The frequencies of a polymorphic inversion, In(2L)t, and of Adh and αGpdh alleles were analyzed in three natural populations of Drosophila melanogaster from Japan. Significant positive correlations between the frequencies of In(2L)t and AdhS or αGpdhF were detected due to tight linkage. An analysis of correlation with latitude showed that the negative cline of AdhS frequency could be explained entirely by its linkage with In(2L)t; the frequency of AdhS on the standard chromosome did not show a latitudinal cline. To the contrary, the cline of αGpdhF frequency itself was positive, and its linkage with In(2L)t makes the positive cline unclear. These results suggest that the two allozymes themselves respond to latitudinal natural selection in different ways. When these populations were transferred to laboratory cages and maintained for a long time, they lost the chromosomal polymorphism but retained stable enzyme polymorphisms, although allele frequencies in the cage were not the same as in nature. The frequencies of Adh and αGpdh alleles were close to those in earlier cage populations of the same geographical origin.  相似文献

In various populations of the cultivated and weedy amaranth species, the electrophoretic patterns of alcohol dehydrogenase (ADH), glutamate dehydrogenase (GDH), malate dehydrogenase (MDH), isocitrate dehydrogenase (IDH) and malic enzyme (Me) were studied. In total, 52 populations and two varieties (Cherginskii and Valentina) have been examined. Allozyme variation of this material was low. Irrespective of species affiliation, 26 populations and two varieties were monomorphic for five enzymes; a slight polymorphism of three, two, and one enzymes was revealed in three, nine, and fourteen populations, respectively. A single amaranth locus, Adh, with two alleles, Adh F and Adh S, controls amaranth ADH. Two alleles, common Gdh S and rare Gdh F, control GDH; no heterozygotes at this locus were found. The MDH pattern has two, the fast- and slow-migrating, zones of activity (I and II, respectively). Under the given electrophoresis conditions, the fast zone is diffuse, whereas slow zone is controlled by two nonallelic genes, monomorphic Mdh 1 and polymorphic Mdh 2 that includes three alleles: Mdh 2-F, Mdh 2-N, and Mdh 2-S. Low polymorphism of IDH and Me was also found, though their genetic control remains unknown.  相似文献

Ten Indian geographical populations of D. melanogaster were assayed electrophoretically for Adh genic variation. The Indian geographical populations of D. melanogaster revealed significant clinal variation (3 % for 1 d? latitude) at Adh locus and AdhF allelic frequency correlated significantly with increase in latitude. It was suggested that the abundance of secondary alcohols in the southern Indian tropical and humid environment might exert selective pressure favouring higher frequency of AdhS allele. Patterns of ethanol utilization as well as ethanol tolerance were analyzed in larval and adult individuals of six geographical populations of D. melanogaster. Latitudinal variation in ethanol tolerance was observed in D. melanogaster populations from India. The parallel occurrence of latitudinal variation it Adh locus as well as ethanol tolerance in Indian geographical populations of D. melanogaster could be maintained by balancing natural selection varying spatially along the north-south axis of the Indian sub-continent.  相似文献

Zymograms of Arabidopsis alcohol dehydrogenase (ADH; EC 1.1.1.1) show a unique anodal migrating band. Three electrophoretic variants were identified among geographical races and designated slow (S), fast (F), and superfast (A), according to their mobility on Tris-citrate starch gels. In plants ADH activity is confined mainly to pollen, seeds, and grains and rapidly declines during the germination process. In callus and suspension cultures, growing on media containing 2,4-D, ADH appeared as one of the major polypeptides. Genetical analysis indicated that the three types of ADH isozymes are under the control of one gene with three alles (Adh 1 s , Adh 1 f , Adh 1 a ), showing codominant expression. Crosses between the electrophoretic types and dissociation-reassociation experiments showed that the Arabidopsis enzyme behaves as a dimer, like ADH from most other species. The molecular weight of the enzyme has been estimated by gel filtration and by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis to be 87,000. The pH optimum for the oxidation of ethanol is 9.0 and two optima for reduction of acetaldehyde have been obtained, 6.0 and 8.5, respectively. The enzyme exhibits a wide substrate specificity for alcohols and is relatively heat resistant.  相似文献

In vitro alcohol dehydrogenase (ADH) activity was measured in adults of species belonging to Drosophila and to the related genus Zaprionus. Data were analyzed according to the known breeding sites and the level of ethanol tolerance of these species. Alcohol dehydrogenase activity was assayed with both ethanol (E) and isopropanol (I). Our results show a very broad range of activities among the 71 species investigated, the ratio of the highest value observed (D. melanogaster) to the lowest (D. pruinosa) being 65:1. A general positive correlation was found between the level of ADH activity and the capacity to detoxify ethanol. Nevertheless, many species show exceptions to this rule. Contrary to a logical expectation, adaptation to high alcoholic resources, which has been a recurrent evolutionary event, was not mediated by a more efficient use of ethanol, that is, an increase of the E/I ratio. This ratio seems to be quite variable according to the phylogeny and is especially low in the subgenus Sophophora as well as in Zaprionus. Alcohol tolerance clearly is related to the larval habitat of the species and shows that adaptation to alcoholic resources has been a major evolutionary challenge in drosophilids. This adaptation is not related to phylogeny, having occurred independently several times during the evolution of the group. Finally, it should be borne in mind that, besides metabolization and detoxification, other physiological processes such as nervous-system tolerance or ethanol excretion may be involved in ethanol tolerance, and such functions also should be investigated. Environmental ethanol, which is certainly a major ecological parameter for many drosophilids, has selected a diversity of physiological adaptations, all related to the Adh locus, but presumably much more complicated than was previously believed.  相似文献

Gametes from each of 19 Australasian collections of Drosophila melanogaster were characterised for polymorphic Adh and Gpdh allozyme loci and the two chromosome-2 common cosmopolitan inversions In(2L)t and In(2R)NS.-No consistent gametic disequilibria for the loosely linked combinations of Adh-Gpdh and Adh-In(2R)NS were found over the 19 collections. However, strong and consistent gametic disequilibria were found for the tightly linked combinations of Adh-In(2L)t and Gpdh-In(2L)t. The results for each of these four combinations agree with those previously reported for Northern-Hemisphere collections. -Notwithstanding its parallel latitudinal cline and consistent gametic disequilibrium with Adh, In(2L)t was found to account for only a small fraction of the Adh latitudinal cline in Australasia. This is consistent with the previous finding that the North American Adh latitudinal cline also exists largely independently of In(2L)t. However, In(2L)t accounts for all of the Australasian Gpdh latitudinal cline and so contrasts the finding from North American collections where the corresponding Gpdh cline exists largely independently of In(2L)t.  相似文献

The Adh and αGpdh allozyme loci (both located on the second chromosome) showed considerable fluctuations in allele frequencies in a seminatural population of Drosophila melanogaster during 1972–97. Both long-term and short-term fluctuations were observed. The short-term fluctuations occurred within almost all years and comparison of allele frequencies between winters and summers showed significantly higher AdhS (P < 0.001) and αGpdhF (P < 0.01) allele frequencies in summers. Frequencies of these alleles were significantly positively correlated with environmental temperature, suggesting the adaptive significance of these allozyme polymorphisms. Frequency changes of the Odh locus (located on the third chromosome) showed no seasonal pattern and were not correlated with environmental temperature. Almost all short-term and long-term increases in AdhS frequency were accompanied by a corresponding decrease in αGpdhS frequency (r = –0.82, P < 0.001) and vice versa. Further analysis showed that gametic disequilibria between the Adh and αGpdh loci, which frequently occurred, were due to the presence of inversion In(2L)t located on the same chromosome arm and In(2L)t frequencies were positively correlated with environmental temperature. Gametic disequilibria between Adh and Odh and between Odh and αGpdh were hardly observed. Because In(2L)t is exclusively associated with the AdhS/αGpdhF allele combination, the observed correlated response in Adh/αGpdh allele frequencies is (at least partly) explained by hitchhiking effects with In(2L)t. This means that the adaptive value of the allozyme polymorphisms has been overestimated by ignoring In(2L)t polymorphism. Fluctuations in Adh allele frequencies are fully explained by selection on In(2L)t polymorphism, whereas we have shown that αGpdh frequency fluctuations are only partly explained by chromosomal hitchhiking, indicating the presence of selective differences among αGpdh genotypes in relation with temperature and independent of In(2L)t. Frequency fluctuations of αGpdh and In(2L)t are consistent with their latitudinal distributions, assuming that temperature is the main environmental factor varying with latitude that causes directly or indirectly these frequency distributions. However, the results of the tropical greenhouse population show no correlation of Adh (independent of In(2L)t) and Odh allele frequencies with environmental temperature, which may indicate that the latitudinal distribution in allele frequencies for these loci is not the result of selection on the F/S polymorphism in a direct way.  相似文献

In the Gram-positive methylotroph Bacillus methanolicus, methanol oxidation is catalyzed by an NAD-dependent methanol dehydrogenase (Mdh) that belongs to the type III alcohol dehydrogenase (Adh) family. It was previously shown that the in vitro activity of B. methanolicus Mdh is increased by the endogenous activator protein Act, a Nudix hydrolase. Here we show that this feature is not unique, but more widespread among type III Adhs in combination with Act or other Act-like Nudix hydrolases. In addition, we studied the effect of site directed mutations in the predicted active site of Mdh and two other type III Adhs with regard to activity and activation by Act.  相似文献

Subcellular localization of isozymes of NAD-dependent malate dehydrogenase (MDH) in sugar beet was studied. Isozymes ss and ll controlled by loci Mdh2 and Mdh3, respectively, were shown to locate in mitochondria, whereas isozyme pp controlled by locus Mdh1, in microbodies. All examined samples lack hybrid MDH isozymes, which could testify to the interaction between products of nonallelic Mdh genes. This can be explained by the localization of nonallelic isozymes in various compartments of the cell and organelles.  相似文献

Alcohol dehydrogenase has been purified from Drosophila melanogaster lines bearing the Adh F, AdhS, and Adh FCh.D. alleles. Biochemical investigations show that the properties of the purified enzymes are very similar to those of crude enzyme extracts except that the pure enzymes are more heat stable. ADH-FCh.D. resembles ADH-S very closely in specific activity, substrate specificity, and a number of kinetic parameters including limiting values for K m(app.) for ethanol. However, it is considerably more heat stable than either of the two common variants. ADH-F differs from ADH-S and ADH-FCh.D. particularly with regard to the rate of oxidation of secondary alcohols. Atomic absorbtion spectroscopy shows that all three allozymes lack zine or other divalent cations as active-site components. Peptide mapping experiments identify one very active cysteinyl residue; and amide residues in the NAD+ binding domain.  相似文献

siren1 and siren2 are novel alcohol dehydrogenase (Adh)-derived chimeric genes in the Drosophila bipectinata complex. D. ananassae, however, harbors a single homolog of these genes. Like other Adh-derived chimeric genes, siren evolved adaptively shortly after it was formed. These changes likely shifted the catalytic activity of siren.  相似文献

Substantial allele-frequency changes were observed at the Adh and αGpdh loci in a seminatural population of Drosophila melanogaster kept in a tropical greenhouse during 1972–1985. Further analysis of the changes at the Adh and αGpdh loci showed that linkage disequilibrium between these loci occurred for a prolonged period due to the presence of In(2L)t, a long inversion on the left arm of the second chromosome. We observed increases in the frequencies of In(2L)t and of short inversions on the left arm of the second chromosome in subpopulations kept at 29.5°C or 33°C. These inversion-frequency increases were accompanied by an increase in Adhs and a decrease in αGpdhs frequency. In populations kept at 20°C and 25°C, inversion frequencies decreased, while αGpdhs allele frequencies decreased at 25°C and increased at 20°C. At 33°C, egg-to-adult survival of individuals possessing In(2L)t, either in the homokaryotypic or the heterokaryotypic state, was higher than that of the other karyotypes of identical allozyme constitution (i.e., Adhs αGpdhF). Thus it seems that In(2L)t has a selective advantage at high temperature. We argue that the observed changes in allele frequencies at the Adh and αGpdh loci are, in part, due to genic selection and are not merely the result of selection acting on the chromosome rearrangements and hitchhiking of the allozymes. The results are discussed with respect to the latitudinal clines found for In(2L)t, Adh, and αGpdh.  相似文献

Microbial fermentation of sugars from plant biomass to alcohols represents an alternative to petroleum-based fuels. The optimal biocatalyst for such fermentations needs to overcome hurdles such as high concentrations of alcohols and toxic compounds. Lactic acid bacteria, especially lactobacilli, have high innate alcohol tolerance and are remarkably adaptive to harsh environments. This study assessed the potential of five Lactobacillus casei strains as biocatalysts for alcohol production. L. casei 12A was selected based upon its innate alcohol tolerance, high transformation efficiency and ability to utilize plant-derived carbohydrates. A 12A derivative engineered to produce ethanol (L. casei E1) was compared to two other bacterial biocatalysts. Maximal growth rate, maximal optical density and ethanol production were determined under conditions similar to those present during alcohol production from lignocellulosic feedstocks. L. casei E1 exhibited higher innate alcohol tolerance, better growth in the presence of corn stover hydrolysate stressors, and resulted in higher ethanol yields.  相似文献

Four Drosophila melanogaster strains, each homozygous for one of the two major ADH aliozymes, Fast and Slow (Adh F1, Adh S , Adh F2 and Adh S2) were used to study the interaction of the Adh locus with ethanol and temperature. At least six nuclear loci are responsible for the genetic control of malate dehydrogenase (L-malate: NAD oxidoreductase; EC 1.1.1.37; MDH) in coleoptiles of maize. Three independently segregating loci (Mdh1, Mdh2, Mdh3) govern the production of MDH isozymes resistant to inactivation by ascorbic acid and found largely or solely in the mitochondria. A rare recessive allele found at a fourth nuclear locus (mmm) causes increased electrophoretic mobility of the MDH isozymes governed by the Mdh1, Mdh2 and Mdh3 loci.—Two loci (Mdh4, Mdh5) govern MDH isozymes that are selectively inactivated by homogenization in an ascorbic acid solution and that appear to be nonmitochondrial (soluble). Mdh4 and Mdh5 segregate independently of each other and independently of Mdh1, Mdh2 and Mdh3. However, there is close linkage between the migration modifier and Mdh4.——Multiple alleles have been found for all of the Mdh loci except the migration modifier, and electrophoretically "null" or near "null" alleles (as expressed in standardized sections of maize coleoptile) have been found for all loci except Mdh4. Duplicate inheritance commonly occurs for Mdh1 and Mdh2 and also for Mdh4 and Mdh5.——Inter- and intragenic heterodimers are formed between sub-units specified by the three loci governing the mitochondrial MDH isozymes. The same is true of the alleles and nonalleles at the two loci governing the soluble variants. No such heterodimers are formed by interactions between mitochondrial and soluble MDH isozymes.  相似文献

The enzyme activity levels of alcohol, malate, isocitrate, glucose-6-phosphate and 6-phosphogluconate dehydrogenases were determined in mature maize scutella in a series of one to four doses of the long arm of chromosome 1, produced by the B-A translocation 1La. Although the Adh structural locus was varied, ADH levels did not exhibit a gene-dosage effect. The levels of G6PDH, 6PGDH and IDH were negatively correlated with the dosage of 1L. MDH was unresponsive. The esterase-8 enzyme, whose structural locus was demonstrated to be elsewhere in the genome, was also negatively correlated with 1L dosage. The portion of the B chromosome involved in the translocation was shown to have no effect on the enzyme levels. Measurements of cell size and hydrolysable DNA per mg dry weight revealed no change in the number of cells through the one, two and three dose series. Lines homozygous for the Adh S and Adh F alleles were extracted from a French and an African natural population of D. melanogaster. The lines from each geographic region were then crossed and the Mendelian F2 constituted the first generation subjected to selection for an increase in adult survival in the presence of 2% ethanol.The responses to selection were similar in the two strains, in spite of their different ethanol tolerance. In less than 10 generations, life span with ethanol increased from about 7 to more than 12 days, with realized heritabilities of 0.14 and 0.18. This extension of longevity was also observed with other concentrations of ethanol but not with water alone. The increase in life span in presence of alcohol appears to result from unknown metabolic processes, which are not obligatorily related to the capacity of the flies to tolerate starvation, nor to their size, their lipid content of their ethanol tolerance. In the two lines, however, the Adh S allele was quickly eliminated, suggesting a selective advantage for the F allele.  相似文献

Flood tolerance of woody plants has been attributed to internal oxygen diffusion from shoot to root, metabolic adaptation within the root, or both. The purpose of this study was to compare several biochemical and physiological responses of birch roots to hypoxia in order to determine the nature of root metabolic adaptation to low oxygen tension. One-year-old seedlings of flood-tolerant river birch (Betula nigra L.) and flood-intolerant European birch (Betula pendula Roth) were transferred to solution culture, and the solutions were bubbled with air or nitrogen. After 18 days of hypoxia, total adenosine phosphate and ATP contents of river birch roots were 35% and 23% of controls, respectively, whereas those of European birch roots were 13% and 8%. Adenylate energy charge of river birch roots decreased between 6 and 12 days of hypoxia. In contrast, energy charge of European birch roots decreased after only 1 day of hypoxia. In vitro activity of cytochrome c oxidase and oxygen consumption capacity of excised roots from both birch species decreased under hypoxia. In vitro activity of alcohol dehydrogenase from roots of both species increased after 1 day of hypoxia. However, alcohol dehydrogenase activity from river birch roots increased 25-fold after 6 days of hypoxia, whereas that from European birch decreased back to control levels. Hypoxia decreased malate content of roots from both species. Metabolic adaptation within the root, rather than internal oxygen diffusion, appears to be responsible for the relative tolerance of river birch to hypoxia.  相似文献
