The seed cotyledons of a euhalophyte, Suaeda physophora, were found to be dark green. The pigment extracted from the cotyledons was proved to be chlorophyll for the absorption spectra curve of extracts the same as that for leaves. Photosynthetic oxygen-exchanging could be detected after the seeds were moistened for 6 h. Microstructure of organelles in cotyledons of ungerminated seeds was detected by transmission electron microscopy. Histochemical pigmentation was used to investigate the degree of damage on the membrane of radicles. A xerophyte, Haloxylon persicum, whose cotyledons of desiccated seeds also contain chlorophyll was used as a comparative species. The results showed that S. physophora maintained the ultra-structure of chloroplasts, the integrity of plasma membranes of radicles kept much better than that of H. persicum, which showed the great adaptability to salinity of the euhalophyte even at the seed-germination stage. Seeds were incubated in 0 and 700 mmol/L NaCl for 10 days in darkness at 20 ℃ , then ungerminated seeds in NaCl solution were transferred to deionised water and reincubated for another 8 days and was recorded the germination recovery. Dry seeds moistened with deionised water germinated gradually in the dark and germination was maximal after 3 or 4 d. Seeds at 700 mmol/L NaCl for 10 d and transferred back to deionised water germinated abruptly, most seeds germinated in one day and photosynthesis was also detected. It is concluded that recovery germination of S. physophora ensured the seeds could germinate rapidly after salinity declines, for example under the mild but transitory favorable edaphic condition in early spring. Photosynthesis function in seeds might also promoted radicle growth and seedling establishment.  相似文献

BACKGROUND AND AIMS: Germination is very important for plant establishment in arid regions. The strategies taken by halophytes during the seed germination stage to adapt to saline environments in an arid zone were investigated in Suaeda physophora (euhalophyte), Haloxylon ammodendron (xero-halophyte) and Haloxylon persicum (xerophyte). METHODS: Seeds of S. physophora, H. ammodendron and H. persicum were exposed to a range of iso-osmotic NaCl and PEG solutions. Seed germination in, and recovery germination from, high NaCl were recorded. The effects of iso-osmotic NaCl and PEG on seed water uptake and changes in ion content were measured. In addition, the structure of seeds and Na+ distribution in the seed coat and embryos of dry seeds were investigated. KEY RESULTS: The relative increase in fresh weight of germinating seeds was markedly reduced in -2.24 MPa PEG compared with that in -2.24 MPa NaCl, while the opposite trend was found in concentration of K+ during the initial 9 h for all species. Haloxylon ammodendron and S. physophora had a higher recovery germination from -3.13 MPa NaCl compared with H. persicum. Seeds of all species had no endosperm. More Na+ was compartmentalized in the seed coats of the two halophytic species compared with that in the xerophyte H. persicum. CONCLUSIONS: The effect of NaCl on seed germination was due to both osmotic stress and ion toxicity for the three species. High soil salinity and a high content of Na+ in seeds may induce more seeds to remain ungerminated in S. physophora and H. ammodendron. Morphological structure and adaptation to salinity during seed germination may determine the geographical distribution of H. ammodendron and S. physophora in certain saline regions.  相似文献

Aims We investigated the impact of salinity on seed germination, chlorophyll content, chloroplast structure and photosynthesis of the green embryos in desiccated seeds of the xerophyte Haloxylon persicum, xero-halophyte Haloxylon ammodendron and euhalophyte Suaeda physophora.Methods Seeds of H. persicum, H. ammodendron and S. physophora were collected from natural environment in Fukang, Xinjiang province. Pretreatment with 700 mM NaCl was carried out to stimulate the natural 'seed priming'; we analyzed the joint effect of salinity and different species on germination physiology and cotyledonal structure and photosynthetic function changes during germination and recovery stage.Important findings We found that seeds did not suffer ion toxicity for the two halophytes H. ammodendron and S. physophora, as evidenced by the high final germination after ungerminated seeds pretreated with 700 mM NaCl were transferred to distilled water, but the final germination of the xerophyte H. persicum was significantly lower than that of control. The Na + concentration in embryos increased under salinity for all species, while K + concentration decreased by salinity only for H. persicum and H. ammodendron, i.e. the concentration of K + in embryos of H. persicum and H. ammodendron decreased by 36% and 46%, respectively. For all species, whether dry intact seeds or cotyledons of dry seeds imbibed in deionized water and NaCl solution, had high chlorophyll content. Treatment with NaCl also caused chloroplast thylakoids to swell and chlorophyll content to decrease in seeds of H. persicum, but no significant change was observed in the more salt-tolerant species S. physophora and H. ammodendron. Fluorescence measurement showed that 700 mM NaCl decreased the Fv/Fm ratio of cotyledons in seeds for all species, especially for H. persicum and H. ammodendron. Photosynthetic oxygen releasing was detected from the seeds that were moistened with distilled water and 700 mM NaCl for 6 or 24 h and from the seeds that were initially moistened with 700 mM NaCl in darkness for 10 days, then transferred to distilled water for another 6 and 24 h. The results indicated that the chlorophyll in cotyledon of desiccated seed had photosynthetic function in early germination stage, even under high-saline condition. In addition, the photosynthesis of chlorophyll in the embryonic cotyledons of desiccated seeds during germination was similar to that in leaves of young seedlings for all species. In conclusion, the chloroplasts of the two halophytes were more salt resistant compared with the xerophyte H. persicum. The photosynthetic function of chlorophyll in cotyledons of mature seeds may be ecologically important for seedling development in early stage for plants growing in extremely saline or arid environments.  相似文献

The stem succulent perennial halophyteHaloxylon recurvum Bungeex. Boiss. grows and produces seed under highly saline conditionsand seeds are deposited in saline soils. Experiments were conductedto determine the effect of salinity and temperature on the germinationof seeds. Results indicate that seeds can germinate at veryhigh salt concentrations (500 mM). However, highest germinationpercentages were obtained in distilled water. Cooler thermoperiodspromoted germination, while high temperatures significantlyinhibited the germination of seeds at all NaCl concentrationstested. Rate of germination decreased with increases in salinity.At higher thermoperiods the rate of germination was significantlylower in comparison to lower thermoperiods. Salinity and nitrogen are two important environmental factors that affect the distribution of halophytes in their natural saline habitats. Seeds of the euhalophyte Suaeda salsa L. were harvested from plants that had been treated with 1 or 500 mm NaCl combined with 0.5 or 5 mm NO3?‐N (nitrate) for 115 days in a glasshouse. Germination was evaluated under different concentrations of NaCl and nitrate. Plants exposed to high salinity (500 mm ) and low nitrate (0.5 mm ) tended to produce heavy seeds. Either high salinity (500 mm ) or high nitrate (5 mm ) increased the brown/black seed ratio. The concentrations of Na+, K+, and Cl? were higher in brown than in black seeds, and NO3? concentrations were higher in black than in brown seeds, regardless of NaCl and nitrate treatments during plant culture. Regardless of NaCl and nitrate concentrations during germination, seeds from plants grown with 0.5 mm nitrate generally germinated more rapidly than seeds from plants grown with 5 mm nitrate, and the difference was greater for black than for brown seeds. Exogenous nitrate during germination enhanced the germination of brown seeds less than that of black seeds. Producing more brown seeds and heavy black or brown seeds appears to be an adaptation of S. suaeda to saline environments. Producing more black seeds, which tend to remain dormant, should reduce competition for nitrogen and appears to be an adaptation to nitrogen‐limited environments. In conclusion, nitrate provided exogenously or by mother plants to black seeds may act as a signal molecule that enhances the germination of black S. suaeda seeds.  相似文献

Seeds and seedlings of the halophyte Atriplex patula were exposed to 0–2% NaCl to determine the effect of salt stress on germination and growth. Seeds germinated and plants survived and grew in solutions of up to 2.0% NaCl. Both seed germination and dry mass production were negatively affected by increased salinity. Dry mass production declined to 1% of controls and seed germination to 17% of controls in the 2% NaCl treatments, indicating that seeds were less inhibited than growing plants. Also, recovery treatments indicated that high salinity did not permanently injure seeds. Percent ash, and Na+ and Cl ions increased in shoots with each salt increment, while the K+ ion content decreased sharply. Atriplex patula is a facultative halophyte, and is limited to low and moderately saline sites because both seed germination and growth are severely reduced at salinities > 1% NaCl.  相似文献

Recovery of seed germination from NaCl salinity of desert shrubs (Haloxylon recurvum and Suaeda fruticosa, and the herbs Zygophyllum simplex and Triglochin maritima was studied under various thermoperiods. The percentage of ungerminated seeds that recovered when they were transferred to distilled water varied significantly with variation in species and thermoperiods. Zygophyllum simplex had little recovery from all NaCl concentrations in all thermoperiods. Haloxylon recurvum, S. fruticosa, and T. maritima showed substantial recovery. Percentage recovery was highest in S. fruticosa, followed by T. maritima, and H. recurvum. Thermoperiodic effects varied with the species investigated. There was little thermoperodic effect on the percentage recovery of S. fruticosa, except in the higher salinity treatment at higher thermoperiods. Variation in thermoperiod appears to play an important role in recovery of germination of halophytes from salt stress when seeds are transferred to distilled water.  相似文献

Haloxylon ammodendron and Haloxylon persicum (as sister taxa) are dominant shrubs in the Gurbantunggut Desert. The former grows in inter-dune lowlands while the latter in sand dunes. However, little information is available regarding the possible role of soil microorganisms in the habitat heterogeneity in the two Haloxylon species from a nutrient perspective. Rhizosphere is the interface of plant–microbe–soil interactions and fertile islands usually occur around the roots of desert shrubs. Given this, we applied quantitative real-time PCR combined with MiSeq amplicon sequencing to compare their rhizosphere effects on microbial abundance and community structures at three soil depths (0–20, 20–40, and 40–60 cm). The rhizosphere effects on microbial activity (respiration) and soil properties had also been estimated. The rhizospheres of both shrubs exerted significant positive effects on microbial activity and abundance (e.g., eukarya, bacteria, and nitrogen-fixing microbes). The rhizosphere effect of H. ammodendron on microbial activity and abundance of bacteria and nitrogen-fixing microbes was greater than that of H. persicum. However, the fertile island effect of H. ammodendron was weaker than that of H. persicum. Moreover, there existed distinct differences in microbial community structure between the two rhizosphere soils. Soil available nitrogen, especially nitrate nitrogen, was shown to be a driver of microbial community differentiation among rhizosphere and non-rhizosphere soils in the desert. In general, the rhizosphere of H. ammodendron recruited more copiotrophs (e.g., Firmicutes, Bacteroidetes, and Proteobacteria), nitrogen-fixing microbes and ammonia-oxidizing bacteria, and with stronger microbial activities. This helps it maintain a competitive advantage in relatively nutrient-rich lowlands. Haloxylon persicum relied more on fungi, actinomycetes, archaea (including ammonia-oxidizing archaea), and eukarya, with higher nutrient use efficiency, which help it adapt to the harsher dune crests. This study provides insights into the microbial mechanisms of habitat heterogeneity in two Haloxylon species in the poor desert soil.  相似文献

The non-native, invasive haplotype of Phragmites australis is rapidly invading tidal and non-tidal wetlands across North America. Phragmites has the potential to spread by seeds and rhizomes. Seed viability and dormancy differences were quantified among 18 patches of non-native Phragmites in subestuarine wetlands in developed (i.e., suburbanized) vs. forested watersheds of the Chesapeake Bay. We used tetrazolium and germination assays to assess seed viability and compared germination percentages and rate of germination among fresh seeds, cold–moist treated seeds, and warm–dry treated seeds to evaluate seed dormancy. Seed viability was <1% in most patches but a few patches produced abundant viable seeds (5–21%). Seed viability, however, did not differ significantly between wetlands in forested vs. developed watersheds. Contrary to studies of Phragmites seed dormancy in European populations, some Phragmites seeds were dormant at maturity; cold–moist treated seeds germinated faster and to higher percentages than fresh seeds or warm–dry treated seeds.  相似文献

梭梭是北方荒漠区重要的固沙植物,具有较强的荒漠环境适应能力。研究梭梭群落能量及蒸散变化对科学管理水资源、保护与建设生态植被等有重大意义。基于波文比自动观测系统在古尔班通古特沙漠南缘丘间地实时连续测定的2018年气象和能量数据,运用波文比-能量平衡法对梭梭群落能量平衡和蒸散特征进行了分析。结果表明:梭梭生长季(4—10月)太阳净辐射和土壤热通量的日变化呈单峰状分布,净辐射通量最高值出现在13:30左右,土壤热通量最大值出现时间比净辐射推迟2 h,波文比在梭梭生长旺盛期全天变化稳定,萌发期和枯落期白天变化较小,夜间波动较大,湍流通量以显热通量为主;在晴天和阴雨天条件下,梭梭生长旺盛期的日蒸散量均明显大于萌发期和枯落期,且日蒸散量在旺盛期呈双峰型,萌发期和枯落期呈单峰型;2018年梭梭生长季蒸散量为279.67 mm,日均蒸散强度1.31 mm/d,梭梭群落的蒸散受到气象、自身生长特性以及土壤水分的影响,4月蒸散强度开始逐渐增大,由于多阴雨天气导致6月的蒸散强度有所降低,7月达到最大,9—10月水分、温度等条件缺乏,梭梭生理活动微弱,蒸散强度急剧减小。  相似文献

In order to determine how salinity and exposure time affect seed viability and germination, seeds of five halophytes, Atriplex prostrata, Hordeum jubatum, Salicornia europaea, Spergularia marina, and Suaeda calceoliformis were exposed to 3.0, 5.0, and 10.0% NaCl solutions for 30, 60, 90, 365, and 730 d. Recovery experiments in distilled water indicated significantly different species responses to salinity over time. Percentage germination and rate of germination in H. jubatum were dramatically reduced following extended exposure and all seeds exposed to 10% NaCl for > 1 yr failed to germinate. Spergularia marina seeds were stimulated following short-term exposure to 3% NaCl; however, germination was delayed and overall germination was significantly reduced with exposure time in the two higher salinity levels. Percentage germination in A. prostrata decreased over time, but salinity level was not related to this reduction. Germination of S. europaea and S. calceoliformis, the most salt-tolerant species being tested, was stimulated by exposure to high salinity. Both species had a significant increase in percentage germination and in the germination rate when compared to seeds germinated in distilled water. Baseline germination data from seeds placed in 0, 1, 2, and 3% NaCl solutions indicated that S. europaea and S. calceoliformis were the only species to germinate in the 3% NaCl solution. Spergularia marina failed to germinate in the 2% NaCl treatment, and germination of A. prostrata and H. jubatum was significantly reduced at this salinity level. It is concluded that prolonged exposure to saline solutions can inhibit or stimulate germination in certain species, and the resulting germination and recovery responses are related to the duration and intensity of their exposure to salt in their natural habitats.  相似文献

Six perennial species endemic to South West Western Australia (Acacia trulliformis, Austrostipa geoffreyi, Banksia oligantha, B. mucronulata, Hakea tuberculata and Orthrosanthus muelleri) were screened for salt tolerance and recovery during seed germination. Growth and survival of 6-month old seedlings of these six plus a further vegetatively propagated species (Myoporum turbinatum) were subsequently examined in response to salt and waterlogging application. Water uptake under elevated saline conditions (200 and 400 mM NaCl) was slow, but not restrictive to germination. Moreover, a large proportion of seeds that were unable to germinate under saline conditions recovered after being transferred to non-saline conditions. Germination, growth and survival varied with species and the salt concentration used. Increasing salt concentrations tended to increase time to germination. Germination of Acacia trulliformis seeds declined exponentially with increasing salinity, and seedlings suffered reduced growth under saline and non-saline waterlogging. Austrostipa geoffreyi seeds were sensitive to saline treatments but seedlings were highly tolerant of both saline and/or waterlogged conditions. Germination of the three proteaceous species declined significantly under highly saline conditions (400 mM NaCl) with seedlings of the two Banksia species not surviving any treatment with the exception of non-saline waterlogging. Seedlings of H. tuberculata were more resilient to treatment conditions. Orthrosanthus muelleri was sensitive to salt stress during germination but was highly resistant to waterlogging, both saline and non-saline. This study provides an insight into the response and resilience of components of the vegetation understorey of saline-affected regions of Western Australia not usually evaluated allowing for more informed restoration.  相似文献

Salt tolerance of halophytes corresponds with the habitat requirement of the species. It is an important factor during the germination phase and it can determine successful establishment. This paper presents the effects of alternating temperature–light regimes (4/8°C, 10/20°C, 20/32°C; 12 h dark: 12 h light) and different salinity levels (0, 200, 400, 600 mmol l21 NaCl) on seed germination of five halophytes, Halimione pedunculata, Bupleurum tenuissimum, Aster tripolium, Triglochin maritimum and Armeria maritima. The five species differ with respect to family and life‐form and spatially correspond to a decreasing salt gradient (i.e. distance from salt water, with H. pedunculata being the most tolerant and A. maritima being the least). Armeria maritima, A. tripolium and T. maritimum seeds were additionally subjected to a cold stratification experiment. The results showed that Halimione pedunculata, an annual therophyte of year‐round heavily saline habitats, was dormant under all experimental conditions. Bupleurum tenuissimum, a species typical to sites of varying salinity prone to leaching during spring and autumn rainfall, germinated best under cold and warm temperatures, but only under non‐saline conditions. Aster tripolium and T. maritimum, close neighbours in salt marshes, showed very similar germination behaviour: seeds of both species tolerated high levels of salinity and germinated best in summer temperatures during periods of highest soil salinity, and germination was significantly promoted by cold. Armeria maritima, a species usually found on the marginal fringes of saline habitats, germinated only under low salt levels and maximum germination was under cold (spring) and warm (autumn) temperatures, with no significant effect of cold stratification.  相似文献

为了研究梭梭种子散布规律,通过布设种子雨收集器结合室内实验分析,对准噶尔盆地东南缘梭梭种子雨特征进行了研究。研究显示:(1)梭梭种子雨的累积密度达到平均189粒/m2,其中有活力种子占约80%;(2)种子散布的高峰集中在11月初到11月15日时间段,其落种量占整个种子雨的65%。其后种子雨密度随时间逐渐减小;(3)整个种子雨过程,不同时期散落的种子雨质量存在差异,表现为不同时期散落种子的萌发率呈现出先增大后减小的趋势;(4)变异函数分析表明,梭梭种子雨在8.12 m的有效变程内,种子雨具有明显的空间格局,其由空间自相关和随机因素引起的空间异质性各占50.0%。准噶尔盆地东南缘梭梭种子雨密度大且质量较高,同时其时空分布异质性较高,这些特征均将影响梭梭种群的分布格局和种群更新。  相似文献

Kosteletzkya virginica (L.) Presl. (Malvaceae) is a perennial that grows in saline or brackish water, and is salt-tolerant in its mature state, but less tolerant during germination. The seeds show a very low permeability to water that increases during storage. The permeability to water differs in seeds harvested in different years. Optimal temperature for germination is 28–30 C. The effect of salinity on imbibition is largely osmotic, but germination is inhibited, apparently, by the combined osmotic and “ionic” effects, especially at high NaCl concentrations. Inhibition of germination by high NaCl concentrations is relatively more severe in scarified than in intact seeds, indicating that the seed coat acts as a partial barrier to Na+ influx. External application of proline or betaine did not improve germination under saline conditions. Dry seeds contain a significant amount of betaine and low levels of proline, but during germination and in the presence of NaCl the betaine content decreased while the proline content increased. Thus, the likely compatible solute in the germinating seed seems to be proline.  相似文献

Restoration of submerged aquatic vegetation from seed has been hampered by a lack of information on the appropriate conditions for collecting, processing, and storing seeds prior to dispersal. Seeds must be processed and stored under conditions that maintain seed viability, meet dormancy requirements, and prevent premature germination. This study examined the effects of collection date, processing technique, aeration, storage and induction temperature and salinity, and storage period on seed germination of two mesohaline aquatic species, Potamogeton perfoliatus and Ruppia maritima. Collection date and processing technique were significant factors affecting seed yield from donor populations. Seeds of both species remained viable and germinated best when stored at 4°C, and then exposed to freshwater induction conditions. However, their responses to other factors differed. Aeration during storage was necessary in order to maintain viability of P. perfoliatus seeds, whereas it was unnecessary for R. maritima seeds. Storage in freshwater at 4°C prevented germination of P. perfoliatus seeds, while high salinity during cold storage was necessary to minimize premature germination of R. maritima. Mean germination time of P. perfoliatus was dependent on storage salinity; in contrast, mean germination time of R. maritima seeds was dependent on induction salinity. These differences indicate that the methods required to produce large quantities of underwater plant seed amenable to large‐scale restoration efforts must be tailored to the specific requirements of individual species and must consider the range of processes from initial harvest through seed testing prior to field establishment.  相似文献

Abstract

Seed priming is a simple biotechnological tool which is potentially able to promote seed germination and invigoration as well as seedlings establishment and stress tolerance. In this study, the effects of seed pre-treatment with water (hydro-priming), 100 (HP-NaCl100) and 200?mM (HP-NaCl200) NaCl (halo-priming) for 6?h on some physiological and biochemical parameters of fenugreek was investigated under saline conditions (100?mM NaCl). For the three priming treatments, no significant changes in the final germination percentage were observed. However, a decrease in seed germination time was observed in hydro- and halo-primed (HP-NaCl200) seeds. Salt stress (100?mM NaCl) reduced growth (shoot and root dry weight), pigment content, disturbed the ionic balance and enhanced malondialdehyde content. Salinity-induced changes in lipid metabolism towards synthesis/accumulation of saturated and monounsaturated fatty acids were observed in stressed plants. Seed priming ameliorated the negative effects of NaCl, ensuring significant amelioration on growth, pigment content, increased the activity of catalase and glutathione peroxidase and enhanced the synthesis of very long chain n-alkanes. Taken together, these data provide compelling evidence that priming is an effective alternative that can be used to promote germination and improves establishment and acclimation of fenugreek seedlings under saline conditions.  相似文献

生物结皮广泛分布于干旱、半干旱区, 强烈影响着土壤表层理化特性, 进而对种子散布、萌发和定居产生影响。目前关于生物结皮与植物种子萌发关系的研究结论存在争议。该文通过室内人工控制实验, 研究了生物结皮对古尔班通古特沙漠5种具不同种子形态特征的荒漠植物白梭梭(Haloxylon persicum)、蛇麻黄(Ephedra distachya)、角果藜(Ceratocarpus arenaarius)、涩芥(Malcolmia africana)和狭果鹤虱(Lappula semiglabra)的种子萌发的影响。结果表明, 在干燥和湿润两种条件下, 生物结皮对不同形态植物种子萌发均具有不同的作用。在干燥条件下, 生物结皮显著抑制了角果藜和涩芥种子的萌发(p<0.05), 对其它3种植物无显著影响; 而湿润条件下, 生物结皮显著抑制了白梭梭、角果藜和狭果鹤虱种子的萌发(p<0.05), 对蛇麻黄、涩芥则无显著影响。  相似文献

The halophyte, Salicornia pacifica var. utahensis (Tiderstorm) Munz produces seed under high salinity conditions, and deposits its seed on saline soil. Experiments were conducted to determine the effect of salinity, temperature and growth regulators on germination. Results indicate that the seeds can germinate at very high salt concentration (5% NaCl). Germination was sensitive to the changes in temperature regimes. At higher 30–20 C, light-dark sequence, no germination occurred at 3, 4 and 5% NaCl treatments. On the other hand, 30% germination did occur at 5% NaCl treatment at a temperature regime of 15–5 C. These seeds required light for germination. Only 50% germination occurred in the non-saline control in the dark and the addition of NaCl further reduced germination. The GA3 partially alleviated the inhibitory effect of NaCl and darkness. Kinetin did not promote germination.  相似文献
