A new type of polymer highly resistant to nonspecific protein adsorption is reported. Poly(N-methyl-β-alanine) (PMeA) and poly(N-ethyl-β-alanine) (PEtA) synthesized via cobalt-catalyzed carbonylative polymerization of N-methylaziridine and N-ethylaziridine were end-functionalized with thiol groups and grafted onto Au surfaces. Protein adsorption was studied by the surface plasmon resonance (SPR) method. The amounts of representative single proteins adsorbed onto the PMeA- and PEtA-grafted surfaces were below the detection limit of SPR at the pg/mm(2) level. After exposure to full blood plasma and serum for 10 min, protein adsorption was at the level of ~ 100 pg/mm(2), similar to the level of protein adsorption on poly(ethylene glycol) surfaces subjected to identical conditions. These poly(β-peptoid)s therefore provide excellent protein resistance comparable to the best antifouling materials known to date. The strong proton-accepting ability when forming hydrogen bonds is suggested to be an important attribute for these poly(β-peptoid)s as well as other poly(tertiary amide)s as antifouling materials.  相似文献

Four aromatic bromo compounds have been isolated from the ethanolic extract of Rytiphlea tinctoria after treatment with diazomethane: 2,4-dibromo-1,3,5-trimethoxy-benzene,5,6,3′,5′-tetrabromo-3,4,2′,4′,6′-pentamethoxydiphenylmethane, 5,6-dibromo-3,4-dimethoxy-benzyl alcohol and its ethyl ether. In addition to sterols, amino acids, this extract also contains quinonoid bromo-pigments which could play a rôle in photosensitisation of chlorophylls, a rôle normally taken by the phycobilins, in other Rhodophyceae.  相似文献

Human cerebrospinal fluid (CSF), produced by the choroid plexus and secreted into the brain ventricles and subarachnoid space, plays critical roles in intra-cerebral transport and the biophysical and immune protection of the brain. CSF composition provides valuable insight into soluble pathogenic bio-markers that may be diagnostic for brain disease. In these experiments we analyzed amyloid beta (Aβ) peptide and micro RNA (miRNA) abundance in CSF and in short post-mortem interval (PMI <2.1 hr) brain tissue-derived extracellular fluid (ECF) from Alzheimer’s disease (AD) and age-matched control neocortex. There was a trend for decreased abundance of Aβ42 in the CSF and ECF in AD but it did not reach statistical significance (mean age ~72 yr; N=12; p~0.06, ANOVA). The most abundant nucleic acids in AD CSF and ECF were miRNAs, and their speciation and inducibility were studied further. Fluorescent miRNA-array-based analysis indicated significant increases in miRNA-9, miRNA-125b, miRNA-146a, miRNA-155 in AD CSF and ECF (N=12; p<0.01, ANOVA). Primary human neuronal-glial (HNG) cell co-cultures stressed with AD-derived ECF also displayed an up-regulation of these miRNAs, an effect that was quenched using the anti-NF-кB agents caffeic acid phenethyl ester (CAPE) or 1-fluoro-2-[2-(4-methoxy-phenyl)-ethenyl]-benzene (CAY10512). Increases in miRNAs were confirmed independently using a highly sensitive LED-Northern dot-blot assay. Several of these NF-кB-sensitive miRNAs are known to be up-regulated in AD brain, and associate with the progressive spreading of inflammatory neurodegeneration. The results indicate that miRNA-9, miRNA-125b, miRNA-146a and miRNA-155 are CSF- and ECF-abundant, NF-кB-sensitive pro-inflammatory miRNAs, and their enrichment in circulating CSF and ECF suggest that they may be involved in the modulation or proliferation of miRNA-triggered pathogenic signaling throughout the brain and central nervous system (CNS).  相似文献

Methanobacterium thermoautotrophicum was grown on a mineral salts medium in a fermenter gassed with H2 and CO2, which were the sole carbon and energy sources. Under the conditions used the bacterium grew exponentially. The dependence of the growth rate () on the concentration of H2 and CO2 in the incoming gas and the dependence of the growth yield ( ) on the growth rate were determined at pH 7 (the pH optimum) and 65° C (the temperature optimum).The curves relating growth rate to the H2 and CO2 concentration were hyperbolic. From reciprocal plots apparent K s values for H2 and CO2 and max were obtained: app. = 20%; app. = 11%; = 0.69 h-1; t (max)=1 h. was 1.6 g mol-1 and almost independent of the growth rate, when the rate of methane formation was not limited by the supply of either H2 or CO2. The yield increased to near 3 g mol-1 when H2 or CO2 were limiting. These findings indicate that methane formation and growth are less tightly coupled at high concentrations of H2 or CO2 in the medium than at low concentrations. The physiological significance of these findings is discussed. Summary Lymphapheresis was performed on a patient with Sézary syndrome. The Sézary cells were purified by removing E-rosette-forming and Fc receptor-bearing cells. Antiserum against these purified Sézary cells was raised in rabbits. This antiserum had cytotoxicity against Sézary cells as well as against normal peripheral blood lymphocytes. Absorption was carried out with chronic lymphocytic leukemia (CLL) and normal lymphocytes. The absorbed antiserum maintained cytotoxicity against Sézary cells but lost cytotoxicity against CLL and normal peripheral blood lymphocytes.Indirect immunofluorescence assay showed that the antiserum reacted against purified Sézary cells and a high percentage (66%) of peripheral blood mononuclear cells from five patients with Sézary syndrome. It also reacted against 5.7% of normal lymphocytes, 8% of CLL cells, 5% of the lymphocytes from a patient who had undergone splenectomy, 2% of lymphocytes from a patient with multiple myeloma, 5% of lymphocytes from a hairy cell leukemia patient, and 1% of acute lymphocytic leukemia cells (T cell). The antiserum did not react against thymocytes but reacted against 34.6% of the bone marrow lymphocytes. This unique marker was designated as sigma () antigen. It was suggested that Sézary syndrome may represent proliferation or malignant transformation of normally present antigen-positive lymphocytes.  相似文献

Here we use sound and movement recording tags to study how deep-diving Blainville’s beaked whales (Mesoplodon densirostris) use echolocation to forage in their natural mesopelagic habitat. These whales ensonify thousands of organisms per dive but select only about 25 prey for capture. They negotiate their cluttered environment by radiating sound in a narrow 20° field of view which they sample with 1.5–3 clicks per metre travelled requiring only some 60 clicks to locate, select and approach each prey. Sampling rates do not appear to be defined by the range to individual targets, but rather by the movement of the predator. Whales sample faster when they encounter patches of prey allowing them to search new water volumes while turning rapidly to stay within a patch. This implies that the Griffin search–approach–capture model of biosonar foraging must be expanded to account for sampling behaviours adapted to the overall prey distribution. Beaked whales can classify prey at more than 15 m range adopting stereotyped motor patterns when approaching some prey. This long detection range relative to swimming speed facilitates a deliberate mode of sensory-motor operation in which prey and capture tactics can be selected to optimize energy returns during long breath-hold dives.  相似文献

Résumé Les observations photoniques et ultramicroscopiques concernant les Blastodinium, parasites de Copépodes, étudiés à tous les stades de leur développement intra-intestinal nous ont apporté les résultats suivants:Le Trophocyte subit de profondes transformations nucléaires avant de commencer à se diviser (éndettement nucléolaire et passage des grains ribonucléoprotéiques dans le cytoplasme); les sphères archoplasmiques liées aux noyaux se divisent avant eux et apportent leur contribution dans les synthèses membranaires; la division nucléaire est une pleuromitose simple, sans cinétochores et sans centrioles: on peut observer tous les stades définis de, la mitose, exceptée l'interphase; la métaphase est très difficile à saisir, surtout dans les espèces à mitoses accélérées. Les faisceaux de microtubules contenus dans les plasmodendrites qui perforent les noyaux toujours en mitoses au cours de la sporogenèse, ne sont jamais en contact avec les chromosomes; nous avons entrevu leur rôle dans la cyclose hyaloplasmique. Durant les phénomènes sporogenétiques, un cycle régulier de condensation chromosomique s'effectue, sans perte de DNA. Dans les stades peu condensés, l'arrangement fibrillaire des chromosomes est inorganisé, puis ceux-ci montrent un agencement en séries d'arceaux réguliers qui disparaît dans les stades les plus condensés.L'enveloppe nucléaire joue un rôle prépondérant dans la traction, l'orientation et la ségrégation du matériel chromosomique. Nous avons recherché également une série d'explications concernant le déterminisme des phénomènes de condensation chromatique.Dans cet exemple précis, noyaux et phénomènes mitotiques démontrent parfaitement cette situation intermédiaire des Dinoflagellés, entre Bactéries et Eucaryotes.
Nuclear structures of Blastodinium Chatton (parasitic dinoflagellates)Division and chromatic Condensation
The mitotic mechanism in Blastodinium Chatton has been studied by photonic and electronic microscopy. Divisions of the archoplasmic spheres precede that of the nuclei and their movements are interconnected. Stages of mitosis follow one another without interphase; they are telescoped, which makes the study of the chromosome cycle more difficult. Progressive condensation of nuclei and chromosomes takes place in the various sporogenetic layers within the same parasite. Cytophotometric measurements show that there is no loss of DNA during successive mitosis. The nuclear envelope plays an important part in the traction, orientation and segregation of the chromosomes. The cytoplasmic invagination which perforate the nuclei contain considerable masses of microtubules whose role in the activation of the hyaloplasmic cyclosis and membranous synthesis is suggested.—In longitudinal section, the chromosomes show a fibrillar arrangement in regular arches in the non-condensed stages and irregular arches in more condensed stages, with a lesser diameter.—The type of nuclear division is a very simple pleuromitosis without kinetochores and centrioles; the role of the nuclear envelope in the chromosomal mechanism appears to be preponderant.

Four chloroplast (cp), one mitochondrial (mt), and one ribosomal nuclear (ITS) DNA regions were studied in four artificial and one natural interspecific Passiflora hybrids. The ITS results confirmed their hybrid origin and all mtDNAs were maternally inherited. The same, however, was not true for cpDNA. The four hybrids (three artificial and one natural) derived from species of the Passiflora subgenus showed a cpDNA paternal inheritance, while the one involving taxa of the Decaloba subgenus gave evidence of maternal transmission. These results are of significance for the ongoing studies which are being performed on the molecular evolution of this genus and furnish important background for investigations aimed at clarifying the factors which determine cpDNA inheritance.  相似文献

Fine structure of the midgut and degeneration of the midgut epithelium of the scorpionfly Sinopanorpa tincta (Navás) adults were investigated using light microscopy and scanning and transmission electron microscopy. The results show that the tubular midgut lacks gastric caeca and is composed of an outer longitudinal and an inner circular muscle layer, a basal lamina, an epithelium and a lumen from the outside to inside. A peritrophic membrane was not found in the lumen. A mass of nodules was observed on the surface of the basal lamina. Three types of cells were recognized in the epithelium: digestive, secretory, and regenerative cells. The digestive cells contain irregular-shaped infoldings in the basal membrane and two types of microvilli in the apical membrane. The secretory cells are characterized by irregular shape and large quantities of secretory granules in the basal cytoplasm. The regenerative cells are triangular in shape and distributed only in the nodules. The epithelial cells are degenerated through programmed cell-death mechanisms (apoptosis and necrosis). The type, function, and degeneration of the epithelial cells of the midgut are briefly discussed.  相似文献

Grass (Poaceae) richness patterns across China’s nature reserves   总被引:1,自引:0,他引:1  
Explaining grass richness patterns over broad geographic scales is a central issue of biogeography and macroecology. In this investigation, we document the richness patterns of grasses across China’s nature reserves, and discuss their possible explanations at national and regional scales using grass richness data coupled with information from climatic and topographical variables. Our results suggest that the water- and energy-related variables and elevation range (a surrogate of topographical heterogeneity) represent the primary explanations for the variation in grass richness across China’s nature reserves. Significant relationships were found between energy and all grasses, water and C3 grasses, and the combination of water and energy and C4 grasses at the national scale. Topographical heterogeneity is strongly associated with grass richness across China, whereas climatic constraints to grass diversity vary regionally. In regions of high rainfall, the presence of C4 grasses is determined by annual potential evapotranspiration and sunshine hours; the climatic constraint to C3 grass richness in a large and varied region is sunshine hours; whereas in regions of low soil temperature, the temperature determines the grass richness pattern. Our results also suggest that water variables alone do not represent the primary predictor of grass richness at the regional scale.  相似文献
