Salinity is one of the major environmental limiting factors that affects growth and productivity of rice (Oryza sativa L.) worldwide. Rice is among the most sensitive crops to salinity, especially at early vegetative stages. In order to get a better understanding of molecular pathways affected in rice mutants showing contrasting responses to salinity, we exploited the power of 2-DE based proteomics to explore the proteome changes associated with salt stress response. Our physiological observations showed that standard evaluation system (SES) scores, Na+ and K+ concentrations in shoots and Na+/K+ ratio were significantly different in contrasting mutants under salt stress condition. Proteomics analysis showed that, out of 854 protein spots which were reproducibly detected, 67 protein spots showed significant responses to salt stress. The tandem mass spectrometry analysis of these significantly differentially accumulated proteins resulted in identification of 34 unique proteins. These proteins are involved in various molecular processes including defense to oxidative stresses, metabolisms, photosynthesis, protein synthesis and processing, signal transduction. Several of the identified proteins were emerged as key participants in salt stress tolerance. The possible implication of salt responsive proteins in plant adaptation to salt stress is discussed.  相似文献

To keep pace with ever growing global population, progressive and sustained increase in rice production is necessary, especially in areas with extremely variable climatic conditions, where rice crop suffers from numerous abiotic stresses including salinity. Designing an effective phenotyping strategy requires thorough understanding of plant survival under stress. The investigation was carried out with four rice cultivars namely FR13A, IR42, Rashpanjor, and Pokkali that differed in salinity tolerance. The study showed that a genotype with initial vigour had some advantage in preserving shoot biomass under salt stress. Though both FR13A and IR42 showed sensitivity to salinity, FR13A with higher initial biomass maintained greater dry weight under saline condition. Increase of Na+:K+ ratio under salinity, due to accelerated absorption of Na+ and lesser absorption of K+ compared to control, was considerably higher in susceptible (118–200 %) than in tolerant (33–48 %) genotypes. While Na+ concentration in shoot increased significantly in both tolerant and susceptible genotypes, decrease in shoot K+ content was noticed only in susceptible genotypes. The imbalance of Na+ and K+ contents led to increased H2O2 production, causing greater peroxidation of membrane lipids and reduction in chlorophyll content and CO2 photosynthetic rate. Certain chlorophyll fluorescence parameters could distinguish between salinity tolerant and sensitive genotypes. To protect the plant from oxidative damage, several enzymatic and nonenzymatic antioxidants such as ascorbate were involved. The genotypes with capacity to assemble antioxidant enzymes in time could detoxify the reactive oxygen species more efficiently, leading to greater protection and reduced impact of salt stress.  相似文献

Abiotic stresses are among the major limiting factors for plant growth and crop productivity. Among these, salinity is one of the major risk factors for plant growth and development in arid to semi-arid regions. Cultivation of salt tolerant crop genotypes is one of the imperative approaches to meet the food demand for increasing population. The current experiment was carried out to access the performance of different rice genotypes under salinity stress and Zinc (Zn) sources. Four rice genotypes were grown in a pot experiment and were exposed to salinity stress (7 dS m−1), and Zn (15 mg kg−1 soil) was applied from two sources, ZnSO4 and Zn-EDTA. A control of both salinity and Zn was kept for comparison. Results showed that based on the biomass accumulation and K+/Na+ ratio, KSK-133 and BAS-198 emerged as salt tolerant and salt sensitive, respectively. Similarly, based on the Zn concentration, BAS-2000 was reported as Zn-in-efficient while IR-6 was a Zn-efficient genotype. Our results also revealed that plant growth, relative water content (RWC), physiological attributes including chlorophyll contents, ionic concentrations in straw and grains of all rice genotypes were decreased under salinity stress. However, salt tolerant and Zn-in-efficient rice genotypes showed significantly higher shoot K+ and Zn concentrations under saline conditions. Zinc application significantly alleviates the harmful effects of salinity by improving morpho-physiological attributes and enhancing antioxidant enzyme activities, and the uptake of K and Zn. The beneficial effect of Zn was more pronounced in salt-tolerant and Zn in-efficient rice genotypes as compared with salt-sensitive and Zn-efficient genotypes. In sum, our results confirmed that Zn application increased overall plant’s performance under saline conditions, particularly in Zn in-efficient and tolerant genotypes as compared with salt-sensitive and Zn efficient rice genotypes.  相似文献

Salinity stress is a major factor limiting plant growth and productivity of many crops including oilseed. The present study investigated the identification of salt tolerant mustard genotypes and better understanding the mechanism of salinity tolerance. Salt stresses significantly reduced relative water content (RWC), chlorophyll (Chl) content, K+ and K+ /Na+ ratio, photosynthetic rate (PN), transpiration rate (Tr), stomatal conductance (gs), intercellular CO2 concentration (Ci) and increased the levels of proline (Pro) and lipid peroxidation (MDA) contents, Na+ , superoxide (O2•− ) and hydrogen peroxide (H2O2) in both tolerant and sensitive mustard genotypes. The tolerant genotypes maintained higher Pro and lower MDA content than the salt sensitive genotypes under stress condition. The activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), glutathione peroxidase (GPX), monodehydroascorbate reductase (MDHAR) and dehydroascorbate reductase (DHAR) were increased with increasing salinity in salt tolerant genotypes, BJ-1603, BARI Sarisha-11 and BARI Sarisha-16, but the activities were unchanged in salt sensitive genotype, BARI Sarisha-14. Besides, the increment of ascorbate peroxidase (APX) activity was higher in salt sensitive genotype as compared to tolerant ones. However, the activities of glutathione reductase (GR) and glutathione S-transferase (GST) were increased sharply at stress conditions in tolerant genotypes as compared to sensitive genotype. Higher accumulation of Pro along with improved physiological and biochemical parameters as well as reduced oxidative damage by up-regulation of antioxidant defense system are the mechanisms of salt tolerance in selected mustard genotypes, BJ-1603 and BARI Sarisha-16.  相似文献

Salinity is a major abiotic stress that limits plant productivity and quality throughout the world. Roots are the sites of salt uptake. To better understand salt stress responses in maize, we performed a comparative proteomic analysis of seedling roots from the salt-tolerant genotype F63 and the salt-sensitive genotype F35 under 160 mM NaCl treatment for 2 days. Under salinity conditions, the shoot fresh weight and relative water content were significantly higher in F63 than in F35, while the osmotic potential was significantly lower and the reduction of the K+/Na+ ratio was significantly less pronounced in F63 than in F35. Using an iTRAQ approach, twenty-eight proteins showed more than 2.0- fold changes in abundance and were regarded as salt-responsive proteins. Among them, twenty-two were specifically regulated in F63 but remained constant in F35. These proteins were mainly involved in signal processing, water conservation, protein synthesis and biotic cross-tolerance, and could be the major contributors to the tolerant genotype of F63. Functional analysis of a salt-responsive protein was performed in yeast as a case study to confirm the salt-related functions of detected proteins. Taken together, the results of this study may be helpful for further elucidating salt tolerance mechanisms in maize.  相似文献

Identification and characterization of proteins involved in salt tolerance are imperative for revealing its genetic mechanisms. In this study, ionic and proteomic responses of a Tibetan wild barley XZ16 and a well‐known salt‐tolerant barley cv. CM72 were analyzed using inductively coupled plasma‐optical emission spectrometer, 2DE, and MALDI‐TOF/TOF MS techniques to determine salt‐induced differences in element and protein profiles between the two genotypes. In total, 41 differentially expressed proteins were identified in roots and leaves, and they were associated with ion homeostasis, cell redox homeostasis, metabolic process, and photosynthesis. Under salinity stress, calmodulin, Na/K transporters, and H+‐ATPases were involved in establishment of ion homeostasis for barley plants. Moreover, ribulose‐1,5‐bisphosphate carboxylase/oxygenase activase and oxygen‐evolving enhancer proteins were significantly upregulated under salinity stress, indicating the great impact of salinity on photosynthesis. In comparison with CM72, XZ16 had greater relative dry weight and lower Na accumulation in the shoots under salinity stress. A higher expression of HvNHX1 in the roots, and some specific proteins responsible for ion homeostasis and cell redox homeostasis, was also found in XZ16 exposed to salt stress. The current results showed that Tibetan wild barley XZ16 and cultivated barley cultivar CM72 differ in the mechanism of salt tolerance.  相似文献

Responses of plants to salinity stress and the development of salt tolerance are extremely complex. Proteomics is a powerful technique to identify proteins associated with a particular environmental or developmental signal. We employed a proteomic approach to further understand the mechanism of plant responses to salinity in a salt-tolerant (Afzal) and a salt-sensitive (Line 527) genotype of barley. At the 4-leaf stage, plants were exposed to 0 (control) or 300 mM NaCl. Salt treatment was maintained for 3 weeks. Total proteins of leaf 4 were extracted and separated by two-dimensional gel electrophoresis. More than 500 protein spots were reproducibly detected. Of these, 44 spots showed significant changes to salt treatment compared to the control: 43 spots were upregulated and 1 spot was downregulated. Using MALDI-TOF-TOF MS, we identified 44 cellular proteins have been identified, which represented 18 different proteins and were classified into seven categories and a group with unknown biological function. These proteins were involved in various many cellular functions. Up regulation of proteins which involved in reactive oxygen species scavenging, signal transduction, protein processing and cell wall may increase plant adaptation to salt stress. The upregulation of the three of four antioxidant proteins (thioredoxin, methionine sulfoxide reductase and dehydroascorbate reductase) in susceptible genotype Line 527 suggesting a different tolerance mechanism (such as tissue tolerance) to tolerate a salinity condition in comparison with the salt sensitive genotype.  相似文献

Physiological responses to salt stress were studied in two cotton cultivars previously selected on the basis of growth under salinity. Plants were grown in nutrient solutions under controlled conditions. In the first experiment, the genotypes were grown at different salt concentrations (0, 100 and 200 mt M NaCl) and growth rates, water contents and ion accumulation were determined. In a second experiment, both genotypes were grown at the same salt concentration (200 mt M NaCl). Dry matter partitioning in individual leaves, stem and roots, water contents, specific leaf area (SLA), ion accumulation (K+, Na+, Cl) and leaf water potentials were measured. Finally, an experiment with low salt levels (2.7 and 27 mt M NaCl) was run to compare K and Na+ uptake and distribution.There were no differences in growth between the cultivars in the absence of salt stress, whereas under stress genotype Z407 had higher leaf area and dry matter accumulation than P792. Leaf water potential and leaf water content were lower in cv P792 than in cv Z407. There were no significant differences in the levels of Cl accumulation between genotypes. The main feature of the tolerant genotype (Z407) was a higher accumulation of Na+ in leaves and an apparent capacity for K+ redistribution to younger leaves.We postulate that the higher tolerance in Z407 is the result of several traits such as a higher Na+ uptake and water content. Adaptation through adequate, but tightly controlled ion uptake, typical of some halophytes, matched with efficient ion compartmentation and redistribution, would result in an improved water uptake capacity under salt stress and lead to maintenance of higher growth rates.  相似文献

Abstract Protein synthesis during seed germination, a stage vulnerable to salinity stress, was investigated. The responses of barley genotypes, CM72 (California Mariout 72) and Prato, toward salinity were different during seed germination. Germination of CM72 was unaffected up to 0.34 kmol m?3 (2%) NaCl, but that of Prato was reduced 30% by 0.17 kmol m 3 NaCl and 75% by 0.34 kmol m?3 NaCl. Therefore, the former genotype is relatively more salt-tolerant than the latter. Protein synthesis in roots, shoots, and embryos was investigated in these two genotypes before and after salinity stress. The uptake of S-methionine and its incorporation into protein were significantly reduced by salinity in both genotypes. The inhibition of global protein synthesis was significant in roots and shoots. Proteins from different tissues were resolved by single and two dimensional gels. The steady-state protein levels were maintained remarkably well during salinity stress in roots and shoots. Likewise, proteins in germinating embryos were stable except for a 42-kilodalton protein unique to the salt tolerant genotype which was apparently degraded during salinity stress. Salinity, around 0.34 kmol m?3 NaCl, induced both quantitative and qualitative changes in the expression of some proteins labelled in vivo. The quantitative changes included repression or enhancement of synthesis of selected groups of proteins. Around 8% of the nearly 400 resolved proteins in a tissue was affected this way. Some of the proteins in this category were specific to each genotype. About 1 % of the total showed qualitative changes; these proteins were expressed only during salinity stress. In roots, two proteins (28, 41.7 kilodaltons) were detected in CM72 and five (28, 45, 60.5, 76.5, 82.5 kilodaltons) in Prato; only the 28-kilodalton protein was common to both genotypes. In shoots, four proteins (45, 60.5, 76.5, 82.5 kilodaltons) were found only in Prato and these were similar to those induced in roots. The four new proteins (32, 37.5, 89, 92 kilodaltons) in germinating embryos were apparently induced only in CM72; these were distinctly different from those detected in developed roots and shoots. The unique protein changes induced by salinity stress during germination (this study) and seedling growth studies reported earlier (Ramagopal, 1987b) are apparently different. The findings demonstrate that ontogeny plays an important role in the expression of tissue-specific proteins during salinity stress in the salt tolerant and sensitive barley genotypes.  相似文献

Soil salinity and drought are the two most common and frequently co‐occurring abiotic stresses limiting cotton growth and productivity. However, physiological mechanisms of tolerance to such condition remain elusive. Greenhouse pot experiments were performed to study genotypic differences in response to single drought (4% soil moisture; D) and salinity (200 mM NaCl; S) stress and combined stresses (D + S) using two cotton genotypes Zhongmian 23 (salt‐tolerant) and Zhongmian 41 (salt‐sensitive). Our results showed that drought and salinity stresses, alone or in combination, caused significant reduction in plant growth, chlorophyll content and photosynthesis in the two cotton genotypes, with the largest impact visible under combined stress. Interestingly, Zhongmian 23 was more tolerant than Zhongmian 41 under the three stresses and displayed higher plant dry weight, photosynthesis and antioxidant enzymes activities such as superoxide dismutase (SOD), peroxidase (POD) catalase (CAT) and ascorbate peroxidase (APX) activities compared to control, while those parameters were significantly decreased in salt‐stresses Zhongmian 41 compared to control. Moreover, Na+/K+‐ATPase activity was more enhanced in Zhongmian 23 than in Zhongmian 41 under salinity stress. However, under single drought stress and D + S stress no significant differences were observed between the two genotypes. No significant differences were detected in Ca2+/Mg2+‐ATPase activity in Zhongmian 41, while in Zhongmian 23 it was increased under salinity stress. Furthermore, Zhongmian 23 accumulated more soluble sugar, glycine‐betaine and K+, but less Na+ under the three stresses compared with Zhongmian 41. Obvious changes in leaf and root tips cell ultrastructure was observed in the two cotton genotypes. However, Zhongmian 23 was less affected than Zhongmian 41 especially under salinity stress. These results give a novel insight into the mechanisms of single and combined effects of drought and salinity stresses on cotton genotypes.  相似文献

To understand the genotypic variation of citrus to mild salt stress, a proteomic approach has been carried out in parallel on two citrus genotypes (‘Cleopatra’ and ‘Willow leaf’ mandarins), which differ for Na+ and Cl accumulation, and their cognate autotetraploids (4×). Using two-dimensional electrophoresis approximately 910 protein spots were reproducibly detected in control and salt-stressed leaves of all genotypes. Among them, 44 protein spots showing significant variations at least in one genotype were subjected to mass spectrometry analysis for identification. Salt-responsive proteins were involved in several functions, including photosynthetic processes, ROS scavenging, stress defence, and signalling. Genotype factors affect the salt-responsive pattern, especially that of carbon metabolism. The no ion accumulator ‘Cleopatra’ mandarin genotype showed the highest number of salt-responsive proteins, and up-regulation of Calvin cycle-related proteins. Conversely the ion accumulator ‘Willow leaf’ mandarin showed high levels of several photorespiration-related enzymes. A common set of proteins (twelve spots) displayed higher levels in salt-stressed leaves of 2× and 4× ‘Cleopatra’ and 4× ‘Willow leaf’ mandarin. Interestingly, antioxidant enzymes and heat shock proteins showed higher constitutive levels in 4× ‘Cleopatra’ mandarin and 4× ‘Willow leaf’ mandarin compared with the cognate 2× genotype. This work provides for the first time information on the effect of 8 weeks of salt stress on citrus genotypes contrasting for ion accumulation and their cognate autotetraploids. Results underline that genetic factors have a predominant effect on the salt response, although a common stress response independent from genotype was also found.  相似文献

Physiological mechanisms of salinity–Cd interactions were investigated in inter- and intracellular leaf compartments of salt-tolerant wheat × Lophopyrum elongatum (Host) A. Löve (syn. Agropyron elongatum) amphiploid and its salt-sensitive wheat parent (Triticum aestivum L. cv Chinese Spring). In comparison with the intracellular fluid, only very low Na+ concentrations (up to about 4 mM) were found in the intercellular leaf compartment of wheat after a 75 mM supply of NaCl. NaCl salinity led to a higher Cd concentration in leaves of the salt-sensitive genotype. Cd in the intercellular leaf compartment was not detectable. Higher K+ concentrations in the intercellular leaf compartment of the salt-sensitive genotype suggest a higher plasma membrane permeability caused by NaCl + Cd stress. Ascorbate peroxidase (APX) activity was increased in leaves of the salt-sensitive genotype under the combined NaCl and Cd stress. The highest non-specific peroxidase activities were detected under the combined stresses. It is suggested that NaCl and Cd stress in combination enhance the production of oxygen radicals and H2O2, especially in leaves of the salt-sensitive genotype. As a consequence, disturbed membrane function may cause elevated Cd concentrations in the intracellular leaf compartment under salinity. Cd did not change protein concentration and pattern in leaves. The protein content in inter-and intracellular leaf compartments of both genotypes was increased under salinity. A different protein pattern was obtained in inter- and intracellular leaf compartments. Thus, several physiological interactions between NaCl stress and Cd were found in the two wheat genotypes.  相似文献

High salinity is one of the most serious environmental stresses that limit crop growth. Expansins are cell wall proteins that regulate plant development and abiotic stress tolerance by mediating cell wall expansion. We studied the function of a wheat expansin gene, TaEXPA2, in salt stress tolerance by overexpressing it in tobacco. Overexpression of TaEXPA2 enhanced the salt stress tolerance of transgenic tobacco plants as indicated by the presence of higher germination rates, longer root length, more lateral roots, higher survival rates and more green leaves under salt stress than in the wild type (WT). Further, when leaf disks of WT plants were incubated in cell wall protein extracts from the transgenic tobacco plants, their chlorophyll content was higher under salt stress, and this improvement from TaEXPA2 overexpression in transgenic tobacco was inhibited by TaEXPA2 protein antibody. The water status of transgenic tobacco plants was improved, perhaps by the accumulation of osmolytes such as proline and soluble sugar. TaEXPA2‐overexpressing tobacco lines exhibited lower Na+ but higher K+ accumulation than WT plants. Antioxidant competence increased in the transgenic plants because of the increased activity of antioxidant enzymes. TaEXPA2 protein abundance in wheat was induced by NaCl, and ABA signaling was involved. Gene expression regulation was involved in the enhanced salt stress tolerance of the TaEXPA2 transgenic plants. Our results suggest that TaEXPA2 overexpression confers salt stress tolerance on the transgenic plants, and this is associated with improved water status, Na+/K+ homeostasis, and antioxidant competence. ABA signaling participates in TaEXPA2‐regulated salt stress tolerance.  相似文献

Although some plant responses to salinity have been characterized, the precise mechanisms by which salt stress damages plants are still poorly understood especially in woody plants. In the present study, the physiological and biochemical responses of Broussonetia papyrifera, a tree species of the family, Moraceae, to salinity were studied. In vitro-produced plantlets of B. papyrifera were treated with varying levels of NaCl (0, 50, 100 and 150 mM) in hydroponic culture. Changes in ion contents, accumulation of H2O2, as well as the activities and isoform profiles of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in the leaves, stems and roots were investigated. Under salt stress, there was higher Na+ accumulation in roots than in stems and leaves, and Ca2 +, Mg2 + and P3 + content, as well as K+/Na+ ratio were affected. NaCl treatment induced an increase in H2O2 contents in the tissues of B. papyrifera. The work demonstrated that activities of antioxidant defense enzymes changed in parallel with the increased H2O2 and salinity appeared to be associated with differential regulation of distinct SOD and POD isoenzymes. Moreover, SDS-PAGE analysis of total proteins extracted from leaves and roots of control and NaCl-treated plantlets revealed that in the leaves salt stress was associated with decrease or disappearance of some protein bands, and induction of a new protein band after exposure to 100 and 150 mM NaCl. In contrast, NaCl stress had little effect on the protein pattern in the roots. In summary, these findings may provide insight into the mechanisms of the response of woody plants to salt stress.  相似文献

Productivity of Indian mustard (Brassica juncea L. Czern. and Coss.) is markedly reduced by salt stress. To develop salt tolerance in this important oilseed crop is a need of the hour. This study, based on analysis of growth parameters and antioxidant profile of fourteen Indian mustard genotypes treated with 50, 100, 150 and 200 mM of sodium chloride, was performed to identify the salt-sensitive and salt-tolerant genotypes. Salinity stress inhibited biomass accumulation and reduced the protein and chlorophyll contents in a dose-dependent manner. The reduction was the highest in genotype Pusa Agrani and lowest in CS-54, depicting their contrasting sensitivity to salt stress. Salt treatments triggered a concentration-dependent overproduction of reactive-oxygen species and a concurrent upregulation of the expression of different antioxidants. Genotype CS-54 showed the least damage and maintained a high antioxidant level with almost each salt treatment, exhibiting its competence to withstand the damage provoked by salinity stress. Genotype Pusa Agrani, on the contrary, depicted a salt-sensitive nature by way of its very high lipid peroxidation and low intensity of antioxidants. These two genotypes were further investigated through gel-based proteomic approach, which resulted in the identification and quantification of 42 salinity-responsive proteins related to different metabolic modifications. Molecular processes, including photosynthesis, redox homeostasis, nitrogen metabolism, ATP synthesis, protein synthesis and degradation, signal transduction and respiratory pathways, have exhibited significant changes. The identified stress-responsive proteins could pave the way to develop salt tolerance in Indian mustard plant, thus sustaining its productivity under salinity.  相似文献

The salt tolerance of the commercial F1 tomato hybrid (Lycopersicon esculentum Mill) Radja (GC-793) has been agronomically and physiologically evaluated under greenhouse conditions, using a control (nutrient solution), a moderate (70 mM NaCl added to the nutrient solution) and a high salt level (140 mM NaCl), applied for 130 days. The results show that Radja is a Na+-excluder genotype, tolerant to moderate salinity. Fruit yield was reduced by 16% and 60% and the shoot biomass by 30% and more than 75% under moderate and high salinities, respectively. At 90 days of salt treatment (DST), the mature leaves feeding the 4th truss at fruiting accumulated little Na+ (178 mmol kg-1 DW). At this time, the sucrose concentration in these leaves even increased with moderate salinity and the amino acid proline was not accumulated under salt conditions as compared to control. At 130 DST, Na+ was accumulated mainly by the roots in proportion to the salt level applied, while in leaves appreciable amounts were found only at high salinity (452 mmol kg-1 DW). In the leaves, Cl- was always accumulated in proportion to the salt level and in a very much greater amounts than Na+ (until 1640 mmol kg-1 DW). The sucrose content was reduced in all plants by salinity, and was distributed preferentially toward the distal stem and peduncle of a truss at fruiting under moderate salinity, and toward the basal stem and root at high salinity. Moreover, proline was accumulated in different organs of the plant only at high salinity, coinciding with Na+ accumulation in leaves. Attempts are made to find a clear relationship between physiological behaviour triggered by stress and the agronomical behaviour, in order to assess the validity of physiological traits used for salt-tolerance selection and breeding in tomato.  相似文献
