【背景】干旱区露天煤矿开采过程中产生的粉尘颗粒物加剧了土壤生态环境的恶化和矿区空气质量的下降,针对煤矿区土壤和粉尘颗粒物的微生物群落组成的研究鲜有报道。【目的】研究新疆哈密南湖乡露天煤矿土壤、粉尘及大气PM2.5颗粒物中的微生物群落结构和多样性特征,并预测潜在的功能类群。【方法】采用高通量测序技术,对煤矿露天采坑区和电厂粉煤灰堆放区的土壤、粉尘及大气PM2.5颗粒物的微生物真菌及细菌群落组成进行比对分析。【结果】矿区优势真菌类群来自子囊菌门(Ascomycota)和担子菌门(Basidiomycota),优势细菌类群来自变形菌门(Proteobacteria)和放线菌门(Actinobacteria)。真菌和细菌群落的丰富度及α多样性在整个矿区内无显著性差异,大气PM2.5颗粒物的细菌群落生态位宽度显著大于露天采坑区和粉煤灰区。矿区内的土壤和PM2.5颗粒物样本中均发现了一些丰度差异显著的功能类群,真菌特征功能类群为腐生营养型类群,细菌特征功能类群主要包括甲烷营养型类群、几丁质酶类细菌类群等。【结论】露天煤矿区粉尘可能对区域内土壤和PM2.5颗粒物的微生物群落结构产生重要影响,具有煤组分降解功能的特定微生物类群可能是维持矿区土壤生态安全的重要微生物学机制之一。  相似文献

通过扩增子测序,分析了枸杞岛附近海域表层水中真菌群落组成及其多样性特征。结果表明,枸杞岛周围海域真菌群落组成稳定,优势真菌菌门为子囊菌门(Ascomycota),优势菌纲为座囊菌纲(Dothideomycetes),优势真菌属包括Hortaea、青霉菌属(Penicillium)、曲霉菌属(Aspergillus)。其中丰度最高的OTU1765属于威尼克外瓶霉(Hortaea werneckii),经FUNGuild功能预测发现这是一种潜在的人类和植物病原真菌。枸杞岛附近海域表层水中真菌群落的生态功能组成比较丰富,主要为植物病原菌和未明确的腐生真菌。研究结果揭示了枸杞岛附近海域表层水中的真菌群落组成特点,为枸杞岛附近海域表层水中微生物资源的开发利用及环境保护提供参考。  相似文献

【目的】中温大曲是浓香型白酒酿造中的糖化发酵剂和生香剂,目前其质量评价主要依赖于感官和理化特征。已有研究采用扩增子测序初步探索了大曲质量等级与微生物群落组成之间的关联性,但是微生物群落功能对大曲质量的影响尚缺乏深入研究。【方法】本研究采用宏基因组学解析和对比了不同质量等级大曲的微生物群落组成和潜在功能的差异,结合理化性质,聚焦分析了酶活、乳酸和乙酸代谢相关的关键酶基因丰度。【结果】两种等级大曲的真菌群落组成差异显著。一级大曲含有更高相对丰度的丝状真菌,主要分布在曲霉属(Aspergillus, 21.6%)、罗萨氏菌属(Rasamsonia, 6.8%)、拟青霉属(Paecilomyces, 5.0%)和篮状菌属(Talaromyces, 4.4%)等。一级大曲中与环境信息处理和细胞过程相关途径的基因相对丰度普遍显著高于二级大曲。一级大曲中薄层菌属(Hymenobacter)和曲霉属等提供了更高基因丰度的α-淀粉酶,使其具有显著较高的液化力。二级大曲则含有更高相对丰度的横梗霉属(Lichtheimia, 11.8%)、根霉属(Rhizopus, 13.4%)和毕赤酵母属(Pichia, 7.2%)以及乳酸菌类微生物,如伴生乳杆菌属(Companilactobacillus)、魏斯氏菌属(Weissella)和黏液乳杆菌属(Limosilactobacillus)等。二级大曲中与代谢相关途径的基因相对丰度显著较高,如碳水化合物代谢、能量代谢和核苷酸代谢等。二级大曲中较多的乳酸菌类群有利于提高乙醇脱氢酶基因丰度,同时,糖多孢菌属(Saccharopolyspora)、葡萄球菌属(Staphylococcus)、曲霉属和高温放线菌属(Thermoactinomyces)等提供了更多的羧酸酯酶基因丰度,分别使其具有较高的发酵力和酯化力。此外,二级大曲中较多的乳酸菌还可能产生更多的乳酸脱氢酶,降解乳酸,且更高基因丰度的乙酸代谢相关酶类可促进乙酸分解代谢,使得二级大曲酸度显著低于一级大曲。【结论】本研究在基因水平上研究了不同质量等级大曲酶活和酸度差异的微生物基础,可为建立完善的大曲质量评价体系以及理性地调控群落功能提供理论支撑。  相似文献

【背景】草莓连年栽培导致土传病害问题突出,施用熏蒸剂进行土壤消毒因效果显著得以广泛应用。但不同熏蒸剂对土壤病原微生物的影响存在较大差异,同时对非靶标微生物和土壤理化性质也会有不同程度的影响。【目的】明确不同熏蒸剂对草莓连作土壤养分和土壤细菌、真菌群落结构的影响,为合理选择熏蒸剂提供科学依据。【方法】以连作土壤为材料设置5个处理:未熏蒸、石灰氮熏蒸、石灰熏蒸、棉隆熏蒸、威百亩熏蒸,测定熏蒸处理后土壤养分含量;采用PacBio测序平台分析土壤微生物多样性的变化。【结果】石灰氮和威百亩处理均增加了碱解氮含量,降低了有机质、有效磷和速效钾含量;棉隆处理土壤中各养分含量均增加;石灰处理除有机质含量增加外,碱解氮、有效磷和速效钾含量均降低;棉隆、石灰和威百亩处理显著降低pH值。5个处理草莓连作土壤样本中获得了1 164个细菌OTU和444个真菌OTU。细菌多样性和丰富度分析发现,4种熏蒸剂处理均增加了土壤细菌群落的丰富度,石灰氮、石灰和威百亩处理增加了土壤细菌菌落的多样性。4种熏蒸剂处理真菌菌落的丰富度低于对照;石灰、棉隆处理真菌菌落的多样性高于对照和石灰氮、威百亩处理低于对照,但差异不显著。在物种组成分析中,从门水平看,变形菌门(Proteobacteria)和芽单胞菌门(Gemmatimonadetes)为优势细菌门;与对照相比,石灰氮、石灰、棉隆处理变形菌门相对丰度增高,威百亩处理相对丰度降低。4种处理均降低了芽单胞菌门的相对丰度。其他门类中,4种处理均增加了浮霉菌门(Planctomycetota)、疣微菌门(Verrucomicrobia)的相对丰度。优势细菌群落分析表明土壤熏蒸减少了芽单胞菌属(Gemmatimonas)、藤黄单胞菌属(Luteimonas)、中慢生根瘤菌属(Mesorhizobium)等细菌的相对丰度,增加了噬几丁质菌属(Chitinophaga)、苍白杆菌属(Ochrobactrum)的相对丰度。子囊菌门(Ascomycota)为优势真菌,石灰氮、石灰、棉隆、威百亩4种处理均增加了子囊菌门的相对丰度。另外还检测到引起草莓根部土传病害的枝孢属(Cladosporium)和镰刀菌属(Fusarium)病菌,熏蒸处理后均降低了枝孢属和镰刀菌属的相对丰度,其中枝孢属在石灰氮、石灰、棉隆、威百亩处理中分别降低了1.35%、1.11%、0.90%和1.31%,镰刀菌属分别降低了0.71%、0.85%、0.19%和0.65%,但差异不显著。4种土壤熏蒸剂均增加了有益真菌毛壳菌属(Chaetomium)的相对丰度。【结论】采用熏蒸剂处理连作土壤改变了微生物群落构成,减少或灭杀土壤中的大部分致病菌属,起到有效防治草莓土传病害的作用,但不能灭杀所有病菌,而且对有益菌和土壤理化性质有不同程度的影响,因此处理后补充有益微生物非常关键。根据对病原菌的灭杀效果,石灰氮、威百亩处理的效果优于其他处理,可作为轮换施用的熏蒸剂,本试验条件下,棉隆是一种弱的处理剂。  相似文献

【背景】传统山西陈醋是在开放环境下由多菌种固态发酵酿制而成,解析酿造过程中微生物群落的组成、互作网络及来源对认识和调控食醋发酵过程都具有重要意义。【目的】揭示山西陈醋酿造微生物群落的组成、演替、互作网络及来源。【方法】基于高通量测序技术揭示山西陈醋酿造过程中微生物群落的组成、演替和多样性;采用SPSS数据统计软件计算物种间的斯皮尔曼相关系数并利用Gephi软件对微生物群落互作网络进行可视化分析;采用快速期望最大化微生物源跟踪(fast expectation-maximization microbial source tracking, FEAST)方法解析发酵微生物群落的来源。【结果】山西陈醋酿造过程中有6个优势细菌属和5个优势真菌属(平均相对丰度大于1%);醋酸杆菌属(Acetobacter)、乳酸杆菌属(Lactobacillus)和复膜孢酵母属(Saccharomycopsis)的平均相对丰度较高,分别为20.76%、30.38%和46.24%;醋酸杆菌属(Acetobacter)的相对丰度在醋酸发酵阶段逐渐增加,而乳酸杆菌属(Lactobacillus)在此阶段的趋势则相反。微生物群落互作网络结果显示魏斯氏菌属(Weissella)、乳球菌属(Lactococcus)、季也蒙酵母属(Meyerozyma)、LoigolactobacillusSchleiferilactobacillus和地杆菌属(Geobacter)具有较高的连接度(连接度≥7);此外,醋酸杆菌属(Acetobacter)与魏斯氏菌属(Weissella)、SchleiferilactobacillusLoigolactobacillus、乳球菌属(Lactococcus)、季也蒙酵母属(Meyerozyma)具有显著的拮抗作用。来源追溯结果显示糖化发酵第2天样品中有1.02%的细菌群落和77.04%的真菌群落来源于大曲;在醋酸发酵第1天样品中有0.93%的细菌群落和52.82%的真菌群落来源于火醅。【结论】为了解山西陈醋酿造过程中微生物群落的组成、演替、互作网络和解释酿造微生物来源提供了重要的理论数据。  相似文献

【背景】土壤真菌主导田间秸秆腐解的过程,秸秆还田配施有机肥可为真菌提供良好的营养物质。【目的】研究麦玉轮作模式下长期秸秆还田配施有机肥对麦田土壤真菌多样性的影响。【方法】依托山西南部麦玉轮作区长达14年的秸秆还田和施有机肥长期定位试验,采用高通量测序开展土壤真菌群落结构和多样性对不施肥+秸秆清茬(CK)、氮磷化肥+秸秆清茬(NP)、氮磷化肥+秸秆还田(SNP)、氮磷化肥+有机肥+秸秆清茬(NPM)及氮磷化肥+秸秆还田+有机肥(SNPM)的响应差异研究。【结果】秸秆还田和施有机肥处理组的物种丰富度指数、Chao1指数和ACE指数均高于CK。5个处理共产生953个分类操作单元(operational taxonomic units, OTU),CK、NP、NPM、SNP和SNPM分别具有398、451、472、462和440个OTU。在门水平上共检测出9个菌门,其中子囊菌门(Ascomycota)、毛霉门(Mucoromycota)和担子菌门(Basidiomycota)为3个主要菌门,各处理中所占丰度差异显著;在属水平上共检测出262个真菌属,其中CK丰度较高的3个属为柄孢壳菌属(Podospora) 18.85%、被孢霉属(Mortierella) 16.67%和镰刀菌属(Fusarium) 7.77%;NP、NPM、SNP和SNPM丰度较高的前3个相同属均为Dendrostilbella、毛葡孢属(Botryotrichum)和被孢霉属(Mortierella),但相对丰度值存在差异。由聚类分析可知,NPM和SNPM群落组成相似度高,归为一类,而CK、NP、SNP各独立为一类。与环境因子间冗余分析发现,TN含量是影响土壤真菌群落结构的关键因子,也受速效磷、pH、速效钾、碱解氮和全磷等环境指标影响。【结论】长期秸秆还田和施有机肥改变了小麦土壤真菌种群结构和多样性。  相似文献

【背景】松树的内生真菌会影响红脂大小蠹及其伴生真菌的生长及扩散,从而影响红脂大小蠹的入侵。【目的】掌握赤峰地区红脂大小蠹寄主树种内生真菌的物种多样性,筛选对其伴生菌有拮抗作用的菌株,为红脂大小蠹的生物防治提供资源。【方法】采用组织分离法、形态学鉴定和internal transcribed spacer (ITS)序列分析相结合的方法对红脂大小蠹寄主树种油松、樟子松和潜在寄主落叶松进行内生真菌多样性研究,并用两点对峙法进行拮抗菌株筛选。【结果】松树内生真菌鉴定为2门6纲10目18科19属,其中落叶松韧皮部分离到的内生真菌数量最多,为39株,隶属于9属12种,真菌检出率为43.33%;油松次之,为30株,隶属于7属8种,真菌检出率为33.33%;樟子松最少,为29株,隶属于10属13种,真菌检出率为32.22%。3个树种的内生真菌相似性较低,无共有的菌种,青霉属(Penicillium)和篮状菌属(Talaromyces)是唯二的共有菌属,且青霉属在3个树种中均为优势菌属。平板对峙结果表明90%以上的树木内生真菌均能够与伴生真菌形成稳定的对峙,抑制率在50%-86%之间,且Phialocephala sp.和Pochonia bulbillosa对伴生菌的抑制率能高达93.7%。【结论】松树韧皮部的内生真菌具有较高的生防潜力,Phialocephala sp.和P. bulbillosa对红脂大小蠹伴生菌有较好的抑制效果,可作为红脂大小蠹的生防资源。  相似文献

【背景】丛枝菌根(arbuscular mycorrhiza,AM)真菌具有广泛的寄主范围、环境适应性和优良的植物促生能力。然而,土壤的高磷水平严重抑制了AM真菌生长及AM形成。【目的】分离鉴定出耐较高有效磷含量的华南土著AM真菌菌株,为菌根学研究工作提供新颖材料。【方法】采用经典形态学和分子系统学方法鉴定高磷土壤中AM真菌。【结果】从有效磷含量为53-131 (平均值±标准差为88.2±17.6) mg/kg的根区土壤中鉴定出7属25种AM真菌,包括无梗囊霉属(Acaulospora) 12种、球囊霉属(Glomus) 7种、隔球囊霉属(Septoglomus) 2种、近明球囊霉属(Claroideoglomus) 1种、根孢囊霉属(Rhizophagus) 1种、硬囊霉属(Sclerocystis) 1种和类球囊霉属(Paraglomus) 1种,其中幼套近明球囊霉(Claroideoglomus etunicatum)和蜜色无梗囊霉(Acaulospora mellea)是优势种。在(87.7±8.0) mg/kg的高磷水平下,AM真菌仍能形成丛枝和泡囊。但当有效磷含量达到(99.7±1.2) mg/kg时,菌根侵染率和丛枝丰度显著下降,但仍能够形成泡囊。【结论】从广州市南沙区有效磷含量为(88.2±17.6) mg/kg的耕地植物根区土壤中,鉴定出具有耐高磷潜力的7属25种AM真菌,幼套近明球囊霉和蜜色无梗囊霉等分离株可作为后续高磷抑制机制解析及耐高磷AM真菌菌剂研发工作的试验菌株。  相似文献

茶轮斑病对茶树叶片内生真菌群落结构的影响   总被引:2,自引:1,他引:1  
[目的] 茶树叶片内生真菌长期与茶树协同进化,互利共生,在生物和非生物胁迫的生态系统中对茶树起着重要的保护作用,其群落结构组成相对稳定,但在外界因素的影响下,也会发生一定的变化。然而,关于生物胁迫对茶树叶片内生真菌群落结构的影响还缺乏系统的研究。因此,对生物胁迫下叶片内生真菌群落结构的多样性研究具有重要意义。[方法] 本研究采用高通量测序技术,测序了茶轮斑病发病茶树叶片和健康茶树叶片的内生真菌ITS rRNA基因的ITS1区序列,对比分析了内生真菌的多样性和群落结构组成。[结果] 结果表明,发病组叶片的内生真菌多样性和物种丰度均低于健康组。在门分类水平上,2组样本的优势菌群均为子囊菌门(Ascomycota),在属分类水平上,发病组的优势菌群为炭疽菌属(Colletotrichum)和假拟盘多毛孢属(Pseudopestalotiopsis),而健康组的优势菌为枝孢属(Cladosporium)。此外,2组样本内生真菌在群落结构组成上也有显著差异,发病组中假拟盘多毛孢属(Pseudopestalotiopsis)、炭疽菌属(Colletotrichum)和节菱孢属(Arthrinium)的相对丰度显著高于健康组,健康组中被孢霉属(Mortierella)、曲霉属(Aspergillus)、织球壳菌属(Plectosphaerella)、Lectera、葡孢霉属(Botryotrichum)、青霉菌属(Penicillium)、赤霉属(Gibberella)、毛壳菌属(Chaetomium)、Lulwoana和轮枝孢属(Verticillium)的相对丰度显著高于发病组。[结论] 综上,茶轮斑病的发生改变了茶树叶片内生真菌的群落结构,使少数物种优势生长。通过研究,明确了真菌病害对茶叶内生真菌群落结构的影响,为病菌的致病机理研究奠定基础,为茶树病害防治提供理论依据。  相似文献

【背景】野生棘线鲬(Grammoplites scaber)具有丰富的营养价值,但关于其胃肠道微生物方面的研究较少。【目的】研究棘线鲬胃肠道微生物群落特征,以揭示其潜在的益生菌和致病菌,为其健康生长的微生物群落调控提供依据。【方法】利用免培养和纯培养技术相结合的方式对来自珠江口的野生棘线鲬胃肠道样品进行了研究。【结果】通过对16S rRNA基因V3区高通量扩增测序,共得到456个操作分类单元(operational taxonomic units,OTU)。分析结果显示,在门水平上,棘线鲬胃肠道内的主要优势类群为变形菌门(Proteobacteria)和厚壁菌门(Firmicutes)。在属级水平上,梭菌属(Clostridium)在棘线鲬胃肠道样品中普遍存在,综合占比为57.11%。基于16S rRNA基因进行表型和功能预测的结果表明,棘线鲬胃肠道内有益菌和潜在致病菌同时存在且有功能相互制约的趋势。纯培养实验采用12种不同的培养基进行选择性分离,共获得纯培养菌株99株,归类于3个门(ProteobacteriaFirmicutesActinobacteria) 4个纲10个目10个科13个属,其中优势类群为变形菌门(占比50.51%),实现纯培养最多的属级类群为嗜冷杆菌属(Psychrobacter)。【结论】揭示了野生棘线鲬胃肠道微环境微生物的物种组成与多样性,可以为硬骨鱼类核心肠道菌群的研究提供基础参考。此外,有益和有害菌群的揭示可为野生棘线鲬作为海洋食物资源利用的食品安全提供一定的借鉴,为发展海洋渔业养殖提供数据支撑。  相似文献

The aim of this work was to identify the main allergy-related Ascomycetes fungal spores present in the atmosphere of Porto, using different and complementary techniques. The atmospheric sampling, performed in the atmosphere of Porto (Portugal) from August 2006 to July 2008, indicated Cladosporium, Penicillium, Aspergillus and Alternaria as the main fungal spore taxa. Alternaria and Cladosporium peaks were registered during summer. Aspergillus and Penicillium highest values were registered from late winter to early spring. Additionally, the Andersen sampler allowed the culture and isolation of the collected viable spores subsequently used for different identification approaches. The internal-transcribed spacer region of the nuclear ribosomal repeat unit sequences of airborne Ascomycetes fungi isolates revealed 11 taxonomically related fungal species. Among the identified taxa, Penicillum and Aspergillus presented the highest diversity, while only one species of Cladosporium and Alternaria, respectively, were identified. All selected fungal spore taxa possessed phosphatase, esterase, leucine arylamidase and β-glucosidase enzymatic activity, while none had lipase, cystine arylamidase, trypsin or β-glucuronidase activity. The association between the spore cell wall morphology, DNA-based techniques and enzymatic activity approaches allowed a more reliable identification procedure of the airborne Ascomycota fungal spores.  相似文献

The deposition of the airborne microorganisms onto cultural heritage is associated closely with the subsequent biodeterioration. In this study, a systematic investigation was carried out to detect the seasonal variation and diversity of airborne fungal concentration at the World Cultural Heritage Site Maijishan Grottoes in western China. A bio-aerosol sampler was deployed to collect samples over four seasons in 2016. The culturable airborne fungi were isolated, purified and then identified with the extraction of genomic DNA, PCR amplification of ITS rRNA region, sequencing, and phylogenetic analysis. The concentrations of culturable fungi ranged from 216 to 1389 CFU/m3, which varied seasonally with significant differences among the sampling sites. Fifteen different fungal genera were confirmed, among them, Cladosporium was the most predominant fungal genus, followed by Penicillium. The fungal community structure and their relationship with environmental factors were also delineated. The spatial–temporal differences of airborne fungi at Maijishan Grottoes were mainly due to height, rainfall, relative humidity, and temperature. The dominant genera Cladosporium and Penicillium may pose potential threats to the ancient painted sculptures and murals, and monitoring of the airborne fungi at such a heritage site could provide supporting data for the pre-warning and control of fungal outbreaks inside the caves for better management.

In this study, we analyzed air samples collected from several sites within the Mogao Grottoes, Dunhuang, China. The samples were collected each month from September 2008 to August 2009 from an open cave (OC), a semi-open cave (SC), a closed cave (CC), and the entrance (EN) of the Mogao Grottoes. Sampling was carried out using a six-stage Andersen FA-I sampler; then samples were cultured and fungal isolates were identified by partial sequencing of their internal transcribed spacer (ITS) region. Eleven different fungal genera were found, and the most prevalent was Cladosporium, followed by Fusarium, Penicillium, Alternaria, and Aspergillus. The fungal community composition varied among the four sites. Fungal community structure was significantly related to site (r = −0.293, p = 0.039) and to time of year (r = −0.523, p = 0.000). The concentrations and abundance of airborne fungi varied greatly throughout the year at the four sampling sites. Meteorological parameters (e.g., temperature, relative humidity) and the number of visitors also influenced both abundance and community structure of airborne fungi in the Mogao Grottoes.  相似文献

Airborne fungi associated with ornamental plant propagation in greenhouses   总被引:1,自引:0,他引:1  
The objective was to determine potential exposure to airborne fungi in greenhouses and to characterize the temporal patterns of airborne fungi in relation to environmental conditions. We analyzed air samples collected in two greenhouses. Results showed that the top 5 fungi in greenhouse 1 were Trichoderma, hyphal fragments, Aspergillus/Penicillium-like, Cladosporium, and Botrytis in a descending order. Those in greenhouse 2 were Aspergillus/Penicillium-like, Cladosporium, Botrytis, yeast-like, and hyphal fragments. Maximum concentrations of Trichoderma and total spores in greenhouse 1 were 36,426 and 49,729 spores/m3, respectively. Maximum concentrations of Aspergillus/Penicillium and total spores in greenhouse 2 were 46,961 and 71,037 spores/m3, respectively. Airborne fungal populations fluctuated dramatically within 2 h during work hours, tenfold for Aspergillus/Penicillium, 66-fold for Trichoderma, and sevenfold for total spores. QPCR detected Trichoderma harzianum ranging from 7 to 3,500 conidia E/m3. Aspergillus/Penicillium and Botrytis showed diurnal patterns, but not Trichoderma. Aspergillus/Penicillium and Cladosporium were positively correlated with temperature, relative humidity, dew point, heat index, and light and negatively with air movement and air pressure. Botrytis and Trichoderma were not correlated with the environmental factors. Greenhouse workers were potentially exposed up to 71,037 spores/m3 of airborne fungi.  相似文献

【背景】广陈皮为药食同源中药材,在高温、高湿且贮存不当的条件下容易发霉,从而产生毒素,严重威胁陈皮的质量安全。【目的】分析广陈皮表面外源真菌的组成及其产生毒素的真菌。【方法】采用平板稀释法分离广陈皮表面外源真菌,利用分生孢子形态特征及DNA序列分析进行真菌鉴定,采用高效液相色谱-三重串联四极杆质谱联用技术对青霉属和曲霉属进行产毒检测。【结果】从广陈皮表面分离外源真菌共132株,鉴定为子囊菌亚门(Ascomycota,98.48%)和毛霉菌亚门(Mucoromycota,1.52%),包括散囊菌纲(Eurotiomycetes,95.45%)、座囊菌纲(Dothideomycetes,3.03%)和毛霉纲(Mucoromycetes,1.52%)。分离活动曲霉属(Aspergillus spp.)菌株共77株,为广陈皮药材外源真菌的优势菌,青霉属(Penicillium spp.)次之。毒素检测筛选出1株产毒真菌,鉴定为黄曲霉(Aspergillus flavus) JXCP1-3。该菌株产生黄曲霉毒素B1 (aflatoxin B1,AFB<...  相似文献

Information about airborne fungal spore is crucial for health risk assessment and management, especially for patients with allergy and asthma. Nonetheless, such data are rarely available from certain areas of the world, including Southeast Asia. The aim of this study was to gain updated information about airborne fungal spore in Bangkok, the capital city of Thailand. A survey was conducted at five sampling sites in Bangkok, using the Rotorod Sampler® for a period of 1 year. High concentrations of spores were found all year with the peak between August and November. The most prominent spore types were Cladosporium, Nigrospora, Puccinia, Aspergillus/Penicillium, and Fusarium. The spore concentrations were positively and significantly correlated with the amount of rainfall and relative humidity, reaching the maximum level in September. Sensitization rates to Cladosporium, Penicillium, and Aspergillus among Thai atopic patients were approximately 16.6, 13.6, and 13.0%, respectively.  相似文献

A quantitative and qualitative survey was carried out for airborne fungus spores coming into contact with horizontally and vertically gravitation sampling oriented surfaces in the atmosphere of Giza city. Czapek Dox agar, malt extract agar, potato dextrose agar and Sabouraud dextrose agar Petri dishes were exposed monthly to the five oriented surfaces of a polystyrene cube, throughout a one-year period. Significant differences (P < 0.01) were observed between the total counts of caught airborne fungi contacting with the horizontal compared to other vertically oriented surfaces. Conversely, there were no significant differences observed between the total catch of airborne fungi using the various sampling media. The results revealed that vertical sampling provides valuable information that may be lost from horizontal sampling alone. A total of 5,053 colonies belonging to 40 fungal organisms were identified. Alternaria (24.26%), Aspergillus (19.2%), Cladosporium (14.5%) and Penicillium (11.43%) were the most predominant fungal genera. Collected fungi were grouped into high, medium, low and rare components depending upon their frequency in the studied atmosphere. Aspergillus niger, Aspergillus parasiticus, Alternaria, Cladosporium and Penicillium were regularly found on all oriented surfaces. However, Arthrobotrys, Biospora, Chaetomium, Pleospora, Trichothecium and Verticillium were rarely found in the air. Positive and/or negative correlations were observed between the total fungal counts and the predominant fungal types with meteorological parameters during sampling days.  相似文献

大量的稀土-重金属通过尾矿坝的浮尘、地表径流和渗滤液排放到周边土壤中,影响了土壤中的微生物群落结构。【目的】分析稀土和重金属复合污染土壤真菌群落结构并分离具有同时吸附稀土和重金属的菌株。【方法】本研究基于ITS基因,采用Illumina-Hiseq测序技术分析了包头稀土尾矿坝周边5份稀土-重金属污染土壤样品和距尾矿区20 km的1份相对未受污染的土壤样品的真菌群落特征,同时采用富集培养法从污染样品中筛选出金属耐性真菌,并对其进行吸附稀土-重金属的特性分析。【结果】群落结构为:在门水平,除了未分类门真菌(unclassified Fungi)外,子囊菌门(Ascomycota)真菌在所有土壤中占比较大(13.5%–90.5%);在纲水平上,除了未分类纲真菌外,粪壳菌纲(Sordariomycetes)真菌在B2 (73.1%)、B3 (28.4%)和B4 (20.8%)的丰度显著高于对照样点C (7.4%),而座囊菌纲(Dothideomycetes)在B5 (11.8%)的丰度明显高于B1 (3.5%);在属水平,除了未分类属,足孢子虫属(Podospora)是C(0.9%)和B3(23.6%)样点的优势种。曲霉属(Aspergillus)、未分类的格孢腔菌目(unclassified Pleosporales)和未分类的戴维迪科(unclassified Davidiellaceae)分别为B1 (3.0%)、B4 (10.5%)和B5(5.8%)的优势种,而蜡蚧属(Lecanicillium)真菌只在B2样点土壤存在且占优(51.6%)。Zn污染对真菌群落结构的影响大于稀土元素污染,且其浓度与优势的未分类真菌相对丰度呈负相关。从污染样品中共分离出6株真菌,它们分属于曲霉属(Aspergillus)(5株)和镰刀霉菌属(Fusarium)(1株)。所有分离菌株对镧(La~(3+))的吸附率均显著高于锌(Zn~(2+)),其中Aspergillus sp. B6-3对La~(3+)和锌Zn~(2+)的吸附率最高,分别为19.7%和3.9%。【结论】该研究为利用真菌去除稀土和重金属以优化生物吸附过程导向的环境生物修复和保护策略提供了机制基础。  相似文献

The occurrence of fungal spores in the upper respiratory tract and in the homes of 86 patients with allergic manifestations and positive skin tests for fungi has been investigated.Alternaria tenuis andCladosporium herbarum have been more frequently found to give positive reactions, followed byAspergillus mix,Candida albicans andPenicillium notatum. Cladosporium andAlternaria represented also the most common moulds isolated from nasal specimens. The prevalent fungi isolated from patients' environments wereCladosporium, Penicillium andAlternaria, occurring respectively in 93%, 78% and 72% of the homes sampled. An examination of individual cases have revealed a surprisingly close correlation, in 47 out of 86 patients, between positive skin test response and presence in high amount of the same fungi in patient's respiratory tract and home. In the majority of cases the fungi involved wereAlternaria and/orCladosporium.  相似文献
