AB-8大孔吸附树脂对红花桑寄生总黄酮静态吸附和动态洗脱的效果,受提取液质量浓度、pH值及环境温度、振速以及洗脱剂乙醇浓度、流速等因素影响。试验表明,提取液质量浓度和pH值对AB-8树脂的吸附效果有显著影响,其吸附分离总黄酮的工艺条件为:浓度为1.2~2.0 mg/ml、pH 3.0~4.0的红花桑寄生提取液,置于摇床上,于室温条件下振荡(振速160 r/min)吸附2~3 h,然后用5倍于树脂体积(5BV)的50%乙醇以1.5 ml/min流速进行柱上动态解吸。AB-8树脂对红花桑寄生总黄酮的饱和吸附量可达29.0 mg/g,动态洗脱率达95.0%,获得产品中黄酮纯度为46.0%,得率为5.5%。  相似文献

AB-8型树脂对无患子皂苷的动态吸附与解吸性能   总被引:1,自引:0,他引:1  
采用单因素实验法,以树脂的饱和吸附量、产品的得率及纯度为指标,考察各因素对动态吸附与解吸的影响,优化大孔树脂动态吸附分离无患子皂苷的工艺条件。结果表明,当pH为4.5的无患子皂苷水提液以流速1 mL/min通过高径比为5.4∶1的吸附柱时,AB-8树脂对无患子皂苷的饱和吸附量达568 mg/g;采用1.5 BV的95%乙醇以1 mL/min的流速洗脱吸附后的树脂,产品得率为83.48%、纯度达93.00%;树脂重复使用8次后其吸附解吸性能基本不变。该方法可以用于无患子皂苷的工业化分离提纯。  相似文献

通过采用11种大孔吸附树脂对榅桲多酚粗提物的静态吸附和解吸试验,筛选出适合分离纯化榅桲总多酚的最优树脂,并对其动态吸附特性和影响因素进行研究。结果表明:最佳纯化工艺条件为:AB-8大孔吸附树脂做吸附填料,上样液质量浓度0.04 g/m L,以2 BV/h的吸附速率进行吸附,上样量为11 BV,用3 BV蒸馏水除杂后,以50%乙醇溶液为洗脱剂,洗脱液用量为4 BV,洗脱流速控制在2 BV/h。经AB-8树脂吸附富集后,样品总多酚质量浓度达到了30.11 mg/g,是粗提物(9.55 mg/g)的3倍。对纯化前后的榅桲总多酚进行PTP1B的抑制作用研究,结果显示,榅桲总多酚对PTP1B有较强的抑制作用,即榅桲总多酚纯化前的IC50为78.14μg/m L,用AB-8树脂纯化后所得总多酚的IC50为16.12μg/m L。  相似文献

探讨了大孔吸附树脂DM-130对甘草酸的吸附性能及原液浓度、pH值、流速对此树脂吸附性能的影响.结果表明,DM-130树脂对甘草酸的吸附性能好,易于洗脱,分离效果好,产品纯度可达94.676%;正交试验表明,pH为5.4±,原液浓度为10 mg/ml,以2 BV/h的流速为最佳处理;洗脱液采用3 BV 10%的乙醇最经济.  相似文献

研究了大孔吸附树脂对乳清分离蛋白(WPI)酶解液的吸附特性。比较了6种大孔吸附树脂对WPI酶解物的静态吸附率与解吸附率。结果表明,DA201-C大孔吸附树脂最适合WPI酶解物的分离,其对WPI酶解液的动态吸附条件为:上样液浓度:10mg/mL;洗脱剂:75%乙醇溶液;洗脱剂流速:1BV/h。  相似文献

通过比较8种大孔吸附树脂对烟草绿原酸的吸附分离性能,筛选出适合分离烟草绿原酸的树脂,并对其动态吸附特性进行研究.结果表明,XDA-1树脂对烟草绿原酸不仅吸附量大,而且解吸率高,适合烟草绿原酸的分离富集.该树脂吸附分离烟草绿原酸的工艺参数为:上柱液浓度3.5 mg/mL,pH 3.0,流速3倍柱床体积/h;以6倍柱床体积的40%乙醇进行洗脱,解吸附效果最佳,绿原酸总回收率为80.06%,初步吸附分离得到的产品中绿原酸含量为39.20 g/100 g.  相似文献

主要研究了从蛹虫草发酵液中提取虫草素的工艺,确定了大孔吸附树脂AB-8及十八烷基键合硅胶反相层析柱对蛹虫草发酵液中虫草素的分离条件;吸附最佳工艺条件为上样液p H6,上样浓度0.4mg/m L,上样量5BV,吸附流速1.0BV/h;解吸最佳工艺条件包括解吸液20%乙醇,解吸体积15BV,解吸流速4BV/h。得到的解吸液进一步以十八烷基键合硅胶分离,条件为上样液p H6,梯度洗脱,流动相为p H6的水和p H6的乙醇。洗脱液通过结晶和重结晶得到精制虫草素。三步得到的虫草素纯度分别达到40%、90%和99%以上。  相似文献

以苹果渣多酚类化合物为吸附模型,制备一种可有效吸附分离苹果渣多酚的磁性树脂,研究其吸附、解吸效能,并就吸附和解吸效能、使用次数与普通大孔树脂进行比较。结果显示,5种普通树脂中D280、H103、AB-8对苹果渣多酚的吸附率均高于80%,AB-8、D3520树脂对苹果渣多酚的解吸率均高于90%,AB-8树脂的吸附率和解吸率最好;磁性大孔树脂吸附0.5 h后即可达到吸附平衡,吸附平衡后剩余液多酚质量浓度低于AB-8树脂吸附平衡后剩余液多酚质量浓度,吸附效能明显优于AB-8树脂;磁性大孔树脂解吸效能也略优于AB-8树脂;使用4 次后,磁性大孔树脂的吸附率仍然高于70%,稳定性优于AB-8树脂。可见,磁性大孔树脂是一种良好的苹果渣多酚吸附剂,有较好的工业化应用前景。  相似文献

研究了大孔树脂吸附法从苦楝树皮的提取液中提纯苦楝素的工艺条件及参数,并筛选出较为理想的大孔吸附树脂。研究结果表明,S-8型吸附树脂的静态饱和吸附量明显大于AB-8型和NKA-9型。该树脂吸附提纯苦楝素的优化吸附条件为吸附温度40℃,溶液pH值8.0,上柱液质量浓度9.127mg/mL,溶液流速2BV/h;优化的解吸条件为:洗脱剂为70%乙醇-水溶液,溶液流速1BV/h,洗脱剂用量为8倍量树脂体积。在优化条件下,可以得到含量达75.2%的苦楝素提取物,表明S-8树脂对苦楝素有良好的吸附选择性。  相似文献

考察大孔吸附树脂吸附分离番石榴叶总黄酮的工艺条件.以静态饱和吸附量、静态洗脱率、动态饱和吸附量、动态洗脱率为考察指标,比较了D101、AB-8两种大孔树脂分离纯化番石榴叶总黄酮的优劣.又以总黄酮回收率为指标,对最佳树脂吸附工艺参数进行了研究.在考察的2种树脂中,AB-8型树脂最适于番石榴叶总黄酮的分离纯化,其工艺条件为:4倍树脂体积50%乙醇洗脱,速度2mL/min.树脂可重复使用4次.其平均总黄酮回收率为87.47%.所得总黄酮纯度为74.03%  相似文献

Equilibrium molecular dynamics simulations were conducted to study the competitive adsorption and diffusion of mixtures containing n-octane and carbon dioxide confined in slit-shaped silica pores of width 1.9 nm. Atomic density profiles substantiate strong interactions between CO2 molecules and the protonated pore walls. Non-monotonic change in n-octane self-diffusion coefficients as a function of CO2 loading was observed. CO2 preferential adsorption to the pore surface is likely to attenuate the surface adsorption of n-octane, lower the activation energy for n-octane diffusivity, and consequently enhance n-octane mobility at low CO2 loading. This observation was confirmed by conducting test simulations for pure n-octane confined in narrower pores. At high CO2 loading, n-octane diffusivity is hindered by molecular crowding. Thus, n-octane diffusivity displays a maximum. In contrast, within the concentration range considered here, the self-diffusion coefficient predicted for CO2 exhibits a monotonic increase with loading, which is attributed to a combination of effects including the saturation of the adsorption capacity of the silica surface. Test simulations suggest that the results are strongly dependent on the pore morphology, and in particular on the presence of edges that can preferentially adsorb CO2 molecules and therefore affect the distribution of these molecules equally on the pore surface, which appears to be required to provide the effective enhancement of n-octane diffusivity.  相似文献

Streamline Direct CST I is a new type of ion exchanger with multi-modal functional groups, specially designed for an expanded bed adsorption (EBA) process, which can capture directly the proteins from the high ionic strength feedstocks with a high binding capacity. In this study, an experimental study is carried out for two-component proteins (BSA and myoglobin) competitive adsorption and desorption in an expanded bed packed with Streamline Direct CST I. Based on the measurements of the single- and two-component bovine serum albumin (BSA)/myoglobin adsorption isotherm on Streamline Direct CST I, the binding and elution conditions for the whole EBA process are selected; and then frontal analysis for a longer timescale and column displacement experiments in a fixed bed (XK16/20 column) are carried out to evaluate the two-component proteins (BSA and myoglobin) competitive adsorption and displacement on Streamline Direct CST I. Finally, the feasibility of capturing both BSA and myoglobin by an expanded bed packed with Streamline Direct CST I is addressed in a Streamline 50 column packed with 300 mL Streamline Direct CST I.  相似文献

The phosphate adsorption maximum as calculated by the Langmuir equation was used to predict the fertilizer P requirement of wheat (Triticum aestivium, cv. Caldwell) on both virgin and cultivated Decatur clay loam (clayey, kaolinitic, thermic, Rhodic Paleudult) and Hartsells sandy loam soils (fine-loamy, siliceous, thermic, Typic Hapludult). Soils with higher adsorption maximum were found to require more fertilizer P than soils with lower adsorption maximum. For soils 25% saturation of the adsorption maximum gave the optimum dry matter yield. This corresponded to equilibrium P concentration of 0.45 mg L−1 for Decatur cultivated and 0.31 mg L−1 for Decatur and Hartsells virgin soils for optimum dry matter yield. These values are within the range of those reported previously by other investigators working with different soils.  相似文献

本文通过考察八种大孔吸附树脂对淫羊藿甙的吸附分离性能,筛选出了AB-8树脂作为分离纯化淫羊藿甙的介质。对该树脂的吸附性能研究表明其对浸提物中的淫羊藿甙有良好的吸附选择性,静态饱和吸附容量和动态吸附容量分别为22.97和16.20mg/mL。通过柱层析实验确定了AB-8树脂分离淫羊藿甙的工艺,经一步层析可将淫羊藿甙的纯度从2.78%提高到27%,回收率达97.3%。  相似文献

An expanded bed adsorption (EBA) column (5 cm diameter) has been modified to allow the abstraction of liquid samples from various positions along the height of an expanded bed. As the adsorbent particles were fluidized, in-bed monitoring of key component concentrations during feedstock application, washing and elution was achieved by the withdrawal of liquid samples from the voids within the expanded bed through ports along the wall of the column. Component levels in the withdrawn streams can be assayed using on-line analytical chromatography or samples can be collected and assayed off-line. On-line monitoring can be used to control the duration of the loading stage and as a tool to provide information about the hydrodynamic and adsorption/desorption processes that occur during expanded bed adsorption. Studies of residence time distributions indicated that the modifications to the column do not significantly affect liquid dispersion. Using the adsorption of glucose-6-phosphate dehydrogenase from yeast homogenate on Streamline DEAE as a model system, comparison of breakthrough curves for runs when in-bed monitoring was and was not performed also suggested that separation efficiency is not appreciably affected by in-bed sampling.  相似文献

采用等温吸附法,研究了重金属铜、铅、镉、镍在膨润土中的吸附特征,发现膨润土对铜、铅的吸附明显强于镉、镍,吸附强度大小顺序为Pb2 >Cu2 >Ni2 >Cd2 。Langmuir和Freundlich方程对这4种金属离子等温吸附的拟合均呈极显著关系。Pb2 、Cd2 、Ni2 分别与Cu2 的双组分竞争吸附表明,黏土矿物对4种离子具有"选择性吸附"。在Pb2 、Ni2 、Cd2 的存在条件下,黏土矿物对Cu2 的吸附产生不同程度的下降;100mg/LCu2 对Pb2 的影响不大,但可完全抑制Ni2 、Cd2 的吸附。建立了IAS和LCA模型来预测Pb2 与Cu2 的双组分竞争吸附,并对LCA模型进行修正,提出了更符合实际情况的竞争吸附模型。文章最后用LCA修正模型对Pb2 与Cu2 的双组分竞争吸附进行了模拟。  相似文献

Adsorption/desorption of toluene on montmorillonite, illite, and kaolinite was studied using the batch equilibrium method. The isotherms measured fit the Freundlich equation (r2 >0.95). Montmorillonite adsorbed more toluene than illite or kaolinite; the adsorption of toluene on illite and kaolinite was not significantly different. Adsorption of toluene by montmorillonite showed an exponential increase as the ratio of toluene to clay was increased from 5 to 100. The rate studies showed that 62% of the adsorption was completed within 6 h. A rapid desorption was observed initially, followed by slow desorption after 1 h. The desorption rate decreased as the time of adsorption was increased. Almost all of the adsorbed toluene was extracted with water from the clay when the adsorption time was 0.1 h, but only 61% of the toluene could be desorbed when the adsorption time was 24 h.  相似文献
