The concentration and composition of seston were measured in the east basin of Lake Erie (U.S.A.–Canada) to determine whether the classical patterns of nearshore versus offshore zonation considered typical of large lakes have been altered by decades of change in nutrient loading and food web structure, and to test the possibility that food quality for seston consumers, as reflected in nutrient and lipid composition, may currently be diminished. Chlorophyll‐a (chl‐a), particulate organic carbon (POC), particulate P, total lipid (TL), lipid classes, fatty acids and major phytoplankton groups were compared between the nearshore (<20 m) and offshore zones in spring and early summer. 2. Particulate organic carbon, chl‐a and TL concentrations were lower nearshore than offshore, which is a reversal of the classical large lake pattern but consistent with other evidence from the Great Lakes that colonization by filter‐feeding dreissenid mussels can cause seston depletion in relatively shallow waters. 3. Terrestrial and bacterial fatty acid markers were slightly higher in the nearshore than the offshore, but there was little difference in nutrient, taxonomic or biochemical composition between zones otherwise. This shows a small differential influence of terrestrial and detrital inputs, and a slight shift away from phytoplankton dominance of seston composition in the nearshore, but little difference in potential food quality of seston. 4. Microscope counts, phytoplankton fluorescence, and fatty acid composition indicated succession from diatom dominance in May to dominance by cryptophyte and chromophyte flagellates in June and July. Depletion of dissolved P and Si, increased contribution of storage lipids, and changes in fatty acid composition suggested a mild degree of nutrient deficiency during this spring to summer transition. 5. Total lipids were dominated by classes typical of pigments and membranes with only moderate amounts of neutral (storage) lipids, polyunsaturated fatty acids (PUFA) were abundant (especially the long chain ω‐3 acids) and the POC : chl‐a and POC : particulate P ratios indicated P sufficiency or slight P‐deficiency. Despite concerns that some important consumers in the Great Lakes food web may be suffering from diminished food quality, these results point to a generally high seston food quality even with highly abundant mussels and incipient P deficiency in the east basin of Lake Erie.  相似文献

The effect of slaughter weight and breed on the composition of intramuscular fat (IMF) of seven Spanish cattle breeds was studied. Lipid classes and fatty acids were evaluated in young bulls of seven local Spanish cattle breeds, Asturiana, Avileña, Morucha, Parda Alpina, Pirenaica, Retinta and Rubia Gallega, slaughtered at a live weight of 320 kg (veal type) and 550 kg (yearling bull type). Higher slaughter weight and early maturating breeds produced higher IMF content in the longissimus muscle (P < 0.001), which was linked to a reduction in phospholipids, monoglycerides, diglycerides, cholesterol and free-fatty acids content, but an increase in triacylglycerols. Besides, heavier animals displayed a higher percentage of saturated and monounsaturated fatty acids (MUFA), but a lower polyunsaturated fatty acids (PUFA) percentage (P < 0.001). The increase in triglycerides (TG), saturated fatty acids (SFA) and MUFA illustrates the increasing importance of lipid storage as fattening proceeds and the acquisition of ruminal functionality. The feeding system based on cereals had a strong influence on the fatty acid profile, giving the studied beef a relatively high PUFA/SFA and n-6/n-3 ratios.  相似文献

Two feeding experiments were conducted to evaluate the suitability of two commercial microdiets as a complementary food for the rearing of larval gilthead seabream Sparus aurata . The effect of these diets on growth, survival and total lipid composition, as well as the histological structure of the liver were examined. The results of the first trial highlighted water quality as an important factor for the survival of microdiet-fed larvae. An increase in water flow rate improved larval survival despite a reduction in live food supply. An effect of tank design on larval growth and survival was observed also and seems to be related with the light level inside the tanks; wider light grey tanks gave better results than narrower black ones. Microdiet feeding significantly improved larval growth and increased hepatocyte diameter and presence of PAS positive vacuoles suggesting an increased glycogen storage in comparison with that of larvae fed live prey only. Artificial diets provided the larvae with a higher amount of lipids and polyunsaturated fatty acids of the n-3 series, resulting in a higher DHA content in the total lipid of the larvae fed on microdiets.  相似文献

Organophosphatic shells of the brachiopod Lingula squarniformis , collected from Scottish Lower Carboniferous shales and mudstones of intertidal to sublittoral provenance, have been studied to ascertain chemico-structural changes resulting from fossilization. Enough original shell has been preserved at ultrastructural and molecular levels to confirm that Carboniferous and Recent integuments are homologous with stratiform successions of apatitic to organic laminae forming rhythmic sets. One of the main organic constituents, the acidic, hydrophilic gel glycosaminoglycans (GAGs), is the dominant component towards the tops of rhythms. During fossilization of the Carboniferous shells, GAGs degraded incrementally without disturbing apatitic ultrastructures, and the spaces so created became partly filled with sheets of recrystal-lized apatite with some kaolinite or with books and plates of kaolinite. The kaolinite in the shells contrasts with the illite of the entombing sediments and suggests that degrading acidic GAGs mediated in clay formation in situ . The sediments also contain framboidal pyrite, which is virtually absent from the shells themselves but is usually even more abundant, with a greater range of trace metals, in the sedimentary fills of complete shells. This imbalance suggests mediation by another gel, the glycocalyx, secreted by the inner epithelium of the brachiopod mantle. The glycocalyx would have lined the shell interior and could have served as a sorption film for dissolved metals precipitated as compounds on decomposition of body tissue.  相似文献

As part of the Keewatin Health Assessment Study, a comprehensive health interview and examination survey of Inuit and non-Inuit in the central Canadian Arctic during 1990–91, plasma samples were analyzed for phospholipid fatty acid composition. Compared to non-Inuit, the Inuit have reduced levels of dihomo-gamma-linoleic (DGLA) and arachidonic acid (ratios of 0.41 and 0.46) and the sum of all n-6 fatty acids (ratio of 0.65), but increased level of eicosapentaenoic (EPA) acid (ratio of 1.37). These trends are consistent with those reported from other circumpolar Inuit populations, especially the reduced arachidonic acid and increased EPA, although the Inuit excess in EPA is much less pronounced due to the greater importance of caribou rather than sea mammals in most of the Keewatin communities. The high linoleic/arachidonic acid ratio suggests increased inhibition of the metabolic pathway regulated by the enzyme Δ-5 desaturase, which can be explained by the presence of high levels of highly unsaturated fatty acids of dietary origin, and/or a genetic deficiency. In multiple linear regression models with the independent variable list consisting of Inuit status, age, sex, education, physical activity, spending time on the land and consumption of wild meat and local fish, Inuit status is independently associated with lower levels of the n-6 acids but not the n-3 acids. This indicates that factors other than diet and lifestyle, perhaps genetic ones, may account for the observed “ethnic” differences. However, for those fatty acids in which Inuit differ from non-Inuit, there is no dose-response relationship in terms of self-reported degree of non-Inuit admixture. Dietary fatty acids play an important role in the risk of cardiovascular diseases and diabetes, diseases of increasing importance in the health transition experienced by the Inuit. Association studies of plasma fatty acids and DNA markers of candidate genes for atherosclerosis and insulin resistance may provide a clearer picture of the genetic basis for the observed differences in plasma fatty acid composition between Inuit and non-Inuit. The developmental time and survival to eclosion of Anopheles hermsi Barr & Guptavanij fed monotypic and mixed diets of ten food types were examined in laboratory studies. Larvae fed monotypic diets containing animal detritus (freeze‐dried rotifers, freeze‐dried Daphnia pulicaria, and TetraMin® fish food flakes) and the mixotrophic protistan Cryptomonas ovata developed faster and survived better than larvae that were fed other monotypic diets. Survival to adulthood of larvae fed several concentrations of the diatom Planothidium (=Achnanthes) lanceolatum was poor (<13%) and larval development time was approximately twice that of larvae fed TetraMin® fish food flakes, the standard laboratory diet. Larvae fed monotypic diets containing prokaryotes (bacteria [Bacillus cereus] and cyanobacteria [Oscillatoria prolifera]) and brewer's yeast (Saccharomyces cerevisiae) failed to survive beyond the 1st and 2nd instar, respectively. Larvae fed only chlorophytes, single‐celled Chlamydomonas reinhardtii and filamentous Spirogyra communis, failed to complete larval development, regardless of the concentration tested. Cohorts fed a combination of food types (mixed diets) usually developed better than cohorts fed monotypic diets. Food types that failed to support complete development when fed alone often facilitated development to adulthood when fed in combination with food types containing >1% C20 polyunsaturated fatty acids as total fat, but regardless of essential fatty acid content, algae that produced mucilage and filaments that sank out of the feeding zone were poor quality diets.  相似文献

Tobacco cell suspensions were grown under controlled conditions to determine whether temperature aftected the fatty acid pattern of the cellular lipids. At any temperature ranging between 17° and 35°, the total fatty acid content and the levels of fatty acids or individual lipids varied during the growth period, The optimum temperature for lipid biosynthesis and polyunsaturated fatty acid accumulation was between 20° and 26°. Increase in the level of polyunsaturated fatty acids was associated with lower temperatures during the active cell division period.  相似文献

Wound healing is a dynamic process comprising multiple events, such as inflammation, re-epithelialization, and tissue remodeling. Re-epithelialization phase is characterized by the engagement of several cell populations, mainly of keratinocytes that sequentially go through cycles of migration, proliferation, and differentiation to restore skin functions. Troubles can arise during the re-epithelialization phase of skin wound healing particularly in keratinocyte migration, resulting in chronic non-healing lesions, which represent a serious clinical problem. Over the last decades, the efforts aimed to find new pharmacological approaches for wound care were made, yet almost all current therapeutic strategies used remain inadequate or even ineffective. As such, it is crucial to identify new drugs that can enable a proper regeneration of the epithelium in wounded skin. Here, we have investigated the effects of the fibrinolytic drug mesoglycan, a glycosaminoglycans mixture derived from porcine intestinal mucosa on HaCaT human keratinocytes that were used as in vitro experimental model of skin re-epithelialization. We found that mesoglycan induces keratinocyte migration and early differentiation by triggering the syndecan-4/PKCα pathway and that these effects were at least in part, because of the formation of the annexin A1/S100A11 complex. Our data suggest that mesoglycan may be useful as a new pro-healing drug for skin wound care.  相似文献

Dietary intake of long-chain n-3 PUFA is now widely advised for public health and in medical practice. However, PUFA are highly prone to oxidation, producing potentially deleterious 4-hydroxy-2-alkenals. Even so, the impact of consuming oxidized n-3 PUFA on metabolic oxidative stress and inflammation is poorly described. We therefore studied such effects and hypothesized the involvement of the intestinal absorption of 4-hydroxy-2-hexenal (4-HHE), an oxidized n-3 PUFA end-product. In vivo, four groups of mice were fed for 8 weeks high-fat diets containing moderately oxidized or unoxidized n-3 PUFA. Other mice were orally administered 4-HHE and euthanized postprandially versus baseline mice. In vitro, human intestinal Caco-2/TC7 cells were incubated with 4-hydroxy-2-alkenals. Oxidized diets increased 4-HHE plasma levels in mice (up to 5-fold, P < 0.01) compared with unoxidized diets. Oxidized diets enhanced plasma inflammatory markers and activation of nuclear factor kappaB (NF-κB) in the small intestine along with decreasing Paneth cell number (up to -19% in the duodenum). Both in vivo and in vitro, intestinal absorption of 4-HHE was associated with formation of 4-HHE-protein adducts and increased expression of glutathione peroxidase 2 (GPx2) and glucose-regulated protein 78 (GRP78). Consumption of oxidized n-3 PUFA results in 4-HHE accumulation in blood after its intestinal absorption and triggers oxidative stress and inflammation in the upper intestine.  相似文献

Military training activities can result in the deposition of depleted uranium (DU) into surface soils. Mechanisms of introduction include the generation of dust from firing and impact as well as the eventual corrosion of projectile fragments and unexploded ordnance. Microorganisms in surface soils have the potential to affect the transport of DU by direct binding of the metal to the cell surface, by altering near field soil chemistry that affects metal solubility, and by microbially influenced corrosion. We investigated the response (in terms of community composition) of a native soil microbiota to the presence of DU in an arid environment. Bacteria in soils outside of the test area were challenged in dilute media with “yellow cake” or U 3 O 8 . At concentrations of 200 to 20,000 mg L? 1 only species of Bacillus were identified. In situ characterizations (by PLFA analysis) of exposed site soils showed an enrichment in sulfate reducing bacterial (i17:1w7c up 39%) and Bacillus species (a15:0 up 35%) biomarkers. Three types of microbial communities were defined (as PLFA profiles) using exploratory statistics and related to three different levels of DU exposure. The community types were then statistically corresponded to site soil chemistry. Observed differences in site soil chemistry were attributed to munitions firing since enrichments (unexposed to exposed) were observed in the minerals magnesium (increase of ~ 18 mg kg?1 ), potassium (increase of ~ 46 mg kg?1 ) and sulfur (increase of ~ 12 mg kg?1 ), all constituents of munitions residues. Increased concentrations of these minerals corresponded with a community type that was associated with an area of extensive DU round use.  相似文献

Niger seeds (Guizotia abyssinica Cass.), which are of interest as a new source of vegetable oils, were subjected to Soxhlet-extraction with n-hexane and the extract analysed using a combination of CC, GC, TLC and normal-phase HPLC. The total lipid content was ca. 300 mg/g seed material, and the fatty acid profile showed a high content of linoleic acid (up to 63%) together with palmitic acid (17%), oleic acid (ca. 11%), and stearic acid (ca. 7%). CC separation over silica gel eluted with solvents of increasing polarity yielded 291 mg/g of neutral lipids, 5.76 mg/g of glycolipids, and 0.84 mg/g of phospholipids. GC analysis showed that the major fatty acid present in all lipid classes was linoleic acid together with minor amounts of palmitic, oleic and stearic acids. Polar lipid fractions, however, were characterised by higher levels of palmitic acid and a lower content of linoleic acid. Phospholipid classes separated by normal-phase HPLC consisted of phosphatidylcholine (ca. 49%), phosphatidylethanolamine (22%), phosphatidylinositol (14%), phosphatidylserine (ca. 8%), and minor amounts (2-3%) of phosphatidylglycerol and lysophosphatidylcholine.  相似文献

Larvae of the three important Central American malaria vectors, Anopheles albimanus, An. vestitipennis, and An. darlingi, are found in distinctly different habitats broadly defined by hydrology and aquatic vegetation, but little is known about the actual food quality and quantity of these habitats. Polyunsaturated fatty acids (PUFA) are of special interest, because mosquitoes require 20:5ω3 (EPA), 20:4ω6 (ARA), and 22:6ω3 (DHA) and without an adequate supply of these PUFAs they are not able to complete their life cycle. We collected samples of larvae and their corresponding habitats and analyzed their fatty acid (FA) composition to reveal if there are any species‐specific and habitat‐specific differences in FA composition, and if habitat FA differences can be linked to differences in the mosquito FA pattern and, ultimately, mosquito performance. We also assessed how FA of wild larvae compare to the laboratory‐reared larvae. Habitats were generally low in essential PUFAs and there were no significant differences among the FA composition of habitat samples. There were significant differences in FA composition of larvae. An. darlingi contained significantly higher amounts of FA, specifically a higher content of ω‐6 PUFA, represented mainly by the linoleic acid (18:2ω‐6). Large differences were found between field‐collected and laboratory‐reared An. vestitipennis larvae, especially in the content of PUFAs. The laboratory‐reared larvae contained significantly more of the total FA, ω3 PUFA, and MUFA. The laboratory‐reared larvae contained three to five times more essential PUFAs, EPA, and DHA. However, there were no differences in the total dry weight of the 4th instar larvae between the wild vs laboratory‐reared larvae. Total FA in both larvae and habitats of An. albimanus and An. darlingi were positively correlated with the concentration of particulate organic carbon and nitrogen (POC, PON) in their respective habitats, but no such correlation was found for An. vestitipennis. PUFA are a good indicator of nutritional quality, although factors controlling the success of anopheline development from larval habitats are likely to be more complex and would include the presence of predators, pathogens, and toxins as interacting factors.  相似文献

Finishing late-maturing bulls on grass may alter the antioxidant/prooxidant balance leading to beef with higher susceptibility to lipid oxidation and a lower colour stability compared to bulls finished on cereal concentrates. In this context, lipid oxidation and colour stability of beef from late-maturing bulls finished on pasture, with or without concentrate supplements, or indoors on concentrate was assessed. Charolais or Limousin sired bulls (n = 48) were assigned to four production systems: (1) pasture only (P), (2) pasture plus 25% dietary DM intake as barley-based concentrate (PC25), (3) pasture plus 50% dietary DM intake as barley-based concentrate (PC50) or (4) a barley-based concentrate ration (C). Following slaughter and postmortem ageing, M. Longissimus thoracis et lumborum was subjected to simulated retail display (4°C, 1000 lux for 12 h out of 24 h) for 3, 7, 10 and 14 days in modified atmosphere packs (O2 : CO2; 80 : 20). Lipid oxidation was determined using the 2-thiobarbituric acid-reactive substances assay; α-tocopherol was determined by HPLC; fatty acid methyl esters were determined using Gas Chromatography. Using a randomised complete block design, treatment means were compared by either ANOVA or repeated measures ANOVA using the MIXED procedure of SAS. Total polyunsaturated fatty acid (PUFA) concentrations were not affected by treatment, n-3 PUFAs were higher (P < 0.001) and the ratio of n-6 to n-3 PUFAs was lower (P < 0.001) in muscle from P, PC25 and PC50 compared to C. α-Tocopherol concentration was higher in muscle from P compared to PC50 and C bulls (P = 0.001) and decreased (P < 0.001) in all samples by day 14. Lipid oxidation was higher in muscle from C compared to P bulls on day 10 and day 14 of storage (P < 0.01). Finishing on pasture without supplementation did not affect beef colour stability and led to lower lipid oxidation, possibly due to the higher α-tocopherol concentration compared to concentrate finished beef.  相似文献

This study was undertaken to provide a thorough analysis of the neutral lipid (NL) and polar lipid (PL) fractions of horse meat that included the content and distribution of acyl and alkenyl moieties in foals under different rearing conditions. Two groups of crossbred horses were studied; the first group was selected from suckling foals produced under grazing conditions and slaughtered at 4 months of age (n=8), and the second group was selected from concentrate-finished foals and slaughtered at 12 months of age (n=7). There were significant differences related to the age and feeding practices of foals which affected the intramuscular (IM) fat content and the fatty acid (FA) composition of NL and PL fractions. Samples from suckling foals were leaner and provided the highest content of methylation products from the plasmalogenic lipids, and total and n-3 polyunsaturated fatty acid (PUFA). By contrast, the meat from concentrate-finished foals had a higher IM fat level resulting in a greater accumulation of 16:0 and total monounsaturated FAs in the NL fraction, whereas the muscle PL fraction retained a similar FA composition between both groups. Linolenic acid was preferentially deposited in the NL fraction, but linoleic acid and the long-chain n-3 and n-6 PUFAs were incorporated into the PL fraction where they served as cell membrane constituents and in eicosanoid formation.  相似文献

Oxygen free radicals damage cells through peroxidation of membrane lipids. Gastrointestinal mucosal membranes were found to be resistant to in vitro lipid peroxidation as judged by malonaldehyde and conjugated diene production and arachidonic acid depletion. The factor responsible for this in this membrane was isolated and chemically characterised as the nonesterified fatty acids (NEFA), specifically monounsaturated fatty acid, oleic acid. Authentic fatty acids when tested in vitro using liver microsomes showed similar inhibition. The possible mechanism by which NEFA inhibit peroxidation is through iron chelation and iron-fatty acid complex is incapable of inducing peroxidation. Free radicals generated independent of iron was found to induce peroxidaton of mucosal membranes. Gastrointestinal mucosal membranes were found to contain unusually large amount of NEFA. Circulating albumin is known to contain NEFA which was found to inhibit iron induced peroxidation whereas fatty acid free albumin did not have any effect. Addition of individual fatty acids to this albumin restored its inhibitory capacity among which monounsaturated fatty acids were more effective. These studies have shown that iron induced lipid peroxidation damage is prevented by the presence of nonesterified fatty acids.  相似文献

Rotifers (Brachionus plicatilis), maintained on baker's yeast, were fed for 24h upon two algal diets, Isochrysis galbana (diet A) and Isochrysis galbana + Nannochloropsis gaditana (diet B). (These algal diets were selected for their potential use as essential fatty acid (EFA) boosters, taking into account the requirements of fish larvae). The effect of these algal diets on total lipid content, lipid classes and fatty acid composition was studied. The total lipid content increased after feeding upon both diets but no significant differences were found between the two types. Neutral lipid and polar lipid contents increased and a positive correlation was observed between the neutral lipids content of rotifers and that of the food supplied. However, the content of polar lipids in rotifers did not depend upon that of the diet. The increase in neutral lipid content was found to be higher in rotifers fed upon diet B, compared to diet A which increased the phospholipid content. Non-enriched rotifers contained only small amounts of polyenoic fatty acids, i.e. 18:3n-6, 18:3n-3, 20:4n-6, 20:5n-3 and 22:6n-3, the contents of which increased significantly by feeding both diets. The EFA composition (20:4n-6, 20:5n-3 and 22:6n-3) of neutral lipids and phopholipids in rotifers reflected the EFA composition of each diet. Diet B-fed rotifers had the highest content in 20:4n-6 and 20:5n-3, whereas rotifers fed diet A and the highest 22:6n-3 content. The mixed diet I. galbana + N. gaditana enhanced substantially the composition of lipid classes i.e. neutral lipids and of n-3 PUFA of rotifers in comparison with Isochrysis or yeast diets.  相似文献
