Strictly simultaneous eyelid conditioning in human Ss on the one hand and forward (ISI: 0.45 s), backward (ISI: -0.45 s), and pseudoconditioning on the other hand were compared in two different experimental situations: one-session experiment (100 reinforcements, 20 isolated CS randomly interspersed), and ten-sessions experiment (20 reinforcements, 4 isolated CS in each session), UCS was a light flash (0.5 J, 20 microseconds duration), CS a tone (1000 Hz, 50 ms, 73 dB). In both the experiments, strictly simultaneous conditioning was significantly different from pseudoconditioning, but significantly less effective than forward conditioning. In the ten-sessions experiment, it was also less effective than backward conditioning, while in the one-session experiment, it did not differ from backward conditioning. Statistical mixture decomposition method applied to the one-session- and the ten-sessions- experimental samples of individual acquisition curves resulted into the division of both samples into 4 subgroups according to the trend of acquisition curves. A significant positive association between the conditioning procedure and the distribution of individual curves into the four subgroups was found.  相似文献

In order to achieve a better understanding of multiple infections and long latency in the dynamics of Mycobacterium tuberculosis infection, we analyze a simple model. Since backward bifurcation is well documented in the literature with respect to the model we are considering, our aim is to illustrate this behavior in terms of the range of variations of the model's parameters. We show that backward bifurcation disappears (and forward bifurcation occurs) if: (a) the latent period is shortened below a critical value; and (b) the rates of super-infection and re-infection are decreased. This result shows that among immunosuppressed individuals, super-infection and/or changes in the latent period could act to facilitate the onset of tuberculosis. When we decrease the incubation period below the critical value, we obtain the curve of the incidence of tuberculosis following forward bifurcation; however, this curve envelops that obtained from the backward bifurcation diagram.  相似文献

The aim of this study was to explore the effect of a change in the speed and direction of shift rotation on the sleep and wakefulness of younger and older workers. A continuous three-shift schedule was changed from a slow backward rotating (EEE-MMMNNN-----) to a fast forward rotating system (MMEENN----). Sixteen subjects (mean age 42 years) were studied before and one year after the change in schedule. Two age groups were compared: ten younger men (mean age 35 years) and six older men (mean age 53 years). The effects of the new work schedule were evaluated by a questionnaire (modified SSI), and on-site registrations with an actigraph and sleep log for one shift cycle (10-15 days) before and after the new schedule. After the change in schedule, subjective sleep problems decreased and alertness increased during the morning shifts. The change in schedule influenced sleep differently in the two age-groups. Both the subjective and objective quality of sleep improved among the older workers. The results indicate that a fast forward rotating shift schedule is more suitable for older workers than a slower backward rotating system.  相似文献

We study the equilibriumbehaviour of a deterministic four-statemutation-selection model as a model for the evolution of a population of nucleotide sequences in sequence space. The mutation model is the Kimura 3ST mutation scheme, and the selection scheme is assumed to be invariant under permutation of sites. Considering the evolution process both forward and backward in time, we use the ancestral distribution as the stationary state of the backward process to derive an expression for the mutational loss (as the difference between ancestral and population mean fitness), and we prove a maximum principle that determines the population mean fitness in mutation-selection balance.  相似文献

A new strategy is used for studying the internal motions of proteins based on measurements of NMR relaxation parameters. The strategy yields values of the so-called spectral density functions J(omega) for N-H bond vectors. The spectral density functions are related to the distribution of frequencies contained in the rotational (overall and internal) motions of these NH bond vectors. No a priori model assumptions about the dynamics are required in this approach. The method involves measurements of six relaxation parameters consisting of 15N longitudinal relaxation rates, transverse relaxation rates of in-phase and antiphase coherence, the relaxation rates of heteronuclear 1H-15N two-spin order, the heteronuclear 1H-15N nuclear Overhauser effects, and longitudinal relaxation rates of the amide protons. The values of the spectral density functions at the five frequencies 0, omega N, omega H + omega N, omega H, and omega H - omega N are determined from the relaxation parameters using analytical relations derived previously [Peng & Wagner (1992) J. Magn. Reson. 98, 308-332]. Here, the method is applied to characterize the backbone dynamics of the 15N-enriched proteinase inhibitor eglin c, a protein of 70 residues. The values for J(0) and J(omega N = 50 MHz) vary significantly with the amino acid sequence, whereas the spectral densities at higher frequencies, J(450 MHz), J(500 MHz), and J(550 MHz), are typically much smaller and show no significant variation with the sequence. The collective behavior of the J(omega) values indicate greater internal motion for the proteinase binding loop residues and the first eight N-terminal residues. The additional internal motion in these regions is in the rate range below 450 MHz. The values of J(omega) are also compared with root mean square deviations (rmsds) of backbone atoms as obtained in NMR structure determinations. Low values of J(0) and J(omega N) are correlated with high rmsds. Spectral densities at higher frequencies, J(450 MHz), J(500 MHz), and J(550 MHz), are small and show no correlation with rmsds. A comparison with the spectral density functions obtained by fitting the experimental data to the functional dependence of the Lipari and Szabo formalism [Lipari & Szabo (1982a) J. Am. Chem. Activation of the ppp(A2'p)nA (2-5A)-dependent RNase was investigated during the abortive infection of BSC40 cells by a temperature-sensitive mutant of vaccinia virus, ts22. At the nonpermissive temperature, ts22 has an abortive late phenotype. At the onset of late-viral-gene expression, viral mRNA is degraded and rRNA is cleaved into discrete fragments in the absence of prior interferon treatment (R. F. Pacha and R. C. Condit, J. Virol. 56:395-403, 1985). Concomitant with rRNA cleavage, an increase in 2-5A occurred late during infection. Discrete 18S- and 28S-rRNA degradation products from BSC40 cells infected with ts22 at the nonpermissive temperature comigrated in denaturing agarose gels with rRNA cleaved fragments produced by the activation of 2-5A-dependent RNase in uninfected cells transfected with exogenous 2-5A. An increase in 2-5A levels and a similar discrete and characteristic degradation of rRNA were observed in BSC40 cells infected with wild-type vaccinia virus in the presence of isatin-beta-thiosemicarbazone. The results show that the ts22 lesion and the action of isatin-beta-thiosemicarbazone may affect the same pathway, leading to the activation of latent 2-5A-dependent RNase and resulting in indiscriminate RNA degradation and inhibition of viral replication.  相似文献

The Leslie population projection matrix may be used to project forward in time the age distribution or age-sex distribution of a population. As it is a singular matrix, it does not have an inverse, and so it is not clear that there is a corresponding procedure for backward projection. Immunohistochemistry of Glut3 (45 kD), an integral membrane peptide mediating the transport of glucose in neurons, was carried out in the hippocampus of 3- and 28-month-old rats to assess the effect of age on energy metabolism. Free-floating sections of fixed-frozen hippocampi were processed for quantitative immunohistochemistry of Glut3. A rabbit affinity-purified antibody identified Glut3 immunoreactivity. Glut3 staining was intense in neuropil, axons, and dendrites, whereas nerve cell bodies were unstained. With aging, Glut3 reactivity was significantly decreased in the inner molecular layer of the hippocampal dentate gyrus (-46%) and the mossy fibers of the CA3 sector (-34%), whereas the stratum radiatum of CA1 did not show any difference due to age. These data document an age-dependent decrease in Glut3 expression in discrete areas of rat hippocampus. Glut3 constitutes the predominant glucose transporter in neurons and is found abundantly in regions with high synaptic density characterized by frequent bursts of function-adequate metabolic activity. Our findings therefore lend further support to the critical role of an impaired metabolism in age-related brain dysfunctions and disease.(J Histochem Cytochem 49:671-672, 2001)  相似文献

This paper presents a mathematical algorithm that computes the sizes and growth rates of breast cancer detected in a hypothetical population that is screened for the disease. The algorithm works by simulating the outcomes of the hypothetical population twice, first without screening and then with screening. The simulation without screening relies on an underlying model of the natural history of the disease. The simulation with screening uses this natural history model to track the growth of breast tumors backwards in the time starting from the time they would have been detected without screening. The method of tracking tumor growth backward in time is different from methods that track tumor growth forward in time by starting from an estimated time of tumor onset. The screening algorithm combines the natural history model, the method tracking of tumor growth backward in time, the age group, the interval between screening exams, and the detection threshold of the screening exam to compute the joint distribution of tumor size and growth rate among screen-detected and interval patients. The algorithm also computes the sensitivity and leadtime distribution. It allows for arbitrary age groups, detection thresholds and screening intervals and may contribute to the design of future screening trials.  相似文献

The kinetic mechanism of rat muscle adenylosuccinate synthetase was studied by determining the rates of isotope exchange at equilibrium. A random sequential binding mechanism was indicated for both the forward and reverse reactions. Aspartate, adenylosuccinate, GDP, and Pi were determined to bind in rapid equilibrium. GTP exchanges with both GDP and Pi at the same rate, which is similar to the exchange rate of IMP with adenylosuccinate. Aspartate exchanges with adenylosuccinate at a higher rate than does IMP over the range of concentrations tested. The slower IMP and GTP exchange rates suggest a forward binding mechanism containing a preferred path in which the quaternary complex is most often formed by aspartate binding to the E-GTP-IMP complex. This preferred path is consistent with an interaction between IMP and GTP in the absence of aspartate as determined by isotope scrambling experiments [Bass, M. B., Fromm, H. J., & Rudolph, F. B. (1984) J. Biol. Chem. 259, 12330-12333]. However, the products of such an interaction are tightly bound to the enzyme as no partial exchange reactions between adenylosuccinate and aspartate in the presence or absence of Pi were detected.  相似文献

(1) Autocorrelation measurements were made of the current fluctuations due to ion-conducting channels produced by gramicidin A in black lipid membranes. (2) Relaxation experiments using a voltage jump were made on the same system. (3) Reciprocal time constants were determined by both methods over a 108 fold range in membrane conductivity. starting from the single channel level. (4) The mean squared amplitudes of the fluctuations were determined from the autocorrelation functions. (5) The data were tentatively rationalized on the basis of a hypothetical dimerization reaction, assuming that gramicidin A dimers form conducting channels. The same forward rate constants are obtained, via (3), by both methods (1) and (2). The backward rate constant agrees excellently with direct measurements of the mean life time of a conducting channel. (6) The unit channel conductance and - assuming again a dimerization - the equilibrium constant can be obtained from the fluctuation amplitude distribution.  相似文献

Hyperelastic anisotropic microplane constitutive model for annulus fibrosus   总被引:3,自引:0,他引:3  
In a recent paper, Peng et al. (2006, "An Anisotropic Hyperelastic Constitutive Model With Fiber-Matrix Interaction for the Human Annulus Fibrosis," ASME J. Appl. Mech., 73(5), pp. 815-824) developed an anisotropic hyperelastic constitutive model for the human annulus fibrosus in which fiber-matrix interaction plays a crucial role in simulating experimental observations reported in the literature. Later, Guo et al. (2006, "A Composites-Based Hyperelastic Constitutive Model for Soft Tissue With Application to the Human Fibrosis," J. Mech. Phys. Solids, 54(9), pp. 1952-1971) used fiber reinforced continuum mechanics theory to formulate a model in which the fiber-matrix interaction was simulated using only composite effect. It was shown in these studies that the classical anisotropic hyperelastic constitutive models for soft tissue, which do not account for this shear interaction, cannot accurately simulate the test data on human annulus fibrosus. In this study, we show that the microplane model for soft tissue developed by Caner and Carol (2006, "Microplane Constitutive Model and Computational Framework for Blood Vessel Tissue," ASME J. Biomech. Eng., 128(3), pp. 419-427) can be adjusted for human annulus fibrosus and the resulting model can accurately simulate the experimental observations without explicit fiber-matrix interaction because, in microplane model, the shear interaction between the individual fibers distributed in the tissue provides the required additional rigidity to explain these experimental facts. The intensity of the shear interaction between the fibers can be adjusted by adjusting the spread in the distribution while keeping the total amount of the fiber constant. A comparison of results obtained from (i) a fiber-matrix parallel coupling model, which does not account for the fiber-matrix interaction, (ii) the same model but enriched with fiber-matrix interaction, and (iii) microplane model for soft tissue adapted to annulus fibrosus with two families of fiber distributions is presented. The conclusions are (i) that varying degrees of fiber-fiber and fiber-matrix shear interaction must be taking place in the human annulus fibrosus, (ii) that this shear interaction is essential to be able to explain the mechanical behavior of human annulus fibrosus, and (iii) that microplane model can be fortified with fiber-matrix interaction in a straightforward manner provided that there are new experimental data on distribution of fibers, which indicate a spread so small that it requires an explicit fiber-matrix interaction to be able to simulate the experimental data.  相似文献

A theoretical investigation is presented which allows the calculation of states of maximal reaction rates for single enzymes and for unbranched enzymatic chains. As an extension to previous papers (Heinrich & Holzhütter, 1985, Biomed. biochim. Acta 44, 959-969; Heinrich et al., 1987, Bull. math. Biol. 49, 539-595) a detailed enzymatic mechanism was taken into consideration. Conclusions are drawn for the optimal values of the microscopic rate constants as well as of the maximal activities and Michaelis constants. Ten solutions are found which depend on the equilibrium constant as well as on the concentrations of substrates and products. It is shown that for high equilibrium constants one of the solutions applies to a very large range of the concentrations of the outer reactants. This solution is characterized by maximal values of the rate constants of all forward reactions and by non-maximal values of the rate constants of all backward reactions. In contrast to previous assumptions (Albery & Knowles, 1976b, Biochemistry 15, 5631-5640; Burbaum et al., 1989, Biochemistry 28, 9293-9305) states of maximal reaction rate are not always characterized by the highest possible values of the second-order rate constants which are related to the diffusion of the substrate and the product to the active site of the enzyme. Predictions are made concerning the ratios of maximal activities in optimal states as well as for the adaptation of the Michaelis constants to the concentrations of the outer reactants. Using metabolic control analysis it is shown that the solutions obtained for single enzymes may also be applied in multi-enzyme systems.  相似文献

The observed temperature dependence of the rate constant for reduction of (Chl a.2H2O)2+. by water is fitted to a theoretical expression for the rate of an intramolecular electronic radiationless transition. The theory assumes a single effective mediating mode and the best-fit value for the frequency of this mode is omega(m) = 680 cm(-1), which corresponds to high-frequency, intermolecular librations in water. From an explicit calculation of the nonadiabatic-coupling matrix element, an average electron-transfer distance of 11 A is obtained, consistent with molecular dimensions available for this system.  相似文献
