Study of the structural organization of gustatory apparatus in rainbow trout Parasalmo mykiss performed using electron scanning microscopy demonstrated that external taste buds are absent in this species in skin covers of the head and in the circumoral region. In the oropharyngeal cavity (oral and gill cavities and pharynx) of the rainbow trout, a well-developed gustatory receptor apparatus was found. In correspondence with specific features of morphology and anatomy of the skull, taste buds form seven gustatory zones. Morphometric analysis demonstrated differences between gustatory zones in the pattern and density of distribution of taste buds, as well as in average sizes of their sensory field. Zones of similar innervation have many common features in morphology. Morphologically similar zones form three regions in the oropharyngeal cavity: rostral, central, and caudal. A tendency for a decrease in the concentration of taste buds in the rostrocaudal direction common for all sensory zones was revealed. The highest concentration of taste buds was recorded at papillae of rostral regions near big teeth. A typical feature of taste buds in rainbow trout is irregular shape of the taste pore. Analysis of ultrastructural specific features of apical processes of taste cells allows us to distinguish five cell shapes in the composition of taste buds. The numeric ratio of cell shapes varies in buds of different localization. The quantitative distribution of taste buds over sensory zones, specific features of morphology and sizes of their sensory field are discussed in relation to the feeding pattern of the species.  相似文献

Investigation of the structure of the gustatory apparatus and morphometric features of the brain in two age groups (1.5 and 6 months) of Siberian sturgeon Acipenser baerii shows a well-developed gustatory system in all studied exemplars. In the oral cavity of the sturgeon, 14 sensory zones are separated, and eight zones are supplied by gustatory receptors. During the growth of the juveniles, morphometric parameters of the zones can change. The zones of the dorsal and ventral linings of the oral cavity are similar by morphology and size, and they are complementary. The largest zone of the oral cavity of the sturgeon with a complex structure is the palate organ. The taste buds of the sturgeon are characterized by a large variation in shapes of apical protrusions of receptor cells. Weight indices of the brain parts and estimated sensory coefficients show an increasing degree of development of the gustatory and olfactory systems during growth of the juveniles. Two age groups of the juveniles are characterized by similar morphometric parameters of the gustatory receptor apparatus with external localization, but the morphology of the gustatory apparatus of the oral cavity is different. The features of the gustatory system morphology are discussed in connection with biology of the species.  相似文献

To confirm the existence in fish of the olfactogustatory interactions of adaptive significance, the study was performed of reactions of the receptor part of the carp gustatory system to the complete olfactory deafferentation with use of the method of scanning electron microscopy. Peculiarities of morphological changes of the taste receptor apparatus of the external and oral localization were revealed, and the dynamic of development of the taste receptor adaptive changes was traced for 1–9 months after production of anosmia. Receptors of the intraoral gustatory system had reactions of low statistical significance which were delayed in time by 5–6 months. The receptor apparatus of the external gustatory system receptors of the intraoral taste system showed pronounced hypertrophic changes that appeared since 2–3 months after the beginning of anosmia. The changes consisted in an increase of the area of the taste bud (TB) sensory field and of their number in the sensory zones of the maxillary and mandibular parts. The taste receptors located in the maxillary barbels and in the mandible gular zone had the most expressed reactions to the olfactory deafferentation. In the fish with anosmia, formation of the additional structures of the “external taste” was detected in the form of epidermal processes under the lower lip, which were covered with taste papillae. Thus, it has been shown that the fish olfactory and gustatory systems are functionally interrelated; under conditions of olfactory deprivation the external gustatory system is able to undergo compensatory morphofunctional changes aimed at vicariation of the lost distant chemical reception.  相似文献

Neurotrophins, neurotrophin receptors and sensory neurons are required for the development of lingual sense organs. For example, neurotrophin 3 sustains lingual somatosensory neurons. In the traditional view, sensory axons will terminate where neurotrophin expression is most pronounced. Yet, lingual somatosensory axons characteristically terminate in each filiform papilla and in each somatosensory prominence within a cluster of cells expressing the p75 neurotrophin receptor (p75NTR), rather than terminating among the adjacent cells that secrete neurotrophin 3. The p75NTR on special specialized clusters of epithelial cells may promote axonal arborization in vivo since its over-expression by fibroblasts enhances neurite outgrowth from overlying somatosensory neurons in vitro. Two classical observations have implicated gustatory neurons in the development and maintenance of mammalian taste buds—the early arrival times of embryonic innervation and the loss of taste buds after their denervation in adults. In the modern era more than a dozen experimental studies have used early denervation or neurotrophin gene mutations to evaluate mammalian gustatory organ development. Necessary for taste organ development, brain-derived neurotrophic factor sustains developing gustatory neurons. The cardinal conclusion is readily summarized: taste buds in the palate and tongue are induced by innervation. Taste buds are unstable: the death and birth of taste receptor cells relentlessly remodels synaptic connections. As receptor cells turn over, the sensory code for taste quality is probably stabilized by selective synapse formation between each type of gustatory axon and its matching taste receptor cell. We anticipate important new discoveries of molecular interactions among the epithelium, the underlying mesenchyme and gustatory innervation that build the gustatory papillae, their specialized epithelial cells, and the resulting taste buds.  相似文献

The Mexican tetra, Astyanax mexicanus, exists as two morphs of a single species, a sighted surface morph and a blind cavefish. In addition to eye regression, cavefish have an increased number of taste buds, maxillary teeth and have an altered craniofacial skeleton compared to the sighted morph. We investigated the effect the lens has on the development of the surrounding skeleton, by ablating the lens at different time points during ontogeny. This unique long-term study sheds light on how early embryonic manipulations on the eye can affect the shape of the adult skull more than a year later, and the developmental window during which time these effects occur. The effects of lens ablation were analyzed by whole-mount bone staining, immunohistochemisty and landmark based morphometric analyzes. Our results indicate that lens ablation has the greatest impact on the skeleton when it is ablated at one day post fertilisation (dpf) compared to at four dpf. Morphometric analyzes indicate that there is a statistically significant difference in the shape of the supraorbital bone and suborbital bones four through six. These bones expand into the eye orbit exhibiting plasticity in their shape. Interestingly, the number of caudal teeth on the lower jaw is also affected by lens ablation. In contrast, the shape of the calvariae, the length of the mandible, and the number of mandibular taste buds are unaltered by lens removal. We demonstrate the plasticity of some craniofacial elements and the stability of others in the skull. Furthermore, this study highlights interactions present between sensory systems during early development and sheds light on the cavefish phenotype.  相似文献

The developing taste buds of vallate papillae were studied with electron microscope in rats during the first 7 days after birth. Two types of cells--light and dark--are identified in the taste buds of a one day old animal. The apical parts of dark cells are characterized by numerous dark granules. A distinguishing feature of light cells is the presence of synaptic contacts with afferent intragemmal nerves. On the 4th day of development on the top of the apical parts of the cell, a microvillar apparatus is seen to form, which does not yet communicate with the oral cavity. On the 7th day, basal cells appear in the taste buds. Some of these cells are seen mitotically dividing. The differentiated microvillar apparatus now communicates with oral cavity. The structure of the taste buds is getting similar to that in the adults. The structural and functional peculiarities of the developing taste buds are discussed in association with the period of ontogenesis under consideration.  相似文献

In almost all mammals a well developed, paired and blind ending vomeronasal Organ (VNO) situated within the basement of the nasal septum, communicates with the oral cavity. This contact is established by two nasopalatine ducts, which penetrate the rostral palate close to the incisors. These ducts open orally into the sulcus which moulds the palatine papilla. In several mammals taste buds were found in the epithelium of the patatine papilla located within the nasopalatine ducts or close to their oral openings. Presumably these taste buds interact with the vomeronasal olfaction. It is likely that they are leading to a chemosensory sensation comparable to the combination of normal taste and smell. As not all mammals with a functionable VNO possess taste buds in this position, an inspection of the rostral part of the tongue which touches the palatine papilla presented an interesting situation concerning the distribution of taste buds. This region of the tongue is almost completely free of taste buds in species like Tupaia glis and Didelphis marsupialis virginiana, which have taste buds in the epithelium of their palatine papilla. In Lemur catta however, where the palatine papilla is lacking taste buds, the respective tongue part is densely covered with them. In this case it appears likely that they in a way of substitution functionally are connected with vomeronasal olfaction.  相似文献

The Mexican tetra Astyanax mexicanus has many of the favorable attributes that have made the zebrafish a model system in developmental biology. The existence of eyed surface (surface fish) and blind cave (cavefish) dwelling forms in Astyanax also provides an attractive system for studying the evolution of developmental mechanisms. The polarity of evolutionary changes and the environmental conditions leading to the cavefish phenotype are known with certainty, and several different cavefish populations have evolved constructive and regressive changes independently. The constructive changes include enhancement of the feeding apparatus (jaws, taste buds, and teeth) and the mechanosensory system of cranial neuromasts. The homeobox gene Prox 1, which is expressed in the expanded taste buds and cranial neuromasts, is one of the genes involved in the constructive changes in sensory organ development. The regressive changes include loss of pigmentation and eye degeneration. Although adult cavefish lack functional eyes, small eye primordia are formed during embryogenesis, which later arrest in development, degenerate, and sink into the orbit. Apoptosis and lens signaling to other eye parts, such as the cornea, iris, and retina, result in the arrest of eye development and ultimate optic degeneration. Accordingly, an eye with restored cornea, iris, and retinal photoreceptor cells is formed when a surface fish lens is transplanted into a cavefish optic cup, indicating that cavefish optic tissues have conserved the ability to respond to lens signaling. Genetic analysis indicates that multiple genes regulate eye degeneration, and molecular studies suggest that Pax6 may be one of the genes controlling cavefish eye degeneration. Further studies of the Astyanax system will contribute to our understanding of the evolution of developmental mechanisms in vertebrates.  相似文献

Analysis of taste buds (TB) on the lips and oropharyngeal cavity in several species of gobies (Gobiidae) and blennies (Blenniidae) from the Red Sea, Mediterranean Sea and Seychelles, revealed three types of these organs: types I and II, which protrude above the surrounding epithelium on lobules of various forms, and type III, which terminate on the level of the epithelium. These TB are composed of either light or dark sensory cells with apical microvillar extensions, and of basal cells situated at the TB base. Synaptic junctions occur between the TB cells and the sub‐epithelial sensory nerves. Numerical distribution and morphology of TB on the lips and in the oral cavity of the species studied revealed patterns that are specific on both species and family levels. In most of the gobies the lips, jaws and oral breathing valves are usually covered by numerous lobules, each of which bears papillae with two to seven type I and type II TB, reaching a total of up to 7500 buds on the lips and in the oropharyngeal cavity in these fishes. The number of TB increases with growth (age) of the fish, and the combined and total sensory area of TB in an adult fish can reach up to 80 000 μm2. In contrast, in blennies the anterior region of the oral cavity is seldom lobulated, with far fewer TB; the majority of TB are found in the more posterior region. It is postulated that these differences in TB density and location between gobies and blennies are connected to differences in foraging strategies and diet, and may represent ecomorphological adaptations.  相似文献

Food searching behavior of the Buenos Aires tetra Hyphessobrycon anisitsi has been investigated before and after experimentally induced anosmia combined with visual deprivation. The threshold concentration of a water extract of familiar food (chironomid larvae) for fish with intact olfaction was 10–6 g/L. Induced anosmia led to complete loss of the search reaction to food odor. The chemosensory behavioral response to the water extract of food used at high concentrations (10-1-10-2 g/L) was not restored in fish after 1.5, 5, or 13.5 months of anosmia. The obtained results show that the external sense of taste is absent or extremely poorly developed in the Buenos Aires tetra.  相似文献

Similarity between the taste preferences of classical taste substances and free L-amino acids in adult three-spined stickleback Gasterosteus aculeatus (forma leiurus) and nine-spined stickleback Pungitius pungitius (forma laevis) was found. The strongest and the most significant responses in both species were evoked by citric acid and cysteine, asparatic, and glutamic acids. Sodium chloride, calcium chloride, sucrose, and the remaining 18 amino acids do not elicit a statistically significant effect on the consumption of agar-agar pellets by the fish or have weak taste attractiveness. Similarity of taste preferences in the three-spined and nine-spined sticklebacks are supported by correlation analysis. Absolute values of the consumption of pellets of the same types are also similar in the two species. There were, however, differences in the behavioral taste response, repeated snaps, and the duration of processing of the pellets. The taste response of the nine-spined stickleback is more similar to taste responses in fish of the limnophilic complex than to the response in the three-spined stickleback. It is hypothesized that taste spectra may be very similar in fish with similar ecology and feeding patterns not only in sticklebacks, but also in other related species.  相似文献

The present study compares dental morphology and taste bud distribution in the oral cavity of four species of the teleost family Kyphosidae, five species of Girellidae and one species of Sparidae. Some of these species are predominantly herbivorous, while others are omnivorous with a high portion of invertebrates in their diets. All the kyphosids feature one row of frontal cusped teeth on their jaws and areas of miniature teeth on the tongue and palate; the girellid species feature 2–5 rows of denticulate teeth, and no teeth inside the oral cavity. The total number of taste buds in the oral cavity is higher in Girella spp. than in the kyphosids, and their number is species-specific, not correlated with fish size. For example, Kyphosus bigibbus of 500 mm standard length bears 1780 taste buds, while the kyphosid Neoscorpis lithophilus of 80 mm bears 3460. The maximum number of taste buds, 7900, is found in Girella punctata of 60 mm standard length, and the minimum number, 1320, in Kyphosus vaigiensis of 175 mm. The higher number of taste buds appears to characterize species that possess a more diversified diet. The present study contributes to our understanding of the differences between the two families Kyphosidae and Girellidae. In particular, it relates the eco-morphological adaptations to the type of diet consumed by the various species.  相似文献

Background

How and why animals lose eyesight during adaptation to the dark and food-limited cave environment has puzzled biologists since the time of Darwin. More recently, several different adaptive hypotheses have been proposed to explain eye degeneration based on studies in the teleost Astyanax mexicanus, which consists of blind cave-dwelling (cavefish) and sighted surface-dwelling (surface fish) forms. One of these hypotheses is that eye regression is the result of indirect selection for constructive characters that are negatively linked to eye development through the pleiotropic effects of Sonic Hedgehog (SHH) signaling. However, subsequent genetic analyses suggested that other mechanisms also contribute to eye regression in Astyanax cavefish. Here, we introduce a new approach to this problem by investigating the phenotypic and genetic relationships between a suite of non-visual constructive traits and eye regression.

Results

Using quantitative genetic analysis of crosses between surface fish, the Pachón cavefish population and their hybrid progeny, we show that the adaptive vibration attraction behavior (VAB) and its sensory receptors, superficial neuromasts (SN) specifically found within the cavefish eye orbit (EO), are genetically correlated with reduced eye size. The quantitative trait loci (QTL) for these three traits form two clusters of congruent or overlapping QTL on Astyanax linkage groups (LG) 2 and 17, but not at the shh locus on LG 13. Ablation of EO SN in cavefish demonstrated a major role for these sensory receptors in VAB expression. Furthermore, experimental induction of eye regression in surface fish via shh overexpression showed that the absence of eyes was insufficient to promote the appearance of VAB or EO SN.

Conclusions

We conclude that natural selection for the enhancement of VAB and EO SN indirectly promotes eye regression in the Pachón cavefish population through an antagonistic relationship involving genetic linkage or pleiotropy among the genetic factors underlying these traits. This study demonstrates a trade-off between the evolution of a non-visual sensory system and eye regression during the adaptive evolution of Astyanax to the cave environment.

SUMMARY Mexican tetra (Astyanax mexicanus) exist as two morphs: a sighted (surface) form and a blind (cavefish) form. In the cavefish, some modules are lost, such as the eye and pigment modules, whereas others are expanded, such as the taste bud and cranial neuromast modules. We suggest that modularity can be viewed as being nested in a manner similar to Baupläne so that modules express unique sets of genes, cells, and processes. In terms of evolution, we conclude that natural selection can act on any of these hierarchical levels within modules or on all the sensory modules as a whole. We discuss interactions within and between modules with reference to the blind cavefish from both genetic and developmental perspectives. The cavefish represents an illuminating example of module interaction, uncoupling of modules, and module expansion.  相似文献

The purpose of the current study was to mask the taste of cetirizine HCl and to incorporate the granules produced in oral disintegrating tablets (ODT). The bitter, active substance was coated by fluidized bed coating using Eudragit® RL30-D at levels between 15% and 40% w/w. The ODTs were developed by varying the ratio of superdisintegrants such as sodium croscarmellose, crospovidone grades and low substituted hydroxypropyl cellulose (L-HPC). A direct compression process was used to compress the ODTs under various compaction forces to optimize tablet robustness. The properties of the compressed tablets including porosity, hardness, friability and dissolution profiles were further investigated. The in vitro and in vivo evaluation of the tablet disintegration times showed almost identical rapid disintegration below 10 s at the optimal levels of each superdisintegrant. Finally, the taste and sensory evaluation in human volunteers demonstrated excellence in masking the bitter active and tablet palatability.  相似文献

Epidermal sensory structures of adults and juveniles of amphihaline migratory fish hilsa Tenualosa ilisha were studied from two habitats, i.e., freshwater (FW) and marine water (MW). Every year, adults and sexually mature hilsa migrate upstream from marine habitat to riverine freshwater habitat for breeding. This report provides evidences of chemoreception on their upstream migration through several characteristic features on their body, especially on the head and oral cavity. Scanning electron microscopy (SEM) reveals that freshwater adult hilsa (FH) has abundant solitary chemosensory cells (SCCs) on the snout epidermis (around the openings of the epidermal pit) and upper lip, whereas marine water adult hilsa (MH) moderately possesses such sensory structures. The juveniles returning to marine water completely lack SCCs. Immunohistochemical studies revealed the expression of PLC β2 on the snout of FH and tongue of both FH and MH. Further analysis (immunofluorescence, immunoblot and densitometry) of the epidermis confirms the presence of chemosensory structures through strong expression and localization of G-proteins (Gαq and Gα s/olf) from the snout as well as tongue in freshwater hilsa. The SEM also confirms the presence of two types of taste buds in FH, viz. type I (TB I) and type III (TB III). Whereas TB I and TB III are observed on the upper palatine and lips, most of the TB III are located on the tongue region of freshwater and marine hilsa. The juvenile hilsa are devoid of such structures. The presence of dense and rich SCCs and taste sensory cells in adults could be a characteristic feature for strong sensory reception to recognize odour and food-related environmental cues from habitats where they often migrate.  相似文献

Espins are multifunctional actin-bundling proteins that are highly enriched in the microvilli of certain chemosensory and mechanosensory cells, where they are believed to regulate the integrity and/or dimensions of the parallel-actin-bundle cytoskeletal scaffold. We have determined that, in rats and mice, affinity purified espin antibody intensely labels the lingual and palatal taste buds of the oral cavity and taste buds in the pharyngo-laryngeal region. Intense immunolabeling was observed in the apical, microvillar region of taste buds, while the level of cytoplasmic labeling in taste bud cells was considerably lower. Taste buds contain tightly packed collections of sensory cells (light, or type II plus type III) and supporting cells (dark, or type I), which can be distinguished by microscopic features and cell type-specific markers. On the basis of results obtained using an antigen-retrieval method in conjunction with double immunofluorescence for espin and sensory taste cell-specific markers, we propose that espins are expressed predominantly in the sensory cells of taste buds. In confocal images of rat circumvallate taste buds, we counted 21.5 ± 0.3 espin-positive cells/taste bud, in agreement with a previous report showing 20.7 ± 1.3 light cells/taste bud when counted at the ultrastructural level. The espin antibody labeled spindle-shaped cells with round nuclei and showed 100% colocalization with cell-specific markers recognizing all type II [inositol 1,4,5-trisphosphate receptor type III (IP3R3), α-gustducin, protein-specific gene product 9.5 (PGP9.5)] and a subpopulation of type III (IP3R3, PGP9.5) taste cells. On average, 72%, 50%, and 32% of the espin-positive taste cells were labeled with antibodies to IP3R3, α-gustducin, and PGP9.5, respectively. Upon sectional analysis, the taste buds of rat circumvallate papillae commonly revealed a multi-tiered, espin-positive apical cytoskeletal apparatus. One espin-positive zone, a collection of ~3 μm-long microvilli occupying the taste pore, was separated by an espin-depleted zone from a second espin-positive zone situated lower within the taste pit. This latter zone included espin-positive rod-like structures that occasionally extended basally to a depth of 10–12 μm into the cytoplasm of taste cells. We propose that the espin-positive zone in the taste pit coincides with actin bundles in association with the microvilli of type II taste cells, whereas the espin-positive microvilli in the taste pore are the single microvilli of type III taste cells.  相似文献

Generally, pellets obtained from extrusion/spheronization, containing microcrystalline cellulose (MCC), do not disintegrate. An attempt has been made to develop melt-in-mouth pellets of taste-masked atomoxetine hydrochloride, using extrusion-spheronization, for pediatric patients. Melt-in-mouth pellets were prepared using extrusion-spheronization method and optimized using 33 FFD. MCC (X1, %), mannitol (X2, %) and Indion 414: Pharmaburst 500 ratio (X3, ratio) were the factors (independent variables) studied, whereas responses studied (dependent variables) were friability (Y1, %), yield (Y2, %) shape (Y3, roundness) in vitro disintegration time (Y4, seconds). The optimized formulation obtained from FFD was characterized for friability, shape and morphology, in vitro disintegration time, porosity, moisture uptake, in vitro release study and in vivo taste and disintegration time in healthy human volunteers. Randomized, two-treatment, two-sequence, two-period, single dose, crossover sensory evaluation study of taste-masked melt-in-mouth pellet was carried out in 10 healthy human subjects. A statistically significant polynomial mathematical relationship was generated between the factors and responses to obtain an optimized formulation. The optimized formulation was characterized (in vitro and in vivo) and exhibited a rapid drug release in vitro attributed to fast disintegration of pellets and high solubility of drug in 0.1 N HCl and buffer (pH 6.8). In vivo, 40% of volunteers ranked taste-masked optimized formulation as slightly bitter while 60% ranked it as no taste. The optimized pellets were conveniently administered in volunteers and exhibited rapid in-vivo disintegration in the oral cavity. Melt-in-mouth pellets can be a used as a platform technology for administering drugs to paediatric patients accurately and conveniently resulting in patient compliance.  相似文献
