Two Sargassum species (S. baccularia and S. polycystum) collected from Teluk Kemang and Cape Rachado, Port Dickson, Negeri Sembilan, Malaysia, which are alike in morphology except for the rhizoidal system and vesicles, were characterised using random amplified polymorphism DNA (RAPD). The genomic DNA of both species was isolated from the leaves using a modified CTAB method. Four random primers, that is, OPA2, OPA3, OPA4 and OPA13, successfully amplified the DNA. The polymorphisms generated by these four primers were analysed using the Dice Coefficient of Similarity and cluster analysis was carried out using GelCompar II Version 2.0 (Applied Maths, Kortrijk, Belgium) based on UPGMA. DNA analysis showed that three primers were able to differentiate the two species. Morphological analysis using Principal Components Analysis (PCA) and discriminant function analysis supported the molecular data. Both species are characterised by heavily muricate main branches, oblong-lanceolate leaves with dentate margins and discoid holdfasts and spherical vesicles; both are dioecious. The only difference is that S. polycystum has secondary holdfasts transformed into stolons. This last characteristic is therefore a very important criterion and may contribute to the difference shown by DNA analysis.  相似文献

The random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) was used for the molecular characterisation and identification of Sargassum spp. A total of 17 samples of Sargassum (Sargassaceae, Fucales) was obtained from various localities around Peninsular Malaysia and Singapore. On the basis of morphological characteristics, the samples were tentatively grouped into five species: Sargassum baccularia, S. glaucescens, S. oligocystum, S. polycystum and S. siliquosum. By RAPD-PCR, five of 31 random primers tested generated reproducible amplification products, and polymorphic loci were detected by four of them (OPA02, OPA03, OPA04, OPA13). The RAPD-PCR profiles did not correlate with the morphological grouping into five species and extensive variation was detected between different isolates of the same species. A 450 base pair fragment generated using OPA13 was detected in 12 of 17 samples of Sargassum. This fragment was also present in profiles from Turbinaria (Sargassaceae). This study suggests that RAPD-PCR is useful in discriminating individual samples of the genus Sargassum and in developing fingerprints for them.  相似文献

Four species of brown seaweeds, namely Sargassum baccularia, Sargassum binderi, Sargassum siliquosum and Turbinaria conoides, harvested from Port Dickson, Negeri Sembilan, Malaysia were analysed for ash content, alginate yield and alginate properties. Seaweeds calcined at 450°C were found to have low amount of non-combustible residue as these were not contaminated by calcareous animals. Alginate was extracted from these seaweeds by two methods: hot and cold. In the hot method, the storing time was 3 h and the processing temperature was 50°C, whilst in the cold method, the sample was stored overnight at room temperature. Higher yield of alginate was obtained by the hot method compared to the cold method, but alginate extracted by the cold method gave higher molecular weight. In the hot method, 49.9% of alginate was extracted from S. siliquosum, followed by T. conoides (41.4%), S. binderi (38.9%) and S. baccularia (26.7%). Alginate extracted from T. conoides has an average molecular weight, M w, of 8.06 × 105 g mol−1, whereas alginate from S. siliquosum was the lowest in M w (4.81 × 105 g mol−1) when the extraction was done at room temperature. Alginate extracted from S. baccularia was found to be very heat-sensitive. Its M w has dropped more than 83%, from 7.52 × 105 to 1.23 × 105 g mol−1, when the extraction temperature was raised. The effect of heat on the extent of depolymerisation of the alginate molecule of the other three brown seaweed species was less significant, with decrease in molecular weight ranging between 13% and 16%.  相似文献

Ten species of Sargassum (Sargassaceae, Phaeophyceae) were found along the Gulf of Thailand. Morphological characteristics of Sargassum baccularia (Mertens) C.A. Agardh, S. binderi Sonder, S. cinereum J.G. Agardh, S.crassifolium J.G. Agardh, S. longifructum Tseng et Lu, S. oligocystum Montagne, S. polycystum C.A. Agardh, S. siliquosum J.G. Agardh, S. swartzii (Turner) C.A. Agardh and one unidentified species were examined and are described in detail. The most common species were S. polycystum distributed widely in almost all the study sites, S. crassifolium restricted to Prachuap Khirikhan Province, S. longifructum restricted to Chumphon Province, S. siliquosum restricted to Surat Thani Province and one unidentified species restricted to Songkhla Province. Three species (S. cinereum, S. longifructum and S. swartzii) are new records for the algal flora of Thailand. Five species (S. baccularia, S. cinereum, S. longifructum, S. polycystum and the unidentified species) belong to the section Zygocarpicae (J.G. Agardh) Setchell.  相似文献

Genomic DNA was extracted from 13 samples of Sargassum polycystum and S. siliquosum collected from various localities around Peninsular Malaysia and Singapore by using four different extraction methods. The yields and the suitability of the DNA to be used as template for the polymerase chain reaction (PCR) was compared. DNA samples were subjected to PCR analysis by using random primers. Only DNA samples that were extracted using the CTAB method were successfully amplified by random amplified polymorphic DNA (RAPD)-PCR. Five of 31 random primers (OPA02, OPA03, OPA04, OPA13 and OPM10) tested amplified sequences of DNA from the DNA samples. Reproducible, amplified products were obtained using these primers and showed some potential to be useful in discriminating individual samples within the genus, in determining relationships between species within a genus and in developing individual fingerprints for individual samples.  相似文献

Sargassum muticum is important in maintaining the structure and function of littoral ecosystems, and is used in aquaculture and alginate production, however, little is known about its population genetic attributes. In this study, random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers were used to investigate the genetic structure of four populations of S. muticum and one outgroup of S. fusiforme (Harv.) Setchell from Shandong peninsula of China. The selected 24 RAPD primers and 19 ISSR primers amplified 164 loci and 122 loci, respectively. Estimates of genetic diversity with different indicators (P%, percentage of polymorphic loci; H, the expected heterozygosity; I, Shannon’s information index) revealed low or moderate level of genetic variations within each S. muticum population, and a high level of genetic differentiations were determined with pairwise unbiased genetic distance (D) and fixation index (F ST ) among the populations. The Mantel test showed that two types of matrices of D and F ST were highly correlated whether from RAPD (r = 0.9706, P = 0.009) or ISSR data (r = 0.9161, P = 0.009). Analysis of molecular variance (AMOVA) was conducted to apportion the variations among and within the S. muticum populations. It indicated that variations among populations were higher than those within populations, being 55.82% verse 44.18% by RAPD and 55.21% verse 44.79% by ISSR, respectively. Furthermore, the Mantel test suggested that genetic differentiations among populations were related to the geographical distances (r > 0.6), namely, conformed to the IBD (isolation by distance) model, as expected from UPGMA (unweighted pair group method with arithmetic averages) cluster analysis. On the whole, the high genetic structuring among the four S. muticum populations along the distant locations was clearly indicated in RAPD and ISSR analyses (r > 0.9, P < 0.05) in our study.  相似文献

Genetic variation of four populations of Sargassum thunbergii (Mert.) O. Kuntze and one outgroup of S. fusiforme (Harv.) Setchell from Shandong peninsula of China was studied with random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers. A total of 28 RAPD primers and 19 ISSR primers were amplified, showing 174 loci and 125 loci, respectively. Calculation of genetic diversity with different indicators (P%, percentage of polymorphic loci; H, the expected heterozygosity; I, Shannon’s information index) revealed low or moderate levels of genetic variations within each S. thunbergii population. High genetic differentiations were determined with pairwise Nei’s unbiased genetic distance (D) and fixation index (F ST ) between the populations. The Mantel test showed that two types of matrices of D and F ST were highly correlated, whether from RAPD or ISSR data, r = 0.9310 (P  = 0.008) and 0.9313 (P = 0.009) respectively. Analysis of molecular variance (AMOVA) was used to apportion the variations between and within the S. thunbergii populations. Matrix polysaccharide from the brown algae Sargassum turbinarioides collected in the coastal waters of Nosy Be (Madagascar) in the Indian Ocean was isolated and its structure was studied by 1H-NMR spectroscopy, FT-IR, SEC-MALLS and HPAEC. An alginate with a molecular weight of 5.528 × 105 g mol−1 was identified as sole polysaccharide. Values of the M/G ratio, F GG, F MM and F GM (or F GM) blocks were measured at respectively 0.94, 0.39, 0.36 and 0.25 and compared with those of alginates from other Sargassum species. This sodium alginate appeared similar to some of the other Sargassum alginates with M/G < 1, high values of homopolymeric blocks (η < 1) and significant polyguluronic block content.  相似文献

The seasonality of standing crop of a Sargassum bed was investigated by conducting monthly sampling from February 1988 to July 1989. Environmental parameters of water movement, salinity, number of daytime minus tides, and water temperature were also measured. An intra-annual pattern of variation in standing crop of Sargassum crassifolium, S. cristaefolium, S. oligocystum, and S. polycystum was observed. Standing crop was generally lowest in February, March, April, or May, and highest in November through January. Sargassum accounted for about 35 to 85% of the monthly algal standing crop of the bed, and the observed variation in overall standing crop of the bed generally reflected the standing crop of Sargassum. The seasonality of the standing crops of the associated algal divisions also followed an annual cycle, but their maximum and minimum standing crops did not coincide with those of Sargassum. Individually, as well as collectively, the standing crops of the Sargassum spp. were poorly correlated with the environmental factors observed.  相似文献

The effects of temperature and nutrient availability on seasonal abundance were compared among Sargassum berberifolium J. Ag., S. polycystum C. Ag., S. siliquosum J. Ag., and S. sandei from a reef in southern Taiwan. Growth temperature limits and optimum were variable between species and between developmental stages. Growth temperature ranges agree with Sargassum periodicity except S. sandei. Sargassum siliquosum can tolerate high temperature as indicated by higher optimum temperature and tolerance limits. Temporal changes in tissue nutrient content and alkaline phosphatase activity and the results from nutrient enrichment bioassay show that S. sandei, S. berberifolium, and S. polycystum are P limited in the early growth period and then N limited, whereas S. siliquosum showed a contrasting trend, reflecting the type of nutrient limitation is variable by time and between species. A smaller nutrient threshold for maximum growth in S. berberifolium, S. polycystum, and S. siliquosum than in S. sandei and higher growth rate in S. berberifolium and S. polycystum indicate the higher growth vigor and nutrient utilization efficiency, the higher abundance. High N reserves in S. berberifolium and S. polycystum exposed to elevated seawater N in the early growth period supported the subsequent growth. The multiple regression analysis indicates that percent cover of S. siliquosum increased with increasing water temperature and dissolved inorganic nitrogen concentrations, whereas that of S. polycystum decreased with increasing water temperature. In conclusion, seasonal dynamics of Sargassum spp. from southern Taiwan are attributable to species and temporal variations in temperature limitation and nutrient utilization strategy.  相似文献

The antioxidant activity of eight edible species of Malaysian North Borneo seaweeds obtained from Sabah waters (Kudat, Tanjung Aru and Semporna) consisting of three red seaweeds (Eucheuma cottonii, E. spinosum and Halymenia durvillaei), two green seaweeds (Caulerpa lentillifera and C. racemosa) and three brown seaweeds (Dictyota dichotoma, Sargassum polycystum and Padina sp.) were determined. Methanol and diethyl ether were used as extraction solvent. The antioxidant activities were determined by two methods, TEAC (trolox equivalent antioxidant capacity) and FRAP (ferric reducing antioxidant power) assays. The total phenolic content of the extract was determined according to the Folin–Ciocalteu method and results were expressed as phloroglucinol equivalents. The methanolic extracts of green seaweeds, C. lentillifera and C. racemosa, and the brown seaweed, S. polycystum showed better radical-scavenging and reducing power ability, and higher phenolic content than the other seaweeds. The TEAC and FRAP assays showed positive and significantly high correlation (R 2 = 0.89). There was a strong correlation (R 2 = 0.96) between the reducing power and the total phenolic content of the seaweeds methanolic dry extracts. These seaweeds could be potential rich sources of natural antioxidants.  相似文献

Ovine pulmonary adenocarcinoma (OPA) and Maedi–Visna (Maedi) are two chronic respiratory diseases of retroviral origin which occur worldwide. It is known that different host genetic factors influence the outcome of viral infections. To determine if variation in the Mhc-DRB1 gene was associated with progression to these ovine diseases, sheep lungs with and without OPA and Maedi lesions were collected. A sequence-based method was applied and 40 different alleles were detected in the sample analysed. In the allele-by-allele association analysis, allele DRB1*0325 had a significant association with susceptibility to Maedi (P = 0.045). For OPA, DRB1*0143 and DRB1*0323 were significantly associated with susceptibility (P = 0.024 and P = 0.029), and allele DRB1*0702 was significantly associated with resistance (P = 0.012). Based on these results, the Mhc-DRB1 alleles were classified by effect in three categories—susceptible (S), resistant (R) and neutral (N)—and animals were reassigned the genotypes as S/S, S/R, S/N, R/R, R/N and N/N. In a second analysis, penalised logistic regression models including a flock effect were run. In Maedi, significant association was detected for the N/S heterozygote (P = 0.0007), but not for the S/S homozygote, probably as a result of the low number of S/S animals. In OPA, association was detected for both the S/S and R/R homozygotes (P = 0.005 and P = 0.047). This allele grouping method may be applied in association studies with highly variable genes. This is the first study demonstrating significant associations between sheep Mhc-DRB1 alleles and susceptibility to OPA and Maedi. Therefore, both diseases are suitable candidates for more comprehensive genetic studies.  相似文献

There have been limited reports on molecular sex markers for macroalgae. We report the use of random amplified polymorphic DNA analysis (RAPD) to identify molecular sex markers for Gracilaria changii (Xia et Abbott) Abbott, Zhang et Xia. Two DNA extraction methods were used: a modified CTAB and phenol-chloroform combination method and the DNeasy Plant Mini Kit. The CTAB and phenol-chloroform method gave the best yield of DNA in quality and quantity and is suitable for larger-sized specimens like G. changii. Sixty-nine RAPD primers were screened to search for sex-linked DNA markers for G. changii, and only one sex-linked marker (716 bp) was identified using OPA 18. RAPD was also used to investigate the molecular characteristics of the three life-stages (male, female, tetrasporophyte) of G. changii. Seven (OPA7, OPA18, S14, S61, S64, S75 and S76) out of the 69 primers showed polymorphism and were selected for interpopulation analysis for DNA isolated from 23 samples collected from Morib and Sungai Pulai in Malaysia. The combination of data produced by the seven primers generated a dendrogram that grouped the specimens into different clades according to their sex and life-stage using the unweighted pair group and arithmetic averages (UPGMA) method. The main objective of this investigation was to examine physiological changes occurring within the olive flounder, Paralichthys olivaceus, in the presence of continuous stress factors. The experiment gave continuous stress, and each stress step is as follows: water level reduction (S1), selection process (S2), confinement (S3), air exposure (S4), transportation (S5), and storing in volume 0.5-t (S6) and 50.0-t (S7) rearing tanks for 24 h after transportation. The cortisol concentration showed a trend to continuously rise in response to consecutive stress from transportation from 7.4 ± 0.6 ng/ml in an experimental opening. The concentration showed the highest level, 25.3 ± 4.4 ng/ml, after confinement stress. Glucose concentration in S3 and S4 were increased significantly to 71.0 ± 13.0 and 78.7 ± 7.0 mg/dl, respectively (P < 0.05). Lactic acid was increased significantly from 0.5 ± 0.1 mM (S0) to 9.0 ± 1.2 mM (S4), but it did not recover until 24 h (7.0 ± 1.0 mM) after transportation (S6 and S7) (P < 0.05). Some contents displayed recovery within 24 h, but a longer time would be required for normal physical metabolism to resume after fish experienced stress. Therefore, it would be endangering the survival of this species if the transport were repeated within a 24-h period.  相似文献

Genetic variation within the Murina species group, which includes S. murina, S. gilberti, S. leucopus, S. dolichura and S. archeri, was examined through analyses of complete 12S rRNA, partial control region mitochondrial DNA sequences and partial omega-globin nuclear DNA sequences. Sminthopsis butleri was found to be an additional member of the Murina group, and appears to be most closely related to S. leucopus rather than the morphologically similar S. archeri. This latter species appears to be the most divergent member of the group, and there is a possible sister relationship between S. murina and S. gilberti, as suggested by previous allozyme evidence. It appears that the systematic affinities of the taxonomically problematic northeastern Queensland populations of both S. murina and S. leucopus and a disjunct population of S. gilberti (from the Western Australia/South Australia border) are indeed with those respective species; although each appears to belong to a distinct morphological and genetic lineage. A specimen of S. leucopus from Queensland was found to be as divergent from each of the southeastern Australian S. leucopus subspecies as they are from each other, suggesting that this northern population of S. leucopus may also warrant recognition as a distinct taxon. Specimens of S. murina murina were found to be genetically divergent from each other, and this subspecies appears to be paraphyletic, as suggested by previous morphological evidence. * This paper is the second part of a series dealing with the systematics and evolution of the dasyurid marsupial genus Sminthopsis. The actions of two novel diselenide-bridged bis(porphyrin)s (1 and 2) on Staphylococcus aureus growth was investigated by microcalorimetry at 37.00°C, compared with that of Na2SeO3. Differences in their capacities to inhibit the growth metabolism of S. aureus were observed. By analyzing the power–time curves, crucial parameters such as the rate constant of bacterial growth (k), inhibitory rate (I), and generation time (t G) were determined. The growth rate constant (k) of S. aureus (in the log phase) in the presence of the drugs decreased with increasing concentrations of the drugs regularly. The relationship of k and c is nearly linear for diselenide-bridged bis(porphyrin) 2. The sequence of the antibacterial activities of these selenium compounds tested was 2 > 1 > Na2SeO3.  相似文献

Trichuris spp. infect the majority of captive primate species along with an estimated 1049 million people worldwide, making it an important zoonosis [Stephenson, L. S., Holland, C. V., & Cooper, E. S. Parasitology, 121(Suppl.), S73–S95, 2000]. We investigated the efficacy of methods used to evaluate the prevalence of Trichuris spp. in 2 groups (n = 12) of socially housed Abyssinian colobus (Colobus guereza kikuyensis) at Paignton Zoo Environmental Park and the factors that may affect density. We collected individual and group fecal samples over 6 mo and estimated burden (egg counts/g of feces) of Trichuris spp. via the McMaster technique. Shedding was significantly higher in the afternoon than in the morning (matched-pairs t-test: t [5] = −4.46, p < 0.01) and in dominant adult male colobus (Spearman rank: r [5] = −0.94, p < 0.01; age: r [5] = 0.89, p < 0.05). Parasitological studies of zoo-housed primates can be a useful tool to explore factors that may affect burdens of Trichuris spp. in them.  相似文献
