Miscanthus is an interesting raw material for pulp production, it is a high yield low maintenance plant with a high cellulose and hemicellulose content. Its semichemical pulp can be beneficial in paper for cardboard production process, which nowadays is usually made from secondary fibers, by increasing the mechanical properties of the paper produced. In this study, the influence of the percentage of NaOH used related to the dry Miscanthus weight, digestion time and refining time on some pulp and paper properties have been studied and compared with pulp obtained from commercial fluting paper (CF). Fiber size distribution of the Miscanthus pulp was found to contain a higher fines (less than 0.2 mm) percentage than the CF pulp. Hand-sheets made from Miscanthus pulp showed better mechanical properties than the ones made with the CF pulp. CMT, RCT and CCT indexes were higher when using 100% Miscanthus pulp or mixtures of Miscanthus and CF pulp. The only property which worsened was Gurley porosity. Of the three operational variables changed, refining time exerts the most significant influence on the pulp and paper properties measured.  相似文献

Recent studies have documented that TGF-beta1 takes part in dental pulp tissue repair. Moreover, dental pulp cells have the potential to differentiate into odontoblast-like cells and produce reparative dentine in this process. However, the molecular mechanisms and potential interactions between TGF-beta1 and dental pulp cells are not clear due to the complexity of the pulp/dentine microenvironment. In this study, we investigated the induction of TGF-beta1 on the dental pulp cells in cell culture, tissue culture and three-dimensional culture patterns. These results demonstrated that TGF-beta1 significantly increased the proliferation of cells and activity of ALPase. Dental pulp cells cultured in the presence of TGF-beta1 formed mineralization nodules. In the organ culture, dental pulp cells treated with TGF-beta1 differentiated into odontoblast-like cells and formed a pulp-dentinal complex; and TGF-beta1 significantly induced synthesis of dentine relative proteins DSPP, DMP-1. The dental pulp cells share some characteristics of the odontoblast, such as a parallel arrangement with columnar form and a unilateral cell process. Together, these data indicate that TGF-beta1 can make dental pulp cells differentiated into odontoblast-like cells and form the pulp-dentinal complex. Moreover, these results suggest that TGF-beta1 is an important regulatory factor in odontoblast differentiation during tooth development and pulp repair.  相似文献

Crude enzymes produced by different strains were used in the production of bleached wheat straw pulp. Pre-treatment with enzymes from Penicillium A10 and Aspergillus L22 at a xylanase dosage of 4 IU/g prior to pulping decreased pulp kappa number by 6.29% and 12.07% respectively as compared to the control. High cellulase activity in crude enzymes has a negative influence on pulping. Xylanase pre-bleaching reduced chlorine charge by 20-30%, or increased final brightness by approximately 4-5% ISO, and improved the pulp strength properties. Xylanase could substitute for alkali extraction in CEH sequence, and be used for treating chemical-bleached pulp, which resulted in higher strength properties for bleached pulp. Modification of bleached pulp with enzymes of 3 IU/g (on xylanase) increased pulp brightness and breaking length by 3-6% ISO and 160-790 m respectively, and decreased post color number and beating degree of pulp by 29-36% and 2.5-5.5 degrees SR respectively, as compared to the original pulp.  相似文献

Both beta-xylanase and beta-xylosidase were purified to homogeneity from a xylose-grown culture of Aureobasidium pullulans. Cellular distribution studies of enzyme activities revealed that beta-xylanase was an extracellular enzyme, during both the exponential and stationary phases, whereas beta-xylosidase was mostly periplasmic associated. The beta-xylanase exhibited very high specificity for xylan extracted from Eucalyptus grandis dissolving pulp, whereas the beta-xylosidase was only active on p-nitrophenyl xyloside and xylobiose. Comparison of kcat/Km ratios showed that the beta-xylanase hydrolyzed xylan from dissolving pulp 1.3, 2.1, and 2. 3 times more efficiently than Eucalyptus hemicellulose B, Eucalyptus hemicellulose A, and larchwood xylan, respectively. The beta-xylosidase exhibited a transxylosylation reaction during the hydrolysis of xylobiose. When applied on acid sulfite pulp, both enzymes released xylose and hydrolyzed xylan to a different extent. Although beta-xylosidase (0.4 U/g pulp) liberated more xylose from pulp than beta-xylanase (4.7 U/g pulp), it was responsible for only 3% of xylan solubilization. Treatment of pulp with beta-xylanase liberated 51.7 microgram of xylose/g and hydrolyzed 10% of xylan. The two enzymes acted additively on pulp and removed 12% of pulp xylan. A synergistic effect in terms of release of xylose from pulp was observed when the enzyme mixture of beta-xylanase and beta-xylosidase was supplemented with beta-mannanase. However, this did not result in further enzymatic degradation of pulp xylan. Both beta-xylanase and beta-xylosidase altered the carbohydrate composition of sulfite pulp by increasing the relative cellulose content at the expense of reduced hemicellulose content of pulp.  相似文献

A high accessibility is an essential prerequisite for a homogeneous substitution of cellulose material. In this study, chemical and enzymatic pretreatments to increase the accessibility of cellulose materials have been investigated. Dissolving pulp has been treated with a monocomponent endoglucanase. Fock's method, a microscale process similar to the viscose process, showed an increase in cellulose yield. Simultaneously, the viscosity decreased. To clarify whether the increase in reactivity was due solely to the decrease in the degree of polymerization, the dissolving pulp was also subjected to acid hydrolysis. At a given viscosity level, the enzymatic pretreated pulp had a higher reactivity than the pulp subjected to acid hydrolysis. To achieve 100% reactivity, according to Fock, the acid-treated pulp showed a lower molecular weight compared to the enzymatic-treated pulp. A monocomponent endoglucanase can thus be used to increase the reactivity and accessibility of dissolving pulp in the viscose process.  相似文献

Studies on mediators of manganese peroxidase for bleaching of wood pulps   总被引:3,自引:0,他引:3  
In order to enhance the bleaching effect of manganese peroxidase (MnP), unsaturated fatty acids, thiol-containing compounds and various other organic compounds were applied in pulp bleaching experiments with MnP. Thiol-containing compounds did not improve the pulp bleaching effect by MnP. Some unsaturated fatty acids, linoleic acid and linolenic acid provided a better pulp bleaching effect than Tween 80. The correlation between the number of C=C bonds in a fatty acid and its pulp bleaching effect was also investigated. The MnP pulp bleaching capability was shown to depend on the carboxylic acid used. A combination of Tween 80 and a carboxylic acid resulted in higher pulp brightness than that obtained with Tween 80 alone. A laccase mediator, 3-hydroxy-1,2,3-benzotriazin-4(3H)-one, could also enhance the MnP pulp bleaching effect.  相似文献

Pretreatment of hemp pulp with xylanase was investigated. Unbleached hemp pulp was treated with commercial xylanase, and then bleached with hydrogen peroxide. Control pulp bleached with out xylanase was compared with xylanase bleached pulp. Application of xylanase was found to have a positive effect on followed peroxide stage in terms of low kappa number and high brightness of pulp.  相似文献

Pulp regeneration using human dental pulp stem cells (hDPSCs) maintains tooth vitality compared with conventional root canal therapy. Our previous study demonstrated that preameloblast-conditioned medium (PA-CM) from murine apical bud cells induces the odontogenic differentiation of hDPSCs and promoted dentin formation in mouse subcutaneous tissue. The purpose of the present study is to evaluate the effects of PA-CM with human whole pulp cells on pulp regeneration in an empty root canal space. Human pulp cells were seeded in the pulp cavities of 5 mm-thick human tooth segments with or without PA-CM treatment, and then transplanted subcutaneously into immunocompromised mice. In the pulp cell-only group, skeletal muscle with pulp-like tissue was generated in the pulp cavity. A reparative dentin-like structure with entrapped cells lined the existing dentin wall. However, in the PA-CM-treated group, only pulp-like tissue was regenerated without muscle or a reparative dentin-like structure. Moreover, human odontoblast-like cells exhibited palisade arrangement around the pulp, and typical odontoblast processes elongated into dentinal tubules. The results suggest that PA-CM can induce pulp regeneration of human pulp cells with physiological structures in an empty root canal space.  相似文献

对香蕉果实贮藏过程中内、外果肉相关生理生化指标及细胞结构的变化进行系统的观察分析,结果显示:(1)在贮藏初期香蕉果实内果肉的硬度小于外果肉,在贮藏过程中的同一时期,均表现出内果肉硬度小于外果肉,且内果肉硬度较外果肉先降到零;(2)在贮藏初期内果肉中多聚乳糖醛酸酶(PG)和淀粉酶的活性均高于外果肉,随着贮藏时间的延长,酶活性在内、外果肉均表现出不断增加,且这两种酶活性在内果肉中早于外果肉达到最高值,但其在内果肉中的最高值均略低于外果肉的最高值;而淀粉含量却相反,在贮藏初期内果肉中淀粉含量低于外果肉,且在贮藏过程中的降解速率高于外果肉;(3)超微结构显示,香蕉果实内果肉中淀粉粒和细胞壁结构的降解均早于外果肉.研究表明,香蕉果实的软化首先由内果肉细胞降解开始,并呈放射状向外逐步延伸.  相似文献

Graphiumputredinis, Trichodermaharzianum and fusant were used in the present study to produce extracellular xylanases, an important industrial enzyme used in pulp and paper industry produced in a minimal medium supplemented with oat spelt xylan (1%, w/v) pH 7.0 at 27+/-2 degrees C. The enzyme was purified to homogeneity by DEAE-Cellulose and Superdex 75 FPLC column, respectively. The enzyme was found to be a monomer as determined by SDS gel electrophoresis. The optimum pH and temperature for purified G. putredinis, T. harzianum and fusant xylanases were 5.0-6.0 and 50-70 degrees C, respectively. Pretreatment of paper pulp with G. putredinis, T. harzianum and fusant xylanases decreased pulp kappa number. Xylanases particularly that of fusant at 5 IU/g pulp concentration and 1.5% pulp consistency at 60 degrees C for 18 h followed by EDED process yielded good quality paper from waste paper pulp. A significant increase in pulp brightness and improvement in various pulp properties, viz. burst capacity, thickness and bulkness of the treated pulp were observed in comparison to the conventional chemical bleaching. Easy purification and high stability of these enzymes makes it amicable for industrial applications.  相似文献

Eight groups of citrus varieties grown in Spain were included in this study. Citrus pulp composition was (dry matter basis) 60–65% peel, 30–35% segment pulp and 0–10% seeds. Citrus seeds had a high percentage of protein, ether extract and crude fibre. Segment pulp contained slightly less fibre than citrus pulp. The proximate composition of citrus pulp was similar to that of the peel fraction and did not differ from previous results. Significant differences in fibre and acid detergent lignin of the dried citrus pulp samples were found between varieties.The drying process was studied. Temperatures over 130°C and the addition of calcium hydroxide affected some fractions of the pulp, mainly by increasing ash and fibre contents.  相似文献

Applying a complex of modern microangiological techniques, the organization of the blood bed in the incisor and molar pulp has been studied in the white rat mandible. Production of the dental tissue components in the zones of dentinogenesis is ensured by an increased density of the capillary and venular bed of the pulp, a tight connection of the capillaries with predentin. The morphometric data obtained make it possible to prognosticate a high intracapillary pressure and the hydratation degree of the interstitial gel in the incisor pulp, which is close to maximal figures. Differences in angio-architectonics of the incisor pulp and the molar pulp depend on the peculiarities of the histostructure and functional state of the pulp, on the geometry of the dental cavity and the radicular canals. The pulp blood bed possesses a complex apparatus that ensures the blood stream regulation (duplication of the nutrition sources, arteriolar and venular area, various types of anastomoses, precapillary sphincters and others).  相似文献

A cellulase-free xylanase produced by Bacillus subtilis C 01 from wheat bran under solid-state cultivation was tested for its efficacy in biobleaching of raw banana fibre and banana pulp obtained through a mechanical pulping process. Banana pulp samples treated with crude xylanase (450 nkat g−1 pulp) resulted in a 19.6% increase in the brightness as compared to untreated pulp. The presence of chromophores, hydrophobic compounds and an increased reducing sugar (10.79 mg g−1 pulp) quantity in the bleached solution after enzymatic treatment indicated the removal of materials that were absorbed at 237 nm from the banana pulp.  相似文献

Despite the frequent use of primary dental pulp cells in dental regenerative research, few systematic studies of stemness for osteogenic and dentinogenic differentiation of human adult pulp cells have been reported. To investigate the stemness of human adult dental pulp cells, pulp tissues were obtained from extracted third molars and used as a source of pulp cells. In FACS analysis and immunophenotyping, the general mesenchymal stem cell markers CD44, CD90, and CD146 were highly expressed in early passages of the pulp cell culture. The stem cell population was dramatically decreased in an expansion culture of human dental pulp cells. When pulp cells were treated with additives such as β-glycerophosphate, ascorbic acid, and dexamethasone, nodule formation was facilitated and mineralization occurred within 2 weeks. Expression of osteogenic markers such as alkaline phosphatase, osteocalcin, and osteonectin was relatively low in undifferentiated cells, but increased significantly under differentiation conditions in whole passages. Dentinogenic markers such as dentin sialophosphoprotein and dentin matrix protein-1 appeared to decrease in their expression with increasing passage number; however, peak levels of expression occurred at around passage 5. These data suggested that stem cells with differentiation potential might exist in the dental pulp primary culture, and that their phenotypes were changed during expansion culture over 8-9 passages. Under these conditions, a dentinogenic population of pulp cells occurred in limited early passages, whereas osteogenic cells occurred throughout the whole passage range.  相似文献

To clarify the role of excreted extracellular enzymes during long-term incubation in a pulp biobleaching system with white rot fungi, we developed a cultivation system in which a membrane filter is used; this membrane filter can prevent direct contact between hyphae and kraft pulp, but allows extracellular enzymes to attack the kraft pulp. Phanerochaete sordida YK-624 brightened the pulp 21.4 points to 54.0% brightness after a 5-day in vitro treatment; this value was significantly higher than the values obtained with Phanerochaete chrysosporium and Coriolus versicolor after a 7-day treatment. Our results indicate that cell-free, membrane-filtered components from the in vitro bleaching system are capable of delignifying unbleached kraft pulp. Obvious candidates for filterable reagents capable of delignifying and bleaching kraft pulp are peroxidase and phenoloxidase proteins. The level of secreted manganese peroxidase activity in the filterable components was substantial during strain YK-624 in vitro bleaching. A positive correlation between the level of manganese peroxidase and brightening of the pulp was observed.  相似文献

This paper describes pathological alterations in the permanent canines (tusks) of four male wild boars. The mandibular tusks of all individuals, and also some of the maxillary tusks, exhibited an extended enamel hypoplasia in their apical portion, denoting an impairment of secretory ameloblast function. Moreover, the pulp cavities of the mandibular tusks were exposed through cleft-like openings in the wear (whetting) surfaces of the teeth. Presence of a plug of reparative dentine within the pulp cavity was observed in a split mandibular tusk of one individual. In a second boar, the presence of a plug of reparative dentine within the pulp cavity of the mandibular tusks was indicated radiographically. These findings suggested a reparation process attempting to demarcate a vital, apical pulp portion from a necrotic, incisal portion. The enamel hypoplasias observed in the teeth are regarded to be sequelae of the pulp inflammation caused by bacterial invasion in the mandibular tusks. Most likely, bacterial invasion of the dental pulp occurred through the cleft-like openings in the tusks whetting surfaces, the openings resulting from insufficient formation of secondary dentine. It is, however, also conceivable that pulp inflammation and partial necrosis occurred as a consequence of bacterial invasion of patent dentinal tubules, and that the openings in the whetting surface developed secondarily as a consequence of the pulp changes. One mandibular tusk showed marked signs of resorption apically, suggesting a spread of the inflammation from the pulp into the periodontium.  相似文献

Paper sheets from olive tree wood pulp obtained by soda, sulphite or kraft pulping were studied to examine the influence of pulp beating on properties of the paper sheets.Paper sheets from kraft and sulphite pulps exhibited the highest resistance, and sulphite pulp the highest brightness. Soda pulp required more intensive beating than did kraft or sulphite pulps; in fact, the PFI beater had be operated at a 40–50% higher number of beating revolutions to obtain soda pulp with 70–80° SR.The breaking length, stretch, burst index and tear index of paper sheets obtained from kraft pulp, beaten to a Shopper–Riegler index of 70–80° SR were 20–30%, 30–50%, 50–60% and 15–35% higher, respectively, than those of sheets obtained from soda pulp.  相似文献

The recombinant xylanase B (XynB) of Thermotoga maritima MSB8 was found to be highly specific towards xylans and exhibit very low activity towards carboxymethylcellulose in previous study. XynB was thermostable at neutral to alkaline pH region at 90°C and retained more than 90% activity after 1 h over the pH range of pH 6.1 to 11.1. The suitability of XynB for use in the biobleaching of wheat straw pulp was investigated. Pretreatment of the pulp with XynB resulted in a substantial improvement in the bleachability of wheat straw pulp. When XynB at 10 U g−1 was used to treat wheat straw pulp, it reduced pulp kappa number by 1.1 point, enhanced pulp brightness by 5.5% (% ISO) and improved other pulp properties, such as tensile index and breaking length. Biobleaching of wheat straw pulp with XynB saved active chlorine up to 34.5% while still maintaining the brightness at the control level. Besides, pretreatment of pulp with XynB was also effective at an alkaline pH as high as pH 10.1. This is the first report on the potential application of XynB from T. maritima MSB8 in the pulp and paper sector.  相似文献
