Culture studies with healthy and virus-infected isolates of Ectocarpus siliculosus, Feldmannia simplex and F. irregularis gave the following results:
–  Virus particles are produced in deformed reproductive organs (sporangia or gametangia) of the hosts and are released into the surrounding seawater.
–  Their infective potential is lost after several days of storage under laboratory conditions.
–  New infections occur when gametes or spores of the host get in contact with virus particles. The virus genome enters all cells of the developing new plant via mitosis.
–  Virus expression is variable, and in many cases the viability of the host is not impaired. Infected host plants may be partly fertile and pass the infection to their daughter plants.
–  Meiosis of the host can eliminate the virus genome and generate healthy progeny.
–  The genome of the Ectocarpus virus consists of dsDNA. Meiotic segregation patterns suggest an intimate association between virus genome and host chromosomes.
–  An extra-generic host range has been demonstrated for the Ectocarpus virus.
–  Field observations suggest that virus infections in ectocarpalean algae occur on all coasts of the world, and many or all Ectocarpus and Feldmannia populations are subject to contact with virus genomes.

The present review describes the effects of light on reproductive processes in fungi, mainly when action spectra are available.The study of these has resulted in three kinds of photoresponses observable in fungi:
–  responses only to UV light (230–380 nm)
–  responses to NUV and blue light (300–520nm)
–  responses to great wavelengths of the visible spectrum. The photomorphogenetic processes on the control of these same photoreceptor pigments are reviewed.

Path analysis was used to determine the importance of long-term disturbance regime and the relative importances of correlations among vegetation patterns, disturbance history, and nitrogen (N) mineralization in old-growth forests of northwestern Minnesota. Leaf biomass (estimated by allometric equations), fire history (from fire scars on Pinus resinosa trees), and N mineralization rates (estimated from incubationsin situ) were determined from sample plots dominated by
–  Betula papyrifera, Populus tremuloides, andP. grandidentata
–  a mixture ofAcer saccharumandTilia americana, or
–  Quercus borealis andOstrya virginiana.

1.  The epithelium of the anterior intestine in the seawater eel shows very high and narrow folds whose composition and possible role in osmotic regulation were studied.
2.  The enterocytes present a basal labyrinth: numerous mitochondria more or less surrounded by infoldings of the basal membrane. In the intestinal folds, the serosal tissue contains lymphocytes, fibrocytes, enterochromaffin cells, capillaries, smooth muscle fibers and collagen fibers. The tissue is less dense at the tip than at the base of the folds.
3.  X-ray microanalyses and electrophysiological studies showed that the serosal, spaces are diffusive compartments sustaining ionic gradients. Na+ and Cl activity decreased and K+ activity increased from the base to the tip of the intestinal folds.
4.  Na+ or Cl gradients are abolished when the co-ion is removed from the bathing solutions. Serosal ouabain has no reproducible effects on the ion gradients, making it impossible to determine whether these gradients result from the activity of the Na+–K+ pump. K+ gradients decreased with serosal barium and increase with mucosal barium.
5.  The ion distribution in the serosal compartments together with ultrastructural characteristics are discussed. It is suggested that water absorption occurs involving a basal ion recycling mechanism.

This study evaluated the impact of P supply on rice plant development and the methane budget of rice fields by 2 different approaches: (1) root growth, exudation and aerenchyma formation were recorded in an experiment with hydroponic solution; (2) dissolved CH4 concentration and CH4 emission were investigated in a pot experiment. In both approaches, we used three different cultivars and three levels of P supply. In the experiment with solution culture (0.5 ppm, 5 ppm, and 10 ppm P), root exudation ranged between 0.5 to 36.7 mol C plant–1 h–1 and increased steadily with plant growth at given P level. Low P supply resulted in
•  depressed shoot growth but increased root growth in culture solution
•  increments in the root/shoot ratio by factors of 1.4 to 1.9 at flowering stage
•  enhanced the development of root aerenchyma, and
•  stimulation of root exudation per plant by factors of 1.3–1.8 as compared to medium P
•  supply and by factors of 2.1–2.4 as compared to high P supply.
However, root exudation did not differ among treatments when related to the dry weight of roots. Thus, high exudation rates were caused by larger root biomass and not by higher activity of the root tissue.The pot experiment was conducted with a P-deficient soil that was either left without amendment or fertilized by 25 and 50 mg P kg soil –1 , respectively. Low P supply resulted in
•  higher CH4 concentrations in soil solution; i.e., at flowering stage the soil solution concentrations were 34–50 M under P deficiency and 10–22 M under ample P supply and · significant increases of CH4 emission rates during the later stages of plant growth.
•  These findings reflect a chain of response mechanisms to P stress, that ultimately lead to higher methane emission rates.

The depth-time distributions of detritus depositions onto lake bottom and the amount of resident organic matter on the upper layer of the bottom have been assessed in a volcanic lake. Depositions were sampled monthly across the lake at four depths (0.2 m; 2 m; 6 m; 30 m) during two years (1983–1984). Organic and ash fractions of sediment cores collected along a depth gradient were assessed in the summer of 1984. The result show:
1.  The mean amount of organic matter deposition (size > 10 µm) is 1.24 gm–2 d–1 (dry weight) (i.e. 452.6 g m–2 year–1):
2.  The metabolism of Large Particulate Organic Matter (L.P.O.M.)) occurs primarily in the littoral zones not farther than 30 m offshore and the dispersibility of the L.P.O.M. may be predicted by a negative exponential model;
3.  The deposition on the bottom comprised under the perimeter of about 80% of the lake surface, is principally autochthonous (planktonic in origin);
4.  The detritus settled out of the water column is not completely processed and about half of the total material enters the slow cycle of the sediment on the deepest zone of the lake.

We identified three electrophoretic pathways by spectrophotometrically following the swelling of isolated yeast mitochondria:
–  --An anion uniport whose activity could only be detected after depletion of divalent cations from the matrix by treatment with 1,10-phenanthroline. This uniport was inhibited by Mg2+ and dicyclohexylcarbodiimide.
–  --A K+(Na+) uniport which was detected only when mitochondria were suspended at low pH and low temperature. This uniport was sensitive to ruthenium red and oleic acid.
–  --A K+ selective uniport which was activated by alkaline pH and ATP depletion. This pathway was sensitive to glibenclamide and to various amphiphilic cations.
Similarities and differences between these three electrophoretic pathways and the electrophoretic systems described in mammalian and plant mitochondria are discussed.  相似文献

1.  In this commentary we discuss results obtained by a micromethod for the study of Cl permeability across single nerve membranes from rabbit Deiters' neurons.
2.  These results showed the presence of GABAA receptors on the nerve cell membrane cytoplasmic side.
3.  We could show that these receptor complexes have a higher affinity for GABA than their extracellularly facing counterparts. Moreover, they present a phenomenon of desensitization. Another distinct property is that upon activation by GABA, they expose positive charges at their cytoplasmic mouths.
4.  We propose that these receptor complexes could functionin situ as a device for extruding Cl anions from the nerve cell interior. This phenomenon would create an electrochemical gradient for Cl penetration into the cell upon the action of extracellular GABA, after its presynaptic release.

We have identified a nerve carrying auditory afferents and characterized their physiological responses in the tiger beetle,Cicindela marutha.
1.  The tympana are located at the lateral margins of the first abdominal tergum. The nerve carrying the tympanal afferents is a branch of the dorsal root from the first abdominal ganglion.
2.  Both male and female auditory afferent responses are sharply tuned to 30 kHz with sensitivities of 50–55 dB SPL.
3.  The auditory afferents show little adaptation and accurately code the temporal characteristics of the stimulus with the limit of a resolution of 6–10 ms.
4.  The difference in threshold between contralateral and ipsilateral afferents for lateral stimuli is greatest at 30 kHz and is at least 10–15 dB.
5.  Ablation studies indicate that the floppy membrane in the anterolateral corner of the tympanum is crucial for transduction while the medial portion of the tympanum is less important.
6.  The tiger beetle and acridid (locust and grasshopper) ears have evolved independently from homologous peripheral structures. The neural precursor of the tympanal organs in both animals is likely the pleural chordotonal organ of the first abdominal segment.

1.  In a concentration-dependent manner neuropeptide Y was found to be a potent inhibitor of the spontaneous activation of human granulocytes and macrophages as well asMytilus edulis immunocytes.
2.  Neuropeptide Y also inhibited the chemotaxic response of these immunocytes to the chemoattractant f-MLP.
3.  Incubation of both the human and the invertebrate immunocytes in f-MLP (10–9 M) causes activation as noted by random locomotion (chemokinesis). Neuropeptide Y also blocked f-MLP-induced chemokinesis.
4.  The results suggest that neuropeptide Y may, in addition to other functions, serve as an endogenous regulator of immunocyte function.

1.  The diffusive hydrogen conductance of chicken eggshell compound membrane was measured in situ on day 16 of incubation, in a direction parallel to the shell and the chorioallantois (lateral conductance). A value of 3.9 mmol d–1 kPa–1 was obtained through a ring 13.29 cm in circumference, 0.0076 cm thick and 0.3 cm long.
2.  Lateral hydrogen conductance for 1 mm2 of shell membrane 76 m thick is 30 times the conductance of one pore serving the same area.
3.  Lateral conductance for H2 is not significantly influenced by chorioallantoic perfusion.
4.  Oxygen consumption change due to partial covering of the hen eggshell indicates that there is a significant resistance to lateral diffusion of oxygen under the shell toward the covered area.

The content of neuraminic acid (NA) of different developmental stages of trout eggs was determined.
1.  The total NA increases from about 13 g NA per egg (6–8 weeks before spawning) to 50 g directly before spawning until hatching.
2.  In freshly hatched fish larvae the NA-content is decreased to about 40 per cent as compared with stages before hatching.
3.  The ratio of bound to free NA decreases from values of about 13.5 (6–8 weeks before spawning) to 0.85–1.2 at the hatching-stage.
4.  The bound NA is almost entirely bound to sialo-glycoproteins.

3DFS is a 3D flexible searching system for lead discovery. Version 1.0 of 3DFS was published recently (Wang, T.; Zhou, J. J. Chem. Inf. Comput. Sci., 1998, 38, 71–77). Here version 1.2 represents a substantial improvement over version 1.0. There are six major changes in version 1.2 compared to version 1.0.
1.  A new rule of aromatic ring recognition.
2.  The inclusion of multiple-type atoms and chains in queries.
3.  The inclusion of more spatial constraints, especially the directions of lone pairs.
4.  The improvement of the query file format.
5.  The addition of genetic search for flexible search.
6.  An output option for generating MOLfiles of hits.

1.  Transepithelial potential differences (PDte) and acidification rates of the bath chamber were measured on isolated perfused posterior gills of the fiddler crabUca tangeri adapted to dilute seawater.
2.  The PDte decreased to almost zero when Na+ was substituted by choline or when ouabain was added to the perfusion saline in high concentrations (10 mmol·l–1). Thus, the rheogenic NaCl-transport across the gill epithelium seems to be totally Na+-dependent.
3.  When Cl was replaced by gluconate, a bath positive PDte occurred which was insensitive to ouabain. This PDte could also be observed when, in addition to Cl removal, Na+ was replaced by TMA+.
4.  Bath acidification under normal conditions could be abolished by ouabain, indicating that there is H+ excretion via electrically silent Na+/H+ exchange. In contrast, bath acidification under Cl-free conditions is only partially blocked by ouabain.
5.  It is concluded that under Cl-free conditions a rheogenic H+-pump in the apical membrane is responsible for the ouabain-insensitive bath acidification as well as for the PDte.

Being able to carry zidovudine (AZT) at known concentrations into CD4+/CD38+ and CD14+ cells permits:
–  - to reduce the drug dosage and to increase the interval for administration (until 1 dose I.V. every week);
–  - to modulate the drug concentration into the CD4+/CD38 an CD14+ cells in relation to the in vitro determined HIV sensitiveness;
–  - to eliminate haematological, medullary and general toxicity;
–  - to be able to treat severely hill patients.
Further studies are necessary in order to:
–  - To find out the better phase to start the therapy;
–  - To use several drugs with different mechanisms of action in order to slow down as much as possible the presence of resistant viral strains.
–  - As for other drugs which are beginning to be used with artificial vehicles, futher studies are required to improve the selectivity and safety of LIPOAZT for the target cells including macrophages.

1.  Coupling mechanisms between ciliary beating and the membrane potential in Paramecium were investigated under voltage clamp applying intracellular pressure injection of cAMP, cGMP and Ca-EGTA buffer. Ciliary responses following step changes in membrane potential were recorded by high-speed video on magnetic tape.
2.  Injections of cAMP and cGMP up to millimolar concentrations caused no detectable changes in the frequency voltage relationship. A minor effect was that the ciliary reorientation towards the anterior cell end (reversal) tended to be inhibited with depolarization up to 10 mV.
3.  Injection of Ca2+ into the cell clamped at the resting potential caused a transient anteriad ciliary reorientation and a simultaneous increase in the beating frequency.
4.  Injection of EGTA (to buffer Ca2+ below 10–8 M) was ineffective in relation to frequency for several minutes. After this time, hyperpolarization- and depolarization activated frequency responses of EGTA-injected cells were increasingly inhibited. The ciliary reorientation following depolarization was not affected by EGTA.
5.  A posterior contraction of the cell diameter was noticed upon membrane hyperpolarization. The contraction coincided in time with the increase in beating frequency.
6.  The results support the view that the voltage-dependent augmentation of the ciliary beating rate is not directly mediated by an intracellular increase in either cAMP or cGMP.
7.  The role of Ca2+ as intracellular messenger in the ciliary and somatic compartments is discussed.

Thiolutin was found to inhibit the utilization of glucose and other growth substrates in Escherichia coli. The inhibition was detected by a sharp drop of the respiration rate after addition of the antibiotic. The actual function affected was allocated to the cytoplasmic membrane of the bacterial cells by the following evidence:
–  - spheroplasts were affected like intact cells,
–  - individual reactions of either the electron transport chain or the glycolytic pathway were not inhibited,
–  - glucose consumption in the culture stopped and the cells accumulated guanosine tetraphosphate as under starvation conditions,
–  - activation of the cell's apo-glucose dehydrogenase restored respiration via bypassing the glucose phosphotransferase system.
It was concluded that the transport of certain substrates across the membrane was inhibited.  相似文献

1.  Umbrellar fragments of the leptomedusaCampanularia johnstoni with or without parts of the radial canal demonstrate a gradient in the potential for manubrium regeneration and in regeneration time.
2.  Implantation experiments exclude the manubrium as a source of inhibition or induction in the regeneration of another manubrium. One special case of inhibition appears to be due to competition for a common substrate.
3.  Medusa fragments consisting of only peripheral umbrella (C-fragments) undergo a considerably different restitution process as compared with fragments including a central portion of the umbrella (A-fragments). Vital stain is seen to disperse in the subumbrellar tissues during this process in C-fragments, whereas vital stain in A-fragments is observed to accumulate and later on is incorporated into the regenerating manubrium.
4.  The mesogloea of different-sized A-fragments retains a stable form when freed of its adhering cellular components, after a 12–24 h regeneration period; for C-fragments, however, the same result is not observed until 72 to 96 h after their excision.
5.  InPodocoryne carnea the observed gradients in manubrium regeneration can be abolished when the subumbrellar tissues are separated from the mesogloea by collagenase treatment.
6.  A model for manubrium regeneration in interradial fragments, based on the influence of tension exerted by the cicatrization process and the counteracting mesogloeal force, is presented and discussed.
