The effect of colonization with the arbuscular mycorrhizal (AM) fungus Glomus mosseae (Nicol. & Gerd.) Gerdemann & Trappe on the growth and physiology of NaCl-stressed maize plants ( Zea mays L. cv. Yedan 13) was examined in the greenhouse. Maize plants were grown in sand with 0 or 100 mM NaCl and at two phosphorus (P) (0.05 and 0.1 mM) levels for 34 days, following 34 days of non-saline pre-treatment. Mycorrhizal plants maintained higher root and shoot dry weights. Concentrations of chlorophyll, P and soluble sugars were higher than in non-mycorrhizal plants under given NaCl and P levels. Sodium concentration in roots or shoots was similar in mycorrhizal and non-mycorrhizal plants. Mycorrhizal plants had higher electrolyte concentrations in roots and lower electrolyte leakage from roots than non-mycorrhizal plants under given NaCl and P levels. Although plants in the low P plus AM fungus treatment and those with high P minus AM fungus had similar P concentrations, the mycorrhizal plants still had higher dry weights, soluble sugars and electrolyte concentrations in roots. Similar relationships were observed regardless of the presence or absence of salt stress. Higher soluble sugars and electrolyte concentrations in mycorrhizal plants suggested a higher osmoregulating capacity of these plants. Alleviation of salt stress of a host plant by AM colonization appears not to be a specific effect. Furthermore, higher requirement for carbohydrates by AM fungi induces higher soluble sugar accumulation in host root tissues, which is independent of improvement in plant P status and enhances resistance to salt-induced osmotic stress in the mycorrhizal plant.  相似文献

The objective of this study was to assess the effect of different Cd and Si concentrations on the maize plants. The following Cd and/or Si treatments were used: 5 Cd; 10 Cd; 100 Cd; 5 Cd + 0.08 Si; 10 Cd + 0.08 Si; 100 Cd + 5 Si treatments (Cd concentration in μM, Si concentration in mM). The plant growth, photosynthetic pigments content, antioxidant enzymes activities (POX, SOD, CAT), Cd and Si accumulation, translocation and cell wall deposition of the maize plants was observed. Changes in the endodermal cell walls development and late metaxylem elements lignification due to Cd and/or Si treatment were also evaluated. The negative effect of Cd (5 and 10 μM) on the growth parameters was alleviated by Si at 0.08 mM. The positive effect of Si was not observed at higher Cd and Si concentrations. This indicates that the alleviating effect of Si on Cd toxicity depends on the Cd and Si concentrations. Plants responded to Cd toxicity by an increase of antioxidant enzyme activity. Silicon addition in Cd + Si treatment stimulated an increase in the activity of antioxidant enzymes in comparison with the Cd treatment. Chlorophyll and carotenoid content in the Cd treated plants was not significantly affected by Si. The young maize plants retained much more Cd in their roots as they translocated into the shoots. 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments correlated with an increase in Cd concentration in the roots and shoots, and in the cell walls. Silicon caused a slight decrease of the Cd translocation into the shoots in 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments. Negative correlation between the root Cd cell wall deposition and Cd translocation was observed. Cadmium and/or Si altered root anatomy. Cadmium enhanced suberin lamellae development and late metaxylem lignification; silicon in Cd + Si treatments accelerated suberin lamellae deposition and enhanced the tertiary endodermal cell walls formation in comparison with Cd treatments. Negative correlation between the endodermal cell walls development and Cd translocation was observed.  相似文献

Salt stress perturbs a multitude of physiological processes such as photosynthesis and growth. To understand the biochemical changes associated with physiological and cellular adaptations to salinity, two lettuce varieties (Verte and Romaine) were grown in a hydroponics culture system supplemented with 0, 100 or 200 mM NaCl. Verte displayed better growth under 100 mM NaCl compared to Romaine, but both genotypes registered relatively similar reductions in growth under 200 mM NaCl treatment. Both varieties showed differences in net photosynthetic activity in the absence of salt and 8 days after salt treatment. These differences diminished subsequently under prolonged salt stress (14 days). Verte showed enhanced leaf proline and restricted total cations especially Na+, lesser malondialdehyde (MDA) formation and lignification in the roots under 100 mM NaCl salinity. Membrane damage estimated by electrolyte leakage increased with elevated salt concentrations in roots of both varieties, but Verte had significantly lower electrolyte leakage relative to Romaine under 100 mM NaCl. Moreover, Verte also accumulated greater levels of carotenoids under increasing NaCl concentrations compared to Romaine. Taken together, these findings suggest that the greater tolerance of Verte to 100 mM NaCl is related to the more restricted accumulation of total cations and toxic Na+ in the roots and enhanced levels of antioxidative metabolites in root and leaf tissue.  相似文献

The present study was carried out to compare the effect of NaCl on growth, cell membrane damage, and antioxidant defences in the halophyte Crithmum maritimum L. (sea fennel). Physiological and biochemical changes were investigated under control (0 mM NaCl) and saline conditions (100 and 300 mM NaCl). Biomass and growth of roots were more sensitive to NaCl than leaves. Roots were distinguished from leaves by increased electrolyte leakage and high malondialdehyde (MDA) concentration. Superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activities, ascorbic acid (AA) and glutathione (GSH) concentrations were lower in the roots than in the leaves of control plants. The different activity patterns of antioxidant enzymes in response to 100 and 300 mM NaCl indicated that leaves and roots reacted differently to salt stress. Leaf CAT, APX and glutathione reductase (GR) activities were lowest at 300 mM NaCl, but they were unaffected by 100 mM NaCl. Only SOD activity was reduced in the latter treatment. Root SOD activity was significantly decreased in response to 300 mM NaCl and root APX activity was significantly higher in plants treated with 100 and 300 mM compared to the controls. The other activities in roots were insensitive to salt. The concentration of AA decreased in leaves at 100 and 300 mM NaCl, and in roots at 300 mM NaCl, when compared to control plants. The concentrations of GSH in NaCl-treated leaves and roots were not significantly different from the controls. In both organs, AA and GSH were predominating in the total pool in ascorbic acid and glutathione, under control or saline conditions.  相似文献

We have investigated the influence of silicon on higher zinc concentration reducing the growth of aboveground parts by ca 50 % in young maize plants (hybrid Novania) grown in hydroponics. Eight different treatments were used: control, Zn (800 μM ZnSO4·7H2O), Si1/Si2.5/Si5 (1/2.5/5 mM Na2SiO7) and Zn+Si (combination of zinc and all silicon concentrations). The concentration of Zn and Si and their distribution in plants was determined. The growth parameters (length of primary seminal root, leaf area of first and second leaves, fresh and dry weight of below- and above-ground plant parts) of plants grown in various Zn+Si treatments were significantly decreased in comparison to all other treatments. Increasing concentration of Si in combination with Zn treatment and selected hybrid (Novania) resulted in increased physiological stress in comparison to Zn treatment. However, roots and shoots of all Zn+Si treated plants contained significantly lower amount of Zn than Zn treatment. The Si concentration in roots was the same in Si and Zn+Si plants. In general, higher amount of Si was observed in shoots than in roots of Si1- and Si2.5-treated plants and opposite was observed in Si5-treated plants. In spite of significantly decreased root and shoot accumulation of Zn in the presence of Si, no positive effect of Si on Zn toxicity in young maize plants under experimental conditions used in this work and used maize hybrid was observed.  相似文献

In this study cashew (Anacardium occidentale) plants were exposed to a short- and long-term exposure to NaCl in order to establish the importance of the salt-induced proteolysis and the glutamine synthetase activity on the proline accumulation. The cashew leaf showed a prominent proline accumulation in response to salt stress. In contrast, the root tissue had no significant changes in proline content even after the drastic injury caused by salinity on the whole plant. The leaf proline accumulation was correlated to protease activity, accumulation of free amino acid and ammonia, and decrease of both total protein and chlorophyll contents. The leaf GS activity was increased by the salt stress whereas in the roots it was slightly lowered. Although the several amino acids in the soluble pool of leaf tissue have showed an intense increment in its concentrations in the salt-treated plants, proline was the unique to show a proportional increment from 50 to 100 mol m-3 NaCl exposure (16.37 to 34.35 mmol kg-1 DM, respectively). Although the leaf glutamate concentration increased in the leaves of the salt-stressed cashew plants, as compared to control, its relative contribution to the total amino acid decreased significantly in stressed leaves when compared to other amino acids. In addition, when the leaf discs were incubated with NaCl in the presence of exogenous precursors (Glu, Gln, Orn or Arg) involved in the proline synthesis pathways, the glutamate was unique in inducing a significant enhancement of the proline accumulation compared to those discs with precursor in the absence of NaCl. These results, together with the salt-induced increase in the GS activity, suggest an increase in the de novo synthesis of proline probably associated with the increase of the concentration of glutamate. Moreover, the prominent salt-induced proline accumulation in the leaves was associated with the higher salt-sensitivity in terms of proteolysis and salt-induced senescence as compared to the roots. In conclusion, the leaf-proline accumulation was due, at least in part, to the increase in the salt-induced proteolysis associated with the increments in the GS activity and hence the increase in the concentration of glutamate precursor in the soluble amino acid pool.  相似文献

The behaviour of enzymes involved in nitrogen metabolism, as well as oxidative stress generation and heme oxygenase gene and protein expression and activity, were analysed in soybean (Glycine max L.) nodules exposed to 50, 100 and 200 mM NaCl concentrations. A significant increase in lipid peroxidation was found with 100 and 200 mM salt treatments. Moreover, superoxide dismutase, catalase and peroxidase activities were decreased under 100 and 200 mM salt. Nitrogenase activity and leghemeoglobin content were diminished and ammonium content increased only under 200 mM NaCl. At 100 mM NaCl, glutamine synthetase (GS) and NADH-glutamate dehydrogenase (GDH) activities were similar to controls, whereas a significant increase (64%) in NADH-glutamate synthase (GOGAT) activity was observed. GS activity did not change at 200 mM salt treatment, but GOGAT and GDH significantly decreased (40 and 50%, respectively). When gene and protein expression of GS and GOGAT were analysed, it was found that they were positively correlated with enzyme activities. In addition, heme oxygenase (HO) activity, protein synthesis and gene expression were significantly increased under 100 mM salt treatment. Our data demonstrated that the up-regulation of HO, as part of antioxidant defence system, could be protecting the soybean nodule nitrogen fixation and assimilation under saline stress conditions.  相似文献

The aim of this study was to determine the effect of exogenously applied Si on the growth and physiological parameters of sorghum [Sorghum bicolor (L.) Moench] cultivated in hydroponics with elevated zinc concentrations (75 μM and 150 μM Zn). Increased concentrations of Zn inhibited root growth and biomass production of roots and shoots. Application of Si individually showed a positive effect on root growth but negatively affected production of fresh and dry biomass of roots and shoots. On the other hand, silicon in combination with Zn significantly reduced the inhibitory effect of Zn on root growth but did not positively affect biomass production of roots and shoots. Accumulation of Zn in plant tissues increased with increasing Zn concentration in nutrient solution, but application of Si in combination with Zn did not significantly influence Zn accumulation in roots. Completely opposite results were found in Si accumulation in roots treated with Si in combination with Zn. Interaction of these ions resulted in considerable increase of Si accumulation in roots which almost doubled in comparison to individal Si treatment. Impact of Zn on the activity of some antioxidant enzymes was equivocal and differences were observed also between two Zn concentrations. Individual application of Si resulted in significant increase in the activity of all studied antioxidant enzymes but Si in combination with Zn mostly negatively affected their activity except the activity of catalase (CAT) which was the highest in roots grown in solution containing both Si and Zn ions. Comparing all obtained data we can assume that Si applied in combination with Zn did not significantly alleviate Zn toxicity in young sorghum except the growth of primary seminal root and further experiments are required for better understanding of their interaction.  相似文献

Glutamine synthetase (GS) level is positively influenced by exogenously supplied sucrose in isolated pea roots (similarly as nitrate reductase - NR), glutamate dehydrogenase (GDH) level negatively. Comparison with previous results shows that GS level decreases more slowly than NR level when sucrose is omitted from the medium; the rate of changes in GS level corresponds rather to that in GDH level. The increase in GDH level in the tips of isolated roots cultivated in the medium lacking sucrose stops after approx. 24 h, but continues for at least 72 h in more mature root parts. GS level decreases during the first 24 h in the tips of isolated roots (compared with roots of intact seedlings) cultivated both with sucrose and without it (without sucrose more), however it again rises in the course of further cultivation with sucrose. The differences in GS and GDH levels caused by omission of sucrose are small in isolated roots from which root tips were removed, the difference in NR level in decapitated roots is similar to that found in isolated roots with root tips left. Decapitated isolated roots cultivated without sucrose contain higher amounts of soluble sugars than corresponding roots with root tips left. These facts are dismissed with regard to sugar consumption, transport, and compartmentalisation, and with respect to production in root tips and other plant parts of unknown compounds involved in GS and GDH regulation. The results obtained suggest that GDH functions in pea roots in the deaminating direction.  相似文献

Tomato plants (Lycopersicon esculentum Mill, cv. Chibli F1) grown for 10 days on control medium were exposed to differing concentrations of NaCl (0, 25, 50, and 100mM). Increasing salinity led to a decrease of dry weight (DW) production and protein contents in the leaves and roots. Conversely, the root to shoot (R/S) DW ratio was increased by salinity. Na(+) and Cl(-) accumulation were correlated with a decline of K(+) and NO(3)(-) in the leaves and roots. Under salinity, the activities of nitrate reductase (NR, EC 1.6.6.1) and glutamine synthetase (GS, EC 6.3.1.2) were repressed in the leaves, while they were enhanced in the roots. Nitrite reductase (NiR, EC 1.7.7.1) activity was decreased in both the leaves and roots. Deaminating activity of glutamate dehydrogenase (GDH, EC 1.4.1.2) was inhibited, whereas the aminating function was significantly stimulated by salinity in the leaves and roots. At a high salt concentration, the nicotinamide adenine dinucleotide reduced (NADH)-GDH activity was stimulated concomitantly with the increasing NH(4)(+) contents and proteolysis activity in the leaves and roots. With respect to salt stress, the distinct sensitivity of the enzymes involved in nitrogen assimilation is discussed.  相似文献

We investigated effect of silicon (Si) on the growth, uptake of sodium (Na), chloride (Cl), boron (B), stomatal resistance (SR), lipid peroxidation (MDA), membrane permeability (MP), lipoxygenase (LOX) activity, proline (PRO) accumulation, H2O2 accumulation, non-enzymatic antioxidant activity (AA) and the activities of major antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT and ascorbate peroxidase, APX) of spinach and tomato grown in sodic-B toxic soil. Si applied to the sodic-B toxic soil at 2.5 and 5.0 mM concentrations significantly increased the Si concentration in the plant species and counteracted the deleterious effects of high concentrations of Na, Cl and B on root and shoot growth by lowering the accumulation of these elements in the plants. Stomatal resistance, MP, MDA and the concentrations of H2O2 and PRO were higher in the plants grown in sodic-B toxic soil without Si: LOX activity of excised leaves of both species was increased by Si. Antioxidant activities of both species were significantly affected by Si, with the activities of SOD, CAT and APX decreased and AA increased by applied Si. For most of the parameters measured, it was found that 5 mM Si was more effective than the 2.5 mM Si. Based on the present work, it can be concluded that Si alleviates sodicity and B toxicity of the plants grown in sodic-B toxic soil by preventing both oxidative membrane damage and also translocation of Na, Cl and B from root to shoots and/or soil to plant, and lowering the phytotoxic effects of Na, Cl and B within plant tissues. It was concluded that tomato was more responsive to Si than spinach since it was more salt sensitive than spinach. To our knowledge, this is the first report that Si improves the combined salt and B tolerance of spinach and tomato grown in naturally sodic-B toxic soil, and which describes membrane-related parameters and antioxidant responses.  相似文献

Zinc deficiency and salinity are well-documented soil problems and often occur simultaneously in cultivated soils. Usually, plants respond to environmental stress factors by activating their antioxidative defense mechanisms. The antioxidative response of wheat genotypes to salinity in relation to Zn nutrition is not well understood. So, we investigated the effect of Zn nutrition on the growth, membrane permeability and sulfhydryl group (–SH groups) content of root cells and antioxidative defense mechanisms of wheat plants exposed to salt stress. In a hydroponic experiment, three bread wheat genotypes (Triticum aestivum L. cvs. Rushan, Kavir, and Cross) with different Zn-deficiency tolerance were exposed to adequate (1 μM Zn) and deficient (no Zn) Zn supply and three salinity levels (0, 60, and 120 mM NaCl). The results obtained showed that adequate Zn nutrition counteracted the detrimental effect of 60 mM NaCl level on the growth of all three wheat genotypes while it had no effect on the root and shoot growth of ‘Rushan’ and ‘Kavir’ at the 120 mM NaCl treatment. At the 0 and 60 mM NaCl treatments, Zn application decreased root membrane permeability while increased –SH group content and root activity of catalase (CAT) and superoxide dismutase (SOD) in ‘Rushan’ and ‘Kavir’. In contrast, Zn had no effect on the root membrane permeability and –SH group content of ‘Rushan’ and ‘Kavir’ exposed to the 120 mM NaCl treatment. At all salinity levels, ‘Cross’ plants supplied with Zn had lower root membrane permeability and higher –SH group content compared to those grown under Zn-deficient conditions. At the 0 and 60 salinity levels, Zn-deficient roots of Kavir and Rushan genotype leaked significantly higher amounts of Fe and K than the Zn-sufficient roots. In contrast, at the 120 mM treatment, Zn application had no effect or slightly increased Fe and K concentration in the root ion leakage of these wheat genotypes. For ‘Cross’, at all salinity levels, Zn-deficient roots leaked significantly higher amounts of Fe and K compared with the Zn-sufficient roots. The differential tolerance to salt stress among wheat genotypes examined in this study was related to their tolerance to Zn-deficiency, –SH group content, and root activity of CAT and SOD. Greater tolerance to salinity of Zn-deficiency tolerant genotype ‘Cross’ is probably associated with its greater antioxidative defense capacity.  相似文献

Ammonium assimilation into glutamine and glutamate is vital for plant growth as these are precursors for almost all nitrogenous compounds. Ammonium can be assimilated onto nitrogenous organic compounds by the concerted action of two enzymes that compose the glutamine synthetase (GS, EC 6.3.1.2) – glutamate synthase (Fd-GOGAT, EC 1.4.7.1; NADH–GOGAT, EC 1.4.1.14) cycle. Ammonium may also be directly incorporated into glutamate by the glutamate dehydrogenase (GDH, EC 1.4.1.2) aminating reaction. However, as GDH reversibly deaminates glutamate, its physiological role in vivo remains controversial. Potato has been classified as moderately tolerant to salinity. Potato GS is encoded by a small multigene family which is differentially regulated in an organ and age-dependent way. In this study, the effect of increasing concentrations of salinity in the soil in GS activity and gene-specific mRNA accumulation levels were studied on potato leaves and roots, as well as the biochemical parameters protein, chlorophyll, lipid peroxidation and proline levels, in order to evaluate the severity of the imposed stress. The data obtained suggests that when potato plants are subjected to salt stress, increased ammonium assimilation occurs in roots, due to an increased GS accumulation, along with a decreased assimilation in leaves. Regarding GS gene-specific mRNA accumulation, an organ-dependent response was also observed that contributes for the detected alteration in the ammonium assimilatory metabolism. This response may be a key feature for future genetic manipulations in order to increase crop productivity in salty soils. The possible contribution of GDH for ammonia assimilation was also investigated.  相似文献

The effects of exogenous sucrose, lactose, d -glucose, d (-)fructose, d -galactose, d -mannose, l -sorbose, l -arabinose and d -xylose on nitrate reductase (NR), glutamine synthetase (GS) and glutamate dehydrogenase (GDH) levels, on anaerobic nitrite production and on respiratory O2 consumption were studied in excised roots of pea (Pisum sativum L. cv. Raman). Sucrose, glucose and fructose increase NR and GS levels and decrease GDH level (when compared with roots cultures without any sugar) at all concentrations used, but the extent of this effect varies. NR induction is enhanced by all sugars within the concentration range studied. Precultivation of roots with mannose and galactose results in an increase in anaerobic nitrite production in a medium consisting of phosphate buffer and KNO3. GS reaches its maximum at lower sugar concentrations, this fact being especially clear-cut with galactose. The decrease in GS level observed in roots cultured without sucrose is enhanced by higher sorbose concentrations. The increase in GDH level occurring in roots cultured without sucrose is depressed by low galactose and mannose concentrations but enhanced by high galactose, mannose, xylose and a wide range of sorbose concentrations. Lactose exerts only slight influence on the enzymes. The effects of sugars are in no case consistent with their effect on respiratory O2 consumption which is most pronounced with NR. The above results show that the effects of sugars on NR, GS and GDH are not mediated by one universal mechanism.  相似文献

Glutamate dehydrogenase (GDH E.C. 1.4.1.2.4), glutamine synthetase (GS E.C. 6.3.1.2) and glutamate synthase (glutamine oxoglutarate amino transferase, GOGAT E.C. 2.6.1.53) activities, protein and organic nitrogen contents and growth of roots and shoots of maize seedlings raised in dark at 25±2°C in half strength Hoagland’s solution containing different ammonium salts as source of nitrogen, were determined to assess the contribution of alternate pathways in ammonium assimilation. Ammonium nitrate or in some cases ammonium chloride appeared to be the best source for both root and shoot growth and for increase in protein, total nitrogen and the enzymes of ammonium assimilation. In roots, NH4-nitrogen appeared to be assimilated by both GDH as well as GS-GOGAT pathways specially in the dark grown seedlings, while in shoots it was primarily by GS-GOGAT pathway.  相似文献

The wide range of plant responses to ammonium nutrition can be used to study the way ammonium interferes with plant metabolism and to assess some characteristics related with ammonium tolerance by plants. In this work we investigated the hypothesis of plant tolerance to ammonium being related with the plants’ capacity to maintain high levels of inorganic nitrogen assimilation in the roots. Plants of several species (Spinacia oleracea L., Lycopersicon esculentum L., Lactuca sativa L., Pisum sativum L. and Lupinus albus L.) were grown in the presence of distinct concentrations (0.5, 1.5, 3 and 6 mM) of nitrate and ammonium. The relative contributions of the activity of the key enzymes glutamine synthetase (GS; under light and dark conditions) and glutamate dehydrogenase (GDH) were determined. The main plant organs of nitrogen assimilation (root or shoot) to plant tolerance to ammonium were assessed. The results show that only plants that are able to maintain high levels of GS activity in the dark (either in leaves or in roots) and high root GDH activities accumulate equal amounts of biomass independently of the nitrogen source available to the root medium and thus are ammonium tolerant. Plant species with high GS activities in the dark coincide with those displaying a high capacity for nitrogen metabolism in the roots. Therefore, the main location of nitrogen metabolism (shoots or roots) and the levels of GS activity in the dark are an important strategy for plant ammonium tolerance. The relative contribution of each of these parameters to species tolerance to ammonium is assessed. The efficient sequestration of ammonium in roots, presumably in the vacuoles, is considered as an additional mechanism contributing to plant tolerance to ammonium nutrition.  相似文献

The effects of NaCl were studied in 6-month-old jack pine (Pinus banksiana Lamb.) seedlings growing in solution culture under hypoxic (approximately 2 mg lу O2) and well-aerated (approximately 8 mg lу O2) conditions. The results showed that hypoxia led to further reduction of stomatal conductance (gs) in plants treated with 45 mM NaCl. This effect was likely due to a reduction in root hydraulic conductance by both stresses. When applied individually or together, neither 45 mM NaCl nor hypoxia affected cell membrane integrity of needles as measured by tissue electrolyte leakage. Hypoxia did not alter shoot Na+ and Clm concentrations in NaCl-treated plants. However, root Na+ concentrations were lower in NaCl-treated hypoxic plants, suggesting that hypoxia affected the ability of roots to store Na+. Hypoxia also induced root electrolyte leakage from NaCl-treated and control plants. The higher root Clm concentrations compared with Na+ and the positive correlation between root Clm concentrations and electrolyte leakage suggest that Clm played a major role in salt injury observed in jack pine seedlings. Roots of well-aerated plants treated for 1 week with NaCl contained almost two-fold higher concentration of total non-structural carbohydrates compared with plants from other experimental treatments and these concentrations decreased in subsequent weeks. We suggest that under prolonged hypoxic conditions, roots lose the ability to prevent Clm uptake resulting in the increase in root Clm concentration, which has damaging effects on root cell membranes.  相似文献

The influence of increased nitrate concentration—14 (control) and 140 mmol L−1 (T)—in hydroponic culture on ammonia assimilation in cucumber (Cucumis sativus L. cv. Xintaimici) seedlings was investigated. The results showed that NH3 accumulation in the roots and leaves of T seedlings increased significantly, indicating that NH3 toxicity might be involved in nitrate stress. Under control conditions, GS and GOGAT activity were much higher in the leaves than in the roots, whereas GDH activity was much higher in the roots than in the leaves. Correlation analysis showed that NH3 concentration had a strong negative linear relationship with GDH activity in the roots but had a strong negative linear relationship with GS and GOGAT activity in the leaves. These results indicate that NH3 might be assimilated primarily via GDH reaction in the roots and via GS/GOGAT cycle in the leaves. Short-term nitrate stress resulted in the increase of GS and GOGAT activity in the roots and GDH activity in the leaves of T seedlings, indicating possible shifts in ammonia assimilation from the normal GDH pathway to GS/GOGAT pathway in the roots and from the normal GS/GOGAT pathway to the GDH pathway in the leaves under nitrate stress, but with the increase of treatment time, GS, GOGAT, and GDH activity in the roots and leaves of T seedlings decreased possibly due to low water potential and NH3 toxicity.  相似文献

Two 14-day-old seedlings of maize (Zea mays L.) cultivars (3223 and Vero) were exposed to different concentrations of lead [0, 2, 5 and 8 mM Pb(NO3)2·4H2O] for 8 days. Exposure of maize cultivars to excess Pb resulted in a significant root growth inhibition though shoot growth and absolute water content remained less affected. The results of chlorophyll a fluorescence indicated that the highly toxic Pb level affected photochemical efficiency in 3223, while no significant effect was observed in the Vero. At the highly toxic Pb concentration, higher membrane leakage was observed in 3223 leaves than that of Vero. This result was related to the accumulation of Pb. On the other hand, the results suggested that there were similar responses in total soluble POD and GR activities with increasing Pb concentrations between both cultivars. But APX activity significantly decreased at highly toxic Pb level in the Vero while a significant increase observed in the 3223. However, SOD activity in 3223 significantly decreased at the highly toxic Pb concentration compared with that at 2 mM Pb concentration. The results of the present study indicated that, Vero withstands excess Pb with its higher Pb accumulation capacity in roots and better upregulated protective mechanisms compared to 3223. Therefore, Vero is more tolerant to Pb toxicity compared to 3223 which was found to be a less tolerant cultivar.  相似文献
