Identification of interspecific hybrids is often a subject of primary concern in the development of conservation strategies. Here we performed a genetic analysis combining mitochondrial DNA (mtDNA), microsatellites and single nucleotide polymorphic sites (SNPs) to assay the level of hybridization and introgression between an introduced babbler, Chinese hwamei Leucodioptron canorus, and its close relative, the endemic Taiwan hwamei L. taewanus in Taiwan. Fifty‐five Chinese hwameis from the Asian mainland and 69 Taiwan hwameis, including nine morphological hybrids, were sampled and analyzed. Evidences of mitochondrial introgression were found in three hybrids and one Taiwan hwamei. Five unlinked interspecific SNPs were identified at nine anonymous nuclear loci with interspecific differentiation (total Fst=0.77) that was much higher than that at seven highly polymorphic microsatellite loci combined (total Fst=0.1). Bayesian cluster analysis based on five interspecific SNP loci and two highly differentiated microsatellite loci (Fst>0.08) suggested that twelve individuals sampled in Taiwan were likely F2 or backcross hybrids, among which eight were morphological intermediates. A total of 20.3% (14/69) individuals sampled in Taiwan were suggested to be hybrids, suggesting that fitness reduction in hybrids might be negligible. These results imply that without an effective management strategy, the entire Taiwan hwamei population could easily become an admixed with Chinese hwamei and loose its evolutionary integrity. To reduce introgressive hybridization, illegal trade of Chinese hwamei should be strictly regulated and only the expensive male Chinese hwameis should be legally imported to minimize the chance for Chinese hwameis being released into the field. In our study we also found interspecific SNP markers to outperform microsatellite loci in detecting hybridization and introgression between two closely related species, which may be ascribed to the lower level of homoplasy of SNP loci.  相似文献

Analysis of mitochondrial 16S rRNA sequences of four speciments from two lizard species (Leiolepis guentherpetersi and L. reevesii) showed identity of 91.1–91.6% and the genetic distances were 8.8–9.3%. The two speciments (C5 and C7) of L. reevesii species have the homology of 96.5–99.4% with L. belliana and L. reevesii, respectively. Whereas, those of L. guentherpetersi species (S4 and S6) have higher homology of 99.6–100% with L. guttata and L. guentherpetersi, respectively. These mitochondrial 16S rRNA sequences of individuals from L. guentherpetersi (S4 and S6) and L. reevesii (C5 and C7) were deposited in GenBank with accession number EU428186, EU428187, EU428188, and EU428189, respectively.  相似文献

Miscanthusgiganteus, a triploid hybrid between tetraploid M. sacchariflorus and diploid M. sinensis, has considerable potential as a bioenergy crop. Currently only one genotype is widely cultivated, increasing its vulnerability to diseases during production. Finding new hybrids is important to broaden genetic resources of M. ×giganteus. Three putative triploid hybrids were discovered in a sympatric population of tetraploid M. sacchariflorus and diploid M. sinensis in Kushima, Japan. The hybrid nature of the triploids was determined by morphological analysis and sequencing the ribosomal DNA internal transcribed spacer (ITS) region. The triploids had awns on their florets, which is a common characteristic of diploid M. sinensis, and sheath hairs, which is typical of tetraploid M. sacchariflorus. All triploids showed heterozygosity in their ribosomal DNA ITS sequences. Based on these results, it is confirmed that the triploids are hybrids and novel genotypes of M. ×giganteus. Natural crossing between tetraploid M. sacchariflorus × diploid M. sinensis may also lead to the production of tetraploid hybrids. ITS analysis of tetraploid plants showed that one maternal parent of the triploid hybrids, K-Ogi-1, had heterozygous ITS, which was different than the other analyzed tetraploid, M. sacchariflorus. Thus, K-Ogi-1 was likely of hybrid origin. These tetraploid hybrids can also be utilized as parents in M. ×giganteus breeding. Since all hybrids identified in this study had tetraploid M. sacchariflorus as maternal parents, collecting and analyzing seeds from tetraploid M. sacchariflorus in sympatric areas could be an effective strategy to identify natural Miscanthus hybrids that can be used as bioenergy crops.  相似文献

Little has been known about the impacts of past vicariance events on the phylogeography and population structure of freshwater fishes in East Asia. The aims of this study are to assess the genetic variability with extensive sampling throughout the range of Chinese spiny loach, Cobitis sinensis, and to infer the genetic structure and evolutionary history of populations. Cobitis sinensis in China may have initiated from two ancestral populations, namely Yangtze and Pearl Rivers, which diverged about 7.24 MYA likely due to drainage systems alteration. In the phylogroup I, a southward dispersal event occurred from East China (Yangtze River) to south ZheMin and Hainan subregions, followed by eastward dispersal from ZheMin to south Taiwan. In the phylogroup II, eastward colonization took place from Pearl River to north Taiwan in the late Pliocene, coupled with loss of genetic diversity in the island populations. This study showed that Cenozoic tectonic movements and climatic and sea‐level fluctuations may have shaped the genetic structure of C. sinensis in concert. Highly diverged mtDNA sequences suggest existence of cryptic species in morphospecies C. sinensis.  相似文献

A field experiment with 15 Miscanthus genotypes including M. × giganteus, M. sacchariflorus, M. sinensis and M. sinensis hybrids was conducted for 14 years at the experimental Ihinger Hof station of the University of Hohenheim in southwest Germany to evaluate interannual yield performance stability over 14 years of harvests of the different genotypes. In this article, a simple formula is presented which could be used to forecast late winter yields using morphological traits in autumn. The data obtained indicated a shorter establishment period to reach a yield plateau of M. × giganteus and M. sacchariflorus than M. sinensis hybrids and M. sinensis genotypes. The best performing genotype was M. × giganteus (Gig‐2, No. 16.21) with a mean spring harvestable yield of 14.1 t DM ha?1 year?1. A correlation analysis with climatic parameters revealed precipitation during the growing period as the key factor for high yields at this site. Likewise, but to a lesser degree, heat sum during the growing period was positively correlated with yields. It could be shown that precipitation/snow during the winter correlated with yield losses, until the harvest date in February/March. Phenological measurements indicated that a high yield potential of the tested genotypes is associated with either an absence of flowering or late flowering. Also, height of the plants and shoot diameter were indicators for high yield potential. Shoot density and plant height at senescence were found to be solid parameters to estimate harvestable biomass in late winter. Yield approximations with a mean accuracy of 80.9% for M.× giganteus genotypes were obtained using the newly developed equation. Yields of M. sinensis hybrid (Sin‐H7) were projected most accurately with this simple formula, resulting in a mean accuracy of 84.5%.  相似文献

Information on genome size, ploidy level, and genomic polymorphisms among accessions of the genus Miscanthus can assist in taxonomic studies, help understand the evolution of the genus, and provide valuable information to biomass crop improvement programs. Taxonomic investigation combining variation in plant morphology, genome size, chromosome numbers, and simple sequence repeat (SSR) marker polymorphisms were applied to characterize 101 Miscanthus accessions. A total of 258 amplicons generated from 17 informative SSR primer pairs was subjected to cluster and principal coordinate analysis and used to characterize genetic variation and relationships among 31 Miscanthus accessions, including four interspecific Miscanthus hybrids created from controlled pollinations, and four Saccharum, six Erianthus, and one Sorghum bicolor accessions. Miscanthus accessions were distinct from accessions in the genera Erianthus and Saccharum. Miscanthus accessions fell into five taxonomic groups, including the existing taxonomic section Miscanthus, diploid and tetraploid Miscanthus sacchariflorus, and a fourth (M. × giganteus) and fifth group (Miscanthus ‘purpurascens’); the last two being intermediate forms. In contrast to previous work, our findings suggest diploid and tetraploid M. sacchariflorus are taxonomically different, the latter more closely related to M. sacchariflorus var lutarioriparius. We also suggest that Miscanthus ‘purpurascens’ accessions are interspecific hybrids between Miscanthus sinensis and diploid M. sacchariflorus based on DNA content and SSR polymorphisms. The evolution of Miscanthus and related genera is discussed based on combined analysis and geographical origin.  相似文献

Abstract

In this study, leaf morphology was assessed in a mixed oak stand (western France) using two geometric morphometric (landmark and outline) datasets and one dataset of 19 leaf measures. Adult oaks (817 oaks), comprising four white oak species (Quercus petraea, Q. robur, Q. pubescens and Q. pyrenaica), were sampled for DNA extraction and genetic analysis (nuclear microsatellites). Leaf morphology was assessed on 336 oaks, comprising pure species and hybrids as determined by genetic assignment. This comparative study of oak leaf morphology, based on the use of two free size geometric morphometric methods and a set of leaf measurements, combined with the genetic assignment of individuals to pure species or hybrids, provided information about the differences among species and the intermediate leaf morphology of their hybrids.  相似文献

为了解五桂山的土沉香(Aquilaria sinensis)种群空间遗传结构,利用18个微卫星位点对143株胸高直径大于5 cm的土沉香个体基因型进行了检测。结果表明,五桂山土沉香种群的观测杂合度和期望杂合度均为0.534;近交系数为0.000 1,说明五桂山土沉香遗传多样性并不低,且种群处于随机交配状况。空间遗传结构分析结果表明,五桂山土沉香缺乏空间遗传结构,不同遗传背景的个体相互混杂,导致相邻个体遗传差异较大。这可能是五桂山土沉香种群发育过程中自疏所致,也可能是这一地区土沉香为人工种植,而非自然发育形成而造成的。  相似文献

Miscanthus is a C4 perennial grass originating from East Asia, the yields of which progressively increase in the first years of growth. Several species for bioenergy have been studied since the mid‐1980s in Europe, in particular (Miscanthus × giganteus [M. × giganteus]), due to its high yields. M. × giganteus is mainly cultivated in France and established from rhizomes. Our study aimed to assess, in field conditions, alternative establishment methods combined with an alternative species, Miscanthus sinensis (M. sinensis). We set up a multi‐environment experimental network. On each trial, we tested two treatments with M. × giganteus, established from rhizomes (G_r‐sd) and from plantlets obtained from rhizomes (G_p‐sd), and two treatments with M. sinensis seedlings transplanted in single (S_p‐sd) and double density (S_p‐dd). ANOVA was performed to compare establishment and regrowth rates across treatments, as well as yields across treatments and site‐years. A logistic model was used to describe yield trends and to compare the maximum yield reached and the rate of yield increase of both species. Results showed that miscanthus establishment from plantlets resulted in higher establishment (between 87% and 92%) and regrowth (between 91% and 94%) rates compared to establishment from rhizomes. Treatments with M. × giganteus obtained higher average yields across site‐years than those with M. sinensis, but more variable yields across site‐years. We showed a strong species effect on yields, yield components (shoot weight, shoot density and shoot number per plant) and light interception (through leaf area index). Lastly, to use M. sinensis established from transplanted plantlets as an alternative to M. × giganteus, research would be required on the breeding of M. sinensis sterile seeds to avoid risks of invasiveness.  相似文献

Miscanthus sinensis Andersson grows naturally at the Hitachi mine. The root‐zone soil was acidic and contained high concentrations of Cu, Pb, Zn and exchangeable Al. Adventitious roots accumulated high concentrations of Al and Fe, but not other heavy metals. The purpose of this study was to elucidate the mechanism of tolerance of Al in M. sinensis, focusing on its chemical interaction with root endophytes. We isolated Chaetomium cupreum, which produced siderophores, from adventitious roots of M. sinensis via CAS assay. In inoculation tests, C. cupreum promoted M. sinensis seedling growth and increased Al and Fe uptake in the roots, although C. cupreum did not stimulate M. sinensis to produce Al detoxicants, such as citric and malic acids. Observation of the pattern of Al localization in the roots clarified that C. cupreum reduced Al toxicity in M. sinensis via compartmentalizing Al into fungal mycelia surrounding the roots and creating a less toxic Al‐localization pattern, allocating Al to the epidermis, endodermis and stele of roots. In conclusion, our results indicated that C. cupreum increases Al tolerance in M. sinensis growing at the acidic mine site.  相似文献

为探讨矿区优势植物粽叶芦(Thysanolaenalatifolia)、类芦(Neyraudiareynaudian)和芒草(Miscanthussinensis)的抗重金属污染能力及生态恢复潜力,对生长在铅镉污染土壤中3种植物的生理指标和重金属含量进行了测定。结果表明,同一铅、镉污染水平下3种植物的过氧化物酶活性和脯氨酸含量为芒草类芦粽叶芦,丙二醛含量为芒草粽叶芦类芦;芒草中的铅、镉含量最高、富集系数最大。3种植物抗铅镉能力为芒草类芦粽叶芦,芒草对重金属污染的耐受性最强。因此,在矿区土壤植被恢复时,芒草可作为备选植物之一。  相似文献

Mitochondrial DNA cytochrome b sequences (1140 bp) in 61 specimens of Chinese spiny loach Cobitis sinensis from 12 drainages in Taiwan were identified as two major clades, exhibiting a southern and a northern distribution, north of TzengWen and south of TzengWen (including TzengWen), respectively. The divergence time between these two phylogroups was estimated at 7·34–9·06 million years before present (b.p. ), but these two phylogroups were formed c. 3·41–4·23 and 2·22–2·75 M b.p. , respectively. Moreover, geological events have been recalculated that Taiwan Island emerged above sea level at an estimate of c. 4–5 M b.p. , and quickly became its present shape at c. 2 M b.p. through mountain building. These results suggest that these two major clades of C. sinensis in Taiwan might originate from two different continental populations, since the island's initial isolation in the Pliocene. Within southern Taiwan, the initial colonization was hypothesized to be in KaoPing River, followed by its northward dispersal. The high divergence between KaoPing and TzengWen was influenced by glaciations and landforms. Within north Taiwan, the colonization was from the Miaoli Plateau through western Taiwan to north‐eastern and northern Taiwan. This dispersal pattern is concordant with the previously proposed hypothesis. Apparently, both geological and phylogeographic evidence suggested that river capture of the upper Takia River by the LanYang River promoted range expansion in freshwater fishes and also indicated that the Central Range within Taiwan did not act as a barrier to the dispersal of C. sinensis.  相似文献

Tapiscia sinensis, a rare endemic woody plant with both male and hermaphrodite individuals, is distributed in southern China. Whether T. sinensis is functionally androdioecious is unknown. In this study, we compare the male fitness between male and hermaphrodite individuals and perform pollination experiments in different habitats, identify the ability of actual siring of pollen from hermaphrodites and males under natural pollination, and discuss the evolution and maintenance of androdioecy in T. sinensis. Research suggests that flowers and fruits grow synchronously on hermaphrodite plants of T. sinensis from April to June. The males of T. sinensis had more than twice the genetic contribution of hermaphrodites through their male function and the fruit set from male pollination and cross‐pollination was the highest in all of the treatments, whereas that from self‐pollination was the lowest. Additionally, paternity analysis showed that the hermaphroditic pollen could result in siring success under natural pollination. The results showed that T. sinensis is a functionally androdioecious tree, that male individuals might evolve from a hermaphroditic ancestor and that the synchronous growth of flowers and fruit in hermaphrodites might facilitate the evolution and maintenance of androdioecy in T. sinensis.  相似文献

Due to its versatility and storability, biomass is an important resource for renewable materials and energy. Miscanthus hybrids combine high yield potential, low input demand, tolerance of certain marginal land types and several ecosystem benefits. To date, miscanthus breeding has focussed on increasing yield potential by maximising radiation interception through: (1) selection for early emergence, (2) increasing the growth rate to reach canopy closure as fast as possible, and (3) delayed flowering and senescence. The objective of this study is to compare early season re-growth in miscanthus hybrids cultivated across Europe. Determination of differences in early canopy development on end-of-year yield traits is required to provide information for breeding decisions to improve future crop performance. For this purpose, a trial was planted with four miscanthus hybrids (two novel seed-based hybrids M. sinensis × sinensis [M sin × sin] and M. sacchariflorus × sinensis [M sac × sin], a novel rhizome-based M sac × sin and a standard Miscanthus × giganteus [M × g] clone) in the UK, Germany, Croatia and Italy, and was monitored in the third and fourth growing season. We determined differences between the hybrids in base temperature, frost sensitivity and emergence strategy. M × g and M sac × sin mainly emerged from belowground plant organs, producing fewer but thicker shoots at the beginning of the growing season but these shoots were susceptible to air frosts (determined by recording 0°C 2 m above ground surface). By contrast, M sin × sin emerged 10 days earlier, avoiding damage by late spring frosts and producing a high number of thinner shoots from aboveground shoots. Therefore, we recommend cultivating M sac × sin at locations with low risk and M sin × sin at locations with higher risk of late spring frosts. Selecting miscanthus hybrids that produce shoots throughout the vegetation period is an effective strategy to limit the risk of late frost damage and avoid reduction in yield from a shortened growing season.  相似文献

We present nine polymorphic di- and tri-nucleotide repeat nuclear microsatellite markers selected specifically for their use in high throughput studies concerning the dioecious allotetraploid Salix albaSalix fragilis willow complex. These taxa and their hybrids are difficult to discriminate using morphological characters. Thus, multiplex reactions were developed for these microsatellite loci and their effectiveness to distinguish individuals, especially hybrids, and their inheritance patterns in controlled crosses were determined. All loci displayed disomic–monogenic inheritance which allowed for the genotype data to be analysed as for a diploid organism. The nine loci produced a total of 67 alleles (mean, 7.4 alleles per locus; range, 3–11 alleles) in a reference panel of 57 individuals from two germplasm collections and natural populations. Gene diversity values (as measured by the expected heterozygosity) ranged from 0.000–0.820. A total of 53 distinct multilocus genotypes were observed, and ordination analysis revealed three separate clusters corresponding to S. alba, S. fragilis and hybrids. The microsatellite loci described here will be used in population genetic studies to investigate genetic variation, gene flow, levels of hybridisation and the extent of introgression in natural populations of the S. albaS. fragilis complex. They are also useful for clonal identification, conservation and sustainable management of germplasm collections, genetic mapping and the selection of individuals and/or certification of controlled crosses for breeding programmes.  相似文献

During gastrulation, one of the most important morphogenetic events in sea urchin embryogenesis, the gut bends toward the ventral side to form an open mouth. Although the involvement of transforming growth factor‐β (TGF‐β) signals in the cell‐fate specification of the ectoderm and endoderm along the dorsal–ventral axis has been well reported, it remains unclear what controls the morphogenetic behavior of gut bending. Here, using two sea urchin species, Hemicentrotus pulcherrimus and Temnopleurus reevesii, we show that TGF‐β signals are required for gut bending toward the ventral side. To search for the common morphogenetic cue in these two species, we initially confirmed the expression patterns of the dorsal–ventral regulatory TGF‐β members, nodal, lefty, bmp2/4, and chordin, in T. reevesii because these factors are appropriate candidates to investigate the cue that starts gut bending, although genetic information about the body axes is entirely lacking in this species. Based on their expression patterns and a functional analysis of Nodal, the dorsal–ventral axis formation of T. reevesii is likely regulated by these TGF‐β members, as in other sea urchins. When the Alk4/5/7 signal was inhibited by its specific inhibitor, SB431542, before the late gastrula stage of T. reevesii, the gut was extended straight toward the anterior tip region, although the ectodermal dorsal–ventral polarity was normal. By contrast, H. pulcherrimus gut bending was sensitive to SB431542 until the prism stage. These data clearly indicate that gut bending is commonly dependent on a TGF‐β signal in sea urchins, but the timing of the response varies in different species.  相似文献

Genome fingerprinting with a hypervariable minisatellite sequence of phage M13 DNA was used to study the genetic variation in individual species of the genera Bosand Bison(subfamily Bovinae) and in their interspecific and intergeneric hybrids. DNA fingerprints were obtained for domestic cow Bos taurus primigenius, vatussy Bos taurus macroceros, banteng Bos javanicus, gaur Bos gaurus, wisent Bison bonasus, bison Bison bison, and for the interspecific and intergeneric hybrids. Compared with the original species, most hybrids showed a greater variation in number and size of hybridization fragments. An association was revealed between the number of hybridization fragments and degree of consanguinity of interspecific hybrids resulting from unique crossing of domestic cow and banteng. Pairwise similarity coefficients were calculated to construct a dendrogram of genetic similarity, which reflected the relationships between the parental species and hybrids varying in degree of consanguinity. The applicability of the method for identifying interspecific and intergeneric hybrids and for studying the consequences of hybridization in the subfamily Bovinae is discussed.  相似文献

Miscanthus, a C4 perennial rhizomatous grass from Asia is a leading candidate for the supply of sustainable biomass needed to grow the bioeconomy. European Miscanthus breeding programmes have recently produced a new range of seeded hybrids with the objective of increasing scalability to large acreages limited by current clonal propagation. For the EU-GRACE project, new replicated field trials were established in seven locations across Europe in 2018 with eight intraspecific M. sinensis hybrids (sin × sin) and six M. sacchariflorus × M. sinensis (sac × sin) from Dutch and UK breeding programmes, respectively, with clonal Miscanthus × giganteus. The planting density of the sin × sin was double that of sac × sin (30,000 & 15,000 plants ha−1), creating commercially relevant upscaling comparisons between systems. Over the first 3 years, the establishment depended on location and hybrid. The mature sin × sin hybrids formed tight tufts of shoots up to 2.5 m tall which flower and senesce earlier than the taller sac × sin hybrids. Following the third growing season, the highest yields were recorded in Northern Italy at a low altitude (average 13.7 (max 21) Mg DM ha−1) and the lowest yielding was on the industrially damaged marginal land site in Northern France (average 7.0 (max 10) Mg DM ha−1). Moisture contents at spring harvest were lowest in Croatia (21.7%) and highest in Wales, UK (41.6%). Overall, lower moisture contents at harvest, which are highly desirable for transport, storage and for most end-use applications, were found in sin × sin hybrids than sac × sin (30% and 40%, respectively). Yield depended on climate interactions with the hybrid and their associated planting systems. The sin × sin hybrids appeared better adapted to northern Europe and sac × sin hybrids to southern Europe. Longer-term yield observations over crop lifespans will be needed to explore the biological (yield persistence) and economic costs and benefits of the different hybrid systems.  相似文献
