L-Tyrosine carboxy-lyase (E. C. 4. 1. 1. 25) was extracted fromthe roots of barley seedlings and purified approximately 25fold. Optimum pH for the enzyme activity was found to be 7.3.The Km value for L-tyrosine was calulated as 4.5?10–4M.D-Isomer did not react with the enzyme. L-DOPA, m-tyrosine ando-tyrosine were decarboxylated to some extent. Pyridoxal phosphateactivated the enzyme 4 fold. Caffeic acid and p-coumaric acidare competitive inhibitors. Ki values were 4.5?10–5M forcaffeic acid and 1.6?10–4M for p-coumaric acid. L-DOPAand m-tyrosine had an inhibitory effect on the decarboxylationof L-tyrosine. Hydroxylamine, semicarbazide, p-CMB, Fe++, Cu++,and Hg++ inhibited the decarboxylation of tyrosine. Enzyme activitywas also found in extracts from Triticum aestivum, Zea maysand Cytisus scoparius. Various plant and environmental factors influence the hydraulicproperties for roots, which were examined using negative hydrostaticpressures applied to the proximal ends of individual excisedroots of a common succulent perennial from the Sonoran Desert,Agave deserti Engelm. The root hydraulic conductivity, Lp, increasedsubstantially with temperature, the approximately 4-fold increasefrom 0.5°C to 40°C representing a Q10 of 1.45. Suchvariations in Lp with temperature must be taken into accountwhen modelling water uptake, as soil temperatures in the rootzone of such a shallow-rooted species vary substantially bothdaily and seasonally. At 20°C, Lp was 2.3 x 10–7 ms{macron}1MPa{macron}1for 3-week-old roots, decreasing to abouthalf this value at 10 weeks and then becoming approximatelyhalved again at 6 months. For a given root age, Lp for rainroots that are induced by watering as lateral branches on theestablished roots (which arise from the stem base) was aboutthe same as Lp for established roots. Hence, the conventionalbelief that rain roots have a higher Lp than do establishedroots is more a reflection of root age, as the rain roots tendto be shed following drought and thus on average are much youngerthan are established roots. Unlike previous measurements onroot respiration, lowering the gas-phase oxygen concentrationfrom 21% to 0% or raising the carbon dioxide concentration from0.1% to 2% had no detectable effect on Lp for rain roots andestablished roots. Lp for rain roots and established roots wasdecreased by an average of 11% and 35% by lowering the soilwater potential from wet conditions (soil=0 kPa) to {macron}40kPa and {macron}80 kPa, respectively. Such decreases in Lp mayreflect reduced water contact between soil particles and theroot surface and should be taken into account when predictingwater uptake by A. deserti. High activity of phosphoenolpyruvate (PEP)-carboxykinase, orADP: oxalacetate (OAA) carboxy-lyase activity (a kind of EC4. 1. 1. 32) was discovered in enzyme extracts or partiallypurified preparations obtained from the brown algae, Eiseniabicyclis, Dictyota dichotoma, Spatoglossum pacificum; and Hizikiafusiformis. Enzyme activities were determined by measuring theradioactivity incorporated in the products of dark 14CO2-fixationand by spectrophotometric determinations. Except for the lowactivity of "malic enzyme" (EC 1. 1. 1.40), no activities ofother carboxylases, i.e. PEP-carboxylase, PEP-carboxytransphosphorylase,and pyruvate carboxylase could be detected in algal extractsprepared under various conditions. Malate dehydrogenase (EC1. 1. 1. 37), fumarase (EC 4. 2. 1. 2), and glutamic: oxalacetictransaminase (EC 2. 6. 1. 1) were also detected. The algal PEP-carboxykinase required ADP and Mn2+ for maximumactivity in the carboxylation reaction; and ATP and Mn2+, butnot GTP, for maximum activity in both the decarboxylation andOAA-14CO2-exchange reactions. The optimum pH of purified PEP-carboxykinase was in the regionof 7.0 to 7.3 in both the carboxylation and decarboxylationreactions, and its Km values for HCO3, PEP, and ADP were10 mM, 0.3 mM, and 0.07 mM, respectively, in the carboxylationreaction, and values for OAA and ATP were 0.05 mM and 0.4 mM,respectively, in the decarboxylation reaction. Furthermore,the decarboxylation reaction was markedly inhibited by 20 mMHCO3. The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 10–7m s–1 MPa–1 for F. acanthodes and 5.2 ? 10–7m s–1 MPa–1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (1–3 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of –0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from–0.016 to –1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Previous papers have shown that abscisic acid can inhibit transportof ions across the root to the xylem vessels, resulting in reducedexudation from excised roots or inhibiting guttation from intactplants. However, it has not been established whether the inhibitionwas due to a reduction in salt transport (Js) or in permeabilityof the roots to water (Lp). This paper investigates the effectof ABA on Lp and Js separately. It is shown that Lp increasedin ABA and then fell, but was about the same as in control rootswhen transport was inhibited. The effect of ABA on exudationtherefore appeared to be mainly due to reduction in Js. Inhibitionof Js was also present in intact, transpiring plants and sowas not due to reduced water flow. The inhibition of ion releaseto the xylem affected Na+, Mg2+, Ca2+, and phosphate as wellas the major ion in the exudate, K+. It is concluded that ABAinhibits salt transport to the shoot by acting on ion transportinto the xylem, and not by reducing water flow coupled withsalt transport.  相似文献

Regulation of Pyruvate Decarboxylase In Vitro and In Vivo   总被引:2,自引:0,他引:2  
Results presented in this paper strongly support the view thatregulation of the key enzyme of alcoholic fermentation, pyruvatedecarboxylase (PDC), is achieved in a number of ways, all associatedwith possible lowering of the cytoplasmic pH during anoxia.These mechanisms include not only the well-known acid pH optimumof PDC, but also long-term, reversible changes in characteristicsof the enzyme established both in vitro and in vivo. Following transfer of desalted extracts from pH 6.0 to 7.4,maximal activity of PDC was decreased, while there was a considerableincrease in the lag before maximal activity was reached. Similarchanges in enzyme characteristics were observed when wheat (Triticumaestivum L. cv. Gamenya) roots and rice (Oryza sativa L. cv.Calrose) coleoptiles were transferred from anoxic to aerobicsolutions, provided PDC was assayed within 10 min of the startof maceration. All of the above changes were usually readilyreversible when extracts were returned to pH 6.0, or when plantswere returned to anoxic solutions. Additional regulation of PDC would be achieved by the S0.5 forpyruvate which is 0.75 mol m–3 at pH 6.0, 1.0 mol m–3at pH 6.8, and 2.5 mol m–3 at pH 7.4; the latter is wellabove estimates for pyruvate concentrations in the cytoplasmof aerated tissues. We assess that the combined effects of the acid pH optimum,the high S0.5 at pH 7.4 and the long-term decreases in activityobserved during incubation at pH 7.4 would reduce PDC activityin aerobic cells to at most 7% of the activity in anoxic cells.Possible additional controls for the pathway of alcoholic fermentationare briefly considered. A morphologically explicit numerical model for analysing wateruptake by individual roots was developed based on a conductornetwork, with specific conductors representing axial or radialconductivities for discrete root segments. Hydraulic conductivity(Lp; m s–1 MPa–1) was measured for roots of Agavedeserti Engelm. and Opuntia ficus-indica (L.) Miller by applyinga partial vacuum to the proximal ends of excised roots in solution.Lp was also measured for 40- to 80-mm segments along a root,followed by measurements of axial conductivity and calculationof radial conductivity. Predicted values of Lp for entire rootsbased on two to ten segments per root averaged 1.04±0.07(mean±s.e. mean for n = 3) of the measured Lp for A.deserti and 1.06±0.10 for O. ficus-indica. The modelalso closely predicted the drop in water potential along theroot xylem (xylem); when a tension of 50 kPa was applied tothe proximal ends of 0.2 m-long roots of A. deserti and O. ficus-indica,the measured xylem to midroot averaged 30 kPa compared witha predicted decrease of 36 kPa. Such steep gradients in xylemsuggest that the driving force for water movement from the soilto young distal roots may be relatively small. The model, whichagreed with an analytical solution for a simple hypotheticalsituation, can quantify situations without analytical solutions,such as when root and soil properties vary arbitrarily alonga root. H+-Transport activity of the vesicles prepared from barley rootswas studied at the early phase after application of NaCl stress.The activity reached maximal level at 3 days after the treatmentwith 200 mM NaCl which moderately reduced the growth. This activityincrease could be suppressed in the presence of cycloheximideand actinomycin D. The properties of the membrane vesicles associated with H+-transportactivity prepared from both control and NaCl-stressed rootssuggested that it was of tonoplast origin based on the followingfindings: optimal pH at 7.5, strong inhibition by nitrate butnot by vanadate, and stimulation by chloride. The density gradient centrifugation of vesicles with DextranT70 did not show any detectable difference in the distributionpatterns of H+-transport activities between control and NaClstressedroots. Furthermore, Km values for ATP of the H+-transport activityof vesicles prepared from control and NaCl-stressed roots werethe same. Guard cell protoplasts (GCP) were prepared from leaves of Commelinacommunis L. and phosphoenolpyruvate carboxylase (PEPc) activityrecorded after injection of the protoplasts directly into theassay medium. The GCP were lysed immediately by the presenceof Triton X-100 and a lowered osmotic concentration in the assaycuvette enabling PEPc activity to be measured with ‘nascent’enzyme. There was no light activation of the enzyme with KmPEP (about 3.7 mol m–3) and Vmax being similar in light-ordark-treated protoplasts. Illumination of the GCP in the presenceof CO2-free air and KCI, a treatment which is known to swellGCP, did not change the kinetics. PEPc activity at saturating PEP was very sensitive to malateinhibition, 20 mmol m–3 (the I50 value) inhibiting activityby about 50%. Inhibition was similar in light- or dark-treatedprotoplasts. Malate inhibition was, however, much less (I50= 500 mmol m–3) if the enzyme source was a protoplastextract kept in the absence of glycerol. Inclusion of 20% glycerolin the extraction medium maintained the enzyme in the malate-sensitiveform as occurred in the in vivo assays. The high apparent KmPEP and the high sensitivity to malate inhibition of GCP PEPcare features unlike those observed with PEPc from leaf tissuesof C4 and CAM plants and from GCP extracts. PEPc activity increased slightly in the presence of KCI in theassay medium up to about 10 mol m–3 and thereafter activityslowly declined as KCI concentrations increased further. Amounts of the enzyme ribulose-1,5-bisphosphate carboxylasewere estimated in seedling leaves of barley (Hordewn vulgareL.) and flag leaves of wheat (Triticum aestitum L.) by radialimmuno diffusion. A fourfold variation among barley varietiesfor amount of RuBPCase at the seedling stage was observed (c.3.5–15mg g–1 fr. wt). Range in variation for amountof flag leaf RuBPCase among wheat varieties was 6-09-9.39 mgRuBPCase g–1 fr. wt. F1 hybrids from interspecific andintergeneric crosses of crested wheatgrasses (Agropyron andElymus spp.) and their amphidiploid analogues were comparedfor amount of RuBPCase in the most recent fully expanded leavesharvested before seed set. Amount of enzyme varied from 3.4to 77.6 mg g–1 fr. wt among the hybrids. No effect chromosomenumber on enzyme concentration was observed among 13 hybridsand their amphidiploid counterparts. Influx of nitrate into the roots of intact barley plants wasfollowed over periods of 1–15 min using nitrogen-13 asa tracer. Based on measurements taken over 15 min from a rangeof external nitrate concentrations (0·2–250 mmolm–3), the kinetic parameters of influx, Imax and Km, werecalculated. Compared with plants grown in the presence of nitrate throughout,plants that had been starved of N for 3 d showed a significantlygreater value ofImax for 13N-nitrate influx (by a factor of1·4–1·8), but a similar value of Km (12–14mmol m–3). Pre-treating N-starved plants with nitratefor about 5 h further increased the subsequent rate of 13N-nitrateinflux, but had little effect in the unstarved controls. Allowingfor this induction of additional nitrate transport, the differencein rates of nitrate influx in control and N-starved plants wassufficient to account for the previously-observed differencein net uptake by the two groups of plants. In barley plants grown without any exposure to nitrate, butwith ammonium as N-source, both Imax and Km for subsequent 13N-nitrateinflux were significantly decreased (by about one-half) comparedwith the corresponding nitrate-grown controls. Barley was grown at a range of oxygen concentrations (0.5–9mg l–1), in nutrient solutions. Growth of both shootsand seminal roots was restricted by O2 concentrations lowerthan 2–3 mg l–1) but nodal root growth was not. Root porosities were increased even at those O2 concentrationswhich did not restrict growth, and were inversely proportionalto the protein levels of the roots. Growth and nitrate uptake kinetics in vegetatively growing barley(Hordeum vulgare L., cvs Laevigatum, Golf, and Mette) were investigatedin solution culture under long-term limitations of externalnitrogen availability. Nitrate was fed to the cultures at relativeaddition rates (RA) ranging from 0.02 to 0.2 d–1. Therelative growth rate (RG, calculated for total plant dry weight)correlated well with RA in the range 0.02 to 0.07 d–1.In the RA range from 0.07 to 0.2 d–1 RG continued to increase,but an increasing fraction of nitrogen, added and absorbed,was apparently stored rather than used for structural growth.The RG of the roots was less affected by RA. Vmax, for net nitrateuptake increased with RA up to 0.11 d–1, but decreasedat higher RA. The decline in Vmax coincided with a build-upof nitrate stores in both roots and shoots. Vmax, expressedper unit nitrogen in the plants (the relative Vmax, was higherthan required for maintenance of growth (up to 30-fold) at lowRA, whereas at higher RA the relative Vmax decreased. Kineticpredictions of steady-state external nitrate concentrationsduring N-limited growth ranged from 0.2 to 5.0 mmol m–3over the RG range 0.02 to 0.11 d–1. It is suggested thatthe nitrate uptake system is not under specific regulation atlow RA, but co-ordinated with root protein synthesis and growthin general. At RA higher than 0.11 d–1, however, specificregulation of nitrate uptake, possibly via root nitrate pools,become important. We studied the course of early leaf area expansion and specificleaf area (SLA) in potato (Solanum tuberosum L.) and wheat (Triticumaestivum L.) genotypes and tested whether air temperature explainsdifferences in these courses within different environments.Such knowledge can be used to improve crop growth modelling.The relative rate of leaf area expansion (RL) of potato andwheat decreased with thermal time, but was nearly linear upto a leaf area index (L) of 1.0. TheRL (L < 1; mean: 17.9x 10-3°C-1 d-1) of potato showed an interaction betweengenotype and environment, and varied with year. TheRL (L <1; mean: 7.1 x 10-3°C-1 d-1) of winter wheat was lower thanthat of spring wheat (mean: 10.9 x 10-3°C-1 d-1), and bothvaried with year. SLAof potato increased nearly linearly withthermal time from 5 to 15 m2 kg-1at 50% emergence, to 20 to25 m2 kg-1at 155°Cd, and then decreased slightly. The SLAofboth winter and spring wheat began at 16 to 23 m2 kg-1and inmost cases increased slightly with thermal time. In potato,regression parameters of SLAwith thermal time were affectedby environment (management conditions and year) and genotype;in wheat they were affected by environment (year and site).Treatment effects on RLof potato were not correlated with thoseon SLA , and were only partly correlated for wheat. Thereforewe conclude that the early foliar expansion of potato is associatedwith a strong increase in SLA , and not so for wheat. The study of ß-glycerophosphatase activity in cell-wallpreparations and in excised root tips from barley seedlingssupports the view that the former, which constitutes about 20per cent of the activity of the whole homogenate, representsthe fraction located at the surface of the roots in vivo. Theactivities of the cell-wall suspension and intact roots arevirtually identical, and further show identical relations topH, substrate concentration (Km), and competitive inhibitionby molybdate and inorganic phosphate (Ki). The enzyme must thereforebe freely exposed to the external solution without any permeabilitybarrier separating it from either substrate or inhibitors. Theabsence of any lag phase in the hydrolysis in excised root tipssuggests that the surface enzyme may be limited to the outermostlayers of the root. The solubilization of some of the activityof the cell-wall preparation by treatment with sodium chlorideand ammonium sulphate suggest that surface activity may havebeen lost from these preparations rather than adsorbed duringhomogenization and extraction. The Km and pH-activity curveof the supernatant activity remaining after centrifugation ofthe cell-wall fraction indicate that only this enzyme and noother detectable glycerophosphatase exists in the roots.  相似文献

The activity of shikimate: NADP oxidoreductase [EC 1. 1. 1.25] in sweet potato root tissue increased soon after slicing.Enzyme preparations obtained from both sliced tissue and fromfresh tissue probably contained a single enzyme component, andthey showed identical chromatographical behaviour. Km values of the enzyme for NADP and shikimate were 1.0x10–4Mand 1.3 x 10–3M, respectively. Enzyme activity was potentlyinhibited by SH-inhibitors such as p-chloromercuribenzoate andoxidized glutathione. Enzyme activity was affected neither by mononucleotides suchas ATP, ADP and AMP, divalent cations, Mg++, Ca++ and Mn++,nor by metabolites such as tryptophan, phenylalanine, tyrosineand t-cinnamic acid which are involved in aromatic compoundsyntheses. The enzyme rapidly lost its activity. This inactivation reactionshowed a time course consisting of two steps of the first-orderreaction. The inactivated enzyme preparation was not reactivatedby thiol compounds such as cysteine, 2-mercaptoethanol and glutathione,although these reagents, to a certain extent, protected theenzyme from inactivation. NADP-malic enzyme (EC 1.1.1.40 [EC] ), which is involved in Crassulaceanacid metabolism (CAM), was purified to electrophoretic homogeneityfrom the leaves of the inducible CAM plant Mesembryanthemumcrystallinum. The NADP-malic enzyme, which was purified 1,146-fold,has a specific activity of 68.8 µmol (mg protein)–1min–1. The molecular weight of the subunits of the enzymewas 64 kDa. The native molecular weight of the enzyme was determinedby gel-filtration to be 390 kDa, indicating that the purifiedNADP-malic enzyme is a hexamer of identical subunits. The optimalpH for activity of the enzyme was around 7.2. Double-reciprocalplots of the enzymatic activity as a function of the concentrationof L-malate yielded straight lines both at pH 7.2 and at pH7.8 and did not reveal any evidence for cooperativity of bindingof L-malate. The Km value for L-malate was 0.35 mM. Hill plotsof the activity as a function of the concentration of NADP+indicated positive cooperativity in the binding of NADP+ tothe enzyme with a Hill coefficient (nH) of 2.0. An S0.5 value(the concentration giving half-maximal activity) of 9.9 µMfor NADP+ was obtained. Oxaloacetate inhibited the activityof the NADP-malic enzyme. Increased extracellular osmolarity ([Os]e) suppresses stimulated hormone secretion from anterior pituitary cells. Ca2+ influx may mediate this effect. We show that increase in [Os]e (by 18–125%) differentially suppresses L-type and T-type Ca2+ channel currents (IL and IT, respectively); IL was more sensitive than IT. Hyperosmotic suppression of IL depended on the magnitude of increase in [Os]e and was correlated with the percent decrease in pituitary cell volume, suggesting that pituitary cell shrinkage can modulate L-type currents. The hyperosmotic suppression of IL and IT persisted after incubation of pituitary cells either with the actin-disrupter cytochalasin D or with the actin stabilizer phalloidin, suggesting that the actin cytoskeleton is not involved in this modulation. The hyperosmotic suppression of Ca2+ influx was not correlated with changes in reversal potential, membrane capacitance, and access resistance. Together, these results suggest that the hyperosmotic suppression of Ca2+ influx involves Ca2+ channel proteins. We therefore recorded the activity of L-type Ca2+ channels from cell-attached patches while exposing the cell outside the patch pipette to hyperosmotic media. Increased [Os]e reduced the activity of Ca2+ channels but did not change single-channel conductance. This hyperosmotic suppression of Ca2+ currents may therefore contribute to the previously reported hyperosmotic suppression of hormone secretion. Barley and rice, at the early tillering stage, were grown inaerated nutrient solutions (> 7 mg O2 l–1) and transferredto solutions of low O2 concentrations (< 0.5 mg l –1). For barley, low O2 concentrations during the first 5 days severelyinhibited growth of seminal roots had less effect on nodal roots,and did not reduce shoot growth. Longer exposure to low O2 concentrationsreduced shoot as well as root growth. Sugar concentrations inroots and shoots increased within 7 h after transfer of plantsto low O2 concentrations. After 5 days at low O2 concentrationssugar concentrations were very high in fast growing nodal rootsand in shoots, as well as in the slower growing seminal roots. In rice, low O2 concentrations increased sugar levels of rootsduring summer, but not during winter. In summer, the highersugar levels at low O2 concentrations persisted throughout adiurnal cycle. In root apices, sugar concentrations were increasedby low O2 concentrations, even though the experiment was donein winter and the bulk of the root system showed no differencein sugar levels. The data indicate that sugar accumulation, at low O2 concentrations,is caused by reduced growth and also that even apices of rootsgrown at low O2 concentrations have sufficient substrates forrespiration.
