河套蜜瓜花粉管通道法转化hALR基因   总被引:1,自引:0,他引:1  
为利用河套蜜瓜为生物反应器生产可用于治疗肝病的蛋白因子,以人胎肝mRNA为模板,经RT-PCR扩增获得了471 bp cDNA片段.克隆到pMD18-T载体,命名为pMD-ALR,测序分析结果与GenBank中报道的hALR编码序列完全相同.酶切回收hALR片段,插入到含有甜瓜果实黄瓜素(Cucumisin)基因启动子的果实特异性表达载体pPZP-CGN中,构建了hALR甜瓜果实特异性表达载体pPZP-CAN.花粉管通道法转化试验大田自花授粉花63朵,收获T0代果实27颗,结实率为42.9%.提取T0代种子无菌苗DNA,经PCR、PCR-southern和斑点杂交检测有13株呈阳性.  相似文献

采用花粉管通道技术,将几丁质酶基因导入14个大豆品种,共导入1125朵花,花朵的成活率为47.8%,将Bt基因导入11个大豆品种,共导入260朵花,花朵的成活率为40.8%。8个转Bt基因品种中的D1代共收获192粒种子,获得幼苗132株,出苗率为68.75%,把转Bt基因的132株植株用X-Gluc溶液检测,没有发现阳性反应,将转Bt基因的132株植株进行PCR检测,得到5株阳性转化植株,对D1代获得的5株阳性转化植株的D2代,进行PCR检测,得到D2代稳定遗传的阳性转化植株2株。  相似文献

利用西瓜有性生殖过程 ,将携带有外源几丁质酶基因的质粒DNA涂抹在授粉后的柱头上 ,使其沿花粉管通道进入到生殖细胞 ,得到转化处理种子。转化T1代植株经除草剂Basta筛选和PCR扩增获得转化植株。对T3 代株系进行田间自然发病和人工接种鉴定获得 3个抗枯萎病的株系。结果表明 :几丁质酶对镰刀菌引起的西瓜枯萎病有一定的抑制作用 ,利用花粉管通道法直接导入西瓜活体植株的技术是可行的。  相似文献

以番木瓜"solo Ⅱ"号植株为受体材料,用花粉管通道法进行了番木瓜环斑病毒外壳蛋白基因(PRSV-CP)278 bp片段的遗传转化.采用质粒DNA和农杆菌菌液两种导入液,分别处理花187和232朵,收获成熟番木瓜105和30个.座果率分别为56.15%和12.93%,随机选择每种载体的种子100粒播种.成株率分别为61%和60%.对T1幼苗(除含空载体外)全部进行PCR检测.检测结果为质粒DNA和农杆菌菌液两种导入液转化所得的幼苗阳性率分别为50.54%和51.22%.  相似文献

利用农杆菌介导法将番茄红素β-环化酶基因(Lycb)转入由玉米自交系天塔五号植株,分析基因在T0转化及后代的遗传情况,结果表明,在27株T0转基因植株中,PCR初步检测后8株呈阳性;将T1代转基因植株以株系为单位用200mg/L草铵膦抗性筛选后,收获抗性植株种子。T2代转基因植株进一步进行PCR、RT-PCR和田间草铵膦涂抹检测,结果表明,PCR、RT-PCR为阳性的6个株系植株均具有草铵膦抗性。选取6株阳性植株提取叶片总类胡萝卜素,经HPLC分析其β-胡萝卜素含量显著高于野生型,表明目的基因Lycb成功的转入玉米,并得到了稳定遗传。  相似文献

采用PPT抗性筛选、PCR扩增、PCR-Southern杂交、ELISA、Western杂交等方法,对花粉管通道法介导IL-4基因转化大白菜的后代进行了分子生物学检测,同时结合统计分析手段研究了IL-4基因在转基因大白菜后代中的遗传规律.结果表明:(1)PPT抗性的转基因大白菜,T4代植株PPT抗性的阳性率为33.43%,IL-4基因PCR扩增的阳性率为1.39%.IL-4的分离情况不符合孟德尔分离定律.(2)对T4代5个株系的14个PCR阳性株进行ELISA检测,其中有7份样品为阳性,IL-4表达量约为326.87 ~ 1233.13 ng/g FW,同一株系内各株间IL-4的表达量差异显著.(3) Western杂交进一步证实IL-4在转基因大白菜后代中能正常表达.PCR-southern杂交表明IL-4基因已整合到大白菜基因组中,且在转基因后代植株中仍然存在.  相似文献

目的:以携带水稻矮缩病毒(RDV)运动蛋白缺陷型(MP-)基因的转基因玉米种子T0为试验材料,通过分子分析、抗病鉴定及农艺性状筛选得到转基因纯合株系。方法:首先对转化种子进行潮霉素抗性筛选,其后对各代转基因材料进行PCR检测、农艺性状调查和抗病鉴定。结果:从645粒T0转化种子得到抗潮霉素植株246株,即T1转基因植株。T1、T2、T3、T4代材料的PCR检测阳性率分别为56.9%、83.9%、94.6%和99.8%,证明RDVMP-基因已被导入玉米自交系中,且目的基因可以稳定遗传到转基因植株及其后代。田间人工接种抗病鉴定结果表明,经过连续筛选,转基因后代植株(系)的抗病性不断提高,T1、T2、T3和T4代中的高抗病材料分别占总材料数的8.9%、31.5%、70.7%和100%;所选纯合系连续2年的发病率均为0,抗病性比相应对照株系提高4级。农艺性状调查结果表明,转基因株系的株高比对照株系高15.0~33.4cm,穗位高提高13.4~20.2cm,;穗长增加2.0~3.8cm,穗粒数增加10.2~22.8粒。结论:根据分子检测、田间抗病鉴定及农艺性状鉴定结果,选育到96C0502、96C0507和96C0513等抗矮花叶病转基因玉米纯合株系。  相似文献

我国棉花抗虫基因大都为Cry1Ab/c,抗性风险日趋增加。本研究依据棉花密码子偏好,人工合成Bt-Cry5Aa抗虫基因,通过花粉管通道法转入棉花,并通过卡那霉素法及PCR方法对不同世代转化株进行鉴定,同时进行了抗虫性测试。结果表明,通过花粉管通道法成功获得转Bt-Cry5Aa基因植株,通过田间卡那霉素鉴定,阳性株率T1为7.76%,T2为73.1%,T3为95.5%;PCR检测显示,T1阳性率为2.35%,T2为55.8%,T3为94.5%;田间抗性试验分析,转Bt-Cry5Aa株系对第2、3、4代棉铃虫校正死亡率分别达到85.42%、75.35%和62.79%,其抗虫性与GK19相比差异不显著;Bt-Cry5Aa能够部分替代目前主流鳞翅目抗虫基因,是棉铃虫的新抗源。  相似文献

利用农杆菌介导的转化系统转化中花11成熟胚愈伤组织,获得1489个独立转化的T-DNA插入再生株系。PCR和Southern杂交的结果表明,69.8%转化株系被整合了T-DNA,通过Tail-PCR也从转化植株中扩增出T-DNA侧翼序列。同时对1066个T1转化株系的抽穗期、株高、单株穗数的调查结果表明,不同株系中分离出了突变植株。  相似文献

为在短期内提高小麦品质,本研究构建了一个含有反义蜡质基因、HMWGS1Dy10基因和抗除草剂基因的重组质粒pWXAB,以优良小麦品种(冀麦24、白玉149、9411等)为材料,采用花粉管通道法进行转基因研究。对转基因后代进行除草剂筛选和SDSPAGE电泳。结果表明,除草剂基因及反义蜡质基因已经整合到小麦基因组中,从抗性水平而言,其转化频率为0.5%。  相似文献

To determine the effects of soil nitrogen on pollen production, pollen size, and pollen performance, two cultivars of zucchini (Cucurbita pepo) were grown under two nitrogen regimes in an experimental garden. The two cultivars were true breeding for alternative alleles for a one gene trait, ovary color. The soil nitrogen treatment had a significant effect on most measures of reproductive output through the female function. The nitrogen treatment did not affect the number of staminate flowers per plant but did have an effect on the number of pollen grains per staminate flower and the mean pollen grain size. A pollen mixture experiment revealed that pollen produced by plants in the high nitrogen treatment sired significantly more seeds than pollen from low nitrogen plants. Moreover, we found that the high nitrogen pollen sired even a greater percentage of seeds in the region of the fruit (ovary) previously shown to be fertilized by the fastest growing pollen tubes. Thus, the difference in the number of seeds sired by pollen from the two nitrogen treatments is due to differences in pollen performance. We conclude that spatial heterogeneity in soil nitrogen can influence the paternity of seeds in a plant population.  相似文献

The percentage of cross-fertilization in the field bean was found to be around 45%, and to be higher on the lower nodes of the plants than on the upper. No differences in the relative pollen tube growth rates of self and foreign pollen were observed. The different lines and genotypes investigated responded differently to open pollination, selfing with manual tripping, and selfing without tripping. Fertilizer application and date of sowing affected the number of flowers produced and the number of seeds set, but did not affect the ratio of seeds to flowers, i.e. the index of self fertility.  相似文献

The visual marker GUS has been utilized in this study to understand the Arabidopsis thaliana vacuum infiltration transformation process by Agrobacterium tumefaciens. High transformation frequencies of up to 394 transgenic seeds per infiltrated plant were achieved. The results showed that the majority of the transgenic seeds from single infiltrated plants were from independent transformation events based on Southern analysis, progeny segregation, distribution of transgenic seeds throughout the infiltrated plants and the microscopic analysis of GUS expression in ovules of infiltrated plants. GUS expression in mature pollen and anthers was monitored daily from 0 to 12 days post-infiltration. In addition, all ovules from a single infiltrated plant were examined every other day. GUS expression frequencies of up to 1% of pollen were observed 3-5 days post-infiltration, whereas frequencies of up to 6% were detected with ovules of unopened flowers 5-11 days post-infiltration. Most importantly, transgenic seeds were obtained only from genetic crosses using infiltrated plants as the pollen recipient but not the pollen donor, demonstrating Agrobacterium transformation through the ovule pathway.  相似文献

The seed-specific phaseolin promoter (Ph/P) was fused to an ipt gene, then was cloned to a plant expression vector containing a gus gene driven by a 35S promoter. Cotton (Gossypium hirsutum L.) plants were transformed through pollen tube pathway methods. After seed germination, histochemical staining of the roots demonstrated that 32 GUS positive plants were obtained and three of which contained the chimeric Ph/P-ipt transgene as confirmed by PCR analysis. An immunosorbent assay showed that two of the three transgenic cotton lines contained higher levels of zeatin equivalents in seeds than the control. Seedling development of these two transgenic lines differed from the control in a reduction of the shoot growth, showing a stunted phenotype as expected, but a surprisingly developed root system with a 3-4 fold fast-growing lateral roots. In addition, fibers (seed-hairs) of the two transgenic cotton lines were considerably shorter than those of the control. These results indicate that genetic engineering may be used to manipulate the development of cotton plants, particularly cotton fibers.  相似文献

Fruits formed after different pollination regimes (flowers hand pollinated, unpollinated, and open pollinated) and the seeds obtained were characterized in the wild chilli pepper Capsicum flexuosum Sendtn. Pollen tube development in vivo and ovary growth were also analyzed. Seedless fruits and empty seeds were abundant among the fruits from hand pollinated and open pollinated flowers, while no more than one seed with embryo was found in a low percentage of fruits from such pollination treatments. Parthenocarpic fruits were formed from unpollinated flowers. Pollen tube growth was arrested in the upper third of the style for almost all pollen tubes except for a single one that may continue elongating occasionally. The ovary size increased continuously after pollination, even without fertilization. The sum of the evidence registered may help to explain the low number of seeds with embryo harvested, the abundance of seedless fruits formed from pollinated flowers (possibly parthenocarpic), and the high rate of parthenocarpic fruits formed from unpollinated flowers.  相似文献

The mean volume of pollen grains and total pollen production varied both within and among plants of Erythronium grandiflorum. The second flowers of two-flowered plants tended to produce smaller and fewer grains than first flowers, but there was no overall relationship between mean pollen grain size and production per flower. I evaluated the effects of pollen size differences within and among plants on two components of male reproductive success: pollen tube growth and postfertilization siring ability. Pollen tubes grown in media were longer for second flowers, but were not correlated with the mean size of pollen grains, suggesting that (1) internal resource content of pollen (i.e., carbohydrates plus lipids) was not associated with the hydrated size of pollen, and that (2) pollen from second flowers contained more resources. I analyzed the growth rate and the fertilization ability of pollen growing in styles. Growth rate differed among donors and recipients, but no effects of pollen or donor characters (i.e., pollen production, grain size, and flower position) were detected. In single donor pollinations, pollen size was negatively correlated with fertilization ability across donors, and positively correlated with postfertilization siring ability of donors. A second experiment used pairs of donors; within-plant differences in pollen size and flower position had effects similar to the single donor experiment on fertilization ability, but among-plant differences were not significant. The results corroborate earlier experiments that suggest that the growth of pollen tubes in the style is probably controlled by the recipient, since donor characters had minimal effects on pollen fertilization ability. Postfertilization siring ability was not affected by within-plant differences in mean grain size and production. For among-donor differences, the number of seeds set for each donor was positively correlated with the mean grain volume, and when a donor producing large pollen fertilized ovules in an ovary, there was increased seed abortion for seeds in the same ovary sired by a second donor. In addition, the total number of seeds produced by a fruit was decreased when both donors had large pollen, apparently due to increased postfertilization abortion. Postfertilization processes appear to be influenced by paternal differences that are expressed through competition among developing seeds for maternal resources.  相似文献

The phenology, major floral characteristics, fruiting levels, and breeding system ofCistus ladanifer L. (Cistaceae), a common western Mediterranean shrub species, were studied in a southern Spanish population. The white, large (64 mm in diameter) flowers of this shrub appear during spring (March–May) and produce abundant pollen and nectar. In the year of study, flowers lasted up to three days, during which they were visited by a diverse array of insects including beetles, flies, and bees. Hand-pollinations revealed that flowers do not set any seed unless cross pollen is applied to the stigma. Microscopical observations indicate that self pollen tubes grow down the stigma but invariably fail to induce fruit maturation. At the plant level, all estimates of fecundity investigated (number of seeds per capsule, proportion of ovules developing into seed, and proportion of flowers setting fruit) were highly dependent on nearest neighbour distance, with isolated plants setting as little as 0% fruit. In contrast, plants within a clump often transformed into fruit as much as 90% of the flowers. At the population level, seed output was estimated to range between 3,000 and 270,000 seeds per plant during 1991.  相似文献
