A model was developed based on an initial ingestion phase, followed by a period of virus multiplication and host death, to describe the effects of codling moth granulosis virus (CpGV) concentration and time from initial infection on the percentage of codling moth larvae (Cydia pornonella) dying from virus infection in the laboratory. With modification, this model also described the effect of CpGV concentration on the reduction of damage to fruit by C. pomonella larvae in the field. In both cases, the effect of CpGV increased with the l/10th power of virus concentration. LT50 (time to death of 50% of larvae) was inversely related to virus concentration in the laboratory. The model predicts the threshold concentration for death of larvae, as well as LD50(C50 and LD90(C 90,) for all ages of larvae. The model also predicts that if the majority of larvae entered through the sides of apple fruits, rather than through the calyx or near the stalk (as they probably did in the field), then the efficacy of CpGV in reducing fruit damage would be lowered. This effect of site of entry is especially marked at low virus concentrations.  相似文献

Studies were undertaken to improve the biological efficacy of the granulovirus (CpGV) of the codling moth, Cydia pomonella , by evaluating the performance of some formulation additives that might improve virus persistence and/or virus uptake by first instar larvae. Laboratory studies, using a leaf disc bioassay, demonstrated that 15% cane molasses incorporated within a formulation of purified CpGV dramatically reduced the median lethal exposure time (LET 50 ) to CpGV for neonate larvae at a CpGV dosage rate of 10 7 occlusion bodies (OBs) ml -1 . Screening of a range of other compounds showed that sucrose, fructose and sorbitol (at 10% concentrations) and extracts of apple flesh and skin also gave significant reductions in the LET 50 of CpGV formulations containing these ingredients. Pectin, malic acid and &#102 -farnesene did not significantly reduce the LET 50 . In a field trial, molasses included at 15% (v/v) in a CpGV formulation, containing a dosage rate of 10 12 OBs ha -1 , gave as good control of codling moth damage as virus formulations containing the 'sticker' 0.2% skimmed milk at higher dosage rates of 10 13 and 10 14 OBs ha -1 . Studies of CpGV persistence on foliage revealed no significant improvement of virus persistence on apple foliage using 10% or 15% molasses formulations. A second field trial demonstrated that 10% molasses, 10% sorbitol or 0.08% &#102 -farnesene significantly reduced codling moth deep damage to fruit when these ingredients were added to formulations of pure CpGV. Substantial sooty-mould growth ( Cladosporium spp.) was observed on apple foliage treated with formulations containing molasses, indicating that this formulation additive has secondary consequences that would need to be taken into account if molasses was to be used in commercial CpGV formulations. Nonetheless, these studies clearly demonstrate that major biological improvements in CpGV performance can be achieved by the incorporation of formulation additives, including molasses and several other compounds, that probably function as attractants and/or feeding stimulants for codling moth larvae.  相似文献

Cydia pomonella granulovirus (CpGV) has received considerable attention as a potential microbial insecticide for the control of the codling moth (Cydia pomonella L.) , a worldwide pest of apples. Laboratory experiments were established to investigate virus uptake by first instar larvae, using a novel leaf disc bioassay technique. Virus uptake was found to be independent of active feeding and larvae became infected simply by walking or browsing on sprayed leaf disc surfaces in as little time as 3.5 min. Infection increased as a function of time spent on the leaf disc surface and a linear log time/probit mortality relationship could be fitted. The bioassay technique used has potential for the realistic laboratory testing of virus spray formulations. A field experiment showed that virus infection could be contracted by newly hatched codling moth larvae both from the surface of sprayed leaves and sprayed fruit. The potential for exploiting this knowledge for improving spray formulations is discussed.  相似文献

The Cydia pomonella granulovirus (CpGV) has been used for many years as biological agent for codling moth control in apple orchards. Resistance to the Mexican strain of CpGV was detected in orchards in Germany, France and Italy. A laboratory insect colony was started from insects collected in a French resistant orchard. It was named RGV. Various virus isolates were identified as active against this resistant insect colony. Field tests were carried out in 2007 to test if the two virus isolates CpGV-I12 and NPP-R1 were effective in the field. Although these virus isolates were not able to reduce insect caused fruit damages, they significantly reduced the overwintering insect populations. NPP-R1 was subjected to eight passages on RGV larvae (NPP-R1.8) that improved its biological activity on RGV larvae. 2008 field trials were set up to test this improved virus strain, compared to CpGV-I12 and Madex plus active on RGV. These tests confirmed the ability to control both in susceptible and resistant insect populations.  相似文献

(E,Z) -2,4-decadienoate (pear ester) is a larval kairomone for the codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae). Orchard studies were conducted in 2005 and 2006 in apple, Malus domestica Borkhausen, and pear, Pyrus communis L., to evaluate a 5% active ingredient (AI), microencapsulated formulation of pear ester (PE-MEC) as an insecticidal additive for the codling moth granulovirus (CpGV). Although CpGV applied at 5-15-d intervals at commercial rates (2.2 X 10(12)-10(13) granules per ha) killed the majority (82-94%) of larvae found inside infested fruit, it did not eliminate significant damage, i.e., 30-92% fruit injury at harvest versus 51-82% in controls. PE-MEC treatments had significant but inconsistent results in our tests. In apple (mixed cultivars), PE-MEC (3.7-4.7 g [AI] /ha) plus CpGV reduced the percentage of fruit injured during the second but not the first larval generation, compared with CpGV alone, but there no was no additional population reduction (live larvae collected from infested fruit and tree bands). In 'Bartlett' pear, PE-MEC (3.7 g [AI] /ha) plus CpGV significantly increased larval mortality and reduced deep fruit entries at harvest over CpGV alone in 2006, but similar improvements were not observed in 2005 when a lower rate (1.5 g [AI] /ha) was tested. Surprisingly, compared with untreated controls, the PE-MEC formulation alone also reduced fruit injury (mid-season in Bartlett) and larval survivorship inside infested fruit at harvest (2006 apple tests and both years in Bartlett). Although pear ester seems amenable as a kairomonal adjuvant for use with insecticides, our inconsistent data with CpGV in apple and pear suggest practical improvements in formulation and application strategies (e.g., to optimize and maintain attractive release rates) are needed.  相似文献

  • 1 Aquatic larvae of the pyralid moth Parapoynx rugosalis Möschler repeatedly construct protective cases by cutting portions (discs) from leaves of the waterlily Nymphaea ampla and sandwich themselves between the disc and the underside of the host leaf. Construction of a new case requires leaving the old case, thus increasing exposure to predators and parasites.
  • 2 In an experiment, larvae with protective cases experienced no mortality due to predation by fish, whereas larvae without cases experienced substantial predation.
  • 3 In a series of choice tests, larvae preferentially selected young, tender leaves over old, tough leaves for construction of cases, and larvae spent significantly less time completing their shelters when cutting discs from young, tender leaves.
  • 4 A partial explanation of why larvae select young, tender leaves for construction of their protective shelters may be that exposure time to predators during construction is minimized.

Inundative applications of the codling moth (CM), Cydia pomonella L., granulovirus (CpGV), which target neonate larvae before or during initial entry into fruit, offer potential for selective control of this key pest. In field tests on apple we compared the persistence and efficacy of single applications of three CpGV products approved for organic orchards in North America. In addition, the success of repeated (2–14) applications of one product (Cyd-X) as a principal control measure for CM in apple orchards was monitored following operational use by cooperating growers at four separate locations. In the first study, an early season application of all products at label rates remained highly effective for the first 24 h (averaging 94% larval mortality relative to controls) and moderately effective after 72 h (averaging 71% mortality) during dry sunny conditions. Significant activity remained up to 14 days, suggesting prolonged survival of the virus in UV-protected locations, such as the calyx of fruit. A second application later in the season was slightly less effective. Data obtained from commercial sites provide circumstantial evidence for the effectiveness of well-timed CpGV applications against CM outbreaks. In all cases where first generation larvae were targeted beginning at egg hatch (≈250 degree days) and treated areas monitored (0.3–1.6 ha plots), fruit damage during the second larval generation was reduced or eliminated. Based on the number of live larvae recovered throughout the season, mortality rates remained high (80.3–100% across sites). The cumulative number of moths caught in pheromone-baited traps was reduced (66–94%) in the second flight. Data from tree bands placed to catch diapause-destined larvae indicated overwintering generations remained low in treated sites (0.18 larvae/band).  相似文献

Sequence analyses indicate that a granulovirus isolated from a laboratory colony of codling moth in British Columbia, Canada is identical or very similar to the Mexican isolate of Cydia pomonella granulovirus (CpGV-M). In addition to codling moth, the vims was found to cause mortality of secondary leafroller pests of North American orchards, including the fruit tree leafroller and the obliquebanded leafroller. A survey in apple growing regions of British Columbia indicates that 23% of the wild population of codling moth larvae carry CpGV.  相似文献

Multiple applications of hydrophobic kaolin particle film in apple orchards suppressed numbers of blossom weevil (Anthonomus pomorum), brown leaf weevil (Phyllobius oblongus), attelabid weevil (Caenorhinus pauxillus), leafhoppers (Empoasca vitis and Zygina flammigera) and green apple aphid (Aphis pomi) colonies. The kaolin treatments reduced the apple sawfly (Hoplocampa testudinea) fruit infestation on cultivar J. Grieve, and the fruit damage caused by oyster scale (Quadraspidiotus ostreaeformis), mussel scale (Lepidosaphes ulmi), early caterpillars, leaf rolling moths (Tortricidae), fruitlet‐mining tortrix moth (Pammene rhediella) and codling moth (Cydia pomonella). There was no effect on the number of colonies of rosy leaf curling aphid (Dysaphis devecta), nor on the fruit damage caused by common earwig (Forficula auricularia) and apple sawfly on cv. G. Delicious. The level of infestation of rosy apple aphid (Dysaphis plantaginea), leaf miner moths (Phyllonorycter blancardella, Lyonetia clerkella), and agromyzid flies (Phytomyza heringiana) increased in the kaolin‐treated plots. Kaolin treatments promoted woolly apple aphid (Eriosoma lanigerum) infestation, which became severe, while it reduced the abundance of polyphagous predators like F. auricularia, predaceous Heteroptera and Coleoptera, the red velvet mite (Allothrombium fuliginosum), spiders (Araneae) and the abundance of common black ant (Lasius niger). The treatments also reduced parasitism of the apple sawfly by the ichneumonid Lathrolestes ensator. Many weeks after ending the kaolin treatments, the number of predaceous Coleoptera and especially the number of spiders remained low in the kaolin‐treated plots.  相似文献

Codling moth granulosis virus (Cp GV) was produced in larvae of Cydia pomonella reared on artificial diet. The average yield of virus (9 × 109capsules/larva) was increased by raising the larvae on diet containing methoprene, a juvenile hormone analogue. In field trials in 1978 and 1979, one or two high-volume applications of Cp GV at 7 × 1010 capsules/litre achieved reductions in numbers of mature larvae and damaged fruit that were little different from those obtained by two applications of azinphos-methyl. A field trial in 1980 showed that at concentrations of Cp GV ≥ 6 × 108 capsules/litre, damaged diminished slowly with increasing virus concentration. Following virus application in 1980, Cp GV infectivity was reduced by half in 3 days, but some activity persisted at least 4–8 wk after spraying. The results indicate that codling moth GV is an effective control agent particularly in reducing numbers of larvae and the more severe forms of fruit damage.  相似文献

The functional response of adult females of the predatory mites Euseius (Amblyseius) finlandicus and Amblyseius andersoni to larvae and adult females of the fruit tree red spider mite Panonychus ulmi was determined on apple and peach leaf disks in the laboratory at 25°C and 16:8 (L:D). For adult females of P. ulmi the predation efficiency of E. finlandicus was higher on peach than on apple, whereas that of A. andersoni was higher on apple than on peach. Efficiency of predation on larvae of P. ulmi by either predator did not differ significantly between apple and peach. On both plants, A. andersoni had a higher predation rate than E. finlandicus on larvae of P. ulmi. It is concluded that in the laboratory the host plant has a substantial effect on predation efficiency of A. andersoni and E. finlandicus when they preyed on adults but not when they preyed on larvae of P. ulmi.  相似文献

The identification of effective solar protectants for field application of the codling moth, Cydia pomonella (L.), granulovirus (CpGV) is of interest to improve its efficacy and commercial viability as a biological pesticide. We evaluated several materials as potential adjuvants to protect CpGV from ultraviolet degradation. In laboratory tests with a solar simulator (9.36×106 J/m2), the addition of kaolin clay (Surround® WP at 3 and 6%, w/v), a paraffin wax-based emulsion (SPLAT? at 5%, v/v), and a bark extract trans-cinnamaldehyde combined with a film agent (both at 1%, v/v) did not significantly reduce larval entries or increase larval mortality in irradiated apples that were treated with a commercial CpGV product (Cyd-X). In semi-field tests in an apple orchard, a spray-dried lignin formulation containing CpGV (6.57×1012 OBs/ha) and a lignin-based adjuvant used with Cyd-X (both applied at 4.7–5.6 kg lignin/ha) significantly improved residual activity of CpGV compared with Cyd-X alone applied at the same rate. However, the benefits were short lived and could not be detected after 7 days. In orchard tests, we evaluated two additional refined lignin-products (Lignosulfonate and Vanisperse? CB at 5.61 kg/ha) and two particle film materials (kaolin clay, ‘Cocoon?’, and calcium with boron ‘Eclipse?’) as adjuvants for UV protection of Cyd-X (6.57×1012 OBs/ha) in tests against a dense codling moth infestation. Although all virus treatments were highly effective (causing ≥90% larval mortality), no significant effects of the adjuvant treatments could be detected. In these latter tests, the use of a silicone based wetting agent at 0.025% (v/v) may have been beneficial at increasing mortality among older larvae inside the fruit.  相似文献

Contact activities of flucycloxuron on immature stages of the two-spotted spider mite (Tetranychus urticae (Koch)) and the European red mite (Panonychus ulmi (Koch)) gradually decrease in the successive developmental stages. The levels of contact activity of flucycloxuron on larvae and protonymphs ofT. urticae andP. ulmi are of the same order. Deutonymphs ofT. urticae are less susceptible to contact activity than the similar stage ofP. ulmi. In adultT. urticae, the transovarial ovicidal activity was used as an indicator for cuticular penetration. More than 90% of the maximal penetration into adult mites occurs within 8 h. Reversibility of the transovarial activity was not observed after 24 h, but did occur after a subsequent 48 h stay on untreated leaves. The ovo-larvicidal activity of flucycloxuron onP. ulmi after treatment of apple leaves is strongly negatively influenced by leaf age, partly by lower retention of the spray liquid on the leaves. Leaf penetration was measured by application of flucycloxuron on leaf uppersides and assessment of the transovarial activity in mites (P. ulmi orT. urticae) infested on the undersides, one day after treatment. In this test system, leaf penetration was found to be strongly species dependent. Penetration was high in cucumber, moderate in French beans, cotton, roses and strawberry, but low in apple and pepper. Leaf penetration in French bean plants is drastically reduced at increasing leaf age. The overall positive effect of increase in relative air humidity on leaf penetration, is statistically highly significant (P=0.001) for French beans and almost significant (P=0.08) for cucumbers. WithT. urticae on French bean it was found that in this test flucycloxuron needs more than one day for maximal leaf penetration. Although in apple leaves penetration from uppersides was low, penetration from undersides was much higher. The influence of the nitrogen-content of apple leaves on the development of the fruit tree red spider mite (Metatetranychus ulmi (Koch)) was studied under laboratory and semi-field conditions.The experiments in the laboratory showed that a higher egg production per female in 24 hours, a lower mortality rate of the eggs, larvae and nymphs, and a shorter period of development of the male as well as the female mites was obtained on the leaves with a high nitrogen-content. However, these differences were statistically significant only in the case of egg production and mortality rate. From the experiments under semi-field conditions it became evident that the egg-female ratio on leaves with high nitrogen content originating from manured rootstocks was also significantly higher than this ratio on leaves with a low nitrogen content. In both series of experiments the differences gave a factor of 1.6.The experiments confirm the results, we obtained earlier from perennial field trials where an effect of manuring of the apple trees on the development of M. ulmi was found.It is concluded that the mortality of predators of M. ulmi, caused by chemical treatment, is not the only reason why the red spider mite populations are stimulated in well kept orchards. The higher level of nitrogen content of the leaves, brought about by cultural practice, in particular manuring, may be of considerable importance also.
Zusammenfassung Der Einfluß des Stickstoffgehaltes der Blätter auf die Entwicklung der Roten Obstbaum-Spinnmilbe, Metatetranychus ulmi (Koch), wurde unter Laboratoriums- und halbfeldmäßigen Bedingungen geprüft. Steigender Stickstoffgehalt ergibt eine höhere Eiproduktion pro Weibchen und je 24 Stunden, eine geringere Absterberate der Eier, Larven und Nymphen und eine höhere Entwicklungsgeschwindigkeit sowohl der Männchen wie der Weibchen. Im Falle der Eiproduktion und der Mortalitätsraten waren die Unterschiede gesichert. Unter den Halb-Freilandbedingungen wurde es deutlich, daß das Ei-Weibchen-Verhältnis bei hohem Stickstoffgehalt der Blätter gedüngter Wurzelstöcke ebenfalls signifikant höher war als auf

Summary Wire cages with different-sized meshes were placed on trunks and around leaves at different heights in oak trees and in forest litter. Gypsy moth, Lymantria dispar, instars II–V tethered with threads were placed in each cage (instars II–III only in leaf cages) as well as outside the cages. Predation of larvae decreased from near ground to mid-crown in trees and was highest in litter and very low on leaves. Predation in litter was not strongly related to cage type, suggesting that small, invertebrate predators were active there, but IV–V instars on trunks were mainly killed by relatively large predators, probably forest mice and shrews. Influences of the time of day and weather on predation were evaluated by observing tethered V–VI instars in litter and on tree trunks hourly. Ants, particularly carpenter ants (Camponotus ferrugineus) and Formica sp., and probably vertebrates, were conspicuous predators in the litter. Ants were most active at lower relative humidities, while other predators were apparently not influenced by humidity. No daily activity rhythms of predation were noted. Invertebrates appear to be important predators of larvae only in the litter whereas vertebrates, such as forest mice and shrews, also readily attack larvae on tree trunks.  相似文献

Trophic cascades may purportedly be more common in aquatic than terrestrial food webs, but herbivory on freshwater vascular plants has historically been considered low. Water lilies are an exception, suffering severe grazing damage by leaf beetles. To test whether a central prediction of cascade models—that predator effects propagate downwards to plants—operates in a macrophyte-based food web, we experimentally manipulated predation pressure on a key herbivore of water lilies in the littoral zone of a lake in Michigan, USA. Field experiments comprised combinations of caging treatments to alter the number of predators (larvae of the ladybird beetle Coleomegilla maculata) that hunt the grazers of the macrophytes (larvae of the leaf beetles Galerucella nymphaeae) on the leaves of the water lily Nuphar advena. Predatory larvae of the ladybird beetles significantly reduced grazing damage to water-lily leaves by 35–43%. The predators reduced plant damage chiefly via density-mediated effects, when lower densities of grazers translated to significant declines in plant damage. Plant damage caused by the surviving herbivores was less than predicted from individual grazing rates under predator-free conditions. This suggests that trait-mediated effects may possibly also operate in this cascade. The observed strong effect of predators on a non-adjacent trophic level concurs with an essential component of the trophic cascade model, and the cascade occurred at the ecotone between aquatic and terrestrial habitats: Nuphar is an aquatic macrophyte with emergent and floating leaves, whereas both beetle species are semi-terrestrial and use the dry, emergent and floating leaves of the water lily as habitat. Also, the cascade is underpinned by freshwater macrophytes—a group for which trophic processes have often been underappreciated in the past.  相似文献

The occurrence of resistance of codling moth (CM, Cydia pomonella L.) to Cydia pomonella granulovirus (CpGV) used as biological control agent revealed the need for fast and reliable resistance monitoring methods. Here, we describe the development of a laboratory resistance test that is directly performed on larvae extracted from infested apples. This test is based on a 14‐day bioassay at a discriminative CpGV concentration of 2 × 105 occlusion bodies/ml diet and can be applied to L1–L4 larvae. Information on virus resistance can be obtained within <4 weeks. In a survey, CM larvae were isolated and tested from 6698 apples from 10 different orchards in Germany, Austria, Switzerland, Italy and the Netherlands. We identified seven CM populations resistant or partly resistant to CpGV isolates. Although some of the orchards were treated with commercial CpGV products, this method allowed us to obtain reliable information about the resistance status of the examined populations.  相似文献

A method for screening codling moth granulovirus (CpGV) formulation sensitivity to sunlight using specially prepared half apples and a solar simulator is described. The half apple preparation allows an even coverage of virus over the surface of the fruit that would not be possible using whole apples. Leaves and artificial medium were not usable for extended periods of exposure in the solar simulator due to excess drying. Fruit was sprayed with 10−3 and 10−5 dilutions of three commercial formulations of CpGV (Carpovirusine, Cyd-X, and Virosoft) and infested with codling moth neonates. Half of the sprayed fruit was exposed to 650 W/m2 for 4 h in an Atlas Suntest CPS solar simulator resulting in an accumulated radiant energy of 9.36 × 106 J/m2 before they were infested with neonate codling moth larvae. Spraying non-irradiated fruit with the 10−3 dilution of Cyd-X and Virosoft resulted in nearly 100% mortality of neonate larvae. Irradiation reduced viral activity by 71-98% at the 10−3 dilution and by up to 32% at the 10−5 dilution relative to non-irradiated fruit. The procedures utilized enabled good preservation of the fruit throughout the incubation period and minimized invasion of the fruit by plant pathogens and saprophytic organisms. This laboratory method for screening candidate formulations and potential UV protectants could conserve time and resources by eliminating adjuvants with less potential in laboratory tests and field testing only the most promising candidates. Different numbers of larvae of the beetle Phaedon cochleariae (F.) and the moth Plutella maculipennis (Curt.) were fed on the leaves of turnip plants (variety Early Milan White) and radish (variety French Breakfast) growing in a constant-environment room. Turnips grew more slowly than radish, were less affected by insect feeding and were affected more by Phaedon than by Plutella. Phaedon larvae severed leaf veins and rasped the leaf surface, thus killing more leaf tissue by desiccation than they ate. They remained on and greatly damaged the older leaves and their feeding reduced yield. Plutella larvae fed on leaves of all ages, which they ate cleanly, leaving the veins. They pupated sooner than Phaedon and did less damage to older leaves, which grew larger and lived longer than corresponding leaves on intact plants. This compensatory growth by turnip prevented loss of yield and may have increased it, as indicated by dry weight of the roots. Both species of insect decreased the yield of radish similarly. Loss of yield of turnip caused by Phaedon or of radish caused by both species of insects was negatively and linearly proportional to the number of larvae that fed on the leaves.  相似文献
