Behavioural changes that occur during the parr–smolt transformation were investigated in juvenile coho salmon Oncorhynchus kisutch. Fish from two populations were examined from the Fraser River catchment in British Columbia, Canada; a short and a long-distance migrating population. Fish showed a significant decrease in condition factor and significant increase in gill Na+K+-ATPase activity during the spring indicating that they became competent smolts, but no difference between populations. Temperature preference trials were conducted using a shuttlebox system throughout the spring. Mean temperature preference did not differ between the two populations, but preferred temperature decreased with development from 16.5 ± 0.3°C for parr to 15.5 ± 0.4°C for smolts. Mean swimming velocity was also greater in smolts than parr, but there was no difference between the two populations. The preference for warmer water temperature observed for parr in early spring may be advantageous for stimulating smolt development. Preference for slightly cooler temperatures observed for smolts would sustain elevated seawater tolerance during the smolt window by a short time and may ensure successful transition to the marine environment.  相似文献

We examined changes on N-methyl-d-aspartate receptors (NRs) in different growth stages (early parr, parr, and early smolt) of chum salmon, Oncorhynchus keta, during parr-smolt transformation from freshwater to seawater. Expression levels of NR genes mRNA and concentration of cortisol, T3, T4, dopamine and Na+/K+-ATPase activity significantly increased at salinity change condition. Moreover, in cultured brain cells, NRs were significantly lower in all groups treated with MK-801 (an antagonist of NRs) than in the early parr stage group in the FW treatment. We confirmed that the reduction in mRNA expression levels of NRs increased from the early parr to the early smolt stage. The information reported here should be taken into account in future studies on the relationship between memory factors of natal streams and homing mechanisms in Salmonidae.  相似文献

To examine the changes in secretion of growth hormone (GH) and prolactin (PRL) with reference to their osmoregulatory roles, changes in pituitary mRNA levels and plasma concentrations of these hormones were examined during seawater adaptation in silvery juveniles (smolts) and precociously mature males (dark parr) of amago salmon (Oncorhynchus rhodurus). Transfer to seawater increased plasma sodium levels in both smolts and dark parr. Smolts adjusted their plasma sodium to the level associated with seawater-adaptation (165 mEq/liter) within 3 days, whereas no adjustment was seen in dark parr; the latter failed to survive in seawater for more than 3 days. In smolts, plasma GH levels increased significantly 1 day after transfer, whereas there was no significant change in dark parr. An increase in GH mRNA levels was observed in smolts in association with increased plasma GH, whereas there was no change in dark parr. In contrast, a reduction in plasma PRL levels was consistently observed in both smolts and dark parr after transfer to seawater. However, there was no significant change in PRL mRNA levels in either smolts or dark parr. These results suggest that both gene expression and release of GH are activated by seawater transfer only in smolts with adequate seawater adaptability, whereas PRL gene expression is decreased after seawater transfer regardless of seawater adaptability.  相似文献

Since zinc mediates the effects of many hormones or is found in the structure of numerous hormone receptors, zinc deficiency leads to various functional impairments in the hormone balance. And also thyroid hormones have important activity on metabolism and feeding. NPY and leptin are affective on food intake and regulation of appetite. The present study is conducted to determine how zinc supplementation and deficiency affect thyroid hormones (free and total T3 and T4), melatonin, leptin, and NPY levels in thyroid dysfunction in rats. The experiment groups in the study were formed as follows: Control (C); Hypothyroidism (PTU); Hypothyroidism+Zinc (PTU+Zn); Hypothyroidism+Zinc deficient; Hyperthyroidism (H); Hyperthyroidism+Zinc (H+Zn); and Hyperthyroidism+Zinc deficient. Thyroid hormone parameters (FT3, FT4, TT3, and TT4) were found to be reduced in hypothyroidism groups and elevated in the hyperthyroidism groups. Melatonin values increased in hyperthyroidism and decreased in hypothyroidism. Leptin and NPY levels both increased in hypo- and hyperthyroidism. Zinc levels, on the other hand, decreased in hypothyroidism and increased in hyperthyroidism. Zinc supplementation, particularly when thyroid function is impaired, has been demonstrated to markedly prevent these changes.  相似文献

Smolt traits (length, age) and timing of smolt migration of wild Atlantic salmon, Salmo salar L., were investigated in the Simojoki River, northern Baltic Sea. The aim was to determine whether they responded to changes in parr length, parr density and temperature from 2000 to 2014. Annual electrofishing surveys and smolt numbers determined parr densities by springtime trapping in the river mouth. During the smolt trapping period captured parr and smolts were aged from scales. Water temperature was measured daily. Mean length decreased from 137 mm (TL) to 129 mm among 2‐year‐old smolts, and from 150 mm to 139 mm among 3‐year‐olds. Median date of the smolt migration was 10 days earlier, from early June to late May during the study period, linked to the rise in air temperature in May at the nearby Kemi‐Tornio airport. However, the median day temperature and the mean daily water temperatures during the second (Q2) and third (Q3) migration quartiles did not change. This implied that migration began when a suitable water temperature was reached, independent of the date.  相似文献

The aim of the present study was to illustrate the combined effect of excess iodine and low-protein diet on the thyroid, and the potential molecular mechanism of this effect. One hundred ninety-two Wistar rats were randomly divided into the following groups: normal; 10- (10HI), 50- (50HI), or 100-fold excess iodine (100HI); low-protein; and low-protein combined with 10- (L10HI), 50- (L50HI), or 100-fold excess iodine (L100HI). At the end of 2, 4, or 6 months, the rats were sacrificed for iodine concentration and thyroid hormone analyses. The histological appearance of the thyroid gland was observed at the end of 6 months. At the end of 6 months, the serum total thyroxine (TT4), free thyroxine (FT4), total triiodothyronine (TT3), and free triiodothyronine (FT3) levels in the 100HI, L10HI, L50HI, and L100HI groups were significantly lower than the control group (P?<?0.05 or P?<?0.01). Serum TT4, FT4, TT3, and FT3 levels in the low-protein excess iodine groups were significantly lower than the groups with an equal amount of excess iodine alone (P?<?0.05 or P?<?0.01). Light and electron microscopy showed that excess iodine caused damage to the ultrastructure of thyroid and apoptosis of follicular epithelial cells. In the 100HI, L50HI, and L100HI groups, thyroid follicular epithelial cells became flattened, and follicles became distended with colloid. The damage was more serious in low-protein groups. The present findings demonstrated that the low-protein diet aggravated the damage to the thyroid caused by excess iodine alone.  相似文献

In Atlantic salmon, the preadaptation to a marine life, i.e., parr-smolt transformation, and melatonin production in the pineal gland are regulated by the photoperiod. However, the clock genes have never been studied in the pineal gland of this species. The aim of the present study was to describe the diurnal expression of clock genes (Per1-like, Cry2, and Clock) in the pineal gland and brain of Atlantic salmon parr and smolts in freshwater, as well as plasma levels of melatonin and cortisol. By employing an out-of-season smolt production model, the parr-smolt transformation was induced by subjecting triplicate groups of parr to 6 wks (wks 0 to 6) under a 12?h:12?h light-dark (LD) regime followed by 6 wks (wks 6 to 12) of continuous light (LL). The measured clock genes in both pineal gland and brain and the plasma levels of melatonin and cortisol showed significant daily variations in parr under LD in wk 6, whereas these rhythms were abolished in smolts under LL in wk 12. In parr, the pineal Per1-like and Cry2 expression peaked in the dark phase, whereas the pineal Clock expression was elevated during the light phase. Although this study presents novel findings on the clock gene system in the teleost pineal gland, the role of this system in the regulation of smoltification needs to be studied in more detail. The frequency distribution of the fork length of 0+ aged masu salmon,Oncorhynchus masou, changed from unimodal to bimodal distribution in autumn of the years from 1982 to 1984 in the Mogusa River of southern Hokkaido, Japan. The bimodal distribution consists of two (upper and lower) modal groups. These two groups resulted from a difference in growth rate of 0+ aged individuals in autumn. Fish belonging to the upper modal group are assumed to be potential 1+ smolts. Whether 0+ aged parr transform into smolt or remain as parr in the following spring may be related to the growth rate of fish in the first autumn.  相似文献

One‐year‐old and 1‐summer‐old salmon parr have been stocked in the Simojoki, a northern Baltic river, to protect the natural salmon stock from extinction. This enhancement practice was studied during a 5‐year stocking period, when an average of 14.3 1‐summer‐old parr and 5.4 1‐year‐old parr was needed to produce one sea running smolt. The mean yield from stocking was 70 smolts per 1000 stocked 1‐summer‐old parr and 186 smolts per 1000 stocked 1‐year‐old parr. The mean cost of smolts produced by stocking 1‐year‐old and 1‐summer‐old parr was 1.2 and 2.6 times the cost of producing one 50 g smolt in the hatchery, respectively. There was a negative relationship between density of wild parr and length of smolts stocked as 1‐summer‐old parr in the river, but no relationship between wild parr density and length of smolts stocked as 1‐year‐old parr. This indicated that 1‐summer‐old parr were more susceptible than the older parr to competition with wild parr. Considering production costs and smolt yield, stocking of 1‐year‐old parr appeared to be more profitable than that of 1‐summer‐old parr in the enhancement of the endangered salmon stock.  相似文献

This study was undertaken to examine the long‐term effects of photoperiod, temperature and their interaction on growth, gill Na+,K+‐ATPase (NKA) activity, seawater tolerance and plasma growth‐hormone levels in Atlantic salmon Salmo salar pre‐smolts and smolts. The fish (mean ± s.e . initial body mass = 15·9 ± 0·4 g) were reared on two photoperiods (continuous light, LL, and simulated natural photoperiod, LDN, 60° 25′ N) and two temperatures (8·3 and 12·7° C) from June to May of the following year. Mean body mass was affected by photoperiod, temperature and their interactions. Both temperature groups on LL developed peak levels in gill NKA activity from October to November, 4–5 months prior to the natural season for the parr–smolt transformation. Fish at 12° C showed peak levels in NKA activity 4–6 weeks before the fish at 8° C. Fish in all four experimental groups showed maximum NKA activity within a similar size range (113–162 g). The present findings further indicate that smoltification in S. salar is to some extent driven by size, and that S. salar will develop smolt characteristics, e.g. a marked increase in NKA activity, within a similar size range. Faster‐growing S. salar will, thus, reach this size threshold at a relatively younger age.  相似文献

Alterations in circulatory levels of total T4 (TT4), total T3 (TT3), free T4 (FT4), free T3 (FT3), thyrotropin (TSH) and T3 uptake (T3U) were studied in male and female sea-level residents (SLR) at sea level, in Armed forces personnel staying at high altitude (3750 m) for prolonged duration (acclimatized lowlanders, ALL) and in high-altitude natives (HAN). Identical studies were also performed on male ALL who trekked to an extreme altitude of 5080 m and stayed at an altitude of more than 6300 m for about 6 months. The total as well as free thyroid hormones were found to be significantly higher in ALL and HAN as compared to SLR values. Both male as well as female HAN had higher levels of thyroid hormones. The rise in hormone levels in different ALL ethnic groups drawn from amongst the southern and northern parts of the country was more or less identical. In both HAN and ALL a decline in FT3 and FT4 occurred when these subjects trekked at subzero temperatures to extreme altitude of 5080 m but the levels were found to be higher in ALL who stayed at 6300 m for a prolonged duration. Plasma TSH did not show any appreciable change at lower altitudes but was found to be decreased at extreme altitude. The increase in thyroid hormones at high altitude was not due to an increase in hormone binding proteins, since T3U was found to be higher at high altitudes. A decline in TSH and hormone binding proteins and an increase in the free moiety of the hormones is indicative of a subtle degree of tissue hyperthyroidism which may be playing an important role in combating the extreme cold and hypoxic environment of high altitudes.  相似文献

Our recent study showed that monocrotophos (MCP) pesticide disrupted the hypothalamic-pituitary-thyroid (HPT) axis in male goldfish (Carassius auratus); however, the effects of MCP on the thyroid system in female goldfish are remain unclear. In the present study, plasma thyroid hormone (TH) and thyroid-stimulating hormone (TSH) levels were evaluated in female goldfish exposed to 0.01, 0.10, and 1.00 mg/L of 40% MCP-based pesticide for 21 days in a semi-static exposure system. Expression profiles of HPT axis-responsive genes, including transthyretin (ttr), deiodinases (d1, d2, and d3), tshβ, thyrotropin-releasing hormone (trh), and corticotrophin-releasing hormone (crh), were determined. The results indicated that MCP decreased the plasma levels of total 3,3′,5-triiodo-l-thyronine (TT3) and the ratio of TT3 to total 3,3′,5,5′-l-thyroxine (TT4), and induced alternative expression of TH-related genes. Exposure to 0.01 and 0.10 mg/L MCP pesticide resulted in the up-regulation of ttr mRNA. The reduction of plasma TT3 levels was partly attributed to an increase in the metabolism of T3 in the liver, as revealed by the highly elevated hepatic d1 and d3 mRNA levels in the MCP treatment groups, and the expression of hepatic d3 showed a negative correlation with the plasma TT3/TT4 levels in females. Moreover, the plasma TSH levels were lower in females exposed to 0.01 and 0.10 mg/L MCP pesticide, whereas the up-regulation of tshβ mRNA levels was compensated by the decreased plasma TT3 levels. These results indicated that MCP had the potential to influence several pathways of HPT axis homeostasis in female goldfish.  相似文献

Salmon ( Salmo salar ) formed the largest proportion by mass offish found in the stomachs of red-breasted mergansers ( Mergus serrator ) shot during the smolt run in north-east Scotland. Salmon parr represented approximately two-thirds by mass of juvenile salmon eaten, whilst smolts were present in a smaller proportion than previously predicted. The median lengths of parr and smolts eaten were 70 and 115 mm, respectively; the latter being significantly smaller than the smolt population sampled during annual production estimates on the river.  相似文献

The Atlantic salmon has a complex life-cycle in which it encounters a salinity barrier initially upon migration to the sea as a young smolt and later as an adult salmon returning to its natal river. Concurrent with seawater migration is a process termed smoltification which is a series of metabolic changes which transform the freshwater parr into smolts adapted for life in the marine environment. To gain an understanding of events occurring at the molecular level in the salmon liver during this developmental process, a cDNA library prepared from post-smolt salmon liver mRNA was screened with total liver cDNA probes synthesised from parr and smolts. Clones which hybridised more strongly to the smolt probe than the parr probe were chosen as candidates, for an analysis of liver gene expression implicated in seawater adaptation. Many of these cDNA clones encoded the iron binding protein transferrin. Transferrin mRNA levels were determined to be significantly higher in seawater smolt salmon than in freshwater smolts implying that transferrin may play a role in seawater adaptation.  相似文献

In general, hatchery salmonid smolts experience higher mortality during migration than wild smolts, which is suggested to be due to domestication effects and that hatchery fish lack experience of the natural environment. However, possible differences in feeding during smolt migration between hatchery and wild smolts have rarely been addressed. We compared the number of feeding smolts and stomach fullness among wild Atlantic salmon smolts, hatchery-reared smolts released as 1-year-old parr, and hatchery-reared smolts released as 2-year-old smolts during their descent to sea in River Tornionjoki. In addition, estimations of prey selection among the smolt groups were conducted. A high proportion of wild smolts and smolts stocked as parr actively fed during the smolt migration. A lower proportion of smolts stocked as smolts was feeding and their stomach fullness were much reduced in comparison with the two other groups. The study also indicated that the feeding of migrating smolts is selective rather than opportunistic. In conclusion, this study suggests that stocked 2-year-old smolts may enter sea with an inferior foraging behaviour and it is a possibility that this may contribute to the observed low post-smolt survival in the Baltic Sea.  相似文献

Abstract

The diurnal patterns of changes in plasma cortisol, growth hormone (GH), somatostatin‐14 (SRIF), thyroid hormones (L‐thyroxine, T4 and triiodo‐L‐thyronine, T3) and glucose were investigated in rainbow trout, Oncorhynchus mykiss, both during single meal‐feeding and during a progressive fast of 13 weeks. All measured variables exhibited a diurnal pattern in fed rainbow trout, most of which appeared to be correlated with the time of feeding (30–60 min after the onset of light), while additional changes, associated with the scotophase were also found for cortisol. Although fasting had no affect on mean daily plasma cortisol or SRIF concentrations, there was a progressive increase in mean daily plasma GH concentrations and a progressive decrease in mean daily plasma thyroid hormone and glucose concentrations associated with fasting. However, for GH, significant changes were not evident until week 10 of the fast. In addition, fasting appeared to phase‐shift the diurnal patterns of plasma GH, cortisol and glucose concentrations; the consequence of such a shift is that conclusions as to the effects of fasting, if based on a single time of sampling, may be flawed.  相似文献

Polychlorinated biphenyls (PCBs) are a group of persistent organochlorine compounds that have the potential to disrupt the homeostasis of thyroid hormones (THs) in fish, particularly juveniles. In this study, thyroid histology, plasma TH levels, and iodothyronine deiodinase (IDs, including ID1, ID2, and ID3) gene expression patterns were examined in juvenile Japanese flounder (Paralichthys olivaceus) following 25- and 50- day waterborne exposure to environmentally relevant concentrations of a commercial PCB mixture, Aroclor 1254 (10, 100, and 1000 ng/L) with two-thirds of the test solutions renewed daily. The results showed that exposure to Aroclor 1254 for 50 d increased follicular cell height, colloid depletion, and hyperplasia. In particular, hypothyroidism, which was induced by the administration of 1000 ng/L Aroclor 1254, significantly decreased plasma TT4, TT3, and FT3 levels. Profiles of the changes in mRNA expression levels of IDs were observed in the liver and kidney after 25 and 50 d PCB exposure, which might be associated with a reduction in plasma THs levels. The expression level of ID2 mRNA in the liver exhibited a dose-dependent increase, indicating that this ID isotype might serve as sensitive and stable indicator for thyroid-disrupting chemical (TDC) exposure. Overall, our study confirmed that environmentally relevant concentrations of Aroclor 1254 cause significant thyroid disruption, with juvenile Japanese flounder being suitable candidates for use in TDC studies.  相似文献

Juvenile stages of rainbow trout, smaller parr and older juveniles, termed smolts, show differences in red muscle contractile properties: parr red muscle has faster kinetics and a faster maximum shortening velocity than smolt red muscle. A developmental reduction in the number of MHC isoforms as detected by SDS-PAGE between parr and smolt has also been observed. To investigate whether this shift in contractile kinetics results from differential gene expression, three different MHC cDNA fragments, one each from red, white, and ventricular muscle, were identified. The red muscle and ventricular forms are novel MHCs, and the white muscle form is identical to a published MHC from adult trout white muscle. Tissue and developmental stage-specific expression patterns of these MHC isoforms were examined using isoform-specific RT-PCR. Ventricular muscle typically showed only the ventricular form; 60% parr and 80% smolts expressed the ventricular form only. Approximately half of the white muscle samples of either parr or smolts, 58% and 50%, respectively, expressed only white muscle MHC. Red muscle samples were the most heterogeneous, with red muscle MHC found in combination with either the white or ventricular form or both. Combining samples from the anterior and posterior, 8% of parr red muscle samples expressed solely the red muscle MHC form, and 30% of smolt red muscle samples expressed the red muscle form alone. Variations in the relative contribution of each MHC to the red muscle of parr and smolt may explain observed differences in protein composition and contractile properties. J. Exp.
