The genetic diversity and population structure of the miiuy croaker (Miichthys miiuy) were investigated. Seventy-six individuals were sampled from six localities of the East China Sea. Genetic variation in DNA sequences was examined from the 5′-end of the mitochondrial DNA control region. High levels of haplotype diversity (h = 0.99333) in the control region were detected, indicating a high level of genetic diversity. A total of 81 polymorphic sites were found, and 65 haplotypes were defined. The results of AMOVA analysis indicated that 97.9% of the genetic variation contained within populations and 2.14% occurred among populations. No significant genealogical branches or clusters were recognized on the NJ tree. Tests of neutral evolution and analysis of mismatch distribution suggested that miiuy croaker might have undergone a population expansion. The knowledge on genetic diversity and genetic structure will be crucial to establish appropriate fishery management stocks for the species.  相似文献

Portunus trituberculatus is a commercially important fishery species. In this study, intraspecific variation was examined by using mitochondrial DNA 16S rRNA gene in 213 individual crabs sampled from six localities along the coast of the East China Sea. Twenty-two polymorphic sites defined 25 distinct haplotypes, revealing a moderately high haplotype diversity and relatively low sequence divergence among the six localities. An excess of within population unique haplotypes at most sample locations were detected, which might influence genetic structure of the swimming crab populations. Neither neighbor-joining tree nor minimum spanning network (MSN) based on the haplotype data indicated distinct patterns of phylogeographic structure among the 25 haplotypes. Analyses of molecular variance (AMOVA) and FST statistics supported the hypothesis that population samples from the East China Sea were genetically nonhomogenous, indicating that gene flow might be restricted across those regions, despite the high potential of dispersal. In addition, tests of neutral evolution and analysis of mismatch distribution suggested that P. trituberculatus might have undergone a population expansion, possibly within the last 127,000 and 429,000 years. Our study unraveled the extant population genetic structure of the P. trituberculatus, and addressed the related fishery management issues including artificial breeding, fishery stock identification and conservation.  相似文献

In order to describe the genetic diversity of five geographical populations of cuttlefish (Sepiella japonica) along with Chinese coast and determined their phylogenetic relationship, partial mitochondrial COI gene (681bp in length) was amplified from 96 individuals collected from these populations and sequenced. The 5 populations of cuttlefish inhabit Yellow Sea, East China Sea and South China Sea. Out of 22 polymorphic nucleotides identified, 8 were represented by a single sequence, 12 were parsimony informative, which defined 22 haplotypes. Haplotype and nucleotide diversity were low among populations. Of 22 haplotypes, 15 appeared only in a single population, 6 appeared in 2 or 3 populations and 1 was shared by all populations. The COI gene was monomorphic in Qingdao population. The haplotypes identified clustered into 2 clades, each covered individuals from 5 populations each. Pairwise FST were not proportional to the geographical distances. Among the 5 populations, relatively high level of genetic diversity was found in Ningde population which was recommended to be the best choice of germplasm resources for artificial releasing project.  相似文献

Portunus trituberculatus is a commercially important species widely spread in the East China Sea. Intraspecific variation of the mitochondrial DNA cytochrome oxidase subunit I (mtDNA COI) gene was investigated in 213 individuals from six localities (Changjiang Estuary, Shengsi Islands, Zhoushan Islands, Dongtou Islands, Dinghai Bay, and Quanzhou Bay) ranging from north (31°21′N) to south (24°55′N) coastal waters of the East China Sea. Overall, a total of 27 mtDNA haplotypes and 21 variable sites were detected in the 787 bp segment of COI gene. Analysis of mtDNA COI sequence data revealed that crabs from the six localities were characterized by moderately high haplotypic diversity (h = 0.787 ± 0.026), while sequence divergence values between haplotypes were relatively low (π = 0.00241 ± 0.00098). Each population was characterized by a single most frequent haplotype, shared among all six localities, and a small number of rare ones, typically present in only one or two individuals and representative of a specific population. However, neither the neighbor-joining tree nor the minimum spanning network (MSN) based on the haplotype data exhibited geographical patterns of the six populations. Mismatch distribution analysis of P. trituberculatus individuals sampled from the six localities suggested that sudden population expansion might have occurred in CJ and SS population that might be consistent with over-exploitation of the swimming crab. Analysis of molecular variance (AMOVA) and F ST statistics showed that significant genetic differentiation existed among the SS, ZS, DT, DH, and QZ populations, suggesting that gene flow might be reduced, even between the geographically close sites, despite the high potential of dispersal. The possible causes of the observed genetic heterogeneity among the P. trituberculatus populations and the potential applications of the mtDNA COI marker in the artificial breeding and fisheries management are discussed. Eleutheronema tetradactylum is an economically important fish species in China water. To investigate the genetic diversity and describe population structure of it, an 1151 base pair (bp) fragment of the mitochondrial DNA Cytb sequence was analyzed in 120 individuals from four populations in the East China Sea and the South China Sea. A total of 16 haplotypes were defined by 24 variable nucleotide sites. High level of haplotype diversity and low nucleotide diversity were observed in all populations. The results of AMOVA detected that 89.44% of the genetic variation occurred within populations. Significant genetic differentiations were detected among populations (0.05097, P < 0.05), but no large-scale regional differences were detected. Analysis of neutral evolution and mismatch distribution suggested no recent population expansion happened. The present results provided new information for genetic assessment, fishery management and conservation of this species.  相似文献

对采自黄海、东海和南海的7个银鲳群体的线粒体COⅠ基因序列变异进行分析,研究银鲳的遗传多样性、遗传结构和群体历史动态。在所分析的111个个体中检测到16个单倍型。7个群体呈现出高的单倍型多样性(h=0.564~0.688) 和低的核苷酸多样性(π=0.001~0.003)。单倍型遗传学关系、两两群体间的FST值和分子方差分析均表明中国近海7个银鲳群体间的遗传分化不显著。中性检验和核苷酸不配对分析均表明中国近海银鲳经历了晚更新世的群体扩张事件,扩张时间约为6.0×104~1.04×105 年前。研究结果表明,银鲳的卵和幼体具有较强的扩散能力、中国近海的海洋环流以及近期的群体扩张可能是造成中国近海银鲳群体在线粒体COⅠ基因序列上存在较高的遗传同质性的原因。  相似文献

Silver pomfret (Pampus argenteus) is an important fisheries resource in China, but harvest management lacks identification of management units (genetic stocks or populations). Using amplified fragment length polymorphism (AFLP), we investigated the population genetic structure of silver pomfret from the Yellow and East China Seas. In 143 individuals examined from six locations (three locations in the Yellow Sea, three in the East China Sea), a total of 194 loci were detected by six primer combinations. In all loci, 149 loci (76.8%) were polymorphic, and the percentage of polymorphic loci varied within samples from 62.9% to 67.0%. Among the six samples, Nei’s genetic diversity was 0.23 ± 0.20–0.25 ± 0.21, and Shannon’s index was 0.34 ± 0.28–0.36 ± 0.29. Analyses revealed significant genetic differentiation among the six samples, indicating at least six separate populations of silver pomfret in the Southern Yellow and East China Seas.  相似文献

The cocktail shrimp Trachypenaeus curvirostris is an ecologically and economically important shrimp species in the Yellow Sea and East China Sea. However, there is no information about its population genetic structure. The population genetic structure and level of gene flow of T. curvirostris from the Yellow Sea and the East China Sea were studied with a 658-bp segment of mtDNA COI gene. In total, 85 individuals were collected from five locations and 13 haplotypes were obtained. The genetic variation of COI gene in five populations was moderate, giving an overall haplotype diversity of 0.6888 ± 0.0432 and nucleotide diversity of 0.0069 ± 0.0038. Conflicting to our expectation, significant genetic differentiation was detected in this species. The result revealed two genetically divergent lineages, displaying clear different geographical distributions in the studied area. The significant genetic differentiation between the Yellow Sea and East China Sea populations might be caused by the Yangtze River outflow. Mismatch distribution revealed that T. curvirostris had undergone population range expansion, possibly before 103,400–109,700 years ago in the last interglaciation, rejecting the sudden demographic expansion.  相似文献

The genetic diversity of silver pomfret (Pampus argenteus) from the Bohai, East China, and South China Seas was investigated using mitochondrial DNA (mtDNA) control region sequence data. We found high levels of variation with 17 haplotypes among the 45 individuals (h = 0.88, π = 0.006). AMOVA analysis detected significant structuring among China Sea silver pomfret (P < 0.05, FST = 0.050), which indicates significant genetic differentiation. No significant differentiation between Bohai Sea and East China Sea samples was detected (P > 0.05). Whether silver pomfret from the South China Sea exhibit a different demographic history than those from the Bohai and East China Seas remains to be determined.  相似文献

The large yellow croaker Larimichthys crocea is an important commercial marine fish species in China. However, information about the population structure of this species is limited. In the present study, mitochondrial DNA (mtDNA) control region was sequenced from four populations of the yellow croaker in the southern Yellow Sea, East China Sea and South China Sea to investigate the genetic structure of this species. A total of 54 haplotypes were detected among 62 individuals of large yellow croaker. High levels of population genetic diversity were observed. Among the four populations, the haplotype diversity was between 0.9895 ± 0.0193 (Xiamen) and 1.0000 ± 0.0524 (Lvsi, Zhoushan). The nucleotide diversity ranged from 0.0208 ± 0.0108 (Xuwen) to 0.0246 ± 0.0138 (Lvsi). The results of AMOVA detected no significant differences among populations. The conventional FST statistics were negative and insignificant values. These indicated lack of population genetic structure throughout the Yellow Sea, East China Sea and South China Sea, and random mixing of individuals among the samples. Biological characters of large yellow croaker and lack of physical barrier in the studied area could be the reasons for lack of genetic structure in this species.  相似文献

The Asian green mussel Perna viridis is ecologically and economically important in the coastal regions of China. In order to characterize the genetic diversity and population connectivity of P. viridis in South China Sea, a 664 bp region of mitochondrial COI gene and a 293 bp region of 16S rRNA gene were sequenced and analyzed for 78 and 92 individuals from four populations in South China Sea, respectively. A total of 15 haplotypes were defined by 14 variable nucleotide sites in COI gene, and 7 haplotypes by 6 variable nucleotide sites in 16S rRNA gene. High haplotype diversity and low nucleotide diversity were observed in COI gene, while moderate haplotype diversity and low nucleotide diversity were observed in 16S rRNA gene. Pairwise FST values of COI gene were all negative and those of 16S rRNA gene ranged from −0.01409 to 0.10289. The results showed that no significant genetic divergence (or shallow genetic structure) and high levels of population connectivity among the four populations of P. viridis in South China Sea.  相似文献

Tea green leafhopper is one of the most damaging tea pests in main tea production regions of East Asia. For lack of recognized morphological characters, the dominant species of tea green leafhoppers in Mainland China, Taiwan and Japan have always been named as Empoasca vitis Göthe, Jacobiasca formosana Paoli and Empoasca onukii MATSUDA, respectively. Furthermore, nothing is known about the genetic relationships among them. In this study, we collected six populations from Mainland China, four populations from Japan and one population from Taiwan, and examined the genetic distances in the COI and 16sRNA regions of mtDNA among them. The results showed that the genetic distances based on single gene or the combined sequences among eleven leafhopper populations were 0.3–1.2%, which were all less than the species boundary of 2%. Moreover, there were at least two haplotypes shared by two distinct populations from different regions. The phylogenetic analysis based on single gene or combined sets also supported that tea green leafhoppers from Mainland China, Taiwan and Japan were closely related to each other, and there were at least two specimens from different regions clustered ahead of those from the same region. Therefore, we propose that the view of recognizing the dominant species of tea green leafhoppers in three adjacent tea production regions of East Asia as different species is unreliable or questionable and suggest that they are a single species.  相似文献

Japanese anchovy Engraulis japonicus was not only a commercial species but also a key link in the marine food chain in the Yellow Sea and East China Sea. However, in recent years, E. japonicus experienced a deep recession caused by over fishing. Therefore, determination of population genetic structure is essential to underpin resource recovery and to aid delineating and monitoring populations for fishery management. In this study, sequence variation of a 402-bp fragment of mitochondrial cytochrome b (cyt b) gene from ten populations of E. japonicus in the Yellow Sea and East China Sea was determined, detecting 73 haplotypes. Haplotype diversities and nucleotide diversities for the ten populations ranged from 0.4706 ± 0.1177 (D) to 0.9935 ± 0.210 (WH), and from 0.0014 ± 0.0013 (D) to 0.0090 ± 0.0053 (WH), respectively. Analysis of molecular variance (AMOVA) showed no significant differences at all hierarchical levels, and almost all FST values were non-significant, indicating no significant population genetic structure in the Yellow Sea and East China Sea. Two indistinct clades with low support values were detected and the analysis of haplotype frequency distribution showed no significant geographic differences among populations. These results supported the null hypothesis that E. japonicus within the Yellow Sea and East China Sea constituted a panmictic mtDNA gene pool. Based on the present study, the existence of separate genetic stocks in the Yellow Sea and East China Sea were not detected. Strong dispersal capacity of planktonic larvae, and ocean current could be the reasons for genetic homogeneity in this species in the Yellow Sea and East China Sea.  相似文献

In order to prove the fisheries‐related hypothesis of the presence of two local populations of the mottled skate, Beringraja pulchra in the Yellow Sea (Sensu Jo et al., 2011 ), the geographic variations of 90 individuals (in molecule) and 129 individuals (in morphology) were investigated. The skates were collected from the middle Yellow Sea (Daechungdo), southern Yellow Sea (Heuksando), and the middle East Sea (Ulleungdo) using gill nets, bottom trawls and longlines. Comparing 471 base pairs of mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) sequences, only three haplotypes (BR1–BR3) were found. The Daechungdo and Heuksando individuals displayed all three haplotypes, whereas the Ulleungdo individuals displayed a single haplotype (BR3). Pairwise fixation indices (FST) showed that the Daechungdo and Heuksando individuals were not genetically distinguishable (with a value of ?0.0344), whereas the Ulleungdo individuals were clearly distinct from both the Daechungdo and Heuksando individuals (with values of 0.6875 and 0.6871, respectively). Similar to the molecular result, a canonical discriminant analysis based on seven morphometric characters showed that the Ulleungdo individuals had positive values on the first axis, whereas those from the other two locations had negative values, slightly differentiating the two populations (East Sea population vs Yellow Sea population). This study is the first to identify two populations of mottled skate in the northwest Pacific.  相似文献

The intraspecific structure of the European anchovy (Engraulis encrasicolus) was studied on the basis of mitochondrial cytochrome b gene (cytb) fragment variability in 84 individuals from seven localities of the Black Sea and the Sea of Azov. The data on haplotype and nucleotide diversity and the values of neutrality tests suggested expansive growth of anchovy populations in the Azov-Black Sea basin. All samples from anchovy populations demonstrated a high level of haplotype diversity (Hd = 0.962). Two dominant haplotypes were identified, the frequencies of which were not directional, and they were present in all localities. Sequence analysis of the mitochondrial cytb gene fragment showed no differentiation between the Sea of Azov and the Black Sea subspecies.  相似文献

The bumblebee, Bombus ardens, is a valuable natural resource, and is most notably utilized for greenhouse pollination. In order to gain a greater understanding of the population genetic structure and the genetic diversity of this species, we sequenced a portion of the mitochondrial COI gene corresponding to the “DNA Barcode” region (658 bp) from 160 individuals collected over 15 Korean localities. Uncorrected pairwise distances among the eight haplotypes suggested low intraspecific genetic diversity, with a maximum sequence divergence of 0.3%. Such a low level of intraspecific genetic diversity was further reflected in local populations, particularly to islet populations, such as those of Youngheungdo, Jakyakdo, and Ulleugdo, which had zero genetic diversity. Geographically, one haplotype (BARBA01) was widespread and dominant, with a frequency of 90.6% (145 among 160 individuals). Other haplotypes were restricted to one to three localities and had low frequency. Overall, a very high rate of per generation female migration ratio (Nm = 4.6 to infinite) and a very low level of genetic fixation (FST = 0 to 0.099) were detected between pairs of localities, suggesting that the B. ardens populations on the Korean peninsula are panmictic, which is consistent with our understanding of their dispersal capability.  相似文献

In this paper, we analyzed population genetic diversity of Scylla paramamosain in Hainan Island of China based on COI gene sequence. Totally 92 individuals were collected from six localities: Haikou, Wenchang, Wanning, Sanya, Dongfang and Danzhou. A 761 bp fragment was sequenced, defining 32 different haplotypes. H4 was the most frequent haplotype, existing in all six localities, while the majority of haplotypes were rare ones, existing in only one or two individuals. Haplotype diversity (h) and nucleotide diversity (π) ranged from 0.625 to 0.914 and from 0.001 to 0.003, with an average of 0.841 and 0.002, respectively. Genetic distance ranged from 0.001 to 0.003 within localities and from 0.002 to 0.003 between localities. The AMOVA analysis indicated a low level of genetic differentiation among six localities (FST = 0.0176, P > 0.05). Neutrality tests and mismatch analysis implied a potential population expansion event for this crab species.  相似文献

为了揭示中国岩羊不同地理种群的遗传差异,探讨岩羊亚种分化的分子机制,采用中国岩羊不同地理种群的细胞色素b(Cyt b)基因的全序列,分析了碱基变异情况、遗传距离以及核苷酸序列差异。用最大似然法和贝叶斯法构建分子系统树并对获得的拓扑结构进行分析。结果发现,西藏亚种与四川亚种Cyt b基因平均序列差异为4.2%(±0.007),处于偶蹄目亚种的序列差异范围内,支持了目前对岩羊西藏亚种的分类地位。四川亚种内部各地理种群之间的遗传距离(0.033±0.0 111)与它们分别到西藏种群的遗传距离(0.042±0.007)差异不显著(t=1.824,P=0.084),说明四川亚种内部各地理种群间已经发生较显著的遗传分化。其中,四川、甘肃和青海种群亲缘关系较近,并与四川亚种内部的其它种群已产生了显著的遗传分化。因此认为四川亚种内部各地理种群的种下分化需要深入研究。  相似文献

Gymnocypris dobula is a commercially important fishery species and mostly distributed in the freshwater of Tibetan plateau. In this study, genetic diversity and intraspecific population differentiation were examined by using mitochondrial DNA D-loop sequences in 97 individuals sampled from three localities (Pali, Lasa and Yanghu) in the Tibetan plateau. Two hundred and fourteen polymorphic sites and 50 haplotypes were defined among the three localities. Genetic diversity analysis showed that the highest genetic diversity level was found in Pali population. Phylogenetic relationships analysis results indicated that closer phylogenetic relationships were found between the Yanghu and Lasa populations. Genetic population differentiation analysis indicated that the majority of variation (84.91%) was attributed to variations among populations and the largest differentiation was found between Pali and Lasa localities (Fst = 0.874, Nm = 0.036). Moreover, the historical demographic events were assessed by implementing the mismatch distribution analysis, Fajima's D test and Fu's Fs test. The results indicated that the Pali population had undergone a demographic expansion, possibly within the last 0.163 MYA (Million Years Ago). Our study firstly identified the population genetic structure of the G. dobula, which could be helpful for artificial breeding, fishery stock identification and resource conservation for this species.  相似文献

To investigate the phylogeographic pattern and historical demography of Saurida elongata, a 463 base pair fragment of the mitochondrial DNA control region was analyzed in 118 individuals collected from five populations in the East China Sea and South China Sea. The results revealed remarkably low nucleotide diversity in S. elongata, which was probably caused by selection during Pleistocene drastic climate fluctuations. Phylogenetic relationship was continuous, but a localized clade was found dominating Beihai population. Mismatch analyses showed that S. elongata experienced both demographic and range expansions. Among these populations, Beihai population expanded latest. Localization of haplotypes clade in Beihai was likely a result of the selection when new habitat was established during a range expansion, and indicated limited gene flow between Beihai and other populations. Our results suggested Beihai population should be managed separately in fishery.  相似文献
