In this report we address the changes in the expression of the genes involved in ROS scavenging and ethylene biosynthesis induced by the inoculation of plant growth-promoting rhizobacteria (PGPR) isolated from potato rhizosphere. The two Bacillus isolates used in this investigation had earlier demonstrated a striking influence on potato tuberization. These isolates showed enhanced 1-aminocyclopropane-1-carboxylic acid deaminase activity, phosphate solubilization, and siderophore production. Potato plants inoculated with these PGPR isolates were subjected to salt, drought, and heavy-metal stresses. The enhanced mRNA expression levels of the various ROS-scavenging enzymes and higher proline content in tubers induced by PGPR-treated plants contributed to increased plant tolerance to these abiotic stresses. Furthermore, the photosynthetic performance indices of PGPR-inoculated plants clearly exhibited a positive influence of these bacterial strains on the PSII photochemistry of the plants. Overall, these results suggest that the PGPR isolates used in this study are able to confer abiotic stress tolerance in potato plants.  相似文献

Ethylene is a gaseous plant growth hormone produced endogenously by almost all plants. It is also produced in soil through a variety of biotic and abiotic mechanisms, and plays a key role in inducing multifarious physiological changes in plants at molecular level. Apart from being a plant growth regulator, ethylene has also been established as a stress hormone. Under stress conditions like those generated by salinity, drought, waterlogging, heavy metals and pathogenicity, the endogenous production of ethylene is accelerated substantially which adversely affects the root growth and consequently the growth of the plant as a whole. Certain plant growth promoting rhizobacteria (PGPR) contain a vital enzyme, 1-aminocyclopropane-1-carboxylate (ACC) deaminase, which regulates ethylene production by metabolizing ACC (an immediate precursor of ethylene biosynthesis in higher plants) into α-ketobutyrate and ammonia. Inoculation with PGPR containing ACC deaminase activity could be helpful in sustaining plant growth and development under stress conditions by reducing stress-induced ethylene production. Lately, efforts have been made to introduce ACC deaminase genes into plants to regulate ethylene level in the plants for optimum growth, particularly under stressed conditions. In this review, the primary focus is on giving account of all aspects of PGPR containing ACC deaminase regarding alleviation of impact of both biotic and abiotic stresses onto plants and of recent trends in terms of introduction of ACC deaminase genes into plant and microbial species.  相似文献

The mitogen-activated protein (MAP) kinase cascade is an important signaling module which is involved in biotic and abiotic stress responses as well as plant growth and development. In this study, we identified 17 tobacco MAPKs including 11 novel tobacco MAPK genes that have not been identified before. Comparative analysis with MAPK gene families from other plants, such as Athaliana thaliana, rice and poplar, suggested that tobacco MAPKs (such as NtMPK1, NtMPK3 and NtMPK8) might play similar functions in response to abiotic and biotic stresses. QRT-PCR analysis revealed that a total of 14 NtMPKs were regulated by SA and/or MeJA, suggesting their potential roles involved in plant defense response. In addition, 6 NtMPKs were induced by drought treatment, implying their roles in response to drought stress. Our results indicated that most of tobacco MAPK might be involved in plant defense response, which provides the basis for further analysis on physiological functions of tobacco MAPKs.  相似文献

In plants, biotic and abiotic stresses regulate the expression and activity of various peroxidase isoforms. Capsicum annuum EXTRACELLULAR PEROXIDASE 2 (CaPO2) was previously shown to play a role in local and systemic reactive oxygen species bursts and disease resistance during bacterial pathogen infection. Here, we report CaPO2 expression patterns and functions during conditions of biotic and abiotic stress. In pepper plants, CaPO2 expression was strongly induced by abscisic acid, but not by defense-related plant hormones such as salicylic acid, ethylene and jasmonic acid. CaPO2 was also strongly induced by abiotic and biotic stress treatments, including drought, cold, high salinity and infection by the hemibiotrophic fungal pathogen Colletotrichum coccodes. Loss-of-function of CaPO2 in virus-induced gene silenced pepper plants led to increased susceptibility to salt- and osmotic-induced stress. In contrast, CaPO2 overexpression in transgenic Arabidopsis thaliana plants conferred enhanced tolerance to high salt, drought, and oxidative stress, while also enhancing resistance to infection by the necrotrophic fungal pathogen Alternaria brassicicola. Taken together, these results provide evidence for the involvement of pepper extracellular peroxidase CaPO2 in plant defense responses to various abiotic stresses and plant fungal pathogens.  相似文献

RNA interference (RNAi) is a promising gene regulatory approach in functional genomics that has significant impact on crop improvement which permits down-regulation in gene expression with greater precise manner without affecting the expression of other genes. RNAi mechanism is expedited by small molecules of interfering RNA to suppress a gene of interest effectively. RNAi has also been exploited in plants for resistance against pathogens, insect/pest, nematodes, and virus that cause significant economic losses. Keeping beside the significance in the genome integrity maintenance as well as growth and development, RNAi induced gene syntheses are vital in plant stress management. Modifying the genes by the interference of small RNAs is one of the ways through which plants react to the environmental stresses. Hence, investigating the role of small RNAs in regulating gene expression assists the researchers to explore the potentiality of small RNAs in abiotic and biotic stress management. This novel approach opens new avenues for crop improvement by developing disease resistant, abiotic or biotic stress tolerant, and high yielding elite varieties.  相似文献

MicroRNAs are small non-coding regulatory RNA molecules that play an important role in the modulation of gene expression during various environmental stresses. Pseudomonas putida RA, a plant growth promoting rhizobacteria (PGPR) colonizes the root surface of plants improving their growth and development during abiotic stresses modulating the expression of stress-responsive genes; however, the impact of RA on stress responsive-miRNA remains elusive. The present study was an attempt to delineate the role of PGPR in modulating stress responsive-miRNAs in a tolerant desi chickpea genotype exposed to drought and salt stresses. The existence of variable expression patterns of individual miRNAs and their target genes under these stresses at different time points indicate a distinct miRNA-mediated perception and response mechanisms operating under these stresses in the presence or absence of RA in chickpea.  相似文献

The expression of resveratrol synthase (RS) genes is induced by biotic and abiotic factors in peanut cell cultures. However, little is known about the regulation of the RS gene expression in peanut plants. The expression of RS genes was investigated in peanut plants with a peanut RS clone, pPRS3C, which encodes two polypeptides that show about a 96% amino acid sequence identity to peanut RS2 and RS3, respectively. A low level of RS mRNA was detected in the roots of peanut plants grown aseptically in vitro. In mature peanut plants that were grown in the field, however, RS mRNAs were present at relatively high levels in both the roots and pods, but at below the detection limit in leaves. RS mRNAs were abundant in young pods and decreased dramatically in mature pods. The RS mRNA expression was induced by yeast extract and UV in leaves and roots, and also by wounding in leaves. Stress hormones, such as ethylene, jasmonic acid, and salicylic acid, induced RS mRNA accumulation in leaves. These results indicate that the RS gene expression is induced by biotic and abiotic stresses through the stress hormones in peanut plants. The induction of the RS gene expression by biotic and abiotic stresses could provide peanut plants with protection from microbial infections through resveratrol synthesis. The RS gene expression in developing pods has significant implications in terms of the role of resveratrol as a phytochemical for human health.  相似文献

Plants encounter many biotic agents, such as viruses, bacteria, nematodes, weeds, and arachnids. These entities induce biotic stress in their hosts by disrupting normal metabolism, and as a result, limit plant growth and/or are the cause of plant mortality. Some biotic agents, however, interact symbiotically or synergistically with their host plants. Some microbes can be beneficial to plants and perform the same role as chemical fertilizers and pesticides, acting as a biofertilizer and/or biopesticide. Plant growth promoting rhizobacteria (PGPR) can significantly enhance plant growth and represent a mutually helpful plant-microbe interaction. Bacillus species are a major type of rhizobacteria that can form spores that can survive in the soil for long period of time under harsh environmental conditions. Plant growth is enhanced by PGPR through the induction of systemic resistance, antibiosis, and competitive omission. Thus, the application of microbes can be used to induce systemic resistance in plants against biotic agents and enhance environmental stress tolerance. Bacillus subtilis exhibits both a direct and indirect biocontrol mechanism to suppress disease caused by pathogens. The direct mechanism includes the synthesis of many secondary metabolites, hormones, cell-wall-degrading enzymes, and antioxidants that assist the plant in its defense against pathogen attack. The indirect mechanism includes the stimulation of plant growth and the induction of acquired systemic resistance. Bacillus subtilis can also solubilize soil P, enhance nitrogen fixation, and produce siderophores that promote its growth and suppresses the growth of pathogens. Bacillus subtilis enhances stress tolerance in their plant hosts by inducing the expression of stress-response genes, phytohormones, and stress-related metabolites. The present review discusses the activity of B. subtilis in the rhizosphere, its role as a root colonizer, its biocontrol potential, the associated mechanisms of biocontrol and the ability of B. subtilis to increase crop productivity under conditions of biotic and abiotic stress.  相似文献

14-3-3 proteins are found in all eukaryotes where they act as regulators of diverse signalling pathways associated with a wide range of biological processes. In this study the functional characterization of the ZmGF14-6 gene encoding a maize 14-3-3 protein is reported. Gene expression analyses indicated that ZmGF14-6 is up-regulated by fungal infection and salt treatment in maize plants, whereas its expression is down-regulated by drought stress. It is reported that rice plants constitutively expressing ZmGF14-6 displayed enhanced tolerance to drought stress which was accompanied by a stronger induction of drought-associated rice genes. However, rice plants expressing ZmGF14-6 either in a constitutive or under a pathogen-inducible regime showed a higher susceptibility to infection by the fungal pathogens Fusarium verticillioides and Magnaporthe oryzae. Under infection conditions, a lower intensity in the expression of defence-related genes occurred in ZmGF14-6 rice plants. These findings support that ZmGF14-6 positively regulates drought tolerance in transgenic rice while negatively modulating the plant defence response to pathogen infection. Transient expression assays of fluorescently labelled ZmGF14-6 protein in onion epidermal cells revealed a widespread distribution of ZmGF14-6 in the cytoplasm and nucleus. Additionally, colocalization experiments of fluorescently labelled ZmGF14-6 with organelle markers, in combination with cell labelling with the endocytic tracer FM4-64, revealed a subcellular localization of ZmGF14-6 in the early endosomes. Taken together, these results improve our understanding of the role of ZmGF14-6 in stress signalling pathways, while indicating that ZmGF14-6 inversely regulates the plant response to biotic and abiotic stresses.  相似文献

Asiatic desi cotton (Gossypium arboreum) shows great potential against biotic and abiotic stresses. The stress resistant nature makes it a best source for the identification of biotic and abiotic stress resistant genes. As in many plants same set of genes show responding behavior against the various abiotic and biotic stresses. Thus in the present study the ESTs from the G. arboreum drought stressed leaves were subjected to find the up-regulated ESTs in abiotic and biotic stresses through homology and in-silico analysis. A cDNA library has been constructed from the drought stressed G. arboreum plant. 778 clones were randomly picked and sequenced. All these sequences were subjected to in-silico identification of biotic and abiotic up-regulated ESTs. Total 39 abiotic and biotic up-regulated ESTs were identified. The results were further validated by real-time PCR; by randomly selection of ten ESTs. These findings will help to develop stress resistant crop varieties for better yield and growth performance under stresses.  相似文献
