As genome and cDNA sequencing projects progress, a tremendous amount of sequence information is becoming publicly available. These sequence resources can be exploited for gene discovery and marker development. Simple sequence repeat (SSR) markers are among the most useful because of their great variability, abundance, and ease of analysis. By in silico analysis of 10,232 non-redundant expressed sequence tags (ESTs) in pepper as a source of SSR markers, 1,201 SSRs were found, corresponding to one SSR in every 3.8 kb of the ESTs. Eighteen percent of the SSR–ESTs were dinucleotide repeats, 66.0% were trinucleotide, 7.7% tetranucleotide, and 8.2% pentanucleotide; AAG (14%) and AG (12.4%) motifs were the most abundant repeat types. Based on the flanking sequences of these 1,201 SSRs, 812 primer pairs that satisfied melting temperature conditions and PCR product sizes were designed. 513 SSRs (63.1%) were successfully amplified and 150 of them (29.2%) showed polymorphism between Capsicum annuum ‘TF68’ and C. chinense ‘Habanero’. Dinucleotide SSRs and EST–SSR markers containing AC-motifs were the most polymorphic. Polymorphism increased with repeat length and repeat number. The polymorphic EST–SSRs were mapped onto the previously generated pepper linkage map, using 107 F2 individuals from an interspecific cross of TF68 × Habanero. One-hundred and thirtynine EST–SSRs were located on the linkage map in addition to 41 previous SSRs and 63 RFLP markers, forming 14 linkage groups (LGs) and spanning 2,201.5 cM. The EST–SSR markers were distributed over all the LGs. Native to Brazil,Schinus terebinthifolius Raddi, of the family Anacardiaceae, has been commonly cultivated in Florida for over 50 years as a dooryard ornamental. Use of its sprays of showy red fruits for Christmas decoration gave rise to the popular misnomer “Florida holly.” Too late it was found to become a large, spreading tree; aggressive seedlings began springing up near and far. Jungles ofSchinus have crowded out native vegetation over vast areas of Florida and the Bahamas, as in all the islands of Hawaii. When in bloom, the tree is a major source of respiratory difficulty and dermatitis; the fruits, in quantity, intoxicate birds and cause fatal trauma in four-footed animals. The abundant nectar yields a spicy commercial honey and beekeepers are opposed to eradication programs.  相似文献

Quantitative trait locus (QTL) mapping for fruit weight and shape in pepper (Capsicum spp.) was performed using C. chinense and C. frutescens introgression lines of chromosomes 2 and 4. In chromosome 2, a single major fruit-weight QTL, fw2.1, was detected in both populations that explained 62% of the trait variation. This QTL, as well as a fruit-shape QTL, fs2.1, which had a more minor effect, were localized to the tomato fruit-shape gene ovate. The cloned tomato fruit-weight QTL, fw2.2, did not play a major role in controlling fruit size variations in pepper. In chromosome 4, two fruit-weight QTLs, fw4.1 and fw4.2, were detected in the same genomic regions in both mapping populations. In addition, a single fruit-shape QTL was detected in each of the mapping populations that co-localized with one of the fruit-weight QTLs, suggesting pleiotropy or close linkage of the genes controlling size and shape. fw2.1 and fw4.2 represent major fruit-weight QTLs that are conserved in the three Capsicum species analyzed to date for fruit-size variations. Co-localization of the pepper QTLs with QTLs identified for similar traits in tomato suggests that the pepper and tomato QTLs are orthologous. Compared to fruit-shape QTLs, fruit-weight QTLs were more often conserved between pepper and tomato. An in vitro regeneration protocol has been developed from half-seed explants of a mild (cv. New Mexico-6) and a pungent (cv. Rajpur Hirapur) chile pepper (Capsicum annuum L). Imbibed seeds were cut into two parts such that one portion contained the cotyledons and a part of the hypocotyl (part A) while the other part had the proximal part of the hypocotyl and the radicle (part B). These explants were cultured on MS medium with or without cytokinins (KIN, BA, ZEA, 2iP). Cytokinins dramatically increased both the percentage of explants forming buds as well as the number of buds per explant, and also hastened the rate of bud production. The relative efficacy of cytokinins in inducing the formation of leafy buds varied in the two cultivars. However, the best response was observed with ZEA in both cultivars. The highest percentage of bud formation was recorded after presoaking part B explants for 72 hours. The elongation growth of leafy buds was severely inhibited in the continuous presence of high concentrations of cytokinins, and frequently the buds became quite thick, ill-defined and vitreous. Within 3–5 weeks of transfer to Magenta boxes containing vermiculite and soil (1:3), 70–85% of the rooted hypocotyls developed 1–2 elongated shoots. Temperature is one of the most important environmental factors that influence plant growth and development. Recent studies imply that plants show various responses to non-extreme ambient temperatures. Previously, we have found that a pepper cultivar cv. Sy-2 (Capsicum chinense) shows developmental defects at temperatures below 24°C. In this study, to gain new insights into the temperature sensitivity of cv. Sy-2, temperature-sensitive genes were screened using microarray techniques. At restrictive temperature of 20°C, almost one-fourth of the 411 up-regulated genes were defense related or predicted to be defense related. Further expression analyses of several defense-related genes showed that defense-related genes in cv. Sy-2 were constitutively expressed at temperatures below 24°C. Moreover, accumulation of high level of salicylic acid (SA) in cv. Sy-2 grown at 20°C suggests that the defense response is activated in the absence of pathogens. To confirm that the defense response is induced in cv. Sy-2 below 24°C, we evaluated the resistance to biotrophic bacterial pathogen Xanthomonas campestris pv. vesicatoria and necrotrophic fungal pathogen Cercospora capsici. Cv. Summary In vitro plantlet regeneration was obtained from cultured cotyledon and young leaf explants of five Indian chile pepper cultivars (Capsicum annuum L. evs. Gujarat-1, Gujarat-2, Guntur-4, Selection-49, and Jwala). Adventitious shoot buds (ASB) were regenerated directly from cotyledon and young leaf explants in all the five cultivars on media containing benzyladenine (BA) alone or in combination with 1-naphthaleneacetic acid (NAA). Regeneration frequency was highly influenced by cultivar explant type, media combination and their interactions, except the interaction between cultivar and explant, for number of ASB per explant. Percent contribution of individual source suggested that selection of explant type followed by medium combination and cultivars was essential for obtaining high-frequency ASB induction. Across different cultivars the young leaf explant was found to be the most responsive explant, while Murashige and Skoog (MS) medium containing BA alone (17.8, 26.6, and 35.5 μM) was found to be the best medium for the production of maximum number of ASB. Between the two explants, shoot elongation was observed with ASB obtained from young leaf explants on MS medium containing BA (2.2 and 4.4 μM) and gibberellie acid (GA3) (1.4, 2.9, 4.3 and 5.8 μM). The MS medium fortified with 4.4 μM BA+2.9μM GA3 was optimum for shoot elongation. Elongated shoots were rooted on liquid MS medium supplemented with 2.9 μM indole-3-acetic acid (IAA) and successfully established ex vitro.  相似文献

The sequence of the 2227 nucleotides of the 3-terminus of chilli vein-banding mottle virus-Chiangmai isolate (CVbMV-CM1) genome was determined. This sequence encodes a putative 287 amino acid coat protein. Downstream of coat protein coding region is a 288 nucleotide untranslated sequence terminated by a polyadenylate tract. The amino acid sequence of coat protein contained aspartic acid–alanine–glycine tripeptide, which may correlate with the high frequency of aphid transmissibility. The cleavage site between large nuclear inclusion protein (NIB) and coat protein was glutamine/serine. Comparison of 3 non-coding region nucleotide and coat protein amino acid sequences of CVbMV-CM1 with those of other 18 potyviruses showed 44–56% and 54–65% homology respectively, suggesting CVbMV should be regarded as new potyvirus.  相似文献

We have constructed a molecular linkage map of pepper (Capsicum spp.) in an interspecific F2 population of 107 plants with 150 RFLP and 430 AFLP markers. The resulting linkage map consists of 11 large (206–60.3 cM) and 5 small (32.6–10.3 cM) linkage groups covering 1,320 cM with an average map distance between framework markers of 7.5 cM. Most (80%) of the RFLP markers were pepper-derived clones, and these markers were evenly distributed across the genome. By using 30 primer combinations, we were able to generate 444 AFLP markers in the F2 population. The majority of the AFLP markers clustered in each linkage group, although PstI/MseI markers were more evenly distributed than EcoRI/MseI markers within the linkage groups. Genes for the biosynthesis of carotenoids and capsaicinoids were mapped on our linkage map. This map will provide the basis of studying secondary metabolites in pepper. Pepper fruit is typically red, but green, orange and yellow cultivars are gaining consumer acceptance. This color variation is mainly due to variations in carotenoid composition. Orange color in pepper can result from a number of carotenoid profiles, but its genetic basis is only partly known. We identified an EMS-induced orange-fruited mutant using the wild-type blocky red-fruited cultivar ‘Maor’ as progenitor. This mutant accumulates mainly β-carotene in its fruit, instead of the complex pattern of red and yellow carotenoids in ‘Maor’. We identified an A709 to G transition in the cDNA of β-CAROTENE HYDROXYLASE2 in the orange pepper and complete co-segregation of this single-nucleotide polymorphism with the mutated phenotype. We therefore hypothesized that β-CAROTENE HYDROXYLASE2 controls the orange mutation in pepper. Interestingly, the expression of β-CAROTENE HYDROXYLASE2 and additional carotenogenesis genes was elevated in the orange fruit compared with the red fruit, indicating possible feedback regulation of genes in the pathway. Because carotenoids serve as precursors for volatile compounds, we compared the volatile profiles of the two parents. The orange pepper contained more volatile compounds than ‘Maor’, with predominant elevation of norisoprenoids derived from β-carotene degradation, while sesquiterpenes predominated in the red fruit. Because of the importance of β-carotene as a provitamin A precursor in the human diet, the orange-fruited mutant might serve as a natural source for pepper fruit biofortification. Moreover, the change in volatile profile may result in a fruit flavor that differs from other pepper cultivars.  相似文献

Fruits, seeds and embryos of Pimenta officinalis Lindl. syn. Pimenta dioica (L.) Merr. (Myrtaceae) have been found in Gdask (northern Poland) in latrines dated to the 18th century. This is one of the first few records of allspice from European archaeobotanical material. The efficacy of Neoseiulus californicus (a generalist predatory mite) for the biological control of Tetranychus urticae, was compared to release of Phytoseiulus persimilis (a specialist predatory mite) and an acaricide treatment in sweet pepper plants grown in greenhouse tunnels in a hot and arid climate. To ensure uniform pest populations, spider mites were spread on pepper plants in two seasons; a natural infestation occurred in one season. Predators were released prophylactically and curatively in separate tunnels when plants were artificially infested with spider mites, and at low and moderate spider mite populations when infestations occurred naturally. Although spider mite populations did not establish well the first year, fewer spider mites were recovered with release of N. californicus than with all other treatments. In the second year, spider mites established and the prophylactic release of N. californicus compared favorably with the acaricide-treated plants. In the course of monitoring arthropod populations, we observed a significant reduction in western flower thrips (Frankliniella occidentalis) populations in tunnels treated with N. californicus as compared with non-treated control tunnels. Our field trials validate results obtained from potted-plant experiments and confirm that N. californicus is a superior spider mite predator at high temperatures and low humidities.  相似文献

The ripening-related pepper endo-1,4--D-glucanase (EGase) CaCel1 was over-expressed in transgenic tomato plants under the control of the constitutive 35S promoter to investigate the effects on plant growth and fruit softening of high levels of a potential cell wall-degrading activity. In transgenic fruit, recombinant CaCel1 protein was associated with a high-salt putative cell wall fraction, and extractable CMCase activity was increased by up to 20-fold relative to controls. However, the effects of high levels of EGase activity on fruit cell wall metabolism were relatively small. The largest consequence observed was a decrease of up to 20% in the amount of matrix glycans in a 24% KOH-soluble fraction consisting of polysaccharides tightly bound to cellulose. This decrease was confined to polysaccharides other than xyloglucan, did not affect the size distribution of remaining molecules, and was not correlated with a corresponding increase in glycans in a 4% KOH-soluble fraction loosely bound to cellulose, suggesting that the missing polymers had been degraded to fragments small enough to be lost from the extracts. The amount of matrix glycans in the 4% KOH-soluble fraction was not substantially changed, but the size distribution showed a small relative increase in the amount of polymers in a peak eluting close to a linear dextran marker of 71 kDa. This could be due either to an increase in the amount of polymers of this size, or to a loss from the extract of other polymers present in peaks of higher molecular weight. Transgenic fruit were not softer than controls but appeared the same or slightly firmer at both green and red developmental stages, and no differences in plant vegetative growth were observed. CaCel1 did not cause depolymerization of tomato fruit xyloglucan in vivo, but differences in the amount or molecular weight profile of other matrix glycans were observed. The data suggest that degradation of a proportion of matrix glycans other than xyloglucan does not result in fruit softening, and that fruit softening is not limited by the amount of EGase activity present during ripening.  相似文献

Lipid transfer proteins (LTPs) were thus named because they facilitate the transfer of lipids between membranes in vitro. This study was triggered by the characterization of a 9-kDa LTP from Capsicum annuum seeds that we call Ca-LTP(1) . Ca-LTP(1) was repurified, and in the last chromatographic purification step, propanol was used as the solvent in place of acetonitrile to maintain the protein's biological activity. Bidimensional electrophoresis of the 9-kDa band, which corresponds to the purified Ca-LTP(1) , showed the presence of three isoforms with isoelectric points (pIs) of 6.0, 8.5 and 9.5. Circular dichroism (CD) analysis suggested a predominance of α-helices, as expected for the structure of an LTP family member. LTPs immunorelated to Ca-LTP(1) from C. annuum were also detected by western blotting in exudates released from C. annuum seeds and also in other Capsicum species. The tissue and subcellular localization of Ca-LTP(1) indicated that it was mainly localized within dense vesicles. In addition, isolated Ca-LTP(1) exhibited antifungal activity against Colletotrichum lindemunthianum, and especially against Candida tropicalis, causing several morphological changes to the cells including the formation of pseudohyphae. Ca-LTP(1) also caused the yeast plasma membrane to be permeable to the dye SYTOX green, as verified by fluorescence microscopy. We also found that Ca-LTP(1) is able to inhibit mammalian α-amylase activity in vitro.  相似文献

To study the resistance of pepper to Phytophthora capsici, we analyzed 94 doubled-haploid (DH) lines derived from the intraspecific F1 hybrid obtained from a cross between Perennial, an Indian pungent resistant line, and Yolo Wonder, an American bell-pepper susceptible line, with 119 DNA markers. Four different criteria were used to evaluate the resistance, corresponding to different steps or mechanisms of the host-pathogen interaction: root-rot index, receptivity, inducibility and stability. Three distinct ANOVA models between DNA marker genotypes and the four disease criteria identified 13 genomic regions, distributed across several linkage groups or unlinked markers, affecting the resistance of pepper to P. capsici. Some QTLs were criterion specific, whereas others affect several criteria, so that the four resistance criteria were controlled by different combinations of QTLs. The QTLs were very different in their quantitative effect (R2 values), including major QTLs which explained 41–55% of the phenotypic variance, intermediate QTLs with additive or/and epistatic action (17–28% of the variance explained) and minor QTLs. Favourable alleles of some minor QTLs were carried in the susceptible parent. The total phenotypic variation accounted for by QTLs reached up to 90% for receptivity, with an important part due to epistasis effects between QTLs (with or without additive effects). The relative impact of resistance QTLs in disease response is discussed.  相似文献

The aim of study was to gain a deeper knowledge about local and systemic changes in photosynthetic processes and sugar production of pepper infected by Obuda pepper virus (ObPV) and Pepper mild mottle virus (PMMoV). PSII efficiency, reflectance, and gas exchange were measured 48 and/or 72 h after inoculation (hpi). Sugar accumulation was checked 72 hpi and 20 d after inoculation (as a systemic response). Inoculation of leaves with ObPV led to appearance of hypersensitive necrotic lesions (incompatible interaction), while PMMoV caused no visible symptoms (compatible interaction). ObPV (but not PMMoV) lowered Fv/Fm (from 0.827 to 0.148 at 72 hpi). Net photosynthesis decreased in ObPV-infected leaves. In ObPV-inoculated leaves, the accumulation of glucose, fructose, and glucose-6-phosphate was accompanied with lowered sucrose, maltoheptose, nystose, and trehalose contents. PMMoV inoculation increased the contents of glucose, maltose, and raffinose in the inoculated leaves, while glucose-6-phosphate accummulated in upper leaves.  相似文献

The ‘Poblano’ pepper crop is economically important in Mexico and throughout the world as it is used as a hot spice in food. The cultivated area of the ‘Poblano’ pepper crop is decreasing yearly for many reasons, among them a wilt disease commonly associated with Fusarium spp. This disease is a problem of field and greenhouse production plants. Moreover, it is not clear whether the pathogens that cause wilt in mature plants are the same as those involved in the damping-off symptoms and death of pepper seedlings in greenhouses. For this reason, the aim of the present study was to identify the causal agent of damping-off in pepper during seedling production, establish its relationship with the causal agent of wilting in mature plants, and determine whether histological damage in seedlings occurs. Isolates were recovered from the crown rot and stem base of 4-month-old infected ‘Poblano’ mature pepper plants and were identified using morphological and phylogenetic approaches. Fusarium oxysporum and F. solani were isolated from the crown rot and base stem, respectively. A pathogenicity test showed that both species caused damping-off in pepper seedlings. Histological studies with inoculated seedlings of both isolates showed several changes in the external cortex, epidermal cells, endodermis, Casparian strips, cell size, and xylem wall. Casparian strip rupture resulted in permeability loss and regulatory activity to maintain the cellular equilibrium inside the vascular bundles. Hence, according to these findings, producers should avoid seedling contamination by infected mature plants because the aggressiveness of Fusarium isolates can cause rapid seedling mortality.  相似文献
