Chinese bayberry (Myrica rubra Sieb. & Zucc.) is one of the most important fruit tree crops in southern China. This paper reports the development of microsatellite
primers for Chinese bayberry. By screening the expressed sequence tag database and other sources, 11 polymorphic loci were
generated and primers were designed. Polymorphism of these 11 loci was assessed in 32 cultivars. All the 11 loci are polymorphic
and the number of alleles (A) ranged from 3 to 12, observed heterozygosity (H
O) and expected heterozygosity (H
E) ranged from 0.1250 to 0.9667 and 0.2359 to 0.8790, respectively, polymorphism information content ranged from 0.2285 to
0.8516. These microsatellite loci should be useful in the studies of genetic diversity of Myrica rubra and will provide useful implications for resource conservation. 相似文献

Characterization of new microsatellite loci in Pieris amamioshimensis (Ericaceae), a species nearly extinct in the wild 下载免费PDF全文
A set of 11 polymorphic microsatellite markers has been developed and characterized for the critically endangered species Pieris amamioshimensis. Fifty‐nine individuals of an ex‐situ population were used to identify these markers. The total number of alleles for each locus ranged from 3 to 9, with an average of 5.4. The expected heterozygosities (HS) and observed heterozygosities (HO) ranged from 0.47 to 0.77 and 0.22 to 0.88, respectively. In total, four loci exhibited significant deviations from Hardy–Weinberg equilibrium: two loci showed significant heterozygosity excess and the other two loci showed significant heterozygosity deficit. The polymorphism information content (0.43 ≤ PIC ≤ 0.73), the probability of exclusions (PE1 = 0.9565, PE2 = 0.9969 and PE3 = 0.9999) and probabilities for identity (PI = 3.78 × 10?9 and PI‐Sib = 2.35 × 10?4) suggest that these markers are useful for estimating not only genetic diversity but also parentage, for the ex‐situ conservation management of populations. 相似文献

We characterize five polymorphic microsatellite loci for the Great Plains toad, Bufo cognatus, which are being used as markers in a study of the genetics and structure of toad populations inhabiting playa lakes on the Llano Estacado plateau of Texas. Polymorphic microsatellite markers were developed for European hazelnut (Corylus
avellana L.) from the sequences of inter-simple sequence repeat (ISSR) fragments and flanking regions. Twenty-five ISSR primers were
used to generate fragments for cloning. Of the 520 unique sequences obtained, 41 contained long internal repeats (≥20 bp)
with flanking sequences sufficient for primer design. From these, we developed 23 new polymorphic microsatellite loci. The
flanking sequences were obtained for fragment ends by chromosome walking, and an additional 47 polymorphic markers were developed.
Two additional polymorphic markers were developed from a GA-enriched library. The 72 new marker loci were characterized using
50 diverse hazelnut accessions. For the internal repeat loci, the number of alleles per locus ranged from 2 to 16, with a
mean of 7.52. Mean values for expected heterozygosity (He), observed heterozygosity (Ho), and polymorphism information content (PIC) were 0.62, 0.59, and 0.58, respectively. The estimated frequency of null alleles
(r) was ≥0.05 at six of the 23 loci. For the 47 marker loci developed from fragment ends, the number of alleles per locus ranged
from 2 to 16, with a mean of 7.30. Mean values for He, Ho, and PIC were 0.62, 0.47, and 0.58, respectively. The estimated frequency of null alleles (r) was ≥0.10 at 18 of the 47 loci. Of the 70 loci developed from ISSR and flanking sequences, 50 segregated in our mapping

The present study reports isolation and characterization of 11 polymorphic microsatellite markers for Nycticorax nycticorax. A total of 82 alleles were detected with an average of 7.5 alleles per locus. The observed heterozygosity (H
O) ranged from 0.25 to 1.00, and the expected heterozygosity (H
E) ranged from 0.51 to 0.88. Analyses revealed no evidence for Linkage disequilibrium between any two loci, and only one locus
was significantly deviated from HWE with the estimation of exact P values by the Markov chain method (P < 0.001). The 11 loci were successfully amplified in 11 other Ardeidae species. These results demonstrate these markers can

Characterization of microsatellite markers in the mosquito Ochlerotatus caspius (Diptera: Culicidae)
Seven polymorphic microsatellite loci were developed for the mosquito species Ochlerotatus caspius, using an enriched genomic library. The number of alleles per locus varied between two and 11; the expected heterozygosity (HE) ranged from 0.18 to 0.77. Here we describe the characterization of six polymorphic microsatellite loci for Lilium philadelphicum (Liliaceae). Polymorphism levels ranged from 7 to 30 alleles per locus with a mean number of 15 alleles per locus. Observed heterozygosity (HO) ranged from 0.17 to 0.82 and did not deviate from Hardy–Weinberg expectations in the three western Montana populations included in the analysis. Six polymorphic microsatellites (eight loci) were used to study the genetic diversity and population structure of common carp
from Dongting Lake (DTC), Poyang Lake (PYC), and the Yangtze River (YZC) in China. The gene diversity was high among populations
with values close to 1. The number of alleles per locus ranged from 2 to 11, and the average number of alleles among 3 populations
ranged from 6.5 to 7.9. The mean observed (H
O) and expected (H
E) heterozygosity ranged from 0.4888 to 0.5162 and from 0.7679 to 0.7708, respectively. Significant deviations from Hardy–Weinberg
Equilibrium expectation were found at majority of the loci and in all three populations in which heterozygote deficits were
apparent. The analysis of molecular variance (AMOVA) indicated that the percent of variance among populations and within populations
were 3.03 and 96.97, respectively. The Fst values between populations indicated that there were significant genetic differentiations for the common carp populations
from the Yangtze River and two largest Chinese freshwater lakes. The factors that may result in genetic divergence and significant

The utility of EST‐simple sequence repeats (EST‐SSRs) was evaluated in the fern Athyrium distentifolium. From 1152 frond cDNA clones, 165 microsatellites, including di‐, tri‐, tetra and penta‐nucleotide repeat motifs, were identified. Primer design was possible for 74 of the SSRs; subsequent screening of 10 loci on 186 individuals from six natural populations revealed between two and seven alleles per locus and expected heterozygosity (HE) estimates ranging from 0.027 to 0.809. Eight of these loci were further examined for cross‐species and cross‐generic amplification in other Woodsiaceae species, and polymorphic products were detected. EST‐derived SSRs provide robust, informative and potentially transferable polymorphic markers suitable for biodiversity research. 相似文献

Seven microsatellite loci were isolated and characterized from the red‐capped robin Petroica goodenovii, using nonradioactive polymerase chain reaction (PCR)‐based techniques to screen an enriched genomic library. Five loci showed no evidence of null alleles and were variable [mean heterozygosity (HE) = 0.440, mean number of alleles = 8]. Cross‐amplification using primers for microsatellites in Phylloscopus occipitalis and Emberiza schoeniclus yielded another two polymorphic loci. The combined set of five red‐capped robin and two cross‐amplified loci are suitable for paternity assignment (exclusion probability for seven unlinked loci = 0.9760). 相似文献

Microsatellites were isolated and characterized for Japanese larch, Larix kaempferi, a conifer species distributed in Japan. A larch genomic DNA library enriched for (AG)n repeats was screened using the colony polymerase chain reaction method and 145 unique microsatellite containing sequences were obtained. Seventy‐two primer pairs were designed and 30 produced single‐locus products, and 19 of them were polymorphic. The expected heterozygosity ranged from 0.566 to 0.951. We developed microsatellite loci for the southern pine beetle (Dendroctonus frontalis). Twelve microsatellite loci were identified. Eight loci were polymorphic and sufficiently variable in 62 individuals (expected heterozygosity ranged from 0.707 to 0.880) to investigate population structure. All loci conformed to HWE except Dfr‐14, which showed heterozygote excess, and no two loci deviated from linkage equilibrium. The loci were tested for cross‐species amplification in four species of Dendroctonus (D. valens, D. terebrans, D. brevicomis, and D. ponderosae). deceptive orchid Ophrys fusca. Microsatellites were isolated from two partially enriched genomic libraries using FIASCO (Fast Isolation by AFLP of Sequences

loci were selected for microsatellite characterization of two Portuguese populations. Total number of alleles per locus ranged
from 10 to 32. All loci showed a high level of observed heterozygosity (H0) ranging from 0.33 to 1 and were possible to amplify in 16 other species of Ophrys using the same primers.

We have isolated and characterized seven polymorphic microsatellite loci in the white‐winged chough (Corcorax melanorhamphos), a highly social, cooperatively breeding bird of Australian eucalypt woodlands. In analyses of 100 samples from 16 family groups, the number of alleles per locus ranged from four to 18, and observed heterozygosity ranged between 0.46 and 0.93. One locus appears to be sex‐linked. The primers were also tested in apostlebirds (Struthidera cinerea), the only other species in the subfamily Corcoracinae. Five loci were successfully amplified and three were polymorphic. 相似文献

Eighteen polymorphic microsatellite loci from the giant mottled eel (Anguilla marmorata) were identified and their characteristics were described. These markers were tested in two wild populations from China (n = 20) and Australia (n = 20) respectively. The number of alleles ranged from 9 to 14 with an average of 11.28 per locus. The mean expected heterozygosity (HE) was 0.8696 in the population from China, 0.8823 in the population from Australia. We identified 14 novel polymorphic microsatellite loci in the black‐capped vireo (Vireo atricapillus). We also attempted to amplify and genotype these loci in other Vireo species, including the white‐eyed vireo (Vireo griseus), red‐eyed vireo (Vireo olivaceus), and blue‐headed vireo (Vireo solitarius). In 33 genotyped black‐capped vireos from two locations, total alleles ranged from six to 20, with observed heterozygosity ranging from 0.58 to 0.91 and expected heterozygosity from 0.65 to 0.93. Two loci had detectable levels of null alleles. Many of the loci were able to be amplified in the related Vireo species. 相似文献
