Ten novel polymorphic microsatellite loci were isolated and characterized from the Balearic shearwater (Puffinus mauretanicus), a critically endangered seabird. The developed loci revealed a relatively low number of alleles per locus, as well as low levels of polymorphism (HO = 0.377 ± 0.241). One of the loci appeared to be W‐linked. All polymorphic loci were successfully amplified in its closely related species, the Yelkouan shearwater (Puffinus yelkouan). These microsatellite markers would be useful for assessing population structure in the Balearic shearwater and the possible hybridization process between both shearwaters species.  相似文献

Polymorphic microsatellite loci were identified in order to study golden eagle (Aquila chrysaetos) population fragmentation. Twenty‐six published Aquila and eight published Haliaeetus microsatellite loci were tested for polymorphism in A. chrysaetos. Fifteen loci were polymorphic with between two and six alleles detected per locus. Observed heterozygosity ranged from 0.15 to 0.77 among 177 unrelated individuals from Scotland. There was no evidence for null alleles. Two pairs of loci (Hal‐10 & Aa15 and Hal‐10 & Aa26) displayed linkage disequilibrium.  相似文献

Twenty‐six polymorphic microsatellite markers were isolated from (AC)n and (AG)n microsatellite‐enhanced genomic libraries of the gray, short‐tailed opossum Monodelphis domestica. All 26 loci showed high allelic diversity, with allele numbers ranging from five to 11 in a subset of 35 animals. Normal Mendelian inheritance was confirmed for 24 loci by analysing allelic segregation in 10, two‐generation, families. Non‐amplifying (null) alleles were detected at two loci, which we recommend be used only if pedigree data are available. We conclude that all of these microsatellite markers would be useful for quantitative trait locus mapping and population genetic studies.  相似文献

Twenty‐six microsatellite loci were isolated and characterized from the mangrove species Rhizophora mangle using (GT)n and (CT)n repeats. Eighty‐four per cent of the clones contained microsatellite sequences; the most common dinucleotides were the (GA/CT) and (CA/GT) repeats. Ten primers were selected to investigate the polymorphism among individuals of R. mangle from two natural populations of the Colombian Pacific Coast. The observed heterozygosity per locus varied from 0.20 to 0.80, the power of discrimination was 0.32–0.84 and the power of exclusion was 0.03–0.75. This set of microsatellites offers an efficient tool for population genetics studies on this species.  相似文献

Fifteen microsatellite loci were isolated from the Peruvian tropical plant‐ant Allomerus octoarticulatus cf. demerarae (Hymenoptera: Myrmicinae) and their polymorphism was characterized. High levels of within‐population variation were observed at most loci, with number of alleles ranging from one to 21, and heterozygosity from 0 to 1 per population sample. Cross‐species amplification of these loci was also tested in one other species of the ant genus Allomerus (Allomerus decemarticulatus), displaying similar life history.  相似文献

High levels of within‐population variation were observed at most loci, with number of alleles ranging from one to 16, and heterozygosity from 0 to 0.929 per population sample. Eight highly polymorphic microsatellite loci were developed for Virola flexuosa from a (CA)n‐enriched genomic library for population and seed dispersal studies in eastern Ecuador. Loci show a high level of variation with the number of alleles ranging from 13 to 27. Observed and expected heterozygosities were from 0.313 to 0.896 and 0.552 to 0.937, respectively. The high levels of polymorphism and exclusionary power of the developed markers will likely prove very useful in direct measurement of seed dispersal.  相似文献

Black locust (Robinia pseudoacacia) is an economically and ecologically important tree species in the world. We isolated seven polymorphic microsatellite loci from R. pseudoacacia using a dual‐suppression‐PCR technique. These loci provided microsatellite markers with high polymorphism ranging from three to 12 alleles per locus and expected heterozygosity between 0.538 and 0.944. The markers are now available for more detailed investigation of population genetic structure and pollen and seed dispersal.  相似文献

Seven polymorphic microsatellite markers were isolated and characterized for golden‐striped salamander, Chioglossa lusitanica (Bocage 1864), a salamandrid endemic to the northwestern part of the Iberian Peninsula, from genomic libraries enriched for (GATA)8 (GACA)8 (ATG)9 and (CA)16. These loci were screened in 37–51 individuals from two populations. The number of alleles per locus ranged from five to 19. Heterozygosity ranged from 0.241 to 1.0. The high level of polymorphism revealed by these loci will be extremely useful for the study of population structure and evolutionary history of this species.  相似文献

Rhizophora stylosa is an ecologically important mangrove tree species that is found in tropical and subtropical regions. We isolated five polymorphic microsatellite loci from R. stylosa using a dual‐suppression‐polymerase chain reaction technique. These loci provided microsatellite markers with polymorphism of four alleles in each locus for overall population. The mean expected heterozygosities ranged from 0.113 to 0.473.  相似文献

We report on the isolation and characterization of five polymorphic microsatellites in the gorgonian Pseudopterogorgia elisabethae from genomic DNA‐enriched libraries. Forty‐four microsatellites were screened from the libraries with the oligonucleotide probe (CA)12. Five of the screened microsatellites were polymorphic. The levels of polymorphism found in 50 genotyped individuals from a single population suggest that microsatellites are useful tools for the study of genetic variation in gorgonians. These are the first microsatellite loci reported from any gorgonian species.  相似文献

High levels of within‐population variation were observed at most loci, with number of alleles ranging from two to 15, and heterozygosity from 0.050 to 0.925. Cristaria plicata was an important freshwater mussel for pearl culture in China. 18 polymorphic microsatellite markers were isolated and characterized using (CA)15-enriched genomic library of C. plicata. These loci showed high levels of genetic polymorphism testing on 60 individuals sampled from Poyang Lake of Jiangxi Province, China. The number of alleles per locus ranged from 4 to 18. The expected (H E) and observed heterozygosities (H O) were 0.7232–0.8961 and 0.0000–1.0000, respectively. Four microsatellite loci were significantly deviated from Hardy–Weinberg equilibrium and no linkage disequilibrium was found. These microsatellite loci will be useful for assessment of genetic diversity and population structure in C. plicata.  相似文献

Here we present nine novel, polymorphic microsatellite loci developed for the scleractinian coral Acropora millepora (Acroporidae) from the Great Barrier Reef. In addition, we have assessed the specificity and polymorphism of five microsatellite loci previously developed for a Caribbean congener and one locus developed for an Indo‐Pacific congener. Only one of the latter six loci produced reliable results, yielding a total of 10 polymorphic microsatellite loci for A. millepora. Variability was tested on 20–23 individuals from a single population, plus another ~10 individuals from each of three different populations, resulting in five to 20 alleles per locus.  相似文献

We isolated 56 common buzzard (Buteo buteo) microsatellite loci from (AC)n‐ and (AAAG)n‐enriched genomic libraries. Eleven loci were tested on 90 individuals from Eastern Westphalia in Germany, yielding two to 17 alleles per locus (average 5.7) and expected heterozygosities ranging from 0.11 to 0.93 (average 0.53). These highly variable microsatellite markers provide a powerful means for investigating population genetic diversity in the common buzzard, a little‐understood aspect of this widespread species.  相似文献

A set of 11 polymorphic microsatellite markers has been developed and characterized for the critically endangered species Pieris amamioshimensis. Fifty‐nine individuals of an ex‐situ population were used to identify these markers. The total number of alleles for each locus ranged from 3 to 9, with an average of 5.4. The expected heterozygosities (HS) and observed heterozygosities (HO) ranged from 0.47 to 0.77 and 0.22 to 0.88, respectively. In total, four loci exhibited significant deviations from Hardy–Weinberg equilibrium: two loci showed significant heterozygosity excess and the other two loci showed significant heterozygosity deficit. The polymorphism information content (0.43 ≤ PIC ≤ 0.73), the probability of exclusions (PE1 = 0.9565, PE2 = 0.9969 and PE3 = 0.9999) and probabilities for identity (PI = 3.78 × 10?9 and PI‐Sib = 2.35 × 10?4) suggest that these markers are useful for estimating not only genetic diversity but also parentage, for the ex‐situ conservation management of populations.  相似文献

The purpose of this study was to assess the genetic characteristics of six breeds of Chinese local sheep using 19 microsatellite loci and to effectively validate statistical methods for individual assignment based on informative microsatellites. All the six breeds deviated from Hardy–Weinberg equilibrium expectations, while the majority of markers complied. The polymorphism information content (PIC) of overall loci for the six populations ranged from 0.283 (SRCRSP5) to 0.852 (OarVH72). Tibetan sheep were the most diverse population with the highest mean allelic richness (6.895), while Ujmuqin (UQ) harboured the lowest allelic richness (6.000). The F‐statistics for the six populations were FIS = ?0.172, FIT = ?0.082 and FST = 0.077, respectively. Furthermore, the pair‐wise FIS revealed a moderate genetic differentiation among populations (P < 0.01), indicating that all breeds can be considered genetically independent entities. The lowest genetic differentiation was between Tengchong (TC) and UQ (FST = 0.041), and the highest one was between TC and Fat‐tailed Han (FST = 0.111). In comparing the three statistical models, we note that the seven microsatellite loci (MAF65, OarJMP58, SRCRSP9, MCM140, OarAE129, BM8125 and SRCRSP5) commonly used for individual assignment will ensure a powerful detection of individual origin, with accuracy up to 91.87%, when the likelihood‐based method is used. Overall, these findings shed light onto the genetic characteristics of Chinese indigenous sheep and offer a set of microsatellite loci that is simple, economic and highly informative for individual assignment of Chinese sheep.  相似文献

Simple sequence repeat‐enriched library screening and expressed sequence tag database mining were adopted to develop microsatellite markers for sea cucumber (Apostichopus japonicus). Eighty‐three microsatellite loci were selected for polymorphism assessment using 48 individuals. The results showed that 45 novel loci were polymorphic. The number of alleles ranged from two to 16, and the values of observed and expected heterozygosities varied from 0 to 0.9375 and from 0.1135 to 0.9674, respectively. No significant linkage disequilibrium between pairs of loci was found and 26 loci conformed to the Hardy–Weinberg equilibrium. These markers are therefore a potential tool for studies in the population structure and linkage map construction for A. japonicus.  相似文献

Microsatellite markers of Crinipellis perniciosa, with three and four repeats, were developed from sequence database and evaluated for their usefulness in detecting genetic polymorphism. Thirty‐three primers produced unambiguous amplification products of 28 microsatellite‐containing loci and 14 microsatellite‐like polymorphic loci, with two to seven alleles at each locus. Three loci were useful to distinguish isolates from different biotypes and isolates from different countries. Amplification of the markers in the closely related fungi Moniliophthora roreri indicates that their usefulness in population's studies may go beyond the present study of the C. perniciosa and may have applications in population genetics of M. roreri.  相似文献

Two microsatellite‐enriched libraries [(CAGA)n, (TAGA)n] were constructed using pooled DNA from three cyprinid species native to the Sacramento–San Joaquin Delta of California: Sacramento splittail (Pogonichthys macrolepidotus); Sacramento pikeminnow (Ptychocheilus grandis); and tui chub (Siphateles bicolor). Primers were designed for 105 loci and tested for levels of polymorphism in five cyprinid species found in the Delta: Sacramento splittail, Sacramento pikeminnow, tui chub, hitch (Lavinia exilicauda), and Sacramento blackfish (Orthodon microlepidotus). Fifty‐one loci were polymorphic for at least one species and 31 loci were polymorphic for multiple species. The number of polymorphic loci per species ranged from 16 to 26.  相似文献
