MicroRNA调控固有免疫应答的分子机制   总被引:2,自引:0,他引:2  
MicroRNA(miRNA)是近几年继siRNA之后非编码RNA研究的又一热点.它通过与靶mRNA的特异性结合,在转录后水平上对基因表达进行调控.研究表明,miRNA可能参与脊椎动物固有免疫应答的多个环节.在病原微生物感染时,它们不仅成为重要的固有免疫受体活化后的信号调节分子,而且能够直接干扰病毒复制而发挥抗病毒效应.miRNA可能与经典的固有免疫应答体系共同组成机体抵御病原微生物入侵的“第一道防线”.同时,病原微生物,特别是病毒还可以通过自己编码miRNA或者改变宿主细胞miRNA表达谱直接或间接地干扰很多宿主免疫相关基因的表达,实现逃逸机体免疫清除的目的.因此,miRNA水平的相瓦作用可能是病原微生物与其宿丰展开免疫“博弈”的重要战场.  相似文献

microRNAs (miRNAs) are small (approximately 22 nucleotide) regulatory RNAs which play fundamental roles in many biological processes. Recent studies have shown that the expression of many miRNAs is altered in various human tumors and some miRNAs may function as oncogenes or tumor suppressor genes. However, with the exception of glioblastoma multiforme, the expression of miRNAs in brain tumors is unknown. Furthermore, methods to profile miRNAs from formalin-fixed, paraffin-embedded (FFPE) archival tissues or to study their cellular and subcellular localization in FFPE tissues have been lacking. Here we report the coordinated miRNA expression analysis from the tissue level to the subcellular level, using the RAKE (RNA-primed, array-based, Klenow Enzyme) miRNA microarray platform in conjunction with Locked Nucleic Acid (LNA)-based in situ hybridization (LNA-ISH) on archival FFPE human brains and oligodendroglial tumors. The ability to profile miRNAs from archival tissues at the tissue level, by RAKE microarrays, and at the cellular level by LNA-ISH, will accelerate studies of miRNAs in human diseases.  相似文献

Kaposi''s sarcoma (KS) is caused by infection with Kaposi''s sarcoma-associated herpesvirus (KSHV). The virus expresses unique microRNAs (miRNAs), but the targets and functions of these miRNAs are not completely understood. In order to identify human targets of viral miRNAs, we measured protein expression changes caused by multiple KSHV miRNAs using pulsed stable labeling with amino acids in cell culture (pSILAC) in primary endothelial cells. This led to the identification of multiple human genes that are repressed at the protein level, but not at the miRNA level. Further analysis also identified that KSHV miRNAs can modulate activity or expression of upstream regulatory factors, resulting in suppressed activation of a protein involved in leukocyte recruitment (ICAM1) following lysophosphatidic acid treatment, as well as up-regulation of a pro-angiogenic protein (HIF1α), and up-regulation of a protein involved in stimulating angiogenesis (HMOX1). This study aids in our understanding of miRNA mechanisms of repression and miRNA contributions to viral pathogenesis.  相似文献

MicroRNAs (miRNAs) are endogenous, small non-coding RNAs (20-22 nucleotides) that negatively regulate gene expression at the translational level by base pairing to the 3' untranslated region of target messenger RNAs. More than 400 miRNAs have been identified in humans and are evolutionally conserved from plants to animals. It has been revealed that miRNAs regulate various biological processes, such as development, cell differentiation, cell proliferation, and cell death. It is predicted that 30% of protein-encoding genes are regulated by miRNAs. Inappropriate expression of miRNAs has been found in cancer. Especially, the expression level of miRNAs that act like anti-oncogenes is frequently reduced in cancers because of chromosome aberrations. In addition, since the processing of miRNAs has been characterized to be enzymatic in nature, the expression levels of miRNAs are closely associated with the activity and levels of such enzymes. In this review, we discuss recent remarkable advances in miRNA biogenesis, bio-networking involving miRNAs, and their roles in carcinogenesis. Further, we discuss the expression of miRNA-143 and -145 in colon cancer and their roles in carcinogenesis. The available data suggest that miRNAs would be potentially useful as diagnostic and therapeutic tools.  相似文献

microRNAs (miRNAs) are critical to heart development and disease. Emerging research indicates that regulated precursor processing can give rise to an unexpected diversity of miRNA variants. We subjected small RNA from murine HL-1 cardiomyocyte cells to next generation sequencing to investigate the relevance of such diversity to cardiac biology. ~40 million tags were mapped to known miRNA hairpin sequences as deposited in miRBase version 16, calling 403 generic miRNAs as appreciably expressed. Hairpin arm bias broadly agreed with miRBase annotation, although 44 miR* were unexpectedly abundant (>20% of tags); conversely, 33 -5p/-3p annotated hairpins were asymmetrically expressed. Overall, variability was infrequent at the 5' start but common at the 3' end of miRNAs (5.2% and 52.3% of tags, respectively). Nevertheless, 105 miRNAs showed marked 5' isomiR expression (>20% of tags). Among these was miR-133a, a miRNA with important cardiac functions, and we demonstrated differential mRNA targeting by two of its prevalent 5' isomiRs. Analyses of miRNA termini and base-pairing patterns around Drosha and Dicer cleavage regions confirmed the known bias towards uridine at the 5' most position of miRNAs, as well as supporting the thermodynamic asymmetry rule for miRNA strand selection and a role for local structural distortions in fine tuning miRNA processing. We further recorded appreciable expression of 5 novel miR*, 38 extreme variants and 8 antisense miRNAs. Analysis of genome-mapped tags revealed 147 novel candidate miRNAs. In summary, we revealed pronounced sequence diversity among cardiomyocyte miRNAs, knowledge of which will underpin future research into the mechanisms involved in miRNA biogenesis and, importantly, cardiac function, disease and therapy.  相似文献
