Spermatozoa morphology and fine structure were studied in the grey mullet, Mugil cephalus using light, scanning and transmission electron microscopy. Light microscopy observations indicate a semi‐cystic type of spermatogenesis in the testis. The electron microscopy micrograph showed that the spermatozoon of M. cephalus is uniflagellated (total length 5.78 ± 1.26 μm), differentiated into an ovoid‐shaped head without acrosome (1.80 ± 0.35 μm in length and 1.91 ± 0.30 μm in diameter), with a short midpiece and a long cylindrical flagellar tail (length 3.60 ± 0.50 μm). The midpiece is characterized by the presence of four to five vacuoles, a cytoplasmic canal, two centriole and two spherical mitochondria having a flat type of cristae. Chromatin granules of the nucleus form an electron‐dense homogeneous mass. The flagellum consists of nine peripheral microtubules and a central pair (9 + 2) surrounded by the plasma membrane with side fins. The results confirm that spermatozoa of M. cephalus are perciform or teleostean type II. Information generated from the present study will be useful in taxonomic classification, cryopreservation and breeding work.  相似文献

Unlike the primitive type of spermatozoon found in most polychaetes, the spermatozoon of Autolytus has a bilateral symmetry with elongated nucleus, and the mitochondria surround the posterior part of the nucleus. A rather large disk-shaped acrosome is situated along one side of the anterior part of the nucleus. From the anterior margin of the distal centriole emerge long striated rootlets, which run along the nuclear envelope to the anterior part of the nucleus. The spermatozoon of Chitinopoma serrula has an elongated, slightly bent nucleus, a thimble-like acrosome apically on the anterior surface of the nucleus, and an elongated middle piece containing 4 rod-like mitochondria developed from spherical mitochondria surrounding the basal part of the tail flagellum. In the spermatozoon of Capitella capitata, both nucleus and middle piece are elongated compared to the primitive type. The large and conical acrosome is placed asymmetrically at the nucleus and consists of an acrosomal vesicle and subacrosomal substance. The greater part of the middle piece forms a collar around the initial part of the tail flagellum. The cytoplasm of the collar contains granular material. One or two small mitochondria lie around the 2 centrioles at the base of the nucleus.

These types of spermatozoa represent early steps in the evolution of modified spermatozoa combined with changed biology of reproduction. The modified spermatozoa are larger than the primitive ones.  相似文献

The spermatozoon of the parasitoid wasp Cotesia congregata is an extremely short gamete measuring less than 7 μm; it is as yet the shortest flagellated sperm to be identified. The mature sperm consists of an acrosome, surrounded by an extra cellular coat, a condensed nucleus, two uncoiled mitochondrial derivatives and a short axoneme. Testes of young adults contain a continuum of differentiation stages. Initially, the flagellum is approximately 5 μm long. It conserves its length in round, elongated and mature spermatids, but is reduced to less than 3 μm in mature spermatozoa. The nucleus is 2 μm in diameter when round, 10 μm long when it becomes a long boat-hull shaped filament, and then reduces to 3.6 μm. Thus, during development the gamete reaches a total length of 15 μm before finally reducing to less than half that length. Some traits of mature sperm anatomy are similar to related species of the Braconidae family, but others seem to be specific and could be due to the shortness of the cell. This uncommon elongation and subsequent shortening of such a tiny flagellated cell constitutes a model for both nucleus and cilium development.  相似文献

The spermatozoa of Bephratelloides pomorum are very long and fine. Each spermatozoon measures about 620 μm in length by 0.38 μm in diameter and, when seen under the light microscope, appears to be wavy along its entire length. The head, which is approximately 105 μm, comprises a small acrosome and a nucleus. The acrosome is made up of a cone-shaped acrosomal vesicle surrounding the perforatorium and the anterior end of the nucleus. Innumerable filaments radiate from it. The perforatorium has a diameter equal to that of the nucleus at their junction, where it fits with a concave base onto the rounded nuclear tip. The nucleus is helicoidal and completely filled with homogeneous compact chromatin. It is attached to the tail by a very long and quite electron-dense centriolar adjunct that extends anteriorly from the centriole in a spiral around the nucleus for approximately 8.5 μm. The tail consists of an axoneme with the 9+9+2 microtubule arrangement pitched in a long helix, as well as a pair of spiraling mitochondrial derivatives (with regularly arranged cristae) that coil around the axoneme, and two small accessory bodies. As well as the spiraling of the nucleus, mitochondrial derivatives and axonemal microtubules, the sperm of B. pomorum present other very different morphological features. These features include the acrosome and centriolar adjunct, both of which differentiate the spermatozoa from the majority of sperm found in other Hymenoptera. In addition these structural variations demonstrate that the sperm of chalcidoids provide characteristics that can certainly prove useful for future phylogenetic analysis at the subfamily level and, possibly, the genus too.  相似文献

Lates niloticus is a valuable commercial fish species with good potential for aquaculture. However, there is limited information on the type and structure of the Nile perch spermatozoon, which could potentially aid in culture of this species. Here, we describe the spermatozoon ultrastructure in L. niloticus using transmission and scanning electron microscopy. The spermatozoon had a round head-shape, medio-laterally flat, no acrosome, a short midpiece located laterally to the nucleus, uniflagella with one wing. The head of the spermatozoon contained the nucleus, centriolar system, proximal part of the flagellum, and cytoplasmic channel. Centrioles were arranged at an angle of 90° to each other, forming a T-shape, parallel to the nucleus. The midpiece was cylindrical, loaded with cytoplasm, five to seven spherical mitochondria; and the flagellum’s plasma membrane extended to form one lateral wing. The spermatozoa were classified as type II spermatozoa. L. niloticus spermatozoon differed from that of its Australian congener L. calcarifer, especially in the centriole arrangement and nuclear shape, length of the midpiece and the number of mitochondria and lateral wings.  相似文献

本文研究卵胎生硬骨鱼褐菖(Sebastiscusmarmoratus)精细胞的成熟变化和精子结构。褐菖精细胞发育晚期已具有硬骨鱼类精子的结构雏形:细胞核的背面较平坦,腹面稍外鼓,呈弧面;染色质浓缩成团块状,核的腹侧和后端的染色质较致密;中心粒复合体由近端中心粒和基体组成,近端中心粒和基体排成“L”形;近端中心粒向细胞核的背侧伸出中心粒附属物,中心粒附属物由9条微管组成,9条微管围成一筒状结构,类似轴丝。在晚期精细胞形成精子的过程中,中心粒附属物和近端中心粒相继退缩以至消失不见,同时细胞核后端的形状也随着发生变化。中心粒附属物和近端中心粒的相继消失可以看作是成熟的最后标志。精子的中心粒复合体由基体及其上方的基体帽组成,袖套接于核的后端,其中约有30~40个线粒体;鞭毛从袖套腔中伸出,鞭毛的中心结构是轴丝;轴丝外方为细胞质形成的侧鳍,在鞭毛的近核段,轴丝两侧的侧鳍较宽且不对称。  相似文献

This study describes morphology and fine structure of the Persian sturgeon (Acipenser persicus) (Acipenseridae, Chondrostei) spermatozoon. The results show that the spermatozoon of A. persicus is differentiated into an elongated head (length: mean±SD: 7.1±0.5μm) with an acrosome (length: 1.2±0.2μm), a cylindrical midpiece (length: 1.8±0.5μm), a flagellum (length: 50.3±5.9μm) and a total length of 59.2±6.2μm. Ten posterolateral projections (PLPs) arise from the posterior edge of the acrosome and there were 3 endonuclear canals that traversed the nucleus from the acrosomal end to the basal nuclear fossa region. Three to six mitochondria were in peripheral midpiece and the proximal and distal centrioles were located near to "implantation fossa" and basement of the flagellum. The axoneme has a typical eukaryotic structure composed of 9 peripheral microtubules and a central pair of single microtubule surrounded by the plasma membrane. Lateral fins were observed along the flagellum. The fins started and ended at 0.5-1μm from midpiece and at 4-6μm from the end of flagellum. There were significant differences in the size of almost all measured morphological parameters between males and flagellar, midpiece and nucleus characters were more isolated parameters that can be considered for detecting inter-individual variations. This study showed that sperm morphology and fine structure are similar among sturgeon species, but the dimensions of the parameters may differ.  相似文献

The Chinese sturgeon (Acipenser sinensis Gray 1835) is an endangered anadromous sturgeon inhabiting the Yangtze River in China. In this study, the ultrastructure and morphology of spermatozoa was studied using transmission and scanning electron microscopy with a cryo-holder. The spermatozoon consisted of an elongated head with a distinct acrosome and nucleus region, a midpiece and a flagellum. The mean length of the head and midpiece, the flagellum and total length of spermatozoon were 4.48, 33.3 and 37.8 microm, respectively. The nucleus was an elongated trapezoid shape with anterior (acrosome) end narrower than the posterior. Granular material and an actin filament were observed within the anterior acrosome. Three to five endonuclear canals were present. The midpiece was eudipleural along its longitudinal axis. Compared to other sturgeon species, the data from the present study suggest a more recent evolutionary linkage between Chinese sturgeon and white sturgeon (Acipenser transmontanus Richardson 1836).  相似文献

In this paper spermatogenesis and sperm ultrastructure of the cockle Anadara granosa are studied using transmission electron microscopy. The spermatocyte presents electron-dense vesicles and the arising axoneme that begins to form the flagellum. During spermatid differentiation, proacrosomal vesicles appear to migrate towards the presumptive anterior pole of the nucleus; eventually these vesicles become acrosome. The spermatozoon of Anadara granosa is of the primitive type. The acrosome, situated at the apex of the nucleus, is cap-shaped and deeply invaginated at the inner side. The spherical nucleus of the spermatozoon contains dense granular chromatin and shows invagination at the posterior poles. The centriole shows the classic nine triplets of microtubules. The middle piece consists of the centriolar complex surrounded by five giant mitochondria. It is shown that the ultrastructure of spermatozoa and spermiogenesis of Anadara granosa reveals a number of features that are common among bivalves. Abstract Spermatogenesis in Marenzelleria viridis was studied by ultrastructural investigation. The testes are formed on the greatly ramified nephridial blood vessel and are enveloped by a thin layer of peritoneal cells. The spermatogonia vary in shape, are about 10 μm in diameter and are not linked by intercellular bridges. Pairs or tetrads of spermatocytes connected by intercellular bridges float freely in the coelomic cavity. A complex acrosome is produced by a Golgi complex. The acrosome consists of four to five different structures, forms cisternae and, in the mature spermatozoon, lies deep in an invagination of the nucleus. Two centrioles are also situated in a deep centriolar fossa, the proximal centriole being perpendicular to the distal one. The mature spermatozoon is an ect-aquasperm measuring about 5 μm in length and 2.5 μm in width. The midpiece consists of five spherical mitochondria arranged around the axoneme behind the nucleus. The axoneme is connected to the plasma membrane by a satellite complex. The microtubules of the flagellum are arranged in a typical 9 × 2 + 2 configuration. The fine structure of the spermatozoa of two spionids is described. The spermatozoon of Prionospio cf. queenslandica is typical of an animal utilizing external fertilization, in having a subspheroidal nucleus, a midpiece composed of unmodified rounded mitochondria surrounding two centrioles and a free flagellum. The acrosome is unusual in showing bilateral symmetry. The spermatozoon of Tripolydora sp. resembles that of spionids utilizing spermatophores, in possessing an extremely elongate nucleus and midpiece. The nucleus is penetrated by the 9+2 axoneme for its entire length, linking with a single centriole at the anterior end. Platelets surround the nucleus and intermingle with the mitochondria of the midpiece, which terminates with an annulus. The acrosome shows some internal vesiculation and substructuring. Sperm structure in relation to reproductive methods is discussed and the view of external fertilization as primitive is questioned.  相似文献

The ultrastructure of the sperm of the common bivalve species Mercenaria stimpsoni and Mactra chinensis from Peter the Great Bay is described. The sperm structure is typical for animals with external insemination. The sperm consists of a head, middle part, and flagellum. The sperm head of M. stimpsoni has a curved crescent form and includes the nucleus and acrosome; the head length is 9.8 μm. The acrosome is subdivided to the acrosome granule and the periacrosomal material. There are 4 mitochondria of about 0.8 μm in size in the middle part of the spermatozoon. The mitochondria surround the centriolar apparatus, which consists of proximal and distal centrioles located at a right angle. The axoneme originates from the distal centriole. The sperm of M. chinensis is barrel-shaped, with a head length of 3.2 μm. The acrosome is relatively larger, and its height is 1–1.2 μm. There are also 4 mitochondria 0.6–0.8 μm in the middle part of the spermatozoon. The sperm structure of the described species is typical of the families to which the mollusks belong, with insignificant variations.  相似文献

This study describes the sperm morphology of the mayfly Hexagenia (Pseudeatonica) albivitta (Ephemeroptera). Its spermatozoon measures approximately 30 μm of which 9 μm corresponds to the head. The head is composed of an approximately round acrosomal vesicle and a cylindrical nucleus. The nucleus has two concavities, one in the anterior tip, where the acrosomal vesicle is inserted and a deeper one at its base, where the flagellum components are inserted. The flagellum is composed of an axoneme, a mitochondrion and a dense rod adjacent to the mitochondrion. A centriolar adjunct is also observed surrounding the axoneme in the initial portion of the flagellum and extends along the flagellum for at least 2 μm, surrounding the axoneme in a half‐moon shape. The axoneme is the longest component of the flagellum, and it follows the 9+9+0 pattern, with no central pair of microtubules. At the posterior region of the flagellum, the mitochondrion has a dumb‐bell shape in cross sections that, together with the rectangular mitochondrial‐associated rod, is responsible for the flattened shape of the flagellum. An internal membrane is observed surrounding both mitochondrion and its associated structure.  相似文献

Summary Sperm ultrastructure and spermiogenesis of the enteropneust hemichordate Saxipendium coronatum conforms to the general pattern of the prototype spermatozoon found in many phyla. The sperm is about 29 m long, including head, middle piece, and tail. The Saxipendium spermatozoon has some unique features. The head is pyramidal in shape and the nucleus has four frontal ridges radiating from the base of the acrosomal region. The acrosome is composed of a large acrosomal vesicle surrounded by periacrosomal material. The acrosomal region projects about 1 m in front of the nucleus and has a width at the base of 1.5 m. The middle piece is dish-shaped and contains a large mitochondrial mass surrounding the centriolar region. The centriolar region is partially located in a centriolar fossa at the basal part of the nucleus. In spermatids, an anchoring fiber apparatus is observed surrounding the centriolar region. The distal ends of the fibers are attached to the plasmalemma by electron-dense thickenings. The tail is a simple flagellum. The sperm of Saxipendium and the small eggs found in the female suggest non-specialized external fertilization and embryogeny leading to a planktotrophic larva. 通过电光学显微镜和透射电子显微镜观察了平疣桑椹石磺精子的形态及其超微结构。平疣桑椹石磺成熟精子属于进化型,由头部、中段和末段组成。头部由顶体和精核构成,顶体长约0.7μm,呈细奶嘴状,内含物分布均匀,电子密度稍低于细胞核。顶体基部与精核前端紧密相连,无间隙。精核长约3.8μm,宽约1.0μm,核质高度浓缩,电子密度高,无核泡,纵切似辣椒状,核后端内凹形成核后窝。中段加长,结构复杂,线粒体演化成线粒体鞘,螺旋状包绕轴丝。精子末段由轴丝及包绕轴丝的质膜组成,轴丝为典型的“9+2”结构。比较了平疣桑椹石磺精子与相关腹足类精子结构的异同,进一步证实了腹足纲贝类精子结构之间的区别主要在于顶体有无及形态,精核的长短与外形、中段线粒体的数目及其排列方式等。  相似文献

The ultrastructure of the spermatid and spermatozoon of Macracanthorhynchus hirudinaceus (Archiacanthocephala) was studied by means of transmission electron microscopy. The flagellum and nucleus in the spermatid gradually expanded simultaneously. The karyoplasma of the spermatid transformed into dense inclusions and a multibarrel structure, which were also found in the spermatozoan body. The multibarrel structure was located close to the flagellum and consisted of many irregular microtubes. The flagellum of the developing spermatozoon was observed in a concavity of the spermatid nucleus. The microtubule arrangement of the flagellum was "9 + 2". No mitochondria or acrosome were observed in spermatozoa.  相似文献

We describe sperm ultrastructure and acrosome differentiation during spermiogenesis in Crassostrea gigas (Mollusca Bivalvia). The sperm cell is a uniflagellated cell of the primitive type. The head region contains a rounded or conical nucleus surmounted by small acrosome. This organelle consists of a membrane-bound acrosomal granule, the contents of which have a homogeneous density, except in the anterior region, which is positive for PTA. The acrosome also surrounds the perforatorium, which includes oriented fibrillar elements: this is the axial body. The middle piece contains four mitochondria encircling two perpendicular centrioles. The distal centriole is provided with a system of mechanical fixation to the plasma membrane, consisting of nine fibers in radial arrangement.
