The ability of brown-rot fungi (BRF) to eliminate DDT in artificially and historically contaminated soil was investigated to determine whether the BRF would be suitable for the bioremediation of DDT in soil. Gloeophyllum trabeum, Fomitopsis pinicola and Daedalea dickinsii showed an ability to eliminate DDT in artificially contaminated sterilized (SL) and un-sterilized (USL) soils. The addition of Fe2+ to the soil system enhanced the ability of some BRF to eliminate DDT. In the contaminated SL soil, the DDT was eliminated by approximately 41%, 9% and 15% by G. trabeum, F. pinicola and D. dickinsii, respectively. Compared with the controls, in the USL soil approximately 43%, 29% and 32% of DDT was eliminated and approximately 20%, 9% and 26% of DDD (1,1-dichloro-2,2-bis (4-chlorophenyl) ethane) was detected as a metabolic product with G. trabeum, F. pinicola and D. dickinsii, respectively. Of the BRF, G. trabeum demonstrated the greatest ability to eliminate DDT both in the SL and USL soils. G. trabeum was applied to a historically contaminated soil which had a DDT concentration more than three times the artificially contaminated soil. G. trabeum remediated about 64% of the initial DDT with the addition of Fe2+. There were no significant differences in the results with or without the addition of Fe2+, indicating that G. trabeum can be used directly for the degradation of DDT in soil without any other additional treatment. This study identified that G. trabeum is the most promising BRF for use in the bioremediation of DDT contaminated soil.  相似文献

The insecticide 1,1,1-trichloro-2,2-bis-(4-chlorophenyl)ethane (DDT) has been used extensively since the 1940s for control of agricultural pests, and is still used in many tropical countries for mosquito control. Despite a ban on DDT use in most industrialized countries since 1972, DDT and its related residues (DDTr) persist in the environment and pose animal and human health risks. Abiotic processes such as volatilization, adsorption, and photolysis contribute to the dissipation of DDTr in soils, often without substantial alteration of the chemical structure. In contrast, biodegradation has the potential to degrade DDTr significantly and reduce soil concentrations in a cost-effective manner. Many bacteria and some fungi transform DDT, forming products with varying recalcitrance to further degradation. DDT biodegradation is typically co-metabolic and includes dechlorination and ring cleavage mechanisms. Factors that influence DDTr biodegradation in soil include the composition and enzymatic activity of the soil microflora, DDTr bioavailability, the presence of soil organic matter as a co-metabolic substrate and (or) inducer, and prevailing soil conditions, including aeration, pH, and temperature. Understanding how these factors affect DDTr biodegradation permits rational design of treatments and amendments to stimulate biodegradation in soils. The DDTr-degrading organisms, processes and approaches that may be useful for bioremediation of DDTr-contaminated soils are discussed, including in situ amendments, ex situ bioreactors and sequential anaerobic and aerobic treatments.  相似文献

White-rot fungi possess a unique oxidative mechanism by which the recalcitrant lignin component of wood is mineralised. The activity of lignin-degrading enzymes, chiefly lignin and manganese peroxidases, depends on several small organic molecules. Some of these (e.g. chloroanisyl alcohols) are chloroaromatics and may act as environmental pollutants in the forest soil, whereas the synthesis of others (e.g. veratryl alcohol) requires chloromethane. Certain white-rot genera, notably Phellinus and Inonotus, release excess quantities of chloromethane into the atmosphere where it acts as a greenhouse gas. On the other hand, their powerful ligninolytic system enables white-rot fungi to degrade a wide range of man-made environmental pollutants, including recalcitrant chloroaromatics such as DDT, PCP, 2,4-D and 2,4,5-T. This review describes the multifarious interactions of white-rot fungi with their environment via the chlorine cycle.  相似文献

Endophytic fungi, mostly belonging to the Ascomycota, are found in the intercellular spaces of the aerial plant parts, particularly in leaf sheaths, sometimes even within the bark and root system without inducing any visual symptoms of their presence. These fungi appear to have a capacity to produce a wide range of enzymes and secondary metabolites exhibiting a variety of biological activities. However, they have been only barely exploited as sources of enzymes of industrial interest. This review emphasizes the suitability and possible advantages of including the endophytic fungi in the screening of new enzyme producing organisms as well as in studies aiming to optimize the production of enzymes through well-known culture processes. Apparently endophytic fungi possess the two types of extracellular enzymatic systems necessary to degrade the vegetal biomass: (1) the hydrolytic system responsible for polysaccharide degradation consisting mainly in xylanases and cellulases; and (2) the unique oxidative ligninolytic system, which degrades lignin and opens phenyl rings, comprises mainly laccases, ligninases and peroxidases. The obvious ability of endophytic fungi to degrade the complex structure of lignocellulose makes them useful in the exploration of the lignocellulosic biomass for the production of fuel ethanol and other value-added commodity chemicals. In addition to this, endophytic fungi may become new sources of industrially useful enzymes such as lipases, amylases and proteases.  相似文献

Microbial pesticide degradation studies have until now mainly focused on bacteria, although fungi have also been shown to degrade pesticides. In this study we clarify the background for the ability of the common soil fungus Mortierella to degrade the phenylurea herbicide diuron. Diuron degradation potentials of five Mortierella strains were compared, and the role of carbon and nitrogen for the degradation process was investigated. Results showed that the ability to degrade diuron varied greatly among the Mortierella strains tested, and the strains able to degrade diuron were closely related. Degradation of diuron was fastest in carbon and nitrogen rich media while suboptimal nutrient levels restricted degradation, making it unlikely that Mortierella utilize diuron as carbon or nitrogen sources. Degradation kinetics showed that diuron degradation was followed by formation of the metabolites 1-(3,4-dichlorophenyl)-3-methylurea, 1-(3,4-dichlorophenyl)urea and an hitherto unknown metabolite suggested to be 1-(3,4-dichlorophenyl)-3-methylideneurea.  相似文献

Lignin is an abundant plant-based biopolymer that has found applications in a variety of industries from construction to bioethanol production. This recalcitrant branched polymer is naturally degraded by many different species of microorganisms, including fungi and bacteria. These microbial lignin degradation mechanisms provide a host of possibilities to overcome the challenges of using harmful chemicals to degrade lignin biowaste in many industries. The classes and mechanisms of different microbial lignin degradation options available in nature form the primary focus of the present review. This review first discusses the chemical building blocks of lignin and the industrial sources and applications of this multifaceted polymer. The review further places emphasis on the degradation of lignin by natural means, discussing in detail the lignin degradation activities of various fungal and bacterial species. The lignin-degrading enzymes produced by various microbial species, specifically white-rot fungi, brown-rot fungi, and bacteria, are described. In the end, possible directions for future lignin biodegradation applications and research investigations have been provided.  相似文献

The aim of this study was to select a bacterial strain able to degrade 1,1,1-trichloro-2,2-bis(4-chlorophenyl) ethane (DDT), and to use it for bioaugmentation in order to decontamination soil. Advenella Kashmirensis MB-PR (A. Kashmirensis MB-PR) was isolated from DDT contaminated soil, and the degradation ability of DDT by this strain in the mineral salt medium was screened by gas chromatography. The efficiency of degradation was 81% after 30 days of bacterial growth. The study of intermediary products during the degradation of DDT showed the appearance and accumulation of DDD and DDE, which emerged from the first days of the experiment. Other metabolites were detected at a lower number of chlorine atoms, such as DBH. DNA samples were isolated and screened for the linA gene, encoding dehydrochlorinase. The bioaugmentation by A. Kashmirensis MB-PR of polluted sterile soil showed that 98% of DDT disappeared after 20 days of experience. This study demonstrates the significant potential use of A. Kashmirensis MB-PR for the bioremediation of DDT in the environment.  相似文献

Over the past 30 years, research on the microbial degradation of polycyclic aromatic hydrocarbons (PAHs) has resulted in the isolation of numerous genera of bacteria, fungi and algae capable of degrading low molecular weight PAHs (compounds containing three or less fused benzene rings). High molecular weight PAHs (compounds containing four or more fused benzene rings) are generally recalcitrant to microbial attack, although some fungi and algae are capable of transforming these compounds. Until recently, only a few genera of bacteria have been isolated with the ability to utilise four-ring PAHs as sole carbon and energy sources while cometabolism of five-ring compounds has been reported. The focuss of this review is on the high molecular weight PAH benzo[a]pyrene (BaP). There is concern about the presence of BaP in the environment because of its carcinogenicity, teratogenicity and toxicity. BaP has been observed to accumulate in marine organisms and plants which could indirectly cause human exposure through food consumption. This review provides an outline of the occurrence of BaP in the environment and the ability of bacteria, fungi and algae to degrade the compound, including pathways for BaP degradation by these organisms. In addition, approaches for improving microbial degradation of BaP are discussed.  相似文献

Zinc oxide (ZnO) has several industrial applications due to its versatile properties, which lead to its continuously increasing demand in different industrial sectors. Additionally, ZnO nanostructures possess unique photocatalytic activity, and because of this, they are being applied to degrade organic dyes through photocatalysis for wastewater treatment. Nevertheless, chemical synthesis methods to develop ZnO nanostructures have raised concerns related to environmental issues, furthermore, these methods are found to be costly and tedious. As a result, the synthesis of ZnO nanostructures using green methods is gaining popularity due to its low cost and eco-friendly mode, while avoiding the use of toxic chemicals. Green synthesis of ZnO nanostructures using different biological approaches involving plants, algae, and different microorganism-derived bioactive compounds has been well reported for diverse applications. Among different applications, ZnO nanostructures that enable photocatalysis to degrade dye have been found to be imperative for wastewater treatments. Therefore, the current review explores recent studies on green synthesis approaches to prepare ZnO nanostructures via adopting different biological methods that rely on plants, algae, and bacterial microorganisms. The properties of ZnO nanostructures, along with their green synthesis routes and feasible mechanisms, have also been discussed in this review. This review focuses on the use and efficiency of green route synthesized ZnO nanostructures as nanophotocatalysts for the degradation of organic dyes in wastewater treatment. Additionally, existing challenges in green synthesis methods and the efficiency of ZnO nanostructures to degrade organic dyes following photocatalysis has been discussed.  相似文献

Chlorinated nitroaromatic compounds (CNAs) are persistent environmental pollutants that have been introduced into the environment due to the anthropogenic activities. Bacteria that utilize CNAs as the sole sources of carbon and energy have been isolated from different contaminated and non-contaminated sites. Microbial metabolism of CNAs has been studied, and several metabolic pathways for degradation of CNAs have been proposed. Detoxification and biotransformation of CNAs have also been studied in various fungi, actinomycetes and bacteria. Several physicochemical methods have been used for treatment of wastewater containing CNAs; however, these methods are not suitable for in situ bioremediation. This review describes the current scenario of the degradation of CNAs.  相似文献

Operational cellulose acetate reverse-osmosis membranes were examined for evidence of biological degradation. Numerous fungi and bacteria were isolated by direct and enrichment techniques. When tested, most of the fungi were active cellulose degraders, but none of the bacteria were. Neither fungi nor bacteria were able to degrade cellulose acetate membrane in vitro, although many fungi were able to degrade cellulose acetate membrane after it had been deacetylated. Organisms did not significantly degrade powdered cellulose acetate in pure or mixed cultures as measured by reduction in acetyl content or intrinsic viscosity or production of reducing sugars. Organisms did not affect the performance of cellulose triacetate fibers when incubated with them. The inability of the organisms to degrade cellulose acetate was attributed to the high degree of acetate substitution of the cellulose polymer. The rate of salt rejection decline was strongly correlated with chlorination of feed water and inversely with densities of microorganisms. These data suggest that microbial degradation of operational cellulose acetate reverse-osmosis membranes is unlikely.  相似文献

Pulp and paper mill effluents pollute water, air and soil, causing a major threat to the environment. Several methods have been attempted by various researchers throughout the world for the removal of colour from pulp and paper mill effluents. The biological colour removal process uses several classes of microorganisms--bacteria, algae and fungi--to degrade the polymeric lignin derived chromophoric material. White rot fungi such as Phanerochaete chrysosporium, Corius versicolor, Trametes versicolor etc., are efficient in decolourizing paper and pulp mill effluents. Gliocladium virens, a saprophytic soil fungus decolourised paper and pulp mill effluents by 42% due to the production of hemicellulase, lignin peroxidase, manganese peroxidase and laccase.  相似文献

农用化学品是农业生产的重要组成部分,其安全问题也成为人们关注的焦点。农用化学品可分为化肥、农药、农膜3类。化肥主要用于提高土壤肥力,增加农作物产量,不合理使用则会改变土壤性质,降低土壤肥力,并且对生态环境以及人类健康造成不可忽视的危害。农药主要用于杀菌、杀虫、除草等,在促进和保障农作物健康生长中发挥重要作用,但农药的滥用造成农药事故频发,农药的毒性富集作用以及残留问题越发引起人们的关注。农膜主要用于农膜覆盖栽培技术,农膜的原料主要是聚乙烯树脂等高分子化学物质,在土壤中很难降解,造成的环境污染问题日益突出。各种农用化学品的不合理使用,可降低农产品的质量,使其在出口贸易中受到极大限制,造成巨大的经济损失。本文在肯定农用化学品在现代化农业生产中的巨大贡献的同时,深入分析了其污染状况及危害,总结了当前国际上对于化肥、农药、农膜的研究进展以及所取得的成果,最后提出了科学使用农用化学品的建议。  相似文献

ABSTRACT Polycyclic aromatic hydrocarbons (PAHs) are present in products made from creosote, coal tar, and asphalt. When wood pile treated with creosote is placed in soil, PAHs can contaminate it. Creosote has been used for wood preservation in the past and is composed of approximately 85% PAHs and 15% phenolic compounds. PAHs cause harmful effects to humans and the environment because of their carcinogenic and mutagenic properties. White rot fungi can degrade not only lignin, but also recalcitrant organic compounds such as PAHs. Among numerous white rot fungi used in previous studies, four species were selected to degrade PAHs in a liquid medium. From this evaluation of the degradation of PAHs by the four fungal isolates, two species were ultimately selected for the highest rates of removal. Following 2 weeks of incubation with Peniophora incarnata KUC8836, the degradation rates of phenanthrene, fluoranthene, and pyrene were 86.5%, 77.4%, and 82.6%, respectively. Mycoaciella bispora KUC8201 showed the highest degradation rate for anthracene (61.8%). Hence, bioremediation of creosote-contaminated soil with an initial concentration of 229.49 mg kg?1 PAHs was carried out using the two selected fungi because they could simultaneously degrade 13 more PAHs than the comparison species. More importantly, isolates of P. incarnata KUC8836 were discovered as powerful degraders of PAHs by producing laccase and manganese-dependent peroxidase (MnP), with 1.7- and 1.1-fold higher than the comparison species, respectively. Therefore, the white rot fungus may be proposed for the removal of PAHs and xenobiotic compounds in contaminated environments.  相似文献

Rhizobium leguminosarum secretes two extracellular glycanases, PlyA and PlyB, that can degrade exopolysaccharide (EPS) and carboxymethyl cellulose (CMC), which is used as a model substrate of plant cell wall cellulose polymers. When grown on agar medium, CMC degradation occurred only directly below colonies of R. leguminosarum, suggesting that the enzymes remain attached to the bacteria. Unexpectedly, when a PlyA-PlyB-secreting colony was grown in close proximity to mutants unable to produce or secrete PlyA and PlyB, CMC degradation occurred below that part of the mutant colonies closest to the wild type. There was no CMC degradation in the region between the colonies. By growing PlyB-secreting colonies on a lawn of CMC-nondegrading mutants, we could observe a halo of CMC degradation around the colony. Using various mutant strains, we demonstrate that PlyB diffuses beyond the edge of the colony but does not degrade CMC unless it is in contact with the appropriate colony surface. PlyA appears to remain attached to the cells since no such diffusion of PlyA activity was observed. EPS defective mutants could secrete both PlyA and PlyB, but these enzymes were inactive unless they came into contact with an EPS(+) strain, indicating that EPS is required for activation of PlyA and PlyB. However, we were unable to activate CMC degradation with a crude EPS fraction, indicating that activation of CMC degradation may require an intermediate in EPS biosynthesis. Transfer of PlyB to Agrobacterium tumefaciens enabled it to degrade CMC, but this was only observed if it was grown on a lawn of R. leguminosarum. This indicates that the surface of A. tumefaciens is inappropriate to activate CMC degradation by PlyB. Analysis of CMC degradation by other rhizobia suggests that activation of secreted glycanases by surface components may occur in other species.  相似文献

To identify regulators of AU-rich element (ARE)-dependent mRNA turnover we have followed a genetic approach using a mutagenized cell line (slowC) that fails to degrade cytokine mRNA. Accordingly, a GFP reporter construct whose mRNA is under control of the ARE from interleukin-3 gives an increased fluorescence signal in slowC. Here we describe rescue of slowC by a retroviral cDNA library. Flow cytometry allowed us to isolate revertants with reconstituted rapid mRNA decay. The cDNA was identified as butyrate response factor-1 (BRF1), encoding a zinc finger protein homologous to tristetraprolin. Mutant slowC carries frame-shift mutations in both BRF1 alleles, whereas slowB with intermediate decay kinetics is heterozygous. By use of small interfering (si)RNA, independent evidence for an active role of BRF1 in mRNA degradation was obtained. In transiently transfected NIH 3T3 cells, BRF1 accelerated mRNA decay and antagonized the stabilizing effect of PI3-kinase, while mutation of the zinc fingers abolished both function and ARE-binding activity. This approach, which identified BRF1 as an essential regulator of ARE-dependent mRNA decay, should also be applicable to other cis-elements of mRNA turnover.  相似文献

1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane (DDT) was used as the substrate for a degradation experiment with the white rot fungi Phlebia lindtneri GB-1027 and Phlebia brevispora TMIC34596, which are capable of degrading polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated biphenyls (PCBs). Pure culture of P. lindtneri and P. brevispora with DDT (25 μmol l−1) showed that 70 and 30% of DDT, respectively, disappeared in a low-nitrogen medium after a 21-day incubation period. The metabolites were analyzed using gas chromatography/mass spectrometry (GC/MS). Both fungi metabolized DDT to 1,1-dichloro-2,2-bis(4-chlorophenyl)ethane (DDD), 2,2-bis(4-chlorophenyl)acetic acid (DDA) and 4,4-dichlorobenzophenone (DBP). Additionally, DDD was converted to DDA and DBP. DDA was converted to DBP and 4,4-dichlorobenzhydrol (DBH). While DBP was treated as substrate, DBH and three hydroxylated metabolites, including one dihydroxylated DBP and two different isomers of monohydroxylated DBH, were produced from fungal cultures, and these hydroxylated metabolites were efficiently inhibited by the addition of a cytochrome P-450 inhibitor, piperonyl butoxide. These results indicate that the white rot fungi P. lindtneri and P. brevispora can degrade DBP/DBH through hydroxylation of the aromatic ring. Moreover, the single-ring aromatic metabolites, such as 4-chlorobenzaldehyde, 4-chlorobenzyl alcohol and 4-chlorobenzoic acid, were found as metabolic products of all substrate, demonstrating that the cleavage reaction of the aliphatic-aryl carbon bond occurs in the biodegradation process of DDT by white rot fungi.  相似文献

Bioremediation is a spontaneous or controlled process in which biological, mainly microbiological, methods are used to degrade or transform contaminants to non or less toxic products, reducing the environmental pollution. The most important parameters to define a contaminated site are: biodegradability, contaminant distribution, lixiviation grade, chemical reactivity of the contaminants, soil type and properties, oxygen availability and occurrence of inhibitory substances. Biological treatments of organic contaminations are based on the degradative abilities of the microorganisms. Therefore the knowledge on the physiology and ecology of the biological species or consortia involved as well as the characteristics of the polluted sites are decisive factors to select an adequate biorremediation protocol. Basidiomycetes which cause white rot decay of wood are able to degrade lignin and a variety of environmentally persistent pollutants. Thus, white rot fungi and their enzymes are thought to be useful not only in some industrial process like biopulping and biobleaching but also in bioremediation. This paper provides a review of different aspects of bioremediation technologies and recent advances on ligninolytic metabolism research.  相似文献

Bioremediation of DDT in soil by genetically improved recombinants of the soil fungus Fusarium solani was studied. The parent strains were isolated from soil enriched with DDD or DDE (immediate anaerobic and aerobic degradation products of DDT), as further degradation of these products are slow processes compared to the parent compound. These naturally occurring strains isolated from soil, however, are poor degraders of DDT and differed in their capability to degrade its metabolites such as DDD, DDE, DDOH and DBP and other organochlorine pesticides viz. kelthane and lindane. Synergistic effect was shown by some of these strains, when grown together in the medium containing DDD and kelthane under mixed culture condition. No synergism in DDE degradation was observed with the strains isolated from enriched soil. DDD-induced proteins extracted from individual culture filtrate (exo-enzyme) when subjected to SDS-Polyacrylamide Gel Electrophoresis (SDS-PAGE) showed complementary polypeptide bands in these strains i.e., each strain produced distinct DDD degrading polypeptide bands and the recombinant or hybrid strains produced all of the bands of the two parents and degraded DDD better than the parental strains. Recombinant hybrid strains with improved dehalogenase activity were raised by parasexual hybridisation of two such complementary isolates viz. isolate 1(P-1) and 4(P-2) showing highest complementation and are compatible for hyphal fusion inducing heterokaryosis. These strains are genetically characterised as Kel+BenRDBP-Lin- and Kel-BenrDBP+Lin+ respectively.Recombinants with mixed genotype, i.e., Kel+BenRDBP+Lin+ showing superior degradation quality for DDT were selected for bioremediation study. Recombination was confirmed by polypeptide band analysis of DDD induced exo-proteins from culture filtrate usingSDS-Polyacrylamide Gel Electrophoresis (PAGE) and RAPD (Random Amplified Polymorphic DNA) of genomic DNA using PCR (Polymerase Chain Reaction) technique. SDS-PAGE showed combination of DDD induced polypeptide bands characteristic of both the parents in the recombinants or the hybrids. PCR study showed the parent specific bands in the recombinant strains confirming gene transformation.  相似文献
