydrolysis involves cleavage of the N-glycosyl bond as the major degradative process, together with another minor process. pH-rate profiles, activation parameters and deuterium isotope solvent effects are discussed.  相似文献

The design of a new clinical candidate histamine-H(3) receptor antagonist for the potential treatment of excessive daytime sleepiness (EDS) is described. Phenethyl-R-2-methylpyrrolidine containing biphenylsulfonamide compounds were modified by replacement of the sulfonamide linkage with a sulfone. One compound from this series, 2j (APD916) increased wakefulness in rodents as measured by polysomnography with a duration of effect consistent with its pharmacokinetic properties. The identification of a suitable salt form of 2j allowed it to be selected for further development.  相似文献

Synthesis of 1-(2, 3, 4-tri-0-acetyl-α-L-rhamnopyranosyl) uracil (3), 1-(α-L-rhamnopyranosyl) uracil (4), 1-(2, 3-0-isopropylidene-α-L-rhamnosyl) uracil (5), and 1-(2, 3-0-isopropylidene-4-keto-α-L-rhamnopyranosyl) uracil (6) are reported. 2-Arylquinazolin-4(3H)-ones 125 were synthesized by reacting anthranilamide with various benzaldehydes using CuCl2·2H2O as a catalyst in ethanol under reflux. Synthetic 2-arylquinazolin-4(3H)-ones 125 were evaluated for their β-glucuronidase inhibitory potential. A trend of inhibition IC50 against the enzyme in the range of 0.6–198.2 μM, was observed and compared with the standard d-saccharic acid 1,4-lactone (IC50 = 45.75 ± 2.16 μM). Compounds 13, 19, 4, 12, 14, 22, 23, 25, 15, 8, 17, 11, 21, 1, 3, 18, 9, 2, and 24 with the IC50 values within the range of 0.6–44.0 μM, indicated that the compounds have superior activity than the standard. The compounds showed no cytotoxic effects against PC-3 cells. A structure–activity relationship is established.  相似文献

Terminal alkynes (R–CC–H, R = 1-naphthyl, 9-anthryl, 4-Me2N–C6H4–, or the longer analogue, 4-(4-Me2N–C6H4–CC–)–C6H4–) react with [Rh(PMe3)4Me] at ambient temperature, with loss of methane and one PMe3 ligand, to form the corresponding mer,trans-[(PMe3)3Rh(CCR)2H] compounds in excellent yield. In this preliminary study, the synthesis and spectroscopic characterization of the four new compounds are reported, along with the single-crystal structure of the R = 4-Me2N–C6H4 derivative.  相似文献

The hydrolysis of α-chloro-N-methyl-4-pyridone was found to be more than five times faster than that of α-chloro-N-methyl-2-pyridone. Structural studies of 2- and 4-pyridones have revealed the higher polarity and greater extent of zwitterionic content in 4-pyridone. The results are thus consistent with the hypothesis that polarization and higher zwitterionic content in the heterocyclic structures enhances the rate of hydrolysis in α-substituted pyridone and uracil derivatives.  相似文献
