Effect of osmotic stress and sodium nitroprusside (SNP, NO donor) pretreatment on growth and proline metabolism of wheat seedlings was investigated. Polyethylene glycol 6000 treatment for 2, 4 and 6 d could be termed as mild, moderate and severe stress, respectively, according to decrease in the relative water content. Severe osmotic stress significantly decreased the growth and photochemical efficiency, and increased proline content due to activation of its synthesis. 0.2 mM SNP pretreatment enhanced growth of wheat seedlings, increased variable to maximum fluorescence ratio (Fv/Fm) and fluorescence yield, while decreased proline content. However, 2 mM SNP retarded the seedlings growth and chlorophyll a fluorescence, and increased proline accumulation. Our results showed that NO might be involved in the regulation of osmotic stress in a concentration-dependent manner.  相似文献

Leaf rolling observed in some crops such as maize, rice, wheat and sorghum is an indicator of decreased water status. Moderate leaf rolling not tightly or early increases the photosynthesis and grain yield of crop cultivars under environmental stresses. Moreover, the effects of exogenous abscisic acid (ABA) on stomatal conductance, water status and synthesis of osmotic compounds are a well-known issue in plants subjected to water deficit. However, it is not clear how the cross-talk of ABA with H2O2 and osmolyte compounds affects the leaf rolling mechanism. Regulation mechanism of leaf rolling by ABA has been first studied in maize seedlings under drought stress induced by polyethylene glycol 6000 (PEG 6000) in this study. ABA treatment under drought stress reduced hydrogen peroxide (H2O2) content and the degree of leaf rolling (%) while the treatment-induced ABA synthesis, osmolyte levels (proline, polyamine and total soluble sugars) and some antioxidant enzyme activities in comparison to the plants that were not treated with ABA. Furthermore, exogenous ABA up-regulated the expression levels of arginine decarboxylase (ADC) and pyrroline-5-carboxylate synthase (P5CS) genes and down-regulated polyamine oxidase (PAO), diamine oxidase (DAO) and proline dehydrogenase (ProDH) gene expressions. When endogenous ABA content was decreased by the treatment of fluoridone (FLU) that is an ABA inhibitor, leaf rolling degree (%), H2O2 content and antioxidant enzyme activities increased, but osmolyte levels, ADC and P5CS gene expressions decreased. Finally, the treatment of ABA to maize seedlings exposed to drought stress resulted in the stimulation of the antioxidant system, osmotic adjustment and reduction of leaf rolling. We concluded that ABA can be a signal compound cross-talking H2O2, proline and polyamines and thus involved in the leaf rolling mechanism by providing osmotic adjustment. The results of this study can be used to provide data for the molecular breeding of maize hybrids with high grain yield by means of moderately rolled leaves.  相似文献

Proline accumulation and mobilization in roots of 7-d-old seedlings of wheat genotypes varying in sensitivity towards water stress were compared. Water stress was induced by polyethylene glycol (PEG-6000; osmotic potential −1.5 MPa) in the presence of 0.1 mM abscisic acid (ABA), 1 mM calcium chloride, 0.5 mM verapamil (Ca2+ channel blocker), 0.5 mM fluridone (inhibitor of ABA biosynthesis). While both the genotypes did not differ in total proline accumulation, rate of proline accumulation and utilization was higher in tolerant genotype C 306 as compared to susceptible genotype HD 2380. The treatment with ABA and CaCl2 caused further increase in proline accumulation during stress and reduced its mobilization during recovery. The membrane stability and elongation rate of roots was observed to be higher at ABA and calcium treatment in both the genotypes under stress. As was evident from inhibitor studies, the tolerant genotype was more responsive to ABA and the susceptible one to calcium. Nitric oxide (NO) is an important plant signaling molecule that has a vital role in abiotic stress tolerance. In the present study, we assessed drought-induced (15 and 30% PEG, polyethylene glycol) damage in wheat (Triticum aestivum L. cv. Prodip) seedlings and mitigation by the synergistic effect of exogenous Arg (0.5 mM l-Arginine) and an NO donor (0.5 mM sodium nitroprusside, SNP). Drought stress sharply decreased the leaf relative water content (RWC) but markedly increased the proline (Pro) content in wheat seedlings. Drought stress caused overproduction of reactive oxygen species (ROS) and methylglyoxal (MG) due to the inefficiency of antioxidant enzymes, the glyoxalase system, and the ascorbate-glutathione pool. However, supplementation with the NO donor and Arg enhanced the antioxidant defense system (both non-enzymatic and enzymatic components) in drought-stressed seedlings. Application of the NO donor and Arg also enhanced the glyoxalase system and reduced the MG content by increasing the activities of the glyoxalase system enzymes (Gly I and Gly II), which restored the leaf RWC and further increased the Pro content under drought stress conditions. Exogenous NO donor and Arg application enhanced the endogenous NO content, which positively regulated the antioxidant system and reduced ROS production. Thus, the present study reveals the crucial roles of Arg and NO in enhancing drought stress tolerance in wheat seedlings by upgrading their water status and reducing oxidative stress and MG toxicity.  相似文献

Salicylic acid (SA) and nitric oxide (NO) are reported to alleviate the damaging effects of stress in plants rather similarly when applied at appropriate low concentrations. An experiment was therefore conducted to study the impact of SA, sodium nitroprusside (SNP; as NO donor), and methylene blue (MB; as a guanylate cyclase inhibitor) on wheat seedling performance under osmotic stress. Osmotic stress significantly reduced shoot fresh weight (SFW), chlorophyll contents (Chla, Chlb, total Chl), and membrane stability index (MSI) and also increased malondialdehyde (MDA) level, lipoxygenase (LOX) activity, and hydrogen peroxide production. Moreover, enzymatic antioxidant activities including superoxide dismutase, guaiacol peroxidase, and glutathione reductase activity were enhanced under osmotic stress. On the contrary, SA or SNP pretreatment reduced the damaging effects of osmotic stress by further enhancing the antioxidant activities that led to increased SFW, Chl, and MSI and reduced MDA level and LOX activity. However, pretreatment of plants with MB reversed or reduced the protective effects of SA and SNP suggesting that the protective effects were likely attributed to NO signaling. Therefore, NO may act as downstream of SA signaling in reduction of induced oxidative damage in wheat seedlings.  相似文献

Signalling process of water stress‐induced abscisic acid (ABA) accumulation was investigated in maize (Zea mays L.) leaf and root tissues. Potent free‐radical scavengers and reducing agents, N‐acetyl cysteine (NAC) and cystein (Cys), significantly inhibited or nearly completely blocked dehydration‐induced ABA accumulation. Dithiothreitol (DTT), a reducing agent but not a free‐radical scavenger, also significantly inhibited such accumulation whereas solely free‐radical scavengers, dimethyl sulphoxide (DMSO) and melatonin (Mela), had no effects. Moreover, water stress‐induced ABA accumulation was not affected either by free radicals, such as superoxide anion and hydrogen peroxide, or by oxidants such as KIO4. These observations suggest that the blocking of water stress‐induced ABA accumulation resulted from the reducing effect, rather than from anything associated with free radicals. The disulphide bond might be crucial to the reactivity of some signal element(s) in the signalling process of water stress‐induced ABA accumulation. To test the hypothesis, we used a sulfhydryl modifier, iodoacetamide (IOA), and found that it nearly totally blocked the water stress‐induced ABA accumulation. Furthermore, an impermeable sulfhydryl modifier, p‐chloromercuriphenylsulphonic acid (PCMBS), could also inhibit the water stress‐induced ABA accumulation in the leaf tissues. These results indicate that water stress‐perception protein(s) or receptor(s) may be located on the plasmalemma and a sulfhydryl group in the extracellular domain is critical to the reactivity of the speculated water stress receptors. Cys, DTT and IOA did not lead to a decrease of the baseline ABA level, i.e. in non‐stressed roots. Result indicates that their blocking of water stress‐induced ABA accumulation occurred upstream of the ABA biosynthesis pathway, i.e. in the signalling process that initiates such accumulation.  相似文献

Hydrogen peroxide (H2O2) functions as a signal molecule in plants under abiotic and biotic stresses. Leaves of detached maize (Zea mays L.) seedlings were used to study the function of H2O2 pretreatment in osmotic stress resistance. Low H2O2 concentration (10 mM) which did not cause a visual symptom of water deficit (leaf rolling) was applied to the seedlings. Exogenous H2O2 alone increased leaf water potential, endogenous H2O2 content, abscisic acid (ABA) concentration, and metabolite levels including soluble sugars, proline, and polyamines while it decreased lipid peroxidation and stomatal conductance. Osmotic stress induced by polyethylene glycol (PEG 6000) decreased leaf water potential and stomatal conductance but enhanced lipid peroxidation, endogenous H2O2 content, the metabolite levels, and ABA content. H2O2 pretreatment also induced the metabolite accumulation and improved water status, stomatal conductance, lipid peroxidation, ABA, and H2O2 levels under osmotic stress. These results indicated that H2O2 pretreatment may alleviate water loss and induce osmotic stress resistance by increasing the levels of soluble sugars, proline, and polyamines thus ABA and H2O2 production slightly decrease in maize seedlings under osmotic stress.  相似文献

不同浓度(0.01~5.00mmol/L)的外源一氧化氮(NO)供体硝普钠(SNP)以浓度依赖性的性式诱导150mmol/LNaCl胁迫下小麦(Triticum aestivum L.cv.Yangmai 158)幼苗叶片脯氨酸的累积.其中0.1 mmol/L的SNP效果最明显,而结合采用NO清除剂c-PTIO和血红蛋白的处理均分别逆转了该效应.研究结果还发现:0.1 mmol/L SNP诱导的脯氨酸累积还可能有利于盐胁迫下小麦幼苗的保水性;0.1 mmol/L的SNP显著激活了内源ABA的合成,而结合血红蛋白的处理则证实,在外源ABA诱导脯氨酸累积的过程中NO可能作用于ABA信号分子的下游,但NO和ABA信号分子在此诱导反应中不存在累积效应.进一步研究脯氨酸合成和降解的酶促反应途径,发现外源NO处理前4天内可能主要是通过提高△'-吡咯啉-5-羧酸合成酶(P5CS)的活性来促进脯氨酸的合成,以后直至第8天主要是通过抑制脯氨酸脱氢酶(ProDH)的活性来抑制脯氨酸的降解;ABA对于P5CS和ProDH活性的调节能力弱于NO.此外,Ca2 在NO诱导的盐胁迫下小麦叶片脯氨酸累积的信号分子途径中起重要的介导作用.  相似文献

The roles of the plasma-membrane (PM) NADPH oxidase in abscisic acid (ABA)- and water stress-induced antioxidant defense were investigated in leaves of maize ( Zea mays L.) seedlings. Treatment by exogenous ABA (100 micro M ABA) or osmotic stress (-0.7 MPa induced by polyethylene glycol) significantly increased the activity of the PM NADPH oxidase, the production of leaf O(2)(-), the activities of several antioxidant enzymes (superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase), and the contents of antioxidant metabolites (ascorbate and reduced glutathione). Pretreatment with three different inhibitors of NADPH oxidase (diphenylene iodonium, imidazole and pyridine) or an inhibitor of ABA biosynthesis (tungstate) reduced the increase in the activity of the PM NADPH oxidase and the production of leaf O(2)(-), and the capacity of antioxidant defense systems mediated by ABA. The inhibitory effects above caused by tungstate were reversed by exogenous ABA. These data indicate that NADPH oxidase is involved in the ABA-induced production of active oxygen species (AOS), and our results depict a minimal chain of events initiated by water stress-induced ABA accumulation, which then triggers the production of AOS by membrane-bound NADPH oxidase, resulting in the induction of antioxidant defense systems against oxidative damage in plants.  相似文献

The effects of water stress and osmotic stress (sorbitol treatment) on the production of putrescine and proline in excised rice leaves were compared. Osmotic stress and water stress were found to affect differentially the levels of putrescine and proline in excised rice leaves. Putrescine accumulation is induced by osmotic stress, whereas proline accumulation is induced by water stress. The effects of ABA on the levels of proline and putrescine are similar to those of water stress, whereas the effects of jasmonic acid methyl ester (JA-Me) are similar to those of osmotic stress. Water stress results in an increase of endogenous ABA is excised rice leaves. However, neither osmotic stress nor JA-Me has effect on endogenous ABA levels in excised rice leaves. Of particular interest is the finding that proline levels increase when putrescine levels induced by osmotic stress or JA-Me are reduced by D-arginine and -methylornithine. L-arginine and L-ornithine applied exogenously also cause an increase in proline levels. Plants accumulate abscisic acid (ABA) under droughted conditions. Genetic variation in the accumulation of ABA in detached and partially dehydrated leaves of rice has previously been reported, and this was found to be associated with variation in leaf size (smaller leaves made more ABA). Correlation analysis failed to distinguish clearly between a causal relationship between the two traits and close genetic linkage between loci controlling the traits. Here we present a detailed genetic analysis of ABA accumulation in detached and partially dehydrated rice leaves, using a population of F2 plants generated from the lowland × upland cross IR20 (high-ABA) × 63-83 (low-ABA) which was mapped with RFLP and AFLP markers. Several highly significant quantitative trait loci (QTLs) for ABA accumulation and leaf weight were identified. Only one of the minor QTLs for ABA accumulation (accounting for only 4% of the phenotypic variance) was coincident with any QTLs for leaf size such that the high-ABA allele was associated with smaller leaves. This analysis, therefore, showed that the association previously found between ABA accumulation and leaf size was probably largely due to genetic linkage and not to a direct effect of leaf size on ABA accumulation or vice versa. Because of the importance of ABA accumulation in regulating responses of plants to drought stress and the effects of plant size on the rate of development of stress, QTLs for drought-induced ABA accumulation, leaf size and tiller number were compared between rice and wheat. In particular, a possible location in rice was sought for a homoeologue of the major wheat vernalization responsive gene, Vrn1, as this gene is also associated with major effects on leaf size, tiller number and ABA accumulation in wheat. The likelihood of homoeologous loci regulating ABA accumulation, leaf size and tiller number in the two crops is discussed.  相似文献

Dynamics of endogenous NO in the leaves of 7-day-old seedlings of spring wheat (Triticum aestivum L., cv. Debyut) and the effect of exogenous NO donor (sodium nitroprusside, SNP) on the development of oxidative stress and activity of antioxidant enzymes in the leaves under water deficiency were investigated. Quick and phasic accumulation of NO in the leaves was observed under growing dehydration (0–3 h) and subsequent rehydration (0–3 h), which points to identical response of NO signal system to opposite changes in the water status of plants. A decrease in relative turgidity of tissues brought about accumulation of H2O2 and MDA therein. Protective effect of NO donor infiltrated in the leaves was associated with an elevation of ascorbate peroxidase and catalase activities and suppression of lipid peroxidation upon dehydration. Pretreatment with SNP (50–250 μM) induced the elevation of NO level in the leaves both before action of the stress agent and in the beginning (0–30 min) of dehydration. The obtained results suggest that brief increase in endogenous NO is necessary for triggering protective and adaptive responses in wheat plants during the development of water deficiency.  相似文献

Nitric oxide (NO) is a multifunctional gaseous signal in plant. In the present study, we found that pretreatment with NO could significantly improve wheat seeds germination and alleviate oxidative stress against copper toxicity. With the enhancement of copper stress, the germination percentage of wheat seeds decreased gradually. Pretreatment during wheat seed imbibition with sodium nitroprusside (SNP), an NO donor, could greatly reverse the inhibitory effect of the following copper stress to wheat seeds germination. SNP-pretreated seeds also tended to retain higher amylase activities than that of the control without SNP pretreatment. On the other hand, there was no apparent difference in the activities of esterase in wheat seeds pretreated with or without SNP. Further investigations showed that pretreatment with NO donor dramatically stimulated the activities of superoxide dismutase (SOD, EC 1.15.1.1) and catalase (CAT, EC 1.11.1.6), decreased the activities of lipoxygenases, sustained a lower level of malondialdehyde, and interfered with hydrogen peroxide (H2O2) excessive accumulation compared with the control, thereby enhancing the antioxidative capacity in wheat seeds under copper stress. In addition, the seed copper contents were not significant different between those pretreated with SNP and the controls, inferring that protective roles of NO was not responsible for preventing Cu uptake. The effects of sodium nitroprusside (SNP, a donor of NO) on cadmium (Cd) toxicity in lettuce seedlings were studied. SNP was added into hydroponic systems or sprayed directly on the leaves of plants grown with and without Cd. Excess supply of Cd (100 μM) caused growth inhibition, dramatically increased Cd accumulation in both leaves and roots, and inhibited the absorption of Ca, Mg, Fe and Cu. Excess Cd also decreased activities of superoxide dismutase peroxidase and catalase in leaves and roots, and increased the accumulation of superoxide anion (O 2 ·? ), hydrogen peroxide (H2O2) and malondialdehyde (MDA). Root or foliar applications of exogenous NO alleviated Cd-induced growth suppression, especially root application of 250 μM SNP and foliar addition of 500 μM SNP. Addition of SNP promoted the chlorophyll synthesis suggesting that the photosynthesis was up-regulated. Exogenous NO increased Cd-decreased activities of antioxidant enzymes and markedly diminished Cd-induced reactive oxygen species (ROS) and MDA accumulation. Moreover, the absorption of Ca, Mg, Fe and Cu was increased, indicating that exogenous NO stimulated H+-ATPase activity to promote sequestration or uptake of ions. In addition, exogenous NO also inhibited Cd transfer from roots to shoots, which may indicate that Cd retention in roots induced by NO plays a significant role in Cd tolerance in lettuce seedlings. These data suggest that under Cd stress, exogenous NO improves photosynthesis by increasing chlorophyll synthesis, protects lettuce seedlings against oxidative damage by scavenging ROS, helps to maintain the uptake of nutrient elements, and inhibits Cd transferred to shoots effectively.  相似文献

Abscisic acid (ABA) accumulated in detached, wilted leaves of spinach (Spinacia oleracea L. cv Savoy Hybrid 612) and reached a maximum level within 3 to 4 hours. The increase in ABA over that found in detached turgid leaves was approximately 10-fold. The effects of water stress could be mimicked by the use of thin slices of spinach leaves incubated in the presence of 0.6 molar mannitol, a compound which causes plasmolysis (loss of turgor). About equal amounts of ABA were found both in the leaf slices and in detached leaves, whereas 2 to 4 times more ABA accumulated in the medium than in the slices. When spinach leaf slices were incubated with ethylene glycol, a compound which rapidly penetrates the cell membrane causing a decrease in the osmotic potential of the tissue and only transient loss of turgor, no ABA accumulated. Ethylene glycol was not inhibitory with respect to ABA accumulation. Spinach leaf slices incubated in both ethylene glycol and mannitol had ABA levels similar to those found when slices were incubated with mannitol alone. Increases similar to those found with mannitol also occurred when Aquacide III, a highly purified form of polyethylene glycol, was used. Aquacide III causes cytorrhysis, a situation similar to that found in wilted leaves. Thus, it appears that loss of turgor is essential for ABA accumulation.

When spinach leaf slices were incubated with solutes which are supposed to disturb membrane integrity (KHSO3, 2-propanol, or KCl) no increase in ABA was observed. These data indicate that, with respect to the accumulation of ABA, mannitol caused a physical stress (loss of turgor) rather than a chemical stress (membrane damage).

The paper studied the effects of drought stress, selenium (Se) supply and their combination on growth and physiological characteristics of wheat (Triticum aestivum L. cv Shijiazhuang NO. 8) seedlings. The experimental design included two water treatments (well-watered, 75% of maximum field capacity; drought stress, 30% of maximum field capacity) and two Se levels (0; 0.5 mg/kg) to determine whether Se can modify the negative impacts of drought stress on seedling growth and physiological traits. Drought stress caused a marked decline in growth parameters and soluble protein content, whereas it induced an increase in root activity, proline content and the activities of peroxidase (POD) and catalase (CAT) of leaf tissue. On the other hand, Se supply induced an increase in biomass accumulation only under well-watered condition. Under drought stress, Se supply increased free proline content, root activity and the activities of POD and CAT in leaf tissue, but did not significantly affect on growth parameters. These results implied that drought stress brought harmful effects on wheat seedlings, and that Se supply was favorable for biomass accumulation of wheat seedlings under well-watered condition. However, it did not significantly affect on biomass accumulation under drought stress, although it increased root activity and activities of some antioxidant index in experimental periods.  相似文献

Barley (Hordeum vulgare L.) and tomato Lycopersicon esculentum Mill.) were grown hydroponically and examined 2, 5, and 10 d after being deprived of nitrogen (N) supply. Leaf elongation rate declined in both species in response to N stress before there was any reduction in rate of dryweight accumulation. Changes in water transport to the shoot could not explain reduced leaf elongation in tomato because leaf water content and water potential were unaffected by N stress at the time leaf elongation began to decline. Tomato maintained its shoot water status in N-stressed plants, despite reduced water absorption per gram root, because the decline in root hydraulic conductance with N stress was matched by a decline in stomatal conductance. In barley the decline in leaf elongation coincided with a small (8%) decline in water content per unit area of young leaves; this decline occurred because root hydraulic conductance was reduced more strongly by N stress than was stomatal conductance. Nitrogen stress caused a rapid decline in tissue NO 3 - pools and in NO 3 - flux to the xylem, particularly in tomato which had smaller tissue NO 3 - reserves. Even in barley, tissue NO 3 - reserves were too small and were mobilized too slowly (60% in 2 d) to support maximal growth for more than a few hours. Organic N mobilized from old leaves provided an additional N source to support continued growth of N-stressed plants. Abscisic acid (ABA) levels increased in leaves of both species within 2 d in response to N stress. Addition of ABA to roots caused an increase in volume of xylem exudate but had no effect upon NO 3 - flux to the xylem. After leaf-elongation rate had been reduced by N stress, photosynthesis declined in both barley and tomato. This decline was associated with increased leaf ABA content, reduced stomatal conductance and a decrease in organic N content. We studied the responses of leaf water potential (Ψw), morphology, biomass accumulation and allocation, and canopy productivity index (CPI) to the combined effects of elevated CO2 and drought stress in Caragana intermedia seedlings. Seedlings were grown at two CO2 concentrations (350 and 700 μmol mol−1) interacted with three water regimes (60–70%, 45–55%, and 30–40% of field capacity of soil). Elevated CO2 significantly increased Ψw, decreased specific leaf area (SLA) and leaf area ratio (LAR) of drought-stressed seedlings, and increased tree height, basal diameter, shoot biomass, root biomass as well as total biomass under the all the three water regimes. Growth responses to elevated CO2 were greater in well-watered seedlings than in drought-stressed seedlings. CPI was significantly increased by elevated CO2, and the increase in CPI became stronger as the level of drought stress increased. There were significant interactions between elevated CO2 and drought stress on leaf water potential, basal diameter, leaf area, and biomass accumulation. Our results suggest that elevated CO2 may enhance drought avoidance and improved water relations, thus weakening the effect of drought stress on growth of C. intermedia seedings.  相似文献

We examined the carry-over effects of ozone (O3) and/or water stress on leaf phenological characteristics and bud frost hardiness of Fagus crenata seedlings. Three-year-old seedlings were exposed to charcoal-filtered air or 60 nl l–1 O3, 7 h a day, from May to October 1999 in naturally-lit growth chambers. Half of the seedlings in each gas treatment received 250 ml of water at 3-day intervals (well-watered treatment), while the rest received 175 ml of water at the same intervals (water-stressed treatment). All the seedlings were moved from the growth chambers to an experimental field on October 1999, and grown until April 2000 under field conditions. The exposure to O3 during the growing season induced early leaf fall and reduction in leaf non-structural carbohydrates concentrations in the early autumn, as well as resulting in late bud break and reduction in the number of leaves per bud in the following spring. However, O3 did not affect bud frost hardiness in the following winter. On the contrary, water stress did not affect leaf phenological characteristics, leaf and bud non-structural carbohydrates concentrations and bud frost hardiness. There were no significant synergistic or antagonistic effects of O3 and water stress on leaf phenological characteristics, concentrations of leaf and bud non-structural carbohydrates and bud frost hardiness of the seedlings. These results show that the carry-over effects of O3 can be found on the phenological characteristics and leaf non-structural carbohydrates concentrations, although there are almost no carry-over effects of water stress on phenological characteristics and winter hardiness of the seedlings.  相似文献
