Total ankle replacement (TAR) is an alternative to fusion, replacing the degenerated joint with a mechanical motion-preserving alternative. Minimal pre-clinical testing has been reported to date and existing wear testing standards lack definition. Ankle gait is complex, therefore the aim of this study was to investigate the effect on wear of a range of different ankle gait kinematic inputs. Five Zenith (Corin Group) TARs were tested in a modified knee simulator for twelve million cycles (Mc). Different combinations of IR rotation and AP displacement were applied every 2Mc to understand the effects of the individual kinematics. Wear was assessed gravimetrically every Mc and surface profilometry undertaken after each condition. With the initial unidirectional input with no AP displacement the wear rate measured 1.2±0.6 mm3/Mc. The addition of 11° rotation and 9 mm of AP displacement caused a statistically significant increase in the wear rate to 25.8±3.1 mm3/Mc. These inputs seen a significant decrease in the surface roughness at the tibial articulation. Following polishing three displacement values were tested; 0, 4 and 9 mm with no significant difference in wear rate ranging 11.8–15.2 mm3/Mc. TAR wear rates were shown to be highly dependent on the addition of internal/external rotation within the gait profile with multidirectional kinematics proving vital in the accurate wear testing of TARs. Prior to surface polishing wear rates were significantly higher but once in a steady state the AP displacement had no significant effect on the wear.  相似文献

After menopause, critically estrogen low levels result in modifications in vaginal wall. This cross-sectional study aims to determine whether there is a change in the number of vessels in the lamina propria of the vagina after menopause in parallel to the ER-alpha expression on the vaginal wall. Twelve women who underwent a genital surgery for genital prolapse up to grade II were selected. They were divided into two groups: a premenopausal group (PG) consisting of six women who were 18–40 years old with FSH levels =12 mIU/ml and regular cycles, and a menopausal group (MG) consisting of six women at least one year after menopause who were <65 years old with FSH levels =40 mIU/ml. Slides were stained for ER-alpha immunohistochemistry, and an endothelial cell marker CD3 was used to label vessels which were identified by using a system for morphometry. The number of vessels was significantly higher in the PG than in the MG both on the anterior wall (PG: 1.055 ± 145.8 vessels/mm2, MG: 346.6 ± 209.9 vessels/mm2, p < 0.0001) and on the posterior wall (PG: 1064 ± 303.3 vessels/mm2, MG: 348.6 ± 167.3 vessels/mm2, p = 0.0005). The ER-alpha score was significantly higher in the PG than the score for the MG on both the anterior and posterior walls (PG: 6.0 ± 0.52, MG: 2.5 ± 0.89, p = 0.007; PG: 5.8 ± 0.79, MG: 2.7 ± 0.95, p = 0.03, respectively). There was a positive correlation between the ER-alpha score and the vessel concentration on the anterior (r = 0.6656, p = 0.018) and posterior (r = 0.6738, p = 0.016) vaginal walls. Age was strongly negatively correlated with vessel concentration on the vaginal walls (respectively r = -0.9033, p < 0.0001, r = -0.7440, p = 0.0055). Therefore, postmenopausal women with genital prolapse have a smaller number of vessels on the vaginal wall compared to normoestrogenic controls with the same pathological condition. Hypoestrogenism and advancing age are factors that are associated to these changes.  相似文献

Knee contact mechanics play an important role in knee implant failure and wear mechanics. Femoral condylar contact loss in total knee arthroplasty has been reported in some studies and it is considered to potentially induce excessive wear of the polyethylene insert.Measuring in vivo forces applied to the tibial plateau with an instrumented prosthesis is a possible approach to assess contact loss in vivo, but this approach is not very practical. Alternatively, single-plane fluoroscopy and pose estimation can be used to derive the relative pose of the femoral component with respect to the tibial plateau and estimate the distance from the medial and lateral parts of the femoral component towards the insert. Two measures are reported in the literature: lift-off is commonly defined as the difference in distance between the medial and lateral condyles of the femoral component with respect to the tibial plateau; separation is determined by the closest distance of each condyle towards the polyethylene insert instead of the tibia plateau.In this validation study, lift-off and separation as measured with single-plane fluoroscopy are compared to in vivo contact forces measured with an instrumented knee implant. In a phantom study, lift-off and separation were compared to measurements with a high quality bi-plane measurement.The results of the in vivo contact-force experiment demonstrate a large discrepancy between single-plane fluoroscopy and the in vivo force data: single-plane fluoroscopy measured up to 5.1 mm of lift-off or separation, whereas the force data never showed actual loss of contact. The phantom study demonstrated that the single-plane setup could introduce an overestimation of 0.22 mm±±0.36 mm. Correcting the out-of-plane position resulted in an underestimation of medial separation by −0.20 mm±±0.29 mm.In conclusion, there is a discrepancy between the in vivo force data and single-plane fluoroscopic measurements. Therefore contact loss may not always be determined reliably by single plane fluoroscopy analysis.  相似文献

The active ingestion of xylem sap by aphids is hypothesised to be an important mechanism for rehydration. When starved bird cherry-oat aphids (Rhopalosiphum padi) were allowed to feed on wheat (Triticum aestivum) treated with a sublethal dose of the xylem-mobile neonicotinoid thiamethoxam, analysis of feeding behaviours using the electrical penetration graph revealed a reduction in xylem feeding that was reversed on removal of the toxin. To test the importance of xylem-feeding behaviour as a rehydration mechanism, the effects of the sublethal dose of thiamethoxam on aphid water content, honeydew excretion, growth and fecundity were investigated. Body water contents of starved R. padi feeding on wheat treated with thiamethoxam were significantly reduced compared to aphids feeding on wheat treated with distilled water (74.5 ± 0.23 and 75.6 ± 0.18%, respectively). In addition, the sublethal dose of thiamethoxam had detrimental effects on aphid performance. At reproductive maturity, aphids that had been born on wheat treated with thiamethoxam were significantly smaller (as measured by body plan area; 1.07 ± 0.03 mm2), lighter (0.31 ± 0.04 mg) and less fecund (2.85 ± 0.36 nymphs/day) than aphids born on wheat treated with distilled water (1.87 ± 0.02 mm2, 0.72 ± 0.03 mg, 11.28 ± 0.58 nymphs/day, respectively). Regardless of whether the observed impairment of xylem feeding is due to a neurotoxic or an antifeedant effect, these results have important implications for commercial crop protection as the behaviour-modifying effects of the sublethal dose of thiamethoxam may change the efficacy of this pesticide throughout the growing season.  相似文献

Wetlands have proven effective at improving water quality of treated wastewater effluent, which in turn promotes increased primary productivity and vertical accretion. Baldcypress (Taxodium distichum) seedlings grown under different conditions (bare root and potted) were planted in four subunits of an effluent assimilation marsh and a control marsh in southeast Louisiana, USA, and basal diameter growth was monitored over one growing season. Mean basal diameter growth for seedlings in the assimilation subunits ranged from 16.1 (±1.4) mm to 9.5 (±0.9) mm, whereas growth for seedlings planted in the control marsh was 6.4 (±0.9) mm. Seedlings planted nearest the outfall experienced greater basal diameter growth (18.1 ± 2.6) compared to those planted 700 m away (8.0 ± 0.9), with growth generally decreasing with distance. Potted seedlings experienced greater growth (19.1 ± 1.0 and 20.6 ± 1.0 for five-month-olds and ten-month olds, respectively) than bare root seedlings (4.6 ± 0.6 and 4.0 ± 0.4 for one-year-olds and two-year olds, respectively). Planting assimilation marshes with baldcypress seedlings can be an effective restoration tool for coastal Louisiana, which will provide hurricane protection and improved surface water quality. Wastewater treatment wetlands may offer an effective tool for restoring coastal baldcypress (T. distichum)-water tupelo (Nyssa aquatic) swamps in Louisiana.  相似文献

This study assessed the efficacy of a protocol combining short-interval cloprostenol-based protocols and “male effect” for estrous synchronization in hair sheep. In Experiment 1, 24 ewes were randomly assigned to three groups (n = 8) and treated with cloprostenol on Days 3, 5 and 7 after ovulation, respectively. Estradiol secretion during the follicular phase was similar among groups. Onset of estrus (P < 0.001) and the timing of maximum LH concentration (P < 0.01) were earlier in group D3 than in D5 and D7 groups. During the subsequent cycle, the number and size of corpora lutea were higher (P < 0.05) in ewes of the groups D3 (1.9 ± 0.3 and 115.1 ± 14.3 mm2) and D5 (1.8 ± 0.2 and 100.2 ± 11.2 mm2) than in group D7 (1.3 ± 0.2 and 75.6 ± 6.4 mm2) group. In Experiment 2, 24 ewes were treated with two cloprostenol injections (7 days apart). Twelve ewes were exposed to “male effect” previous to an isolation period (ME group), whereas the remaining ewes were controls without male exposure (CTR group). Male effect induced earlier preovulatory LH surge (P < 0.05) and ovulation (P < 0.001) than CTR group. In Experiment 3, the estrus was synchronized in 68 ewes. Nineteen of them (group FGA) were treated using intravaginal sponges impregnated with fluorogestone acetate for 12 days and inseminated at 55 h. Forty-nine females (group ME) were treated like ME group. Twenty-four (ME48 group) and 25 ewes (ME55 group) were inseminated at 48 and 55 h after treatment, respectively. The fertility rate was numerically higher in ME48 than ME55 and FGA groups (62.5, 44.0 and 47.4%, respectively). In conclusions, the combined use of short-interval cloprostenol treatment and “male effect” may be an adequate alternative for synchronizing estrus and applying artificial insemination in hair sheep throughout the entire year.  相似文献

The sperm-egg interaction assay is a good predictor of the fertilizing potential of rooster semen; the ability of chicken sperm to interact with the egg can be assessed by counting the number of holes in the inner perivitelline layer (IPVL) of a freshly laid egg. Atlhough isolated IPVL can be stored for up to 24 h, preservation of IPVL for prolonged intervals in liquid nitrogen would facilitate the sperm-egg interaction assay. The objective of this study was to adapt the technique of vitrifying swine oocytes for use with the IPVL. Our hypothesis was that vitrification would not alter the ability of the membrane to bind sperm; therefore, there would be no difference between vitrified and fresh IVPL in the number of hydrolysis holes made by sperm. Our hypothesis was supported; there were no differences in the mean ± SEM number of holes made by the same sample of sperm in vitrified and in fresh membranes (146.0 ± 17.7 holes/mm2 IPVL and 159.5 ± 17.7 holes/mm2 IPVL, respectively, P > 0.05; n = 123 IVPLs tested). Furthermore, 80% of frozen-thawed membranes were recovered intact. Because vitrification did not significantly change the ability of membranes to bind sperm, vitrified membranes can be safely used for the sperm-egg interaction assay. Vitrified IVPL would ensure availability for sperm evaluation and facilitate wide distribution of IPVL, enabling assays to be conducted even in the absence of facilities or expertise to prepare membranes.  相似文献

Purpose

In this study, we set out to establish an in vivo animal model of oxidative stress in the retinal ganglion cells (RGCs) and determine whether there is a link between oxidative stress in the RGCs and the activation of calpain, a major part of the apoptotic pathway.

Materials and methods

Oxidative stress was induced in the RGCs of C57BL/6 mice by the intravitreal administration of 2,2′-azobis (2-amidinopropane) dihydrochloride (AAPH, 30 mM, 2 μl). Control eyes were injected with 2 μl of vehicle. Surviving Fluorogold (FG)-labeled RGCs were then counted in retinal flat mounts. Double staining with CellROX and Annexin V was performed to investigate the co-localization of free radical generation and apoptosis. An immunoblot assay was used both to indirectly evaluate calpain activation in the AAPH-treated eyes by confirming α-fodrin cleavage, and also to evaluate the effect of SNJ-1945 (a specific calpain inhibitor: 4% w/v, 100 mg/kg, intraperitoneal administration) in these eyes.

Results

Intravitreal administration of AAPH led to a significant decrease in FG-labeled RGCs 7 days after treatment (control: 3806.7 ± 575.2 RGCs/mm2, AAPH: 3156.1 ± 371.2 RGCs/mm2, P < 0.01). CellROX and Annexin V signals were co-localized in the FG-labeled RGCs 24 h after AAPH injection. An immunoblot assay revealed a cleaved α-fodrin band that increased significantly 24 h after AAPH administration. Intraperitoneally administered SNJ-1945 prevented the cleavage of α-fodrin and had a neuroprotective effect against AAPH-induced RGC death (AAPH: 3354.0 ± 226.9 RGCs/mm2, AAPH+SNJ-1945: 3717.1 ± 614.6 RGCs/mm2, P < 0.01).

Conclusion

AAPH administration was an effective model of oxidative stress in the RGCs, showing that oxidative stress directly activated the calpain pathway and induced RGC death. Furthermore, inhibition of the calpain pathway protected the RGCs after AAPH administration.  相似文献

The neuromusculoskeletal system interacts with the external environment via end-segments, e.g. feet. A person with trans-tibial amputation (TTAmp) has lost a foot and ankle; hence the residuum with prosthesis becomes the new end-segment. We investigated changes in kinetics and muscle activity in TTAmps during cycling with this altered interface with the environment. Nine unilateral TTAmps and nine subjects without amputation (NoAmp) pedaled at a constant torque of 15 Nm and a constant cadence of 90 rpm (~150watts). Pedal forces and limb kinematics were used to calculate resultant joint moments. Electromyographic activity was recorded to determine its magnitude and timing. Biomechanical and EMG variables of the amputated limb were compared to those of the TTAmp sound limb and to the dominant limb in the NoAmp group using a one-way ANOVA. Results showed maximum angular displacement between the residuum and prosthesis was 4.8±1.8 deg. The amputated limb compared to sound limb and NoAmp group produced lower extensor moments averaged over the cycle about the ankle (13±2.3, 20±5.7, and 19±5.3 Nm, respectfully) and knee (8.4±5.0, 15±4.5, and 12.7±5.9 Nm, respectfully) (p<0.05). Gastrocnemius and rectus femoris peak activity in the TTAmps shifted to later in the crank cycle (by 36° and 75°, respectfully; p<0.05). These data suggest gastrocnemius was utilized as a one-joint knee flexor in combination with rectus femoris for prosthetic socket control and highlight prosthetic control as an interaction between the residuum, prosthesis and external environment.  相似文献

Purpose

We investigated the influence of Leptin (LEP) and leptin receptor (LEPR) SNPs on habitual physical activity (PA) and body composition response to a unilateral, upper body resistance training (RT) program.

Methods

European-derived American volunteers (men = 111, women = 131, 23.4 ± 5.4 yr, 24.4 ± 4.6 kg·m− 2) were genotyped for LEP 19 G>A (rs2167270), and LEPR 326 A>G (rs1137100), 668 A>G (rs1137101), 3057 G>A (rs1805096), and 1968 G>C (rs8179183). They completed the Paffenbarger PA Questionnaire. Arm muscle and subcutaneous fat volumes were measured before and after 12 wk of supervised RT with MRI. Multivariate and repeated measures ANCOVA tested differences among phenotypes by genotype and gender with age and body mass index as covariates.

Results

Adults with the LEP 19 GG genotype reported more kcal/wk in vigorous intensity PA (1273.3 ± 176.8, p = 0.017) and sports/recreation (1922.8 ± 226.0, p < 0.04) than A allele carriers (718.0 ± 147.2, 1328.6 ± 188.2, respectively). Those with the LEP 19 GG genotype spent more h/wk in light intensity PA (39.7 ± 1.6) than A allele carriers (35.0 ± 1.4, p = 0.03). In response to RT, adults with the LEPR 668 G allele gained greater arm muscle volume (67,687.05 ± 3186.7 vs. 52,321.87 ± 5125.05 mm3, p = 0.01) and subcutaneous fat volume (10,599.89 ± 3683.57 vs. − 5224.73 ± 5923.98 mm3, p = 0.02) than adults with the LEPR 668 AA genotype, respectively.

Conclusion

LEP19 G>A and LEPR 668 A>G associated with habitual PA and the body composition response to RT. These LEP and LEPR SNPs are located in coding exons likely influencing LEP and LEPR function. Further investigation is needed to confirm our findings and establish mechanisms for LEP and LEPR genotype and PA and body composition associations we observed.  相似文献

A new species, Huernia humpatana Bruyns (Apocynaceae–Ceropegieae), closely related to H. similis N.E.Br., is described from the Chela Mountains of Huila Province in southern Angola. The two species are distinguished by the 5-angled and erect stems with more prominent tubercles up to 6 mm long joined into clear angles and separated by V-shaped grooves in H. humpatana as opposed to very obtusely 4-angled stems with tubercles only 2 mm long and only indistinct grooves between the angles in H. similis. Furthermore, in H. similis the nodding corolla is ± 9 mm in diameter with sepals ± 2 mm long, while in H. humpatana the horizontally facing corolla is 18–20 mm in diameter with sepals 4–6 mm long.  相似文献

Objective

Angiotensin-(1-7) [ANG-(1-7)] has been reported to attenuate neointimal formation after vascular injury and stent implantation in rats, but the mechanism remains mostly unresolved. Interestingly, the levels of circulating transforming growth factor-beta1 (TGF-β1) after myocardial infarction were suppressed by ANG-(1-7), which suggests a possible downstream target for the anti-remodeling action of ANG-(1-7). Our study focused on the effects of ANG-(1-7) on vascular remodeling, including neointimal formation and collagen synthesis, and determining whether or not these effects were dependent upon the TGF-β signaling pathway.

Methods

Thirty-two New Zealand white rabbits underwent sham surgery or angioplasty in abdominal aorta. The animals were divided into four groups, which were sham, control, ANG-(1-7), and ANG-(1-7) + A-779. Subsequently, an osmotic minipump was implanted to deliver saline, ANG-(1-7) (576 μg kg−1 d−1) or ANG-(1-7) + A-779 (576 μg kg−1 d−1) for 4 weeks.

Results

The ANG-(1-7) group displayed a significant reduction in neointimal thickness (207.51 ± 16.70 μm vs. 448.08 ± 15.30 μm, P < 0.001), neointimal area (0.266 ± 0.009 mm2 vs. 0.408 ± 0.002 mm2, P < 0.001), and restenosis rate (28.13 ± 2.74% vs. 40.13 ± 2.74%, P < 0.001) when compared to the control group. ANG-(1-7) also inhibited collagen synthesis by significantly decreasing the mRNA expression of Collagen I and Collagen III (vs. Control group: 0.2190 ± 0.0036 vs. 0.3852 ± 0.0212, P < 0.001 and 1.1328 ± 0.0554 vs. 1.7378 ± 0.1164, P < 0.001, respectively). Furthermore, the expression of TGF-β1 and phosphor-Smad2 (p-Smad2) were significantly suppressed by ANG-(1-7) (vs. Control group: 1.21 ± 0.07 vs. 1.54 ± 0.08, P < 0.001 and 0.31 ± 0.01 vs. 0.43 ± 0.02, P < 0.001, respectively), but no effect on p38 phosphorylation was observed. [d-Ala7]-ANG-(1-7) (A-779), showed a tendency to attenuate the anti-remodeling effects of ANG-(1-7).

Conclusion

ANG-(1-7) decreases the amount of vascular remodeling, including a reduction in neointimal formation and collagen synthesis, after angioplasty in rabbits. The responsible mechanism may function through the possible down-regulation of TGF-β1 levels and inhibition of the Smad2 pathway.  相似文献

The present study deals with the development of novel pH-sensitive tamarind seed polysaccharide (TSP)-alginate composite beads for controlled diclofenac sodium delivery using response surface methodology by full 32 factorial design. The effect of polymer-blend ratio (sodium alginate:TSP) and cross-linker (CaCl2) concentration on the drug encapsulation efficiency (DEE, %) and drug release from diclofenac sodium loaded TSP-alginate composite beads prepared by ionotropic gelation was optimized. The observed responses were coincided well with the predicted values by the experimental design. The DEE (%) of these beads containing diclofenac sodium was within the range between 72.23 ± 2.14 and 97.32 ± 4.03% with sustained in vitro drug release (69.08 ± 2.36-96.07 ± 3.54% in 10 h). The in vitro drug release from TSP-alginate composite beads containing diclofenac sodium was followed by controlled-release pattern (zero-order kinetics) with case-II transport mechanism. Particle size range of these beads was 0.71 ± 0.03-1.33 ± 0.04 mm. The swelling and degradation of the developed beads were influenced by different pH of the test medium. The FTIR and NMR analyses confirmed the compatibility of the diclofenac sodium with TSP and sodium alginate used to prepare the diclofenac sodium loaded TSP-alginate composite beads. The newly developed TSP-alginate composite beads are suitable for controlled delivery of diclofenac sodium for prolonged period.  相似文献

Aims

Dietary flavonoid intake shows a significant inverse association with mortality from coronary heart disease, incidence of myocardial infarction and stroke. Quercetin is one of the most common flavonoids in our diet and has several favorable biological activities. Quercetin glucosides, which are enzymatically trans-glycosylated isoquercitrin, have high water-solubility and bioavailability compared with quercetin. Here, we investigated the effects of quercetin glucosides on collateral development in a murine hindlimb ischemia model.

Main methods

We induced hindlimb ischemia in 24- to 32-week-old male C3H/HeJ mice by resecting the right femoral artery. Then, 0.5% carboxymethyl cellulose (control) or quercetin glucosides (100 mg/kg/day) were administered daily by gavage. Blood flow was monitored weekly by laser Doppler imaging.

Key findings

Recovery of blood flow to the ischemic leg was significantly enhanced by quercetin glucosides (blood flow ratio at 4 weeks: control, 0.57 ± 0.11; quercetin glucosides, 0.95 ± 0.10, p < 0.05). Furthermore, anti-CD31 immunostaining revealed that quercetin glucosides increased capillary density in the ischemic muscle (control, 200 ± 24/mm2; quercetin glucosides, 364 ± 41/mm2, p < 0.01). Quercetin glucosides did not promote tumor growth. The beneficial effect of quercetin glucosides was abrogated in eNOS-deficient mice.

Significance

These results suggest that quercetin glucosides may have therapeutic potential to promote angiogenesis in ischemic tissue.  相似文献

Sicilo-Sarde is a local breed from northern Tunisia resulting from crossing Sarda and Comisana dairy sheep and is traditionally used for cheese production after lamb weaning. A sample of 52 adult Sicilo-Sarde lactating ewes was used for studying their udder morphological traits and milk yield potential during weeks 4-8 (milking-suckling period) and 10 (milking period) of lactation. Daily milk yield was estimated on a daily basis by using the double oxytocin injection method 4 h after machine-milking at d 30 and 45. Udder and teat morphology were also measured at d 45 of lactation. Cisternal area (by ultrasonography) and udder compartments (cisternal and alveolar milk) were evaluated 8 h after milking by using atosiban and oxytocin on d 72 of lactation. Milk yield averaged 0.56 ± 0.10 L/d and ewes had small (volume, 496 ± 28 mL) and healthy udders (CMT, <1), with medium sized teats (length, 18.5 ± 4.9 mm; diameter, 10.0 ± 2.0 mm) attached at 45 ± 10°. A drop in milk production (49%) was found in the transition from suckling (day 30) to milking (week 10). Udder cisterns were multilocular and small sized (half udder area, 11.6 ± 4.5 cm2), although cisternal milk accounted for 54% of the total milk in the udder. Correlation between cisternal milk and cisternal area was moderate (R2 = 0.48; P < 0.05). Lag time and total milking time were 1.9 ± 0.1 and 31 ± 5 s, respectively. In conclusion, the Sicilo-Sarde ewes evaluated showed medium sized cisterns and teats which were morphologically adequate for machine milking, although milk production needs to be improved.  相似文献

Multiphoton microscopy (MPM) holds promise as a noninvasive imaging technique for characterizing collagen structure, and thus mechanical properties, through imaging second harmonic generation (SHG) and two-photon fluorescence in engineered and real connective tissues. Controlling polymerization pH to manipulate collagen gel microstructure, we quantified pore and fiber dimensions using both standard methods and image correlation spectroscopy (ICS) on MPM, scanning electron, and darkfield microscopy images. The latter two techniques are used to confirm microstructural measurements made from MPM images. As polymerization pH increased from 5.5 to 8.5, mean fiber diameter decreased from 3.7 ± 0.7 μm to 1.6 ± 0.3 μm, the average pore size decreased from 81.7 ± 3.7 μm2 to 7.8 ± 0.4 μm2, and the pore area fraction decreased from 56.8% ± 0.8% to 18.0% ± 1.3% (measured from SHG images), whereas the storage modulus G′ and loss modulus G′, components of the shear modulus, increased ∼33-fold and ∼16-fold, respectively. A characteristic length scale measured using ICS, WICS, correlates well with the mean fiber diameter from SHG images (R2 = 0.95). Semiflexible network theory predicts a scaling relationship of the collagen gel storage modulus (G′) depending upon mesh size and fiber diameter, which are estimated from SHG images using ICS. We conclude that MPM and ICS are an effective combination to assess bulk mechanical properties of collagen hydrogels in a noninvasive, objective, and systematic fashion and may be useful for specific in vivo applications.  相似文献

Calcium oxalate crystal and silicophytolith assemblages were investigated in leaves of eleven aquatic species from habitats of the Argentinean Pampean Plain. Calcium oxalate druses and raphide bundles mainly occurred in leaf parenchyma and aerenchyma. Druses were observed in Alternanthera philoxeroides, Ludwigia peploides, Polygonum hydropiperoides and Rumex crispus; and raphide bundles in L. peploides and Typha latifolia. No calcium oxalate crystals were observed in Bidens laevis, Mikania parodii, Solanum glaucophyllum, Ranunculus apiifolius or the Juncaceae family. Druse density ranged between 14.62 ± 3.09 (R. crispus) to 39.6 ± 7.2 crystals mm−2 (P. hydropiperoides), and raphide bundle density ranged between 7.55 ± 1.36 (L. peploides) to 20.55 ± 5.19 crystals mm−2 (T. latifolia). Silicification mainly occurred in epidermal cells and xylem. Juncus spp., S. glaucophyllum, B. laevis, M. parodii and R. apiifolius produced abundant and diverse phytolith morphotypes. The rest of the species produced very few (L. peploides, P. hydropiperoides, A. philoxeroides) or no identifiable phytoliths (T. latifolia, R. crispus). The aim of this study was to compare the pharmacokinetics of baicalin and wogonoside in rats following oral administration of Xiaochaihu Tang (Minor Radix Bupleuri Decoction) and Radix scutellariae extract. Thus, a specific LC–MS method was developed and validated for the determination of these flavonoids in the plasma of rats after oral administration Xiaochaihu Tang and Radix scutellariae extract. Chromatographic separation was performed on a Zorbax SB C18 column (150 mm × 4.6 mm, i.d.: 5 μm) with 0.1% formic acid in water and acetonitrile by linear gradient elution. Baicalin, wogonoside and carbamazepine (internal standard, I.S.) were detected in select-ion-monitoring (SIM) mode with a positive electrospray ionization (ESI) interface. The following ions: m/z 447 for baicalin, m/z 461 for wogonoside and m/z 237 for the I.S. were used for quantitative determination. The calibration curves were linear over the concentration ranges from 0.1231 to 6.156 μg mL−1 for baicalin and 0.08832 to 4.416 μg mL−1 for wogonoside. The lower limit of detection (LLOD) based on a signal-to-noise ratio of 2 was 0.06155 μg mL−1 for baicalin and 0.04416 μg mL−1 for wogonoside. Intra-day and inter-day precisions (RSD%) were within 10% and accuracy (RE%) ranged from −6.4 to 4.4%. The extraction recovery at three QC concentrations ranged from 74.7 to 86.0% for baicalin and from 71.3 to 83.7% for wogonoside. The plasma concentrations of baicalin and wogonoside in rats at designated time periods after oral administration were successfully determined using the validated method, pharmacokinetic parameters were estimated by a non-compartment model. Following oral administration of Xiaochaihu Tang and Radix scutellariae extract, the t1/2 of baicalin was 3.60 ± 0.90 and 5.64 ± 1.67, the Cmax1 was 1.64 ± 0.99 and 5.66 ± 2.02, the tmax1 was 0.13 ± 0.05 and 0.20 ± 0.07, the Cmax2 was 2.43 ± 0.46 and 3.18 ± 1.66, and the tmax2 were 6.40 ± 1.67 and 5.66 ± 2.02, respectively. Following oral administration of Xiaochaihu Tang and Radix scutellariae extract, the t1/2 of wogonoside was 4.97 ± 1.68 and 7.71 ± 1.55, the Cmax1 was 1.39 ± 0.83 and 1.45 ± 0.37, the tmax1 was 0.21 ± 0.20 and 0.17 ± 0.01, the Cmax2 was 1.90 ± 0.55 and 1.42 ± 0.70, and the tmax2 was 5.60 ± 1.67 and 5.20 ± 1.79, respectively. A significant difference (p < 0.05) was observed for t1/2, and the elimination of baicalin and wogonoside in Xiaochaihu Tang was increased.  相似文献

The objective of this study was to investigate the effect of somatic cell count (SCC), body condition score (BCS) or lameness score on ovarian follicular growth and ovulation in dairy cows. Seventy four animals 30-80 days post-partum were monitored for all three conditions before synchronization of ovarian follicular phases by administration of gonadotrophin releasing hormone (GnRH) followed seven days later with prostaglandin F2alpha (PG). Ultrasonography of both ovaries twice daily throughout the follicular phase revealed that fewer animals with combined high SCC and lameness (4/9) ovulated compared to healthy animals (19/21; P = 0.006) or animals with only high SCC (11/11; P = 0.004) or only lameness (21/27; P = 0.06). Overall, regardless of the presence of other concurrent conditions, fewer lame cows ovulated than Non Lame animals (30/42 and 30/32; P = 0.015). Mean follicular growth and maximum follicular diameter were unaffected by any of the three conditions. However, dominant follicle growth and maximum diameter were greater in the 60 animals that ovulated compared to the 14 that did not; 1.83 ± 0.16 versus 0.96 ± 0.26 mm/day (P = 0.014) and 19.4 ± 0.4 versus 16.4 ± 1.2 mm (P = 0.003), respectively. In conclusion, lameness reduced the proportion of cows that ovulated and the synergistic effect of high SCC and lameness reduced that proportion further. However, follicular growth and maximum follicular diameter were unaffected by high SCC, low BCS or lameness.  相似文献

The evaluation of the glenohumeral joint laxity requires the estimate of displacements of the humeral head centre (HHC) with respect to the glenoid. To the authors? knowledge, several studies have been conducted to estimate HHC translations in vivo but data under anterior loading conditions has not been collected yet. Aim of this study was to develop a non-invasive experimental methodology based on magnetic resonance (MR) imaging for the in vivo evaluation of the HHC translations due to an anteriorly directed force. Fourteen asymptomatic shoulders were acquired using a horizontal open MR scanner with the subjects in the supine position both at 15° and 90° of arm abduction with and without an anterior force of 20 N applied at the HHC level. When no load was applied, from 15° to 90° of arm abduction, the HHC moved, anteriorly (1.5±1.3 mm) and superiorly (1.8±1.3 mm) while smaller displacements were observed medio-laterally (0.4±0.7 mm). Under the application of the anterior force the 3D displacement of the HHC with respect to the glenoid was 1.6±1.2 mm and 1.3 ±0.7 mm, respectively at 15° and 90° of arm abduction. The level of precision associated to the GHJ translation was less than 0.33 mm along all directions i.e. one order of magnitude smaller than the relevant translations. In conclusion, the MRI-based methodology allowed for the analysis of HHC displacements under conditions of anterior loads within an acceptable level of reliability.  相似文献
