For many genes, ray-finned fish (Actinopterygii) have two paralogous copies, where only one ortholog is present in tetrapods. The discovery of an additional, almost-complete set of Hox clusters in teleosts (zebrafish, pufferfish, medaka, and cichlid) but not in basal actinopterygian lineages (Polypterus) led to the formulation of the fish-specific genome duplication hypothesis. The phylogenetic timing of this genome duplication during the evolution of ray-finned fish is unknown, since only a few species of basal fish lineages have been investigated so far. In this study, three nuclear genes (fzd8, sox11, tyrosinase) were sequenced from sturgeons (Acipenseriformes), gars (Semionotiformes), bony tongues (Osteoglossomorpha), and a tenpounder (Elopomorpha). For these three genes, two copies have been described previously teleosts (e.g., zebrafish, pufferfish), but only one orthologous copy is found in tetrapods. Individual gene trees for these three genes and a concatenated dataset support the hypothesis that the fish-specific genome duplication event took place after the split of the Acipenseriformes and the Semionotiformes from the lineage leading to teleost fish but before the divergence of Osteoglossiformes. If these three genes were duplicated during the proposed fish-specific genome duplication event, then this event separates the species-poor early-branching lineages from the species-rich teleost lineage. The haemoglobin systems of the order Clupeiformes have been studied by several researchers in 41 species belonging to three out of its five families. Most of them were investigated in the native form using electrophoretic methods, and a few were also examined from the functional point of view. Both approaches corroborate the widespread view that acidic and basic haemoglobin components, which are structurally and functionally distinct, may be present in teleost fish. However, the former are always present, whereas the latter are often lacking, depending on the taxonomic group. Both kinds of components are found in families Clupeidae and Pristigasteridae, but only acidic ones in Engraulidae.Most electrophoretic patterns show high multiplicity, and chiefly concern the acidic components. Ontogenetic variation was described in three species. Individual variants were also observed in other species, although some of these might be due to ontogenetic variations rather than genetic polymorphism.  相似文献

Summary Proteins in sieve tube exudate from Cucumis melo L., Cucumis sativus L. and Cucurbita maxima Duch. were analysed by gel electrophoresis and isoelectric focusing. Estimated molecular weights and isoelectric points for the major and minor proteins from each plant species are presented. Electrophoresis revealed striking differences between the protein complements of exudatc from the two genera investigated. Similarly, although a few exudate proteins from the two species of Cucumis possessed identical molecular weights, several major proteins were peculiar to each species. Isoelectric focusing of proteins in exudate samples from the three plants confirmed the marked differences in their protein complements. Furthermore, focusing also revealed differences between cultivars of Cucumis sativus. Both Cucumis sativus and Cucurbita maxima possessed relatively large amounts of basic proteins; these were absent in exudate from Cucumis melo. Little is known about early development of the near-threatened bonefish (Albula vulpes), a member of superorder Elopomorpha. Members of Elopomorpha are partially defined by their synapomorphic leptocephalus larval stage, for which the nutritional requirements are not well understood. Characterizing the nutritional profile, including major nutrients (such as lipids) used for energetic processes, can help to gain a better understanding of the nutritional requirements for leptocephalus larvae. A total of 24 settlement stage A. vulpes leptocephalus larvae were collected at Long Caye Island, Belize. Samples were used to determine various biochemical characteristics including lipid class, fatty acid and glycosaminoglycan compositions. Each of these biochemical components plays a role in early developmental processes such as cellular membrane formation and is crucial for healthy development. Biochemical characteristics of settlement stage A. vulpes leptocephalus are presented in this study for the first time. The dominant lipid classes and fatty acids detected in these samples were consistent with prior studies using closely related species like the Japanese eel, indicating possible similarities in diets at this stage. In the future, similar analyses can be applied to other species that share the leptocephalus life stage to determine if nutritional requirements at this stage of development are unique to this species. The findings in this study will also help to facilitate the establishment of adequate aquaculture systems for captive bonefish, ultimately leading to improved management strategies for wild bonefish habitats.  相似文献

Of the two proteins making up volvatoxin, a mushroom toxin, the smaller, volvatoxin A2, of molecular weight 25,000, is able to synergistically increase the low neurotoxicity of crotoxin B, the larger basic component of the Crotalusdurissusterrificus neurotoxin, crotoxin. No other combination of volvatoxin and crotoxin components shows this effect. Gel filtration suggests that the stable complex is composed of one molecule of each, crotoxin B and volvatoxin A2, although highest activation may be favored by the presence of an additional molecule of crotoxin B. Upon sucrose density gradient electrophoresis, the complex of volvatoxin A2 and crotoxin B behaves as a single species of greater anodic mobility than volvatoxin A2 alone. The complex formation between these two toxin components is not a nonspecific interaction of an acidic with a basic protein, since crotoxin B is not activated, nor complexed by other proteins of similar isoelectric points to volvatoxin A2.  相似文献

The acid phosphatase secreted by the biA1 strain of the mould Aspergillus nidulans was separated into at least nine isoforms by isoelectric focusing (IEF). The components visualized by activity were predominantly acidic proteins with isoelectric points ranging from pH 4.0 to 6.5. Almost the same isoforms were secreted by strains pabaA1 and palD8 biA1. Furthermore, the isoforms secreted by strain pacA1 biA1 were not visualized by staining after IEF, indicating that these isoforms are encoded by gene pacA. Treatment of the secreted enzyme with endoglycosidase H also reduced the number of isoforms visualized by staining after IEF and enhanced the Rf (electrophoretic mobility) value of this enzyme visualized after PAGE.  相似文献

The large- and small-subunit polypeptide composition of fraction 1 protein contained in seven species of Lycopersicon and Solanum pennellii was determined by electrofocusing. The eight species of protein had large subunits composed of three polypeptides separated by about 0.05 pH unit, but there was no difference in the isoelectric points of the clusters of three polypeptides. By this criterion, no surviving mutations have appeared in the extranuclear DNA coding for the cluster of large-subunit polypeptides during a period of evolution which generated the eight species of plants. The genus Lycopersicon appears to be much younger than its sister genus Nicotiana in the family Solanaceae, where four types of polypeptide clusters have evolved. Three different small-subunit polypeptides whose isoelectric points are coded by nuclear DNA have arisen among the seven Lycopersicon species, and L. hirsutum and S. pennellii have proteins containing single polypeptides and are therefore considered older than L. chilense, L. chimielewskii, and L. parviflorum, whose proteins contain two polypeptides. L. cheesemanii, L. pimpinellifolium, and L. esculentum (and probably L. peruvianum) seem to be the most recently evolved species since their fraction 1 proteins have small subunits composed of three polypeptides.This research was supported by NSF Grant 75-07368 and Contract No. EY-76-S-03-0034, P. Sandy bottoms are a ubiquitous environment found from sea bottoms to intertidal and freshwater zones. They are inhabited by many invertebrates and vertebrates which have developed morphological and physiological adaptations to sustain life under these particular conditions. Sandy habitats exhibit three potential constraints: abrasion, hypoxia and mechanical resistance. Here, three teleost species living in sandy environments were investigated: Ammodytes tobianus (Ammodytidae), Gorgasia preclara and Heteroconger hassi (Congridae). These teleost fishes were studied for their integument and muscular systems, which are potentially subject to sand abrasion and hypoxia, respectively. Based on histochemistry and transmission electron microscopy, we found the complex mucus system of G. preclara and H. hassi consists of two types of goblet cells and one type of sacciform cell. The secretions of both species are made of complex polysaccharides. In contrast, the scaly integument of A. tobianus has only a few goblet cells and no sacciform cells. We also highlighted, by immunohistochemistry, that the epidermal cell proliferation was much higher for this latter species, potentially resulting from the high rate of sand abrasion when A. tobianus buries itself quickly in the substrate. For all species, the major muscle fibre type was revealed by histoenzymology and corresponds to fast glycolytic fibres followed by intermediate fibres with slow fibres in the lowest proportion. Ammodytes tobianus possesses the highest fast fibre proportion (about 87% for A. tobianus and 75–78% for both garden eels). Our results provide new insights into the previously poorly studied teleost species, such as G. preclara, and allow us to highlight the complex skin histology of both garden eel species. Furthermore, the previously unknown muscle typing of these three species was determined.  相似文献

Summary The innervation of the swimbladder in four different teleost species has been studied by the use of immunohistochemical methods. The teleosts examined belong to two different groups regarding their swimbladder morphology: physoclists (the cod, Gadus morhua and the goldsinny wrasse, Ctenolabrus rupestris) and physostomes (the eel, Anguilla anguilla and the rainbow trout, Salmo gairdneri). Vasoactive intestinal polypeptide-like immunoreactivity was demonstrated in nerves of the swimbladder walls of all four species, and in the gas glands of the cod and the goldsinny wrasse. Substance P-like immunoreactivity was shown in swimbladders of the cod, eel and rainbow trout but not the goldsinny wrasse. Immunoreactivity to met-enkephalin antiserum was revealed in the swimbladder walls of the eel and the goldsinny wrasse, while neurotensin-like immunoreactivity was present in the goldsinny wrasse and rainbow trout swimbladders. Neurotensin-like immunoreactivity was also seen in the gas gland of the goldsinny wrasse. 5-Hydroxytryptamine immunoreactivity was found in endocrine cells in the pneumatic duct of the eel and in the swimbladder walls of the goldsinny wrasse and the rainbow trout. In conclusion, all teleosts examined showed a very close resemblance in the peptidergic/tryptaminergic innervation of the swimbladder to that of the gut, inasmuch as the immunoreactivity present in the swimbladders always occurred in the gut of the same species.  相似文献

Forest vegetation is known to increase hillslope stability by reinforcing soil shear resistance and by influencing hydrologic conditions of soil. Although the importance of plant root systems for hillslope stability has received considerable attention in recent years, the quantification of such an effect needs more investigation. In this paper, we present a synthesis of the data gathered in the last 5 years for some species in different locations of the Alps and Prealps of Lombardy (Northern Italy) with the aim to increase our knowledge on root tensile strength and on Root Area Ratio distribution within the soil. Concerning root tensile strength we developed tensile strength–diameter relationships for eight species: green alder (Alnus viridis(Chaix) D.C.), beech (Fagus sylvatica L.), red willow (Salix purpurea L.), goat willow (Salix caprea L.), hazel (Corylus avellana L.), European ash (Fraxinus excelsior L.), Norway spruce (Picea abies (L.) Karst.) and European larch (Larix decidua Mill.). Results show a great variability among the different species and also for the same species. In general, however, root strength (in terms of tension) tends to decrease with diameter according to a power law, as observed by other Authors. Comparing the power law fitting curves for the considered species, it can be observed that they fall in a relatively narrow band, with the exception of hazel, which appears the most resistant. Concerning the evaluation of root distribution within the soil we estimated the Root Area Ratio (the ratio between the area occupied by roots in a unit area of soil) according to its depth for five species (beech, Norway spruce, European larch, mixed hazel and ash) in three locations of Lombardy. Results show that there is a great variability of root density for the same species well as for different points at the same locality. The general behaviour of root density, in any case, is to decrease with depth according to a gamma function for all the studied species. The results presented in this paper contribute to expanding the knowledge on root resistance behaviour and on root density distribution within the soil. The studied location have allowed the implementation of soil–root reinforcement models and the evaluation of the vegetation contribution to soil stability.  相似文献

Summary The present immunocytochemical study concerns the distribution of serotonin in the epidermis of three species of teleost fish. Serotonin-like immunoreactivity was found in the club cells of Heteropneustes fossilis and Carapus acus but not in those from the sea eel Conger conger. This study is the first immunocytochemical identification of serotonin in the club cells of teleost epidermis. By comparing data from the literature (Zaccone et al. 1986, 1987, 1988) regarding the occurrence of serotonin and GRP/bombesin in the exocrine sacciform gland cells of piscine skin, it is worthy mentioning here that the serotonin contained in the club cells of the species studied may have the ability to affect the pheromonal or other possible functions of these cells. The presence of serotonin in these systems has been correlated with the capacity of the exocrine glands of fish skin to secrete, ectopically, amine messengers in contrast to those produced eutopically i.e. in the neuron-paraneuron system in some vertebrates (Fujita et al. 1988).  相似文献

In cyanobacteria, plastocyanin and cytochrome c 6, the alternate donor proteins to Photosystem I, can be acidic, neutral or basic; the role of electrostatics in their interaction with photosystem I varies accordingly. In order to elucidate whether these changes in the electron donors’ properties correlate with complementary changes in the docking site of the corresponding photosystem, we have investigated the kinetics of reactions between three cytochrome c 6 with isoelectric points of 5.6, 7.0 and 9.0, with Photosystem I particles from the same three genera of cyanobacteria which provided the cytochromes. The model systems compared here thus sample the full range of charge properties observed in cytochromes c 6: acidic, basic and neutral. The rate constants and dependence on ionic strength for photosystem I reduction were distinctive for each cytochrome c 6, but independent of Photosystem I. We conclude that the specific structural features of each cytochrome c 6 dictate their different kinetic behaviours, whereas the three photosystems are relatively indiscriminate in docking with the electron donors.  相似文献

An electrophoretic procedure for the selective separation and determination of the closely similar ribosomal proteins L7 and L12 (which are specifically involved in the GTPase reactions of the ribosome) from the total protein mixture extracted from unwashed ribosomes is described. In this procedure, which takes advantage of their unusually low isoelectric points. L7 and L12 (and a few other acidic proteins) migrate into gel asanions, while the bulk of ribosomal proteins which are basic remain behind. The positions of L7 and L12 were determined with authentic, pure proteins. It was further determined by means of 2-dimensional gel electrophoresis that no other protein components present in unwashed ribosomes comigrate with the bands of L7 and L12.  相似文献

Climate change is expected to promote migration of species. In ecotones, areas of ecological tension, disturbances may provide opportunities for some migrating species to establish in otherwise competitive environments. The size of and time since disturbance may determine the establishment ability of these species. We investigated gap dynamics of an old-growth red pine (Pinus resinosa Sol. ex Aiton) forest in the Great Lakes–St. Lawrence forest in northern Ontario, Canada, a transition zone between temperate and boreal forest. We investigated the effects of gaps of different sizes and ages on tree species abundance and basal area. Our results show that tree species from the temperate forest further south, such as red maple (Acer rubrum L.), red oak (Quercus rubra L.), and white pine (Pinus strobus L.), establish more often in large, old gaps; however, tree species that have more northern distributions, such as black spruce (Picea mariana Mill.), paper birch (Betula papyrifera Marsh.), and red pine show no difference in establishment ability with gap size or age. These differences in composition could not be attributed to autogenic succession. We conclude that treefall gaps in this forest facilitate the establishment of northward migrating species, potentially providing a pathway for future forest migration in response to recent changes in climate.  相似文献

The isoelectric points of isolated cuticles from Citrus aurantium L. (3.15), Prunus armeniaca L. (3.45), and Pyrus communis L. (2.90) leaves were determined from membrane potentials. At pH values below the isoelectric point, cuticular membranes carry a net positive charge and are permselective to anions (determined using 82Br). Above the isoelectric point, they carry a net negative charge and are permselective to cations (determined using 24Na+). There are no gradients of fixed charges across the cuticular membranes as indicated by the absence of asymmetry potentials. Positive charges in the membranes originate from residues of basic amino acids of proteins or polypeptides contained in a nonextractable form within the cuticle. The exchange capacity of basic fixed groups in the cuticles of six species (Lycopersicon esculentum Mill., Capsicum annuum L. fruit cuticles, and Brassaia spec. leaf cuticles in addition to the above species) varied between 0.010 and 0.025 meq g−1 cuticle. Fixed acidic groups were donated by residues of acidic amino acids, polygalacturonic acid, and nonesterified -COOH groups of the cutin polymer. At pH 8, total cation exchange capacity as determined using 45Ca2+ varied between 0.26 (Citrus) and 0.30 (apricot) meq g−1.  相似文献

The effect of temperature and hypoxic acclimation on the haemoglobin system and intraerythrocytic organic phosphate concentrations in the South African mudfish, Labeo capensis, have been investigated. Exposure to hypoxia or increased temperature raised haemoglobin concentration and decreased NTP/Hb ratio. Temperature acclimation did not effect the oxygenation characteristics of the haemolysate or haemoglobin multiplicity, as evident from isoelectric focussing experiments that showed one cathodic (Hb I) and three anodic haemoglobins (Hb II, III and IV). Oxygen equilibria of the isolated components showed a smaller Bohr effect and lower temperature and organic phosphate sensitivities in the cathodic than in the anodic haemoglobins. Unlike the trout and eel haemoglobin systems, both the anodic and cathodic haemoglobins from L. capensis exhibited sensitivity to organic phosphates but the effect was smaller in the latter. The results indicate that oxygen transport in mudfish blood is supported by variations in the red cell organic phosphate\haemoglobin ratio and the functional differentiation between anodic and cathodic haemoglobins.  相似文献

The phospholipid and the fatty acid composition of the main phospholipid families of erythrocyte plasma membranes was studied in two species of cartilaginous fish: the common thrasher (Raja clavata L.) and the common stingray (Dasyatis pastinaca) and in three bony fish species: the scorpion fish (Scorpaena porcus L.), the smarida (Spicara flexuosa Raf.), and the horse mackerel (Trachurus mediterraneus ponticus Aleev). It was shown that in the studied fish, 70.0–80.0% of all membrane phospholipids were composed of phosphatidylcholine and phosphatidylethanolamine. Phosphatidylserine, monophosphoinositide, and sphingomyelin were minor components whose content in the erythrocyte membrane fluctuated from 3.0% to 13.0%. The fatty acid phospholipids composition was represented by a large specter of acids. From saturated acids, basic for plasma membranes are palmitic (C16: 0) and stearic (C18: 0) acids. From unsaturated acids, the larger part belongs to mono-, tetra-, penta-, and hexaenoic acids in fish phospholipids. The calculation of the double bond index and of the unsaturation coefficient showed difference in the deformation ability of erythrocyte membranes of the studied fish.  相似文献

Anglers' records of fish weight (ARFW) were analysed in Poland for the periods 1965–1989 (successive increases in inland water pollution), and 1990–2010 (considerable improvements in water quality). For the initial ARFW analysis, the entire study was divided into nine intervals of 5 years each; the period that followed was divided into three catchments: the Vistula, Oder and Pomeranian rivers, whereby 10 species were chosen to represent obligatory riverine and facultative species. With a few exceptions, the investigated species showed unexpected and varying reactions to the changes in water quality. The ARFW of obligatory riverine barbel Barbus barbus (L.) decreased during the first periods (1965–1989), then increased significantly from 1990 to 2010. For obligatory riverine ide Leuciscus idus (L.), tendencies were less clear but as significant. However, other obligatory riverine species have not shown a relationship between ARFW and water quality. Discrepancies were also recorded in the facultative riverine pike (Esox lucius L.) and brown trout (Salmo trutta L.). Considering the number of medals recorded, there were some interesting tendencies related to the decline or improvement in the water quality, although not in all investigated species. The species displaying a decrease in medals awarded from 1966 to 1989 and increasing after 1990, was chub Leuciscus cephalus (L.), but only until 2005. Species which showed successive increases in awards from 1989 to 2010 were: pike, perch (Perca fluviatilis L.), ide, and wels (Silurus glanis L.). However, for species in the period when the water quality actually deteriorated (1966–1990), the number of medals awarded increased: barbel, brown trout (S. trutta L.), and grayling Thymallus thymallus (L.).  相似文献

The ricefield eel (Monopterus albus Zuiew), a burrowing eel-like synbranchoid teleost, undergoes a natural sex change from female to male during its life history. Since the teleost pineal gland and its melatoninergic output have been suggested as regulators in seasonal reproduction and sexual maturation in many fish species, it is reasonable to postulate that melatonin may play important roles in the ricefield eel’s sex-change process. This hypothesis was tested by examining secretional characteristics and reproductive effects of melatonin in the ricefield eel. Results indicate that serum melatonin (mainly secreted from the pineal complex, retinae and gastrointestinal tract) is involved in sex change of this species. It seems that, within a reproductive cycle, relatively lower mid-night serum melatonin (MNSM) levels are necessary for natural spawning, but relatively higher MNSM levels after spawning permit initiation of the sex-change process. A putative model is presented to clarify the involvement of melatonin in natural sex change of the ricefield eel, although the precise mechanisms are still under further investigation.  相似文献

An acidic lethal protein, Mojave toxin, has been isolated from the venom of Crotalus scutulatus scutulatus. The purified toxin had an i.v. LD50 of 0.056 μg/g in white mice. Disc polycrylamide gel electrophoresis at pH values of 9.6 and 3.8 and isoelectric focusing in polyacrylamide gels with a pH 3.5–10 Ampholyte gradient were used to establish the presence of one major protein band. The pI of the most abundant form of the toxin was determined to be 5.5 by polyacrylamide gel isoelectric focusing experiments. The molecular weight was established to be 24,310 daltons from amino acid composition data. Mojave toxin was shown to consist of two subunits, one acidic and one basic with isoelectric point (pI) values of 3.6 and 9.6, respectively. Amino acid analyses established molecular weights of 9593 for the acidic component and 14,673 for the basic component. The acidic subunit consisted of three peptide chains intermolecularly linked by cystine residues. The basic subunit was a single polypeptide chain with six intramolecular disulfide bonds. The basic subunit was lethal to test animals with an intravenous LD50 of 0.58 μg/g. Following recombination of the subunits a recombinant toxin was isolated which was identical to the native toxin by comparisons of electrophoretic mobility and toxicities. Comparisons of circular dichroism spectra also indicated reassociation to the native toxin structure. Phospholytic activity was associated with Mojave toxin and the basic subunit was responsible for this enzymic activity. Phospholipase activity of the basic subunit was inhibited by addition of the acidic subunit.  相似文献
