施氮水平对小麦籽粒发育过程中氨基酸含量的影响   总被引:11,自引:0,他引:11  
施氮能提高小麦籽粒蛋白质氨基酸的含量,并与施氮水平呈正相关;但对普通小麦必需氨基酸与蛋白质氨基酸的比值没有影响,而硬粒小麦4286随施氮水平的提高,该比值下降。在开花后32d以前,籽粒发育过程中游离氨基酸与施氮水平呈正相关,以后,籽粒中游离氨基酸趋于相近,表明施氮增加了游离氨基酸的库源,不同基因型小麦对施氮水平的反应不同,在同等施氮水平和栽培条件下,籽粒中蛋白质氨基酸和游离氨基酸含量为硬粒小麦4286>小偃6号>小偃107,不同施氮水平下,籽粒中氨基酸含量为高氮>中氮>低氮。  相似文献

Developmental changes of amino acids in ovine fetal fluids   总被引:3,自引:0,他引:3  
We recently reported an unusual abundance of arginine (4-6 mM) in porcine allantoic fluid during early gestation. However, it is not known whether such high concentrations of arginine are unique for porcine allantoic fluid or whether they represent an important physiological phenomenon for mammals. The present study was conducted to test the hypothesis that arginine is also the most abundant amino acid in ovine allantoic fluid. Allantoic and amniotic fluids, as well as fetal and maternal plasma samples, were obtained from ewes between Days 30 and 140 of gestation. Glycine was the most abundant amino acid in maternal uterine arterial plasma, representing approximately 25% of total alpha-amino acids. Alanine, glutamine, glycine, plus serine contributed approximately 50% of total alpha-amino acids in fetal plasma. Fetal:maternal plasma ratios for amino acids varied greatly, being less than 1 for glutamate during late gestation, 1.5-3 for most amino acids throughout gestation, and greater than 10 for serine during late gestation. Marked changes were observed in amino acid concentrations in amniotic and allantoic fluids associated with conceptus development. Concentrations of alanine, citrulline, and glutamine in allantoic fluid increased by 20-, 34-, and 18-fold, respectively, between Days 30 and 60 of gestation and were 24.7, 9.7, and 23.5 mM, respectively, on Day 60 of gestation (compared with 0.8 mM arginine). Remarkably, alanine, citrulline, plus glutamine accounted for approximately 80% of total alpha-amino acids in allantoic fluid during early gestation. Serine (16.5 mM) contributed approximately 60% of total alpha-amino acids in allantoic fluid on Day 140 of gestation. These novel findings of the unusual abundance of traditionally classified nonessential amino acids in allantoic fluid raise important questions regarding their roles in ovine conceptus development.  相似文献

Summary The effect of nitrogen applications on the amino acid composition of wheat grain was studied in a pot experiment, in which spring wheat was supplied with increasing amounts of ammonium sulphate. The ensuing yield curve extended from very small N-applications to applications resulting in a toxic effect on the yield.The contents of dry matter, total N, 15 amino acids and ammonia were determined in samples taken from 9 representative treatments. The relationship between the yield of dry matter and the concentration in the dry matte rof the individual amino acids showed the same characteristic variations as those previously found for total N and for a number of plant nutrients.In accordance with the results cited in the literature, increasing applications of N caused an increase of glumatic acid and proline on a nitrogen basis, whereas lysine and arginine decreased.  相似文献

We have previously shown, using truncated soluble recombinant receptors, that substituting the 62 N-terminal amino acids of the alpha subunit from the insulin-like growth factor I receptor (IGFIR) with the corresponding 68 amino acids from the insulin receptor (IR) results in a chimeric receptor with an approximately 200-fold increase in affinity for insulin and only a 5-fold decrease in insulin-like growth factor I (IGFI) affinity (Kjeldsen, T., Andersen, A. S., Wiberg, F. C., Rasmussen, J. S., Sch?ffer, L., Balschmidt, P., M?ller, K. B., and M?ller, N. P. H. (1991) Proc. Natl. Acad. Sci. U.S.A. 88, 4404-4408). We demonstrate that these 68 N-terminal amino acids of the IR also confer insulin affinity on the intact IGFI holoreceptor both in the membrane-bound state and when solubilized by Triton X-100. Furthermore, this domain can be subdivided into two regions (amino acids 1-27 and 28-68 of the IR alpha subunit) that, when replacing the corresponding IGFIR sequences, increases the insulin affinity of truncated soluble receptor chimeras 8- and 20-fold, respectively, with only minor effects on the IGFI affinity. Within the latter of these two regions, we found that amino acids 38-68 of the IR, representing 13 amino acid differences from IGFIR, confer the same 20-fold increase in insulin affinity on the IGFIR. Finally, the amino acids from position 42 to 50 are not responsible for this increase in insulin affinity. We thus propose that at least two determinants within the 68 N-terminal amino acids of the insulin receptor are involved in defining the ligand specificity of the insulin receptor, and that one or a combination of the remaining seven amino acid differences between position 38 and 68 are involved in conferring insulin affinity on the insulin receptor.  相似文献

The influence of protein quality on the growth-depressing effect of excessive amount of 12 individual essential and semiessential amino acids was examined. Growing rats were fed for 3 weeks diets containing either 10.5% egg albumin or 11.6% wheat gluten (equivalent to the protein content of a 10% casein diet) supplemented with 5% of each of the l-amino acids. In general, the pattern of growth depression produced by the addition of excess amino acids to the egg albumin or the wheat gluten diet was similar to that of the case of casein diet obtained previously under the same experimental conditions. However, the extent of these effects was dependent not only upon the kind of amino acid supplemented with but also upon the source of protein used, and the depressing effect of each of excess amino acids added to the wheat gluten diet was usually severer than those added to casein and egg albumin diets. No evidence was noted of any striking changes in the liver protein and nucleic acid concentrations by either diets, but total liver protein, RNA and DNA contents were decreased in some amino acid groups of the egg albumin diet and in all amino acid groups of the wheat gluten diet except the lysine addition. The free amino acid level in plasma generally showed extreme elevation for the amino acid supplemented in excess in the diet, and in most cases the extent of the elevation was correlated with the growth depression.  相似文献

For most prokaryotic organisms, amino acid biosynthesis represents a significant portion of their overall energy budget. The difference in the cost of synthesis between amino acids can be striking, differing by as much as 7-fold. Two prokaryotic organisms, Escherichia coli and Bacillus subtilis, have been shown to preferentially utilize less costly amino acids in highly expressed genes, indicating that parsimony in amino acid selection may confer a selective advantage for prokaryotes. This study confirms those findings and extends them to 4 additional prokaryotic organisms: Chlamydia trachomatis, Chlamydophila pneumoniae AR39, Synechocystis sp. PCC 6803, and Thermus thermophilus HB27. Adherence to codon-usage biases for each of these 6 organisms is inversely correlated with a coding region's average amino acid biosynthetic cost in a fashion that is independent of chemoheterotrophic, photoautotrophic, or thermophilic lifestyle. The obligate parasites C. trachomatis and C. pneumoniae AR39 are incapable of synthesizing many of the 20 common amino acids. Removing auxotrophic amino acids from consideration in these organisms does not alter the overall trend of preferential use of energetically inexpensive amino acids in highly expressed genes.  相似文献

本文采用高速氨基酸自动分析仪对太子参四大种植主产区(安徽、江苏、福建、贵州)的栽培品及河南的野生品,共13个批次太子参药材的氨基酸组成和含量进行测定。结果显示氨基酸含量基本顺序为:精氨酸(Arg)>谷氨酸(Glu)>天冬氨酸(Asp)>脯氨酸(Pro),其中安徽宣城产太子参样品(S2)的16种氨基酸总含量和7种人体必需氨基酸含量最高。以上述4种主要氨基酸作为指标,探讨了不同产区太子参药材的内在品质。采用聚类分析法对氨基酸含量进行多元统计分析,表明氨基酸含量分析可体现样品产区间和产区内的差异性,可区别野生品与栽培品。这些结果为发展太子参道地药材的质量标准控制提供了依据。  相似文献

Trends in the time course, changes in the moisture, soluble amino acids, proline, abscisic acid contents and nitrate reductase activity determined byin vivo method in the developing seeds of wheat were studied. Maximum dry matter augmentation in the seed took place in the period between 10–30 days after anthesis. Per cent moisture and moisture content started declining 15 days and 25 days after anthesis, respectively. Levels of soluble amino acids, proline and nitrate reductase activity were higher during initial stages of seed development, but decreased with increasing magnitude of dehydration and accumulation of abscisic acid (ABA) in the maturing seeds.  相似文献

以中华猕猴桃(Actinidia chinensis Planch.)品种‘H-1’为材料,研究有机栽培和生态绿色栽培模式对其可溶性固形物、总糖、总酸、Vc、氨基酸、矿物元素和香气等果实品质性状的影响。结果显示:生态绿色栽培的‘H-1’中,果实可溶性固形物、总糖和Vc含量比有机栽培的果实分别高出15.90%、26.30%和29.25%,其中Vc的差异最为明显;总酸含量与有机栽培种相差不大;有机栽培的‘H-1’中必需氨基酸的含量是生态绿色栽培种的1.69倍,且锌和钙的含量也高于生态绿色栽培,但钾元素含量相差不明显;生态绿色栽培的‘H-1’果实中可检出的果香成分比有机栽培多5种。研究结果表明栽培方式不同,猕猴桃果实品质表现出一定差异,可根据果实鲜食及加工用途的不同选择合适的栽培方式。  相似文献

NaCl-stress induced a pronounced suppression in growth of wheat seedlings. The most abundant amino acids (cysteine, arginine, methionine) constituting about 55 % of total free amino acid content in control wheat were reduced in 100 mM NaCl-treated plants. However, valine, isoleucine, aspartic acid and proline accumulated in response to NaCl stress and NaCl-treated wheat seedlings showed 1.6 fold increase in total free amino acids compared to the control. Addition of 2 [micro ]M thiamine alleviated the effects of NaCl on the amino acid composition and the amount of total free amino acids decreased to that in the control. Content of 26 kDa protein increased in NaCl-treated plants, stimulation was more pronounced in roots than in shoots. In contrast, the contents of 13 and 20 kDa proteins decreased. After addition of thiamine, the 24 kDa protein, which disappeared with NaCl treatment, has been initiated again. Moreover, thiamine treatment stimulated the accumulation of the 20 kDa protein.  相似文献

利用氨基酸自动分析仪对板蓝根的新鲜植物组织和不同加工程度的板蓝根药材中的总氨基酸和游离氨基酸含量进行测定。结果表明:板蓝根的新鲜植物组织和不同加工程度的板蓝根药材氨基酸在含量上有明显差别。结论为:不同加工程度的板蓝根药材中氨基酸含量差别明显,其中碱性氨基酸含量的差别尤其显著,该差异可能是由于加工过程中发生美拉德反应造成的。  相似文献

Summary Mutagenesis and the subsequent selection of mesophyll diploid protoplasts ofNicotiana sylvestris on growth inhibitory concentrations of lysine plus threonine has led to the isolation of an LT-resistant mutant. Regeneration of this line (RLT 70) and analysis of its descendants demonstrated the dominant monogenic nuclear character of the resistance gene, further namedak-LT1. When the inhibition properties of aspartate kinase were examined in the homozygous mutant, lysine-sensitive activity could no longer be detected. In comparison, 70%–80% of the wild-type enzyme activity was usually inhibited by lysine, and the rest by threonine. Evidence for the existence of at least two AK isoenzymes was obtained by ion-exchange chromatography, where two peaks of activity could be detected: the first one to be eluted is lysine sensitive, and the second one threonine sensitive. One consequence of the altered regulation of AK in the mutant was the enhanced production of soluble threonine. Threonine accumulation was observed to occur throughout the life cycle of the mutant plant as well as in its different organs. In particular, leaves exhibited a 45-fold increment of soluble threonine, which corresponds to a 13-fold increase in total threonine: almost one-third of the total amino acids was free and proteinbound threonine. In RLT 70 seeds, 20% of the free amino acid pool was in the form of threonine (70-fold accumulation compared to the wild type), and total threonine content was increased five fold. As a general rule, the other amino acids were also more abundant in RLT 70 seeds, such that the total of amino acids present was between two to four times higher, but in contrast with the situation encountered in leaves, this was also due to a higher protein-bound amino acid content.  相似文献

Despite increasing recognition that free amino acids can be an important source of N for plant uptake, we have a poor understanding of environmental variation in the availability of amino-acid N in soils outside of arctic, alpine and boreal regions. I investigated patterns of amino-acid availability along a temperate forest fertility gradient ranging from low mineral N availability, oak-dominated forests to high mineral N availability, maple-basswood forests (5 sites). I measured standing pools of free amino acids, soluble peptides, ammonium and nitrate, rates of amino acid production (native proteolysis activity) and rates of consumption of a 15N-labeled leucine tracer. Standing pools of amino acid N decreased consistently along the fertility gradient from the low fertility black oak/white oak system to the high fertility sugar maple/basswood system, with a 25-fold difference in pool sizes between the poorest and richest sites. Standing pools of soluble peptides varied little among sites, instead, the relationship between free amino acids and peptides changed markedly across the gradient. At low fertility sites free amino acids were positively correlated with soluble peptides, whereas free amino acid pools were universally low at high fertility sites, regardless of peptide pools. Assays for native proteolysis activity indicated that amino acid production did not vary significantly among sites. Recovery of leucine tracer in inorganic (NH4 + and NO3 ) pools and in residual soil organic matter both increased with increasing soil fertility; however, total consumption of the added amino-acid tracer did not vary among sites. Results from this study demonstrate that free amino acids can make an important contribution to potentially plant-available N pools in temperate forest soils, particularly at low fertility sites.  相似文献

Changes in free amino acids (FAA) in the hemolymph of the giant freshwater prawn, Macrobrachium rosenbergii, were examined in individuals exposed to varying salinities for up to 1 week. In freshwater and under conditions of low salinity, total FAA concentrations were maintained between approximately 0.85 and 1 mM and did not exhibit changes in response to salinity exposure. Under high salinities, total FAA concentrations increased dramatically, reaching up to 2.1 mM depending on treatment. Examination of individual amino acid concentrations revealed that these increases were based on specific changes in glycine, arginine, alanine, proline and lysine. Among these, alanine showed the greatest increases, resulting in levels six-fold higher under high salinity than in freshwater and under low salinity. The other amino acid species showed increases of 2.5-fold compared to original values. These five FAAs in freshwater and under low salinity together occupied approximately 45% of total FAA contents and under high salinity comprised more than 70% of total FAA contents. These results suggest that specific hemolymph FAAs are involved in mediating response to salinity exposure in freshwater prawns.  相似文献

Amino acid composition and fatty acid composition were determined on seed samples of a range of white lupin (Lupinus albus) cultivars and accessions grown in either of two environments.Variability between genotypes was found for lysine, arginine and glutamic acid content, but not for the concentrations of other amino acids. The deficiency in sulphurcontaining amino acids, typical of legume proteins, was evident, with methionine and cyst(e)ine totalling only 2.2% of the protein. Variability was limited, indicating that improvement by breeding would be impracticable. Lupinus albus differed slightly from other lupin species in amino acid composition, having higher levels of threonine, tyrosine and isoleucine, but a lower level of glutamic acid than both L. angustifolius and L. luteus. Four low-alkaloid lines of L albus each had higher lysine content than the high-alkaloid line, but ‘Kiev Mutant’, despite earlier claims, had a lysine level no higher than the other three low-alkaloid lines.Fatty acid composition of the seed oil varied considerably between genotypes. Oleic acid ranged from 43.6 to 54.4% and linolenic acid from 6.7 to 15.2%, these two fatty acids being negatively correlated at one site. Linoleic acid content varied between 17.2 and 26.9% and was not correlated with other fatty acids. Total oil content averaged 9.6% with little variability between lines.It is concluded that, relative to other lupin species, L. albus has a more favourable amino acid profile for its utilisation in cereal-based diets for animals, particularly if the energy source is wheat, which is deficient in threonine. The higher oil content would be an important energy benefit to such diets and may allow their protein/energy balance to be maintained at higher levels of incorporation of L. albus seed meal than is possible with other lupin species.  相似文献

采用现代化学分析手段对平菇菌丝体和子实体的营养成分进行分析比较。结果表明,用PDA固体培养的菌丝体的灰分、可溶物、总糖和粗蛋白的含量均明显低于子实体,而粗脂肪和粗纤维的含量则显著高于子实体;菌丝体的Fe、Ca、Zn的含量显著高于子实体,而Mg和K的含量明显低于子实体,Na的含量两者比较接近;菌丝体和子实体的氨基酸组成相似,都含有17种氨基酸,但子实体的各氨基酸的含量均高于菌丝体,从各项氨基酸含量的比值来看,菌丝体则更优于子实体。两者都不失是极好的营养食品源。  相似文献

利用氨基酸自动分析仪对10个山东栽培瓜蒌样品中的水解氨基酸和游离氨基酸进行含量测定.结果发现山东栽培瓜萎中氨基酸的种类较为丰富,均含有19种相同的游离氨基酸和水解氨基酸;氨基酸含量在各个样品之间差异显著,从游离总氨基酸的含量考虑,糖瓜蒌略优于仁瓜萎,仁瓜蒌优于地瓜萎.  相似文献

This paper presents stable carboxypeptidase A (CPA)-glyoxyl derivatives, to be used in the controlled hydrolysis of proteins. They were produced after immobilizing-stabilizing CPA on cross-linked 6% agarose beads, activated with low and high concentrations of aldehyde groups, and different immobilization times. The CPA-glyoxyl derivatives were compared to other agarose derivatives, prepared using glutaraldehyde as activation reactant. The most stabilized CPA-glyoxyl derivative was produced using 48 h of immobilization time and high activation grade of the support. This derivative was approximately 260-fold more stable than the soluble enzyme and presented approximately 42% of the activity of the soluble enzyme for the hydrolysis of long-chain peptides (e.g., cheese whey proteins previously hydrolyzed with immobilized trypsin and chymotrypsin) and of the small substrate N-benzoylglycyl-l-phenylalanine (hippuryl-l-Phe). These results were much better than those achieved using the conventional support, glutaraldehyde-agarose. Amino acid analysis of the products of the acid hydrolysis of CPA (both soluble and immobilized) showed that approximately four lysine residues were linked on the glyoxyl agarose beads, suggesting the existence of an intense multipoint covalent attachment between the enzyme and the support. The maximum temperature of hydrolysis was increased from 50 degrees C (soluble enzyme) to 70 degrees C (most stable CPA-glyoxyl derivative). The most stable CPA-glyoxyl derivative could be efficiently used in the hydrolysis of long-chain peptides at high temperature (e.g., 60 degrees C), being able to release 2-fold more aromatic amino acids (Tyr, Phe, and Trp) than the soluble enzyme, under the same operational conditions. This new CPA derivative greatly increased the feasibility of using this protease in the production of protein hydrolysates that must be free of aromatic amino acids.  相似文献
