Lipid class and fatty acid compositions were determined in Limacina helicina and Clione limacina from an Arctic fjord and the marginal ice zone around Svalbard. C. limacina had higher levels of neutral lipids, including both alkyldiacylglycerols (ADG) and triacylglycerols (TAG), than L. helicina, which contained mainly TAG. However, considerable heterogeneity in the lipid classes and their fatty acids/alcohols were observed in C. limacina in that only two out of the seven specimens analysed were lipid-rich and contained both ADG and TAG, the others having only low percentages of TAG. In specimens of C. limacina containing ADG, 15:0 and 17:1n-8 were prominent fatty acids in both ADG and TAG. The fatty acids of the TAG of L. helicina were variable but 15:0 and 17:1n-8 were absent. We consider the heterogeneity in the fatty acid compositions of L. helicina to reflect temporal and spatial variability in the animals' predominantly phytoplanktonic and particulate diet, which occasionally includes small copepods. We further consider L. helicina to be the prime food for C. limacina and the noticeable amounts of 22:1 found in one sample of C. limacina to reflect significant input of Calanus either directly or indirectly through their prime food, L. helicina. We view the heterogeneity in the fatty acid compositions of both L. helicina and C. limacina, as well as the ability of C. limacina to biosynthesise WE, ADG, 15:0, and 17:1n-8, as adaptations to a large variation of food availability that enables C. limacina to synthesise lipids rapidly and flexibly. The Arctic pteropod Clione limacina was collected in Kongsfjorden, Svalbard, in mid June 2004, to study the lipid metabolism within the sites of lipid storage structures during long-term starvation. Animals survived in an aquarium without any food for nearly 1 year (356 days). Size, number of lipid droplets, dry and lipid mass, lipid class and fatty acid compositions of C. limacina were determined and separately analysed for the digestive gland and the remaining integument. During the starvation period, animals shrunk from 22.4 to 12 mm in length on average, and the number of lipid droplets decreased from 1,600 to 1,000 per animal. Dry mass (DM) and total lipid mass both dropped by about 80% from day 200 to the end. The lipid content as percentage DM of the total organism did not decrease significantly ranging from 43.8 to 32.3%DM. The lipid content of the trunk was moderate with about 20%DM. The digestive gland was very rich in lipids with more than 70%DM throughout the experiment and is the major site of lipid metabolism and storage. Triacylglycerols (TAG) decreased, in the total organism, from high initial levels of 62.6 to 43% of total lipid at the end. In contrast, the proportions of 1-O-alkyldiacylglycerols [diacylglycerol ethers (DAGE)] remained almost constant, varying between 20.4 and 28.4%. In the digestive gland, TAG ranged from 60.3 to 64.8% and DAGE from 23.6 to 32.2% from day 200 to the end of the experiment. TAG and DAGE of the trunk were most likely located in the lipid droplets and were almost depleted at the end of starvation. Besides their function as lipid deposit DAGE may also act as protecting substance against bacterial and fungal infections. During the first 200 days of starvation, the fatty acid compositions showed only small variations. Thereafter, fatty acids typical for storage lipids decreased in all body compartments. In adaptation to long periods of food scarcity, C. limacina has evolved various strategies as body shrinkage, utilisation of body constituents not essential for survival, a very low metabolism and slow lipid consumption.  相似文献

This study examines ontogenetic changes in lipid class composition in relation to survival and growth of five batches of sea scallops, Placopecten magellanicus, from egg to dissoconch stages. It also investigates the effects of antibiotic addition during metamorphosis to determine whether transient treatment during this critical period is effective in increasing metamorphic success. The top-performing larvae (growth rate: 4.7-5.0 μm day− 1, dissoconch survival: 32-57%) were characterized by a pronounced increase in absolute (ng larva− 1) and relative concentrations (wt.% of total lipid) of triacylglycerol (TAG) during the pre-metamorphic period, followed by utilization during metamorphosis (0.5-1.4 μg day− 1 larva− 1). In contrast, the low-performing scallops (growth rate: 3.6-4.5 μm day− 1, dissoconch survival < 1%) exhibited a constant, low level of TAG. These results strongly suggest that the accumulation of TAG during the pre-metamorphic period is a good predictor of scallop performance as measured by survival to dissoconch stage. Antibiotic treatment enhanced absolute and relative TAG concentrations in pre-metamorphic scallops and resulted in higher post-metamorphic survival and TAG concentration than in non-treated controls. However, the proportion of dissoconch to live larvae at the end of the experiment was significantly lower with antibiotic treatment (8% vs. 20% in controls), thus resulting in comparable yields of dissoconch larvae ∼ 8% irrespective of treatment. A possible negative effect of antibiotic treatment on bacterial induction of settlement and metamorphosis of sea scallop larvae is suggested.  相似文献

The objective of this study was to document and compare the lipid class and fatty acid composition of the green microalga Chlorella zofingiensis cultivated under photoautotrophic and heterotrophic conditions. Compared with photoautotrophic cells, a 900% increase in lipid yield was achieved in heterotrophic cells fed with 30 g L−1 of glucose. Furthermore heterotrophic cells accumulated predominantly neutral lipids (NL) that accounted for 79.5% of total lipids with 88.7% being triacylglycerol (TAG); whereas photoautotrophic cells contained mainly the membrane lipids glycolipids (GL) and phospholipids (PL). Together with the much higher content of oleic acid (C18:1) (35.2% of total fatty acids), oils from heterotrophic C. zofingiensis appear to be more feasible for biodiesel production. Our study highlights the possibility of using heterotrophic algae for producing high quality biodiesel.  相似文献

The effect of meal size on the specific dynamic action (SDA) of the juvenile snakehead (Channa argus) was assessed at 25 °C. The fish were fed with test diets at meal sizes of 0.5, 1, 2, 3, 4, and 5% body mass and the postprandial oxygen consumption rate was determined at 1-h intervals until it returned to the pre-prandial level. The peak metabolic rate increased from 237.4 to 283.2 mg O2 kg− 1 h− 1 as the relative meal size increased from 0.5% to 3% and leveled off at 4% and 5%. Factorial metabolic scope increased from 1.53 to 1.99 and SDA duration increased from 11.7 to 32.3 h as the relative meal size increased from 0.5% to 5%. The relationship between SDA duration (D) and relative meal size (M) was described as: D = 4.28 M + 10.62 (r2 = 0.752, P < 0.05, n = 50). The energy expended on SDA increased while the SDA coefficient decreased with increasing meal size. The results of the present study suggest that the snakehead may adopt different feeding strategies when taking in different amounts of food.  相似文献

To understand how a major cosmopolitan pest responds to two very different insecticidal proteins and to determine whether herbivorous insects and their frass could be environmental sources of recombinant proteins from transgenic plants, Spodoptera litura (Fab.) (Lepidoptera, Noctuidae) larvae were fed on tobacco leaves expressing either the biotin-binding protein, avidin, or the protease inhibitor, aprotinin. Control larvae received non-transgenic tobacco. Samples of larvae were taken after 5, 6 or 7 days’ feeding and frass was collected after two 24-h periods at 6 and 7 days. Insects in all treatments grew significantly during the experiment, but the avidin-fed larvae were significantly smaller than the others on Day 7. Avidin was found in all samples of avidin-fed larvae (7.0±0.86 ng mg−1, n=45), at a lower level than in their frass (31.9±5.08 ng mg−1, n=30), and these frass levels were lower than those of the the leaves fed to the larvae (69.0±6.71 ng mg−1, n=45). All of the avidin detected in these samples was capable of binding biotin. On average, between 10 and 28% of avidin was recovered with the methods used, whereas almost full recovery of aprotinin was effected. Aprotinin levels in larvae (8.2±0.53 ng mg−1, n=45) were also lower than aprotinin levels in frass (77.4±6.9 ng mg−1, n=30), which were somewhat lower than those in the leaves fed to the larvae (88.6±2.51 ng mg−1, n=45). Approximately half the trypsin-binding ability of aprotinin was lost in larvae, and in frass, aprotinin had lost about 90% of its ability to bind trypsin.  相似文献

The mass fragmentation patterns of stilbene glycosides isolated from the genus Lysidice were investigated by negative ion electrospray ionization tandem mass spectrometry, and the influence of collision energy on their fragmentation behavior is discussed. It is found that the presence of the Y0 and B0 ions in the MS2 spectra is characteristic for 1 → 6 linked diglycosyl stilbenes, while the Y0, Y1, and Z1 ions are representative ions of 1 → 2 linked diglycosyl stilbenes. These results indicate that ESI-MSn in the negative ion mode can be used to differentiate 1 → 6 and 1 → 2 linked diglycosyl stilbenes. Based on the fragmentation rules, 9 new trace constituents were identified or tentatively characterized in a fraction of Lysidice brevicalyx by using HPLC/HRMS and HPLC-DAD/ESI-MSn. The results of the present study can assist in on-line structural identification of analogous constituents and targeted isolation of novel compounds from crude plant extracts.  相似文献

The microalgae, Chlorella sp., were cultivated in various culture modes to assess biomass and lipid productivity in this study. In the batch mode, the biomass concentrations and lipid content of Chlorella sp. cultivated in a medium containing 0.025–0.200 g L−1 urea were 0.464–2.027 g L−1 and 0.661–0.326 g g−1, respectively. The maximum lipid productivity of 0.124 g d−1 L−1 occurred in a medium containing 0.100 g L−1 urea. In the fed-batch cultivation, the highest lipid content was obtained by feeding 0.025 g L−1 of urea during the stationary phase, but the lipid productivity was not significantly increased. However, a semi-continuous process was carried out by harvesting the culture and renewing urea at 0.025 g L−1 each time when the cultivation achieved the early stationary phase. The maximum lipid productivity of 0.139 g d−1 L−1 in the semi-continuous culture was highest in comparison with those in the batch and fed-batch cultivations.  相似文献

Effects of nitrate feeding on the cell growth and lipid accumulation of marine microalgae Isochrysis zhangjiangensis were investigated. When nitrate was supplied at interval of 24 h, instead of 72 h, a high lipid content of 40.9% and a biomass density of 3.1 g L−1 were obtained. To confirm whether I. zhangjiangensis accumulates lipid during nitrogen-repletion, a two-stage cultivation method was applied. This algal strain had a high lipid content during sustained nitrate addition and showed a high carbohydrate content under nitrate-depletion conditions. These results revealed that this algal strain can accumulate lipids under nitrogen-repletion conditions and accumulate carbohydrate under nitrogen-depletion conditions. When cultured in an extremely high nitrate concentration, 9 g L−1 at 24 h intervals, the growth of algal cells was suppressed, but the highest lipid content of 53% was attained. This special characteristic of lipid accumulation makes I. zhangjiangensis an ideal candidate for producing biodiesel using N-rich wastewater.  相似文献

The discovery of natural and natural-based compounds has resulted in its application as an alternative to synthetic algicides to control harmful algae in aquatic systems. Of the many natural-product-based algicides, sorgoleone, a natural plant product from Sorghum bicolor root exudates has been investigated for its controlling effect on different algal species and its acute fish toxicity. Growth of the blue green algal species Microcystis aeruginosa Kützing was completely inhibited by the crude methanol extract of sorghum root at 20 μg mL−1. The most noticeable inhibition was observed in the bioassay of n-hexane soluble extract, where 98% growth inhibition occurred in M. aeruginosa at the concentration of 1.25 μg mL−1. Sorgoleone very effectively controlled blue green algae inhibiting 97% of M. aeruginosa at 0.5 μg mL−1 and 99% of Anabaena affinis Lemmermann at 4 μg mL−1. In contrast, inhibition of the green algae species Chlorella vulgaris Beijerinck and Scenedensmus spp. at 16 μg mL−1 sorgoleone was 87 and 68%, respectively. There were no mortalities or adverse effects observed in any of the fish exposed to water control, solvent control, and a nominal concentration of 1 μg mL−1 during the test period. The no observed effect concentration (NOEC) value was 1.5 μg mL−1 for the tested fish (O. latipes). Sorgoleone can be considered as an effective and an ecologically and environmentally sustainable approach to controlling harmful algae.  相似文献

The possibility of elevating the omega-3 fatty acid contents in mammalian cells using the sdd17 gene from Saprolegnia diclina was investigated in the current study. The nucleotide sequence of the sdd17 gene was optimized and the pSDD17-IRES-GFP plasmid was introduced into murine 3T3 fibroblast cells by electroporation, following which its heterologous expression was evaluated by fatty acid analysis. Evaluation of GFP co-expression and RT-PCR analysis indicated that sdd17 could be expressed at very high levels in mammalian cells. Total cellular lipid analysis of transformed cells fed with arachidonic acid (20:4 n  6) as a substrate showed that the sdd17 expression resulted in an 82-155% (p < 0.05) increase in eicosapentaenoic acid (20:5 n  3) compared with the control. This expression also reduced the arachidonic acid/(eicosapentaenoic + docosapentaenoic + docosahexaenoic acid) ratio from approximately 4:1 in control cells to 1.5:1 in sdd17-transformed cells (p < 0.05). This study demonstrated that the foreign sdd17 gene from EPA-rich fungus was expressed at a high efficiency and caused the omega-3 fatty acid contents in mammalian cells to be elevated. It also provided a basis for potential applications of this gene in animal transgenesis.  相似文献

Previous work demonstrated that a mixture of NH4Cl and KNO3 as nitrogen source was beneficial to fed-batch Arthrospira (Spirulina) platensis cultivation, in terms of either lower costs or higher cell concentration. On the basis of those results, this study focused on the use of a cheaper nitrogen source mixture, namely (NH4)2SO4 plus NaNO3, varying the ammonium feeding time (T = 7-15 days), either controlling the pH by CO2 addition or not. A. platensis was cultivated in mini-tanks at 30 °C, 156 μmol photons m−2 s−1, and starting cell concentration of 400 mg L−1, on a modified Schlösser medium. T = 13 days under pH control were selected as optimum conditions, ensuring the best results in terms of biomass production (maximum cell concentration of 2911 mg L−1, cell productivity of 179 mg L−1 d−1 and specific growth rate of 0.77 d−1) and satisfactory protein and lipid contents (around 30% each).  相似文献

Industrial wastewater treatment comprises several processes to fulfill the discharge permits or to enable the reuse of wastewater. For tannery wastewater, constructed wetlands (CWs) may be an interesting treatment option. Two-stage series of horizontal subsurface flow CWs with Phragmites australis (UP series) and Typha latifolia (UT series) provided high removal of organics from tannery wastewater, up to 88% of biochemical oxygen demand (BOD5) (from an inlet of 420 to 1000 mg L−1) and 92% of chemical oxygen demand (COD) (from an inlet of 808 to 2449 mg L−1), and of other contaminants, such as nitrogen, operating at hydraulic retention times of 2, 5 and 7 days. No significant (P < 0.05) differences in performance were found between both the series. Overall mass removals of up to 1294 kg COD ha−1 d−1 and 529 kg BOD5 ha−1 d−1 were achieved for a loading ranging from 242 to 1925 kg COD ha−1 d−1 and from 126 to 900 kg BOD5 ha−1 d−1. Plants were resilient to the conditions imposed, however P. australis exceeded T. latifolia in terms of propagation.  相似文献

The analysis of tissue's naturally occurring stable carbon and nitrogen isotope ratios is a useful tool to delineate trophic relationships. However, the interpretation of δ13C and δ15N is complicated by the influence of multiple factors such as the tissue-specific lipid content. The aim of this work was to evaluate the effects of lipid extraction on δ13C and δ15N compositions in muscle, hepatopancreas and gonads of a marine decapod crustacean, the spider crab Maja brachydactyla. Samples were analyzed for stable isotopes before and after lipid removal, using a derived Soxhlet extraction method. Differences in δ13C and δ15N were measured among tissues before and after treatment. Lipid extraction of muscle did not have a significant effect on either δ13C or δ15N. By contrast, ecologically significant shifts for both carbon and nitrogen stable isotopes ratios (+ 2.9 ± 0.8‰ for δ13C, and + 1.2 ± 0.7‰ for δ15N) were noticed in the hepatopancreas. In regard to gonads, lipid extraction led to a shift only on δ13C (+ 1.3 ± 0.3‰). Finally, the derived Soxhlet extraction method removed the lipid influence for δ13C, and had an effect on δ15N composition for lipid-rich samples. We recommend this treatment for carbon stable isotope studies on decapod crustacean lipid-rich tissues.  相似文献

The specific metabolic rate (SMR) and haemolymph osmolality (HO) of the mud crab Rhithropanopeus harrisii Gould, 1841 from Baltic brackish waters were measured at a habitat salinity of 7 psu (T = 15 °C, full air saturation) and after step-wise acclimation to a salinity range of 3-27 psu. Values of SMR at 7 psu varied between 0.40 and 3.89 J g− 1 WW h− 1 (n = 25, wet weight range 0.051-1.142 g) and were significantly (p < 0.05) related to the specimen's wet weight (WW) according to the power regression SMR = 0.94 WW 0.41 (R2 = 0.68). The SMR of females did not differ significantly (p > 0.05) from those of males. When exposed to higher salinities, the SMR of R. harrisii decreased significantly (p < 0.05) and reached a minimum value at 23 psu (0.55 ± 0.05 J g− 1 WW h− 1, n = 6). Mean haemolymph osmolality at 7 psu amounted to 581 ± 26 mOsm kg− 1 (n = 5) and was 2.9 times higher than that of the external medium. R. harrisii hyperosmoregulated its body fluids up to 24 psu (727 mOsm kg− 1) at which salinity the isosmotic point was reached.  相似文献

Omega-3 (n − 3) fatty acids are emerging as bioactive agents protective against cardiovascular disease. However, their cellular delivery pathways are poorly defined. Here we questioned whether the uptake of n − 3 triglyceride-rich particles (TGRP) is mediated by cell surface proteoglycans (PG) using LDL receptor (LDLR)+/+ and LDLR−/− cell models. LDLR+/+ but not LDLR−/− cells showed higher n − 6 over n − 3 TGRP uptake. Removal of cell surface proteins and receptors by pronase markedly enhanced the uptake of n − 3 but not n − 6 TGRP. Lactoferrin blockage of apoE-mediated pathways decreased the uptake of n − 6 TGRP by up to 85% (p < 0.05) but had insignificant effect on n − 3 TGRP uptake. PG removal by sodium chlorate in LDLR+/+ cells substantially reduced n − 3 TGRP uptake but had little effect on n − 6 TGRP uptake. Thus, while n − 6 TGRP uptake is preferentially mediated by LDLR-dependent pathways, the uptake of n − 3 TGRP depends more on PG and non-LDLR cell surface anchoring.  相似文献

The progression of baculovirus (BmNPV, BmCysPD, AcMNPV or AcAaIT) infection in larval Bombyx mori and Heliothis virescens (1st, 3rd or 5th instar) was investigated following various starvation regimes. When the larvae were starved for 12 or 24 h immediately following inoculation, the median lethal time to death (LT50) was delayed by 9.5-19.2 h in comparison to non-starved controls. This corresponded to a delay of 10-23% depending upon the larval stage and virus that was used for inoculation. When a 24 h-long starvation period was initiated at 1 or 2 days post inoculation (p.i.), a statistically significant difference in LT50 was not found indicating that the early stages of infection are more sensitive to the effects of starvation. Viral titers in the hemolymph of 5th instar B. mori that were starved for 24 h immediately following inoculation were 10-fold lower (p < 0.01) than that found in non-starved control larvae. Histochemical analyses indicated that virus transmission was reduced in 5th instar B. mori that were starved for 24 h immediately following inoculation in comparison to non-starved control larvae. In general, the mass of larvae that were starved immediately after inoculation was 30% lower than that of non-starved control insects. Our findings indicate that starvation of the larval host at the time of baculovirus exposure has a negative effect on the rate baculovirus transmission and pathogenesis.  相似文献

The acid biocoagulants produced from non-sterile lactic acid fermentation by Lactobacillus casei TISTR 1500 were used to settle colloidal protein, mainly casein, at the isoelectric point in dairy effluent prior to secondary treatment. High concentration of azo dye (Ponceau 4R) in the dairy wastewater and the stress of starvation decreased the efficiencies of the micro-aerobic SBR. Consequently, low casein recovery obtained and organic removal suffered a decline. The number of lactic acid bacteria (LAB) also declined from log 7.4 to log 5.30 in the system fed with 400 mg L−1 of the dye containing wastewater. The recovery of the system, however, showed that 25,000 mg COD L−1 influent with 200 mg L−1 of the dye maintained the growth of LAB in the range of log 7.74–8.12, with lactic and acetic production (2597 and 197 mg L−1) and 83% protein removal. The results in this study suggested that the inhibitory effects were compensated with high organic content feeding.  相似文献

The energetics and behavior of the parapodial-swimming Aplysia brasiliana were investigated in order to compare net cost of transport (COTnet) between swimming and crawling, and to compare transport costs with other swimmers. Oxygen consumption (VO2) increased with increasing animal mass for resting, crawling, and swimming animals. Slopes of the regressions of log VO2 on log mass were 0.90, 0.91, and 0.89 for resting, crawling, and swimming, respectively. The regression for resting VO2 on mass was significantly lower than regressions of crawling and swimming on mass, which fell into a statistically homogenous subgroup. During 4-h swimming bouts, parapodial beat frequency dropped by less than 10% of starting values after 2 h and then stabilized for the remainder of the trial, whereas velocity steadily decreased to about 70% of starting values over the 4-h period. Initial beat frequency (at the start of a swimming bout) was negatively related to body mass, varying from 1.1 beat s− 1 for a 34 g individual to 0.7 beats s− 1 for a 500 g individual. Final beat frequency (at the end of a swimming bout) was also negatively related to body mass, but had a significantly lower intercept than initial beat frequency. Neither initial swimming velocity nor final swimming velocity was related to mass, but final velocity was significantly lower than initial velocity. A 250 g A. brasiliana swam at 345 m h− 1 and crawled at 7 m h− 1. Swimming COTnet (0.1 ml O2 kg− 1 m− 1) for a 250 g A. brasiliana was 50 times less than crawling COTnet (5.3 ml O2 kg− 1 m− 1). While the crawling COTnet for A. brasiliana fell within the range of other marine gastropods, swimming COTnet was less than that of swimming crustaceans, and much less than another gastropod, Melibe leonina, that uses lateral bending to swim.  相似文献

Crude glycerol is a major byproduct of the biodiesel industry; previous research has proved the feasibility of producing docosahexaenoic acid (DHA, 22:6 n − 3) through fermentation of the algae Schizochytrium limacinum on crude glycerol. The objective of this work is to investigate the cell growth kinetics, substrate utilization efficiency, and DHA production of the algae through a continuous culture. Steady-state biomass yield, biomass productivity, growth yield on glycerol, specific glycerol consumption rate, and fatty acid composition were investigated within the range of dilution rate (D) from 0.2 to 0.6 day−1, and the range of feed crude glycerol concentration (S0) from 15 to 120 g/L. The maximum specific growth rate was determined as 0.692 day−1. The cells had a true growth yield of 0.283 g/g but with a relatively high maintenance coefficient (0.2216 day−1). The highest biomass productivity of 3.88 g/L-day was obtained at D = 0.3 day−1 and S0 = 60 g/L, while the highest DHA productivity (0.52 g/L-day) was obtained at D = 0.3 day−1 and S0 = 90 g/L due to the higher DHA content at S0 = 90 g/L. The biomass and DHA productivity of the continuous culture was comparable to those of batch culture, while lower than the fed-batch culture, mainly because of the lower DHA content obtained by the continuous culture. Overall, the results show that continuous culture is a powerful tool to investigate the cell growth kinetics and physiological behaviors of the algae growing on biodiesel-derived crude glycerol.  相似文献
