Summary Ten cultivars of tomato were screened for their ability to produce shoots and shoot primordia on media containing a range of 6-benzyladenine (BA) concentrations (0.0–4.0 mgl−1; 0.0–17.7 μM) in combination with indole-3-acetic acid (IAA; 0.0, 0.1, 0.2 mgl−1; 0.0, 0.57, 1.14 μM, respectively). Both genotypes and growth regulator levels showed significant differences at α=0.05. However, their interaction was not significantly different. A comparison of the mean number of shoots produced six cultivar classes, with UC82, UC97-3, Castlerock, Red Rock, and Peto86 producing the highest means. There were four cultiver frequency classes, the highest including cultivars Peto Rock (93.7%), Peto86 (92.3%), and Strain B (90.7%). Growth regulator mean comparisons produced eight classes, the highest included three different BA/IAA combinations, with media containing 2.0 mgl−1 (8.87 μM) BA and 0.2 mgl−1 (1.14. μM) IAA giving the highest mean and frequency (94.1%). The effect of carbon source was studied using glucose, fructose, maltose, and sucrose at 3.0% concentration. Three high regenerating genotypes and four media were used in combination with each sugar. Sugars were significantly different with maltose giving the highest number of shoots (31.6) followed by glucose (20.9). Growth regulator means, sugar-growth regulator interaction, and cultivar-sugar interaction were significant. A three-way interaction was nonsignificant. The effect of maltose and sucrose concentration (1.5–9.0%) showed the positive effect of maltose over sucrose in inducing shoots and in reducing loss rates of shoots on the regeneration media. Maltose at 1.5, 3.0, and 4.5% was the most effective. Six pH values (4.0–6.5) were used to test their effect on shoot induction in three cultivars (Peto86, UC97-3 and Castlerock). pH effects and cultivar-pH interaction were not significant.  相似文献

Variation in fruit morphology is a prevalent characteristic among cultivated tomato. The genetic and developmental mechanisms underlying similarities and differences in shape between the fruit of two elongated tomato varieties were investigated. Fruit from two F2 populations constructed from either Solanum lycopersicum cv. Howard German or cv. Banana Legs crossed with S. pimpinellifolium accession LA1589, and one BC1 population constructed with S. lycopersicum Howard German as the recurrent parent, were analysed for shape by using a new software program Tomato Analyzer. Quantitative trait loci (QTLs) controlling 15 individual shape attributes were mapped by both single and multitrait composite interval mapping in each population. In addition, principal components analysis and canonical discriminant analysis were conducted on these shape attributes to determine the greatest sources of variation among and between the populations. Individual principal components and canonical variates were subjected to QTL analysis to map regions of the genome influencing fruit shape in the cultivars. Common and unique regions, as well as previously known and novel QTLs, underlying fruit morphology in tomato were identified. Four major loci were found to control multiple fruit shape traits, principal components, and canonical variates in the populations. In addition, QTLs associated with the principal components better revealed regions of the genome that varied among populations than did the QTL associated with canonical variates. The QTL identified can be compared across additional populations of tomato and other fruit-bearing crop species.  相似文献

Two commercial cultivars of tomato, Alta and Peto 95, the accession line number LA716 of Lycopersicon pennellii and lines 94GH-006 and 94GH-033 (backcrosses between Peto 95 and LA716), with different leaf acyl sugar contents were screened for resistance to Bemisia argentifolii Bellows & Perring (corresponding to the Spanish B-biotype of Bemisia tabaci (Gennadius)), in greenhouse- and field-no-choice experiments. There was no oviposition on LA716 (with the highest acyl sugar content) while the greatest fecundity and fertility values were observed on the cultivar Alta (no acyl sugar content). However, no clear relationship was found between the low acyl sugar content in the other tomato cultivars tested and whitefly reproduction. Thus, resistance to B. tabaci did not appear to correlate with acyl sugar content below a threshold level of 37.8 microg cm-2 leaf. In a greenhouse choice-assay, B. tabaci exhibited reduced host preference and reproduction on the commercial tomato cultivars Motelle, VFN8 and Ronita all of which carry the Mi gene resistance to Meloidogyne nematodes and the aphid Macrosiphum euphorbiae (Thomas), than on the Mi-lacking cultivars Moneymaker, Rio Fuego and Roma. When data of Mi-bearing plants were pooled, the mean values for daily infestation and pupal production of B. tabaci were significantly lower than those of Mi-lacking plants. This reflected a level of antixenosis- and antibiosis-based resistance in commercial tomato and indicated that Mi, or another closely linked gene, might be implicated in a partial resistance which was not associated either with the presence of glandular trichomes or their exudates. These findings support the general hypothesis for the existence of similarities among the resistance mechanisms to whiteflies, aphids and nematodes in commercial tomato plants.  相似文献

The study of phenotypic and genetic diversity in landrace collections is important for germplasm conservation. In addition, the characterisation of very diversified materials with molecular markers offers a unique opportunity to define significant marker-trait associations of biological and agronomic interest. Here, 50 tomato landraces (mainly collected in central Italy), nine vintage and modern cultivars, and two wild outgroups were grown at two locations in central Italy and characterised for 15 morpho-physiological traits and 29 simple sequence repeat (SSR) loci. The markers were selected to include a group of loci in regions harbouring reported quantitative trait loci (QTLs) that affect fruit size and/or shape (Q-SSRs) and a group of markers that have not been mapped or shown to have a priori known linkage (NQ-SSRs). As revealed by univariate and multivariate analyses of morphological data, the landraces grouped according to vegetative and reproductive traits, with emphasis on fruit size, shape and final destination of the product. Compared to the low molecular polymorphism reported in tomato modern cultivars, our data reveal a high level of molecular diversity in landraces. Such diversity has allowed the inference of the existence of a genetic structure that was factored into the association analysis. As the proportion of significant associations is higher between the Q-SSR subset of markers and the subset of traits related to fruit size and shape than for all of the other combinations, we conclude that this approach is valid for establishing true-positive marker-trait relationships in tomato. Fruit set and fruit development in tomato is largely affected by changes in environmental conditions, therefore autonomous fruit set independent of fertilization is a highly desirable trait in tomato. Here, we report the production and characterization of male‐sterile transgenic plants that produce parthenocarpic fruits in two tomato cultivars (Micro‐Tom and Moneymaker). We generated male‐sterility using the cytotoxic gene barnase targeted to the anthers with the PsEND1 anther‐specific promoter. The ovaries of these plants grew in the absence of fertilization producing seedless, parthenocarpic fruits. Early anther ablation is essential to trigger the developing of the transgenic ovaries into fruits, in the absence of the signals usually generated during pollination and fertilization. Ovaries are fully functional and can be manually pollinated to obtain seeds. The transgenic plants obtained in the commercial cultivar Moneymaker show that the parthenocarpic development of the fruit does not have negative consequences in fruit quality. Throughout metabolomic analyses of the tomato fruits, we have identified two elite lines which showed increased levels of several health promoting metabolites and volatile compounds. Thus, early anther ablation can be considered a useful tool to promote fruit set and to obtain seedless and good quality fruits in tomato plants. Tomato is among important vegetable crops cultivated in different climates; however, heat stress can greatly affect fruit quality and overall yield. Crop reflectance measurements based on ground reflectance sensor data are reliable indicators of crop tolerance to abiotic stresses. Here, we report on using non-destructive spectral vegetation indices to monitor yield traits of 10 tomato genotypes transplanted on three different dates (Aug. 2, Sept. 3 and Oct. 1) during 2019 growing season in the Riyadh region. The ten genotypes comprised six commercial cultivars–(Pearson Improved, Strain B, Valentine, Marmande VF, Super Strain B, and Pearson early) ––and four local Saudi cultivars (Al-Ahsa, Al-Qatif, Hail and Najran). Spectral reflectance data were utilized using a FieldSpec 3 spectroradiometer in the range of 350–2500 nm to calculate nine vegetation indices (VIs): Normalized Water Band Index (NWBI), Difference Water Index (NDWI), Photochemical Reflectance Index (PRI), Normalized Difference Vegetation Index (NDVI), Green Normalized Difference Vegetation Index (GNDVI), Red Edge Normalized Difference Vegetation Index, Soil Adjusted Vegetation Index (SAVI), Red Edge Normalized Difference Vegetation Index (RENDVI), Renormalized Difference Vegetation Index (RDVI), and Normalized Difference Nitrogen Index (NDNI). VIs and yield parameters (total fruit yield, harvest index) revealed that second transplanting date was optimal for all the genotypes. Valentine showed the best growth performance followed by Najran, Hail, Super Strain B and finally Pearson early. For all the three transplanting dates, Valentine recorded the highest total fruit yield. Additionally, some genotypes had no significant differences in the VIs values or the total fruit yield between the second and third transplanting dates. This study indicated that yield parameters could be linked to rapid, non-destructive hyperspectral reflectance data to predict tomato production under heat stress.  相似文献

Tomato (Lycopersicon esculentum Mill.) plants, which normally do not accumulate glycinebetaine (GB), are susceptible to chilling stress. Exposure to temperatures below 10 degrees C causes various injuries and greatly decreases fruit set in most cultivars. We have transformed tomato (cv. Moneymaker) with a chloroplast-targeted codA gene of Arthrobacter globiformis, which encodes choline oxidase to catalyze the conversion of choline to GB. These transgenic plants express codA and synthesize choline oxidase, while accumulating GB in their leaves and reproductive organs up to 0.3 and 1.2 micromol g(-1) fresh weight (FW), respectively. Their chloroplasts contain up to 86% of total leaf GB. Over various developmental phases, from seed germination to fruit production, these GB-accumulating plants are more tolerant of chilling stress than their wild-type counterparts. During reproduction, they yield, on average, 10-30% more fruit following chilling stress. Endogenous GB contents as low as 0.1 micromol g(-1) FW are apparently sufficient to confer high levels of tolerance in tomato plants, as achieved via transformation with the codA gene. Exogenous application of either GB or H2O2 improves both chilling and oxidative tolerance concomitant with enhanced catalase activity. These moderately increased levels of H2O2 in codA transgenic plants, as a byproduct of choline oxidase-catalyzed GB synthesis, might activate the H2O2-inducible protective mechanism, resulting in improved chilling and oxidative tolerances in GB-accumulating codA transgenic plants. Thus, introducing the biosynthetic pathway of GB into tomato through metabolic engineering is an effective strategy for improving chilling tolerance.  相似文献

Abstract

So far, many QTL mapping studies in tomato have been performed for progeny obtained from crosses between two genetically distant parents, e.g., domesticated tomatoes and wild relatives. However, QTL information of quantitative traits related to yield (e.g., flower or fruit number, and total or average weight of fruits) in such intercross populations would be of limited use for breeding commercial tomato cultivars because individuals in the populations have specific genetic backgrounds underlying extremely different phenotypes between the parents such as large fruit in domesticated tomatoes and small fruit in wild relatives, which may not be reflective of the genetic variation in tomato breeding populations. In this study, we constructed F2 population derived from a cross between two commercial F1 cultivars in tomato to extract QTL information practical for tomato breeding. This cross corresponded to a four-way cross, because the four parental lines of the two F1 cultivars were considered to be the founders. We developed 2510 new expressed sequence tag (EST)-based (euchromatin-derived) genomic SSR markers and selected 262 markers from these new SSR markers and publicly available SSR markers to construct a linkage map. QTL analysis for ten agricultural traits of tomato was performed based on the phenotypes and marker genotypes of F2 plants using a flexible Bayesian method. As results, 13 QTL regions were detected for six traits by the Bayesian method developed in this study.

Tomato line LA1478 and Pusa Ruby were resistant to tomato leaf curl virus (TLCV) disease. They registered higher plant height, number of branches, total phenol content and yield per plant than the other cultivars. Variety Peto 86 was tolerant to the disease while the other popular tomato cultivars, i.e. Ace, Early Pack, Money Maker, Prichard and Strain B were highly susceptible to the disease. Plant height and number of branches per plant revealed significantly positive association with fruit yield per plant. The disease index of TLCV exhibited significant negative correlations with plant height, total phenol content and fruit yield per plant – 0–4 and 5–25 adult whiteflies were observed on resistant susceptible cultivars. In the case of epiphytically colonisation by Beauveria bassiana conidia, not all developing hyphae on the leaf surface penetrated the whitefly cuticle. Many of the germ tubes elongated to a short distance before terminating its growth. On the other hand, the rapid staining of tomato tissues injected with B. bassiana conidial suspension indicates that the entomopathogenic fungus was established inside tomato tissues until the end time of the trial. The direct injection with the spore suspension yielded high post-colonisation, where the fungus was recovered from sites distant from the point of inoculation. This indicates that the fungus has the potential to move throughout the plant tissues. Laboratory bioassay of tomato whitefly feeding on tomato tissues containing B. bassiana conidial spores indicates that plant endophytic colonisation with entomopathogenic fungi may reduce insect survival on these plants. LT50 values of the test diet were between three and four days. The mortality of Bemisia tabaci was high in the case of endophytically colonisation compared to epiphytically one (90.0% compared to 10.0% during three days) for whiteflies fed tomato tissues containing 1.5 × 107 B. bassiana spores/ml. Application of B. bassiana as an artificial endophyte inside tomato plants can be an important component in the integrated control of tomato whiteflies. The endophytic colonising can achieve biocontrol effect based on induced disease resistance in plant tissues. According the available references, this is the first report on B. tabaci controlling by plant endophytic treatment.  相似文献

Non-destructive methods have been widely recognized for evaluating fruit quality traits of many horticultural crops and food processing industry. Destructive (analytical) test, and non-destructive evaluation of the quality traits were investigated and compared for ‘Red Rose’ tomato (Solanum lycopersicum L.) fruit grown under protected environment. Fresh tomato fruit at five distinctive maturity stages namely; breaker (BK), turning (TG), pink (PK), light-red (LR), and red (RD) were labeled and scanned using the handheld near infra-red (NIR) enhanced spectrometer at a wavelength range of 285–1200 nm. The labeled tomato samples were then measured analytically for flesh firmness, lycopene, β-carotene, total phenolic content (TPC) and total flavonoids content (TFC). The results revealed that quality traits could be estimated using NIR spectroscopy with a relatively high coefficient of determination (R2): 0.834 for total phenolic content, 0.864 for lycopene, 0.790 for total flavonoid content, 0.708 for β-carotene; and 0.679 for flesh firmness. The accumulation of Lyco and β-Car rapidly increased in tomatoes harvested between the TG and the LR maturity stages. Harvesting tomatoes at BK maturity stage resulted in significantly higher flesh firmness than harvesting at the later maturity stages. Tomato fruits had the lowest TPC and TFC contents at the earliest maturity stage (BK), while they had intermediate TPC and TFC levels at LR and RD maturity stages. NIR spectroscopic measurements of fruit firmness and lipophilic antioxidants in tomato fruit at various maturity stages were partially in accordance with those estimated by destructive (analytical) methods. Based on these findings, we recommend using non-destructive NIR spectroscopy as an effective tool for predicting tomato fruit quality during harvest stage and postharvest processing.  相似文献

The salinity stress is one of the most relevant abiotic stresses that affects the agricultural production. The present study was performed to study the improvement of the salt tolerance of tomato plants which is known for their susceptibility to salt stress. The present study aimed to assess to what extent strain Azospirillum brasilense (N040) and Saccharomyces cerevisiae improve the salt tolerance to tomato plants treated with different salt concentration. The inoculant strain A. brasilense (N040) was previously adapted to survive up to 7% NaCl in the basal media. A greenhouse experiment was conducted to evaluate the effect of this inoculation on growth parameter such as: plant height, root length, fresh and dry weight, fruits fresh weight, chlorophyll content, proline and total soluble sugar in tomato plants under salt stress condition. The results revealed that co-inoculation of Azospirillum brasilense (N040) and Saccharomyces cerevisiae significantly increased the level of proline (8.63 mg/g FW) and total soluble sugar (120 mg/g FW) of leaves under salinity condition comparing to non-inoculated plants (2.3 mg/g FW and 70 mg/g FW, respectively). Plants co-inoculated with adapted strain of A. brasilense and S. cerevisiae showed the highest significant (p < 0.01) increase in fruit yield (1166.6 g/plant), plant high (115 cm) and roots length (52.6) compared whit un-inoculated control plants (42 g/pant, 43.3 cm and 29.6 cm, respectively). In contrast, Na+ ion content was significantly decreased in the leaves of salt stressed plants treated with the A. brasilense (N040) and S. cerevisiae. Finally, the results showed that dual benefits provided by both A. brasilense (N040) and S. cerevisiae can provide a major way to improve tomato yields in saline soils.  相似文献

This work aims to determine the phytochemical characterization of the pericarp of Chamaedorea radicalis Mart. fruit as a non-timber product with potential to obtain phytochemicals with potential applications in the industry. Fruit from C. radicalis were grouped in four ripening stages named as S1, S2, S3 and S4, according to maturity; S1 the most unripe stage and S4 the completely ripe stage. Determinations of total phenolic compounds, free radical scavenging activities and total flavonoid contents using spectrophotometric methods were done. Also, the tentative identification of phytochemicals during fruit ripening was done using UPLC-MS-MS. Total phenolic compound (TPC) content ranged from 7.24 to 12.53 mg gallic acid equivalents per gram of fresh weight (mg GAE/ g FW). Total flavonoids (TF) contents ranged from 0.163 to 0.23 mg of quercetin equivalents per g FW (mg QE/g FW). Free radical scavenging activity against DPPH and ABTS radicals varied from 40.80 to 53.68 and from 22.29 to 37.76 mmol Trolox equivalents g FW (mmol TE/g FW), respectively. Antioxidant assay in vitro by FRAP (ferric reducing antioxidant power) method showed that S3 was the highest level with antioxidant power while S4 was the lowest with Red ripeness stage showed the lowest contents for all determinations. Mass spectrometry allowed detection of 26 compounds, including phenolics, alkaloids and saponins. Afzelin, Kaempferol 3-neohesperidoside and the four saponins identified were present in all ripeness stages. Preliminary phytochemical identi- fication and the spectrophotometric determinations showed that the pericarp of C. radicalis presented antioxidants and compounds related to alkaloids, phenolics and saponins. The presence and abundance of each phytochemical regarding each ripeness stage should be considered.  相似文献

Quantitative Trait Locus (QTL) allelic variation was studied by analyzing near-isogenic lines (NILs) carrying homologous introgressions on chromosome 4 from three green-fruited wild tomato species. The NILs affect agronomic (yield, brix, fruit weight) and fruit (fruit shape, color, epidermal reticulation) traits in a similar manner. However, significant differences were detected in the magnitudes of the effects, the dominance deviations and epistatic interactions, indicating that those species carry different alleles for the QTL. As the QTL did not show any interaction across environments, gene-tic backgrounds or other QTLs, it can be used to introduce novel genetic variation into a broad range of cultivars. Analysis of new recombinant NILs showed that fruit traits are controlled by several linked genetic loci, whereas multiple genetic loci control the agronomic traits within the original introgression. The hypothesis that QTLs may be composed of multiple linked genes can not be rejected prior to implement projects for QTL isolation and cloning. Loci involved in color enhancement could not be related to any known gene involved in the carotenoid biosynthesis pathway, therefore it is hypothesized that the function of those loci must be related to the genetic regulation of the carotenoid biosynthetic pathway. A set of 94 peach cultivars including Spanish native peach and foreign commercial cultivars were analyzed using 15 SSR markers, selected for their high level of polymorphism. The number of alleles obtained varied from two to 11 with an average of 6.73 giving 185 different genotypes. All the cultivars showed a unique genetic profile, each one using different genotypic combination of all loci. BPPCT001 was the most informative locus showing also the highest discrimination power. Only six loci allowed the unambiguous separation of all the Spanish native cultivars studied, and the genotypic combination of only eight loci permitted the total differentiation of the 94 peach cultivars analyzed. The six selected loci (BPPCT001, BPPCT006, BPPCT008, PS9f8, UDP98-022, and UDP98-412) seem to be very useful for future Spanish peach identification works, and they will help to establish a molecular data base for native peach cultivars. UPGMA analysis was performed from the genetic distance matrix, and allowed the arrangement of all genotypes according to their genetic diversity. The genetic diversity among cultivars, observed in this work, led to their separation according to their regional origin, their morphological characteristics, and especially according to their fruit traits. Analysis of molecular variance was performed for seven populations from different regions of Spain and USA to examine the distribution of genetic variation of the studied accessions, showing that the major variation occurred within populations in each geographic site. The results reveal the existence of two diversity regions in Spain for peach germplasm.  相似文献

比较了SSR标记和形态标记在研究番茄栽培品种遗传多样性上的应用。用7对SSR引物检测了11个番茄栽培品种的遗传多样性,共得到53条带,每一个位点上扩增出2-9条带,平均为6条;品种间遗传相似系数在0.39-0.84之间,平均遗传相似系数为0.60。根据11个形态学性状表型值计算的遗传相似系数在0.27-0.72之间,平均遗传相似系数0.58。用UPGMA进行聚类分析,SSR标记和形态标记都可依果肉颜色和果实大小将供试材料分组,即黄色和红色果肉,樱桃小番茄和大、中果形的混合型;两种方法对番茄栽培品种遗传多样性的评价相近。  相似文献

Observations were made on the role of glandular leaf hairs of four tomato cultivars in trapping cotton whitefly, Bemisia tabaci, adults. Dead, trapped whitefly occurred in similar numbers on both the upper and lower surfaces of leaves even though glandular hairs were denser on the former. Females constituted 58% of untrapped but 86% of trapped adults, many becoming trapped during oviposition which occurred mainly on the lower surfaces. Of the four cultivars studied, Strain B trapped more whitefly than Ace, Marglobe or Moneymaker.  相似文献

The availability of favorable genetic diversity is a thriving vitality for the success of a breeding program. It provides a firm basis of selecting superior breeding lines for the development of high yielding crop genotypes. In this context, present investigation aimed to generate information on genetic divergence and character association in a diversity panel of 123 local and exotic soybean genotypes. Analysis of variance revealed significant response of the evaluated genotypes based on studied attributes. It depicted the probability of selecting desirable soybean genotypes by focusing on character association studies and genetic diversity analysis. Correlation analysis revealed that seed yield per plant showed high positive correlation with 100-seed weight followed by pods per plant and plant height. Furthermore, path coefficient analysis exposed that pods per plant had maximum direct contribution in seed yield per plant followed by 100-seed weight, days to flowering and SPAD measurement. Genotype named “G-10” showed maximum yield per plant followed by 24607, G-52, 24593, Arisoy, 24566, 17426, A-3127, 24570 and 24567. Genetic diversity analysis grouped the evaluated germplasm into 17 clusters. All clusters showed zero intra-cluster variability; while inter-cluster divergence ranged from 9.00 to 91.11. Cluster V showed maximum inter-cluster distance with cluster XII followed by that of between V and VIII. Moreover, cluster IV with superior genotypes (G-10, 24607, 24593 and 24566), VI (17426 and 24567), XIII (24570) and X (Arisoy and G-52) showed above mean values for most of the studied characters. Overall, the results of hybridization between the superior genotypes of these cluster pairs might be useful for soybean breeding with improving agronomic traits and adaptability.  相似文献
