The impact of low humidity in ambient air on water relations,nitrate uptake, and translocation of recently absorbed nitrogen,was investigated in 5-week-old tomato (Lycopersicon esculentumMill cv. Ailsa Craig) plants grown hydroponically in a completenutrient solution. Plants were subjected to dry air (relativehumidity 2–4% for 6 h. The transpiration rate increasedseveral-fold and the shoot water content decreased by almost20%, whereas root water content was unaffected. No effect onin vitro nitrate reductase (NR) activity was detected when usingan EDTA-contraining assay buffer. Replacement of EDTA with Mg2+revealed a significant decline in shoot NR activity, which suggestsphosphorylation of the enzyme during the stress treatment. Plantswere grown in a split-root system, in which one root half wasfed 15N-nitrate during the treatment, in order to determinenitrate uptake and translocation of recently absorbed nitrogenin the plants. Uptake of nitrate was substantially inhibited,but the proportion of absorbed 15N that was translocated tothe shoots was only slightly affected. In untreated plants,71% of the 15N recovered in roots had been retranslocated fromthe shoots, whereas in plants subjected to stress the deliveryof 15N from shoots to roots appeared to be completely inhibited.The data show that lowered humidity in air has significant effectson both uptake of nitrate as well as translocation of nitrogenwithin the plants. Some of these effects appear to be commonwith those observed in plants subjected to reduced water potentialsin the root environment and point to the possibility of theshoot water relations being highly influential on nitrogen uptakeand translocation. l4N nuclear magnetic resonance (NMR) spectroscopy was used todetect simple nitrogen compounds in root tissues from barley(Hordeum vulgare), maize (Zea mays) and pea (Pisum sativum)seedlings. The spectra included contributions from amino andamide groups but, as expected, these peaks were broad and poorlydefined in comparison with the resonances from nitrate and ammonium.A quantitative analysis of the spectra of nitrate-grown barleyroots showed that the nitrate resonance intensity accountedfor the total nitrate content of the tissue. Timecourses ofthe tissue nitrate intensity in response to changes in the externalnitrate composition followed single exponentials, and couldbe discussed in terms of nitrate exchange and possibly reduction.There was no evidence from these data, or from spin-latticerelaxation measurements, for the existence of more than onelarge pool of nitrate in the roots, and the results were consistentwith a model in which the bulk of the nitrate is vacuolar. Incontrast, timecourses of the ammonium resonance in ammonium-grownbarley roots were biphasic under certain external conditions,suggesting some form of compartmentation of ammonium in thetissue. Carob seedlings were grown hydroponically for 9 weeks under360 and 800 µl l-1CO2. One of two nitrogen sources, nitrateor ammonium, was added to the nutrient medium at concentrationsof 3 mol m-3. Root systems of the developing plants suppliedwith nitrate compared to those supplied with ammonium were characterizedby:(a)more biomass on the lower part of the root;(b)fewer lateralroots of first and second order;(c)longer roots;(d)higher specificroot length;(e)a smaller root diameter. The morphology of theroot systems of nitrate-fed plants changed in the presence ofelevated carbon dioxide concentrations, resembling, more closely,that of ammonium-fed plants. Total leaf area was higher in ammonium-than in nitrate-fed plants. Nitrate-fed plants had greater totalleaf area in the presence of high carbon dioxide than in normalCO2, due to an increase in epidermal cell size that led to developmentof larger leaflets with lower stomatal frequency. The observedchanges in the morphology of roots and shoots agreed with theresults observed for total biomass production. Nitrate-fed plantsincreased their biomass production by 100% in the presence ofelevated CO2compared to 15% in ammonium-fed plants, indicatingthat the response of carob to high CO2concentrations is verydependent on the nitrogen source. Under elevated CO2, nitrate-grownplants had a larger content of sucrose in both roots and shoots,while no significant difference was observed in the contentof sucrose in ammonium-grown plants, whether in ambient or enrichedcarbon dioxide. The pattern of lateral root initiation in seminal roots of wheat(Triticum aestivumL. cv. Alexandria) and the location, scaleand time-course for adjustments in initiation were studied afterchanges in C and N supply. Macroscopically visible primordiaappeared in a non-acropetal sequence with the frequency (numberper unit length) increasing with distance behind the main rootapex to a maximum at 40–50 mm behind the root tip. Pruningthe root system to a single seminal axis increased the primordiafrequency by 23% within 15 h. After longer periods, the effectof root-pruning was greater. The enhanced primordia frequencywas first observed in tissue located 0–10 mm behind theapex at the start of treatment. Feeding glucose (50 mM) alsoincreased primordia frequency within 15 h, but to a greaterextent, and here additional primordia were initiated in tissuelocated 0–10and10–20 mm behind the apex at the startof treatment. Withdrawing NO3-from one part of a split-rootsystem, whilst maintaining the supply to the other, reducedprimordia frequency in the non-fed roots and, in some cases,a compensatory increase in the NO3--fed roots was observed.The location and scale of the adjustments were similar to thosefound with root-pruning and glucose-feeding, but were slightlyslower to appear. In spite of some differences in detail, therewas a broad similarity in site, scale and time-course for adjustmentsin lateral root initiation with these treatments, which is consistentwith the operation of a common mechanism. Whenever an increasein primordia frequency was observed, it was associated withan increase in the ethanol-soluble sugar content of the tissue.However, the reduction in frequency in NO3--deprived roots wasalso accompanied by an increase in sugar content. There wasno consistent relationship between total N content of the tissueand primordia frequency, but there was between primordia frequencyand the rate of net NO3-uptake. The possible mechanisms controllinglateral root initiation are discussed. Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate. Nitrate ion uptake by the roots of hydroponically grown maizeseedlings was measured using the short-lived isotope 13N. Itis shown to be described by a four compartment model, recognizablynitrogen in the root bathing solution, nitrogen which is readilyexchangeable from the root, nitrogen bound in the root, andnitrogen transported from the root. Some of the absorbed activity leaks back into the root bathingsolution with the efflux from the root, as a fraction of theinflux, increasing with concentration to be greater than 0–8at external nitrate ion concentrations above about 1.0 mol m–3.The capacity of the exchangeable root pool increases with externalnitrate ion concentration, approaching the expected cytoplasmicnitrate ion content at the highest external nitrate ion concentrationsstudied (70 mol m–3). The investigation has highlighted the problems of interpretinguptake profiles in experiments for which the 10 min half-lifeof 13N dictates experimental times that are comparable withthe times for saturation of root pools. An investigation to determine the magnitude of the back reactionswhich occur during net ammonium uptake by roots and during netammonium assimilation within roots was undertaken with maize(Zea mays L.). Ten-day-old seedlings, which had been grown on250 mmol m–3 ammonium at pH 4 or 6, were pretreated for3 h in the absence or presence of 500 mmol m –3 MSX (methionine-DL-sulphoximine),an inhibitor of the glutamine synthetase-catalysed pathway ofammonium assimilation. They were then exposed for 2 h to 99A% 15N-ammonium ± MSX. Substantial ammonium cycling occurredduring net ammonium uptake. Efflux was enhanced by MSX treatment,reflecting a 2- to 3-fold accumulation of ammonium in the roottissue. Influx of ammonium was also increased by treatment withMSX, indicating that influx was enhanced when products of ammoniumassimilation were dissipated. The decline in root 14N-ammoniumaccounted for only a small fraction of the 14N-ammonium recoveredin the ambient 15N-ammonium solution, revealing a substantialgeneration of endogenous 14N-ammonium during the 2 h exposure.The net quantity of ammonium generated was increased appreciablywhen assimilation of ammonium was restricted by MSX and it wasestimated to occur at least 50% faster than net ammonium uptake.Presence of MSX severely decreased translocation of 15N to shootsbut had a smaller influence on incorporation of 15N into macromoleculesof the root tissue. At low nitrogen (N) supply, it is well known that rye has ahigher biomass production than wheat. This study investigateswhether these species differences can be explained by differencesin dry matter and nitrogen partitioning, specific leaf area,specific root length and net assimilation rate, which determineboth N acquisition and carbon assimilation during vegetativegrowth. Winter rye (Secale cereale L.), wheat (Triticum aestivumL.) and triticale (X Triticosecale) were grown in solution cultureat relative addition rates (RN) of nitrate-N supply rangingfrom 0.03–0.18 d-1and at non-limiting N supply under controlledconditions. The relative growth rate (RW) was closely equalto RNin the range 0.03–0.15 d-1. The maximalRW at non-limitingnitrate nutrition was approx. 0.18 d-1. The biomass allocationto the roots showed a considerable plasticity but did not differbetween species. There were no interspecific differences ineither net assimilation rate or specific leaf area. Higher accumulationof N in the plant, despite the same relative growth rate atnon-limiting N supplies, suggests that rye has a greater abilityto accumulate reserves of nitrogen. Rye had a higher specificroot length over a wide range of sub-optimal N rates than wheat,especially at extreme N deficiency (RN=0.03–0.06 d-1).Triticale had a similar specific root length as that of wheatbut had the ability to accumulate N to the same amount as ryeunder conditions of free N access. It is concluded that thebetter adaptation of rye to low N availability compared to wheatis related to higher specific root length in rye. Four maize (Zea mays L.) hybrids were grown hydroponically for4 weeks with 20 mM ammonium or nitrate as the sole nitrogensource. Dry matter production was strongly depressed by ammoniumnutrition in the hybrid Helga relative to plants grown on nitrate,and moderately decreased in the hybrid Melina. Ammonium hadno inhibitory effect on total yield in the other two hybrids(Ramses and DK 261). The relative growth rate (RGR) of rootsand shoots of the sensitive hybrid Helga decreased significantlyunder ammonium nutrition during the first 2 weeks of the experiment,while at the end of the experiment nitrogen form had no effecton the RGR in any of the four hybrids. The strong reductionin RGR of Helga in the early seedling stage was correlated withthe accumulation of twice the concentration of free ammoniumin the shoot tissue relative to the other hybrids. Helga wastherefore unable to sufficiently detoxify ammonia in the roots.Root concentrations of water soluble carbohydrates (WSC) inHelga and Melina in the early seedling stage did not differunder ammonium and nitrate nutrition. In contrast, Ramses andDK 261 both had elevated WSC concentrations in ammonium-fedroots. It is hypothesized that a sufficient supply of carbonskeletons for ammonium assimilation in the roots is requiredfor maximum growth under high ammonium concentrations, and thatthere is genotypic variability in this physiological trait. Spruce (Picea abies (L.) Karst.) seedlings were asepticallycultivated and the effects of different N-nutrition on net uptakeand reduction of nitrate were investigated. The characteristicsof nitrate uptake were calculated, Ks as 0?2 mol m–3 andVmax as 18 µmol g–1 d–1. Low pH, and Al3+ in the medium caused adecrease in nitrate uptake rate. An in vivo assay was set upwhich allowed the measurement of NRA in both roots and needlesof spruce seedlings. The in vivo nitrate reductase activitywas repressed by ammonium and stimulated by nitrate. Nitratereduction was similar to nitrate uptake, negatively affectedby low pH and ammonium. The form of fine roots of Sitka spruce (Picea sitchensis Bong.Carr.) when grown in immediate proximity to roots of Scots pine(Pinus sylvestris L.) and when grown separately was comparedusing split-root systems. When spruce roots were intimatelymixed with pine roots the mean length of individual spruce lateralswas significantly greater, while the length: weight ratio andnumber of root tips: weight ratio were smaller than when grownalone. Apple seedlings, Pyrus malus L., were grown in complete nutrient solutions containing nitrate, ammonium, or ammonium plus nitrate as the nitrogen source. Uptake of nitrogen was calculated from depletion measurements of the nutrient solutions and by using 15N labelled nitrate and ammonium salts. If the plants received nitrogen as ammonium only or as nitrate only, the amounts of nitrogen taken up were similar. However, if the seedlings were supplied with ammonium nitrate, the amount of nitrate-nitrogen assimilated was only half that of ammonium. Nevertheless, if ammonium and nitrate were supplied to a plant with a split-root system, with each root half receiving a different ion, the uptakes were similar. The possibility of independent inhibition by ammonium of both nitrate uptake and reduction in the roots is discussed.  相似文献

The primary roots of the "Golden Cross Bantam 70" cultivar ofZea mays are agravitropic in darkness and their orthogravitropismis light-dependent. Analysis of the agravitropic roots providesimportant information about the mechanism of orthogravitropism.However, the underlying mechanism of the agravitropic responsein darkness is unknown. We found that the growth of intact primaryroots was inhibited by gravitropic stimulation (i.e., changingthe orientation of the roots from vertical to horizontal) indarkness, but that of detipped roots was not. The role of calciumin this gravistimulation-dependent inhibition of growth wasinvestigated using apical 5-mm segments of the primary roots.The gravistimulation-dependent inhibition of growth was preventedby applying 10 mM MES-KOH buffer at pH 6.0 to the root cap.By contrast, the application of 0.1–1 mM buffer at pH6.0 and 10 mM buffer at pH 4.5–5.0 allowed the gravistimulation-dependentinhibition of growth. Furthermore, when the buffer of 10 mM(pH 6.0) contained 1–5 mM CaCl2, the gravistimulation-dependentinhibition of growth was apparent. By contrast, when weak (1mM) buffer at pH 6.0 or 10 mM buffer at pH 4.5 contained 5 mMEGTA, no gravistimulation-dependent inhibition of growth wasobserved. Thus, the gravistimulation-dependent inhibition ofgrowth in darkness seemed to be mediated by an increase in thelevel of free Ca2+ in the root tip. These results suggest thatfree Ca2+ in the apoplast of the root tip plays an importantrole in the agravitropic response in darkness as well as inorthogravitropism under light of the roots of this cultivarof Zea mays. Methods are described for studying the uptake, by hydroponicallygrown Zea mays seedlings, of ammonium and nitrate ions labelledwith radioactive nitrogen-13, which has a half-life often minutes.For nitrate only, some of the activity absorbed by the rootexchanges back out again into the root bathing solution. Theamount of this activity is about five times too large to beattributable to exchange with ions in the root cortical apoplasm.Much of it must be transferred from the root symplasm with ahalf-time of exchange of 2–5 min. After exposing the rootto the labelled solution, equilibrium rates of transport to,and distribution in the shoot were attained within 2 min, fornitrate, or 5 min, for ammonium. The pools within the root,and the transport pathway through which the label passes musttherefore rapidly attain the specific activity of the nutrientsolution. Primary nitrogen metabolism in transformed root cultures ofDatura stramonium was observed by in vivo 15N NMR. Treatmentof the root cultures with the plant growth regulators -naphthaleneaceticacid (NAA) and kinetin caused a de-differentiation of the roottissue, together with perturbation of primary and secondarynitrogen metabolism. The levels of newly-synthesized glutamineand glutamate during ammonium assimilation were depleted relativeto control cultures, whereas GABA biosynthesis was enhanced.Although GABA production could be stimulated by a decrease incytoplasmic pH (whether imposed artificially or induced by hypoxia),observation of the roots during phytohormone treatment by 31PNMR showed that the cytoplasmic pH remained stable, indicatingthat the perturbation of nitrogen metabolism in the de-differentiatedroots must be due to other causes. Morgan, M. A. and Jackson, W. A. 1988. Inward and outward movementof ammonium in root systems: transient responses during recoveryfrom nitrogen deprivation in presence of ammonium.— J.exp. Bot. 39: 179-191. Net ammonium uptake by 20-d-old wheat (Triticum aestivum cv.Kleiber) and oat (Avena sativa cv. Tarok) seedlings was increased5- to 10-fold when the seedlings were deprived of nitrate duringthe 14-20 d period. The effect of nitrogen deprivation was toincrease net 15N-ammonium influx and decrease net 14N-ammoniumefflux during a 1 h assay period. The sizeable rate of net 15N-ammoniuminflux resulting from nitrogen deprivation was stimulated furtherby prior exposure of the seedlings to 14N-ammonium for 5 h.Additional exposure to 14N-ammonium caused the stimulated rateof 15N-ammonium influx to decline. During the 1 h assays in15N-ammonium, net 14N-ammonium efflux increased after 2 h exposureto 14N-ammonium, peaked at 5–10 h, and then declined.The consequence of the differential response of the influx andefflux processes in wheat was a marked decrease in net ammoniumuptake in the initial 2–5 h, followed by a recovery which,in turn, was followed by a slow decline. In oat, there was norecovery in net ammonium uptake after 2–5 h. Interference in ammonium assimilation by presence of methioninesulphoximine after 5 h did not inhibit expression of the ammonium-stimulatednet 15N-ammonium influx at 10 h but did substantially increasenet 14N-ammonium efflux. In nitrogen depleted seedlings, andin those exposed to 14N-ammonium for 2 h, subsequent net 14N-ammoniumefflux during 1 h in 15N-ammonium exceeded the quantity of 14N-ammoniuminitially in the roots. The increase in 15N-ammonium influx upon nitrogen deprivation,its further stimulation with 5-10 h exposure to ammonium andits subsequent decline, are discussed as possibly resultingfrom (a) the operation of two ammonium influx systems (b) theinterplay of tissue ammonium and a product of its assimilationrespectively acting as positive and negative effectors of asingle influx system and (c) variations in energy supply fromthe shoots. The xylem exudation of detopped 7-d-old seedlings of Zea maysL. doubled when KCI was present in the root medium comparedto seedlings maintained on water. It was further enhanced whenKCI was replaced by nitrogen compounds such as nitrate, ammoniumand glutamine. The role of the nitrate assimilation pathwayon the enhancement of xylem exudation rate was investigatedusing tungstate, an inhibitor of nitrate reductase (NR) activity,and phosphinothricin or methionine sulphoximine, inhibitorsof glutamine synthetase (GS) activity. The sap levels of NO3,NH4+, glutamine, and asparagine was used to ascertain the invivo inhibition of both enzymes. The tungstate effects werealso checked by measuring leaf in vitro NA activity and NR proteincontent. Xylem exudation rate of detopped seedlings fed withKNO3 decreased when the nitrate assimilation pathway was blockedeither at the NR or at GS sites. This decrease was preventedwhen urea (acting as NH4+ supply) was given simultaneously withtungstate. KNO3 does not act directly on exudation, but throughthe involvement of NH4+. The involvement of glutamine was alsoshown since GS inhibition resulted in a cancellation of theenhancing effect of KNO3 on exudation. As change of exudationrate was not linked to change in sap osmolarity, it is assumedthat the assimilation chain could modify root water conductance.The role of glutamine was discussed. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. Theflows of nitrate, ammonium, and malate between root and shootwere modelled to identify the site(s) of inorganic nitrogenassimilation and to show the possible role of malate in a pH-statmechanism. Only in the xylem of nitrate-fed plants did inorganicnitrogen, in the form of nitrate, play a role as the transportsolute. The nitrate percentage of total nitrogen in the xylemsap generally increased in parallel with the external nitrateconcentration. The contribution of the shoot to nitrate reductionincreased with higher nitrate supply. Under salt treatment relativelymore nitrate was reduced in the root as compared with non-treatedplants. Ammonium was almost totally assimilated in the root,with only a minor recycling via the phloem. Nitrate reductaseactivities measured in vitro roughly matched, or were somewhatlower than, calculated rates of nitrate reduction. From therates of nitrate reduction (OH -production) and rates of malatesynthesis (2H+-production) it was calculated that malate accumulationcontributed 76, 45, or 39% to the pH-stat system during nitratereduction in plants fed with 0.2, 1.0 or 4.0 mM nitrate, malateflow in the phloem played no role. In tissues of ammonium-fedplants no malate accumulation was found and malate flows inxylem and phloem were also relative low. Cultures of nine potato genotypes (seven Solanum tuberosum oneS. sparsipilum and one S. oplocense genotypes) were examinedfor their response to growing on medium containing either 60mol m–3, 40 mol m–3 or 20 mol m–3 nitrogen.Genotypes differed in their response to nitrogen. Reducing thenitrogen regime tended to produce taller plants with longerinternodes, shoots had larger leaves but contained less chlorophyll.No change in fresh weight or number of nodes was observed. Genotypex nitrogen interactions were significant for chlorophyll content,shoot length and internode length. Results suggest that thechanges observed were as a result of changes in the total nitrogenlevel rather than changes in the ammonium : nitrate ratio.
