Salt Tolerance in the Triticeae: Ion Discrimination in Rye and Triticale   总被引:5,自引:0,他引:5  
When rye and triticale accessions were grown in saline hydroponicculture they exhibited the low Na and high K concentrationsin their leaves which are characteristic of the enhanced K/Nadiscrimination trait originally found in the D genome of wheat.This trait was not consistently improved by the presence ofthe D genome in octaploid triticale or in D genome substitutionlines of hexaploid triticale. The presence of the rye genomedid not significantly affect anion concentrations within theleaves. At high salt concentrations (250 mol m–3 NaCl+12.5mol m–3 CaCl2) the triticales were more tolerant thanthe rye accessions or a DDRR-genome tetraploid, with two triticalelines being almost as salt-tolerant as barley. Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. Photochemical properties were determined for undegraded phytochromepurified from lyophilized rye seedlings (Secale cereale cv.Cougar). The preparation was shown to be a two-component systemduring phototransformations and dark transformation from red-light-inducedphotostationary state. The mole fraction of PFR at the 665-nm-light-inducedphotostationary state was 0.84. The metabolism of winter rye seedlings (Secale cereale, L. ev.Winter) cultured in 99.8 per cent D2O was investigated. Comparedwith water-grown seedlings, the protein content was much lowerin the D2O-cultured seedlings and the pattern of incorporationof [3H]leucine and [3H]phenylalanine into protein was substantiallydifferent. Seedlings cultured in D2O incorporated [3H]thymidineinto DNA, but did not take up [3H]uridine. The results suggestthat some of the toxic effects of D2O culture on higher plantscan be attributed to a partial block of protein synthesis.  相似文献

The single-gene mutation afila in pea (Pisum sativum L.) resultsin the replacement of proximal leaflets with branched tendrils,thereby reducing leaf area. This study investigated whethertheafila line could adjust biomass partitioning when exposedto varying nutrient regimes, to compensate for reduced leafarea, compared with wild-type plants. Wild-type and afila near-isogeniclines were grown in solution culture with nitrate-N added toinitially N-starved seedlings at relative addition rates (RN)of 0.06, 0.12, 0.15 and 0.50 d-1. The relative growth rate (RW)of the whole plants closely matched RNat 0.06 and 0.12 d-1,but higher RNresulted in a slightly higher growth rate. At agiven RN, the wild-type line had lower plant nitrogen statusthan the afila line. RWof the roots of the afila line was lessthan RWof the roots of the wild-type at the three higher ratesof N supply despite a greater accumulation of N in the rootsof the afila plants. Consequently, plant nitrogen productivity(growth rate per unit nitrogen) was lower for afila. Dry matterallocation was strongly influenced by nitrogen status, but nodifferences in shoot–root dry matter allocation were foundbetween wild-type and afila with the same plant N status. Theseresults imply that decreased leaf area as a result of the single-genemutation afila affects dry matter allocation, but only accordingto its effect on the nitrogen status. Changes in freezing tolerance of winter rye (Secale cerealeL. cv. Voima) were determined for leaf tissues during a 1-weekcold stress, which was performed by transferring the 7-d-oldseedlings from a greenhouse (25°C, long day) to 3°Cand short day conditions. The development of cold hardeningwas shown by using an ion leakage test and by determining theamounts of carbohydrates, soluble proteins and RNA. The firstevidence of the development of freezing resistance was foundafter 1 d at low temperature, i.e. an LT50 value increased from-5 to -7°C. Plants cold treated for 7 d reached an LT50value of -9°C. This increase in freezing tolerance was foundto be associated with the increased levels of soluble carbohydrates,total RNA and soluble proteins. We studied the course of early leaf area expansion and specificleaf area (SLA) in potato (Solanum tuberosum L.) and wheat (Triticumaestivum L.) genotypes and tested whether air temperature explainsdifferences in these courses within different environments.Such knowledge can be used to improve crop growth modelling.The relative rate of leaf area expansion (RL) of potato andwheat decreased with thermal time, but was nearly linear upto a leaf area index (L) of 1.0. TheRL (L < 1; mean: 17.9x 10-3°C-1 d-1) of potato showed an interaction betweengenotype and environment, and varied with year. TheRL (L <1; mean: 7.1 x 10-3°C-1 d-1) of winter wheat was lower thanthat of spring wheat (mean: 10.9 x 10-3°C-1 d-1), and bothvaried with year. SLAof potato increased nearly linearly withthermal time from 5 to 15 m2 kg-1at 50% emergence, to 20 to25 m2 kg-1at 155°Cd, and then decreased slightly. The SLAofboth winter and spring wheat began at 16 to 23 m2 kg-1and inmost cases increased slightly with thermal time. In potato,regression parameters of SLAwith thermal time were affectedby environment (management conditions and year) and genotype;in wheat they were affected by environment (year and site).Treatment effects on RLof potato were not correlated with thoseon SLA , and were only partly correlated for wheat. Thereforewe conclude that the early foliar expansion of potato is associatedwith a strong increase in SLA , and not so for wheat. Rye (Secale cereale cv. Rheidol) and wheat (Triticum aestivumcv. Mardler) were grown at shoot/root temperatures of 20/20°C (warm grown, WG plants), 8/8 °C (cold grown, CG plants)and 20/8 °C (differential grown, DG plants). Plants fromcontrasting growth temperature regimes were standardized andcompared using a developmental timescale based on accumulatedthermal time (°C d) at the shoot meristem. Accumulationof dry matter, nitrogen and potassium were exponential overthe time period studied (150–550 °C d). In rye, therates of plant dry matter and f. wt accumulation were linearlyrelated to the temperature of the shoot meristem. However, inwheat, although the rates of plant dry matter and f. wt accumulationwere temperature dependent, the linear relationship with shootmeristem temperature was weaker than in rye. The shoot/rootratio of rye was stable irrespective of growth temperature treatment,but the shoot/root ratio of wheat varied with growth temperaturetreatment. The shoot/root ratio of DG wheat was 50% greaterthan WG wheat. In both cereals, nutrient concentrations anddry matter content tended to be greater in organs exposed directlyto low temperatures. The mean specific absorption rates of nutrientswere calculated for the whole period studied for each species/temperaturecombination and were positively correlated with both plant shoot/rootratio and relative growth rate. The data suggest that nutrientuptake rates were influenced primarily by plant demand, withno indication of specific nutrient limitations at low temperatures. Summary The heat shock protein profiles from Secale cereale L. cv Imperial, Triticum aestivum L. cv Chinese Spring, S. cereale x T. aestivum amphiploid, and the seven disomic S. cereale addition lines to T. aestivum were used to compare the wheat, rye, and triticale Heat Shock Protein profiles and to study the influence of the rye genome on heat shock protein expression in triticale. Three-day-old seedlings were heat shocked for 2 h at 40 °C in the presence of 35S-methionine, and polypeptides from root tissues were subjected to one- or two-dimensional gel electrophoresis. The wheat and rye heat shock protein profiles each consisted of > 150 heat shock proteins, of which 94 were sufficiently reproducible to construct a standard map. There were 11 unique rye heat shock proteins compared to 22 unique wheat heat shock proteins. The triticale heat shock protein profile resembled the rye parent more than the wheat parent. There were 22 heat shock proteins expressed uniquely by wheat that were not expressed in triticale. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. In situ hybridization with cloned, repetitive DNA probes andtotal genomic DNA enables the parental origin of all chromosomesto be established in metaphases of triticale tritordeum F1hybrids (2n=6x=42). Nuclei contain seven chromosomes of Hordeumchilense origin, seven from Secale cereale and 28 of wheat origin.When used as a probe, total genomic rye DNA labelled the ryechromosomes strongly and uniformly along their lengths, withbrighter regions coincident with the terminal heterochromatin.The probe labelled the wheat-origin chromosomes weakly and wasalmost undetectable on the H. chilense-origin chromosomes. Incontrast, under the same conditions, H. chilense DNA hybridizedstrongly to the H. chilense- and, with intermediate strength,to the S. cereale-origin chromosomes, excluding the subtelomericheterochromatin: it hybridized only weakly to the wheat chromosomes,in some experiments revealing characteristic bands on wheatchromosomes. Cloned repetitive DNA probes from rye and H. chilensewere used as probes to identify the linkage groups of all oftheir own-species chromosomes. Analysis of hybridization patternsof various probes to prophase and interphase nuclei indicatedthat there are many non-random features in the localizationof both repetitive DNA and whole chromosomes, although generalpatterns of nuclear organization have yet to emerge. Both theparticular lines used and the techniques developed here arelikely to be valuable for production and characterization ofplant breeding material. Summary The effects of A1 on the growth and mineral composition of different cultivars of triticale (X Triticosecale, Wittmack), wheat (Triticum aestivum L.) and rye (Secale cereale L.) growing in 1/5 strength Steinberg solutions containing 0 or 6 ppm A1 were evaluated after 32 days. Aluminum increased the concentrations of P and K in the roots and K in the tops of most of the cultivars tested. A1 tolerant triticale retained a lower concentration of Mg in the roots and tops than the A1 sensitive triticale, when subjected to A1 stress. In addition, A1 treatments resulted in smaller increases in root P for the A1 tolerant triticale than for the A1 sensitive cultivars.The concentration of root Ca and P of the A1 tolerant wheat cultivars were significantly below that of the more sensitive plants. Aluminum tolerance in rye appeared to be associated with lower Ca and higher Mg concentrations in the tops. The accumulation of P and A1 in the roots was characteristic of sensitivity in triticale, wheat and rye.  相似文献

The specific respiration rates of nodulated root systems, ofnodules and of roots were determined during active nitrogenfixation in soya bean, navy bean, pea, lucerne, red clover andwhite clover, by measurements on whole plants before and afterthe removal of nodule populations. Similar measurements weremade on comparable populations of the six legumes, lacking nodulesbut receiving abundant nitrate-nitrogen, to determine the specificrespiration of their roots. All plants were grown in a controlled-environmentclimate which fostered rapid growth. The specific respiration rates of nodulated root systems ofthe three grain and three forage legumes during a 7–14-dayperiod of vegetative growth varied between 10 and 17 mg CO2g–1 (dry weight) h–1. This mean value consistedof two components: a specific root respiration rate of 6–9mg CO2 g–1 h–1 and a specific nodule respirationrate of 22–46 mg CO2 g–1 h–1. Nodule respirationaccounted for 42–70 per cent of nodulated root respiration;nodule weight accounted for 12–40 per cent of nodulatedroot weight. The specific respiration rates of roots lackingnodules and utilizing nitrate nitrogen were generally 20–30per cent greater than the equivalent rates of roots from nodulatedplants. The measured respiratory effluxes are discussed in thecontext of nitrogen nitrogen fixation, nitrate assimilation. Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control,rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. Seedlings of Secale cereale cv. Rheidol and Triticum aestivumcv. Mardler were grown at shoot/root temperatures of 20/20 °C,20/8 °C and 8/8 °C. During vegetative growth both cerealsproduced leaves, tillers and roots in a defined pattern, ata species-specific rate which was linearly related to the temperatureof the shoot meristem. Thus, plant development could be standardizedon a temperature x time (°C d) basis despite contrastinggrowth-temperature treatments.
