Anatomical changes in the shoot apex of Chenopodium rubrum L.treated with an extract from flowering tobacco plants and cultivatedin non-inductive conditions are described. They are comparedwith the anatomy of non-treated vegetative apices and with apicesof plants induced with a short day. Treatment with the extractresulted in both activation of cell division in the upper partof the apex and in apex elongation. Acceleration of leaf primordiainitiation and stimulation of branching took place. The effectcorresponds to the sequence of changes in photoperiodically-inducedplants but is more pronounced. Elongation following 10–4M GA3 treatment was of a differentnature; there was only a slight stimulation in the upper partof the apex in contrast with a strong stimulation of growthin length in the lower internodes. These preliminary resultssuggest a similarity between apical changes evoked by a stimulusproduced by short days and an exogenously applied floral stimulus.The changes differed from those caused by exogenous phytohormones. Indol-3yl acetic acid (10–4M) was applied to the plumulesof Chenopodium rubrum. Effects on the anatomical structure andthe growth pattern in the apical meristem, as well as DNA synthesisand nucleolus size were investigated. When auxin is applied before or during photoperiodic inductionit inhibits DNA synthesis and meristematic activity. The axillarymeristem (i.e. a group of cells in the axils of the leaf primordia)is most affected. A similar inhibition of the axillary meristemwas also observed in non-induced control plants grown in continuouslight. Auxin applied simultaneously with photoperiodic inductioncounteracts the reduction of apical dominance in the apex andthus inhibits the onset of floral differentiation. Auxin appliedfollowing induction inhibits the previously-formed buds andmakes possible a more complete development of the apical flower. The dual effect of IAA on flowering, inhibitory and stimulatory,manifests itself as a growth response at different stages ofthe changing shoot apex.  相似文献

Abscisic acid (ABA) applied to the decapitated second internodeof runner bean plants enhanced outgrowth of lateral buds onlywhen internode stumps were no longer elongating. Applied toelongating internodes of slightly younger plants, ABA causesinhibition of bud outgrowth. Together with 10–4 M indol-3-ylacetic acid (IAA), ABA stimulated internode elongation and interactedadditively in the inhibition of bud outgrowth. A mixture of10–5 M ABA and 10–6 M gibberellic acid (GA3 ) causedsimilar effects on internode growth as IAA + ABA, but was mutuallyantagonistic in effect on growth of the lateral buds. The occurrence and endogenous level of various plant hormoneswere measured for the short-day plants Lemna paucicostata 151and 381 and the long-day plant Lemna gibba G3 to determine whetherany of them are involved in the photoperiodic control of flowering.ABA, IAA, GA1, GA29, GA34, GA53, trans- and cis-zeatin, trans-and cis-ribosyl zeatin, N6-(2-isopentenyl) adenine and N6-(2-isopentenyl)adenosine were definitely detected in each species, while GA4was only detected in L. gibba G3 and GA20 was only detectedin L. paucicostata 151. The endogenous levels of ABA and IAAwere in the range of 1–7 ng/g fr wt and were not significantlydifferent in vegetative and flowering plants. The endogenousgibberellin levels were generally higher in Lemna grown underlong-day rather than short-day conditions. The endogenous cytokininlevels were almost the same in both flowering and vegetativeplants of L. paucicostata 151 and 381. In L. gibba G3, however,the level of cis-ribosyl zeatin, N6-(2-isopentenyl) adenineand N6-(2-sopentenyl) adenosine were higher in vegetative thanin flowering plants. These results indicate that there is not necessarily a directrelation between endogenous plant hormone levels and flowering,and that the chemical basis for the photoperiodic control offlowering cannot be explained solely by changes in hormone levels.The possibility remains, however, that one or more of the planthormones has some influence of secondary importance on the floweringprocess in Lemna. Removal of the blade from the leaf subtending the first flowerbud on Cyclamen persicum ‘Swan Lake’ plants causedthe petiole of that leaf to senesce, but had no effect on thegrowth of the flower peduncle in the debladed petiole's axil.A 10 mg NAA l–1 application generally had no effect onpetiole senescence, peduncle elongation or flowering date whenapplied to the cut end of the petiole after blade removal. A25 mg GA3 l–1 application or a combination of 25 mg GA3l–1 application or a combination of 25 mg GA3 l–1plus 10 mg NAA l–1 delayed petiole senescence and enhancedpeduncle elongation and subsequent flowering. Gibberellins GA1, GA4, GA8, GA9, GA19, GA20, GA29, GA44, GA81,indole-3-acetic acid (IAA) and abscisic acid (ABA) were identifiedin cambial region tissues of Eucalyptus globulus by comparingmass spectra and Kovats retention indices with those of authenticstandards. Using stable isotope labelled internal standardsGA19, GA20 and GA44 were quantified at levels of 2–7 ng(g fr wt)-1, other GAs were present at levels < 1 ng (g frwt)-1. Levels of IAA and ABA ranged from 417–1, 140 ng(g fr wt)-1 and 86–305 ng (g fr wt)-1 respectively. It has been established that Kalanchoe blossfeldiana and Xanthiumpensylvanicum require CO2 during the light period of short daysfor successful photoperiodic induction of flowering, even ifall but the induced leaf are held in normal air. In X. pensylvanicumfloral induction in normal air was independent of the starchstatus of the leaves but when reserves were reduced, lack ofCO2 in the light suppressed floral induction to an even greaterextent. Injection into the induced leaf (Kalanchoe) or leaftip feeding (Xanthium) of carbohydrates, organic and amino acidsor several other metabolites failed to substitute for the CO2requirement for induction. A small response was produced by10 mg ml–1 sucrose in X. pensylvanicum while in normalair 25 parts 10–6 ATP reduced the time to flowering inK. blossfeldiana and 10–4 M proline was inhibitory. Anexperiment on the light requirement established a need for redlight ( max 660 nm) during photoperiods but red light alonedid not facilitate maximal induction. It is concluded that someearly, possibly labile, product of photosynthetic CO2 fixationis essential to floral induction in these species.  相似文献

Kinetin (1•10–4 M and 1•10–3 M) was appliedto the plumules of 6-day-old Chenopodium rubrum plants. Effectson growth, anatomical structure and organogenesis in the apicalmeristem were followed. Floral differentiation as affected bykinetin was also investigated in plants induced to flower byshort-day treatment. Kinetin increased mitotic activity in the apical meristems inboth induced and non-induced plants. The effect was most pronouncedin the peripheral and subcentral zone. An increase in nucleolussize and a higher degree of pyroninophilia in the peripheralzone was also observed, indicating a localized promotion ofRNA synthesis. A higher rate of leaf initiation and a stimulationof leaf and stem growth was subse quentiy recorded. The growthof axillary meristems and of bud primordia was promoted onlyat the lower concentration of kinetin (1•10–4 M),in both photoperiodically-induced and non-induced plants. However,the pattern of lateral bud growth differed from that found innormal floral differentiation. In kinetintreated plants, thebud primordia are isolated from the summit of the shoot apexby a succession of rapidly growing leaves. The enhancement ofleaf growth leads to correlative inhibition of axillary budpriniordia and results, finally, in a suppression of floraldifferentiation. The inhibitory effect of kinetin on floweringwas compared with that of auxin. Inhibition of flowering occurredin both cases but is achieved in two different ways.  相似文献

Plants of Lamium amplexicaule, grown under short-day field conditionsin Northern California produce predominantly closed flowers.Under long-day field conditions, plants may produce up to 50per cent open flowers, the same total number of flowers beingproduced under each daylength regime. A 20 ml drop of GA3 or GA7 (100µM) was applied to themain shoot apex of plants growing under short-day conditions,and all subsequently produced flowers opened. CCC, an inhibitorof gibberellin synthesis, was applied (0.06 per cent) to thesoil of seedlings grown under long-day conditions. The CCC-treatedplants were dwarfed and bore only closed flowers. With GA7 appliedexogenously to the CCC-treated shoots, inhibition was released,resulting in elongated internodes and open flower production.The timing of flower production and internodal extension inLamium amplexicaule are positively correlated. When floral primordiawere removed from main shoots, the average internode lengthsdecreased. The number of nodes produced in treated plants wasincreased as a result of flower removal. Exogenous GA7 (10 µM)applied to the nodes from which flowers were removed resultedin internodal extension but had no effect on node number. Twoprocesses that may contribute to the control of the productionof open and closed flowers in Lamium amplexicaule are: (1) anincreasing anther sac size from lower to upper node flowersthat may exert control locally, via GN production, on corollaexpansion, and (2) a photoperiodic control. Green plants and plants devoid of photosynthetic pigments were compared with regard to their ability to flower under various growth conditions. Green plants of Chenopodium rubrum L. and plants treated with norflurazon SANDOZ-9789 (SAN) were grown on sucrose-containing media with or without hormones (GA3, BA, IAA, ABA) under short-day photoperiodic or continuous illumination with white, blue, or red light. Green and SAN-treated albino plants produced flowers only under short-day conditions. The flowering of green plants was independent of the presence of sucrose and hormones in the medium as well as of the light quality. The albino plants produced flowers under white and blue light but did not flower in red light. The addition of GA3 or BA to the medium induced flowering of albino plants exposed to red light. The functional interaction of photoreceptors in the flowering control is discussed.  相似文献

Dormancy was induced during storage of Liatris spicata cormsgrown in Israeli summer conditions, but plants left in soilcontinued vegetative growth. Corms of winter-grown plants sproutedfreely. Treatment with GA3 restored both sprouting and floweringin summer-grown corms, but in winter corms GA2 was effectiveonly after corms were stored at low temperature. All the plantsflowered after 4 weeks at 2 °C and GA3 treatment. The content of gibberellins in the main bud of freshly excavatedcorms decreased during the first 18 d of storage but increasedto the initial level after 4 months of cold storage. The numberof flowering stems increased to 2.5 per corm when corms werecold-stored up to 75 d, but decreased with a longer storage. Effects of three phytohormones (IAA1, GA3 and kinetin) on grain-fillingand the pattern of 32P translocation from individual leavesto grains were studied at intervals of 7 days during the progressof reproductive development of rice (Oryza saliva L. cv. Jaya).The plants were sprayed with 100 µg ml–1 aqueoussolutions of the hormones at 100 days, when the plants wereentering the reproductive stage. Kinetin produced a pronouncedeffect on grain-filling as well as on 32P mobilization fromindividual leaf to grains and increased yield, possibly by increasingleaf longevity. GA3 and IAA also increased the grain-fillingand 32P mobilization significantly over control but the effectswere less marked than those of kinetin. Abscisic acid (ABA) was applied in a concentration of 1. 10?3 M and 1. 10?4 M to the quantitative SD plantChenopodium rubrum under various light regimes. ABA did not influence flowering in plants under continuous illumination, enhanced flowering in plants subjected to long days and inhibited it in plants induced by short days. It was concluded that ABA can not substitute for inductive treatment but its action may be additive to initial stages of reproductive morphogenesis (enhanced growth rate and branching of the apical meristem) as evoked by long days.  相似文献

The medium forin vitro culture of green and SANDOZ herbicides-treatedChenopodium rubrum L. plants contained saccharides and hormones in different concentrations. Five days after sowing, the plants were exposed to non-inductive (15 long days—LD) or inductive (6 short days—SD + 9 LD) photoperiodic conditions. The length of hypocotyl and cotyledon blade were measured and percentage of flowering was scored. Gibberellic acid (GA3) stimulated hypocotyl growth of green and photobleached plants under SD and inhibited under LD conditions. Indole-3-acetic acid (IAA) slightly stimulated hypocotyl growth of green plants only under LD conditions. Benzylaminopurine (BAP) inhibited hypocotyl growth regardless of photoperiodic regime. The optimal concentration of glucose or saccharose for flowering in green and SANDOZ-treated plants was 5%. In green SAN 9785-treated plants exogenous saccharides compensated lack of photosynthates to bring about full flowering, but SAN 9789-treated plants needed in addition GA3.  相似文献

Inhibition of the biosynthesis of gibberellins by prohexadione,3,5-dioxo-4-propionylcyclo-hexanecarboxylic acid, was studiedwith cell-free systems derived from immature seeds of Cucur-bitamaxima, Phaseolus vulgaris and Pisum sativum. Prohexadione,at a concentration of 10–4 M, inhibited C-7 oxidationof GA12-aldehyde, C-20 oxidation of GA15, conversion of C20-gib-berellinsto C19-gibberellins, 3ß-hydroxylation, 2,3-dehydrogenationof GA20, 2,3-epoxidation of GA5 and 2ß-hydroxylationof GA9 and GA20. The 3ß-hydroxylase activity appearedto be more sensitive to prohexadione than were the C-20 oxygenaseand the 2ß-hydroxylase activities. The conversionof mevalonic acid to GA12-aldehyde and the 13-hydroxylationof GA12 were not affected by prohexadione at a concentrationof 3 ? 10–4 M. All of the steps inhibited by prohexadioneare oxidation steps catalyzed by soluble enzymes that require2-oxoglutarate, Fe2+ and oxygen, and all of them occur distalto the synthesis of GA12-aldehyde in the biosynthesis of gibberellins. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. The effects of various plant growth regulators and that of pHon the in vitro growth and development of young inflorescencesof Brassica napus L. cv. Westar were examined. A cytokinin wasrequired for normal maturation of floral buds, including thecompletion of microsporogenesis, and it stimulated the initiationof additional buds on the inflorescence axis. Benzylaminopurine(BAP) was the most effective of the cytokinins tested. Gibberellicacid (GA3) and naphthaleneacetic acid (NAA) alone were ineffective.In combination with BAP, both reduced the positive influenceof the cytokinin but GA3 was more inhibitory than NAA. At alow initial pH (3.9–4.6), the percentage of cultures whichproduced normal buds was significantly higher, especially inthe presence of 10-7 M or 5 ? 10-7 M BAP, in comparison to cultureswith a pH of 5.3-6.0, the standard range for plant tissue culture.  相似文献

The distribution of exogenously-supplied radioactive labelledindol-3-yl-acetic acid (IAA) and gibberellin A1 (GA1) in geotropicallystimulated roots of Norway spruce (Picea abies (L.) Karst.)has been demonstrated. Seedlings were positioned with theirroot tips in 2.1 x 10–6 M [14C]IAA or 1.3 x 10–8m 3H-GA1 for 4 and 20 h, respectively. After geotropic stimulationfor 90 min in the horizontal position the root tips were cutlongitudinally in 50 µm thick sections, using a freeze-microtome.The radioactivity in the 14C-IAA treated roots occurred in higherconcentration in the lower than in the upper halves (ratio 1.25:1). A similar trend was observed in the [3H]GA1-treated rootswhere the ratio lower: upper halves was 2.04: 1. The ratio ofradioactivity in right and left halves of vertical roots wasapproximately the same in roots supplied with [14C]IAA and [3H]GA1(1.09: 1). The supplied radioactive compounds were analysed chromatographicallyafter extraction in methanol of 6 mm apical root segments. Onlya small fraction (7–8 per cent) of the supplied [14C]IAAwas revealed unchanged in the segments. The major part of thechromatographed, labelled compound has not been identified,but on basis of its RF value it is suggested that it may beindol-3-acetyl-aspartic acid (IAAasp). The chromatographic analysis of the [3H]GA,-treated segmentsshowed that only small fractions of this gibberellin has beenconverted to other compounds. These results have been discussed and correlated with knowledgeof plant growth regulators and their participation in root geotropism. Feeding experiments on tillers of a non-flowering red cloversegregate indicated that the promotive effect of gibberellinupon stem extension and flowering occured only under inductivelong days (LD). Furthermore, floral initiation took place inLD only when free gibberellin exceeded an unspecified criticallevel. Various intermediates in fungal gibberellin synthesis were fedin a similar fashion but none exerted any effect upon reproductivegrowth. Studies using 3H-gibberellin A3 (GA3) showed that innormal (flowering) material GA3 was more rapidly metabolizedin short days than in long days. Qualitative differences ingibberellin metabolism between normal and non-flowering genotypeswere revealed by radio-isotope studies. Non-flowering materialrapidly degraded GA3 under LD conditions to a biologically inactivecompound chromatographically indentical with allo-gibberic acid,whereas normal plants metabolized GA3 more slowly producinga compund similar to gibberellenic acid. The implications ofthese results are discussed in terms of the mechanism of stemelongation and floral initiation.  相似文献

The activity of IAA-oxidase increased in the leaves of Impatiensbalsamina plants receiving inductive photoperiodic cycles andin plants receiving treatments with gibberellic acid (GA3) and/ortannic acid (TA), even under non-inductive photoperiods; theactivity also increased in the stem receiving inductive photoperiodiccycles (8 h). Treatment with GA3 and TA mimics the effect ofSD cycles in the development of some isoenzymes of IAA-oxidase.Thus a new isoenzyme at Rf 0.48 developed in the leaves andone at Rf 0.82 developed in both the stem and the leaves ofall plants receiving inductive treatments – photoperiodicor chemical – but not in water-treated controls undernon-inductive photoperiods.
