Hairy root cultures of Crambe abyssinica were obtained through infection of leaves with two wild-type agropine strains of Agrobacterium rhizogenes. The efficiency of transformation was about 16 %. The presence of T-DNA from A. rhizogenes in the hairy roots genome was confirmed by PCR using specific primers for rolB and rolC genes. Selected clones of hairy roots and non-Agrobacterium induced roots from sterile cultures were used for analyses of acyl-lipids. The total amount of acyl-lipids per mg of dry weight was similar in both the non-Agrobacterium induced roots and the hairy roots in good physiological condition, and ranged from 38 to 53 nmol. However, in the clones which showed symptoms of ageing, the lipid content was severely reduced. Also the lipid composition of hairy roots appears to be similar to the composition of non-transformed roots. Polar lipids were the dominant class of lipids in both types of roots (about 75 %). Furthermore, we found diacylglycerols, free fatty acids (FFA), triacylglycerols, sterol esters, and an unidentified lipid class. The dominant fatty acids in the lipids of both types of roots were α-linolenic acid, palmitic acid, and linoleic acid (over 12 % of total FA). Among the lipids of both hairy roots and non-Agrobacterium induced roots of C. abyssinica, an unidentified FA was found (over 16 % of total FAs). The present study is the first example of establishment of hairy roots cultures of C. abyssinica. It also includes the first analysis of the lipids in hairy roots and non-Agrobacterium induced roots of this species.  相似文献

Analysis of extracts obtained from shoots, roots and exudates of Brassica alba revealed the presence of 3,5,6,7,8-pentahydroxy-4'-methoxy flavone in shoots, as well as 2',3',4',5',6'-pentahydroxy chalcone and 3,5,6,7,8-pentahydroxy flavone in roots and exudates. Apigenin was also found in the shoots and roots, but not in the root exudates.  相似文献

Auxin binding onto membrane fractions of primary roots of maize seedlings has been demonstrated using naphth-1yl-acetic acid (NAA) and indol-3yl-acetic acid (IAA) as ligands. This binding is compared with the already well characterized interaction between auxins and coleoptile membranes. The results indicate that while kinetic parameters are of the same order for root and coleoptile binding, a number of differences occur with respect to location in cells and relative affinity. Nodulin gene expression is an integral and highly specific part of the formation of nitrogen-fixing nodules on the roots of leguminous plants. Dependent on the time of expression during root nodule development, nodulin genes can be divided into early and late nodulin genes. A brief overview of the functions assigned to early and late nodulins is presented. We hypothesize that nodulin genes originate from regular plant genes that evolved to fit the regulatory and/or physiological constraints of symbiotic nitrogen fixation. Data on nodulins and nodulin genes, nodulation taxonomy and nodule development are evaluated in the light of this hypothesis.  相似文献

The direction of root growth can be studied by analyzing the trajectories of roots growing in soil. Both the primary seminal root and nodal roots of maize attain a preferred, or liminal, angle of growth that deviates from the vertical. These roots are said to be plagiogravitropic. Experiments using plants grown in soil-filled boxes revealed that the primary seminal root is truly plagiogravitropic. It shows both positive and negative gravitropism in response to gravity stimuli and tends to maintain its direction even after growing around obstacles. These are experimental results suggesting that plagiogravitropic growth is controlled by internal factors. The orientation of the grain affects the establishment of the liminal angle of the primary seminal root, and both the position of their node of origin and the root diameter are closely related to the plagiogravitropic behaviour of nodal roots. Several external factors are also known to influence plagiogravitropism. Low soil water content causes a decrease in the angle of growth and soil mechanical resistance suppresses the gravitropic curvature. Plagiogravitropic behaviour of both seminal and nodal roots plays a significant role in shaping the root system.  相似文献

Summary The net uptake and movement of radioactivity by 12-mm root segments of Zea mays have been studied as a function of time at 5, 15 and 25° C. Segments were supplied with an agar donor block containing 1 M IAA-1-14C or IAA-2-14C continuously or for a limited period of time (pulse-labelling). In the latter case the original donor block was replaced either by a plain agar block or by one containing 1 M unlabelled IAA. Receiver blocks were placed at the other end of the segments.The net uptake of radioactivity from the donor block at 15° C was greater at the basal end than at the apical end of the segment. At 5 and 15° C, the net uptake from a basal donor was virtually linear with time but at 25° C the rate of net accumulation decreased after about 10 h. Decarboxylation of IAA undoubtedly occurred at 15 and 25° C when the concentration in the tissue attained a high value.An acropetally polarised movement of radioactivity into the receiver blocks occurred regardless of whether the results were based on the actual amounts of radioactivity in the receiver block, or on the amounts in the receiver block expressed as a percentage of the net total radioactivity accumulated from the donor block. Only one radioactive substance was present in the receiver block and it ran to the same Rf as IAA in the isopropanol: ammonium: water solvent system.The amounts of radioactivity moving into that part of the root segment at least 6 mm distant from the end in contact with either an apical or a basal donor block were assessed. An acropetal polarity in the movement of radioactivity was observed on the basis of the actual amounts of radioactivity in these distal parts of the segments, but no such polarity was evident when the amounts of radioactivity were expressed as a percentage of the net total accumulated from the donor block. At least 3 radioactive substances were present in the tissue in addition to the substance running to the same Rf as IAA. The distribution of radioactivity in the segment cannot therefore be used to assess the distribution of IAA.Acropetal movement of radioactivity into an apical receiver block is not dependent upon the continued uptake of IAA at the basal end of the segment. No distinct pulses of radioactivity were detected moving through the root segments.Only a small part of the radioactivity in the root segment appears to be located in the polar transport system, while the bulk is not. The polarity found in the movement of the bulk radioactivity within the segment seems to be related to the polarity in IAA uptake from the donor blocks.  相似文献

Summary Nitrate-reducing capability was demonstrated for root segments of red alder and, at more than twice that rate, for alder nodules. Root segments of Douglas-fir failed to reduce nitrate despite various treatments designed to induce such activity. The reported response of Douglas-fir to nitrate fertilizer may be ascribed either to microbial assimilation of nitrate ions with subsequent liberation of ammonium in the soil or to nitrate assimilation by fungi that form mycorrhizae with Douglas-fir roots.  相似文献

Summary The reasons underlying the initial increase and subsequent decrease in the amount of radioactivity in the receiver block at the apical end of a Zea root segment supplied with a basal donor block containing labelled IAA have been investigated.The phenomenon was observed in segments supplied with IAA-1-14C, IAA-2-14C and IAA-5-3H. An acropetal polarity in the movement of radioactivity into the receiver blocks was observed using donor blocks containing IAA-5-3H at concentrations as low as 10-10M.The decrease in the amount of radioactivity in the receiver block begins after 6–8 h of transport at 25° C, and is unaffected by renewal of the donor block every 2 h, or the presence of 2% sucrose in the donor and receiver blocks.The net export of radioactivity into the receiver block at the apical end of the segment virtually ceases after 6–8 h of transport at 25° C, and is not prolonged by the presence of 2% sucrose in the donor and receiver blocks. At 10° C, net export of radioactivity continues for at least the first 50 h of transport, and the amount of radioactivity in a continuously applied receiver block continues to increase over this period.Receiver blocks removed from the apical end of segments after 8 h of transport and placed on planchettes show little or no decrease in the amount of radioactivity they contain as a function of time, in marked contrast to those left in contact with the segment.There is a marked, and metabolically dependent, resorption of radioactivity from the receiver block at the apical end of the segment after about 8 h of transport at 25° C; most of the resorbed radioactivity remains in the apical 2–4 mm of the segment.There is a loss of radioactive CO2 from segments supplied with a basal donor block containing 10-6M IAA-1-14C at 25° C, the emission beginning after 6–8 h of transport. Segments similarly supplied with 10-6M IAA-2-14C did not begin to lose radioactive CO2 until after about 10–12 h of transport.The ability of the segments to transport radioactivity in a polar manner declines with time after they are excised from the root, regardless of whether their cut ends are kept in the intervening period in contact with plain agar blocks, or ones containing unlabelled IAA at 10-6M. By the 6th h after excision at 25° C no transport of radioactivity through the segments and into the receiver blocks could be detected in either the aropetal or basipetal direction.The decrease in radioactivity in the receiver block after transport periods of 6–8 h at 25° C is therefore due to (1) a cessation of net export of radioactivity into the block, and (2) the onset of a metabolically-dependent, net resorption of radioactivity. At this time substantial amounts of radioactive CO2 begin to be evolved from segments supplied with IAA-1-14C, whereas with IAA-2-14C radioactive CO2 is not evolved for a further 4–6 h.  相似文献

Summary A method for identifying of the Y-Body in cells from hair root sheaths by fluorescent staining is described. In 52 normal males the Y-Body is found between 15 and 64%. 2 bodies were observed in 60 out of 5200 cells. In the female controls only rarely a fluorescent body was seen. The method is useful for screening tests in population genetics for identification of male sex-chromosome aberrations.
Zusammenfassung Es wird über eine Methode zur Identifizierung von Y-Körperchen in Haarwurzelzellen mittels Fluorescenzfärbung berichtet. In den Zellen von 52 normalen Männern konnte das Y-Körperchen in einem Prozentsatz zwischen 15 und 64 nachgewiesen werden. 2 Y-Körperchen fanden sich unter 5200 Zellen nur 60mal. Bei normalen Frauen findet sich ein Fluorescenzkörperchen nur äußerst selten. Die Methode eignet sich für Screening-Untersuchungen zum Auffinden Y-chromosomaler Aberrationen.

Usually, roots are looked at as rather perfect osmometers with the endodermis being the root membrane which is equivalent to the plasma membrane of cells. However, this single-equivalent-membrane model of the root does not explain the findings of a variable hydraulic resistance of roots as well as of differences between hydraulic and osmotic water flow and of low reflection coefficients of roots. Recent work with the root pressure probe is reviewed and discussed which indicates that the simple osmometer model of the root has to be extended by incorporating its composite structure, i.e. the fact that there are different parallel pathways for water in the root, namely, the cell-to-cell and apoplasmic path. The new composite transport model of the root readily explains the experimental findings mentioned above. Pressure probe work with roots in which the endodermis was punctured to create an additional parallel path as well as anatomical studies support the model.  相似文献

Phosphofructokinase was partially purified from carrot root extracts. Monovalent cations stimulated carrot phosphofructokinase activity. The enzyme was strongly inhibited by P-enolpyruvate and this inhibition was relieved by NACl or KCl. Pi inhibited the enzyme at pH 7.9 but was stimulatory at pH 6.6.  相似文献

Phenolase activity is not found in germinating spinach embryos, but it appears in the radicles when the vascular tissues have developed, and then increases progressively. Unlike the two phenolases detected earlier in the chloroplasts, the root enzyme is a single protein with higher MW occurring both in 3000 g precipitate and 28 000 g supernatant fractions. The phenolase in 3000 g fraction is not activated by treatment with detergents and trypsin. The enzyme is contained mainly in xylem parenchymatous cells adjacent to primary vessels. It also occurs to a lesser degree in the dermal parts, including epidermis and cortex. Similar tissue-level distribution patterns of this enzyme are also observed in the roots of other angiosperms, especially in Compositae.  相似文献
