Molecular markers provide the opportunity to identify marker-quantitative trait locus (QTL) associations in different environments and populations. Two soybean [Glycine max (L.) Merr.] populations, Young x PI 416 937 and PI 97100 x Coker 237, were evaluated with restriction fragment length polymorphism (RFLP) markers to identify additional QTLs related to seed protein and oil. For the Young x PI 416937 population, 120 F4-derived lines were secored for segregation at 155 RFLP loci. The F4-derived lines and two parents were grown at Plains, G.a., and Windblow and Plymouth, N.C. in 1994, and evaluated for seed protein and oil. For the PI 97100 x Coker 237 population, 111 F2-derived lines were evaluated for segregation at 153 RFLP loci. Phenotypic data for seed protein and oil were obtained in two different locations (Athens, G.a., and Blackville, S.C.) in 1994. Based on single-factor analysis of variance (ANOVA) for the Young x PI 416937 population, five of seven independent markers associated with seed protein, and all four independent markers associated with seed oil in the combined analysis over locations were detected at all three locations. For the PI 97 100 x Coker 237 population, both single-factor ANOVA and interval mapping were used to detect QTLs. Using single-factor ANOVA, three of four independent markers for seed protein and two of three independent markers for seed oil were detected at both locations. In both populations, singlefactor ANOVA, revealed the consistency of QTLs across locations, which might be due to the high heritability and the relatively few QTLs with large effects conditioning these traits. However, interval mapping of the PI 97100 x Coker 237 population indicated that QTLs identified at Athens for seed protein and oil were different from those at Blackville. This might result from the power of QTL mapping being dependent on the level of saturation of the genetic map. Increased seed protein was associated with decreased seed oil in the PI 97100 x Coker 237 population (r = –0.61). There were various common markers (P0.05) on linkage groups (LG) E, G,H,K, and UNK2 identified for both seed protein and oil. One QTL on LG E was associated with seed protein in both populations. The other QTLs for protein and oil were population specific.  相似文献

Abscisic acid (ABA) sensitivity in embryos is one of the key factors in the seed dormancy of wheat. Many ABA signaling genes have been isolated in Arabidopsis, while only a few wheat homologues have been identified. In the present study, diploid wheat homologues to Arabidopsis ABA signaling genes were identified by in silico analysis, and mapped them using a population of diploid wheat recombinant inbred lines derived from a cross between Triticum monococcum (Tm) and T. boeoticum (Tb). Four diploid wheat homologues, TmVP1, TmABF, TmABI8 and TmERA1 were located on chromosome 3Am and TmERA3 was on the centromere region of chromosome 5Am. In two consecutive year trials, one major QTL on the long arm of 5Am, two minor QTLs on the long arm of 3Am and one minor QTL on the long arm of 4Am were detected. The 5Am QTL explained 20–27% of the phenotypic variations and the other three QTLs each accounted for approximately 10% of the phenotypic variations. Map positions of the loci of TmABF and TmABI8 matched the LOD peaks of the two QTLs on 3Am, indicating that these two homologues are possible candidate genes for seed dormancy QTLs. Moreover, we have found two SNPs result in amino acid substitutions in TmABF between Tb and Tm. Comparison of the marker positions of QTLs for seed dormancy of barley revealed that the largest QTL on 5Am may be orthologous to the barley seed dormancy QTL, SD1, whereas there seems no orthologous QTL to the corresponding barley SD2 locus.  A study was conducted on a two-row/six-row cross of barley to (1) determine the yield potential, (2) detect epistasis and genetic correlations, (3) estimate the heritabilities of six agronomic traits, and (4) study the effect of the V locus on the agronomic traits in the barley cross. The effects of five other marker loci (Re2, s, R, Est1, and Est5) on the six agronomic traits were also studied. One hundred and ninety doubled-haploid (DH) lines were derived from a ‘Leger’/CI9831 cross using the bulbosum method. The DH lines and the two parents were tested for grain yield, test weight, seed weight, plant height, lodging, and heading/maturity at two locations in Eastern Canada in 1993. Additive×additive epistasis and genetic correlations were detected for some of the agronomic traits. Many of the heritability estimates were high; however, significant progress in yield improvement would be difficult to achieve because of a low mean yield of the DH lines. Under the growing conditions in Eastern Canada, six-row lines outyielded two-row by 20–27%. Six-row lines, however, were associated with low test weight, low seed weight, and severe lodging. Some two-row lines yielded higher than the two-row parent CI9831, but none of the six-row lines yielded higher than the six-row parent ‘Leger’. The R, s, and Est5 loci were associated with the six agronomic traits, but the Est1 locus was apparently not associated with the agronomic traits. The effect of the Re2 locus was probably due to its close linkage with the V locus. Further studies are needed to determine if superior six-row lines can be developed from two-row/six-row crosses. Drought is a major abiotic stress that adversely affects wheat production in many regions of the world. The objective of this study was to identify quantitative trait loci (QTL) controlling grain yield and yield components under reduced moisture. A cross between common wheat cultivars ‘Dharwar Dry’ (drought tolerant) and ‘Sitta’ was the source of one hundred twenty-seven recombinant inbred lines evaluated for two-seasons in a field under differing soil moisture regimes in Ciudad Obregon, Sonora, Mexico. An SSR/EST-STS marker map was constructed and a grain yield QTL on the proximal region of chromosome 4AL was found to have a significant impact on performance under reduced moisture. This region was associated with QTL for grain yield, grain fill rate, spike density, grains m−2, biomass production, biomass production rate, and drought susceptibility index (DSI). Molecular markers associated with these traits explained 20, 33, 15, 23, 30, 26, and 41% of phenotypic variation, respectively on chromosome 4A. Microsatellite locus Xwmc89 was associated with all significant QTL covering a 7.7 centiMorgans (cM) region and generally explained the greatest proportion of phenotypic variation. The alleles associated with enhanced performance under drought stress were contributed by Dharwar Dry. Microsatellite marker wmc89 may be useful for marker assisted selection to enhance drought tolerance.  相似文献

Fifty-four susceptible soybean, Glycine max, cultivars or plant introductions were evaluated for tolerance to H. glycines, the soybean cyst nematode (SCN). Seed yields of genotypes were compared in nematicide-treated (1,2-dibromo-3-chloropropane, 58 kg a.i./ha) and nontreated plots at two SCN-infested locations over 3 years. Distinct and consistent levels of tolerance to SCN were observed among soybean genotypes. PI 97100, an introduction from Korea, exhibited the highest level of tolerance with an average tolerance index ([yield in nontreated plot ÷ yield in nematicide-treated plot] × 100) of 96 over 2 years. Coker 156 and Wright had moderate levels of tolerance (range in index values 68 to 95) compared to the intolerant cuhivars Bragg and Coker 237 (range in index values 33 to 68). Most of the soybean genotypes evaluated were intolerant to SCN. The rankings of five genotypes for tolerance to SCN and Hoplolaimus columbus were similar. Tolerance for seed yield was more consistently correlated with tolerance for plant height (r = 0.55 to 0.64) than for seed weight (r = 0.23 to 0.65) among genotypes.  相似文献

An association panel consisting of 185 accessions representative of the barley germplasm cultivated in the Mediterranean basin was used to localise quantitative trait loci (QTL) controlling grain yield and yield related traits. The germplasm set was genotyped with 1,536 SNP markers and tested for associations with phenotypic data gathered over 2?years for a total of 24?year?×?location combinations under a broad range of environmental conditions. Analysis of multi-environmental trial (MET) data by fitting a mixed model with kinship estimates detected from two to seven QTL for the major components of yield including 1000 kernel weight, grains per spike and spikes per m2, as well as heading date, harvest index and plant height. Several of the associations involved SNPs tightly linked to known major genes determining spike morphology in barley (vrs1 and int-c). Similarly, the largest QTL for heading date co-locates with SNPs linked with eam6, a major locus for heading date in barley for autumn sown conditions. Co-localization of several QTL related to yield components traits suggest that major developmental loci may be linked to most of the associations. This study highlights the potential of association genetics to identify genetic variants controlling complex traits.  相似文献

A total of 206 random, healthy individuals belonging to five distinct ethnic groups (Ezhavas, Arayas, Nairs, Vishwakarmas and Muslims) were analyzed for 18 bp VNTR repeat polymorphism present in the 5í flanking region of the Thiopurine Methyl Transferase gene (TPMT). In the present study, the population data of TPMT minisatellite was compared with the population data of other loci and the utility of minisatellite was evaluated in population studies. Human tandem repeat alleles of the TPMT minisatellite locus were characterized for the length polymorphism. The expected and observed heterozygosity did not show any significant difference. All five populations were in Hardy-Weinberg equilibrium. High polymorphism information iontent (PIC) (≥0.658) and power of discrimination (PD) (ranging from 0.775–0.860) value of this VNTR showed that this marker is informative. The combined power of discrimination of TPMT minisatellite along with other two loci studied earlier in our lab was 0.9964. The paternity exclusion power (PE) of TPMT VNTR ranged from 0.203 to 0.533 and the combined power of paternity exclusion (with two other loci D8S315 and D2S1328) was ≥0.8285. All these parameters (heterozygosity, PD, PIC, PE) of TPMT minisatellite locus showed that this marker is informative and can be used for DNA typing and population studies besides being used in clinical investigation in checking thiopurine drug sensitivity of individuals. 氮磷钾配比施肥对广金钱草产量及质量的影响   总被引:1,自引:0,他引:1  
测定了不同氮磷钾配施条件下广金钱草的株高、地径、种子产量、药材干重及夏佛塔苷含量。结果表明:施肥组生长状况及产量与不施肥组具有极显著差异,其中单施钾肥30g·m-2的广金钱草株高及药材干重最高,分别为174.04cm、94.50g;单施氮肥40g·m-2地径最粗,达到16.79mm;施氮、磷肥各80g·m-2,钾肥30g·m-2种子产量最高,增产率达到179.30%;施氮肥40g·m-2,磷肥80g·m-2,钾肥80g·m-2夏佛塔苷含量最高,达到0.175%。由此可知氮磷钾的合理配施能够促进广金钱草的生长并提高产量,同时影响夏佛塔苷的积累。以株高、地径、种子产量、药材干重、夏佛塔苷含量为评价标准。根据氮磷钾肥料多元二次曲线回归方程数学模型计算结果,得出氮最佳施肥量:42.4~64.5g·m-2,磷:49.5~59.0g·m-2,钾:30.0~41.0g·m-2。氮磷钾最佳施肥组合为N∶P∶K=5.4∶5.5∶3.7。  相似文献

The use of molecular markers to identify quantitative trait loci (QTLs) has the potential to enhance the efficiency of trait selection in plant breeding. The purpose of the present study was to identify additional QTLs for plant height, lodging, and maturity in a soybean, Glycine max (L.) Merr., population segregating for growth habit. In this study, 153 restriction fragment length polymorphisms (RFLP) and one morphological marker (Dt1) were used to identify QTLs associated with plant height, lodging, and maturity in 111 F2-derived lines from a cross of PI 97100 and Coker 237. The F2-derived lines and two parents were grown at Athens, Ga., and Blackville, S.C., in 1994 and evaluated for phenotypic traits. The genetic linkage map of these 143 loci covered about 1600 cM and converged into 23 linkage groups. Eleven markers remained unlinked. Using interval-mapping analysis for linked markers and single-factor analysis of variance (ANOVA), loci were tested for association with phenotypic data taken at each location as well as mean values over the two locations. In the combined analysis over locations, the major locus associated with plant height was identified as Dt1 on linkage group (LG) L. The Dt1 locus was also associated with lodging. This locus explained 67.7% of the total variation for plant height, and 56.4% for lodging. In addition, two QTLs for plant height (K007 on LG H and A516b on LG N) and one QTL for lodging (cr517 on LG J) were identified. For maturity, two independent QTLs were identified in intervals between R051 and N100, and between B032 and CpTI, on LG K. These QTLs explained 31.2% and 26.2% of the total variation for maturity, respectively. The same QTLs were identified for all traits at each location. This consistency of QTLs may be related to a few QTLs with large effects conditioning plant height, lodging, and maturity in this population.  相似文献

The effects of host genotype and initial nematode population densities (Pi) on yield of soybean and soil population densities of Heterodera glycines (Hg) race 3 and Meloidogyne incognita (Mi) race 3 were studied in a greenhouse and field microplots in 1983 and 1984. Centennial (resistant to Hg and Mi), Braxton (resistant to Mi, susceptible to Hg), and Coker 237 (susceptible to Hg and Mi) were planted in soil infested with 0, 31, or 124 eggs of Hg and Mi, individually and in all combinations, per 100 cm³ soil. Yield responses of the soybean cultivars to individual and combined infestations of Hg and Mi were primarily dependent on soybean resistance or susceptibility to each species separately. Yield of Centennial was stimulated or unaffected by nematode treatments, yield of Braxton was suppressed by Hg only, and yield suppressions caused by Hg and Mi were additive and dependent on Pi for Coker 237. Other plant responses to nematodes were also dependent on host resistance or susceptibility. Population densities of Mi second-stage juveniles (J2) in soil were related to Mi Pi and remained constant in the presence of Hg for all three cultivars. Population densities of Hg J2 on the two Hg-susceptible Cultivars, Braxton and Coker 237, were suppressed in the presence of Mi at low Hg Pi.  相似文献

Pisum sativum L. is known for high seed and protein yields but also for.yield instability. Because legumes utilize two sources of nitrogen (atmospheric N2 fixed in nodules and assimilation of soil mineral N), studies on their nitrogen nutrition is more complex than in other plants. In this work, pea symbiotic mutants (with no nodules at all ([Nod-]), with inefficient nodules ([Nod+Fix-]) or showing an hypernodulating and a ‘nitrate-tolerant symbiosis’ character ([Nod++Nts]), their semi-leafless isogenic homologues and the parental control line cv Frisson were fertilized with three levels of mineral nitrogen (0, 25 or 50 g N m-2) to generate a range of mineral nitrogen regimes in the same genetic background. Impact of the source and level of nitrogen nutrition was measured on reproductive development, growth, nitrogen accumulation and seed yield. It was shown that a N deficiency induced flowering termination. It also led to a large decrease in the number of seeds produced and the amount of N accumulated in forage and in seeds, when little effect was observed on the progression rates of reproductive stages along the stem. The single seed weight and the amount of dry matter accumulated in forage neither responded strongly to N deficiency. The source of nitrogen was shown to be of little importance to yield but the application of about 50 g N m-2 was necessary to reach the yield of the control cv Frisson when exclusive assimilation was ensuring the N requirements of the plant. Despite the fact that the nitrate-tolerant and hypernodulating mutant P64 used in this study did not yield as well as the parent cv Frisson, it is proposed that [Nod++Nts] characters could act as a yield regulating factor.  相似文献

Mating systems are generally thought to be important factors in determining the amount and nature of genetic variability in a population. Nearly 1,000 individuals at a single location (Lucknow) and over two years were crossed and subsequently scored for selfing versus outcrossing in 9–10 monohybrid populations of opium poppy (Papaver somniferum). Three different alleles of two marker loci (two—R/r and P/p—for anthocyanin locus and B/b for capsule size locus) were used to determine the male gametes that had effected fertilizations in F2 recessives (rr, pp and bb). The estimates of the gene frequency-based outcrossing parameter (α) were found to vary with year, cross and marker locus used (α range: 7.21–71.03%). Study of the two dihybrid crosses concerning the two marker loci simultaneously, further confirmed that outcrossing at the R/r or P/p locus was significantly greater than that at the B/b locus. The nature of the outcrossing was, in general, nonrandom. In this species, in general, selfing predominated, with one exception in respect of monohybrid crosses involving the purple form of anthocyanin locus, in which outcrossing predominated.  相似文献

The role of seed bank, seed rain, and regeneration from seedlings and sprouts after swidden agriculture was compared in 5-, 10- and 20-year-old secondary forest and in a primary forest in Bragantina, Pará, Brazil. The seed bank (0–5 cm soil depth) was largest in the 5-year-old forest (1190 ± 284 seeds m−2) and decreased nearly ten-fold with age to 137 ± 19 seeds m−2 in the primary forest. The highest seed rain was in the 5-year-old forest (883 ± 230 seeds m−2 year−1) and the least in the primary forest (220 ± 80 seeds m−2 year−1). Large plants (≥5 cm dbh) had more individuals and species that regenerated from sprouts than from seeds and the most abundant tree species in the secondary forest stands of all ages appear to be maintained by sprouting. The smaller individuals (≥1 m tall, <5 cm dbh) in the 5-year-old forest were mainly from sprouts, but those in the older secondary forests originated mainly from seeds. These results show that at the beginning of succession, although many species can be introduced to swidden fallow from seed bank and seed rain, it is the sprout that is the main source of recruits of primary forest species in secondary forests in Bragantina.  相似文献

Phytochemicals such as phenolics and flavonoids in rice grain are antioxidants that are associated with reduced risk of developing chronic diseases including cardiovascular disease, type-2 diabetes and some cancers. Understanding the genetic basis of these traits is necessary for the improvement of nutritional quality by breeding. Association mapping based on linkage disequilibrium has emerged as a powerful strategy for identifying genes or quantitative trait loci (QTL) underlying complex traits in plants. In this study, genome-wide association mapping using models controlling both population structure (Q) and relative kinship (K) were performed to identify the marker loci/QTLs underlying the naturally occurring variations of grain color and nutritional quality traits in 416 rice germplasm accessions including red and black rice. A total of 41 marker loci were identified for all the traits, and it was confirmed that Ra (i.e., Prp-b for purple pericarp) and Rc (brown pericarp and seed coat) genes were main-effect loci for rice grain color and nutritional quality traits. RM228, RM339, fgr (fragrance gene) and RM316 were important markers associated with most of the traits. Association mapping for the traits of the 361 white or non-pigmented rice accessions (i.e., excluding the red and black rice) revealed a total of 11 markers for four color parameters, and one marker (RM346) for phenolic content. Among them, Wx gene locus was identified for the color parameters of lightness (L*), redness (a*) and hue angle (H o). Our study suggested that the markers identified in this study can feasibly be used to improve nutritional quality or health benefit properties of rice by marker-assisted selection if the co-segregations of the marker–trait associations are validated in segregating populations.  相似文献

Experiments conducted over four years (1980–1983) with parsnip cv. Avonre-sister examined the effects of plant density, harvest date and method on seed yield and the components of yield. In 1980, using the root-to-seed method, the yield of seed increased by 50% for an increase in plant density from 5.6 to 35 plants m-2; the maximum yield at the optimum harvest date was 3015 kg h-1. In the other three years, using the seed-to-seed method, seed yield increased to a maximum with an increase in plant density from c. 1 to 10 plants m-2 but at higher densities yields were lower. At c. 10 plants m-2, seed yields were 1567, 4029 and 1040 kg ha-1, in 1981, 1982 and 1983, respectively, when harvested at the optimum time. An increase in plant density increased the number of non-seeding plants in the population, reduced the number of umbels per plant and reduced the number of seeds per umbel. Mean seed weight was, in general, unaffected by plant density and so effects of density on yield reflected changes in seed numbers. Increasing the plant density increased the proportion of primary-umbel seed. Maximum yields of seed were obtained from crops harvested at a seed-moisture content of 50 to 70%, which occurred 46 ± 2·4 days after flowering. Delay in harvesting after this date led to a loss of yield of 33, 35, 139 and 32 kg ha-1wk-1 in 1980, 1981, 1982 and 1983, respectively. Plants cut, placed in the windrow to dry and then threshed gave similar yields to those harvested at the same time but dried in bins.  相似文献

We applied a recently developed multilocus association testing method (localized haplotype clustering) to Wellcome Trust Case Control Consortium data (14,000 cases of seven common diseases and 3,000 shared controls genotyped on the Affymetrix 500 K array). After rigorous data quality filtering, we identified three disease-associated loci with strong statistical support from localized haplotype cluster tests but with only marginal significance in single marker tests. These loci are chromosomes 10p15.1 with type 1 diabetes (p = 5.1 × 10−9), 12q15 with type 2 diabetes (p = 1.9 × 10−7) and 15q26.2 with hypertension (p = 2.8 × 10−8). We also detected the association of chromosome 9p21.3 with type 2 diabetes (p = 2.8 × 10−8), although this locus did not pass our stringent genotype quality filters. The association of 10p15.1 with type 1 diabetes and 9p21.3 with type 2 diabetes have both been replicated in other studies using independent data sets. Overall, localized haplotype cluster analysis had better success detecting disease associated variants than a previous single-marker analysis of imputed HapMap SNPs. We found that stringent application of quality score thresholds to genotype data substantially reduced false-positive results arising from genotype error. In addition, we demonstrate that it is possible to simultaneously phase 16,000 individuals genotyped on genome-wide data (450 K markers) using the Beagle software package. Genetic maps of wheat chromosome 1D consisting of 57 microsatellite marker loci were constructed using Chinese Spring (CS) × Chiyacao F2 and the International Triticeae Mapping Initiative (ITMI) recombinant inbred lines (RILs) mapping populations. Marker order was consistent, but genetic distances of neighboring markers were different in two populations. Physical bin map of 57 microsatellite marker loci was generated by means of 10 CS 1D deletion lines. The physical bin mapping indicated that microsatellite marker loci were not randomly distributed on chromosome 1D. Nineteen of the 24 (79.2%) microsatellite markers were mapped in the distal 30% genomic region of 1DS, whereas 25 of the 33 (75.8%) markers were assigned to the distal 59% region of 1DL. The powdery mildew resistance gene Pm24, originating from the Chinese wheat landrace Chiyacao, was previously mapped in the vicinity of the centromere on the short arm of chromosome 1D. A high density genetic map of chromosome 1D was constructed, consisting of 36 markers and Pm24, with a total map length of 292.7 cM. Twelve marker loci were found to be closely linked to Pm24. Pm24 was flanked by Xgwm789 (Xgwm603) and Xbarc229 with genetic distances of 2.4 and 3.6 cM, respectively, whereas a microsatellite marker Xgwm1291 co-segregated with Pm24. The microsatellite marker Xgwm1291 was assigned to the bin 1DS5-0.70-1.00 of the chromosome arm 1DS. It could be concluded that Pm24 is located in the ‘1S0.8 gene-rich region’, a highly recombinogenic region of wheat. The results presented here would provide a start point for the map-based cloning of Pm24.  相似文献

Celery (Apium graveolens L.) plants cv. Jason overwintered in a polythene tunnel flowered earlier and grew taller than similar plants given a 10-week cold-treatment at 5°C prior to transplanting in the same tunnel in mid-February. However, there was no significant difference in the yield of seeds obtained from both treatments, plants grown at a density of 4m-2 yielded less seeds than those at 2m-2, though the yield per unit area was slightly higher from the high density treatment. Treatment with 100 mgl-1 GA3 applied twice just prior to flowering and during anthesis increased flower stalk, flower pedicel and stamen length but delayed flower opening and seed ripening and decreased seed set and seed yield. Treatment with a mixture of 1000 mgl-1 GA4 and GA7 plus 1000 mgl-1 ethephon on three occasions during seed ripening decreased seed yield and reduced seed germination though those seeds capable of germinating were less dormant than seeds from untreated plants. The size distribution of seeds was unaffected by any treatment other than the preseeding spray with GA3 which reduced the percentage of medium-size seeds.  相似文献
