Abstract:  The potential of azadirachtin as an insect growth regulator was evaluated in Nezara viridula L. by applying different doses to fifth instar nymphs. At doses of 200–500 ng per insect almost all the individuals died during moulting to adults and the survivors showed a great number of nymphal characteristics, and also died very soon after. At lower doses, from 2 to 50 ng per insect, approximately 50% of the adults seemed normal. No effect was observed during the last nymphal instar, even at the highest dose applied. The fertility and fecundity of adults, males and females, developing from nymphs treated with different low doses of azadirachtin were also studied. When azadirachtin was applied at 20 ng per insect on fifth instar nymphs, females developing from these nymphs showed a conspicuous reduction of fecundity. This same effect was evident when untreated females mated with males obtained from treated nymphs. Finally, azadirachtin applied at 1  μ g/cm2 to the oviposition surface showed a remarkable oviposition repellency.  相似文献

The side-effects of three neem formulations (Neem-Amin EC, Stardoor and B.P. 20/S) were tested on the mirid predator Macrolophus caliginosus Wagner in the laboratory. Direct toxicity tests on 1st instar nymphs exposed to fresh dry residues on glass plates at different doses demonstrated that all the products are harmful to the insects with LD50 values much lower than the maximum recommended rate (1.217, 0.264, 1.083 mg a.i./l instead of 15, 31.5 and 80 mg a.i./l for Neem-Amin EC, Stardoor and B.P. 20/S, respectively). Moreover a reduction of fecundity of the surviving females was assessed with Neem-Amin EC and B.P. 20/S. Persistence tests were carried out on sharp pepper plants treated at the maximum recommended rate. High mortality was recorded when the insects were introduced onto the plants just after the treatment, but no significant differences compared with the controls were observed 5 days after the treatment and any consequence on the fecundity of surviving females was detected. Our experiments showed how azadirachtin can be noxious to M. caliginosus , but the short persistence makes this active ingredient a promising solution in integrated pest management programmes, when a lapse of time is guaranteed between the treatment and the introduction of the predator.  相似文献

The insecticidal activity of 38 commercialized pesticides against Atractomorpha lata nymphs in Korea was studied. Seven pesticides showed greater than 70% insecticidal activity against A. lata nymphs after 48 h of treatment, and similar results were observed for adult A. lata using the spray method. The insecticidal activity of the seven pesticides, except for thiacloprid, against Oxya sinuosa adults was similar to that against A. lata. Clothianidin, tetraniliprole, and thiacloprid showed higher toxicity when plants and insects were treated together than when insects were treated directly, indicating that pesticides have ingestion toxicity against A. lata. Clothianidin and thiacloprid have been observed to have knockdown effects on both A. lata nymphs and adults. Tetraniliprole showed 86.7% mortality until 7 days after treatment in the residual effect test of pesticide exposure under field conditions. Therefore, this study provides basic data for A. lata control; tetraniliprole was found to be an effective insecticide for the control of A. lata nymphs and adults.  相似文献

To elucidate the specific associations of insects with glandular plants, the relationship between a plant bug and a wild azalea was examined. The plant bug Orthotylus gotoi Yasunaga (Hemiptera: Miridae) was found only on shrubs of Rhododendron macrosepalum Maxim. (Ericaceae) in Kyoto, central Japan. R. macrosepalum has leaves, stems, and sepals with dense stalked glands that trap and kill many arthropods. However, nymphs and adults of O. gotoi are able to move freely on R .  macrosepalum . O. gotoi frequently fed on carcasses of various arthropods trapped on R .  macrosepalum , whereas it rarely fed upon plant tissues. In laboratory experiments, O. gotoi nymphs mainly fed on the carcasses of insects, which resulted in enhanced rates of growth and metamorphosis, and adult O. gotoi lived longer when they were provided with insect carcasses. These observations and experiments suggest that O. gotoi is a scavenger. Abstract Habitat instability associated with seasonal crop succession in broad-acre farming systems presents a problem for the conservation and utilisation of beneficial insects in annual field crops. The present paper describes two experiments used to measure the potential of seven plant species to be utilised as winter refuges to support and conserve the predatory bug Pristhesancus plagipennis (Walker). In the first experiment, replicated plots of canola ( Brassica napus ), red salvia ( Salvia coccinea ), niger ( Guizotia abyssinica ), linseed ( Linum usitatissimum ), lupins ( Lupinus angustifolius ), and lucerne ( Medicago falcata ) were planted in a randomized experiment during Autumn 1998. Upon crop establishment, adults and nymphs of P. plagipennis were released into treatment plots and their numbers were assessed, along with those of their potential prey, throughout the ensuing winter months. Post-release sampling suggested that canola and niger retained a proportion of adult P. plagipennis , while niger, lucerne and canola retained some nymphs. The other plant species failed to support P. plagipennis nymphs and adults postrelease. In the second experiment, niger was compared with two lines of sunflower ( Helianthus annus ). Both sunflower lines harboured significantly higher ( P  < 0.05) densities of P. plagipennis nymphs than did niger. The more successful refuge treatments (sunflower, niger and canola) had an abundance of yellow flowers that were attractive to pollinating insects, which served as supplementary prey on which P. plagipennis were observed to feed. Sunflower and niger also supported high densities of the prey insect Creontiades dilutus (Stål) and provided protective leafy canopies which supplied shelter during the winter months. The potential and limitations for using each plant species as a winter refuge to retain P. plagipennis during winter are discussed.  相似文献

RNA interference (RNAi) is a powerful means to study functional genomics in insects. The delivery of dsRNA is a challenging step in the development of RNAi assay. Here, we describe a new delivery method to increase the effectiveness of RNAi in the Asian citrus psyllid Diaphorina citri. Bromophenol blue droplets were topically applied to fifth instar nymphs and adults on the ventral side of the thorax between the three pairs of legs. In addition to video recordings that showed sucking of the bromophenol blue by the stylets, dissected guts turned blue indicating that the uptake was through feeding. Thus, we called the method topical feeding. We targeted the abnormal wing disc gene (awd), also called nucleoside diphosphate kinase (NDPK), as a reporter gene to prove the uptake of dsRNA via this method of delivery. Our results showed that dsRNA‐awd caused reduction of awd expression and nymph mortality. Survival and lifespan of adults emerged from treated nymphs and treated adults were affected. Silencing awd caused wing malformation in the adults emerged from treated nymphs. Topical feeding as a delivery of dsRNA is highly efficient for both nymphs and adults. The described method could be used to increase the efficiency of RNAi in D. citri and other sap piercing‐sucking hemipterans.  相似文献

Abstract:  Axinoscymnus cardilobus is an indigenous coccinellid predator in South China, which feeds on many whitefly species. Its development, consumption rate, survivorship, longevity and fecundity on a diet of eggs and nymphs of Bemisia tabaci were evaluated. Larval consumption decreased sharply as instars of B. tabaci become bigger, and development time of beetle larvae was significantly different when fed on different stages of B. tabaci . The sex ratio was 0.44. The performances were better after feeding on whitefly eggs than nymphs. Axinoscymnus cardilobus showed good potential for biological control of B. tabaci , especially in greenhouse situations.  相似文献

Abstract.  Opisoplatia orientalis is an ovoviviparous cockroach living in the subtropical areas in Japan. Both adults and nymphs overwinter on Hachijo Island (33°N). The nymphs sampled before and after overwintering showed a similar pattern in frequency distribution of head widths with a definite peak of fifth instars. The present study was conducted to determine how this pattern was formed by investigating the effects of photoperiod and temperature on development and reproduction. Photoperiod influenced the number of nymphal instars, resulting in a longer duration of nymphal development at LD 12 : 12 h than at LD 16 : 8 h. However, the rate of development at each instar was only affected to a small extent by photoperiod and no sign of diapause was detected. It was suggested that the photoperiodic response controlling the number of nymphal instars might have evolved to adjust the timing of adult emergence and reproduction to the favourable season. The prereproductive period and time intervals between nymph depositions were prolonged as temperature declined, but there was no evidence for diapause in adults. Mortality occurred in eggs and embryos inside of the body of the females during winter. Thus, it was inferred that female adults would reset ovarian development in spring and deposit nymphs in summer simultaneously, and these nymphs would reach the fifth instar before winter comes. This winter mortality hypothesis was supported by experiments in which reproductive activity and mortality were monitored for field-collected adults under either constant or changing temperature conditions simulating those in the field.  相似文献

The Bolivian Chaco is part of the endemic region of Chagas disease and an area where pyrethroid resistant Triatoma infestans (Hemiptera: Reduviidae) populations has been reported. The World Health Organization identified these resistant populations as an important focus for research. The objective of this study was to evaluate the residual effect of a micro-encapsulated formulation containing organophosphate active ingredients and a juvenile hormone analogue (Inesfly 5A IGR) on the mortality of T. infestans. Studies took place in rural houses of the Bolivian Chaco that were treated up to 34 months before and evaluated the susceptibility to pyrethroids of the offspring of field collected insects. Thirty houses were randomly selected within three communities to carry out wall bio-assays with T. infestans nymphs. Mortality was recorded 24, 48 and 72 h after wall contact. Eggs laid by females collected in the area were used to obtain first-instar nymphs and carry out pyrethroid susceptibility tests. The wall bio-assays showed that the micro-encapsulated insecticide eliminates T. infestans populations and produces detectable mortality of insects exposed to walls treated 34 months prior to the tests. The discriminant dose of deltamethrin (0.01 mg/mL) showed 65% nymph survival, whereas at the highest tested dose (1.0 mg/mL) 14% of the nymphs survived. These results show that Inesfly 5A IGR is an appropriate tool for the elimination of intradomestic and peridomestic populations of T. infestans resistant to pyrethroids.  相似文献

Male German cockroaches possess secretory glands that secrete fluid into a pair of transverse depressions on the seventh and eighth abdominal tergites. We investigated the effects of altered juvenoid titer during the first part of the last instar on tergal gland secretory tissue development and the production of tergal gland secretion proteins. Male fifth (last) instar nymphs (1-3 days post-emergent) were topically treated with the JH analog pyriproxyfen. Light and transmission electron microscopy demonstrated that treatment with pyriproxyfen produced a visible decrease in the amount of tergal gland tissue present, a deformation of the overall shape of the gland located on tergite seven, and a less orderly arrangement of the secretory cells in the tissue. The protein fraction of tergal gland secretion was examined in pyriproxyfen-treated and control insects 1, 5, and 15 days after the insects molted to the adult stage. Amounts of all tergal secretion proteins were reduced in treated insects.  相似文献

N,N-diethyl-3-methylbenzamide (DEET) is widely used as an insect repellent; however, little is known about its mode of action. On the other hand, nitric oxide (NO) participates in the olfaction transduction pathway of insects. In this work, nitroso-acetyl-cysteine (SNAC), a nitric oxide donor, or dibutyril-cyclic-GMP (db-cGMP), the cyclic nucleotide analog, were applied on fifth instar nymphs of Rhodnius prolixus before exposing them to DEET, to obtain information about the possible role of NO/cGMP system in the olfaction process. In the first place, we exposed the nymphs to several DEET concentrations (70, 700, 1,750, and 3,500 microg/cm2). All these concentrations produced a repellent effect. A decrease in repellency during the course of the experiment was observed when the nymphs were exposed to high concentrations of DEET (700 and 1,750 microg/cm2), suggesting an adaptation phenomenon. The pre-treatment of the insects with 15 microg /insect of SNAC or 2 microg/insect of db-cGMP produced a reduction of the repellency. An increase in locomotor activity was observed in insects exposed to 350 or 700 microg/cm2 DEET. Although exposure to 70 microg/cm2 DEET produced a high repellency response, it did not modify the insects' locomotor activity. Insects treated with two doses of SNAC before being exposed to 350 microg/cm2 of DEET showed no differences in locomotor activity compared to controls.  相似文献

The planthopper Delphacodes kuscheli is the main vector of Mal de Río Cuarto virus in Argentina, disease that severely affects maize production. In this study, we investigated the effects of heat stress on fitness traits and on the number of its obligate yeast‐like symbionts (YLS). The exposition of newly‐hatched nymphs to 35°C for 3 days, a well‐known procedure used to reduce the number of YLS in planthoppers, was applied. To compare different fitness components between control and heat‐treated insects, we estimated nymphal instars development time, nymphal survival, adult body length, longevity, fecundity and fertility. Also, correlates of fitness, as proportion of sexes and wing forms of the emerging adults, were evaluated. In heat‐treated group, the nymphal developmental time increased due to an increase in the fifth instar duration, and the nymphal survival, body length of adults and fecundity were reduced when compared to control. There was a significant association between treatments (control and heat‐treated insects) and wing morphs. The heat treatment successfully reduced the number of YLS in third instar nymphs of D. kuscheli. Our results revealed the negative effect of heat stress on development, survival and reproduction of D. kuscheli and on the load of its YLS endosymbionts suggesting that YLS could play a crucial role in the development and reproduction of these planthoppers.  相似文献

Insect feeding trials were carried out to determine the effects of incorporating a range of plant derived proteins into artificial diets fed to leafhopper and planthopper pests of rice. The lectins Galanthus nivalis agglutinin (GNA) and wheat germ agglutinin (WGA), and the enzyme soy bean lipoxygenase (LPO) were shown to exhibit significant antimetabolic effects towards first and third instar nymphs of rice brown planthopper (Nilaparvata lugens Stål) when incorporated into artificial diet at 0.1% (w/v), 0.1% (w/v) and 0.08% (w/v) levels respectively. The lectin GNA was also shown to exhibit a significant antimetabolic effect towards third instar nymphs of the rice green leafhopper (Nephotettix cinciteps Uhler). A number of inert proteins, lectins, protein inhibitors and enzymes also tested showed relatively little or no effect towards both insects.  相似文献

This study investigated the effects of sub-lethal high temperatures on the development and reproduction of the brown plant hopper Nilaparvata lugens (Stål). When first instar nymphs were exposed at their ULT50 (41.8°C) mean development time to adult was increased in both males and females, from 15.2±0.3 and 18.2±0.3 days respectively in the control to 18.7±0.2 and 19±0.2 days in the treated insects. These differences in development arising from heat stress experienced in the first instar nymph did not persist into the adult stage (adult longevity of 23.5±1.1 and 24.4±1.1 days for treated males and females compared with 25.7±1.0 and 20.6±1.1 days in the control groups), although untreated males lived longer than untreated females. Total mean longevity was increased from 38.8±0.1 to 43.4±1.0 days in treated females, but male longevity was not affected (40.9±0.9 and 42.2±1.1 days respectively). When male and female first instar nymphs were exposed at their ULT50 of 41.8°C and allowed to mate on reaching adult, mean fecundity was reduced from 403.8±13.7 to 128.0±16.6 eggs per female in the treated insects. Following exposure of adult insects at their equivalent ULT50 (42.5°C), the three mating combinations of treated male x treated female, treated male x untreated female, and untreated male x treated female produced 169.3±14.7, 249.6±21.3 and 233.4±17.2 eggs per female respectively, all significantly lower than the control. Exposure of nymphs and adults at their respective ULT50 temperatures also significantly extended the time required for their progeny to complete egg development for all mating combinations compared with control. Overall, sub-lethal heat stress inhibited nymphal development, lowered fecundity and extended egg development time.  相似文献

Chitin synthases are critical enzymes for synthesis of chitin and thus for subsequent growth and development in insects. We identified the cDNA of chitin synthase gene (CHS) in Aphis glycines, the soybean aphid, which is a serious pest of soybean. The full-length cDNA of CHS in A. glycines (AyCHS) was 5802 bp long with an open reading frame of 4704 bp that encoded for a 1567 amino acid residues protein. The predicted AyCHS protein had a molecular mass of 180.05 kDa and its amino acid sequence contained all the signature motifs (EDR, QRRRW and TWGTR) of chitin synthases. The quantitative real-time PCR (qPCR) analysis revealed that AyCHS was expressed in all major tissues (gut, fat body and integument); however, it had the highest expression in integument (~3.5 fold compared to gut). Interestingly, the expression of AyCHS in developing embryos was nearly 7 fold higher compared to adult integument, which probably is a reflection of embryonic molts in hemimetabolus insects. Expression analysis in different developmental stages of A. glycines revealed a consistent AyCHS expression in all stages. Further, through leaf dip bioassay, we tested the effect of diflubenzuron (DFB, Dimilin ®), a chitin-synthesis inhibitor, on A. glycines'' survival, fecundity and body weight. When fed with soybean leaves previously dipped in 50 ppm DFB solution, A. glycines nymphs suffered significantly higher mortality compared to control. A. glycines nymphs feeding on diflubenzuron treated leaves showed a slightly enhanced expression (1.67 fold) of AyCHS compared to nymphs on untreated leaves. We discussed the potential applications of the current study to develop novel management strategies using chitin-synthesis inhibitors and using RNAi by knocking down AyCHS expression.  相似文献

The commercial adoption of transgenic Bacillus thuringiensis (Bt) cotton (Bollgard II®) reduced the use of insecticides to control Helicoverpa spp. However, the ineffectiveness of the Bt toxin against sucking pests such as silverleaf whiteflies (Bemisia tabaci) resulted in a marked increase in B. tabaci populations and in the use of insecticides to control this pest. The effect of the entomopathogenic fungus Aspergillus sp. BC 639 on B. tabaci and beneficial insects (predominantly predatory insects) was studied in commercial cotton field trials. The results showed that oil-based extracts of the entomopathogenic fungus BC 639 control the number of B. tabaci adults and nymphs in commercial transgenic cotton crops. The BC 639 fungus caused 60.0%, 67.2%, and 68.8% mortality in adults, and 54.6%, 62.3%, and 51.7% in nymphs at 7, 14, and 21 days after treatment, respectively, relative to the unsprayed controls. The effect of BC 639 at concentrations of 125, 250, and 500?ml/ha on low-density B. tabaci (~10 nymphs/leaf) did not differ significantly from that of the commercial insecticide (pyriproxifen). However, at higher densities (>50 nymphs per leaf), low concentrations of BC 639 (125 and 250?ml/ha) were not as effective as 500?ml/ha BC 639 in successfully controlling the pest. A simple graphic analysis suggested that the more B. tabaci nymphs per leaf, the fewer adults per leaf, and that once the number of nymphs increased to ~70 per leaf, a negative feedback regulatory effect reduced the survivorship of the nymphs and adults and/or caused the emigration of the adults from the contaminated leaves in search of new resources. Therefore, the ability of BC 639 to control B. tabaci adults and nymphs with minimal effects on predatory insects indicates its potential utility in supplementing integrated pest management programmes for cotton crops.  相似文献

The cuticle, an essential structure for insects, is produced from cuticular proteins and chitin via a series of biochemical reactions. Tweedle genes are important members of the cuticular protein family and have four conserved motifs binding to chitin. Tweedle family genes have been found to play a profound effect on cuticle development. Here, we report that the cuticular protein gene LmTwdl1 of Locusta migratoria belongs to the Tweedle family. In situ hybridization showed that LmTwdl1 is localized to epidermal cells of the cuticle. The expression patterns of LmTwdl1 showed low expression in the cuticle during the early and middle stages of the fifth‐instar nymphs; in contrast, its expression rapidly increased in the late stages of fifth‐instar nymphs. We performed RNA interference to examine the function of LmTwdl1 in locusts. Silencing of LmTwdl1 resulted in high mortality during the molting process before the next stage. Also, the epicuticle of nymphs failed to molt, tended to be thinner and the arrangement of chitin in the procuticle appeared to be disordered compare to the control group. These results demonstrate that LmTwdl1 plays a critical role in molting, which contributes to a better understanding of the distinct functions of the Tweedle family in locusts.  相似文献

In the current study, midgut α-amylase from Sunn pest ( Eurygaster integriceps Puton) (Hemiptera: Scutelleridae), one of the most serious pests of wheat and barley in the wide area of the Near and Middle East, West Asia, and many of the new independent states of central Asia, were purified and characterized. Amylase activity was detected in the midgut of the insects which were collected from both over-wintering sites during winter and feeding insects during spring. Amylase activities in the midgut of over-wintering and feeding insects were 5.71 and 3.43 U/mg protein, respectively. Initially, a native electrophoretic analysis of E. integriceps crude midgut extract showed that there are two major amylase forms in the midgut. Through the sequence of ammonium sulfate precipitation, first by gel filtration chromatography (Sephadex G-75), anion exchange chromatography (diethylaminoethylcellulose) and second by gel filtration chromatography, specific activity of α-amylase of E. integriceps increased 44-fold from approximately 3 to 133 U/mg protein. Analysis of purified amylases by sodium dodecylsulfate polyacrylamide gel electrophoresis showed that these proteins had estimated molecular masses of 49 and 52 kDa. Optimum temperature was determined to be 30–40°C. The optimum pH value was 6.5 and the K mapp for soluble starch was 0.54%.  相似文献

A 13-d continuous dietary exposure bioassay using nymphs of the insidious flower bug, Orius insidiosus (Say) (Heteroptera: Anthocoridae), was developed to assess the potential dietary effects of insecticidal substances that have little or no contact toxicity. The nymphs were fed a bee pollen diet treated with different concentrations of an inorganic stomach poison, potassium arsenate, and a cysteine protease inhibitor, E-64. The results showed that the test system was capable of detecting the dietary effects of both substances on the survival and development of O. insidiosus from the nymph to the adult stage in a dose-dependent manner. For the potassium arsenate treatments, approximately 25% of the nymphs survived and developed to the adult stage by 13 d of dietary exposure at 3.8 microg/g of diet, whereas no test nymphs survived to adulthood at or above 15 microg/g of diet. The assay time required for a 75% mortality response ranged from approximately 7 d at 30 microg/g of diet to 13 d at 3.8 microg/g of diet. For the E-64 treatments, no test insects survived to adulthood at any of the concentration tested (75-600 microg/g of diet) by 13 d of dietary exposure, and the assay time required for a 75% mortality response ranged from 5 to 11 d at dietary rates of 600 and 75 microg/g, respectively. The research presented here describes a robust test system that is useful for evaluating potential adverse effects (or toxicity) of plant-incorporated protectants on nontarget heteropteran predators such as O. insidiosus.  相似文献
