Preparations of Type II immune induced Interferon enhanced the plaque forming cell response of mice to sheep red blood cells both in vivo and in vitro. The enhancement of the antibody response was dependent on the dosage of interferon used and the time of administration of interferon. The expression of the antiviral and immuno-enhancing activities of Type II interferon preparations shared several physical-chemical properties, including pH 2 lability and heat stability. The plaque forming cell response to lipopolysaccharide, a T-independent antigen, could not be enhanced by treatment with Type II interferon. In addition, treatment of spleen cell cultures of nude thymic deficient mice with Type II interferon could not cause an enhancement of the plaque forming cell response to lipopolysaccharide. These data suggest that the immunoenhancing effect of Type II interferon on antibody responses is produced by an effect on T lymphocytes in contrast with the immunosuppressive effect which appears to be mediated through an effect on B lymphocytes.  相似文献

Fumonisins have been reported to have diverse effects on animals, including immunosuppression in chickens and feeder calves; therefore, the effects of fumonisin B1 (FB1) on immune function in BALB/c mice was investigated. When administered i.p. with sheep red blood cells (SRBC), 5 to 100 µg of FB1 reduced the number of plaque-forming cells (PFC) produced against SRBC; however, when administered daily, 1 to 50 µg of FB1 caused a 4 to 12-fold increase in the number of PFC after SRBC injection. Therefore, FB1 is not only immunosuppressive; but also, immunostimulatory. To test the possibility that there may have been an immune response to FB1 as an antigen, FB1 was injected into mice and the number of splenic cells forming rosettes on FB1-treated SRBC was determined. There were dose-dependent increases in the antigen-binding cells, with up to 4.9- and 4.6-fold increases, respectively, upon primary and secondary immunization. FB1-binding immunoglobulins could be detected in sera from some treated mice, but this response was not obtained in every experiment. The effect of Sulforaphane on the immune system was studied using BALB/c mice. Intraperitoneal administration of five doses of Sulforaphane (500 microg/dose/animal/day) was found to enhance the total WBC count (12,950 cells/mm3) on 9th day. Bone marrow cellularity (23 x 10(6) cells/femur) and number of alpha-esterase positive cells (1346.66/4000 cells) were also increased by the administration of Sulforaphane. Treatment with Sulforaphane along with the antigen, sheep red blood cells (SRBC), produced an enhancement in the circulating antibody titre and the number of plaque forming cells (PFC) in the spleen. Maximum number of PFC (315.83 PFC/10(6) spleen cells) was obtained on the 6th day. Administration of Sulforaphane also showed an enhancement in the phagocytic activity of peritoneal macrophages. Moreover administration of Sulforaphane significantly reduced the elevated level of TNF-alpha production by LPS stimulated macrophages. These results indicate the immunomodulatory activity of Sulforaphane.  相似文献

The dynamics of the titers and number of the antibody forming cells was studied within 26 hours after a single injection of anthracycline to immunized animals at various phases of the immune response. The specificity of the "rapid" effect of anthracyclines suggests that the low differentiated precursors of the antibody forming cells are the main target for the rubomycin effect and the immune competent cells in the state of multiplication and differentiation under the effect of the antigen are the main target for the carminomycin effect. The effects of graded doses of a chemically standardized aqueous alcoholic (1:1) root extract (AGB) of Withania somnifera on the immune system of SRBC immunized BALB/c mice were investigated. Mice were administrated AGB orally for 15 days. AGB stimulated cell mediated immunity, IgM and IgG titers reaching peak value with 30 mg/kg b.wt. Flow cytometric analysis of lymphocyte surface markers of T cells (CD3(+), CD4(+) and CD8(+)) and B cells (CD19(+)) indicated prominent enhancement in proliferation and differentiation of lymphocytes. The extract selectively, induced type 1 immunity because it guided enhanced expression of T helper cells (Th)1 cytokines interferon (IFN)-gamma and interleukin (IL)-2 while Th2 cytokine IL-4 observed a moderate decline. Confirmation of Th1 polarization was obtained from augmented levels of IgG2a over IgG1 in the blood sera of AGB treated groups. Withanolide-A, a major constituent of AGB appeared responsible for Th1 skewing effect of the extract as it significantly increased the levels of Th1 cytokines, decreased moderately IL-4 and significantly restored the selective dexamethasone inhibition of Th1 cytokines in mouse splenocytes cultures in vitro. In addition, AGB also strongly activated macrophage functions ex vivo and in vitro indicated by enhanced secretion of nitrite, IL-12 and TNF-alpha. In contrast IL-10 remained unchanged again suggesting that AGB critically influenced Th1 profile of the cytokines. The studies suggested that AGB supports predominantly Th1 immunity with increase in macrophage functions. The standardized root extract of no toxicological consequences might therefore, find useful applications against the intracellular pathogens and in the management of immune suppressed diseases.  相似文献

Immunosuppressive effect of Entamoeba histolytica extract on hamsters   总被引:1,自引:0,他引:1  
The immune response to sheep red blood cells (SRBC) in mice and hamsters injected with an extract of entamoeba histolytica was studied. Both the primary and secondary immune response, measured by anti-SRBC antibody titers, were unaltered in the mouse, while a significant depression of the primary, but not the secondary, response was observed in the hamster. The effect was greatest when the amebic extract (AE) and SRBC were injected on the same day. The number of anti-SRBC rosettes formed in the spleen cells of hamsters treated with both AE and SRBC on day 0 was measured from days 1-16. The response peaked on day 13, while cells from animals injected with SRBC alone gave a maximal response on day 5. The formation of anti-SRBC rosettes in T-lymphocyte-enriched spleen cells treated with anti-gamma globulin serum and complement was almost abolished for the duration of the experiment. It is suggested that the mechanism responsible for this immunosuppressive phenomenon could involve early interference in the afferent limb of the immune response.  相似文献

There is no published data on Cell Mediated Immune Responses in experimental animals to Rhinosporidium seeberi the causative agent of human and animal rhinosporidiosis. The quantitative mouse foot-pad model was used to assay the Delayed-Type Hypersensitivity (DTH) cell-mediated immune response to extracts of purified endospores and sporangia of R. seeberi. Histological examination was used to confirm that the foot-pad reactions were compatible with DTH reactions in the mouse. We report that sonically disintegrated rhinosporidial endospores/sporangia induced DTH responses in the foot-pads of sensitized mice which were comparable in intensity and histological profile to that induced by sheep red blood cells in SRBC sensitized mice. Anti-rhinosporidial antibody was also induced. Filtrates of the soluble antigens in sonicated suspensions failed to evoke a DTH-foot-pad (DTH-FP) response in sensitized mice although an anti-rhinosporidial antibody response to this preparation was detected. Prolonged pre-treatment with sonicated suspensions of endospores and sporangia resulted in a decrease of DTH reactivity as compared with reactions following pre-treatment of a shorter duration. A single injection of pyran copolymer has been shown to greatly increase the number of hemolytic plaque forming cells to sheep erythrocytes (sRBC). Pyran given from 1 day before to 2 days after sRBC inoculation increased both specific activity and plaques/spleen, suggesting that macrophage activation was probably not responsible for the enhancement seen. In addition, pyran given 1 day prior to the primary injection of sRBC was found to increase the secondary response to SRBC given alone. As similar experiments using thymectomized irradiated bone marrow reconstituted mice showed no increase in specific activity following pyran administration, it was unlikely that pyran was acting directly on B cells. Furthermore, experiments measuring the antibody response to Escherichia coli lipopolysaccharide, a thymic independent antigen, pyran did not increase the response to this antigen. In contrast to the above, pyran delayed and depressed cell mediated cytotoxicity to the allogeneic DBA/2 P815 mastocytoma. However, no difference in the titers of cytotoxic antibody against mastocytoma cells was seen between pyran-treated and normal animals. Pyran was mitogenic for spleen cells in vitro. However, following the administration of pyran in vivo, mitogen induced blastogenesis in vitro to PHA and LPS was inhibited and this inhibition was determined to be macrophage-dependent. These results are consistent with a model in which the immunoregulatory effects of pyran act through macrophages and T-lymphocytes.  相似文献

Intravenous administration of sheep red blood cells resulted in a deficient initial antibody response and provoked a much lower concentration of plaque forming cells in the blood of splenectomized dogs than in the control group. The in vitro serum opsonizing activity on Escherichia coli M-S-15 remained normal after splenectomy, and no difference was found between the test and the control group in the intravascular clearance rate of bacteria injected intravenously.  相似文献

Three experiments were conducted in which weanling mice were fed a nutritionally complete diet either ad libitum or in restricted quantities such that they lost about 30% of their initial weight over a 14-day period. In Experiments 1 and 2, half the animals from each group received dietary triiodothyronine (T3) supplements. In Experiment 3, food-intake-restricted mice were fed graded levels of potassium iodide. Malnutrition reduced the number of nucleated cells per spleen, the number of splenic IgG plaque-forming cells (PFC) per 10(6) cells, and the serum antibody titers against sheep red blood cells. T3 supplements increased antibody titers, the number of nucleated cells per spleen, and both IgM and IgG PFC per 10(6) spleen cells in malnourished mice, but had no effect on well-nourished mice. The beneficial effect of T3 was not a result of improved protein, energy, or iodine status in the malnourished mice.  相似文献

Previous studies have shown that cellular and humoral antibody production to type III pneumococcal polysaccharide (SSS-III) is not appreciably altered in neonatally thymectomized mice and is enhanced in animals which have been treated with ALS. In order to determine what effect ALS has on the response to another antigen which does appear to require helper T cells, immunity to E. coli 055:B5 has been investigated. BALB/c mice were injected i.p. with 0.25 ml of ALS on days ?1, 0, and +1 relative to the day of immunization (d.0) with a killed E. coli bacterial vaccine. Splenic plaque forming cells (PFC) and serum hemolysin and hemagglutinin titers were determined 6 days later using sheep erythrocytes which had been coated with purified E. coli lipopolysaccharide (LPS). Mice treated with ALS or normal heterologous serum and immunized with an optimal immunogenic dose of bacteria (150 × 106) had similar numbers of splenic PFC and serum antibody titers. No significant immunosuppressive effect was noted over a wide range of antigen (0.015–1500 × 106) although dose related variations were seen. In contrast to its effect on the response to SSS-III, no enhancement was noted. ALS treated mice which had been simultaneously immunized with E. coli and sheep RBC had specific depression of the T helper dependent response to SRBC but not to LPS. The lack of immunosuppressive effect on antibody production to E. coli LPS provides strong evidence that ALS preferentially acts on T lymphocytes. It further indicates that enhancement occurs with some but not all T helper independent antigens.  相似文献

The specific delayed-type hypersensitivity (DTH) response was evaluated in resistant (A/SN) and susceptible (B10.A) mice intraperitoneally infected with yeasts from a virulent (Pb18) or from a non-virulent (Pb265)Paracoccidioides brasiliensis isolates. Both strains of mice were footpad challenged with homologous antigens. Pb18 infected A/SN mice developed an evident and persistent DTH response late in the course of the disease (90th day on) whereas B10.A animals mounted a discrete and ephemeral DTH response at the 14th day post-infection. A/SN mice infected with Pb265 developed cellular immune responses whereas B10.A mice were almost always anergic. Histological analysis of the footpads of infected mice at 48 hours after challenge showed a mixed infiltrate consisting of predominantly mononuclear cells. Previous infection of resistant and susceptible mice with Pb18 did not alter their DTH responses against heterologous unrelated antigens (sheep red blood cells and dinitrofluorobenzene) indicating that the observed cellular anergy was antigen-specific. When fungal related antigens (candidin and histoplasmin) were tested in resistant mice, absence of cross-reactivity was noted. Female A/J mice were immunized with sheep red blood cells (SRBC) before mating and boosted a few days before delivery. The progeny of these mothers was immunologically tolerant against SRBC at the level of plaque forming cells (PFC). The state of unresponsiveness was antigen specific. Exchange of the newborn mice between control mothers and immunized ones shows clearly that the tolerance is induced by factors present in the milk or the colostrum, respectively. Some others findings suggest that antibodies of the mothers and not small amounts of the injected antigen are responsible for the nearly complete suppression of the immune response of the offspring.  相似文献

Summary Rat fibrosarcoma cells infected with Friend leukemia virus (FV-KMT-17) grow for a short time and then regress spontaneously in syngeneic hosts. This regression mechanism was examined by analyzing the immunomodulating action of the antitumor drugs busulfan (BU) and cyclophosphamide (CY). In preliminary experiments, the optimum dosages of BU and CY for the enhancement of DTH responses to SRBC were 10 mg/kg and 40 mg/kg respectively. Treatment of rats with BU (10 mg/kg) on day 5 induced the regression of KMT-17 cells, while in contrast, the same drug delayed the spontaneous regression of FV-KMT-17 cells. Pretreatment with CY (40 mg/kg) on day 5 did not affect the growth of KMT-17 or FV-KMT-17 cells. After the same treatment schedule, BU inhibited humoral antibody formation against SRBC and against virus-associated antigen (VAA), NK cell activity, and ADCC effector cell activity. On the other hand, CY did not affect the activities of NK cells or ADCC effector cells, although it significantly augmented the DTH responses to SRBC and the production of antibody to VAA but had no effect on production of antibodies to SRBC. Administering dextran 2 hr before giving antigen (sheep red blood cells) to X-irradiated mice repopulated with B and T cells caused alterations in antibody synthesis. Besides a slight increase in the number of cells making IgM antibodies, cells producing IgG antibodies were detected at a time when none are usually present in untreated control animals.Repopulating X-irradiated mice with B cells treated with dextran either in vivo or in vitro and immunizing with sheep red blood cells resulted in twice the background number of sheep red blood cell-specific IgM-plaque-forming cells at 4.5 days of immunization as well as a small number of IgG-plaque-forming cells at 8.5 days. At these times, control animals given only B cells and sheep red blood cells possessed a background number of IgM-plaque forming cells and no IgG plaque-forming cells.Incubating B cells with θ-specific antiserum and complement to remove residual T cells prior to transplantation obliterated dextran's stimulus. Dextran's alterations of immunological responses towards unrelated antigens therefore appears to be manifested through T cells. The latter must be in company with B cells at the time of exposure to dextran. Thus, T cells upon contact with dextran apparently release B cell stimulatory factor (s) responsible for increasing the number of IgM-forming cells and for triggering IgG-forming cells.  相似文献

Weanling mice were fed ad libitum from age 23 to 37 days either an 18 or an 0.6% protein diet. Half the animals in each dietary group received supplemental triiodothyronine (T3, 0.2 mg/kg diet). T3 increased the primary in vivo antibody response of protein-deficient mice to sheep red blood cells, as measured by both splenic plaque-forming cells (PFC) per 10(6) nucleated spleen cells and serum hemagglutinin titers. T3 also increased PFC/spleen in well-nourished mice. The effect on protein-deficient animals was achieved although nutritional status in these animals, as estimated by weight loss and carcass composition, was further impaired by T3 supplementation. These results support the hypothesis that immune functions can be improved independently of nutritional status in severe (wasting) malnutrition. Insofar as T3 was effective in a model of malnutrition that does not reduce serum total or free T3 levels, the phenomenon appears to represent a pharmacological action of the hormone.  相似文献

Humoral antibody response to sheep red blood cells and cellular immune response to bovine serum albumin were studied in Mycoplasma pulmonis infected, adult, male Sprague-Dawley rats. The hemagglutinating antibody response to sheep red blood cells was evaluated at 0, 3, 5, 7, 14, 21 and 28 days postinfection. Antibody titers during all days postinfection were depressed significantly (p less than 0.05) in infected rats as compared to noninfected controls. Cellular immune responses were evaluated by a delayed hypersensitivity response. Rats were sensitized at 0, 3, 5, 7, 14, 21 or 28 days postinfection with bovine serum albumin and challenged with heat aggregated bovine serum albumin 7 days later. Cell-mediated immune responses in infected rats were not significantly different at any point from controls. These results indicate that M. pulmonis infection in rats suppresses the humoral antibody response to sheep red blood cells, but not the cellular immune response to bovine serum albumin.  相似文献

The potential of autoclaved and heat-killed antigen of Leishmania donovani to induce cell-mediated and humoral response has been evaluated in the present study. The vaccines were delivered thrice subcutaneously at an interval of 2 weeks. Two weeks after second booster, BALB/c mice were challenged with 107 stationary phase promastigotes of L. donovani. Significant protection was achieved in immunized mice against L. donovani challenge with 69% to 76% and 59% to 64% reduction in parasite load in the liver and spleen respectively. Immunization induced significantly higher level of delayed type hypersensitivity (DTH) response in mice immunized with heat-killed antigen followed by autoclaved antigen. The immune response was assessed by quantifying Leishmania-specific antibodies and cytokine production. The antibody response was predominantly of IgG type with increased IgG2a production and lesser amount of IgM. The immunization preferentially stimulates the production of IFN-γ and IL-2 in splenocytes which suggests a Th1 type response with a concomitant down-regulation of IL-10 and IL-4. These results indicate a potential for the heat-killed and autoclaved antigen as a vaccine which could trigger cell-mediated immune response.  相似文献

Rooibos tea contains a large amount of flavonoids and acts as a potent antioxidant. In this study, we examined the effects of Rooibos tea extract on antigen-specific antibody production and cytokine generation in vitro and in vivo. The primary in vitro anti-ovalbumin (anti-OVA) or sheep red blood cell (SRBC) antibody production in murine splenocytes was markedly stimulated by the addition of the tea extract at concentrations of 1-100 microg/ml. On the other hand, a nonspecific antibody response elicited with lipopolysaccharide (LPS) in purified splenic B-cells was not modified by the extract. Rooibos tea extract caused an increase in the generation of interleukin 2 (IL-2) both in OVA- and anti-CD3-primed splenocytes at concentrations ranging from 10 microg/ml to 1000 microg/ml. In contrast, this tea extract suppressed the generation of interleukin 4 (IL-4) in OVA-primed splenocytes. Moreover, the reduction of OVA-induced antibody production in serum of the cyclosporin A (CyA) -treated rats can be significantly restored and the IL-2 generation in murine splenocytes was stimulated, following oral administrations of Rooibos tea extract. Thus, our findings suggested that Rooibos tea extract may facilitate the antigen-specific antibody production through selective augmentation of IL-2 generation both in vitro and in vivo. Collectively, Rooibos tea intake may be of value in prophylaxis of the diseases involving a severe defect in Th1 immune response such as cancer, allergy, AIDS, and other infections.  相似文献

A study was made of the influence of yeast polysaccharides of definite structure and mol wt on the accumulation of anitbody forming cells (AFC) in the spleen of mice belonging to different strains. Yeast mannans Rh. rubra and Sp. species, and also glucan Aur. pullulans with all or chiefly beta-bonds between the monosaccharide units were capable of activating the cell antibody formation, this being expressed in increased AFC production in the animals with a high immune reaction to the administration of sheep red blood cells and in the intensification of the immune response in mice with a low reaction to the antigen administered. The activity of dextrans directly depended on their mol wt. Besides, there was revealed a different reaction to polysaccharides in the animals of different genotypes.  相似文献
