Summary An efficient regeneration system was developed by culturing immature cotyledons and embryo axes of Arachis hypogaea L. cv. Georgia Green on Murashige and Skoog basal medium (MS) supplemented with various concentrations of thidiazuron (TDZ; 1, 5, 10, and 15 μM). Highly morphogenic callus was produced from 100% of the explants comprising the cotyledon with attached embryo axis when cultured in the dark on 10 μM TDZ. Upon excision and continued culture in the dark on 10 μM TDZ, morphogenic callus grew repetitively during monthly subcultures and retained its regeneration potential. For organogenesis, a gradual reduction in TDZ concentration and exposure to light were necessary before transfer to MS basal medium. Inclusion of indole-3-butyric acid in liquid MS medium favored rooting of recovered shoots. A distinct feature of this investigation is the induction of highly morphogenic callus by TDZ and regeneration of morphologically normal, fertile peanut plants after 8 months of callus subculture.  相似文献

简单重复序列在各种生物基因组中广泛存在,同分子进化、遗传多样性、分子标记和某些遗传性疾病等密切相关.本文以全部32种18 bp三核苷酸双链重复序列作为研究对象,对它们在聚合酶作用下的扩展合成进行了系统研究.探讨了反应温度、序列本身等对扩展效率和产物长度的影响.结果显示,几乎所有的序列都能扩展变长.但序列对其扩展效率有很大影响:GC含量较多的短链,尤其是一条链中同时有G和C的短链扩展效率较高;全由AC和GT组成的双链也较易扩展.短链的最适扩展温度与短链GC组成呈一定的正相关关系.琼脂糖凝胶电泳和聚丙烯酰胺凝胶电泳分析发现,大部分产物单一性良好,且产物分子质量的大小随反应时间线性增加;随着反应温度升高,产物差异性增大.最后分析了双链重复序列的“复制滑移”扩展机理,有望为进一步研究重复序列的分子进化和基因检测中的非特异性扩增等奠定基础.  相似文献

Somatic embryogenesis in semul, Bombax ceiba L. (Bombacaceae) was achieved from immature zygotic embryo explants on MS medium. The cytokinin BA induced somatic embryogenesis at higher frequencies than the auxin 2,4-d. The rate of somatic embryogenesis was inversely related to the concentration of BA. A constant supply of 0.44 M BA was necessary for a high multiplication rate. Conversion of somatic embryos into plantlets is reported.  相似文献

Swine genomic DNA segments containing repetitive sequences were isolated from a porcine genomic library using genomic DNA as a probe. Three fragments containing the repetitive sequences from two of the primary phage clones were subcloned for sequence analysis, which revealed six new PRE-1 repetitive families other than those reported earlier by Singer et al. (Nucleic Acids Research 15, 2780, 1987). The frequency of the repetitive sequences in the swine genome was estimated at 2 x 10(6) per diploid genome. Sequence analysis revealed similarities between these repetitive sequences and that of arginine-tRNA gene.  相似文献

Characteristics of genome organization in the sulfur-dependent thermoacidophilic archaebacterium Sulfolobus acidocaldarius have been studied. By means of hybridization analysis it is shown that the genome of S. acidocaldarius, unlike the genome of the extremely halophilic archaebacterium Halobacterium halobium, does not contain repetitive sequences.  相似文献

A primer pair, PB and BSH, which amplified alts, a portion of Candida albicans-specific repetitive sequence, RPS, gave stable and reproducible fingerprint patterns of the strains by polymerase chain reaction (PCR). We applied this method to clinical isolates of C. albicans for strain discrimination. Using PCR fingerprint patterns, we could analyze the relatedness of C. albicans strains including those isolated from children with leukemia and their bedside parents. The results indicated that PCR analysis targeting an alt region gives rise to the same conclusion as the previous study obtained by SmaI RFLP analysis.  相似文献

Nuclear genome analysis using RFLPs and RAPDs has been assessed within different species of the genus Brachypodium P. Beauv. and representatives of other grasses in order to determine the characteristics of the Brachypodium genome and to establish its evolutionary position in relation to other Pooideae. Distinctive features of the Brachypodium genome are its small size, the low amount of repetitive DNA, the lack of restriction fragment length polymorphisms within the genus for the assayed probe/enzyme combinations, and the genomic variability demonstrated at species level by random DNA amplification. These molecular studies confirm Brachypodium as an isolated ancient genus best placed in its own tribe (Brachypodieae). Its relationships to other tribes Bromeae, Triticeae, Poeae are resolved, Brachypodieae being the earliest tribe to diverge from this core of pooids. Within the genus two major Old World clades are distinguishable: an annual clade, represented only by B. distachyon; and a perennial clade, represented by all the other species studied (except B. mexicanum). The perennial American species B. mexicanum appears equally attached to these two clades. RFLP data were found to be useful in obtaining phylogenies at generic and higher rank levels, whereas the highly variable RAPD data were more suitable for resolving interspecific and intraspecific evolutionary pathways.  相似文献

Site-directed mutagenesis is often a prerequisite for elucidation of the functional significance of cis- and trans-factors involved in gene regulation. The aim of this study was to delete the primary binding site for heterogeneous nuclear ribonucleoprotein I (hnRNPI) within the inducible nitric oxide synthase (iNOS) 3′ untranslated region mRNA. The binding site consists of a 53-nucleotide CU-rich region within a long stretch of polypyrimidines. As a result of primer pair annealing, the repetitive sequence limited the use of several deletion methods based on polymerase chain reaction. Therefore, a megaprimer approach was chosen. The megaprimer was produced by a forward primer outside the polypyrimidine-rich region, and a mutagenic reverse primer annealing to flanking regions of the desired deletion, thereby looping out the target sequence. Subsequently, this megaprimer was used to create the final deletion recombinant. The deletion was verified by sequencing and by ultraviolet cross-linking mouse liver protein extracts with radiolabeled mutant and wild-type RNAs. In conclusion, the megaprimer method offers a solution for generating large internal deletions in repetitive sequences, which facilitates investigations on large repetitive DNA or RNA regions interacting with trans-factors.  相似文献

脑缺氧和缺氧后的再灌注常导致一系列复杂的生理生化改变.通过小鼠重复缺氧后,观察其脑组织中的神经节苷脂和单胺类递质水平变化情况,发现重复缺氧时,随着缺氧次数的增加,a.神经节苷脂水平(以唾液酸含量表示)非常明显地持续下降(P<0.01),其中GM1与GD1b相对组分百分比值下降尤为突出(GM1:P<0.05,GD1b:P<0.01); b.单胺类神经递质中NE和DOPA水平下降,DA和HIAA水平升高(P<0.01).结果提示脑组织经重复缺氧后,中枢神经组织细胞膜受到一定程度的损伤,其结果可能影响到单胺类神经递质的合成、释放、重摄取和贮存的全过程;据此推测脑组织缺氧时神经节苷脂与单胺类神经递质水平改变时存在着相互联系.  相似文献

A single system is presented, where both genetic and epigenetic control of tumor induction can be studied at the same time. This system is offered by the amphidiploid tumorous hybrid Nicotiana glauca × N. langsdorffii, a nontumorous mutant of it and the nontumorous parent species N. glauca and N. langsdorffii. The aim of the present paper is to compare long-term in vitro cultures of tumorous (genetic and habituated), and nontumorous strains, through the characterization of their genomes according to several physico-chemical parameters. The data reported show that both qualitative and quantitative differences in DNA complexity are correlated with the tumorous transformation. Particularly, a high degree of mismatching between the DNAs of the tumorous and nontumorous hybrids and the lack, in the second genotype (nontumorous), of three DNA peaks in Ag+?Cs2SO4 analytical ultracentrifugation profile seem to support the hypothesis, suggested in a previous paper, of the presence, in the nontumorous mutant, of a gross chromosomal rearrangement, probably a deletion. Amplification and underreplication of specific sequences also seemed to be correlated with changes from the normal to the tumorous state, highly repetitive sequences being present in higher amounts in the normal strains and in the habituated N. glauca than in the case of the tumorous hybrid. Finally, DNA bound ion contents were found to be strikingly higher in tumorous than in nontumorous tissues. The results are discussed in the frame of the general hypothesis of high somatic genomic plasticity in plants.  相似文献

In Drosophila melanogaster the tandemly arranged repetitive sequences coding for 18S and 28S rRNA are heterogenous at the level of the spacers between units and insertions that interrupt many 28S rRNA genes. This heterogeneity contrasts with the homogeneity of the regions transcribed into 18S and 28S rRNA. Homogenization and evolution of repetitive genes are usually explained by conversion, amplification events or unequal crossovers. In this paper we studied the change in rDNA patterns associated with partial reversion of bobbed mutations. In most cases, no increase in rDNA gene number, but a new repartition of gene types were found.  相似文献

In the neuromuscular junction, blockade of potassium channels can produce multiple discharges after single nerve stimulation. In the large spacer of the rDNA of Vicia faba, multiples of a 0.32 kilobasepair (kb) sequence reiterate to various degrees. We sequenced the repetitious region consisting of the repeating sequences and its flanking regions using two cloned plasmids, which contain V. faba rDNA segments encompassing the whole region of the large spacer. The repetitious region was found to consist of multiple complete copies and one truncated copy of a 325 bp repeat unit and to be flanked by direct repeat sequences of about 150 bp. The set of direct repeats located at either side of the repetitious region differed from each other with about 10% sequence heterogeneity. However, nucleotide sequences of the direct repeats were well conserved between the two clones examined. Southern blot hybridization indicated a widespread distribution within the whole V. faba genome of some related sequences with high homologies to the 325 bp repeat unit and to the direct repeats.  相似文献

由于草菇是一种同宗结合的食用真菌,这给草菇的杂交育种带来了一定的困难。本文在建立草菇部分基因文库的基础之上,对草菇的基因文库进行了鉴定。在草菇基因文库中任意抽取72个克隆,利用专一的PCR方法,测出在草菇基因文库中,草菇基因组DNA的平均大小为1156个碱基对。在基因文库中任意选择53个克隆,利用专一的PCR进行DNA扩增以及dig非同位素标记,用于和草菇基因组DNA杂交。在测试的53个克隆中有8%的高度重复序列,36%中度重复序列和56%的低度重复序列。  相似文献

Interspersed elements are ubiquitous in the genomes of higher eukaryotes and account for over a third of the genomic DNA (Smit 1996). In swine the short interspersed elements, SINEs or PREs (porcine repetitive elements), have been found in a number of introns and 3' untranslated regions of different genes. However, compared to human Alu repeats the number of available PRE DNA sequences is still limited. In this study we have compared 85 PREs selected from DNA sequence database entries. The PREs were aligned and for each nucleotide position the relative frequencies of the four bases were calculated. A consensus sequence was derived from the first base usage. Similar to studies of SINEs in other species, the analysis showed that most mutations in PREs occur at CpG dinucleotide hot spots. The position variability for the two most frequent bases shows a bimodal distribution. The analysis suggests that the porcine SINEs can be divided into three major subfamilies sharing conserved nucleotide similarities.  相似文献
