Conclusion Culture collections are key to the success of biotechnology companies. Protection of patent strains and manufacturing inoculum; standardization of biological materials for research, development and manufacturing; and documentation of organism transfers are essential functions provided by a culture collection in a biotechnology company. The need to recruit qualified and experienced employees is a major concern of many organizations engaged in biotechnology. Universities and industry are scoured to find the right people, but where are we to find promising new candidates for the most numerous, and arguably most talented section of the biotechnological workforce — microorganisms and other types of cells? The international network of culture collections offers the best answer, and a proper understanding of their roles and capabilities can save biotechnologists an immense amount of time and money.Among the millions of microbial, plant and animal cells stored in the world's culture collections it is often possible to find one that exactly suits a biotechnologist's particular requirements or one that can be developed to perform a specific task. Although the principles and the necessity for good laboratory practice (GLP) guidelines to confirm the credibility, integrity, and quality of non-clinical laboratory studies have been known for more than a decade, culture collection activities are not subject to them. Because of recent advances in biotechnology, culture collections face increased demands not only for quality cultures but also current information. When applied in culture collections, GLP guidelines prove to be an excellent management tool as well as a cost-effective system of providing authentic and reliable microbial and cell cultures and associated data.  相似文献

Collections of micro-organisms are a crucial element of life science research infrastructure but are vulnerable to loss and damage caused by natural or man-made disasters, the untimely death or retirement of personnel, or the loss of research funding. Preservation of biological collections has risen in priority due to a new appreciation for discoveries linked to preserved specimens, emerging hurdles to international collecting and decreased funding for new collecting. While many historic collections have been lost, several have been preserved, some with dramatic rescue stories. Rescued microbes have been used for discoveries in areas of health, biotechnology and basic life science. Suggestions for long-term planning for microbial stocks are listed, as well as inducements for long-term preservation.  相似文献

Arguably, the richest source of knowledge (as opposed to fact and data collections) about biology and biotechnology is captured in natural-language documents such as technical reports, conference proceedings and research articles. The automatic exploitation of this rich knowledge base for decision making, hypothesis management (generation and testing) and knowledge discovery constitutes a formidable challenge. Recently, a set of technologies collectively referred to as knowledge discovery in text (KDT) has been advocated as a promising approach to tackle this challenge. KDT comprises three main tasks: information retrieval, information extraction and text mining. These tasks are the focus of much recent scientific research and many algorithms have been developed and applied to documents and text in biology and biotechnology. This article introduces the basic concepts of KDT, provides an overview of some of these efforts in the field of bioscience and biotechnology, and presents a framework of commonly used techniques for evaluating KDT methods, tools and systems.  相似文献

The mission of the United States Culture Collection Network (USCCN; http://usccn.org) is “to facilitate the safe and responsible utilization of microbial resources for research, education, industry, medicine, and agriculture for the betterment of human kind.” Microbial culture collections are a key component of life science research, biotechnology, and emerging global biobased economies. Establishment of national biological resource centres (BRCs) is of special concern and requires harmonization of regulations on microorganisms’ handling, improvement of legal control pertaining to intellectual property right, access to genetic resources and fair benefit sharing arising from their biotechnology application. As exemplified by the Regional Specialized Collection of Alkanotrophic Microorganisms (acronym IEGM, World Federation for Culture Collections # 768, ) hosted at the Institute of Ecology and Genetics of Microorganisms, the role of specialized microbial collections is emphasized as the governing factors of innovative development of biotechnology and bioindustry. The publication aims at drawing attention to the regional BRC being formed in the Perm Krai which provides the appropriate information on the holdings and is responsible for screening, study and maintenance of valuable microbial gene pool to meet the needs of ecology, industry and biotechnology, and for developing novel methodological approaches to studying extremotolerant microorganisms. This centre also contributes to the development and application of advanced achievements in enzymatic transformation of carbon compounds, production of fodder using non-traditional raw material, oil- and gas-prospecting activities, monitoring and bioremediation of contaminated sites.  相似文献

The manipulation of DNA is routine practice in botanical research and has made a huge impact on plant breeding, biotechnology and biodiversity evaluation. DNA is easy to extract from most plant tissues and can be stored for long periods in DNA banks. Curation methods are well developed for other botanical resources such as herbaria, seed banks and botanic gardens, but procedures for the establishment and maintenance of DNA banks have not been well documented. This paper reviews the curation of DNA banks for the characterisation and utilisation of biodiversity and provides guidelines for DNA bank management. It surveys existing DNA banks and outlines their operation. It includes a review of plant DNA collection, preservation, isolation, storage, database management and exchange procedures. We stress that DNA banks require full integration with existing collections such as botanic gardens, herbaria and seed banks, and information retrieval systems that link such facilities, bioinformatic resources and other DNA banks. They also require efficient and well-regulated sample exchange procedures. Only with appropriate curation will maximum utilisation of DNA collections be achieved.  相似文献

In microalgal culture collections, as in many biological resource centres, cryoconservation is the most attractive method for the long-term, secure storage of living material. Nannochloropsis oculata, a marine unicellular alga, is of interest in the field of biotechnology due to its high lipid content. Of various cryoprotectants tested for their toxicity and for their ability to prevent cryoinjury, glycerol (final concentration 1.1 M) was the most efficient. When glycerol-treated cultures were submitted to a strictly regulated cooling rate (-3 degrees C min(-1)), they attained the control culture density within 13 d after thawing.  相似文献

Culture collections are valuable resources for the sustainable use of microbial diversity and its conservation. Advances in biotechnology have further increased their importance and some of these have been recognized as International Depositary Authority (IDA) for the deposition of patent cultures. Microbial Culture Collection at National Centre for Cell Science was established by the Department of Biotechnology, Government of India is country’s newest culture collection with largest holdings. It is recognized as an IDA under the Budapest Treaty and Designated National Repository under the Biodiversity Act 2002. This article describes its various service related activities.  相似文献

Environmental research delivers valuable bacterial resources for biotechnology. We believe that systematic long-term preservation of bacteria will promote future biotechnological innovations, by safeguarding the accessibility of bacteria already recognized to have interesting features and providing a “pool” of bacterial resources for novel applied research. To this end, we want to advocate the incorporation of preservation tests in environmental or applied microbiological research. This paper introduces non-specialists to different preservation methods for bacteria. Several parameters that influence long-term storage of bacterial resources are explained and practical tips and guidelines are formulated. Also, the vital role of public culture collections is highlighted and the state-of-the-art of preservation of non-pure cultures is described.  相似文献

Essential to the ambition of characterising fully the human proteome are systematic and comprehensive collections of specific affinity reagents directed against all human proteins, including splice variants and modifications. Although a large number of affinity reagents are available commercially, their quality is often questionable and only a fraction of the proteome is covered. In order for more targets to be examined, there is a need for broad availability of panels of affinity reagents, including binders against proteins of unknown functions. The most familiar affinity reagents are antibodies and their fragments, but engineered forms of protein scaffolds and nucleic acid aptamers with similar diversity and binding properties are becoming viable alternatives. Recent initiatives in Europe and the USA have been established to improve both the availability and quality of reagents for affinity proteomics, with the ultimate aim of creating standardised collections of well-validated binding molecules for proteome analysis. As well as coordinating affinity reagent production through existing resources and technology providers, these projects aim to benchmark key molecular entities, tools, and applications, and establish the bioinformatics framework and databases needed. The benefits of such reagent resources will be seen in basic research, medicine and the biotechnology and pharmaceutical industries.  相似文献

Development of the theory and practice of building and maintenance of collections of reference cultures of pathogens strains is actual in conservation of a biodiversity, and as for maintenance with standardized superfine starting materials fundamental and applied researches in fields of microbiology, an immunology, biotechnology, ecology, and biosafety. Various approaches to taxonomy of viruses, and also definition of concepts "strain" and "species" are discussed in the article. Formulations of the concepts most conforming to museum collections activity are given. The methodology of collecting pathogens, including fifteen-year experience of maintenance of the National collection of hemorrhagic fever viruses belonging to group I of pathogenicity, is summarized. The systemic approach is suggested as a basis of methodology of collecting. Selection of pathogen species and their specific strains according to purpose and on the basis of complex study of their biological and other characteristics, as well as standardization and unification of methods of pathogens maintenance and preparing on their basis working materials, diagnostic kits and other preparations, create a real basis of optimization, unification and continuity of the subsequent applied researches. The offer about formation at the state level of a new independent direction--pathogens collecting with establishment of federal centers based on pathogens' classes has been made.  相似文献

Contemporary collections frequently contain man-made materials. Although synthetic materials are considered more resistant to chemical, physical and biological damage than natural materials, they can also undergo rapid deterioration. In this Opinion article, we claim that biotechnology can help to identify biodeteriogens and prevent colonisation of polymeric surfaces through the application of biological products that reduce cell adhesion. We report the study of 'Futuro', made in 1965 by the Finnish architect Matti Suuronne. This ski-cabin, constructed of glassfibre-reinforced polyester, polyester-polyurethane, and poly(methylmethacrylate), was significantly degraded by conspicuous growth of microorganisms, identified as Cyanobacteria and Archaea using fluorescent in situ hybridisation. Ultimately, if biodeteriogens are able to adhere to the polymer surfaces, molecules with enzymatic activity can help to prevent the formation of biofilms--a main cause of deterioration--and aid the work of the conservator.  相似文献
