We have investigated the development of chemosensilla and the secretion of odorant‐binding proteins (OBPs) and chemosensory proteins (CSPs) in the embryo of Locusta migratoria manilensis. We first report the changes of each sensillum in embryo just preceding hatch in detail and show that different sensilla have different developmental processes. Trichogen cells are first involved in forming the structure of pegs, and then, after retraction, they start secreting OBPs and CSPs in the sensillar lymph. The synthesis of LmigOBP1 starts during the embryogenesis about 0.5 h preceding hatching, specifically in sensilla trichodea and basiconica of the antenna. LmigOBP2, instead, was only found in the outer sensillum lymph (oSl) of sensilla chaetica of the antenna, while we could not detect LmigOBP3 in any type of sensilla of the antenna. The ontogenesis of CSPs in the embryos is similar to that of OBPs. Expression of CSPI homolog in Locusta migratoria is detected using the antiserum raised against SgreCSPI. CSPI is specifically expressed in the outer sensillum lymph of sensilla chaetica of the antenna, and anti‐LmigCSPII dose not label any sensilla of the embryos. These data indicate that in locusts, OBPs and CSPs follow different temporal expression patterns, and also that OBPs are expressed in different types of sensilla. © 2009 Wiley Periodicals, Inc.  相似文献

We describe the number, distribution, and function of sensilla located on different organs of Lobesia botrana (Lepidoptera: Tortricidae) females using scanning electron microscopy, selective staining, and contact electrophysiology. The tarsi of the prothoracic legs bear contact chemo‐mechanoreceptor sensilla chaetica (5–13 per tarsomere), arranged in rings mainly concentrated on ventral surfaces, and different mechanosensory structures (sensilla chaetica, sensilla squamiformia, sensilla campaniformia, and spines). A single contact chemo‐mechanoreceptor sensillum chaeticum is present between the claws on the pretarsus. The ventral surface of the ovipositor lobes is covered with numerous mechanosensory sensilla chaetica of different types, out of which 10 have a contact chemosensory function. Putative contact chemo‐mechanoreceptor sensilla were also observed on the proboscis and antenna. Longitudinal rows of alternated sensilla styloconica and basiconica are present on the distal part of the proboscis, and rings of sensilla chaetica are present at the antennal tip. The sensilla on these body parts may play different roles in the selection of an oviposition site.  相似文献

Sensilla chaetica, which protrude above all other sensilla on the antenna of Psylliodes chrysocephala L., the cabbage stem flea beetle, were investigated ultrastructurally and found to be innervated by five to six sensory neurones. A dendrite from one of these neurones terminates in a tubular body at the shaft base, whereas dendrites from the others run unbranched to a pore at the shaft tip. Such a structure typifies a sensillum with a combined gustatory/mechanosensory function. Electrophysiological recordings using the tip-recording technique confirmed that this sensillum contains one mechanosensory cell and several chemosensory cells. The chemosensory cells were responsive to host plant chemicals. Sensilla chaetica were also found to be responsive to glucosinolates. One of the sensilla chaetica emerges from a domed area of cuticle on antennomere six. This was found to be relatively less responsive to the chemical stimuli tested and more responsive to mechanical stimulation. It is suggested that the sensilla chaetica are contact chemosensilla, that respond to chemicals present in plant surface waxes when P. chrysocephala antennates a leaf.  相似文献

The hairs on the wings of Locusta migratoria were observed and mapped using light microscopy, as well as by scanning and transmission electron microscopy. Based on their ultrastructure, we can distinguish four main types of hairs on the wings of adult L. migratoria , viz, short, medium and long hairs, and sensilla chaetica. The long hairs are located only on the ventral surface of the hindwing, whereas the other three types are present both on the dorsal and ventral surfaces of forewing and hindwing in both sexes. Medium hairs and sensilla chaetica are significantly more abundant on the dorsal surface of forewings in both females and males, than on the ventral surface, whereas the opposite was observed for short hairs (P < 0.01). No significant difference between males and females was observed in the density of any type of hairs (P > 0.1). Several dendritic branches, enveloped by a dendrite sheath, are situated in the lymph cavity of sensilla chaetica. Instead, no dendritic structure was observed in the cavity of the other three types of hairs. Immunocytochemical localization of chemosensory proteins (CSPs) was performed on ultrathin sections of hairs on wings. The antiserum against chemosensory proteins from L. migratoria (LmigCSP-II) strongly labelled sensilla chaetica, with gold granules only found in the outer sensillum lymph. In addition, the epidermal cell membrane of the wing was stained by the antiserum against LmigCSP-II. The other three types of hairs were never labelled. The results indicate that the wings might involve in contact chemoreception process.  相似文献

Highly developed chemoreception allows insects to detect foods, find mates, and escape natural enemies. We described the structures and distributions of antennal chemosensilla in Adelphocoris fasciaticollis Reuter by scanning electron microscopy and transmission electron microscopy. Seven major types of antennal sensilla were identified in adults of both sexes. Types 1 and 2 are sensilla chaetica and have thick cuticular walls with conspicuous grooves at their surfaces. Types 3 and 4 are multiporous sensilla trichodea with 1–3 dendrites located at the sensillum lymph, indicating a putative olfactory function. Types 5 and 6 are typical sensilla basiconica but share different characteristics in both external morphology and internal ultrastructure, and may be involved in the perception of host-associated odorants. The last sensilla were Böhm bristles. In addition to the morphological characterization, electrophysiological responses of antennal chemosensilla to 51 semiochemicals were investigated based on electroantennogram (EAG) recordings. Results revealed that different chemical stimuli elicited significantly different dose-dependent EAG responses, in which potential sex pheromone components and green leaf volatiles showed relatively higher EAG responses, but neither monoterpenes nor sesquiterpenes can elicit favorable EAG values. The results provided direct morphological and electrophysiological evidence that the adult antennae of A. fasciaticollis could function in searching for mates and host plants.  相似文献

The sensilla on labial palps in Locusta migratoria were observed and mapped using light microscopy, scanning and transmission electron microscopy. A dome region on the tip of the fourth segment (distal segment) of labial palps is mainly covered with sensilla chaetica (about 98%), and few sensilla basiconica (2%). The total number of both types of sensilla is significantly higher in females than in males. Sensilla chaetica can be further subdivided into three groups containing 6, 7 or 10 neurons. Immunocytochemical localization of odorant-binding protein (OBP) and chemosensory proteins (CSPs) was performed on ultrathin sections of sensilla on labial palps. The antiserum against odorant-binding protein from Locusta migratoria (LmigOBP) only labelled sensilla basiconica, with gold granules only found in the sensillum lymph. Chemosensory protein instead was specifically present in the outer sensillum lymph of all three subgroups of sensilla chaetica with antiserum against CSP-I from Schistocerca gregaria (SgreCSP-I). In contrast these three subgroups were never labelled with antiserum against CSP-II from Locusta migratoria (LmigCSP-II). In addition, a few sensilla chaetica could not be stained with any of the antisera used.  相似文献

The nut weevil, Curculio nucum (Linnaeus, 1758), is the main pest in hazelnut orchards (Corylus avellana L.). Semiochemicals are interesting bio control tools that could be used to manipulate the pest behaviour and to control pest populations. The study of the sensorial equipment of the insect antennae provides information on the importance of olfaction in the adult life for host plants and mate findings as well as on the putative other senses. Before electrophysiological investigation, the knowledge of antennae equipment is also necessary. The aim of this study is to determine the types, number and location of sensilla on the antennae of male and female adult C. nucum in order to determine their implication in seeking a sexual partner and a host plant. The 12-segmented antenna comprises a scape, a 7-segmented funicle and a 4-segmented club. Out of the nine sensillum types listed, three are present on the scape and the funicle and seven types on the club which gathers 71–73% of the total of sensilla. Tactile aporous sensilla chaetica C1, gustatory uniporous sensilla chaetica C2, olfactory multiporous sensilla basiconica B1 and B2 are found on both the dorsal and ventral surfaces of the club in both sexes. Thermo-hygroreceptive dome-shaped sensilla D, olfactory multiporous sensilla basiconica B3 and olfactory multiporous fluted sensilla basiconica F are found exclusively on the ventral surface of the club, suggesting that these sensilla are utilized in host plant acceptance during antennal tapping. The sexual dimorphism concerns only the numbers of sensilla chaetica C1 and sensilla basiconica B2.  相似文献

The ultramorphology of the antennae and mouthparts of the adult Loxocephala perpunctata Jacobi was studied through a scanning electron microscope. Seven types of sensilla were found on antennomeres, including a Böhm bristle on the scape, sensillum trichoideum and plaque organ on the pedicel, two subtypes of sensilla chaetica and two subtypes of sensilla campaniformia on these two antennomeres; and Bourgoin's organ with sensory pegs and sensilla basiconicum on the basal bulb of the flagellum. The mouthparts of L. perpunctata are of the typical piercing-sucking type, similar to mouthparts found in other hemipteran insects. In general, six types of sensilla (i.e., four subtypes of sensilla chaetica, sensillum basiconicum, subapical labial sensillum, uniporous peg-like sensillum, multiporous peg-like sensillum and two subtypes of bristle-like sensilla) were detected on different locations of the labium, with the last three, and numerous cuticular processes, present on the labial tip. The potential functions of these sensilla are discussed.  相似文献

The antennae of Lepidoptera Neopseustidae were examined with the scanning electron microscope. The studied species, Nematocentropus cfr. omeiensis, Neopseustis meyricki, Synempora andesae, Apoplania valdiviana and Apoplania penai possess nine types of antennal flagellum sensilla: multiporous large sensilla basiconica, multiporous thin sensilla basiconica, multiporous small sensilla basiconica, multiporous sensilla trichodea, multiporous sensilla coeloconica; uniporous sensilla chaetica; aporous sensilla chaetica, aporous stylus-shaped sensilla chaetica, aporous sensilla styloconica. The multiporous sensillum type here termed “multiporous large sensillum basiconicum” is unknown from other Lepidoptera and probably constitutes an autapomorphy of the family Neopseustidae. This sensillum type is remarkable by having a single base in female Apoplania and Synempora while in male Apoplania it has a bifid or trifid base, and in male Synempora it is composed of two or three incompletely separated hairs. This may be the first recorded example of a sexually dimorphic lepidopteran sensillum type. The stylus-shaped sensillum chaeticum is a primitive type which occurs only in some lower Lepidoptera.  相似文献

The number and distribution of chemosensilla located on different organs of Busseola fusca (Fuller) (Lepidoptera: Noctuidae) males and females are described based on observations using scanning electron microscopy, selective staining with silver nitrate, and gustatory electrophysiological recording. The antennae and the fifth tarsomere of the prothoracic legs of both sexes bear chemosensilla: uniporous chaetica and multiporous trichoidea sensilla. However, there is a sexual dimorphism in the number and size of sensilla on these organs. The distal part of the ovipositor has uniporous gustatory chemosensilla of the chaetica type. The involvement of these sensilla in oviposition site selection by B. fusca is discussed.  相似文献

The antennal sensilla of alate Myzus persicae were mapped using transmission electron microscopy and the ultrastructure of sensilla trichoidea, coeloconica, and placoidea are described. Trichoid sensilla, located on the tip of the antennae, are innervated by 2–4 neurons, with some outer dendrites reaching the distal end of the hair. Coeloconic sensilla in primary rhinaria are of two morphological types, both equipped with two dendrites. Dendrites of Type II coeloconic sensilla are enveloped in the dendrite sheath, containing the sensillum lymph. In sensilla coeloconica of Type I, instead, dendrites are enclosed by an electron opaque solid cuticle, with no space left for the sensillum lymph. The ultrastructure of big placoid sensillum reveals the presence of three groups of neurons, with 2–3 dendrites in each neuron group, while both small placoid sensilla are equipped with a single group of neurons, consisting of three dendrites. Both large and small placoid sensilla bear multiple pores on the outer cuticle. The function of these sensilla is also discussed. J. Olfactory responses of Dasineura tetensi (Rubs) (Diptera; Cecidomyiidae) to leaf volatiles of blackcurrant (Ribes nigrum) were tested in a 4-way olfactometer. Newly emerged virgin females showed no response to the leaf volatiles emitted from a blackcurrant shoot. Newly emerged males (which are known to respond to a pheromone released by the female) also showed no response to the leaf volatiles. Two hours after mating females responded positively, indicating that leaf volatiles may play a role in host plant finding. Scanning (SEM) and transmission (TEM) electron microscopy of the antennae of D. tetensi showed that males and females share five sensillum types; sensilla chaetica (mechanoreceptors), sensilla trichodea, sensilla basiconica, uniporous peg sensilla and circumfila (chemoreceptors). The sensilla chaetica and sensilla trichodea resemble those found on other insects. Sensilla basiconica were found on all antennal subsegments except the tip. These are multiporous receptors with five unbranched dendrites filling the lumen. Small peg sensilla located on the tips of both male and female antennae may function as contact-chemoreceptors. The circumfila, which are a unique type of sensilla found only on cecidomyiid antennae, form loops around each of the antennal subsegments, being attached to the surface by a series of stalks. TEM revealed that each stalk consisted of one sensillum containing a single highly branched dendrite. The distal regions of the walls of each sensillum are fused together to form the circumfila. Circumfila have multiporous walls and a lumen filled with multiple branches of dendrites. Their structure suggests that they are important olfactory receptors in both the male and female.  相似文献

The two sensillum lymph cavities of contact chemosensitive sensilla of insects are separated by the dendrite sheath. The composition of the dendrite sheath in the contact chemosensitive sensilla of the ventral sensory field on the maxillary palps of Periplaneta americana L. has been studied using lectin cytochemistry. The dendrite sheaths can be isolated from the surrounding tissue and the autofluorescent cuticle by incubation of cryostat sections in phosphate-buffered saline containing 1% Triton X-100. Various lectins bind specifically to the dendrite sheath indicating the presence of sugar residues. Enzymatic digestion with hyaluronidase or trypsin prior to lectin cytochemistry decreases staining intensity, whereas treatment with neuraminidase and chitinase causes no visible change. The material of the dendrite sheath can thus be assumed to contain acid mucopolysaccharides that probably are similar to those of the receptor lymph cavities. The negatively charged, hydrophilic acid mucopolysaccharides may make the dendrite sheath permeable for small cations. These findings are discussed in the context of the functional significance of the dendrite sheath in contact chemosensory sensilla of insects.  相似文献

Antennal sensilla were compared in females and males of two sympatric mymarid Hymenoptera, Anaphes victus and A. listronoti which are, respectively, solitary and gregarious parasitoids of eggs of the carrot weevil Listronotus oregonensis (Coleoptera, Curculionidae). Both species are morphologically very similar in the area where they are sympatric. The external morphology of the sensilla was studied using scanning electron microscopy. Female antennae have seven different types of sensilla, morphologically similar in the two species: trichoid sensilla, which are putative mechanosensilla, sensilla chaetica types 1, 3 and 4, which are presumably contact chemosensilla, and sensilla chaetica type 2 and basiconic and placoid sensilla, which are presumed to be olfactory sensilla. The major difference between the two species is the number of sensilla chaetica type 4, of which 6–9 are found on the antennal club in A. victus, while 10–12 are present in A. listronoti. The antennae of the males of both species are similar in morphology and in the number and distribution of their four types of sensilla, i.e. trichoid sensilla, sensilla chaetica type 1 and basiconic and placoid sensilla. Nine different types of sensilla have been identified on the antenna of the cassava mealybug Phenacoccus manihoti (Homoptera : Pseudococcidae) with scanning and transmission electron microscopes. Trichoid sensilla, distributed on all segments of the antenna and innervated by a single mechanoreceptive dendrite, have the characteristics of exteroceptors. A campaniform sensillum located on the pedicel and one basiconic sensillum on the flagellum have the characteristics of proprioceptors. Coeloconic sensilla, located ventrally on the pedicel and flagellum, related to poreless sensilla with inflexible sockets, have the characteristics of thermo/hygroreceptors. Uniporous sensilla with a mechanoreceptive dendrite (smooth pegs P1 and P2, grooved pegs P3) and multiporous chemosensilla (grooved pegs P4 and P5), present on the tip of the flagellum, have, respectively, the characteristics of gustatory and olfactory receptors. The results of this study seem to suggest that the cassava mealybug has sensory equipment on its antennae that can detect, by olfaction and contact, chemicals released by the plant.  相似文献

Summary The ventral organs of the cephalic lobes of the house fly larvae, Musca domestica L., were studied by scanning and transmission electron microscopy. Four sensilla were found. Three of them are each innervated by a single dendrite whose ending possesses a tubular body and communicates to the exterior through an opening. These sensilla are assumed to be mechanoreceptors. The 4th sensillum is supplied by 2 bipolar neurons with the unbranched dendritic tips (without tubular bodies) exposed to the exterior through a single opening and is probably a contact chemoreceptor. This research was supported in part by the Office of Naval Research and NIH Training Grant ES-00069. The general morphology of the antennae and the antennal sensilla of both sexes of Boarmia (Ascotis) selenaria SchiffermÜller are described. The following types of sensilla were found: Bohm bristles and sensilla chaetica, trichodea, coeloconica, auricillica and styloconica. Antennal sexual dimorphism is marked, but mainly confined to the presence of long sensilla trichodea in males and the absence in females. The sensilla trichodea are the most abundant sensillum type.  相似文献
