In order to characterize the significance of sulfur (S) nutrition in protein expression under iron (Fe)-deficient conditions, gel-based proteomic analysis was performed with the leaves of Brassica napus exposed to S and Fe combined treatments: sufficient in S and Fe (+S/+Fe, control), sufficient S but Fe deprived (+S/?Fe), deprived S but sufficient Fe (?S/+Fe), and deprived S and Fe (?S/?Fe). The resulting data showed that 15 proteins were down-regulated due to production of oxidative damage as indicated by H2O2 and O 2 ?1 localizations and due to leaf chlorosis in leaves in S-deprived leaves either in presence (?S/+Fe) or absence of Fe (?S/?Fe), whereas these down-regulated proteins were well expressed in the presence of S (+S/?Fe) compared to control (+S/+Fe). In addition, two proteins were up-regulated under S-deprived condition in presence (?S/+Fe) and absence of (?S/?Fe) Fe. The functional classification of these identified proteins was estimated that 40 % of the proteins belong to chloroplast precursor, and rest of the proteins belongs to hypothetical proteins, RNA binding, secondary metabolism and unknown proteins. On the other hand, five protein spots from S deprived (?S/+Fe) and ten spots from Fe deprived (?S/?Fe) conditions were absent, whereas they were well expressed in presence of S (+S/?Fe) compared to control plants (+S/+Fe). These results suggest that sulfur nutrition plays an important role in alleviating protein damage in Fe-deficient plants and adaptation to Fe-deficiency in oilseed rape.  相似文献

A pot experiment was conducted to determine the effects of Glomus mosseae inoculation on growth and some biochemical activities in roots and shoots of pepper (Capsicum annuum L. cv. Zhongjiao 105) plants subjected to four levels of NaCl [0 (control), 25 (low), 50 (medium), and 100 (high) mM] for 30 days, after 30 days of establishment under non-saline conditions. In mycorrhizal (M) plants, root colonization varied from 48 to 16 %. M plants had higher root and shoot dry weight and leaf area compared with non-mycorrhizal (NM) plants. Under salinity stress, M plants accumulated higher amounts of leaf photosynthetic pigments as well as soluble sugar, soluble protein, and total free amino acids in roots and shoots than those of NM plants. In contrast, the accumulation of proline was less intense in M plants than NM plants. Salt stress induced oxidative stress by increasing malondialdehyde (MDA) content; however, the extent of oxidative damage in M plants was less compared with NM plants due to G. mosseae-enhanced activity of superoxide dismutase (SOD) and peroxidase (POD). We concluded that inoculation with G. mosseae improved growth performance and enhanced salt tolerance of pepper plants via improving photosynthetic pigments and the accumulation of organic solutes (except proline), reducing oxidative stress, and enhancing antioxidant activities of the SOD-POD system.  相似文献

The heavy metal Cadmium (Cd), added to the water bodies through weathering of rocks and human activities, constitutes one of the major environmental pollutants toxic to plants. This study examines the proteome changes in roots of actively growing Kandelia candel (L.) Druce when challenged with Cd. This mangrove-like species proliferates in estuaries and bays and is a potential choice for phytoremediation of Cd. A total of 53 proteins were up- or down-regulated following a short-term Cd treatment. The identities of the differentially expressed proteins were determined by MALDI-TOF/TOF. Approximately half of the up-regulated proteins are involved in oxidative response, including antioxidant enzymes, enzymes required for glutathione biosynthesis, enzymes in TCA and PPP cycles for generating ATP, NADH and NADPH. These results support the prediction that a prompt antioxidative response is necessary for the reduction of the oxidative stress caused by Cd and set the stage for further investigating of Cd up-regulated proteins in Kandelia candel.  相似文献

Owing to a weak availability in soil, plants have developed numerous morphological, physiological and biochemical adaptations to acquire phosphate (Pi). Identification and characterisation of key genes involved in the initial steps of Pi-signalling might provide clues about the regulation of the complex Pi deficiency adaptation mechanism. A two-dimensional gel electrophoresis approach was performed to investigate proteome responses to Pi starvation in Arabidopsis. Two ecotypes were selected according to contrasting responses of their root system architecture to low availability of Pi. Thirty protein spots were shown to be affected by Pi deficiency. Fourteen proteins appeared to be up-regulated and ten down-regulated with ecotype Be-0, wheras only thirteen proteins were observed as down-regulated for ecotype Ll-0. Furthermore, systematic and opposite responses to Pi deficiency were observed between the two ecotypes. The sequences of these 30 differentially expressed protein spots were identified using mass spectrometry, and most of the proteins were involved in oxidative stress, carbohydrate and proteins metabolism. The results suggested that the modulation of alcohol dehydrogenase, malic enzyme and aconitate hydratase may contribute to the contrasted adaptation strategy to Pi deficiency of Be-0 and Ll-0 ecotypes. A focus on aconitate hydratase highlighted a complex reverse response of the pattern of corresponding spots between the two ecotypes. This protein, also potentially involved in iron homeostasis, was speculated to contribute, at least indirectly, to the root architecture response of these ecotypes.  相似文献

Changes in leaf soluble proteome were explored in 3‐month‐old plants of metallicolous (M) and nonmetallicolous (NM) Agrostis capillaris L. populations exposed to increasing Cu concentrations (1–50 μM) to investigate molecular mechanisms underlying plant responses to Cu excess and tolerance of M plants. Plants were cultivated on perlite (CuSO4 spiked‐nutrient solution). Soluble proteins, extracted by the trichloroacetic acid/acetone procedure, were separated with 2‐DE (linear 4–7 pH gradient). Analysis of CCB‐stained gels (PDQuest) reproducibly detected 214 spots, and 64 proteins differentially expressed were identified using LC‐MS/MS. In both populations, Cu excess impacted both light‐dependent (OEE, cytochrome b6‐f complex, and chlorophyll a‐b binding protein), and ‐independent (RuBisCO) photosynthesis reactions, more intensively in NM leaves (ferredoxin‐NADP reductase and metalloprotease FTSH2). In both populations, upregulation of isocitrate dehydrogenase and cysteine/methionine synthases respectively suggested increased isocitrate oxidation and enhanced need for S‐containing amino‐acids, likely for chelation and detoxification. In NM leaves, an increasing need for energetic compounds was indicated by the stimulation of ATPases, glycolysis, pentose phosphate pathway, and Calvin cycle enzymes; impacts on protein metabolism and oxidative stress increase were respectively suggested by the rise of chaperones and redox enzymes. Overexpression of a HSP70 may be pivotal for M Cu tolerance by protecting protein metabolism. Rice overexpressed thaumatin-like protein gene and the proteins from the leaf blades of 2-week-old transgenic rice seedlings were fractionated into cytosolic and membrane fractions, and separated by two-dimensional polyacrylamide gel electrophoresis and stained with Commassie brilliant blue. Among of 440 detected proteins, 5 proteins were up-regulated and 5 proteins were down-regulated by the overexpression of thaumatin-like protein. In the sense thaumatin-like protein transgenic rice and/or in rice inoculated with Xanthomonas oryzae pv. oryzae (Xo7435), 2-cys peroxiredoxin, thaumatin-like protein and glycine cleavage H protein were up-regulated, while oxygen evolving complex protein 2 was down-regulated. These results suggest that thaumatin-like protein-mediated disease resistance of rice against bacterial blight disease is the results of changes in proteins related to oxidative stress and energy metabolism in addition to changes in proteins related to defence.  相似文献

With the aim to examine their potentials as renewable resources to decontaminate polluted soils, growth, photosynthesis and nitrogen balance were analyzed in two poplar species (Populus x canadensis, Adda clone and Populus alba, Villafranca clone) to investigate the tolerance to high copper (Cu) concentrations. The two clones showed different responses to Cu in terms of tolerance and metal allocation: P. x canadensis accumulated Cu in roots, displaying features sought in plants suitable for phytostabilization, while P. alba accumulated the metal in leaves, like an indicator species.  相似文献

Secondary metabolites released by invasive plants can increase their competitive ability by affecting native plants, herbivores, and pathogens at the invaded land. Whether these secondary metabolites affect the invasive plant itself, directly or indirectly through microorganisms, however, has not been well documented. Here we tested whether activated carbon (AC), a well-known absorbent for secondary metabolites, affect arbuscular mycorrhizal (AM) symbioses and competitive ability in an invasive plant. We conducted three experiments (experiments 1–3) with the invasive forb Solidago canadensis and the native Kummerowia striata. Experiment 1 determined whether AC altered soil properties, levels of the main secondary metabolites in the soil, plant growth, and AMF communities associated with S. canadensis and K. striata. Experiment 2 determined whether AC affected colonization of S. canadensis by five AMF, which were added to sterilized soil. Experiment 3 determined the competitive ability of S. canadensis in the presence and absence of AMF and AC. In experiment 1, AC greatly decreased the concentrations of the main secondary metabolites in soil, and the changes in concentrations were closely related with the changes of AMF in S. canadensis roots. In experiment 2, AC inhibited the AMF Glomus versiforme and G. geosporum but promoted G. mosseae and G. diaphanum in the soil and also in S. canadensis roots. In experiment 3, AC reduced S. canadensis competitive ability in the presence but not in the absence of AMF. Our results provided indirect evidence that the secondary metabolites (which can be absorbed by AC) of the invasive plant S. canadensis may promote S. canadensis competitiveness by enhancing its own AMF symbionts.  相似文献

Background

The consecutive monoculture for most of medicinal plants, such as Rehmannia glutinosa, results in a significant reduction in the yield and quality. There is an urgent need to study for the sustainable development of Chinese herbaceous medicine.

Methodology/Principal Findings

Comparative metaproteomics of rhizosphere soil was developed and used to analyze the underlying mechanism of the consecutive monoculture problems of R. glutinosa. The 2D-gel patterns of protein spots for the soil samples showed a strong matrix dependency. Among the spots, 103 spots with high resolution and repeatability were randomly selected and successfully identified by MALDI TOF-TOF MS for a rhizosphere soil metaproteomic profile analysis. These proteins originating from plants and microorganisms play important roles in nutrient cycles and energy flow in rhizospheric soil ecosystem. They function in protein, nucleotide and secondary metabolisms, signal transduction and resistance. Comparative metaproteomics analysis revealed 33 differentially expressed protein spots in rhizosphere soil in response to increasing years of monoculture. Among them, plant proteins related to carbon and nitrogen metabolism and stress response, were mostly up-regulated except a down-regulated protein (glutathione S-transferase) involving detoxification. The phenylalanine ammonia-lyase was believed to participate in the phenylpropanoid metabolism as shown with a considerable increase in total phenolic acid content with increasing years of monoculture. Microbial proteins related to protein metabolism and cell wall biosynthesis, were up-regulated except a down-regulated protein (geranylgeranyl pyrophosphate synthase) functioning in diterpenoid synthesis. The results suggest that the consecutive monoculture of R. glutinosa changes the soil microbial ecology due to the exudates accumulation, as a result, the nutrient cycles are affected, leading to the retardation of plant growth and development.

Conclusions/Significance

Our results demonstrated the interactions among plant, soil and microflora in the proteomic level are crucial for the productivity and quality of R. glutinosa in consecutive monoculture system.  相似文献

A greenhouse pot experiment with different phosphorus supply was conducted to study growth, photosynthesis and free polyamine (PA) content in Plantago lanceolata L. plants in relation to arbuscular mycorrhizal (AM) colonization. Inoculum of Glomus fasciculatum (BEG 53) was used. Inoculated plants had high colonization intensities which were related to the P supply. Non-mycorrhizal (NM) plants showed a typical yield response curve for P availability. Dry masses of mycorrhizal (M) plants were higher at the lowest soil P content than those of NM plants, but the opposite was found at the highest P supply. P contents in M plants were always higher. There were no differences in chlorophyll (Chl) concentrations (except the lowest soil P content) and ratios of variable to maximum Chl fluorescence (Fv/Fm) values between M and NM plants, whereas M plants had higher ratios of leaf area to fresh mass (A/f.m.) at low soil P contents and they had significantly higher CO2 fixation capacities per unit leaf area. Free putrescine (Put), spermidine (Spd) and spermine (Spm) contents in NM plants were usually highest at the lowest P supply. The ratios of Put/(Spd+Spm) were identical in M and NM leaves. They were significantly higher, however, in NM roots at the two low P doses. It is concluded, that a P nutritional status might exist, below which PA concentrations and ratio are increased drastically, possibly indicating P deficiency or a certain state of plant development with a higher demand for AM symbiosis. The accumulation of heavy metals (Cu, Ni, Zn, Mn, and Fe), parameters of the photosynthetic apparatus, and flavonoid content in leaves of two widespread species of submerged hydrophytes (Elodea canadensis Michx. and Potamogeton perfoliatus L.) from habitats with different levels of pollutions have been studied. The investigations are carried out in the surroundings of the metallurgical plant in the town of Revda, Sverdlovsk oblast, Russia (Revda River, upstream and downstream the confluence of the sewage). It is shown that hydrophytes from polluted habitats (impact site) differ from the plants of a less polluted zone (background site) by a higher accumulation of heavy metals (HMs), greater leaf thickness, and larger cells and chloroplasts. Plants from the polluted site are characterized by a high content of photosynthetic pigments, while flavonoid content in the leaves is lower. The identified trends indicate a high adaptive potential of these species of plants and their ability to inhabit in an aquatic environment polluted with metals and other pollutants.  相似文献

Redox homeostasis is important for plants to be able to maintain cellular metabolism, and disrupting cellular redox homeostasis will cause oxidative damage to cells and adversely affect plant growth. In this study, a cotton CCCH-type tandem zinc finger gene defined as GhTZF1, which was isolated from a cotton cell wall regeneration SSH library in our previous research, was characterized. GhTZF1 was predominantly expressed during early cell wall regeneration, and it was expressed in various vegetative and reproductive tissues. The expression of GhTZF1 was substantially up-regulated by a variety of abiotic stresses, such as PEG and salt. GhTZF1 also responds to methyl jasmonate (MeJA) and H2O2 treatment. Overexpression of GhTZF1 enhanced drought tolerance and delayed drought-induced leaf senescence in transgenic Arabidopsis. Subsequent experiments indicated that dark- and MeJA-induced leaf senescence was also attenuated in transgenic plants. The amount of H2O2 in transgenic plants was attenuated under both drought conditions and with MeJA-treatment. The activity of superoxide dismutase and peroxidase was higher in transgenic plants than in wild type plants under drought conditions. Quantitative real-time PCR analysis revealed that overexpression of GhTZF1 reduced the expression of oxidative-related senescence-associated genes (SAGs) under drought conditions. Overexpression of GhTZF1 also enhanced oxidative stress tolerance, which was determined by measuring the expression of a set of antioxidant genes and SAGs that were altered in transgenic plants during H2O2 treatment. Hence, we conclude that GhTZF1 may serve as a regulator in mediating drought stress tolerance and subsequent leaf senescence by modulating the reactive oxygen species homeostasis.  相似文献

Streptomyces lydicus has been reported to produce antibiotic streptolydigin. Pitching ratios play crucial roles in primary and secondary metabolism of Streptomyces bacteria. The higher pitching ratio (30%, v/v) significantly enhanced the levels of streptolydigin products in S. lydicus. Proteome analysis revealed that betaglucosidase and UTP-glucose-1-phosphate uridylyltransferase were up-regulated to accelerate the starch hydrolyzation at the high pitching ratios. Enhancement in the levels of UDPN-acetylmuramoylalanyl-D-glutamate-2, 6-diaminopimelate ligase and glycine cleavage system aminomethyltransferase were involved in the conversion of amino acids into secondary metabolites. Additionally, the expression levels of PfkA2, PfkA3, Zwf2, SucD, GalE1, GatB, TktA1 and ThcA, associated with glycolysis, pentose phosphate pathway, TCA cycle and amino acid metabolism, were dramatically elevated at high pitching ratios, which play important roles in the enhanced streptolydigin production in S. lydicus E9. Interestingly, the levels of proteins (glutamine synthetase I, glutamate synthase subunit beta and glutamine synthetase) were down-regulated with the increases of pitching ratios and fermentation progress, revealing that pitching ratio altered the glutamine synthetase levels and consequently regulated the streptolydigin production of S. lydicus E9. The up-regulation of proteins (eg, aldehyde dehydrogenase and alkyl hydroperoxide reductase) was involved in the redox-based regulation network triggered by an imbalance of the intracellular cell redox homeostasis and by crosstalk with secondary metabolism at the higher pitching ratio. These results settle new insights into physiological facts of S. lydicus E9 in responses to pitching ratios and will eventually improve the antibiotic production schemes in industry.  相似文献

As one of the largest gene families, F-box domain proteins have been found to play important roles in abiotic stress responses via the ubiquitin pathway. TaFBA1 encodes a homologous F-box protein contained in E3 ubiquitin ligases. In our previous study, we found that the overexpression of TaFBA1 enhanced drought tolerance in transgenic plants. To investigate the mechanisms involved, in this study, we investigated the tolerance of the transgenic plants to oxidative stress. Methyl viologen was used to induce oxidative stress conditions. Real-time PCR and western blot analysis revealed that TaFBA1 expression was up-regulated by oxidative stress treatments. Under oxidative stress conditions, the transgenic tobacco plants showed a higher germination rate, higher root length and less growth inhibition than wild type (WT). The enhanced oxidative stress tolerance of the transgenic plants was also indicated by lower reactive oxygen species (ROS) accumulation, malondialdehyde (MDA) content and cell membrane damage under oxidative stress compared with WT. Higher activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and peroxidase (POD), were observed in the transgenic plants than those in WT, which may be related to the upregulated expression of some antioxidant genes via the overexpression of TaFBA1. In others, some stress responsive elements were found in the promoter region of TaFBA1, and TaFBA1 was located in the nucleus, cytoplasm and plasma membrane. These results suggest that TaFBA1 plays an important role in the oxidative stress tolerance of plants. This is important for understanding the functions of F-box proteins in plants’ tolerance to multiple stress conditions.  相似文献

To better understand abscisic acid (ABA) regulation of the synthesis of chloroplast proteins in maize (Zea mays L.) in response to drought and light, we compared leaf proteome differences between maize ABA-deficient mutant vp5 and corresponding wild-type Vp5 green and etiolated seedlings exposed to drought stress. Proteins extracted from the leaves of Vp5 and vp5 seedlings were used for two-dimensional electrophoresis (2-DE) and subsequent matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). After Coomassie brilliant blue staining, approximately 450 protein spots were reproducibly detected on 2-DE gels. A total of 36 differentially expressed protein spots in response to drought and light were identified using MALDI-TOF MS and their subcellular localization was determined based on the annotation of reviewed accession in UniProt Knowledgebase and the software prediction. As a result, corresponding 13 proteins of the 24 differentially expressed protein spots were definitely localized in chloroplasts and their expression was in an ABA-dependent way, including 6 up-regulated by both drought and light, 5 up-regulated by drought but down-regulated by light, 5 up-regulated by light but down-regulated by drought; 5 proteins down-regulated by drought were mainly those involved in photosynthesis and ATP synthesis. Thus, the results in the present study supported the vital role of ABA in regulating the synthesis of drought- and/or light-induced proteins in maize chloroplasts and would facilitate the functional characterization of ABA-induced chloroplast proteins in C4 plants.  相似文献

The purpose of this study was to test the hypothesis that vesicular arbuscular mycorrhizal (VAM) fungi affect net assimilation of CO2 (A) of different-aged citrus leaves independent of mineral nutrition effects of mycorrhizae. Citrus aurantium L., sour orange plants were grown for 6 months in a sandy soil low in phosphorus that was either infested with the VAM fungus, Glomus intraradices Schenck & Smith, or fertilized with additional phosphorus and left nonmycorrhizal (NM). Net CO2 assimilation, stomatal conductance, water use efficiency, and mineral nutrient status for expanding, recently expanded, and mature leaves were evaluated as well as plant size and relative growth rate of leaves. Nutrient status and net gas exchange varied with leaf age. G. intraradices-inoculated plants had well-established colonization (79% of root length) and were comparable in relative growth rate and size at final harvest with NM plants. Leaf mineral concentrations were generally the same for VAM and NM plants except for nitrogen. Although leaf nitrogen was apparently sufficient for high rates of A, VAM plants did have higher nitrogen concentrations than NM at the time of gas exchange measurements. G. intraradices had no effect on A, stomatal conductance, or water use efficiency, irrespective of leaf age. These results show that well-established VAM colonization does not affect net gas exchange of citrus plants that are comparable in size, growth rate, and nutritional status with NM plants.  相似文献

外来植物对入侵地土壤动物群落及理化性质影响的研究不仅有助于评估入侵植物对生态系统的影响,而且对探索外来植物入侵的土壤动物学响应机制尤为重要。为了了解加拿大一枝黄花对入侵地土壤动物的群落结构及理化性质的影响,本文分季节分层次对加拿大一枝黄花不同程度的入侵地进行取样,获得土壤动物9900个,隶属3门11纲14目,弹尾目和蜱螨类均为优势类群。入侵程度不同的样地中土壤动物个体数量和类群组成不同。土壤动物个体数量和类群数量表现为轻度入侵区>中度入侵区>重度入侵区;多样性指数和均匀性指数表现为轻度入侵区<中度入侵区<重度入侵区,优势度指数表现与前两指数相反。加拿大一枝黄花的入侵没有改变土壤动物表聚性特点。非度量多维标度排序分析表明,不同入侵程度下的土壤动物分为3类,即轻度入侵类、中度入侵类、重度入侵类。不同入侵区域土壤的pH、有机质含量、铵态氮、速效钾和速效磷差异显著(P<0.05),土壤的含水量和温度差异不显著(P>0.05)。灰色关联分析表明,入侵区域土壤铵态氮对土壤动物关联最大,有机质含量次之,再次是速效磷和pH,土壤含水量的影响最小。因而,加拿大一枝黄花的入侵,改变了入侵地土壤理化性质(尤其是对铵态氮的调控),进而改变了土壤动物的群落结构,创造了利于自身生长、竞争有利的土壤环境。  相似文献
